  • Role of Mannose-binding Lectin and Association with Microbial Sensitization in a Cohort of Patients with Atopic Dermatitis

    • Atopic dermatitis is a relapsing inflammatory skin condition, in which bacteria, fungi and viruses may colonize the skin and aggravate the condition. Mannose-binding lectin is part of the innate immune system. Polymorphism in the mannose-binding lectin gene can result in deficiency of mannose-binding lectin, which may affect defence against microbes. The aim of this study was to investigate whether polymorphisms in the mannose-binding lectin gene affect the extent of sensitization to common skin microbes, the skin barrier function, or the severity of the disease in a cohort of patients with atopic dermatitis. Genetic testing of mannose-binding lectin polymorphism was performed in 60 patients with atopic dermatitis. The disease severity, skin barrier function, and serum levels of specific immunoglobulin E against skin microbes were measured. In patients with low mannose-binding lectin genotype (group 1) 6 of 8 (75%) were sensitized to Candida albicans, compared to 14 of 22 (63.6%) patients with intermediate mannose-binding genotype (group 2) and 10 of 30 (33.3%) patients with high mannose-binding genotype (group 3). Group 1 (low mannose-binding lectin) was more likely to be sensitized to Candida albicans compared with group 3 (high mannose-binding lectin) (odds ratio 6.34, p-value 0.045). In this cohort of patients with atopic dermatitis, mannose-binding lectin deficiency was associated with increased sensitization to Candida albicans.

  • Carbon Fibers from Lignin-Cellulose Precursors : Effect of Stabilization Conditions

    • There is an increasing demand for lightweight composites reinforced with carbon fibers (CFs). Due to its high availability and carbon content, kraft lignin has gained attention as a potential low-cost CF precursor. CFs with promising properties can be made from flexible dry-jet wet spun precursor fibers (PFs) from blends (70:30) of softwood kraft lignin and fully bleached softwood kraft pulp. This study focused on reducing the stabilization time, which is critical in CF manufacturing. The impact of stabilization conditions on chemical structure, yield, and mechanical properties was investigated. It was possible to reduce the oxidative stabilization time of the PFs from about 16 h to less than 2 h, or even omitting the stabilization step, without fusion of fibers. The main reactions involved in the stabilization stage were dehydration and oxidation. The results suggest that the isothermal stabilization at 250 °C override the importance of having a slow heating rate. For CFs with a commercial diameter, stabilization of less than 2 h rendered in tensile modulus 76 GPa and tensile strength 1070 MPa. Impregnation with ammonium dihydrogen phosphate significantly increased the CF yield, from 31-38 to 46-50 wt %, but at the expense of the mechanical properties.

  • Continuous Stabilization and Carbonization of a Lignin–Cellulose Precursor to Carbon Fiber

    • : The demand for carbon fibers (CFs) based onrenewable raw materials as the reinforcing fiber in composites forlightweight applications is growing. Lignin−cellulose precursorfibers (PFs) are a promising alternative, but so far, there is limitedknowledge of how to continuously convert these PFs underindustrial-like conditions into CFs. Continuous conversion is vitalfor the industrial production of CFs. In this work, we havecompared the continuous conversion of lignin−cellulose PFs (50wt % softwood kraft lignin and 50 wt % dissolving-grade kraft pulp)with batchwise conversion. The PFs were successfully stabilizedand carbonized continuously over a total time of 1.0−1.5 h,comparable to the industrial production of CFs from polyacrylonitrile. CFs derived continuously at 1000 °C with a relative stretch of−10% (fiber contraction) had a conversion yield of 29 wt %, a diameter of 12−15 μm, a Young’s modulus of 46−51 GPa, and atensile strength of 710−920 MPa. In comparison, CFs obtained at 1000 °C via batchwise conversion (12−15 μm diameter) with arelative stretch of 0% and a conversion time of 7 h (due to the low heating and cooling rates) had a higher conversion yield of 34 wt%, a higher Young’s modulus (63−67 GPa) but a similar tensile strength (800−920 MPa). This suggests that the Young’s moduluscan be improved by the optimization of the fiber tension, residence time, and temperature profile during continuous conversion,while a higher tensile strength can be achieved by reducing the fiber diameter as it minimizes the risk of critical defects.

    • To meet the demand for carbon-fibre-reinforced composites in lightweight applications, cost-efficient processing and new raw materials are sought for. Cellulose and kraft lignin are each interesting renewables for this purpose due to their high availability. The molecular order of cellulose is an excellent property, as is the high carbon content of lignin. By co-processing cellulose and lignin, the advantages of these macromolecules are synergistic for producing carbon fibre (CF) of commercial grade in high yields. CFs were prepared from precursor fibres (PFs) made from 70:30 blends of softwood kraft lignin (SW-KL) and cellulose by dry-jet wet spinning with the ionic liquid (IL) 1-ethyl-3-methylimidazolium acetate ([EMIm][OAc]) as a solvent. In focus was the impact of the molecular mass of lignin and the type of cellulose source on the CF yield and properties, while membrane-filtrated kraft lignin and cellulose from dissolving kraft pulp and fully bleached paper-grade SW-KP (kraft pulp) served as sources. Under the investigated conditions, the yield increased from around 22% for CF from neat cellulose to about 40% in the presence of lignin, irrespective of the type of SW-KL. The yield increment was also higher relative to the theoretical one for CF made from blends (69%) compared to those made from neat celluloses (48-51%). No difference in the mechanical properties of the produced CF was observed.

    • This study explores patients' knowledge of cardiac risk factors (CRFs), analyses how information and advice about CRFs are documented in clinical practice, and assesses patient adherence to received instructions to decrease CRFs. Systemic lupus erythematosus (SLE) patients with >= 4 ACR criteria participated through completing a validated cardiovascular health questionnaire (CHQ). Kappa statistics were used to compare medical records with the self-reported CHQ (agreement) and to evaluate adherence. Two hundred and eleven (72%) of the known patients with SLE participated. The mean age of the patients was 55 years. More than 70% of the SLE patients considered hypertension, obesity, smoking and hypercholesterolaemia to be very important CRFs. The agreement between medical record documentation and patients' reports was moderate for hypertension, overweight and hypercholesterolaemia (kappa 0.42-0.60) but substantial for diabetes (kappa 0.66). Patients' self-reported adherence to advice they had received regarding medication was substantial to perfect (kappa 0.65-1.0). For lifestyle changes in patients with hypertension and overweight, adherence was only fair to moderate (kappa 0.13-0.47). Swedish SLE patients' awareness of traditional CRFs was good in this study. However, the agreement between patients' self-reports and medical record documentation of CRF profiles, and patients' adherence to medical advice to CRF profiles, could be improved. In this paper we systematically reviewed the extant literature and estimate the overall magnitude of such programmes and how they can inform future interventions. Specifically, we aimed to: (a) conduct a systematic review on the literature of interpersonal CDPs and PDPs within the youth sport context; (b) examine the effects of such interventions on youth athlete outcomes via a meta-analysis. English written peer-reviewed publications and grey literature was identified through electronic search in databases and manual searches of reference lists. By utilising a priori criteria for inclusion and exclusion, 33 studies describing interpersonal CDPs, and PDPs were identified in the systematic review. Studies that presented required data for estimation of Hedge's g effect sizes were included in the meta-analysis (k = 27). By and large, the included studies used a quasi-experimental design (58%), sampled from team sports (79%), and reported several delivery methods (e.g., workshops, audio feedback, observations, peer group discussions) and outcome measures (e.g., anxiety, autonomous motivation, self-confidence). Some interventions were based on the same delivery protocols (e.g., Coach Effectiveness Training, Mastery Approach to Coaching) or theoretical frameworks (e.g., Achievement Goal Theory, Self-Determination Theory). The meta-analysis showed statistically significant small, and medium, effect sizes on a subsample of youth athlete outcomes (e.g., task-related climate, fun and enjoyment, anxiety), indicating that coach interpersonal skills can contribute to positive youth sport experiences. Theory-based interpersonal CDPs and PDPs are recommended to expand the knowledge in this field of research.

  • Preventing fiber-fiber adhesion of lignin-cellulose precursors and carbon fibers with spin finish application

    • Adhesion of fibers within a spun tow, including carbon fibers and precursors, is undesirable as it may interrupt the manufacturing process and entail inferior fiber properties. In this work, softwood kraft lignin was used together with a dissolving pulp to spin carbon fiber precursors. Lignin-cellulose precursors have previously been found to be prone to fiber fusion, both post-spinning and during carbon fiber conversion. In this study, the efficiency of applying different kinds of spin finishes, with respect to rendering separable precursors and carbon fibers, has been investigated. It was found that applying a cationic surfactant, and to a similar extent a nonionic surfactant, resulted in well separated lignin-cellulose precursor tows. Furthermore, the fiber separability after carbon fiber conversion was evaluated, and notably, precursors treated with a silicone-based spin finish generated the most well-separated carbon fibers. The underlying mechanism of fiber fusion post-spinning and converted carbon fibers is discussed.

  • Novel risk genes for systemic lupus erythematosus predicted by random forest classification

    • Genome-wide association studies have identified risk loci for SLE, but a large proportion of the genetic contribution to SLE still remains unexplained. To detect novel risk genes, and to predict an individual's SLE risk we designed a random forest classifier using SNP genotype data generated on the "Immunochip" from 1,160 patients with SLE and 2,711 controls. Using gene importance scores defined by the random forest classifier, we identified 15 potential novel risk genes for SLE. Of them 12 are associated with other autoimmune diseases than SLE, whereas three genes (ZNF804A, CDK1, and MANF) have not previously been associated with autoimmunity. Random forest classification also allowed prediction of patients at risk for lupus nephritis with an area under the curve of 0.94. By allele-specific gene expression analysis we detected cis-regulatory SNPs that affect the expression levels of six of the top 40 genes designed by the random forest analysis, indicating a regulatory role for the identified risk variants. The 40 top genes from the prediction were overrepresented for differential expression in B and T cells according to RNA-sequencing of samples from five healthy donors, with more frequent over-expression in B cells compared to T cells.

Furthermore, a direct and an indirect evaluation have been conducted compared to stationary rain gauges and C-band radar. In the indirect analysis, the input data sets were integrated into runoff models for Ellinge and Lundåkra WWTP where the results were compared to measured incoming flow to the sewage plants.

    • Objectives: The main objectives of this study were to calculate total costs of illness and cost -driving disease features among patients with systemic lupus erythematosus (SLE) in Sweden. Methods: Five cohorts of well-defined SLE patients, located in different parts of the country were merged. Incident and prevalent cases from 2003 through 2010 were included. The American College of Rheumatology (ACR) classification criteria was used. From the local cohorts, data on demographics, disease activity (SLEDAI 2K), and organ damage (SDI) were collected. Costs for inpatient care, specialist outpatient care and drugs were retrieved from national registries at the National Board of Health and Welfare. Indirect costs were calculated based on sickness leave and disability pensions from the Swedish Social Insurance Agency. Results: In total, 1029 SLE patients, 88% females, were included, and approximately 75% were below 65 years at the end of follow-up, and thus in working age. The mean number of annual specialist physician visits varied from six to seven; mean annual inpatient days were 3.1-3.6, and mean annual sick leave was 123-148 days, all per patient. The total annual cost was 208,555 SEK ($33,369 = 22,941(sic)), of which direct cost was 63,672kr ($10,188 = 7004(sic)) and the indirect cost was 144,883 SEK ($23,181 = 15,937(sic)), all per patient. The costs for patients with short disease duration were higher. Higher disease activity as measured by a SLEDAI 2K score > 3 was associated with approximately 50% increase in both indirect and direct costs. Damage in the neuropsychiatric and musculoskeletal domains were also linked to higher direct and indirect costs, while organ damage in the renal and ocular systems increased direct costs. Conclusion: Based on this study and an estimate of slightly more than 6000 SLE patients in Sweden, the total annual cost for SLE in the country is estimated at $188 million (= 129.5 million (sic)). Both direct (30%) and indirect costs (70%) are substantial. Medication accounts for less than 10% of the total cost. The tax paid national systems for health care and social security in Sweden ensure equal access to health care, sick leave reimbursements, and disability pensions nationwide. Our extrapolated annual costs for SLE in Sweden are therefore the best supported estimations thus far, and they clearly underline the importance of improved management, especially to reduce the indirect costs.   • Simulator performance, psychometrics and personality testing guiding the choice of clinical discipline

    • The complexity of surgical training has increased and surgery put high demands on personal abilities that cannot be met by all even after training. Selection of personnel is critical to many professions, including surgery, but the selection procedure of surgical apprenticeship is not well developed. It would be of value to get an early assessment of important personal features like the ability to learn complex procedures. Further, individuals learn in different ways and the personality may influence this ability. Other important aspects are visuospatial abilities, working memory and executive functioning. These variables are measured in the present study by: learning curves in a TURP VR-simulator, scores from the personality test TCI-R, Rey complex figure and recognition trial, Tower of London (dx) and tests from WAISIII. Twenty-four residents in urology performed three real TUR-P procedures and their performances were analyzed with OSATS and video-recordings. The learning curves from the OR were compared with the results from the simulation practice, personality tests and psychometrics using multiple linear regression. The findings from personality and psychometric data were also compared with the general population to see if there are any indications of a “surgical personality”. The urology residents in this sample have a welldeveloped character (effective, mature, reliable, goal-oriented, empathetic, tolerant, supportive, cooperative) and with high reward dependence (tender-hearted, dedicated, sociable) together with better executive planning abilities and better verbal working memory than normal. The connections between the operation learning curves and the variables above indicate that a better learning score is associated with a good learning score in a simulated environment, goal-directedness, a high level of impulse control, anticipation of harmful events and responsibility, a balanced attachment style and a good visual spatial memory.

  • Coronary Heart Disease in Systemic Lupus Erythematosus Is Associated With Interferon Regulatory Factor-8 Gene Variants

    • Background- Patients with systemic lupus erythematosus have increased morbidity and mortality in coronary heart disease (CHD). We asked whether there was a genetic influence on CHD in systemic lupus erythematosus. Methods and Results- The association between single-nucleotide polymorphisms (SNPs) and CHD in 2 populations of patients with systemic lupus erythematosus was assessed. Patients were genotyped on a custom 12k Illumina Array. The allele frequencies were compared between patients with (n=66) and without (n=509) CHD. We found 61 SNPs with an association (P<0.01) to CHD, with the strongest association for 3 SNPs located in the interferon regulatory factor-8 (IRF8) gene. Comparison of the allele frequencies of these 61 SNPs in patients with (n=27) and without (n=212) CHD in the second study population revealed that 2 SNPs, rs925994 and rs10514610 in IRF8 (linkage disequilibrium, r(2)=0.84), were associated with CHD in both study populations. Meta-analysis of the SNP rs925994 gave an odds ratio of 3.6 (2.1-6.3), P value 1.9x10(-6). The identified IRF8 allele remained as a risk factor for CHD after adjustment for traditional CHD risk factors. The IRF8 risk allele was associated with the presence of carotid plaques (P<0.001) and increased intima-media thickness (P=0.01). By electrophoretic mobility shift assays, we show weaker binding of protein to the risk allele of the highly linked SNP rs11117415, and by flow cytometry, a reduced frequency of circulating B cells was detected in patients with the IRF8 risk allele. Conclusions- There is a considerable genetic component for CHD in systemic lupus erythematosus, with IRF8 as a strong susceptibility locus.

    • OBJECTIVES: Patients with systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA) have increased risk of cardiovascular disease (CVD). We investigated whether single nucleotide polymorphisms (SNPs) at autoimmunity risk loci were associated with CVD in SLE and RA.METHODS: Patients with SLE (n=1045) were genotyped using the 200K Immunochip SNP array (Illumina). The allele frequency was compared between patients with and without different manifestations of CVD. Results were replicated in a second SLE cohort (n=1043) and in an RA cohort (n=824). We analysed publicly available genetic data from general population, performed electrophoretic mobility shift assays and measured cytokine levels and occurrence of antiphospholipid antibodies (aPLs).RESULTS: We identified two new putative risk loci associated with increased risk for CVD in two SLE populations, which remained after adjustment for traditional CVD risk factors. An IL19 risk allele, rs17581834(T) was associated with stroke/myocardial infarction (MI) in SLE (OR 2.3 (1.5 to 3.4), P=8.5×10-5) and RA (OR 2.8 (1.4 to 5.6), P=3.8×10-3), meta-analysis (OR 2.5 (2.0 to 2.9), P=3.5×10-7), but not in population controls. The IL19 risk allele affected protein binding, and SLE patients with the risk allele had increased levels of plasma-IL10 (P=0.004) and aPL (P=0.01). An SRP54-AS1 risk allele, rs799454(G) was associated with stroke/transient ischaemic attack in SLE (OR 1.7 (1.3 to 2.2), P=2.5×10-5) but not in RA. The SRP54-AS1 risk allele is an expression quantitative trait locus for four genes.CONCLUSIONS: The IL19 risk allele was associated with stroke/MI in SLE and RA, but not in the general population, indicating that shared immune pathways may be involved in the CVD pathogenesis in inflammatory rheumatic diseases.

    • Background Systemic lupus erythematosus (SLE) is a complex disease characterized by autoimmunity toward apoptotic cells, excessive amounts of circulating immune complexes, and complement activation. A decreased platelet size has been observed in SLE and their nonhemostatic functions may play an active role in the disease. The main objective of this study was to find clues that could explain their decreased size and functional role, analyzing the entire platelet proteome. Methods Platelets were isolated from 23 patients with SLE. The five individuals with the highest and lowest average platelet forward scatter were selected for further analysis. Platelet protein content was analyzed using liquid chromatography with tandem mass spectrometry (LC-MS/MS) and compared with platelets from five healthy controls. Data are available via ProteomeXchange with identifier PXD031202. Results Out of 2,572 proteins identified, 396 had significantly different levels (ANOVA q-value ≤ 0.01). Forty proteins, including immunoglobulin-, complement- and phosphatidylserinebinding proteins had higher abundance in platelets from SLE patients, largely independent of size (fold difference of ≥1.5 and a t-test p-value of ≤0.05 as cut-off). Functional characterization revealed increased degranulation and skewed hemostatic balance in platelets from SLE patients. In the SLE proteome, immunoglobulin proteins were negatively correlated to serum complement C3 and C4 and the highest relative levels were detected in platelets of normal size. Conclusion Platelets from SLE patients shared a specific protein profile, including immunoglobulins, complement proteins, and autoantigens, largely independent of the platelet size and in agreement with an integrated role for platelets in SLE.

    • OBJECTIVE: We previously reported high Systemic Lupus International Collaborating Clinics (SLICC) scores in fatal cases of systemic lupus erythematosus (SLE) from our inception cohort. This study was done to clarify if the SLICC damage scores 5 years after diagnosis predicted the outcome. METHODS: We studied 80 patients with SLE (70 women, 10 men), all enrolled and diagnosed during the years 1981 through 1991 in our inception cohort, and all alive 5 years after inclusion into the cohort. In all patients the SLICC/American College of Rheumatology (ACR) damage index (DI) was scored at 5 years after SLE diagnosis, and these scores were tested for predictive value. The outcomes were survival or late mortality within the following median observation period of 7 years. All surviving patients were followed through 1999, and no patient was lost to followup. RESULTS: At study entry, 5 years after the diagnosis of SLE, 37 patients had no damage to score with SLICC. Of the remaining 43 patients, 25 had a score of 1 and 18 had a score of 2 or more. In total, 14 fatalities occurred within 7 years after study entry, 7 among the 18 with initial SLICC/ACR DI of 2 or more compared with 7 fatalities among the 62 with less or no damage (p < 0.01). Cardiovascular or cerebrovascular SLICC/ACR DI items were more common in fatal cases than in survivors (p < 0.001). A SLICC score at 5 years of 2 or more increased the relative risk for fatality by 3.4 (95% CI 1.5-14.4), and had a predictive value of 38%. A SLICC score of 0 at 5 years gave an odds ratio in favor of survival of 0.06 (95% CI 0.0-0.5) and had a predictive value for survival of 97%. During an extended followup for one more year the predictive value of damage for fatalities was even more pronounced (p = 0.003, log-rank). CONCLUSION: SLICC damage scores registered 5 years after SLE diagnosis have a high predictive value for survival during the following median observation time of 7 years. These data provide strong evidence that the items included in the SLICC score are clinically relevant.

    • OBJECTIVES: To investigate associations between a high genetic disease risk and disease severity in patients with systemic lupus erythematosus (SLE).METHODS: Patients with SLE (n=1001, discovery cohort and n=5524, replication cohort) and healthy controls (n=2802 and n=9859) were genotyped using a 200K Immunochip single nucleotide polymorphism array. A genetic risk score (GRS) was assigned to each individual based on 57 SLE risk loci.RESULTS: SLE was more prevalent in the high, compared with the low, GRS-quartile (OR 12.32 (9.53 to 15.71), p=7.9×10-86 and OR 7.48 (6.73 to 8.32), p=2.2×10-304 for the discovery and the replication cohorts, respectively). In the discovery cohort, patients in the high GRS-quartile had a 6-year earlier mean disease onset (HR 1.47 (1.22 to 1.75), p=4.3×10-5), displayed higher prevalence of damage accrual (OR 1.47 (1.06 to 2.04), p=2.0×10-2), renal disorder (OR 2.22 (1.50 to 3.27), p=5.9×10-5), anti-dsDNA (OR 1.83 (1.19 to 2.81), p=6.1×10-3), end-stage renal disease (ESRD) (OR 5.58 (1.50 to 20.79), p=1.0×10-2), proliferative nephritis (OR 2.42 (1.30 to 4.49), p=5.1×10-3), anti-cardiolipin-IgG (OR 1.89 (1.13 to 3.18), p=1.6×10-2), anti-β2-glycoprotein-I-IgG (OR 2.29 (1.29 to 4.06), p=4.8×10-3) and positive lupus anticoagulant test (OR 2.12 (1.16 to 3.89), p=1.5×10-2) compared with patients in the low GRS-quartile. Survival analysis showed earlier onset of the first organ damage (HR 1.51 (1.04 to 2.25), p=3.7×10-2), first cardiovascular event (HR 1.65 (1.03 to 2.64), p=2.6×10-2), nephritis (HR 2.53 (1.72 to 3.71), p=9.6×10-7), ESRD (HR 6.78 (1.78 to 26.86), p=6.5×10-3) and decreased overall survival (HR 1.83 (1.02 to 3.30), p=4.3×10-2) in high to low quartile comparison.CONCLUSIONS: A high GRS is associated with increased risk of organ damage, renal dysfunction and all-cause mortality. Our results indicate that genetic profiling may be useful for predicting outcomes in patients with SLE.

    • Background/Purpose: Systemic lupus erythematosus (SLE) is a chronic, autoimmune disease with a complex genetic etiology. Over 80 risk genes for SLE have been identified and some genetic variants have demonstrated association with specific disease manifestations, such as STAT4 and nephritis. The overall effect of a patient’s hereditary risk factors on disease severity has so far not been studied. We therefore assessed the relationship between high genetic risk and development of organ damage in SLE.Methods: Patients with SLE, who met at least 4 ACR criteria (n = 1012), were genotyped using a 200K Immunochip SNP Array (Illumina). A genetic risk score (GRS) was assigned to each patient based on the single nucleotide polymorphisms (SNPs) which in previous studies have shown association (p<5×10-8) with SLE according to Morris, et al (Nat Genet, 2016. 48(8): p. 940-6). For 32 loci the SLE GWAS SNP was available on the ImmunoChip. For each SNP, the natural logarithm of the odds ratio (OR) for SLE susceptibility was multiplied by the number of risk alleles in each individual. The sum of all products for each patient was defined as the GRS. Information regarding organ damage according to Systemic Lupus International Collaborating Clinics / American College of Rheumatology Damage Index (SLICC-DI), disease manifestations, antibody profile, medication, current disease activity, age at diagnosis and sex was retrieved from medical records. Statistical analyzes were performed using Statistica 13.2 (Statsoft).Results: In an ordinal regression model, with SLICC-DI (0, 1, 2, 3, 4 and >4 points) as outcome and age and GRS as independent variables, an association was found between GRS and SLICC-DI (OR1.16 (1.03-1.31), p=0.015). The relationship was more pronounced for patients under 60 years of age (OR1.30 (1.11-1.52) p=7.1×10-4). Using a linear regression model, a negative relationship was observed between GRS and age at diagnosis (β = -0.13, p=1.5×10-5).When analyzing the 11 SLE criteria (ACR-82) using a logistic regression model associations were observed between GRS and nephritis (OR 1.26 (1.09-1.45), p=0.0015), the immunological criteria (OR 1.31 (1.13-1.51), p = 3.2×10-4) and arthritis (OR 0.84 (0.71-1.00), p=0.044). A high GRS was also associated with presence of anti-dsDNA (OR 1.37 (1.15-1.62), p=9.4×10-7) and low complement levels (OR 1.32 (1.03-1.68), p=0.044). No association was observed between GRS and disease activity at the time of follow-up and there was no difference in GRS between men and women with SLE.Conclusion: In patients with SLE, there is an association between a high genetic risk score and early disease onset. In addition, patients with high genetic risk scores have a higher risk of developing permanent organ damage compared to individuals with fewer risk genes. Our findings indicate that genetic profiling of patients with SLE may provide a tool for predicting severity of the disease.

    • Objectives: Systemic lupus erythematosus (SLE) is an autoimmune disease with extensive heterogeneity in disease presentation between patients, which is likely due to an underlying molecular diversity. Here, we aimed at elucidating the genetic aetiology of SLE from the immunity pathway level to the single variant level, and stratify patients with SLE into distinguishable molecular subgroups, which could inform treatment choices in SLE. Methods: We undertook a pathway-centred approach, using sequencing of immunological pathway genes. Altogether 1832 candidate genes were analysed in 958 Swedish patients with SLE and 1026 healthy individuals. Aggregate and single variant association testing was performed, and we generated pathway polygenic risk scores (PRS). Results: We identified two main independent pathways involved in SLE susceptibility: T lymphocyte differentiation and innate immunity, characterised by HLA and interferon, respectively. Pathway PRS defined pathways in individual patients, who on average were positive for seven pathways. We found that SLE organ damage was more pronounced in patients positive for the T or B cell receptor signalling pathways. Further, pathway PRS-based clustering allowed stratification of patients into four groups with different risk score profiles. Studying sets of genes with priors for involvement in SLE, we observed an aggregate common variant contribution to SLE at genes previously reported for monogenic SLE as well as at interferonopathy genes. Conclusions: Our results show that pathway risk scores have the potential to stratify patients with SLE beyond clinical manifestations into molecular subsets, which may have implications for clinical follow-up and therapy selection.

The presented architecture holds promise for quantum simulation with tunable-range interactions and photon transport experiments in the nonlinear regime.

    • The ability to control and measure the temperature of propagating microwave modes down to very low temperatures is indispensable for quantum information processing and may open opportunities for studies of heat transport at the nanoscale, also in the quantum regime. Here, we propose and experimentally demonstrate primary thermometry of propagating microwaves using a transmon-type superconducting circuit. Our device operates continuously, with a sensitivity down to 4×10-4 photons/Hz and a bandwidth of 40 MHz. We measure the thermal occupation of the modes of a highly attenuated coaxial cable in a range of 0.001 to 0.4 thermal photons, corresponding to a temperature range from 35 mK to 210 mK at a frequency around 5 GHz. To increase the radiation temperature in a controlled fashion, we either inject calibrated, wideband digital noise, or heat the device and its environment. This thermometry scheme can find applications in benchmarking and characterization of cryogenic microwave setups, temperature measurements in hybrid quantum systems, and quantum thermodynamics.

    • Platelets have recently emerged as important immune modulators in systemic lupus erythematosus (SLE), in addition to their role in thrombosis and cardiovascular disease. However, studies investigating mean platelet volume (MPV) in SLE are often scarce, conflicting and cross-sectional. In this study, MPV was measured in clinical routine throughout a defined time-period to quantify both individual MPV fluctuations and investigate if such variations are associated with disease activity and clinical phenotypes of SLE. Of our 212 patients, 34 patients had only one MPV value reported with the remaining 178 patients having between 2 and 19 visits with recorded MPV values. The intra-individual MPV variation was low, with a median variation of 0.7 fL. This was further supported by the finding that 84% of patients stayed within their reference interval category (i.e., small, normal or large) over time. In our cohort, no correlation between disease activity and MPV neither cross-sectionally nor longitudinally was found. Mean platelet volume values were significantly smaller in SLE patients (mean 10.5 fL) compared to controls (mean 10.8 fL), p < 0.0001. Based on the reference interval, 2.4% (n = 5) of patients had large-sized platelets, 84.4% (n = 179) had normal-sized and 13.2% (n = 28) had small-sized. A larger proportion (85.7%) of patients with small-sized platelets met the anti-dsDNA criterion (ACR10b; p = 0.003) compared to patients with normal and large (57.6%) sized platelets. In conclusion, the intra-individual MPV variation was of low magnitude and fluctuations in disease activity did not have any significant impact on MPV longitudinally. This lack of variability in MPV over time indicates that measuring MPV at any time-point is sufficient. Further studies are warranted to evaluate MPV as a possible biomarker in SLE, as well as to determine the underlying mechanisms influencing platelet size in SLE.

    • BACKGROUND: Neuropsychiatric (NP) involvement and fatigue are major problems in systemic lupus erythematosus (SLE). S100A8/A9 is a marker of inflammation and responds to therapy in SLE patients. S100A8/A9 has an immunopathogenic role in various neurological diseases. We investigated S100A8/A9 in relation to NP-involvement and fatigue in SLE.METHODS: 72 consecutive SLE outpatients at a tertiary centre and 26 healthy controls were included in this cross-sectional study. NPSLE was determined by specialists in rheumatology and neurology and defined according to three attribution models: "ACR", "SLICC A" and "SLICC B". Cerebral MRI was assessed by a neuroradiologist and neurocognitive testing by a neuropsychologist. The individuals were assessed by scores of pain (VAS), fatigue (VAS and FSS), and depression (MADRS-S). Concentrations of S100A8/A9 in serum and cerebrospinal fluid were measured with ELISA. Statistical calculations were performed using non-parametric methods.RESULTS: Serum concentrations of S100A8/A9 were higher in SLE patients compared with controls (medians 1230 ng/ml; 790 ng/ml, p = 0.023). The concentrations were higher in NPSLE patients compared with non-NPSLE patients when applying the SLICC A and ACR models, but not significant when applying the SLICC B model (medians 1400 ng/ml; 920 ng/ml, p = 0.011; 1560 ng/ml; 1090 ng/ml, p = 0.050; 1460 ng/ml; 1090 ng/ml, p = 0.083, respectively). No differences of CSF S100A8/A9 concentrations were observed between NPSLE and non-NPSLE patients. SLE patients with depression or cognitive dysfunction as an ACR NPSLE manifestation had higher serum S100A8/A9 concentrations than non-NPSLE patients (median 1460 ng/ml, p = 0.007 and 1380 ng/ml, p = 0.013, respectively). Higher serum S100A8/A9 correlated with higher VAS fatigue (r = 0.31; p = 0.008) and VAS pain (r = 0.27, p = 0.021) in SLE patients. Serum S100A8/A9 was not independently associated with NPSLE when adjusting for scores of fatigue (FSS) and pain (VAS) (OR 1.86, 95% CI 0.93-3.73, p = 0.08).CONCLUSIONS: Serum S100A8/A9 concentrations may be associated with NPSLE and fatigue. S100A8/A9 may be of interest in evaluating NPSLE, although further investigations are needed.

    • Systemic lupus erythematosus (SLE, OMIM 152700) is a systemic autoimmune disease with a complex etiology. The mode of inheritance of the genetic risk beyond familial SLE cases is currently unknown. Additionally, the contribution of heterozygous variants in genes known to cause monogenic SLE is not fully understood. Whole-genome sequencing of DNA samples from 71 Swedish patients with SLE and their healthy biological parents was performed to investigate the general genetic risk of SLE using known SLE GWAS risk loci identified using the ImmunoChip, variants in genes associated to monogenic SLE, and the mode of inheritance of SLE risk alleles in these families. A random forest model for predicting genetic risk for SLE showed that the SLE risk variants were mainly inherited from one of the parents. In the 71 patients, we detected a significant enrichment of ultra-rare (0.1%) missense and nonsense mutations in 22 genes known to cause monogenic forms of SLE. We identified one previously reported homozygous nonsense mutation in the C1QC (Complement C1q C Chain) gene, which explains the immunodeficiency and severe SLE phenotype of that patient. We also identified seven ultra-rare, coding heterozygous variants in five genes (C1S, DNASE1L3, DNASE1, IFIH1, and RNASEH2A) involved in monogenic SLE. Our findings indicate a complex contribution to the overall genetic risk of SLE by rare variants in genes associated with monogenic forms of SLE. The rare variants were inherited from the other parent than the one who passed on the more common risk variants leading to an increased genetic burden for SLE in the child. Higher frequency SLE risk variants are mostly passed from one of the parents to the offspring affected with SLE. In contrast, the other parent, in seven cases, contributed heterozygous rare variants in genes associated with monogenic forms of SLE, suggesting a larger impact of rare variants in SLE than hitherto reported.

    • Little is known about the metabolic or cardiovascular effects of selective ER modulators (SERMs), such as raloxifene hydrochloride (RLX), in postmenopausal women with type 2 diabetes mellitus (DM). Therefore, the effect of RLX vs. placebo (PL) on glycemic control, insulin sensitivity, as well as effects on a number of hormone, lipid, coagulation, and safety factors were determined in 30 postmenopausal women with type 2 DM in a randomized, double blind, cross-over trial. All participants had a SHBG serum concentration below 60 nmol/liter at baseline and had stable diabetes controlled by either oral hypoglycemic agents or diet for 1 month. In the first treatment period, participants received 12 wk of either PL or RLX, followed by an 8-wk washout before the second treatment period. In the second treatment period, participants were crossed over to the other treatment. Compared with PL, RLX did not significantly affect fasting blood glucose, hemoglobin A(1c), lipids, fasting insulin, or insulin sensitivity (as measured by the euglycemic clamp technique). Compared with PL, RLX reduced fibrinogen levels by 0.77 g/liter (P < 0.001), IGF-I by 2.4 nmol/liter (P < 0.001), and free T by 0.73 pmol/liter (P = 0.038) and increased SHBG by 5.5 nmol/liter (P = 0.001) and IGF-binding protein-3 by 0.57 ng/ml (P = 0.007). Our results demonstrate that RLX does not significantly affect glycemic control and has favorable or neutral effects on selected surrogate markers of cardiovascular risk in postmenopausal women with type 2 diabetes mellitus while decreasing hyperandrogenicity in these patients.

    • Background: To evaluate the effect of one lesson of continuing medical education (CME) of subjective assessment of optic nerve head appearance on sensitivity and specificity for the diagnosis of glaucoma. Methods: Ophthalmologists and residents in ophthalmology attending an international glaucoma meeting arranged at Malmo University Hospital, Malmo, Sweden, were asked to grade optic nerve head (ONH) photographs of healthy and glaucomatous subjects at two sessions separated by a lecture on glaucoma diagnosis by ONH assessment. Each grader had access to an individual portfolio of 50 ONH photographs randomly selected from a web-based data bank including ONH photographs of 73 glaucoma patients and 123 healthy subjects. The individual portfolio of photographs was graded before and after the lecture, but in different randomized order. Results: Ninety-six doctors, 91% of all attending the meeting, completed both assessment sessions. The number of correct classifications increased from 69 to 72% on the average. Diagnostic sensitivity increased significantly (p < 0.0001) from 70% to 80%, and the number of photographs classified as uncertain decreased significantly (p < 0.0001) from 22% to 13%. Specificity remained at 68%, and intra-grader agreement decreased. Conclusion: CME had only a small effect on the assessment of ONH for the glaucoma diagnosis. Sensitivity increased and the amount of uncertain classifications decreased, while specificity was unchanged.

    • The influence of inorganic elements on lignin ‐based carbon fibre (CF) quality was studied using sulphates of Na +, K+, Mg2+, Fe2+, Al3+. The metal sulphates were added to wet spun prefibres made from softwood kraft lignin (SKL):cellulose (70:30) and melt spun prefibres made from low molecular mass SKL. An increase in concentration from 0.1 w% to about 0.4 w% did neither affect the mechanical properties nor the morphology as observed by SEM. In contrast, metal sulphates added to the initial 0.45 w% to a total range 1.5 to 5.0 w%, was found detrimental to the melt spinning and to the final CF quality. Thus, the recommendation of <0.1 w% ash in kraft lignin may be exceeded, but more research is needed to establish the upper concentration limit.

    • ObjectiveSLE, primary Sjogrens syndrome (pSS) and systemic sclerosis (SSc) are heterogeneous autoimmune diseases with a dysregulated type I interferon (IFN) system. The diseases often show overlapping clinical manifestations, which may result in diagnostic challenges. We asked to which extent SSc-associated autoantibodies are present in SLE and pSS, and whether these link to serum IFN-alpha, clinical phenotypes and sex. Samples with clinical data from patients with SSc and healthy blood donors (HBDs) served as controls. Finally, the diagnostic performance of SSc-associated autoantibodies was evaluated.MethodsSamples from well-characterised subjects with SLE (n=510), pSS (n=116), SSc (n=57) and HBDs (n=236) were analysed using a commercially available immunoassay (EuroLine Systemic Sclerosis Profile (IgG)). IFN-alpha was quantified by ELISA. Self-reported data on Raynauds phenomenon (RP) were available.ResultsWith exceptions for anti-Ro52/SSA and anti-Th/To, SSc-associated autoantibodies were more frequent in SSc than in SLE, pSS and HBDs regardless of sex. IFN-alpha levels correlated with the number of positive SSc-associated autoantibodies (r=0.29, p<0.0001) and associated with Ro52/SSA positivity (p<0.0001). By using data from SLE, SSc and HBDs, RP was significantly associated with topoisomerase I, centromere protein (CENP)-B, RNA polymerase III 11 kDa, RNA polymerase III 155 kDa and PM-Scl100 whereas Ro52/SSA associated inversely with RP. In SLE, CENP-A was associated with immunological disorder, CENP-B with serositis and Ku with lupus nephritis. By combining analysis of ANA (immunofluorescence) with SSc-associated autoantibodies, the diagnostic sensitivity reached 98% and the specificity 33%.ConclusionsThe 13 specificities included in the EuroLine immunoassay are commonly detected in SSc, but they are also frequent among individuals with other diseases imprinted by type I IFNs. These findings are valuable when interpreting serological data on patients with suspected SSc, especially as patients may present with disease manifestations overlapping different rheumatological diseases. In SLE, we observed associations between manifestations and SSc-associated autoantibodies which have not previously been reported.

    • Carbon fibers (CFs) have excellent mechanical properties and a low density, making themattractive as a reinforcing fiber in composites. The use of CFs is limited to high-end applications,since they are produced from an expensive fossil-based precursor via an energy-intensivemanufacturing process, explaining the need for cheaper CFs from renewables. CFs can be madefrom strong cellulosic precursors, but the low carbon content of cellulose results in a lowconversion yield, and thus an expensive CF. Lignin has a higher carbon content than cellulose butCFs from melt spun lignin precursors have presented challenges, since these precursors have a lowstrength and are difficult to convert to CF in a realistic conversion time.In the present work, CFs from solution spun precursors consisting of blends of softwood kraftlignin and cellulose have been developed. The lignin-cellulose precursors (up to 70% lignin) wereprepared with air-gap spinning and wet spinning, using an ionic liquid and a water-based solventsystem for co-dissolution, respectively. Co-processing of cellulose and lignin was beneficial as theformer made the precursor strong and easy to handle, whereas the latter gave a higher conversionyield than precursors based solely on cellulose. The precursors were converted to CFs via bothbatchwise and continuous conversion, using industrially relevant times (< 2 h), with a yield up to45 wt% after incorporation of a flame retardant.These CFs have a moderate Young’s modulus and tensile strength up to 75–77 GPa and 1.2 GPa,respectively, i.e. similar to the values for CFs from fossil-based isotropic pitch and they can thusbe classified as general-grade CFs. These biobased CFs have a disordered turbostratic graphitestructure, and their tensile properties are affected by the precursor structure, the conversionconditions, and the final diameter. These CFs can potentially be used as a sustainable componentin non-structural and semi-structural applications.

    • Carbon fibers (CFs) are gaining increasing importance in lightweight composites, but their high price and reliance on fossil-based raw materials stress the need for renewable and cost-efficient alternatives. Kraft lignin and cellulose are renewable macromolecules available in high quantities, making them interesting candidates for CF production. Dry-jet wet spun precursor fibers (PFs) from a 70/30 w/w blend of softwood kraft lignin (SKL) and fully bleached softwood kraft pulp (KP) were converted into CFs under fixation. The focus was to investigate the effect of carbonization temperature and time on the CF structure and properties. Reducing the carbonization time from 708 to 24 min had no significant impact on the tensile properties. Increasing the carbonization temperature from 600 to 800 °C resulted in a large increase in the carbon content and tensile properties, suggesting that this is a critical region during carbonization of SKL:KP PFs. The highest Young's modulus (77 GPa) was obtained after carbonization at 1600 °C, explained by the gradual transition from amorphous to nanocrystalline graphite observed by Raman spectroscopy. On the other hand, the highest tensile strength (1050 MPa) was achieved at 1000 °C, a decrease being observed thereafter, which may be explained by an increase in radial heterogeneity.

  • Carbon Fibers from Wet-Spun Cellulose-Lignin Precursors Using the Cold Alkali Process

    • In recent years, there has been extensive research into the development of cheaper and more sustainable carbon fiber (CF) precursors, and air-gap-spun cellulose-lignin precursors have gained considerable attention where ionic liquids have been used for the co-dissolution of cellulose and lignin. However, ionic liquids are expensive and difficult to recycle. In the present work, an aqueous solvent system, cold alkali, was used to prepare cellulose-lignin CF precursors by wet spinning solutions containing co-dissolved dissolving-grade kraft pulp and softwood kraft lignin. Precursors containing up to 30 wt% lignin were successfully spun using two different coagulation bath compositions, where one of them introduced a flame retardant into the precursor to increase the CF conversion yield. The precursors were converted to CFs via batchwise and continuous conversion. The precursor and conversion conditions had a significant effect on the conversion yield (12-44 wt%), the Young's modulus (33-77 GPa), and the tensile strength (0.48-1.17 GPa), while the precursor morphology was preserved. Structural characterization of the precursors and CFs showed that a more oriented and crystalline precursor gave a more ordered CF structure with higher tensile properties. The continuous conversion trials highlighted the importance of tension control to increase the mechanical properties of the CFs.

    • It is in the nature of the human species to find solutions of complex technical problems and always strive for improvements. The development of new materials is not an exception. One of the many man-made materials is carbon fibre (CF). Its excellent mechanical properties and low density have made it attractive as the reinforcing agent in lightweight composites. However, the high price of CF originating from expensive production is currently limiting CF from wider utilisation, e.g. in the automotive sector. The dominating raw material used in CF production is petroleum-based polyacrylonitrile (PAN). The usage of fossil-based precursors and the high price of CF explain the strong driving force of finding cheaper and renewable alternatives. Lignin and cellulose are renewable macromolecules available in high quantities. The high carbon content of lignin is an excellent property, while its structural heterogeneity yields in CF with poor mechanical properties. In contrast, cellulose has a beneficial molecular orientation, while its low carbon content gives a low processing yield and thus elevates processing costs. This work shows that several challenges associated with CF processing of each macromolecule can be mastered by co-processing. Dry-jet wet spun precursor fibres (PFs) made of blends of softwood kraft lignin and kraft pulps were converted into CF. The corresponding CFs demonstrated significant improvement in processing yield with negligible loss in mechanical properties relative to cellulose-derived CFs. Unfractionated softwood kraft lignin and paper grade kraft pulp performed as good as more expensive retentate lignins and dissolving grade kraft pulp, which is beneficial from an economic point of view. The stabilisation stage is considered the most time-consuming step in CF manufacturing. Here it was shown that the PFs could be oxidatively stabilised in less than 2 h or instantly carbonised without any fibre fusion, suggesting a time-efficient processing route. It was demonstrated that PF impregnation with ammonium dihydrogen phosphate significantly improves the yield but at the expense of mechanical properties. A reduction in fibre diameter was beneficial for the mechanical properties of the CFs made from unfractionated softwood kraft lignin and paper grade kraft pulp. Short oxidative stabilisation (<2 h) of thin PFs ultimately provided CFs with tensile modulus and strength of 76 GPa and 1070 MPa, respectively. Considering the high yield (39 wt%), short stabilisation time and promising mechanical properties, the concept of preparing CF from lignin:cellulose blends is a very promising route.

    • Ultrasound optical tomography (UOT) is an imaging technique based on the acousto-optic effect that can perform optical imaging with ultrasound resolution inside turbid media, and is thus interesting for biomedical applications, e.g. for assessing tissue blood oxygenation. In this paper, we present near background free measurements of UOT signal strengths using slow light filter signal detection. We carefully analyze each part of our experimental setup and match measured signal strengths with calculations based on diffusion theory. This agreement between experiment and theory allows us to assert the deep tissue imaging potential of similar to 5 cm for UOT of real human tissues predicted by previous theoretical studies [Biomed. Opt. Express 8, 4523 (2017)] with greater confidence, and indicate that future theoretical analysis of optimized UOT systems can be expected to be reliable.     • Stabilisation of the prefibre is a time-consuming step in carbon fibre (CF) production. In this paper the stabilisation condition of dry-jet wet-spun lignin-cellulose (LC) prefibres (70:30  t%) are reported. The impact of prefibre-impregnation by ammonium dihydrogen phosphate (ADHP) and various thermal conditions were evaluated by measuring the yield and mechanical properties of the final CFs. The addition of ADHP improved the CF yields from 32-40 wt% to 45-47 wt% but had a slight negative impact on the tensile modulus (TM) whereas no significant difference in tensile strength (TS) was observed. The absence of fibre fusion and glass transition temperature (Tg) indicate successful stabilisation of all prefibres. This implies possibilities of using short stabilisation times of LC prefibres in CF production.

  • Degenerate and nondegenerate Josephson parametric oscillators for quantum information applications

    • Parametric oscillations are well studied phenomena, with applications in amplification, quantum optics, and quantum information processing. They can occur as a parameter of a system, such as the resonant frequency, is being modulated or “pumped” by an external field. A nonlinearity of the system can then transfer power from the pumping frequency to two frequencies called signal and idler. When the signal falls within a resonance, and when the pump amplitude exceeds an instability threshold, parametric oscillations occur. When signal and idler fall within the same resonance, the pumping is called degenerate.Due to somewhat different implementations, in combination with rich nonlinear physics, the details of the parametric oscillations can differ in different systems. In this work, we expand on the research done on parametric oscillations using a superconducting microwave resonator with a tunable boundary condition. Previously, a degenerate Josephson parametric oscillator (JPO) has been demonstrated by modulation of this boundary condition at twice the resonator's resonant frequency. We further use the JPO to implement a novel read-out method for a superconducting qubit. Moreover, we demonstrate a different, nondegenerate regime of the JPO, which we denote the NJPO, by modulating the frequency at the sum of two resonant frequencies of a multimode resonator.Both the JPO qubit read-out and the NJPO have applications within quantum information processing. The perhaps most promising platform for quantum information processing is represented by the circuit-QED architecture with superconducting resonators and artificial atoms, where the two lowest atomic levels represent the qubit. We make these circuits using lithographic techniques, and control and measure them at low temperature using microwaves. The qubit's state is fragile and difficult to read out with high fidelity. To this end, we have developed a method based on the JPO, in which the qubit state is mapped onto a zero-amplitude or a large-amplitude state of the JPO. We achieved a large contrast (185±15 photons), therefore eliminating the need for a following quantum-limited parametric amplifier. Our readout fidelity was 81.5 %, and by carefully modeling we can distinguish fidelity loss from the qubit and the readout, respectively. This analysis displays an actual readout fidelity of 98.7 %.An alternative model for quantum computing is based on continuous variables, and uses harmonic oscillators instead of qubits. Entangled states, which can be used as quantum resources, can then be created, for example, by two-mode squeezing of the oscillator ground state. The state of the NJPO produces correlated oscillations in its two modes, which might be useful for continuous-variable quantum computing. In this work we fully characterize the classical properties of the NJPO, and show good quantitative agreement with a theoretical model.

    • Present-day, noisy, small or intermediate-scale quantum processors-although far from fault tolerant-support the execution of heuristic quantum algorithms, which might enable a quantum advantage, for example, when applied to combinatorial optimization problems. On small-scale quantum processors, validations of such algorithms serve as important technology demonstrators. We implement the quantum approximate optimization algorithm on our hardware platform, consisting of two superconducting transmon qubits and one parametrically modulated coupler. We solve small instances of the NP (nondeterministic polynomial time)-complete exact-cover problem, with 96.6% success probability, by iterating the algorithm up to level two.

    • We investigate nondegenerate parametric oscillations in a superconducting microwave multimode resonator that is terminated by a superconducting quantum interference device (SQUID). The parametric effect is achieved by modulating magnetic flux through the SQUID at a frequency close to the sum of two resonator-mode frequencies. For modulation amplitudes exceeding an instability threshold, self-sustained oscillations are observed in both modes. The amplitudes of these oscillations s how good quantitative agreement with a theoretical model. The oscillation phases are found to be correlated and exhibit strong fluctuations which broaden the oscillation spectral linewidths. These linewidths are significantly reduced by applying a weak on-resonant tone, which also suppresses the phase fluctuations. When the weak tone is detuned, we observe synchronization of the oscillation frequency with the frequency of the input. For the detuned input, we also observe an emergence of three idlers in the output. This observation is in agreement with theory indicating four-mode amplification and squeezing of a coherent input.

  • Quantum information processing with tunable and low-loss superconducting circuits

    • The perhaps most promising platform for quantum information processing is the circuit-QED architecture based on superconducting circuits representing quantum bits. These circuits must be made with low losses so that the quantum information is retained for as long as possible. We developed fabrication processes achieving state-of-the-art coherence times of over 100 µs. We identified the primary source of loss to be parasitic two-level systems by studying fluctuations of qubit relaxation times. Using our high-coherence circuits, we implemented a quantum processor built on fixed-frequency qubits and frequency-tunable couplers. The tunable couplers were lumped-element LC resonators, where the inductance came from a superconducting quantum interference device (SQUID). We achieved a controlled-phase gate with a fidelity of 99% by parametric modulation of the coupler frequency. Using this device, and another similar to it, we demonstrated two different quantum algorithms, the quantum approximate optimization algorithm, and density matrix exponentiation. We achieved high algorithmic fidelities, aided by our carefully calibrated gates. Additionally, we researched parametric oscillations using frequency-tunable resonators. Previously, degenerate parametric oscillations have been demonstrated by modulation of the resonant frequency at twice that frequency. We use this phenomenon to implement a readout method for a superconducting qubit with a fidelity of 98.7%. We demonstrated correlated radiation in nondegenerate parametric oscillations by modulating at the sum of two resonant frequencies of a multimode resonator. We showed an excellent quantitative agreement between the classical properties of the oscillations with a theoretical model. Moreover, we studied higher-order modulation at up to five times their resonant frequencies. These types of parametric oscillation states might be used as a quantum resource for continuous-variable quantum computing.

  • The effects of extramural language : Relationships between engagement in Japanese language activities and general Japanese language proficiency

    • Many L2 learners believe they learn their target language by using it extramurally, i.e. outside the classroom. This topic, language learning as a result of engagement in L2 use, has garnered increased attention in the last decade. However, there is still a distinct lack of research. To date, most research offers snapshot studies of single groups of learners at similar L2 proficiency levels, and English is by far the most commonly researched target language.The present study quantitatively investigates the relationship between extramural Japanese and general Japanese language proficiency among adult learners studying at university in Sweden. It aims to do so both cross sectionally, using five different Levels of proficiency to investigate the moderating impact of proficiency at a macro level, and longitudinally, in order to move beyond correlations and provide empirical data on causal direction. In total, data were gathered from 168 individuals, many of whom participated in the study at different Levels, during a period of two and a half years. Data were analysed using non-parametric correlations and Bayesian regression.Results show that the participants engaged in extramural Japanese for considerable amounts of time every week. Some forms of Japanese language use (e.g. reading text and speaking face-to-face) related to general Japanese language proficiency, whereas others did not (e.g. listening to music and watching video with TL subtitles). These relationships were largely positive, although some relationships were negative or mixed. Proficiency level was a moderating factor, and different forms of language use had varying relationships with general Japanese language proficiency depending on participants’ Levels of study. In addition, the results suggest that extramural L2 use influenced proficiency but proficiency did not influence extramural L2 use. This is among the first studies to show such an effect empirically.Overall, the study makes significant contributions to our understanding of the connection between extramural L2 use and proficiency, which has implications on the applicability of results to other languages. Furthermore, the study also provides an innovative research framework for investigating extramural L2 use, which should be robust and valid for any target language.

    • Study Objectives: The aim was to evaluate nasal polyposis as a risk factor for nonadherence to continuous positive airway pressure (CPAP) treatment in patients with obstructive sleep apnea (OSA). Methods: This was a population-based, longitudinal analysis of patients starting CPAP treatment for OSA in the Swedish quality registry Swedevox between 2010 and 2018. Data were cross-linked with national registries. The impact of nasal polyposis on CPAP adherence was analyzed using uni-and multivariable logistic and linear regression models. Relevant confounders (age, sex, usage of nasal and oral steroids) were identified using a direct acyclic graph. Results: Of 20,521 patients with OSA on CPAP treatment (29.5% females), 331 (1.6%) had a diagnosis of nasal polyposis at baseline. At the 1-year follow-up, nasal polyposis was associated with an increased risk of CPAP usage < 4 hours/night (unadjusted odds ratio [OR] 1.21; 95% confidence interval [CI] 0.95-1.55); adjusted OR 1.38; 95% CI 1.08-1.77). In this group, unadjusted nocturnal mean CPAP usage was 15.4 minutes (95% CI-31.62 to 0.83) shorter and was an adjusted 24.1 minutes (95% CI-40.6 to-7.7) shorter compared with patients with OSA without nasal polyposis. Conclusions: Nasal polyposis is associated with reduced CPAP usage per night. These results highlight the importance of diagnosing nasal polyposis in patients with OSA before the start of CPAP treatment. Treatment of the condition may improve adherence, efficacy, and patient outcomes.

    • Macrophages are a functionally heterogeneous group of immune cells abundant in atherosclerotic plaques. Macrophages expressing CD163 are associated with intraplaque hemorrhage and have previously been considered atheroprotective. However, in a recent study CD163-deficient atherosclerotic ApoE−/− mice exhibited smaller and less complex plaques, suggesting a proatherogenic role of CD163. Previous smaller studies on CD163+ macrophages and plaque stability in humans have yielded diverging results. Here we assessed the association of CD163+ cells to plaque vulnerability in a large cohort of human carotid plaques. CD163 protein expression was analyzed by immunohistochemistry in 200 human carotid plaques removed by endarterectomy from 103 patients with and 93 patients without cerebrovascular symptoms. Furthermore, CD163 mRNA expression was analyzed in 66 of the plaques. Both protein and mRNA expression of CD163 was higher in plaques from symptomatic patients and in plaques with high vulnerability index. CD163+ macrophages were primarily found in shoulder regions and in the center of the plaques. The present data show that CD163 is associated with increased plaque vulnerability in human carotid plaques, supporting the notion that CD163+ macrophages could contribute to clinical events.
