  • Impaired IFN-gamma Production after Stimulation with Bacterial Components by Natural Killer Cells from Gastric Cancer Patients

    • Gastric adenocarcinoma is a major health problem world-wide, as this is the second most common cause of cancer death in the world. It has been estimated that infection by Helicobacter pylori cause at least half of the gastric cancers. Previously, we have demonstrated that H. pylori antigens directly activate NK cells to secrete IFN-γ. There is also a marked synergistic effect in NK cells stimulated with bacterial lysate and low levels of IL-12, a cytokine which is produced by macrophages and dendritic cells in the H. pylori-infected stomach. The present study was designed to investigate whether NK cells from gastric cancer patients display an altered ability to respond to components from H. pylori and other bacteria. The results show that NK cells from peripheral blood of gastric cancer patients have a severely suppressed ability to produce IFN-γ after stimulation with H. pylori lysate and the synthetic bacterial lipoprotein FSL-1. Furthermore, the synergistic effect of IL-12 and lysate is absent in gastric cancer patients, unless the concentration of IL-12 is increased 10-fold. We also demonstrate that there is a similar lack of IFN-γ production from NK cells isolated from the gastric mucosa of cancer patients. In addition, we propose that the observed suppression is due to tumour-derived TGF-β and that increased expression of the transcription factor GATA-3 may be responsible for the TGF-β induced suppression.

  • High prevalence of pharyngeal bacterial pathogens among healthy adolescents and young adults

    • The pharyngeal mucosa can be colonized with bacteria that have potential to cause pharyngotonsillitis. By the use of culturing techniques and PCR, we aimed to assess the prevalence of bacterial pharyngeal pathogens among healthy adolescents and young adults. We performed a cross-sectional study in a community-based cohort of 217 healthy individuals between 16 and 25 years of age. Samples were analyzed for Group A streptococci (GAS), Group C/G streptococci (SDSE), Fusobacterium necrophorum, and Arcanobacterium haemolyticum. Compared to culturing, the PCR method resulted in more frequent detection, albeit in most cases with low levels of DNA, of GAS (20/217 vs. 5/217; p < 0.01) and F. necrophorum (20/217 vs. 8/217; p < 0.01). Culturing and PCR yielded similar rates of SDSE detection (14/217 vs. 12/217; p = 0.73). Arcanobacterium haemolyticum was rarely detected (3/217), and only by PCR. Overall, in 25.3% (55/217) of these healthy adolescents and young adults at least one of these pathogens was detected, a rate that is higher than previously described. Further studies are needed before clinical adoption of PCR-based detection methods for pharyngeal bacterial pathogens, as our findings suggest a high incidence of asymptomatic carriage among adolescents and young adults without throat infections.

    • Both the environment and the spatial configuration of habitat patches are important factors that shape community composition and affect species diversity patterns. Species have traits that allow them to respond to their environment. Our current knowledge on environment to species traits relationships is limited in spite of its potential importance for understanding community assembly and ecosystem function. The aim of our study was to examine the relative roles of environmental and spatial variables for the small-scale variation in Collembola (springtail) communities in a Dutch salt marsh. We used a trait-based approach in combination with spatial statistics and variance partitioning, between environmental and spatial variables, to examine the important ecological factors that drive community composition. Turnover of trait diversity across space was lower than for species diversity. Most of the variation in community composition was explained by small-scale spatial variation in topography, on a scale of 4-6 m, most likely because it determines the effect of inundation, which restricts where habitat generalists can persist. There were only small pure spatial effects on species and trait diversity, indicating that biotic interactions or dispersal limitation probably were less important for structuring the community at this scale. Our results suggest that for springtails, life form (i.e. whether they live in the soil or litter or on the surface/in vegetation) is an important and useful trait to understand community assembly. Hence, using traits in addition to species identity when analysing environment-organism relationships results in a better understanding of the factors affecting community composition.

  • The effect of landscape structure and habitat composition on the presence of the threatened parasitic sand-living beetle Apalus bimaculatus (Coleoptera: Meloidae)

    • Apalus bimaculatus (Linnaeus) (Coleoptera: Meloidae) is a beetle currently managed for conservation in Sweden. The species inhabits at-risk ephemeral and patchily distributed sandy habitats. However, little is known about its ecology and the factors important for its distribution. We censused 158 discrete sand patches within 31 potential sites for A. bimaculatus and examined which environmental variables predicted the probability of finding the beetle. Apalus bimaculatus was found at 17 sites, its presence at sand-patch scale was positively correlated with sand-patch area, sand temperature, and medium-sized sand grains. Although the beetle is assumed to be a parasite on the solitary bee, Colletes cunicularius (Linnaeus) (Hymenoptera: Colletidae), presence of the bee was a very weak predictor for A. bimaculatus, while other sand-living Hymenoptera were a strong predictor. At site-level scale, the beetle was positively correlated with total amount of sandy habitat and presence of sand-living Hymenoptera. Our study suggests that management strategies for this species should not consider sandy habitats equally. Rather, management efforts should focus on maintaining sites with larger total sandy areas, creating larger sand patches with medium-grained sand and a high degree of sun exposure. We also highlight that biotic interactions between the beetle and sand-living Hymenoptera are still poorly understood but potentially important for successful A. bimaculatus management.

    • The blister beetle (Apalus bimaculatus, Coleoptera: Meloidae) is a beetle managed for conservation in Sweden. The species inhabits at-risk ephemeral and patchily distributed sandy habitats and are dependent on stable colonies of the bee species (Colletes cunicularius) on which it parasitizes. The beetle is not considered threatened at a global or European level. It has previously been categorized as Vulnerable (VU, 2000) and near threatened (NT, 2005 & 2010) in the Swedish National List, but are now considered as Least Concern (LC, 2015). The change in categorization is due to that more populations have been found, which is believed to be an effect of overlooked populations in the past. The species is still considered as declining in the country, due to a general decrease in area of suitable habitat e.g. from sandpits becoming overgrown after excavation has ceased, and when sand dunes and other sandy areas are used for human development. The beetle population in this project has until now inhabited a sandy area between pine-dominated forest and areas used for human activities. The site is in Uppsala, Sweden, and it is located in a part of the city previously little used for human development but where major building work is planned and ongoing.

  • Using citizen-reported data to predict distributions of two non-native insect species in Sweden

    • Information on exotic species' current and potential distribution is vital for decisions on management. Species distribution models can predict where colorizations are likely; however, the collection of species' distribution data over large areas in order to parameterize the models is costly. Therefore, modelling methods that are able to use low-cost information such as citizen-reported data are potentially very useful. In this study, we used the species habitat modelling program Maxent to predict the potential geographical distribution of two non-native insect species in Sweden, the butterfly Araschnia levana and the shield bug Graphosoma lineatum. For this we used citizen-reported presence-only open-access data in combination with climate and land cover data from national databases. Our models showed that presence of A. levana was best predicted by winter temperature and habitats related to open grasslands. For G. lineatum, summer temperature and open green areas, in both urban and rural areas were the best predictors for species presence. These models show that large areas of non-colonized potential habitats exist within Sweden. For A. levana these yet-to-be-colonized habitats are mainly in the south, while for G. lineatum these habitats occur in the south and along the Baltic Sea coast. Comparisons of temporal patterns in species reporting for A. levana and G. lineatum to similar insects with known stable populations revealed large 'willingness to report' effects that could potentially bias range expansion rates. Once corrected for, current distribution expansion rates were estimated as 1.9 km/yr and 1.07 km/yr respectively. The study shows the use of public reports in conservation science as a way of gathering species information over large areas. This increases the data sources available for researchers to predict the distribution of species and have the additional value of the involvement of the public in conservation efforts.

    • This article documents the addition of 411 microsatellite marker loci and 15 pairs of Single Nucleotide Polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Acanthopagrus schlegeli, Anopheles lesteri, Aspergillus clavatus, Aspergillus flavus, Aspergillus fumigatus, Aspergillus oryzae, Aspergillus terreus, Branchiostoma japonicum, Branchiostoma belcheri, Colias behrii, Coryphopterus personatus, Cynogolssus semilaevis, Cynoglossus semilaevis, Dendrobium officinale, Dendrobium officinale, Dysoxylum malabaricum, Metrioptera roeselii, Myrmeciza exsul, Ochotona thibetana, Neosartorya fischeri, Nothofagus pumilio, Onychodactylus fischeri, Phoenicopterus roseus, Salvia officinalis L., Scylla paramamosain, Silene latifo, Sula sula, and Vulpes vulpes.     • Net Zero Energy Buildings constitute one measure to reduce energy use and increase use of energy from renewable sources. Hence, it is important share knowledge and experiences from completed projects. This case study show that it is possible to build Net Zero Energy Buildings with existing techniques. However, a common strategy to prevent or limit the build-up of ice and frost in ventilation heat exchangers, Supply fan shut off, were not suitable this project, since it is air tight buildings. After occurring problems in the first winter, ventilation pre-heater were installed to prevent the build-up of ice and frost. Thanks to placement of temperature sensor after the pre-heater, the increased energy use for pre-heater may be expected to be low, roughly 1 kWh/m2a.

    • Normalisation of measured energy use in buildings is important in order to verify their performance in user phase. Two methods for normalisation have been presented in Sweden, static and dynamic normalisation. The static normalisation considers deviating hot water use, indoor temperature, internal loads and external climate. The dynamic normalisation is based on repeated simulation, meaning that the initial simulation, carried out during the design phase, is repeated with updated conditions regarding actual use of the building and exterior climate. The ratio between the first and second simulation is used as a factor for normalisation. A pre-study has been initiated in Sweden to enable further development of the two methods. This paper present the two methods, the initiated pre-study, and some early findings. The early findings show that there is need for further development of the methods presented.

    • This article presents a Swedish case study of the regional effects of academic spin-offs. It is based on empirical data from Halmstad University and the Halmstad region, on the west coast of Sweden. The Halmstad case functions as an illustration of co-existing territorial and functional rationales, where a “rurban” lifestyle is combined with an increasingly “pracademic” knowledge. Spillover effects of university research (directly as well as indirectly) in the form of academic spin-offs have been traced over time. In doing so, we are able to identify two waves of academic entrepreneurship with direct and indirect regional effects. These waves contribute to the strengthening of the regional entrepreneurship and the attractiveness of the region. The establishment and (early) actions of the university can be seen as a stone that was thrown in the water, causing several waves to appear in the region. Whether these waves will be reinforced enough to create an entrepreneurial region, or just slowly disappear as rings on the water, also depends on the creation of an absorptive capacity and construction of the regional innovation system.

  • Student Entrepreneurs : The Influence of University, Entrepreneurship Education and Research

    • This licentiate thesis deals with the questions if and how entrepreneurship education and access to research at university stimulate student start-ups. The phenomenon of student entrepreneurs is approached in three empirical studies. In the first study mail questionnaires were sent out to former entrepreneurship students at three universities in Sweden, in order to find out if the students’ entrepreneurial behaviour was influenced by education. The second study explores differences in communication skills and the handling of liabilities of newness between entrepreneurs with entrepreneurship education at university and/or previous start-up experience. Student entrepreneurs are compared with a control group of randomly chosen Swedish entrepreneurs. In the third study, the influence from university research on student entrepreneurship as well as direct and indirect regional effects are examined through interviews with university spin-offs around Halmstad University. Entrepreneurship education at university is found to influence entrepreneurial behaviour, to increase the student’s awareness of business opportunities and to result in high start-up frequency early in life. The student entrepreneurs perceive fewer problems to handle some aspects of liability of newness as well as communicative skills in terms of openness and adaptation. Other-orientation is on the other hand supported by previous start-up experience. Hence, with experience from both entrepreneurship education and previous start-up, the students seem well prepared for future start-ups. The case study show multiple and long term indirect effects from student entrepreneurship in developing an entrepreneurial region. Established student entrepreneurs support less experienced student entrepreneurs by linking them to supplementary resources. Students are found to shoulder an important role as academic entrepreneurs in interaction with other individuals connected to the university.

  • Alternative EBNA1 expression in organ transplant patients.

    • In order to identify patients at risk for developing post-transplant lymphoproliferative disease (PTLD), a sensitive nested RT-PCR method for detection of EBNA1 gene expression in peripheral blood cells was used. EBNA1 expression in peripheral blood samples from 60 organ recipients was analyzed and compared with 24 healthy controls in a retrospective study. Overall, EBNA1-positive samples were detected at least once in 43% of the transplant patients with post-transplant lymphoproliferative disease, in 18% of the other transplant patients and in none of the healthy controls. The odds ratio for EBNA1 expression in patients with post-transplant lymphoproliferative disease was 3.42 (95% CI=1.02-11.54) compared to other transplant recipients. Together with normal EBV Q promoter initiated EBNA1 transcripts, an alternatively spliced form was expressed in peripheral blood cells in the above-mentioned transplant patients. This transcript lacks the U leader exon in the 5'-untranslated region (UTR). We have previously identified and characterized a functional internal ribosome entry site, the EBNA IRES, in the untranslated U leader exon of EBNA1. Transfection experiments with EBNA1 coding plasmids followed by Western blot showed that the EBNA IRES promotes cap-independent translation and increases the EBNA1 protein level. The alternative EBNA1 transcript lacking this function is expressed in the majority of the investigated EBNA1-positive patient samples as well as in some EBV-positive B-cell lines. Alternative splicing in this form gives EBV potential to regulate the translation of EBNA1 by modifying the 5' UTR. These findings indicate a new mechanism for EBNA1 expression in vivo.

  • Effects of geriatric interdisciplinary home rehabilitation on complications and readmissions after hip fracture : a randomized controlled trial

    • Objective: This pre-planned secondary analysis of geriatric interdisciplinary home rehabilitation, which was initially found to shorten the postoperative length of stay in hospital for older individuals following hip fracture, investigated whether such rehabilitation reduced the numbers of complications, readmissions, and total days spent in hospital after discharge during a 12-month follow-up period compared with conventional geriatric care and rehabilitation.Design: Randomized controlled trial.Setting: Geriatric department, participants' residential care facilities, and ordinary housing.Subjects: Individuals aged ⩾70 years with acute hip fracture (n = 205) were included.Intervention: Geriatric interdisciplinary home rehabilitation was individually designed and aimed at early discharge with the intention to prevent, detect, and treat complications after discharge.Main measures: Complications, readmissions, and days spent in hospital were registered from patients' digital records and interviews conducted during hospitalization and at 3- and 12-month follow-up visits.Results: No significant difference in outcomes was observed. Between discharge and the 12-month follow-up, among participants in the geriatric interdisciplinary home rehabilitation group (n = 106) and control group (n = 93), 57 (53.8%) and 44 (47.3%) had complications (P = 0.443), 46 (43.4%) and 38 (40.9%) fell (P = 0.828), and 38 (35.8%) and 27 (29.0%) were readmitted to hospital (P = 0.383); the median total days spent in hospital were 11.5 and 11.0 (P = 0.353), respectively.Conclusion: Geriatric interdisciplinary home rehabilitation for older individuals following hip fracture resulted in similar proportions of complications, readmissions, and total days spent in hospital after discharge compared with conventional geriatric care and rehabilitation.

    • This paper will focus on a sense of place in prehistory. The chapter argues for the importance of the sensuous experiences of places, and in this case natural places, in the creation of memory and shared conceptions of place and landscape. Natural places played important roles in the process of creating the societal order in prehistory and in this respect can be considered similar to monuments in defining a sense of place. Here a fen in Malmö in Sweden is discussed, where artefacts were deposited during a period that includes most of the Neolithic period.The topography, the varying vegetation, the water table, as well as the deposited artefacts, created spatial structures that were experienced through the senses by those who moved around this place and performed the acts of deposition. These experiences resulted in a differentiation of this place from the surroundings, which in many cases acted as a ritualization strategy for the acts that took place there. This gave the place a special significance in thelandscape. The fact that this was a recurring practice at the fen, also underlines the historic significance of the place. The sensuous experiences of the acts, the place and the objects also created relations between people, and as such, a social organization. Variations in the social identities that were created at the fen during the course of the Neolithic are discussed in connection to the surrounding society. It seems the fen was mainly used by those that were not a part of the competition for the highest status in society, i.e. the elite, but rather a social stratum of society less likely to have, and dispose of, prestige objects. This part of society did not consist of a grey mass of people but rather a complex web of social relations.

    • The emerging insects-as-food industry is increasingly promoted as a sustainable alternative to other animal protein production systems. However, the exact nature of its environmental benefits are uncertain because of the overwhelming lack of knowledge concerning almost every aspect of production: from suitable species, their housing and feed requirements, and potential for accidental release. If ecological sustainability is to be a hallmark of mass insect rearing for consumption, ecologists need to engage in research related to sustainability criteria that are directly linked to key elements of the development of the industry. There is more to this subject than simply comparing feed-conversion ratios (FCRs) of insects to traditional livestock production, and we highlight areas where research needs to be immediately focused.

    • The art of population modelling is to incorporate factors essential for capturing a population's dynamics while otherwise keeping the model as simple as possible. However, it is unclear how optimal model complexity should be assessed, and whether this optimal complexity has been affected by recent advances in modelling methodology. This issue is particularly relevant to small populations because they are subject to complex dynamics but inferences about those dynamics are often constrained by small sample sizes. We fitted Bayesian hierarchical models to long-term data on vital rates (survival and reproduction) for the toutouwai Petroica longipes population reintroduced to Tiritiri Matangi, a 220-ha New Zealand island, and quantified the performance of those models in terms of their likelihood of replicating the observed population dynamics. These dynamics consisted of overall growth from 33 (+/- 0.3) to 160 (+/- 6) birds from 1992-2018, including recoveries following five harvest events for further reintroductions to other sites. We initially included all factors found to affect vital rates, which included inbreeding, post-release effects (PRE), density-dependence, sex, age and random annual variation, then progressively removed these factors. We also compared performance of models where data analysis and simulations were done simultaneously to those produced with the traditional two-step approach, where vital rates are estimated first then fed into a separate simulation model. Parametric uncertainty and demographic stochasticity were incorporated in all projections. The essential factors for replicating the population's dynamics were density-dependence in juvenile survival and PRE, i.e. initial depression of survival and reproduction in translocated birds. Inclusion of other factors reduced the precision of projections, and therefore the likelihood of matching observed dynamics. However, this reduction was modest when the modelling was done in an integrated framework. In contrast, projections were much less precise when done with a two-step modelling approach, and the cost of additional parameters was much higher under the two-step approach. These results suggest that minimization of complexity may be less important than accounting for covariances in parameter estimates, which is facilitated by integrating data analysis and population projections using Bayesian methods.

    • The area around the villages of Kvarnby and S.dra Sallerup in south-west Scania is the only known flint-mining site in Sweden. Radiocarbon dates show that the flint was mined mainly during the earliest phase of the Early Neolithic, between c. 4000 and 3600 BC, thus coinciding with the earliest evidence of the Funnel Beaker Culture in the region. The type of flint, the size of the flint nodules, production debris in the mining area and the concentration of point-butted axes to south-west Scania all suggest that the mining was related to the extraction of flint for the production of point-butted axes. However, considering the abundance of easily available flint elsewhere in the region, it seems clear that the mining was not motivated purely by economic reasons. We suggest that the very extraction of flint from pits and shafts in the chalk was socially and symbolically significant in itself.

    • Despite the growing interest in relationships between ecological variables and individual immune function, few empirical data have been available from wild populations. In this study, I assayed the immune response from 370 wild-caught bush-crickets, Metrioptera roeseli, from 20 experimentally introduced populations, by measuring individual encapsulation responses to a surgically implanted nylon monofilament. Bush-crickets descended from populations introduced into larger habitat patches showed a greater immune response when compared to individuals from smaller habitat-area introductions. Also, there was a significant positive correlation between immune response and the amount of linear elements at the introduction site. However, there was a lack of effect of population variables (i.e., propagule size and rate of population growth) on immune response. These results suggest that large-scale environmental parameters, such as patch size and connectivity, can be important for an individual's physiological health and its ability to defend against disease-causing agents. Such effects are likely to compound the negative impacts associated with isolation of sub-populations and habitat fragmentation.

Samtidigt som tidigare definitioner av begreppet har problematiserats, har det också vidgats till att omfatta allt fler kategorier av handlingar. Ämnet har också rört sig från sin religionshistoriska vagga och blivit en egen disciplin: Ritual Studies, som förutom religionsvetenskap och socialantropologi också inkluderar ämnen som konstvetenskap, teatervetenskap, litteraturvetenskap, etologi, etnologi, psykologi, sociologi osv. Vi vill här visa att också arkeologin har en plats i denna diskussion.

    • Field archaeology entails everything archaeologists do outside of the office; however, when scrutinizing the concept of the field in field archaeology, it is a rather complex issue. A short history of archaeological fieldwork shows that social factors influenced knowledge production in the field and still do today. An account of the archaeological process, concerning both commercial and research projects, illuminates what archaeologists do in the field during the various phases. Challenges and possibilities ahead affect how fieldwork will be carried out in the future. Among those are increasing demands on societal relevance and inclusive practices, increasing and accelerating technical and digital development and increasing adaption to market principles.

    • In order to understand ritual in the past, archaeology has long relied on theories developed in other disciplines. While these theories, which often rely on written or oral information, have added many important dimensions to our interpretation of the archaeological record, they have often proven difficult to successfully articulate with the archaeological sources. Moreover, archaeology has tended to remain on the receiving end of the formulation of social theory, and has only rarely participated in the theoretical development and critique. In this article we argue that we see a central role for archaeology to contribute to the development of ritual theory. Through two case studies from Scandinavian prehistory we illustrate how the application of a practice-based ritual theory allows us to more firmly connect the theoretical framework to our archaeological sources. This connection not only leads us toward a synchronization of materials, methods and theories, but it also allows us to engage in the broader interdisciplinary theoretical discussion about ritual. The specific challenges posed by the archaeological sources and the archaeological process of interpretation point to new questions relating to the application of theoretical frameworks, and may even suggest some solutions.

    • This thesis deals with questions concerning theoretical perspectives and archaeological interpretation. It is also a thesis about prehistoric wetland depositions, as they illustrate the discussion. The starting point of the study is the use of the concept of sacrifice (or offering) in Scandinavian archaeology. There are several assumptions, both explicit and implicit, that are considered problematic. The assumed universality of sacrifice is one example. In some cases artefacts deposited in wetlands are categorized as ritual deposits. The use of this term warrants an examination of the concept of ritual as well, and it too is associated with similar problems. Instead of using the concept of sacrifice another approach is proposed. In this study, practice theory, and especially the concept of ritualization, are used as a starting point. This means that the term sacrifice is not used as a category of interpretation. In fact, the question whether the material is sacrificed or not becomes irrelevant with this perspective. Instead alternative results that may complement or replace the sacrificial category are explored. In short, the aim of this study is to apply practice theory to material deposited in wetland – traditionally interpreted as sacrifices – to enable an empirically informed discussion of archaeological interpretations and results. To achieve this aim the tools made available in practice theory are used, mainly the concepts of embodiment and objectification. The significance of the past in the past is also discussed. The archaeological material from a fen at Hindbygården, Malmö, in south-west Scania in southern Sweden is mainly dated to a period covering more than three thousand years, from the Late Mesolithic to the Early Bronze Age. There were also a few later activities. The long period of use, along with its variation and complexity, makes this material well suited for this kind of study. An archaeobotanical study also enables a discussion of the role of the physical environment. The environment is important in this study, as the spatial structures were a part of what was created by the actions that took place at the fen, and the environment influenced the experience of the depositions.
