Multi-ancestry GWAS of the electrocardiographic PR interval identifies 202 loci underlying cardiac conduction

- The electrocardiographic PR interval reflects atrioventricular conduction, and is associated with conduction abnormalities, pacemaker implantation, atrial fibrillation (AF), and cardiovascular mortality. Here we report a multi-ancestry (N=293,051) genome-wide association meta-analysis for the PR interval, discovering 202 loci of which 141 have not previously been reported. Variants at identified loci increase the percentage of heritability explained, from 33.5% to 62.6%. We observe enrichment for cardiac muscle developmental/contractile and cytoskeletal genes, highlighting key regulation processes for atrioventricular conduction. Additionally, 8 loci not previously reported harbor genes underlying inherited arrhythmic syndromes and/or cardiomyopathies suggesting a role for these genes in cardiovascular pathology in the general population. We show that polygenic predisposition to PR interval duration is an endophenotype for cardiovascular disease, including distal conduction disease, AF, and atrioventricular pre-excitation. These findings advance our understanding of the polygenic basis of cardiac conduction, and the genetic relationship between PR interval duration and cardiovascular disease. On the electrocardiogram, the PR interval reflects conduction from the atria to ventricles and also serves as risk indicator of cardiovascular morbidity and mortality. Here, the authors perform genome-wide meta-analyses for PR interval in multiple ancestries and identify 141 previously unreported genetic loci.

Genetic insights into resting heart rate and its role in cardiovascular disease

- The genetics and clinical consequences of resting heart rate (RHR) remain incompletely understood. Here, the authors discover new genetic variants associated with RHR and find that higher genetically predicted RHR decreases risk of atrial fibrillation and ischemic stroke. Resting heart rate is associated with cardiovascular diseases and mortality in observational and Mendelian randomization studies. The aims of this study are to extend the number of resting heart rate associated genetic variants and to obtain further insights in resting heart rate biology and its clinical consequences. A genome-wide meta-analysis of 100 studies in up to 835,465 individuals reveals 493 independent genetic variants in 352 loci, including 68 genetic variants outside previously identified resting heart rate associated loci. We prioritize 670 genes and in silico annotations point to their enrichment in cardiomyocytes and provide insights in their ECG signature. Two-sample Mendelian randomization analyses indicate that higher genetically predicted resting heart rate increases risk of dilated cardiomyopathy, but decreases risk of developing atrial fibrillation, ischemic stroke, and cardio-embolic stroke. We do not find evidence for a linear or non-linear genetic association between resting heart rate and all-cause mortality in contrast to our previous Mendelian randomization study. Systematic alteration of key differences between the current and previous Mendelian randomization study indicates that the most likely cause of the discrepancy between these studies arises from false positive findings in previous one-sample MR analyses caused by weak-instrument bias at lower P-value thresholds. The results extend our understanding of resting heart rate biology and give additional insights in its role in cardiovascular disease development.

- My good friend, the artist Peter Christensson, one day showed me some of his new artwork. The pictures were abstract but got the form of early expeditions to the Arctic. Peter told me, everything started with a dream about a polar swimmer and the world this person met on his journey to the North Pole. With my music, I want to enhance mood, pulse, nuances in the art and poetry of Peter Christensson. The irregular rhythm reinforces the feeling of unease, and the music ultimately aims upwards in the harmonics – a symbol for survival. At horizon or below, a poem begins.

A lot of great composers have written for the instruments and through them, I got the inspiration to compose for flute and percussion. After composing Dark Shadows, Kinkaku-ji and Second Concerto for Flute & Percussion, all published by Edition Svitzer, I’ve got a question and wish from many flute players to also write something for piccolo and percussion. They are right, there are not so many pieces composed for that combination of instruments. Working close to the young virtuous performers, Laura Michelin & Filip Korošec, I had the opportunity to write them Bird of Prey. In the pulsing rhythms, we hear the bird hunting its prey and the harmonies enhance the wildlife, to always be on its guard. In the middle of the piece, the listener is getting a high-flying journey to the lamenting song of a Bird of Prey.

- The Swedish composer Lars-Erik Larsson wrote in the late 40’s 12 Concertini – twelve smaller solo concerts for various instruments accompanied by string orchestra, all composed to be performed by amateur orchestras while the solo parts made higher demands on the instrumentalists’ skill. With inspiration from Lars-Erik Larsson, Daniel Berg has written a piece in two movements for a virtuoso solo marimba in combination with a soft string timbre – in classical style, but with clear elements of jazz harmonics. Daniel has, along with experienced string teachers, written parts that can be played by amateur orchestras along with a skilled marimba soloist. In his Concertino for Marimba and Strings, Daniel wants to highlight the marimba as a melodic percussion instrument, and at the same time inspire young musicians to discover classical percussion.

- Some years ago, I wanted to discover the flute and wrote SECOND CONCERTO for flute and percussion in five movements. Since then, the piece has been quite popular worldwide and when I was asked to write a new piece for the very talent Louise Pollock, principal trombonist with the Gothenburg Opera in Sweden, I took my chance. The structure of the concerto has the same as the concerto for flute. Five movements in different styles with different techniques for the solo instrument. 1st movement has a challenging solo part with an ostinato in percussion (this time I found inspiration from American composer Gene Koshinski). 2nd mvt is built on the favorite chord I often use in my music (for example in KINKAKU-JI for alto flute and vibraphone). 3rd mvt has the humor in focus – it’s a game of cat and mouse in rhythms. In the 4th mvt I wanted to write beautiful and sad for trombone with percussion effects. For me the sound of a trombone also reminds me about great brass musicians like Nils Landgren and the soul group Earth Wind and Fire and in the 5th mvt you will get some of this funky groove.

- For many years, the successful Duo Nimm (flautist Åsa Karlberg and percussionist Olle Pettersson) have performed my work Kinkaku-ji, a meditation for Flauto Alto and Vibraphone. One day, Åsa told me about her new Robert Dick Glissando Headjoint and at the same time asked if I could possibly write a new piece for flute and percussion with her new mouthpiece in mind. In contrast to the calm Kinkaku-ji, I began to compose a work with a lot of speed. At this time, I was in a state of darkness where people in my presence showed a shadow of hatred and evil I didn't think existed. The power of the music is incredibly strong and through the composition I was able to somehow react and express my feelings. The harmonies are therefore dark, the rhythms give a feeling of worry and the flute's glissando amplifies the inner pain - an attempt to musically describe the Dark Shadows.

I gave it some thought and answered, “everybody can join!”. And yes, if you have fun pieces to play, you can arrange them for all levels so everybody can join. In this booklet, Per Björkqvist has written drum parts for three of my pieces from Easy Duets (previously published by Edition Svitzer). These parts have their roots in “warm up exercises” often used in the world of drum corps. Per and I hope that these pieces can get everyone to play together and further strengthen their interest in playing percussion ensemble music. Daniel Berg As a percussion teacher, you need to inspire your students to play different instruments, musical styles and concepts. We need to work on lots of layers to get the students to groove and learn new things, often inspired by their friends in a social community. There are three important roles for having a successful climate to grooving: 1. you need to play together with others; 2. you need to understand what it’s about and; 3. you have to learn new things every time. Often, there is too little time to rehearse everything that you want to work on in a rehearsal; warm ups, time keeping, period playing, rudimental drumming, new tunes and so on. In these three tunes, we tried to combine a lot of these ideas. Our first goal is to let the students play a lot and have fun, (it’s much more fun to play then to count rests). You can add one or more of the percussion parts to the great pieces that Daniel has written, depending on the setting of your ensemble and level of your students. Good luck and have fun! - As a young percussionist I played timpani with S:t. Matteus Symfoniorkester (SMSO) in Stockholm. During a production we played among others Gloria by Francis Poulenc, for choir and orchestra. The music, and above all the harmony, had a big impact on me. I was immediately inspired to study more music from Poulenc together with other French composers like Oliver Messiaen. Fantasia and Toccata is strongly inspired by the harmonies of both Poulenc and Messiaen which I have tried to adapt through the marimba.

- My passion for chamber music started when I played my first pieces for percussion duo. To create something of its own together and find a personal means of expression has since led me to new ways and new combinations of instruments. I hope these four duets may inspire you to play more chamber music focusing on percussion. Presently, I give about 100 concerts a year with any of my chamber music formations.
