  • Pesticide use in rice farming and its impacts on climbing perch (Anabas testudineus) in the Mekong Delta of Vietnam

    • The intensification of agricultural production in the Mekong Delta has faced serious challenges with respect to increased use of agrochemicals and especially pesticides. The indiscriminate use of pesticide could potentially impact on the long-term food production, environmental and human health in the delta. The aim of this thesis was to investigate the negative side effects of the current use of pesticides on climbing perch (Anabas testudineus) in rice fields using brain acetylcholinesterase (hereafter referred to as AChE) activity as a biomarker. The empirical work, on which this thesis is based, includes structured questionnaires, laboratory and field experiments. First, a field survey using questionnaires was carried out to gain a better understanding of the current state of rice farming systems, the use of pesticides and attitude to pest management strategies among rice and rice-fish farmers, as well as to provide basic information for the set-up of the laboratory and field experiments. Secondly, laboratory studies were conducted to clarify if the selected insecticides applied alone and in mixtures caused negative side effects on climbing perch fingerlings. Thirdly, further toxicity studies were carried out, under rice field conditions, to further investigate the toxicity effects of the insecticides, applied alone, in mixtures and under sequential applications, on climbing perch fingerlings.The results showed that although there were a more selective use of pesticides and an increased awareness among farmers of the negative side effects of pesticides in 2007 as compared to 1999, the current use of pesticide in the Mekong Delta still cause many problems to the environment and human health. Chlorpyrifos ethyl (hereafter referred to as CPF) was found to cause a significant and more prolonged inhibition on the brain AChE activity in climbing perch than fenobucarb (hereafter referred to as F). The inhibition by the mixture of CPF and F were significantly higher than the inhibition by only F, but less prolonged and significant lower than the inhibition by only CPF. The results suggest that the combined effect from a mixture of F and CPF can create both additive effects initially and later antagonistic effects.CPF and F applied at concentrations used by farmers, either as separate doses, in a mixture or in sequential doses, decreased the brain AChE activity, growth and survival rates in climbing perch. The results demonstrate that brain AChE activity in climbing perch is a relevant biomarker for monitoring of exposure to, and sub-lethal impacts from organophosphates and carbamates under tropical conditions. The result also shows that 2-PAM re-activate the brain AChE activity, and can be used as an alternative method to assess the AChE inhibition level in organisms recently exposed to OP’s, in situation where it may be difficult to find unexposed individuals as controls.In conclusion, this thesis shows that the current use of pesticides in the Mekong Delta has a negative effect on climbing perch living in rice fields. It indicates that a sustained long-term food production in the Mekong Delta must be based on ecological principles, taking advantages of ecosystem biodiversity and productivity, and not through intensified use of pesticides.

  • Commercialising social media : a study of fashion (blogo)spheres

    • A common characteristic of the theoretical developments within the field of social media marketing is that activities to which consumers devote themselves in social media settings shift power from firms to consumers. Extant literature has therefore analysed the practices of consumers within social media and their potential implications for marketing. The current state of social media, however, suggests that these settings are undergoing a process of transformation. Although social media were initially characterised as non-commercial in nature, firms have started to manage interactions within these digital landscapes. From initially being characterised by its social base, this development implies that social media have become increasingly commercialised.The aim of this dissertation is to expand the literature on social media by describing the process through which they evolve from their initially social character to a commercial utility. More specifically, it seeks to develop a conceptual framework that captures the role of marketing processes that lead to the commercialisation of these spheres. This is done mainly through a netnographic study of the Swedish fashion blogosphere in order to explain how and why consumers and professionals interact, organise, create and appropriate commercial values in the fashion blogosphere.Drawing on theory of spheres, this dissertation proposes a sphereological understanding of social media that expands the role of marketing. It is suggested that social media may be understood as a collection of micro-spheres that, together, comprise a densely connected foam of spatiality and place. In these spheres, consumers, together with commercial actors, take part in practices that become increasingly commercial. In that sense, marketing takes the roles of navigating social media in search of symbolic meanings of value, and of affecting, negotiating and redefining atmospheres of places in the social media landscape.

  • Towards co-management of Gialova Lagoon : A Natura 2000 coastal wetland in Messinia, Greece

    • The management of Natura 2000 sites is considered as the cornerstone for the conservation and restoration of biodiversity within Europe. However, protected ecosystems provide a plethora of benefits to local societies, and support the local economy. Thus, to seek solutions for complex environmental issues within Natura 2000 sties it is imperative to approach the site of concern as a connected social-ecological system, and to strengthen the participation of stakeholders in decision-making following a co-management approach.Gialova Lagoon wetland, in Messinia, Greece represents an example of Natura 2000 site which needs to be managed. The overall aim of the PhD thesis was to assess the problem of lagoon salinization, and provide policy recommendations for wetland restoration and management of associated freshwater resources under a changing climate. The thesis has followed a social-ecological approach, by integrating DPSIR framework with participatory Systems Dynamic modelling and the concept of ecosystem services. Knowledge gaps about major social and ecological components were assessed by applying a variety of methods, namely (a) field monitoring and observations, (b) GIS analyses, (c) consultation with stakeholders, (d) modelling and scenarios.The thesis results suggested that past human interventions had multi-fold effects on the Gialova Lagoon wetland, namely hydrology alteration, ecosystem fragmentation, loss and transformation of natural habitats. Furthermore, the combined effects of alterations in hydrology and climate change have led to increased salinity in the wetland over time. These alterations had profound implications on wetland ecosystem services such as the diversity of habitats and waterbirds and the provision of fish. Under contemporary hydrological connectivity and on-going climatic conditions, the mean annual salinity of the lagoon has increased from approximately 35 g/L during the period 2016-2018 to approximately 40 g/L during the period 2021-2023 indicating a salinization increase of approximately 1 g/L per year. To identify restoration alternatives, the work under the PhD thesis has engaged scientists with local stakeholders from the sectors of agriculture, fishing, tourism, and public administration, in a co-management approach. The end product, an SDM (Systems-Dynamics model) co-created with stakeholders, was suitable for exploring scenarios for salinity regulation and management of associated freshwater resources, under a changing climate (RCP 4.5). The derived management suggestions, namely restoration of the connectivity with the surrounding freshwater bodies (river, artesian springs) and between habitats (e.g., lagoon-marshes), could result in the de-salinization of the lagoon within a 10-year period, and could be applied within the Natura 2000 framework as they consider social and ecological needs (e.g. enhancement of biodiversity and fish production). However, under current abstraction rates for irrigation and municipal water-supply, there is a high risk of groundwater scarcity during years with dry conditions, and thus investments in water-saving technologies (e.g. smart irrigation) should be promoted to ensure adequate water availability for restoration, and enhanced resilience of the local economy against groundwater scarcity.

  • Managing Mangrove Ecosystem Services for Local Livelihoods and Adaptations in Tanzania

    • Recognizing the importance of ecosystem services (ES) for peoples' livelihoods and well-being is important for decision-making processes on conservation. Mangrove ecosystems in Tanzania are protected by law, but they continue to be exposed to degradation and loss, and there is still limited information about the awareness, preferences, status and trends of the ES they provide. This thesis aims to explore the link between mangrove ecosystem services (MES) and community livelihoods, changes in MES and associated drivers of change, adaptation options and their management in Tanzania, using the Rufiji Delta and Pangani Estuary as case studies. A mixed framework of methods including focus group discussions, key informant interviews, household surveys, direct observations, and literature reviews was used to gather data. Provisioning services were the most commonly identified MES, and they were more often reported to be deteriorating than regulating, cultural, and supporting services. Proximity to mangrove forest and residence time were positively associated with communities' awareness of all identified MES. Poles for building, firewood for cooking, coastal protection, and fisheries habitats were perceived as the most important MES for sustaining local livelihoods, though perceptions varied between sites. Reliance on mangrove resources was significantly predicted by household residence time, household main occupation, household size, and the cost of alternative resources to substitute mangrove wood as a source of domestic fuel. Illegal harvesting of mangrove poles, rice cultivation, climate change and inadequate governance and conservation measures were identified as the most critical drivers of mangrove degradation, but differed significantly from place to place. Fishing was perceived as the most impacted livelihood occupation compared to the other groups of occupations. Potential ways to adapt to environmental changes in the study areas included reliance on MES for ecosystem-based adaptation (EbA), switching of occupation, diversifying crops, offshore fishing, and migrating to other areas. This thesis argues that the linkage between MES and human well-being is site-specific, and drivers impacting on mangroves and their associated services vary spatially and is greatly accelerated by anthropogenic disturbances. Raising more awareness about the multifunctionality of mangroves and committing to participatory forest management that involves local people, as well as reforming the current forest policy by incorporating clear legal mechanisms for engaging communities around mangrove management and diversifying livelihood options are re-emphasized as appropriate ways to improve mangrove conservation. Furthermore, investment by providing adequate funding for conservation in long run rather than relying on short-term international donor-funded projects are recommended to government institutions as a basis for sustainable management of mangrove forests in Tanzania.

  • Ceramic Core–Shell Particles : Synthesis and Use within Dentistry

    • Dentin hypersensitivity is one of the most prevalent conditions related to oral health, affecting a large share of the adult population. Shortcomings with the available treatment options are related to non-ideal particle sizes and degradation properties. An improved clinical outcome could possibly be obtained using a bioactive occluding agent that can offer a high, continuous release of ions, as well as having a particle size that allows for penetration into the dentin tubules. The work in this thesis focused on the development and investigation of a synthesis approach for calcium phosphate core–shell particles and the use of those in the treatment of dentin hypersensitivity. The overall aim was to increase the knowledge about the synthesis and to evaluate the in vitro performance of amorphous calcium magnesium phosphate (ACMP) particles when used as an occluding agent. The synthesis of the core-shell particles was based on precipitation reactions in aqueous solutions and the synthesized materials were studied in terms of morphological, structural, and compositional aspects. Resulting particles had diameters ranging from 400 nm–1. 5 µm (depending on reaction conditions), with morphologies and structures that were shown to correlate with the ionic radius and the concentration of the substituting ion. This insight resulted in the possibility to control the outcome of the reaction and to extend the synthesis to other alkaline earth phosphates. The mechanism of formation was suggested to be the simultaneous precipitation of primary nanoparticles (NPs) and the formation of gas bubbles that could function as soft templates.A study of the degradation properties together with a series of in vitro studies, using a dentin-disc model, indicated that the ACMP particles may be a promising candidate for clinical use. The material was shown to offer a rapid and continuous release of Ca2+, Mg2+, and phosphate, aiding surface, as well as intratubular occlusion and mineralization. Additional use of a fluoride toothpaste resulted in incorporation of F– in the mineralized material. This could enhance the in vivo performance due to the known benefits of including F– in dental tissues, e.g. decreased solubility. The ACMP particles were, furthermore, shown to be more efficient in terms of degree of occlusion when compared to other similar products available on the market. The intratubular mineralization was additionally mitigating the effect of an acid attack, which is of importance for a long-lasting effect in clinical use.

  • Integrated Agriculture and Aquaculture Systems (IAA) for Enhanced Food Production and Income Diversification in Tanzania

    • Aquaculture production in Tanzania has increased in recent years, responding to an increased demand for fish, partly because of an increasing population and declining catches of wild fish. However, the current aquaculture production is still low, dominated by small scale farming systems, that are struggling with a number of challenges such as lack of improved fish breeds, feeds, technical skills and low adoption rates.This thesis aims to assess the status of fish pond farming with a focus on integrated agriculture and aquaculture (IAA) systems in Tanzania, and to provide guidance on how these systems could be further developed to help local farmers to diversify their food production and income generation.Methodologies included field observations, interviews, structured questionnaires, field experiments and laboratory analyses. Two field surveys in six districts and seven regions of Tanzania provided insights on the current status and the contribution of small-scale pond farming and IAA to household income and food production. Differences between IAA and non-IAA fish farming practices, and their influence on farm productivity and profitability were assessed. Also, opportunities, constraints and farmers’ future plans for fish farming were explored. Two field experiments were conducted on selected IAA systems identified during the surveys. One assessed the effect of including amaranth (Amaranthus hybridus) waste in locally produced fish feed on the yield of Nile tilapia (Oreochromis niloticus). Another investigated how the combination of different vegetables (A. hybridus and Brassica rapa pekinensis) and stocking densities of O. niloticus affected the water use efficiency, need for synthetic fertilizers and overall farm productivity and profitability.The results show that small scale fish farming contributed with 13% of the farmers’ income. 38% of the fish was used for consumption and the remaining part was sold, contributing to both food security and livelihood diversification. Integration of tilapia and vegetables was the most common type of IAA system applied. Local feed ingredients contained medium to high content of crude protein, with high fat content in some animal by-products. The fish yield from the IAA ponds was 60% higher compared to non-IAA ponds because of a more frequent use of on-farm resources and better management. The net income from IAA ponds was 175% higher compared to non-IAA ponds, due to reduced costs and higher yields. Major constraints included water problems, lack of technical skills and good quality fish seeds and feeds. Overall, fish farmers had a positive attitude towards fish farming and the majority were willing to continue with fish farming, with IAA farmers being more positive than non-IAA farmers. The use of amaranth wastes as a fish feed ingredient in a tilapia-amaranth integrated system did not affect the fish growth and yield, but rather improved the feed conversion ratio. The multiple use of water in integrated tilapia-vegetable systems increased the water use efficiency and reduced the need for synthetic fertilizers. Although water from high fish stocking densities increased the vegetable yield, the overall farm productivity in the IAA system was negatively affected because of low fish growth and survival rates. Since agriculture is already popular and fish farming is expanding, promotion of IAA systems could provide an entry point for an increased adoption of aquaculture among rural farmers.

  • ACP-Mg particles for treatment of dental hypersensitivity : a mode of action study

    • Introduction/purpose:Dental hypersensitivity is a common clinical condition usually associated with exposed dentinal tubules. Occlusion of those, hindering fluid movement as a response to external stimuli, is one approach to reduce pain.[1] The occluding agent should induce mineralization and have a good filling effect. In this study, particles (200-400 nm) of Amorphous Calcium Phosphate with Mg (ACP-Mg) were used aiming to evaluate the mode of action with in-vitro studies on dentine as well as degradation properties in buffered systems.Methods:Degradation properties and ion-release was monitored by dispersing the particles in Tris-HCl (10 mg/mL) storing the dispersions at 37 °C.  In-vitro testing was performed on 1 mm thick dentine discs cut from extracted human molars. Specimens were etched in 35 % phosphoric acid, rinsed with DI-water followed by treatment by applying a gel formulation containing the particles using a soft bristled toothbrush, brushing 1 min on each side and leaving the specimen to rest for 3 min. Treatment was repeated four times during one day and samples were incubated at 37 °C in artificial saliva.Results:In vitro tests and degradation studies showed that ACP-Mg particles induced formation of Hydroxyapatite (HA). ICP-OES, XRD and SEM showed that there was a rapid release of all ions up to 6 hours followed by a re-precipitation of HA at 24 hours with a lower Mg-content. The crystallinity increased with time as the concentration of all ions decreased in the solutions. Morphological evaluation for the in-vitro tests showed that HA-like structures formed already after 24 hours on the dentine surface.  Cross-sections revealed that the particles reached as far as 80 µm from the surface and tubules, fully occluded by HA-like structures, was visible at comparable depths after 7 days.Conclusions:ACP-Mg particles can be used to reduce dentine hypersensitivity by effective occlusion of dentine tubules via rapid formation of surface and intra-tubular HA.References:1.     • Occlusion of exposed dentin tubules may eliminate or reduce dentin hypersensitivity by hindering fluid movements within the tubules. In this study, the mode of action of spherical particles of amorphous calcium magnesium phosphate (180-440 nm in diameter) was studied. A degradation study of the particles in Tris-HCl buffer showed that the particles continuously released Ca2+, Mg2+, and phosphate, and XRD analysis revealed the formation of hydroxyapatite (HA) after 1 week. The occluding effect and efficacy of the spherical particles as an occluding agent were evaluated in an in vitro study. The ACMP particles were incorporated in a gel intended for at-home use and tested on extracted human molars. Application of the particles followed by incubation in artificial saliva resulted in occlusion of exposed tubules, and examination with SEM showed that the particles could penetrate the tubules down to 100 mu m from the dentin surface. Transformation of the particles into nanocrystalline HA-structures (nanoHA) was initiated at the dentin surface within 12 h of application, and tubule penetration of the particles, accompanied by further ion release and diffusion of ions, resulted in deep intratubular occlusion in the majority of the tubules within 3 days from application. NanoHA was tightly adhered to the tubule walls, filling the entire tubule volume after 7 days. The results of this study demonstrate the mode of action of the amorphous calcium magnesium phosphate particles in occluding exposed dentin tubules. Interaction with saliva and transformation of the particles within the tubules inducing further mineralization indicate that the particles may be used as an effective treatment to reduce dentin hypersensitivity.

    • This study aimed to evaluate the occluding/remineralization performance and resistance to acid attacks of the mineralization layer formed by a tooth-desensitizing gel containing amorphous calcium magnesium phosphate (ACMP) particles and compare it to six other desensitizing products available on the market. Similar comprehensive studies are few and there is especially a lack of studies that are up to date. A dentin-disc model was used for in vitro evaluation of the desensitizing toothpastes/gels. Application of the products was performed twice daily for seven days. One set of specimens were evaluated using scanning electron microscopy (SEM) directly after the final treatment and another set was evaluated after an acid challenge, exposing specimens to 2 wt% citric acid. The ACMP desensitizing gel was the only product resulting in complete occlusion by the formation of mineralized material on the dentin surface and inside the tubules. Particle deposition was dominant after treatment with the other desensitizing products, with little or no mineralization, resulting in partial occlusion only. Sensodyne Repair & Protect and Oral-B Pro-Expert showed the highest resistance toward acid attacks. Material inside the tubules remained relatively unaffected by acid attacks in all specimens. The results in this study indicated a great variability among the occluding agents in terms of occlusion and acid resistance of the mineralization layer. The high degree of occlusion and intra-tubular mineralization that could mitigate the effect of acid solubilization indicate that the ACMP desensitizing gel may be a superior option for the treatment of dentin hypersensitivity.

    • Dentin hypersensitivity can be reduced by the use of a remineralization agent to hinder movement of fluids within the dentin tubules. Penetration of particles into the tubules and a continuous release of Ca2+ and phosphate ions can induce the mineralization of a material mimicking the mineral component of dentin, sealing the tubules. In this work, we have used complementary electron microscopy techniques to investigate the ultrastructure of dentin and crystallization and occlusion effects when using amorphous calcium magnesium phosphate (ACMP) microspheres on extracted human molars. Application of the particles in a gel intended for athome use resulted in intra-tubular mineralization of a carbonate substituted hydroxyapatite (HA). Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) showed that crystallization was initiated on the peritubular dentin (PTD) with undirected crystal growth leading to the formation of a porous material. We additionally investigated the effects from using a fluoride toothpaste to potentially improve the remineralization and anti-cariogenic properties of the ACMP microspheres. Energy dispersive x-ray spectroscopy (EDX) using TEM in scanning mode (STEM) showed that fluoride incorporation resulted in an increase in aspect ratio of the crystals, crystal growth directed towards the center of the tubule lumen and densification of the mineralized material. Thus, ACMP microspheres are promising alternatives as occluding agents and the efficacy of the particles could be further improved with the complementary use of a fluoride toothpaste.

    • How to precipitate ceramic nano- and microspheres in water based solutions only using inorganic ions is a challenge. In this study, spherical particles of alkaline earth phosphates and fluorides were synthesized using a precipitation reaction. Substituting ions, through inhibition of crystal growth, was used to induce sphere formation and to alter the morphology, size and composition of the spheres. The difference in ionic radius between the substituting ion (Mg, Ca and Sr) and the main cation (Sr and Ba) influenced the critical concentration to allow for sphere formation as well as the crystallinity. The larger difference, the lower was the concentration needed to form spheres. Low concentrations of Mg was enough to generate amorphous spheres of Sr- and Ba- phosphates whereas higher concentrations were needed if the radius difference were smaller. An increasing degree of substitution leads to a decrease in crystallinity of precipitated particles. The degree of substitution was determined to 16-55% where a low degree of ion substitution in the phosphates resulted in the formation of spheres (500-800 nm) with rough surfaces composed of apatite like phases. A higher degree of substitution resulted in amorphous spheres (500 nm- (1) mu m) with smooth surfaces.

  • Synthesis of Ion Substituted Ceramic Core-Shell Particles for Dental Applications

    • Calcium phosphate spheres are interesting alternatives for dental applications due to their chemical similarity to teeth and biocompatibility. A spherical shape with a hollow cores allows for loading of therapeutic agents for drug delivery which potentially could be combined with other applications such as tooth remineralization and treatment of hypersensitivity.Precipitation reactions are one of the techniques used for synthesizing spherical particles, but little is known about the mechanism behind the sphere formation, which makes tuning of the material properties challenging. Previously, it has been shown that substituting ions can influence the crystallization process, which can enable greater control during the synthesis.In this study, several different substituting ions has been used in the synthesis of alkaline earth phosphates, to further investigate their role in sphere formation and to develop a robust synthesis technique.Particles of alkaline earth phosphates (Ca, Sr and Ba) were synthesized with a precipitation reaction. Solutions with constituent anions and cations were mixed at room temperature, and substituting ions (Mg, Ca or Sr) were added before heating at 60-100 °C. Reaction times varied between 10 minutes to 24 hours. Characterization of precipitates was performed with SEM, DLS and FIB to analyze morphology, size and cross-sections of the spheres. Crystal structure and atomic composition was analyzed with XRD and ICP-OES.Without substituting ions, precipitates had no specific shape and crystallized in an apatitic structure or as a hydrogenated phosphate. Substituting ions stabilized the initial amorphous phase during the reaction, hindering rapid crystal growth which allowed for self-assembly into hollow, spherical particles with a diameter between 300-700 nm. The phase composition and degree of ion substitution in the precipitates depended on the size and concentration of the substituting ions. The amount of substitution was determined in the range between 5-30 %, where precipitates with a low degree of substitution crystallized in a structure similar to β-tricalcium phosphate, whereas materials with a higher degree of substitution had an amorphous structure.In this study it was shown that it was possible to obtain hollow, spherical particles of calcium-, strontium- and barium phosphate, by using substituting ions during a precipitation reaction. This indicates that the approach can be used to tailor the properties of spherical particles intended for dental applications.
