    • Background: We aimed to evaluate differences in outcome between patients admitted to intensive care unit (ICU) after elective versus acute surgery in a multinational cohort of very old patients (80 years; VIP). Predictors of mortality, with special emphasis on frailty, were assessed.Methods: In total, 5063 VIPs were induded in this analysis, 922 were admitted after elective surgery or intervention, 4141 acutely, with 402 after acute surgery. Differences were calculated using Mann-Whitney-U test and Wilcoxon test. Univariate and multivariable logistic regression were used to assess associations with mortality.Results: Compared patients admitted after acute surgery, patients admitted after elective surgery suffered less often from frailty as defined as CFS (28% vs 46%; p < 0.001), evidenced lower SOFA scores (4 +/- 5 vs 7 +/- 7; p < 0.001). Presence of frailty (CFS >4) was associated with significantly increased mortality both in elective surgery patients (7% vs 12%; p = 0.01), in acute surgery (7% vs 12%; p = 0.02).Conclusions: VIPs admitted to ICU after elective surgery evidenced favorable outcome over patients after acute surgery even after correction for relevant confounders. Frailty might be used to guide clinicians in risk stratification in both patients admitted after elective and acute surgery.

    • A new source of pectin with a cytotoxic effect on glioblastoma cells is presented. A homogeneous GWP-FP-S fraction (M-w, of 29,170 g mol(-1)) was obtained by fractionating the crude pectin extract (GW) from Campomanesia xanthocarpa pulp. According to the monosaccharide composition, the GWP-FP-S was composed of galacturonic acid (58.8%), arabinose (28.5%), galactose (11.3%) and rhamnose (1.1%), comprising 57.7% of homogalacturonans (HG) and 42.0% of type I rhamnogalacturonans (RG-I). These structures were characterized by chromatographic and spectroscopic methods; GW and GWP-FP-S fractions were evaluated by MIT and crystal violet assays for their cytotoxic effects. Both fractions induced cytotoxicity (15.55-37.65%) with concomitant increase in the cellular ROS levels in human glioblastoma cells at 25-400 mu g mL(-)(1), after 48 h of treatment, whereas no cytotoxicity was observed for normal NIH 3T3 cells. This is the first report of in vitro bioactivity and the first investigation of the antitumor potential of gabiroba pectins.

    • Southwest of the well-known Skellefte District in northern Sweden a new ore province is presently being explored, the so called Gold Line. Today the largest known gold deposit in the Gold Line is the Fäboliden orogenic gold deposit.The gold mineralization is commonly hosted in quartz veins, which parallel the steep main foliation, within a shear zone in the metagreywacke host rocks. The fine-grained (2-40 μm) gold is closely associated with arsenopyrite in the quartz veins.Two main groups of fluid inclusions are present in the Fäboliden quartz veins. 1) Primary inclusions with a CO2-CH4 or a H2S (±CH4) composition (the latter recognized for the first time in a Swedish ore deposit). 2) Secondary fluid inclusions composed of pure CH4 and low-salinity aqueous fluids. The primary fluid inclusions are associated with arsenopyrite (+gold) and the CO2-CH4 fluid was also involved in precipitation of graphite. The graphite-forming reactions should generate a H2O phase as well. However, the presence of a H2O phase was not detected in any of the primary fluid inclusions and is suggested to have been consumed by wall rock reactions, generating hydrated alteration minerals such as Ca-amphibole, biotite, and minor tourmaline. Fluid inclusion data indicate arsenopyrite and graphite deposition at a pressure condition of ~4 kbars. Graphite is useful as an indicator of the metamorphic grade because the graphitization process is irreversible with no effects on the graphite structure during retrogression (Beyssac et al., 2002). Graphite in the mineralized quartz veins at Fäboliden indicates maximum temperatures of 520-560°C for the hydrothermal alteration system.Pyrrhotite was deposited after a subsequent pressure decrease and a later input of pure CH4 and low-salinity aqueous fluids, as suggested by the secondary fluid inclusions. These later fluids were trapped at a substantially lower pressure of ~0.3 kbars and a temperature of ~400°C.

    • The nucleus Os-171 has been populated using the reaction Sn-116(Ni-58,2pn). Four new bands are identified, and the previously known bands are extended in spin, to a maximum of 53/2 (h) over bar. One- and three-quasiparticle configurations are identified, and beta, gamma, and octupole configurations are assigned tentatively. The effects of a possible intruder configuration on the negative parity bands are tested using band-mixing calculations.

Gamma-rays were assigned to the nucleus by the recoil-decay tagging method, The ground-state band has a structure consistent with an h(11/2) proton coupled to a core of large triaxial deformation. At high spins, a bandcrossing occurs which is interpreted as a change in shape to a prolate deformation, Band-mixing calculations are performed for Ir171-175. These show that shape-coexistence between triaxial and prolate states in these nuclei follows the same systematics found in their Pt and Os neighbours. The systematics are also compared with deformations calculated for Ir171-179 using the code "Ultimate Cranker". Dipole bands were also observed, but tilted axis cranking calculations suggest that they are associated with a collective rotation.

    • SNAP-25 is essential to activity-dependent vesicle fusion and neurotransmitter release in the nervous system. During early development and adulthood, SNAP-25 appears to have differential influences on short- and long-term synaptic plasticity. The involvement of SNAP-25 in these processes may be different at hippocampal and neocortical synapses because of the presence of two different splice variants, which are developmentally regulated. We show here that the isoform SNAP-25a, which is expressed first developmentally in rodent brain, contributes to developmental regulation of the expression of both long-term depression (LTD) and long-term potentiation (LTP) at Schaffer collateral-CA1 synapses in the hippocampus. In one month old mice lacking the developmentally later expressed isoform SNAP-25b, Schaffer collateral-CA1 synapses showed faster release kinetics, decreased LTP and enhanced LTD. By four months of age, SNAP-25b-deficient mice appeared to have compensated for the lack of the adult SNAP-25b isoform, now exhibiting larger LTP and no differences in LTD compared to wild type mice. Interestingly, learning a hippocampus-dependent task reversed the reductions in LTP, but not LTD, seen at one month of age. In four month old adult mice, learning prevented the compensatory up-regulation of LTD that we observed prior to training. These findings support the hypothesis that SNAP-25b promotes stronger LTP and weakens LTD at Schaffer collateral-CA1 synapses in young mice, and suggest that compensatory mechanisms can reverse alterations in synaptic plasticity associated with a lack of SNAP-25b, once mice reach adulthood.

The structure of the (pi g(9/2))(-2) two-proton-hole spectrum below a T-1/2 = 0.48(16) mu s isomer is deduced and compared to shell-model predictions. A tentative I-pi = (8(+)) assignment, as suggested by systematics, yields a strongly reduced B(E2,8(+) --> 6(+)) = 0.44((+20)(-10)) W.u., corresponding to an effective proton charge of e(pi) = 0.85((+20)(-10))e, which is at variance with existing theoretical predictions.

    • SNAP-25 exists as two developmentally regulated alternatively spliced isoforms, SNAP-25a and SNAP-25b. We explored the function of SNAP-25a and SNAP-25b at Schaffer collateral-CA1 synapses in hippocampus using 4-week-old wild-type (WT) and SNAP-25b-deficient (MT) mice. Characterizing the protein expression of individual SNAP-25 isoforms revealed that WT females had higher levels of SNAP-25a than WT males, suggesting a sex-dependent delay of the alternative splicing switch from SNAP-25a to SNAP-25b. MT mice expressed normal levels of total SNAP-25, Syntaxin 1A and SNAP-47 in the hippocampus, but females expressed lower levels of VAMP2. Electrophysiological recordings in in vitro hippocampal slices revealed significantly reduced magnitude of LTP in MT mice. We also found reduction in paired-pulse facilitation after induction of LTP in WT males, but not in WT females, possibly related to the difference in SNAP-25a/SNAP-25b ratios, suggesting that the splicing switch may play a sex-specific role in LTP-associated increases in presynaptic release probability. Basal synaptic transmission measured in input-output relations revealed that the ability to discriminate between the intensity of presynaptic stimuli was affected in SNAP-25b-deficient mice. Learning in a behavioural paradigm of active-avoidance was impaired in MT mice, strengthening the conclusion that SNAP-25b is important for cognitive performance by altering activity-dependent synaptic plasticity.

    • High-spin states in Tl-198 were studied using the Au-197(alpha, 3n) reaction. The level scheme was considerably extended including two new bands and several non-yrast levels. One of the new bands is possibly a chiral partner to the yrast pi h(9/2) circle times vi(13/2)(-1) band. Two-quasiparticle-plus-triaxial-rotor model calculations suggest an aplanar orientation of the total angular momenta for these bands, thus supporting possible chirality.

    • A candidate for chiral bands was found in Tl-198 for the first time in a mass region of oblate (or nonaxial with gamma >= 30 degrees) deformed nuclei. Two bands show very similar quasiparticle alignments, moments of inertia, and B(M1)/B(E2) ratios. They have a relative excitation energy of about 500 keV and different patterns of energy staggering. Calculations using the two-quasiparticle-plus-triaxial-rotor model with residual proton-neutron interaction included show that a triaxial deformation with gamma similar to 44 degrees agrees very well with all the experimental observations. Furthennore, considerable energy staggering for both partner bands was calculated for this pi h(9/2) circle times vi(13/2)(-1) configuration at gamma - 30 degrees, suggesting that chiral bands may have substantial energy staggering.

    • Cyclin-dependent kinase 5 (Cdk5) is widely expressed although kinase activity has been described preferentially in neuronal systems. Cdk5 has an impact on actin polymerization during neuronal migration and neurite outgrowth and deregulation of the kinase has been implicated in the promotion of neurodegeneration. Recently it was shown that Cdk5 modulates dopamine signaling in neurons by regulating DARPP-32 function. In addition, Cdk5 phosphorylates munc-18 and synapsin I, two essential components of the exocytotic machinery. We have shown by reverse transcriptase-polymerase chain reaction, immunocytochemistry, and Western blotting that Cdk5 is present in the insulin-secreting pancreatic beta-cell. Subcellular fractionation of isolated beta-cells revealed a glucose-induced translocation of membrane-bound Cdk5 protein to lower density fractions. Inhibition of Cdk5 with roscovitine reduced insulin secretion with approximately 35% compared with control after glucose stimulation and with approximately 65% after depolarization with glucose and KCl. Capacitance measurements performed on single beta-cells that expressed a dominant-negative Cdk5 mutant showed impaired exocytosis. The effect on exocytosis by Cdk5 appeared to be independent of changes in free cytoplasmic Ca(2+) concentration. Taken together these results show that Cdk5 is present in beta-cells and acts as a positive regulator of insulin exocytosis.

    • Excited states in 102Sn have been identified for the first time, in an in-beam γ-ray spectroscopic experiment. Two γ-ray transitions with energies 1472 and 497 keV following the decay of the seniority 6+ isomer with t1/2 = 1.0(5) μs were unambiguously assigned to 102Sn. Due to the very low cross section of about 2 μb for producing 102Sn in the reaction 50Cr(58Ni,1α2n), a highly selective detector setup utilizing NORDBALL ancillary detectors and a recoil catcher device was used. High γ-ray detection efficiency was achieved with two EUROBALL Ge cluster detectors.

To verify bacterial translocation in this strain of rats, another six rats underwent controlled haemorrhage for 60 min to reduce the blood pressure to 55 mm Hg mean arterial pressure (MAP). These rats had free access to food and water before haemorrhage but were allowed only water until samples were taken 24 h after haemorrhage.Main Outcomes Measures: Presence and number of coliform bacteria in samples taken from caecal contents, caecal epithelium, MLN, and blood.Results: Starvation for 24 h increased the number of coliform bacteria (colony forming units (CFU)/g) in the caecal contents 25-fold (p < 0.05). Starvation for 48 h further increased the number by a factor of 100. The number of coliform bacteria that adhered to the caecal epithelium increased 3,000 times in rats that had been starved for 48 h (p < 0.001). There was no significant difference in translocation (as indicated by cultures from MLN) between rats that had been fed and those that had been starved for 48 h. In 4 of the 6 rats that were bled and then starved for 24 h there were signs of bacterial translocation, which was significantly more than the 1/19 in fed rats (p < 0.05).Conclusion: Starvation increases the number of bacteria in the caecal contents and increases bacterial adherence to the caecal epithelium. These changes may contribute to the previously reported increase in bacterial translocation in starved compared wit fed rats that were subjected to stress. The same changes in the gut were observed in animals subjected to haemorrhagic stress in addition to starvation, and in which bacterial translocation was evident.

  • Six-quasiparticle isomer in Nd-140

    • A search for isomeric states was performed in the nucleus Nd-140(60)80 using in-beam gamma-ray spectroscopy and the Te-126(O-18,4n) reaction. Prompt and delayed gamma-ray coincidences were measured with the AFRODITE spectrometer using the pulsed beam delivered by the Separated Sector Cyclotron of iThemba LABS. One new isomer was identified, with spin-parity I-pi=20(+) and lifetime T-1/2 >= 400 ns, at an excitation energy E-x=7430 keV. The lifetime of the 10(+) isomer at E-x=3619 keV was determined to be 32.9(1.8) ns, confirming the previously reported value. The configuration of the I-pi=20(+) isomer is assigned based on configuration-dependent cranked Nilsson-Strutinsky (CNS) calculations as a state pi(d(5/2)g(7/2))(10)(+-4)circle times nu(h(11/2)(-2))(10)(+), with the spin vectors of the six holes in the Gd-146(64)82 core fully aligned.
