    • An experimental investigation of the nucleus 165Lu, using the reactions 138Ba(31P,4n) 165Lu and 150Sm(19F,4n) 165Lu at beam energies of E = 155 and 95 MeV, respectively, has been performed. Among other additions to the existing level scheme, a new band, with transition energies almost identical to a strongly deformed (β2 0.42) πi13/2[660 1/2+] band recently discovered in 163Lu has been established. A theoretical analysis of the structure of the two Lu isotopes, 165Lu and 163Lu is carried out by detailed calculations of total potential energy surfaces for specific configurations. By a diabatic treatment of crossings specific proton configurations as πi13/2[660 1/2+] are identified throughout the deformation space and as a function of spin. It is found as a general feature that well deformed local minima of considerable nonaxial symmetry coexist with a normal deformed global minimum. The depth of these local minima depend on configuration. The structure of the different global and local minima found in these surfaces are analysed and discussed in terms of occupation of available basis configurations and their orientation relative to the rotation axis. The strongly deformed minima are found to belong to a group of superdeformed triaxial structures, expected to appear at low energies for certain favourable combinations of proton and neutron numbers.

  • Antigen delivery to early endosomes eliminates the superiority of human blood BDCA3+ dendritic cells at cross presentation

    • Human BDCA3+ dendritic cells (DCs), the proposed equivalent to mouse CD8α+ DCs, are widely thought to cross present antigens on MHC class I (MHCI) molecules more efficiently than other DC populations. If true, it is unclear whether this reflects specialization for cross presentation or a generally enhanced ability to present antigens on MHCI. We compared presentation by BDCA3+ DCs with BDCA1+ DCs using a quantitative approach whereby antigens were targeted to distinct intracellular compartments by receptor-mediated internalization. As expected, BDCA3+ DCs were superior at cross presentation of antigens delivered to late endosomes and lysosomes by uptake of anti-DEC205 antibody conjugated to antigen. This difference may reflect a greater efficiency of antigen escape from BDCA3+ DC lysosomes. In contrast, if antigens were delivered to early endosomes through CD40 or CD11c, BDCA1+ DCs were as efficient at cross presentation as BDCA3+ DCs. Because BDCA3+ DCs and BDCA1+ DCs were also equivalent at presenting peptides and endogenously synthesized antigens, BDCA3+ DCs are not likely to possess mechanisms for cross presentation that are specific to this subset. Thus, multiple DC populations may be comparably effective at presenting exogenous antigens to CD8+ T cells as long as the antigen is delivered to early endocytic compartments.

    • PURPOSE: To compare conventional MR imaging, echo-planar diffusion-weighted imaging (EP-DWI) and spin-echo diffusion-weighted imaging (SE)-DWI at radiological diagnosis of acute stroke. MATERIAL AND METHODS: Twenty-seven patients (30-85 years old) were examined. Clinical examination was performed before MR imaging. All MR examinations were assessed by an experienced neuroradiologist blinded to clinical findings. RESULTS: In EP-DWI, every patient had a lesion corresponding to the clinical findings. EP-DWI was used as the gold standard. In conventional PD+T2 imaging, 23/59 focal lesions were interpreted as acute, which was false in 11 lesions, and 36/59 lesions were considered to be old, 6 were in fact acute. Nine acute lesions were only detected retrospectively and 12 acute lesions were not detected at all on PD+T2. SE-DWI including the apparent diffusion coefficient correlated fairly well with EP-DWI but the procedure was impractical. CONCLUSION: EP-DWI is reliable for diagnosis of early ischemic stroke, while SE-DWI performs reasonably well. Conventional PD+T2 imaging is not reliable for diagnosis of early ischemia.
