    • The Horizon 2020 Project EuPRAXIA ("European Plasma Research Accelerator with eXcellence In Applications") is preparing a conceptual design report of a highly compact and cost-effective European facility with multi-GeV electron beams using plasma as the acceleration medium. The accelerator facility will be based on a laser and/or a beam driven plasma acceleration approach and will be used for photon science, high-energy physics (HEP) detector tests, and other applications such as compact X-ray sources for medical imaging or material processing. EuPRAXIA started in November 2015 and will deliver the design report in October 2019. EuPRAXIA aims to be included on the ESFRI roadmap in 2020.

  • Landscape of BCL2 Resistance Mutations in a Real-World Cohort of Patients with Relapsed/Refractory Chronic Lymphocytic Leukemia Treated with Venetoclax

    • The oral, highly selective Bcl2 inhibitor venetoclax has substantially improved the therapeutic landscape of chronic lymphocytic leukemia (CLL). Despite the remarkable response rates in patients with relapsed/refractory (R/R) disease, acquired resistance is the leading cause of treatment failure, with somatic BCL2 mutations being the predominant genetic drivers underpinning venetoclax resistance. To assess the correlation between disease progression and the most common BCL2 mutations G101V and D103Y, sensitive (10−4) screening for the most common BCL2 mutations G101V and D103Y was performed in 67 R/R CLL patients during venetoclax single-agent or venetoclax–rituximab combination therapy. With a median follow-up time of 23 months, BCL2 G101V and D103Y were detected in 10.4% (7/67) and 11.9% (8/67) of the cases, respectively, with four patients harboring both resistance mutations. Ten out of eleven patients carrying BCL2 G101V and/or D103Y experienced relapse during the follow-up period, representing 43.5% of the cases (10/23) showing clinical signs of disease progression. All BCL2 G101V or D103Y variants were detected in patients receiving venetoclax as a continuous single-agent treatment while these mutations were not observed during or after fixed-duration venetoclax therapy. Targeted ultra-deep sequencing of BCL2 uncovered three additional variants in four patient samples obtained at relapse, suggesting convergent evolution and implying a cooperating role of BCL2 mutations in driving venetoclax resistance. This cohort is the largest R/R CLL patient population reported to date in which BCL2 resistance mutations were investigated. Our study demonstrates the feasibility and clinical value of sensitive screening for BCL2 resistance mutations in R/R CLL.

    • Next generation ion therapy magnets both for gantry and for accelerator (synchrotron) are under investigation in a recently launched European collaboration that, in the frame of the European H2020 HITRIplus and I.FAST programmes, has obtained some funding for work packages on superconducting magnets. Design and technology of superconducting magnets will be developed for ion therapy synchrotron and -especially- gantry, taking as reference beams of 430 MeV/nucleon ions (C-ions) with 10(10) ions/pulse. The magnets are about 60-90 mm diameter, 4 to 5 T peak field with a field change of about 0.3 T/s and good field quality. The paper will illustrate the organization of the collaboration and the technical program. Various superconductor options (LTS, MgB2 or HTS) and different magnet shapes, like classical CosTheta or innovative Canted CosTheta (CCT), with curved multifunction (dipole and quadrupole), are under evaluation, CCT being the baseline. These studies should provide design inputs for a new superconducting gantry design for existing facilities and, on a longer time scale, for a brand-new hadron therapy centre to be placed in the South East Europe (SEEIIST project).

    • In this paper, we analyse points of connection between the feminist movement, the European Union, and the gendered division of labour in East-Central Europe with the methodology of world-systems analysis. The method and theoretical framework of world-systems analysis enables us to understand, first, that the European Union is the product of a certain historical-world economic phase. Second, that the southern and eastern expansion of the EU is embedded into a different world economic phase. Third, that the history of (feminist) movements of given nation states or regions cannot be understood merely through the social history of those nation states; rather, it has to be considered that (feminist) movements are also embedded in economic macro-processes. Fourth, that relations of dependency arising from unequal development are expressed in them. And fifth, that the relationship between informal and formal work is a hierarchical one:through theircombination, informal work acts as a subsidy to the cost of formal labour, thus contributing to the accumulation of value on higher levels of the chain and within the household relation; female reproductive labour generally acts as a subordinated subsidy to male labour; and the contradicting logics of accumulation through unequal household relations, and love and care as part of reproductive relations, makes the household an intrinsic front of battles and compromises of human life within capitalism.

  • Multi‑μJ harmonic emission energy from laser‑driven plasma

    • We report on simultaneous efficiency and divergence measurements for harmonics from solid targets generated by the relativistic oscillating mirror mechanism. For a value of the normalized vector potential of aL≃1.5aL≃1.5, we demonstrate the generation of 30 μJ high-harmonic radiation in a 17±317±3 mrad divergence cone. This corresponds to a conversion efficiency of ≳≳ 10−4 in the 17–80 nm range into a well-confined beam. Presuming phase-locked harmonics, our results predict unprecedented levels of average power for a single attosecond pulse in the generated pulse train. Results of PIC simulations raise the prospect of attaining efficiencies of a few percent at higher laser intensities.

  • Detection of RNA-DNA binding sites in long noncoding RNAs.

    • Long non-coding RNAs (lncRNAs) can act as scaffolds that promote the interaction of proteins, RNA, and DNA. There is increasing evidence of sequence-specific interactions of lncRNAs with DNA via triple-helix (triplex) formation. This process allows lncRNAs to recruit protein complexes to specific genomic regions and regulate gene expression. Here we propose a computational method called Triplex Domain Finder (TDF) to detect triplexes and characterize DNA-binding domains and DNA targets statistically. Case studies showed that this approach can detect the known domains of lncRNAs Fendrr, HOTAIR and MEG3. Moreover, we validated a novel DNA-binding domain in MEG3 by a genome-wide sequencing method. We used TDF to perform a systematic analysis of the triplex-forming potential of lncRNAs relevant to human cardiac differentiation. We demonstrated that the lncRNA with the highest triplex-forming potential, GATA6-AS, forms triple helices in the promoter of genes relevant to cardiac development. Moreover, down-regulation of GATA6-AS impairs GATA6 expression and cardiac development. These data indicate the unique ability of our computational tool to identify novel triplex-forming lncRNAs and their target genes.
