    • Subcortical brain structures are integral to motion, consciousness, emotions and learning. We identified common genetic variation related to the volumes of the nucleus accumbens, amygdala, brainstem, caudate nucleus, globus pallidus, putamen and thalamus, using genome-wide association analyses in almost 40,000 individuals from CHARGE, ENIGMA and UK Biobank. We show that variability in subcortical volumes is heritable, and identify 48 significantly associated loci (40 novel at the time of analysis). Annotation of these loci by utilizing gene expression, methylation and neuropathological data identified 199 genes putatively implicated in neurodevelopment, synaptic signaling, axonal transport, apoptosis, inflammation/infection and susceptibility to neurological disorders. This set of genes is significantly enriched for Drosophila orthologs associated with neurodevelopmental phenotypes, suggesting evolutionarily conserved mechanisms. Our findings uncover novel biology and potential drug targets underlying brain development and disease.

  • DNA-streckkodning av marina växtplankton : Ett nytt verktyg i miljöövervakningen

    • Växtplankton utgör grunden i den marina näringsväven och används världen över för att bestämma miljöstatus i hav och sjöar. De ingår exempelvis som en kvalitetsfaktor i EU:s vattendirektiv och havsmiljödirektiv. Det finns långa tidsserier av växtplanktonövervakning där analyserna utförts med mikroskopi. Det sker nu en snabb internationell utveckling av DNA-metoder för att övervaka växtplankton. Målsättningen med detta projekt var att utveckla en praktisk och robust DNA analys­metod som kan implementeras i svensk marin miljöövervakning. Vårt tillvägagångssätt var att följa med på ordinarie marina miljöövervakningsexpeditioner under ett års tid (2019–2020) och ta parallella havsvattenprov för så kallad DNA-streckkodning av växtplankton. Sammanlagt tog vi prov vid 19 övervakningsstationer som var spridda från Bottenviken i norr till Skagerrak i söder. Provtagningsfrekvensen var cirka 1 gång per månad. Praktiska metoder utarbetades för fältprovtagning, DNA-extraktion, sekvensering, bioinformatisk analys och taxo­nomisk annotering. Vi har även tagit fram system för datahantering hos nationell datavärd och gett förslag på en ny datatyp för nationellt datavärdskap för marin­biologi och oceanografi vid Svenskt Oceanografiskt Datacentrum (https://sharkweb. smhi.se/hamta-data/). En viktig del har varit att jämföra resultaten av DNA-streckkodning och mikroskopi. Resultaten visar att DNA-streckkodning ger ungefär dubbelt så högt biodiversitetsmått än mikroskopering, även om det skiljer sig åt mellan olika grupper av växtplankton. För att undersöka om DNA-streckkodning kan användas för kvantitativ analys tillsatte vi en intern standard till proverna bestående av syntetiskt DNA, men eftersom resultatet varierade så behöver man arbeta vidare med detta. Den relativa fördelningen av vanliga eukaryota växtplanktongrupper visade sig ha relativt bra överensstämmelse mellan DNA-streckkodning och mikroskopimåttet kolbiomassa, medan biovolym och abundans skiljde sig åt mer. DNA-streckkodning visade sig ge detaljerade utbredningsmönster av skadliga alger, till exempel för släktet Prymnesium bland häftalgerna (Coccolithophyceae). Vi har inom projektet kunnat utvärdera ekologiska drivkrafter för växtplankton­samhällets diversitet och artsammansättning, genom att miljöövervakningen mäter många fysikalisk-kemiska parametrar. Både salthalt och närsalter visade sig ha stor inverkan på växtplanktonsamhällets sammansättning och diversitet. Sammanfattningsvis ser vi att den framtagna DNA-streckkodningsmetoden skulle vara ett bra komplement till den etablerade växtplanktonövervakningen.

  • Dielectric strength of γ-radiation cross-linked, high vinyl-content polyethylene

    • We explore γ-radiation cross-linking of high vinyl-content low-density polyethylene (LDPE) and its potential use as a high-voltage insulation material. Of the three investigated resins containing 1, 0.5 and 0.17 vinyl groups per 1000 carbons, respectively, only the highest vinyl-content material featured a sufficiently high gel content of more than 70% and hot-set elongation below 175%, when cross-linked with a γ-radiation dose of at least 68 kGy. Differential scanning calorimetry (DSC) and small-angle X-ray scattering (SAXS) reveal that neither the crystallinity nor the lamellar thickness of the highest vinyl-content LDPE are negatively affected by γ-radiation cross-linking. As a result, we find that the dielectric strength, as characterized by electrical tree initiation experiments, can be maintained upon γ-radiation cross-linking.

  • Coincidence spectroscopy for increased sensitivity in radionuclide monitoring

    • The majority of the energy in a nuclear explosion is released in the immediate blast and the initial radiation accounts. The remaining fraction is released through radioactive decay of the explosion's fission products and neutron activation products over a longer time span. This allows for the detection of a nuclear explosion by detecting the presence of residual decay. Radionuclide monitoring stations for detection of radioactive emissions to the atmosphere is thereby an important tool in the verification of compliance with nuclear disarmament treaties. In particular, the globally spanning radionuclide station network of the International Monitoring System (IMS) has been implemented for verification of the Comprehensive Nuclear-Test-Ban Treaty.High Purity Germanium (HPGe) detectors are workhorses in radionuclide monitoring. The detection of characteristic gamma rays can be used to disclose the presence of signature nuclides produced innuclear weapon tests. A particular development that has potential to improve the sensitivity of radionuclide monitoring is the coincidence technique where decaying nuclides that emit several coincident gamma rays can be detected at much smaller activity concentrations than with conventional gamma spectroscopy.In this project, dedicated gamma-gamma coincidence detectors are being developed, utilizing electronically segmented HPGe detectors. These detectors are expected to be highly sensitive to low-activity samples of nuclides that present coincident emissions of gamma rays. In this paper we present the concept, define performance parameters, and explore the performance of such detectors to a subset of radionuclides of particular CTBT relevance. In addition, we discuss the path forward in developing a next generation gamma-gamma coincidence spectroscopy system of segmented HPGe.

    • Poly(methyl methacrylate) microspheres have been prepared by the internal phase separation method using either of the three conventional dispersants poly(vinyl alcohol) (PVA), poly(methacrylic acid) (PMAA), or the amphiphilic block copolymer poly(methyl methacrylate)-block-poly(sodium methacrylate). The block copolymer based microsphere, which has a polyelectrolyte brush on the surface, was surface modified with up to two poly(diallyldimethylammonium chloride)-poly(sodium methacrylate) bilayers. The microspheres were loaded with the hydrophobic dye 2-(4-(2-chloro-4-nitrophenylazo)-N-ethylphenylamino)ethanol (Disperse Red 13) and its release from aqueous dispersions of microspheres with the different surface compositions was measured by spectrophotometry. The burst fraction, burst rate and the diffusion constant were determined from a model combining burst and diffusive release. Out of the three dispersants, the block copolymer gave the slowest release of the dye, with respect to both burst release and diffusive release. A very pronounced further reduction of the diffusion constant was obtained by applying polyelectrolyte multilayers on top of the microspheres. However, the diffusion constant was very weakly dependent on further polyelectrolyte adsorption and one polyelectrolyte bilayer appeared to suffice.

    • The formation of strongly elastic physical gels based on poly(alkylene oxide)-grafted hyaluronan or carboxymethylcellulose, exhibiting both shear-thickening and strain-hardening have been studied using rheometry and explained using a slightly different interpretation of the transient network theory. The graft copolymers were prepared by a quantitative coupling reaction. Their aqueous solutions displayed a thermoreversible continuous transition from Newtonian fluid to viscoelastic solid which could be controlled by the reaction conditions. The evolution of all material properties of the gel could be categorized into two distinct temperature regimes with a fast evolution at low temperatures followed by a slow evolution at high temperatures. The activation energy of the zero shear viscosity and the relaxation time of the graft inside the interconnecting microdomains were almost identical to each other in both temperature regimes. This suggests that the number of microdomains remained approximately constant whereas the aggregation number inside the microdomains increased according to the binodal curve of the thermosensitive graft.

A series of thermoresponsive graft copolymers, gelling at physiological conditions in aqueous solution and cell growth media, have been synthesized using quantitative coupling between a small set of amino-functionalized poly(alkylene oxide) copolymers (PAO) and the carboxylate of the biologically important polysaccharides (PSa) carboxymethylcellulose and the less reactive hyaluronate. Quantitative grafting enables the establishment of structure-function relationship which is imperative for controlling the properties of in situ gelling hydrogels. The EDC/NHS-mediated reaction was monitored using SEC-MALLS, which revealed that all PAOs were grafted onto the PSa backbone. Aqueous solutions of the graft copolymers were Newtonian fluids at room temperatures and formed reversible physical gels at elevated temperatures which were noncytotoxic toward chondrocytes. The established structure-function relationship was most clearly demonstrated by inspecting the thermogelling strength and the onset of thermogelling in a phase diagram. The onset of the thermogelling function could be controlled by the global PAO concentration, independent of graft ratio.

  • The impact of interfaces in laminated packaging on transport of carboxylic acids

    • The permeability of oleic and acetic acid through low density polyethylene (LDPE) and ethylene acrylic acid (EAA) have been measured using diffusion cells. In addition, the permeability through combinations of LDPE and EAA in the form of laminates with different numbers of layers has been determined. Oleic acid shows an almost 30 times higher permeability compared to acetic acid, which was partly explained by the adsorption of oleic acid to the film surface during the permeability experiment. In addition, the permeability is lower for both oleic and acetic acid in the laminates compared to the pure films. The decreased permeability can be explained by the presence of crystalline domains close to the interface. This is supported by SAXS data which suggests an ordering of polymer chains in the EAA film close to the interface. In summary, the results show that it is possible to create barrier materials with decreased permeability, which is interesting for example in the packaging industry.

  • Assessment of bio-based fuel ash effects on magnesia refractory materials in quicklime production kilns

    • Limestone is calcined into quicklime in lime kilns at temperatures above 1000°C. Heat is supplied through combustion inside the kilns, which are insulated with a lining of refractory bricks to mitigate heat loss and to protect the kiln from the hot, chemically aggressive, and mechanically abrasive environment. While magnesia bricks have proven to be effective lining materials, they are still susceptible to extensive wear in lime kilns, especially in the burn zone. Refractory corrosion, in particular, can occur when melted fuel ash infiltrates the refractory materials through pores and small cracks. This resultant wear can lead to high maintenance and operational costs, often due to unplanned kiln shutdowns.To reduce the release of fossil-based carbon dioxide into the atmosphere from lime production kilns, there is a growing interest in introducing bio-based fuels with only relatively minor modifications to the process. Biomass fuels can be sourced from bio-based waste streams from industries or be specifically cultivated for combustion. However, the ash content and properties of bio-based fuels tend to be problematic from an ash chemistry perspective. Therefore, before introducing a new fuel source, it is essential to investigate its potential effects on the kiln lining material. In this thesis work, the interactions between melted olive pomace ash and coal ash with commercially available magnesia refractory materials, primarily composed of periclase (MgO) with minor amounts of spinel (MgAl2O4), were studied. A procedure for quantifying the intrusion depths was described. Refractory samples were exposed to the fuel ashes under a simulated lime kiln atmosphere with high CO2 levels at 1200 and 1400°C for 15 and 60 minutes. Cold crushing strength tests were conducted on refractory samples exposed to coal and olive pomace ash, along with CaO powder, at 1400°C for 96 hours. Additionally, postmortem analyses of spent MgO-based refractory bricks were carried out to investigate their chemical characterization and resistance to slag attack after serving as part of the lining in a quick lime rotary kiln for six months.The morphology and elemental compositions of the exposed samples were examined using scanning electron microscopy and energy-dispersive X-ray spectroscopy. Crystalline phases were investigated with powder X-ray diffraction. Thermodynamic equilibrium calculations were performed to further investigate the ash’s melting behavior in contact with the refractories.The results indicated that the potassium-rich olive pomace ash exhibited a greater tendency to infiltrate compared to the silicon-rich coal ash, while the latter formed a glassy melt layer on top of the refractory samples. The ash slags primarily infiltrated through the porous matrix and grain boundaries of the refractory materials. Also, refractory phases were observed in both types of ashes, indicating migration of refractory constituents. K2MgSiO4 phase was found in the olive pomace ash residues on top of the samples, both for the 1200°C and 1400°C exposures. Similarly, Al6Si2O13 phase was dominant in the residual coal ash, in both the 1200°C and 1400°C exposed samples. None of these phases were present in the original ashes.The results of the postmortem analysis revealed that there was no potassium (K) from the fuel ash present on the hot side of the refractory bricks. However, some K was detected in the middle and back parts of the bricks. On the other hand, some phases, possibly connected to degradation, could be found on the hot side of the bricks, where most of the wear was observed.The crushing strengths increased after exposure for all samples, except for those exposed exclusively to coal ash. One possible explanation for this is that the refractory materials exhibited a sintered structure, as a result of their interaction with the ashes and CaO. However, in the samples exposed to coal ash, forsterite (Mg2SiO4) was identified, which can be considered a corrosion product.

    • Magnesium oxide (MgO)-based refractories are commonly used in quicklime and cement rotary kilns. At the high temperatures in the kiln burn zone, the infiltration of molten fuel ash into the refractory can occur. Subsequent chemical interactions can cause refractory wear that inflicts high maintenance costs and loss of production. To improve refractory reliability, it is necessary to increase the understanding of the interactions between fuel ash slag and refractory liner materials. Three commercially available MgO-based refractory materials were exposed to coal ash at 1200 °C and 1400 °C for between 15 and 60 min under a CO2-rich gaseous environment. Hot slag from the coal ash infiltrated the refractories and the infiltration depths were estimated with scanning electron microscope with energy dispersive X-ray spectroscopy. Based on detailed elemental and microstructure analyses, the interactions between ash and refractory were examined. Molten silicates infiltrated the refractory through grain boundaries and pores into depths of up to 2.8 mm. Powder X-ray diffraction of the exposed refractory samples indicated that MgO grains reacted with SiO2-containing phases to form Mg2SiO4. This was identified as a corrosion product whose formation was supported by thermochemical equilibrium calculations. Elevated Mg content was found in the ash residue on top of the samples, indicating the dissolution or dislocation of refractory components. In addition, phases such as MgO were identified in the ash residue.

    • MgO-based refractories are used in lime kilns to withstand the high temperature and chemical environment. Efforts to reduce CO2 emissions have led to an increased interest to use bio-based fuels as alternatives to traditional fossil sources. The potential for refractory corrosion from a potassium-rich biomass ash was investigated by studying the infiltration of olive pomace ash into magnesia/spinel refractories. Refractory samples were exposed to the ash at up to 1400 °C for 15–60 min in a CO2–rich atmosphere. Molten ash infiltrated the refractories through pores and grain boundaries to a depth of up to 9.6 mm, which was quantified with a new systematic procedure. The phase KAlO2 was identified inside the refractories after exposure, indicating an attack of spinel components by potassium. Phases found in the ash residues also indicated the migration of refractory constituents. Thermochemical equilibrium calculations were also used to investigate the ash/refractory chemistry.

    • In quicklime production, limestone is calcined at temperatures above 1000°C, depending on the desired product quality. Heat is supplied to the process from combustion inside the kilns that are insulated to reduce heat loss. The kilns are lined with insulating refractory bricks to withstand the hot, chemically aggressive, and mechanically abrasive environment. Magnesia bricks have emerged as well-performinglining materials, but they are still prone to extensive wear in kilns that are operated at higher temperatures. In particular, refractory corrosion can be caused by fuel ash infiltration that results inmaterial wear, which can incur high maintenance and operational costs through unplanned shutdowns of the kilns. At the same time, to reduce the release of fossil-based carbon to the atmosphere, it is of interest to introduce bio-based fuels into the kilns with only relatively small modifications to the process. Biobased waste streams from existing industries are preferable rather than biomass grown with the sole purpose of combustion. The ash content and properties of these types of waste residues do, however, tend to be problematic from a fuel ash chemistry point of view. Therefore, before introducing a new fuel, their potential effects on kiln lining material should be investigated. In this study, the infiltration of olivepomace ash and coal ash into commercially available refractory materials composed of mainly periclase(MgO) with minor amounts of spinel (MgAl2O4) were compared. They were exposed to the fuel ashes under a simulated lime kiln high CO2 atmosphere at 1200 and 1400°C for 15 and 60 minutes. The morphology and elemental composition of the exposed samples were investigated with scanning electron microscopy equipped with energy-dispersive X-ray spectroscopy. Ash-forming elements infiltrated the porous parts of the materials. The analytical results are complemented with thermodynamic equilibrium calculations to investigate the ash melting behavior. Crystalline phases in the residual ashes were investigated with X-ray diffraction. Refractory phases could be found in both ashes, indicating migration of refractory constituents. Olive pomace ash formed new crystalline compounds together with the refractory components whereas this was not observed for the coal ash, indicating that the former is more of a risk for material failure.

  • A comprehensive dataset on spatiotemporal variation of microbial plankton communities in the Baltic Sea

    • The Baltic Sea is one of the largest brackish water environments on earth and is characterised by pronounced physicochemical gradients and seasonal dynamics. Although the Baltic Sea has a long history of microscopy-based plankton monitoring, DNA-based metabarcoding has so far mainly been limited to individual transect cruises or time-series of single stations. Here we report a dataset covering spatiotemporal variation in prokaryotic and eukaryotic microbial communities and physicochemical parameters. Within 13-months between January 2019 and February 2020, 341 water samples were collected at 22 stations during monthly cruises along the salinity gradient. Both salinity and seasonality are strongly reflected in the data. Since the dataset was generated with both metabarcoding and microscopy-based methods, it provides unique opportunities for both technical and ecological analyses, and is a valuable biodiversity reference for future studies, in the prospect of climate change.

    • Metapopulation theory developed in terrestrial ecology provides applicable frameworks for interpreting the role of local and regional processes in shaping species distribution patterns. Yet, empirical testing of metapopulation models on microbial communities is essentially lacking. We determined regional bacterioplankton dynamics from monthly transect sampling in the Baltic Sea Proper using 16S rRNA gene sequencing. A strong positive trend was found between local relative abundance and occupancy of populations. Notably, the occupancy-frequency distributions were significantly bimodal with a satellite mode of rare endemic populations and a core mode of abundant cosmopolitan populations (e.g. Synechococcus, SAR11 and SAR86 clade members). Temporal changes in population distributions supported several theoretical frameworks. Still, bimodality was found among bacterioplankton communities across the entire Baltic Sea, and was also frequent in globally distributed datasets. Datasets spanning waters with widely different physicochemical characteristics or environmental gradients typically lacked significant bimodal patterns. When such datasets were divided into subsets with coherent environmental conditions, bimodal patterns emerged, highlighting the importance of positive feedbacks between local abundance and occupancy within specific biomes. Thus, metapopulation theory applied to microbial biogeography can provide novel insights into the mechanisms governing shifts in biodiversity resulting from natural or anthropogenically induced changes in the environment.

    • In this paper, we present our process to establish a PICO and a sentiment annotated corpus of clinical trial publications. PICO stands for Population, Intervention, Comparison and Outcome - these four classes can be used for more advanced and specific search queries. For example, a physician can determine how well a drug works only in the subgroup of children. Additionally to the PICO extraction, we conducted a sentiment annotation, where the sentiment refers to whether the conclusion of a trial was positive, negative or neutral. We created both corpora with the help of medical experts and non-experts as annotators.

    • We present detailed continuous wave (cw) and transient photoinduced absorption (PA) measurements in thin films of a novel alternating polyfluorene copolymer, poly[2,7-(9,9-dioctyl-fluorene)-alt-5,5-(4',7'-di-2-thienyl-2',1',3-benzo-thiadiazole)] (DiO-PFDTBT), and its blends with the sol. fullerene deriv. [6,6]-phenyl-C61-butyric acid Me ester (PCBM) in wt. ratios of 1:0, 4:1 and 1:4. We measure the frequency, intensity and temp. dependence of the PA signal in the frequency domain, and compare with the results obtained from the transient PA decay measurements in the time domain. In all blends, the PA spectrum shows a broad high energy PA band ranging from .apprx.1 eV to 2 eV as well as a low energy band peaking at .apprx.0.35 eV. We attribute the low energy band to the P1 transition of polarons and part of the high energy band to the correlated P2 transition of polarons. Both frequency and time domain measurements show that the high energy band has two decay components, a faster component in the microsecond time regime and a slower component in the millisecond time regime. The slow component is strongly dispersive, whereas the fast component is practically non-dispersive. [on SciFinder (R)]

  • Determinants of morbidity and mortality following emergency abdominal surgery in children in low-income and middle-income countries

    • Background: Child health is a key priority on the global health agenda, yet the provision of essential and emergency surgery in children is patchy in resource-poor regions. This study was aimed to determine the mortality risk for emergency abdominal paediatric surgery in low-income countries globally.Methods: Multicentre, international, prospective, cohort study. Self-selected surgical units performing emergency abdominal surgery submitted prespecified data for consecutive children aged <16 years during a 2-week period between July and December 2014. The United Nation's Human Development Index (HDI) was used to stratify countries. The main outcome measure was 30-day postoperative mortality, analysed by multilevel logistic regression.Results: This study included 1409 patients from 253 centres in 43 countries; 282 children were under 2 years of age. Among them, 265 (18.8%) were from low-HDI, 450 (31.9%) from middle-HDI and 694 (49.3%) from high-HDI countries. The most common operations performed were appendectomy, small bowel resection, pyloromyotomy and correction of intussusception. After adjustment for patient and hospital risk factors, child mortality at 30 days was significantly higher in low-HDI (adjusted OR 7.14 (95% CI 2.52 to 20.23), p<0.001) and middle-HDI (4.42 (1.44 to 13.56), p=0.009) countries compared with high-HDI countries, translating to 40 excess deaths per 1000 procedures performed.Conclusions: Adjusted mortality in children following emergency abdominal surgery may be as high as 7 times greater in low-HDI and middle-HDI countries compared with high-HDI countries. Effective provision of emergency essential surgery should be a key priority for global child health agendas.

    • In this paper we describe and evaluate a tool for paradigm induction and lexicon extraction that has been applied to Old Swedish. The tool is semi-supervised and uses a small seed lexicon and unannotated corpora to derive full inflection tables for input lemmata. In the work presented here, the tool has been modified to deal with the rich spelling variation found in Old Swedish texts. We also present some initial experiments, which are the first steps towards creating a large-scale morphology for Old Swedish.

    • Språkbanken, the Swedish Language Bank, is a language technology research unit at the Department of Swedish, University of Gothenburg. We develop language resources – such as corpora, lexical resources, and analytical tools – for all variants of Swedish, from Old Swedish laws to present-day social media. Historical texts offer exciting theoretical and methodological challenges for language technology because they often defy the assumption inherent in most automatic analysis tools that the texts contain a standardized written language. In this article, we describe our ongoing work on the development of annotated historical corpora, as well as our efforts on linking various resources (both corpora and lexical resources). This research advances the state of the art of language technology as well as enables new research for scholars in other disciplines.

    • We present a case study on supervised classification of Swedish pseudo-coordination (SPC). The classification is attempted on the type-level with data collected from two data sets: a blog corpus and a fiction corpus. Two small experiments were designed to evaluate the feasability of this task. The first experiment explored a classifier’s ability to discriminate pseudo-coordinations from ordinary verb coordinations, given a small labeled data set created during the experiment. The second experiment evaluated how well the classifier performed at detecting and ranking SPCs in a set of unlabeled verb coordinations, to investigate if it could be used as a semi-automatic discovery procedure to find new SPCs.

    • © 2016 Federation of European Biochemical Societies Arsenite is widely present in nature; therefore, cells have evolved mechanisms to prevent arsenite influx and promote efflux. In yeast (Saccharomyces cerevisiae), the aquaglyceroporin Fps1 mediates arsenite influx and efflux. The mitogen-activated protein kinase (MAPK) Hog1 has previously been shown to restrict arsenite influx through Fps1. In this study, we show that another MAPK, Slt2, is transiently phosphorylated in response to arsenite influx. Our findings indicate that the protein kinase activity of Slt2 is required for its role in arsenite tolerance. While Hog1 prevents arsenite influx via phosphorylation of T231 at the N-terminal domain of Fps1, Slt2 promotes arsenite efflux through phosphorylation of S537 at the C terminus. Our data suggest that Slt2 physically interacts with Fps1 and that this interaction depends on phosphorylation of S537. We hypothesize that Hog1 and Slt2 may affect each other's binding to Fps1, thereby controlling the opening and closing of the channel.

    • BackgroundThe immunogenicity of repeated vaccination and hybrid immunity in vulnerable patients remains unclear.MethodsWe studied the impact of iterative Covid-19 mRNA vaccination and hybrid immunity on antibody levels in immunosuppressed subjects. Patients with liver cirrhosis (n = 38), survivors of allogeneic haematopoietic stem cell transplantation (allo-HSCT) (n = 36) and patients with autoimmune liver disease (n = 14) along with healthy controls (n = 20) were monitored for SARS-CoV-2-S1 IgG after their 1st-3rd vaccine doses, 31 of whom became infected with the Omicron variant after the 2nd dose. Ten uninfected allo-HSCT recipients received an additional 4th vaccine dose.ResultsUnexpectedly, immunosuppressed patients achieved antibody levels in parity with controls after the 3rd vaccine dose. In all study cohorts, hybrid immunity (effect of vaccination and natural infection) resulted in approximately 10-fold higher antibody levels than vaccine-induced immunity alone.ConclusionsThree doses of the Covid-19 mRNA vaccine entailed high antibody concentrations even in immunocompromised individuals, and hybrid-immunity resulted further augmented levels than vaccination alone.

  • Adaptive radiation of Darwin's finches revisited using whole genome sequencing

    • We recently used genome sequencing to study the evolutionary history of the Darwin's finches. A prominent feature of our data was that different polymorphic sites in the genome tended to indicate different genetic relationships among these closely related species. Such patterns are expected in recently diverged genomes as a result of incomplete lineage sorting. However, we uncovered conclusive evidence that these patterns have also been influenced by interspecies hybridisation, a process that has likely played an important role in the radiation of Darwin's finches. A major discovery was that segregation of two haplotypes at the ALX1 locus underlies variation in beak shape among the Darwin's finches, and that differences between the two haplotypes in a 240 kb region in blunt and pointed beaked birds involve both coding and regulatory changes. As we review herein, the evolution of such adaptive haplotypes comprising multiple causal changes appears to be an important mechanism contributing to the evolution of biodiversity.

Molecular techniques now allow for obtaining genome sequences of uncultivated prokaryotic taxa, facilitating in-depth analyses that may ultimately improve our understanding of these key organisms. Results: We compared results from two culture-independent strategies for recovering bacterial genomes: single-amplified genomes and metagenome-assembled genomes. Single-amplified genomes were obtained from samples collected at an offshore station in the Baltic Sea Proper and compared to previously obtained metagenome-assembled genomes from a time series at the same station. Among 16 single-amplified genomes analyzed, seven were found to match metagenome-assembled genomes, affiliated with a diverse set of taxa. Notably, genome pairs between the two approaches were nearly identical (average 99.51% sequence identity; range 98.77-99.84%) across overlapping regions (30-80% of each genome). Within matching pairs, the single-amplified genomes were consistently smaller and less complete, whereas the genetic functional profiles were maintained. For the metagenome-assembled genomes, only on average 3.6% of the bases were estimated to be missing from the genomes due to wrongly binned contigs. Conclusions: The strong agreement between the single-amplified and metagenome-assembled genomes emphasizes that both methods generate accurate genome information from uncultivated bacteria. Importantly, this implies that the research questions and the available resources are allowed to determine the selection of genomics approach for microbiome studies.

    • File-sharing has become synonym with the “digital economy” where large music conglomerates as well as certain artist voice strong concern over the impact on their bottom line. This research analyzes the music industry, which has been heavily impacted by a major technological shift i.e. the invention, and rollout, of the Internet. We look to the technological shift that has enabled the downloading phenomenon, as well as analyze the uniqueness of the music industries situation. By comparing the book publishing industry as well as the software industry, which are arguably also influenced by intellectual property rights and plagiarism, we try to find similarities as well as dissimilarities with the music industry. We find that the music industry has used alliances as well as Mergers and Acquisitions in order to consolidate their positions in an attempt to slow down change. There is no consensus on the exact extent of ill effects of file sharing. We point to an unwillingness to achieve convergence of purpose between the IT-community and the much of the music industry. Finally we point to the historical fact that consumers always get what they want in the end, which should indicate a need to find a viable e-commerce solution.

    • We discuss what happens at the end of the QCD cascades. We show that, with just a few reasonable assumptions, the emission of soft gluons is constrained to produce an ordered field in the form of a helix. We describe how to modify the Lund fragmentation scheme in order to fragment such a field. Our modified fragmentation scheme yields results which are consistent with current experimental measurements, but predicts at least one signature which should be observable.

    • We derive a partition function for the Lund fragmentation model and compare it with that of a classical gas. For a fixed rapidity “volume” this partition function corresponds to a multiplicity distribution which is very close to a binomial distribution. We compare our results with the multiplicity distributions obtained from the JETSET Monte Carlo for several scenarios. Firstly, for the fragmentation vertices of the Lund string. Secondly, for the final state particles both with and without decays.

    • We show how a difference in the correlation length longitudinally and transversely, with respect to the jet axis in e+e− annihilation, arises naturally in a model for Bose-Einstein correlations based on the Lund string model. In genuine three-particle correlations the difference is even more apparent and they provide therefore a good probe for the longitudinal stretching of the string field. The correlation length between pion pairs is found to be rather independent of the pion multiplicity and the kaon content of the final state.

    • Cerebrospinal fluid (CSF) neurofilament light (NfL) concentration has reproducibly been shown to reflect neurodegeneration in brain disorders, including Alzheimer's disease (AD). NfL concentration in blood correlates with the corresponding CSF levels, but few studies have directly compared the reliability of these 2 markers in sporadic AD. Herein, we measured plasma and CSF concentrations of NfL in 478 cognitively unimpaired (CU) subjects, 227 patients with mild cognitive impairment, and 113 patients with AD dementia. We found that the concentration of NfL in CSF, but not in plasma, was increased in response to Aβ pathology in CU subjects. Both CSF and plasma NfL concentrations were increased in patients with mild cognitive impairment and AD dementia. Furthermore, only NfL in CSF was associated with reduced white matter microstructure in CU subjects. Finally, in a transgenic mouse model of AD, CSF NfL increased before serum NfL in response to the development of Aβ pathology. In conclusion, NfL in CSF may be a more reliable biomarker of neurodegeneration than NfL in blood in preclinical sporadic AD.

    • Background/Aims Coronary heart diseases, including myocardial infarction, remain a major health problem worldwide. Little is known about the cardiac care experiences post myocardial infarction of younger people and their next of kin. This study aimed to describe the cardiac care experiences of post-myocardial infarction younger people (<55 years old) and their next of kin. Method This study employed an exploratory inductive qualitative design involving individual interviews with younger people (n=17) and their next of kin (n=13). Results Three categories were identified and patients were deemed to be in need of: tailored information, recognition or a post-discharge rehabilitation plan Conclusions Findings demonstrated the importance of using the experiences of younger people and their next of kin as the starting point when planning care and cardiac rehabilitation. Health professionals must systematically and purposefully provide person-centred care, both in hospital and post-discharge settings. Next of kin must be regarded as an important resource in post-myocardial infarction care for younger people, as they tend to be engaged in providing their relatives with support, which is valuable for the recovery of young people post myocardial infarction. Further research is warranted to investigate what makes person-centred interventions directed towards post-myocardial infarction younger people and their next of kin effective.

    • Thermally expandable microspheres are extensively used in industry as a lightweight filler for many products. The spheres can expand up to 60 times the initial size and are used for different purposes, including material reduction and surface modification. In fused filament fabrication (FFF), a material is deposited in a layer-by-layer process. Typically, FFF objects need not be solid because such objects are typically used for applications with low mechanical stress. Low material infill percentages are commonly used inside a solid outer shell to reduce material usage, weight, and manufacturing time. This paper proposes a new composite filament for FFF consisting of polylactic acid (PLA) and thermally expandable Expancel microspheres in the form of masterbatch granules. These filaments contain unexpanded microspheres that can be expanded during printing by heating. Two types of filaments containing 2 wt% and 5 wt% of masterbatch granules were manufactured and tested. The filaments were successfully used with a commercial 3D printer to manufacture objects with a density of 45% compared to objects manufactured using standard PLA. The tensile strength of these objects changed linearly with density and was comparable to that of PLA objects of the same density prepared using infill patterns. The composite filaments are advantageous in that they can reduce the amount of material used, as is currently done by using different amounts of infill in a pattern. Further, by varying the nozzle temperature, their density can be adjusted directly during printing as well as during fabrication to produce layers of different densities in the same object.

    • In this paper we present an audio synthesizer that creates audio signals, intended to be reproduced as sound by a loudspeaker. This paper details the design, implementation and verification of an analog, subtractive audio synthesizer. The synthesizer produces a variety of waveforms spanning approximately 3 octaves. The frequency is controlled by an external voltage and the harmonic content is regulated by a tunable filter. The system is fabricated in 130 nm CMOS. Current consumption measures between 1.8 and 2.1 mA from a supply of 1.2 V.

    • Polymer brushes are widely used to alter the properties of interfaces. In particular, poly(ethylene glycol) (PEG) and similar polymers can make surfaces inert toward biomolecular adsorption. Neutral hydrophilic brushes are normally considered to have static properties at a given temperature. As an example, PEG is not responsive to pH or ionic strength. Here we show that, by simply introducing a polymeric acid such as poly(methacrylic acid) (PMAA), the highly hydrated brush barrier can change its properties entirely. This is caused by multivalent hydrogen bonds in an extremely pH-sensitive process. Remarkably, it is sufficient to reduce the pH to 5 for complexation to occur at the interface, which is two units higher than in the corresponding bulk systems. Below this critical pH, PMAA starts to bind to PEG in large amounts (comparable to the PEG amount), causing the brush to gradually compact and dehydrate. The brush also undergoes major rheology changes, from viscoelastic to rigid. Furthermore, the protein repelling ability of PEG is lost after reaching a threshold in the amount of PMAA bound. The changes in brush properties are tunable and become more pronounced when more PMAA is bound. The initial brush state is fully recovered when releasing PMAA by returning to physiological pH. Our findings are relevant for many applications involving functional interfaces, such as capture-release of biomolecules.

  • Hand, Wrist, and Forearm Injuries in Male Professional Soccer Players: A Prospective Cohort Study of 558 Team-Seasons From 2001-2002 to 2018-2019

    • Background: The literature on upper extremity injuries in professional soccer players is scarce, and further insight into the onset and cause of these injuries as well as potential differences between goalkeepers and outfield players is important. Purpose: To investigate the epidemiology of hand, wrist, and forearm injuries in male professional soccer players between 2001 and 2019. Study Design: Descriptive epidemiology study. Methods: Between the 2001-2002 and 2018-2019 seasons, 120 European male soccer teams were followed prospectively for a varying number of seasons (558 team-seasons in total). Time-loss injuries and player-exposures to training sessions and matches were recorded on an individual basis in 6754 unique players. Injury incidence was reported as the number of injuries per 1000 player-hours, and between-group differences were analyzed using Z statistics and rate ratios (RRs) with 95% CIs. Between-group differences in layoff time were analyzed. Results: In total, 25,462 injuries were recorded, with 238 (0.9%) of these affecting the hand (71.4%; n = 170), wrist (16.8%; n = 40), and forearm (11.8%; n = 28), producing an incidence of 0.065 injuries per 1000 hours. A majority of the injuries were traumatic with an acute onset (98.7%; n = 235). Fractures were the most common injuries recorded (58.8%; n = 140), often involving the metacarpal bones (25.2%; n = 60) and phalanges (10.1%; n = 24). The injury incidence was significantly higher for goalkeepers (115 injuries; 0.265 per 1000 hours) compared with outfield players (123 injuries; 0.038 per 1000 hours) (RR, 7.0 [95% CI, 5.4-9.0]). Goalkeepers also had a significantly longer mean layoff time than outfield players (23 +/- 27 vs 15 +/- 27 days; P = .016). Conclusion: Injuries to the hand, wrist, and forearm constituted less than 1% of all time-loss injuries in male professional soccer players. Fractures were most common and constituted more than half of all injuries. Goalkeepers had a 7-fold higher incidence and an over 1-week longer mean layoff time compared with outfield players.

    • Abrasive water jet cutting is a technique which utilizes water mixed with abrasives, projected at high velocities to cut a wide range of materials. With a 5-axis solution for angled cutting chamfered cuts of material e.g. for parts that later are going to be welded and thus reducing one manufacturing step can be performed. This paper reports project work made in the MF2004 Machine Design advanced course at KTH. It describes the design and analysis of a first prototype (alpha prototype) of a 5-axis solution as well as the design of a second prototype (beta prototype). The selected concept is a tool center point (TCP) solution where the chamfer angle is realised by two rotations and the TCP position is independent of these rotations.

Och många av dem kommer från den vetenskapliga disciplinen Operationsanalys.Syftet med den här antologin är att introducera studerande i militärteknik på Försvarshögskolan till två av de mest refererade boktitlarna i operationsanalys – Methods for conducting military operational analysis editerad av Andrew G. Loerch och Larry B. Rainey samt Military Operations Research, quantitative decision making av N.K. Jaiswal.Kapitlen utgörs av essäer skrivna av studerande på den högre stabsofficersutbildningen med teknisk inriktning, som examinationsuppgift i en fördjupningskurs.     • Synthetic imidazole ligands are typically substituted at the N-1 ((1)-Im) position while natural imidazole ligands are substituted at the C-4 ((4)-Im) position. To outline the difference in coordination properties, the methyl-substituted imidazoles Me(4)-Im and Me(1)-Im were complexed with CuCl2 and ZnCl2 and investigated by NMR relaxometry, electron paramagnetic resonance, far-Fourier transform IR vibrational spectroscopy, and ab initio calculations. Me(4)-Im, Me(1)-Im, and Im in excess form the usual tetragonal D-4h [CuL4X2] complexes with CuCl2 whereas the methylated imidazoles form pseudotetrahedral C-2v, complexes instead of the usual octahedral O-h [ZnIm(6)](2+) complex. All imidazoles display a high degree of covalence in the M-L sigma- and pi-bonds and the pi-interaction strength affects the relative energies of complexation. Opportunities to tailor complexes by the chemical properties of the substituents are envisaged due to the role of the inductive and hyperconjugative effects, rather than position.
