  • Transcriptome sequencing in pediatric acute lymphoblastic leukemia identifies fusion genes associated with distinct DNA methylation profiles

    • Background: Structural chromosomal rearrangements that lead to expressed fusion genes are a hallmark of acute lymphoblastic leukemia (ALL). In this study, we performed transcriptome sequencing of 134 primary ALL patient samples to comprehensively detect fusion transcripts. Methods: We combined fusion gene detection with genome-wide DNA methylation analysis, gene expression profiling, and targeted sequencing to determine molecular signatures of emerging ALL subtypes. Results: We identified 64 unique fusion events distributed among 80 individual patients, of which over 50% have not previously been reported in ALL. Although the majority of the fusion genes were found only in a single patient, we identified several recurrent fusion gene families defined by promiscuous fusion gene partners, such as ETV6, RUNX1, PAX5, and ZNF384, or recurrent fusion genes, such as DUX4-IGH. Our data show that patients harboring these fusion genes displayed characteristic genome-wide DNA methylation and gene expression signatures in addition to distinct patterns in single nucleotide variants and recurrent copy number alterations. Conclusion: Our study delineates the fusion gene landscape in pediatric ALL, including both known and novel fusion genes, and highlights fusion gene families with shared molecular etiologies, which may provide additional information for prognosis and therapeutic options in the future.

    • Greenhouse gas emissions from drained agricultural peatlands contribute significantly to global warming. In a laboratory study using intact cores of peat soil from eight different sites in Sweden, factors controlling the emission of the greenhouse gases nitrous oxide (N2O) and methane (CH4) were examined. Soil properties, and the abundance of the total microbial community (16S rRNA gene abundance), and genes encoding for functions controlling N2O emissions (bacterial and archaeal amoA, nirS, nirK, nosZI, and nosZII) were analyzed and compared against measured greenhouse gas emissions. Emissions were measured at different drainage levels, i.e., higher soil water suction values, since drainage is an important factor controlling greenhouse gas emissions from peat soils. The results showed that N2O and CH4 emissions were generally low, except for N2O emissions at near water-saturated conditions, for which three soils displayed high values and large variations in fluxes. Relationships between N2O emissions and soil properties were mainly linked to soil pH, with higher emissions at lower pH. However, specific assemblages of nitrogen cycling guilds that included nosZII, typically present in non-denitrifying N2O reducers, were detected in soils with low N2O emissions. Overall, these results indicate that both pH and biotic controls determine net N2O fluxes.

    • We had the fortune as PhD-students and scientists in Animal Ecology at Uppsala University, to spend joyful and creative field work summers at Klubban Biological Station, during the 1980-90’s. A reunion in June 2018 resulted in this report highlighting research on pipefishes, gobies, sticklebacks and nudibranchs. Our research on these animals have provided novel insights and knowledge of the process of sexual selection and paternal care. These animals have, in many aspects, now become model organisms in evolutionary behavioral ecology in marine environments. Our list of publications provides many examples of how environmental factors influence how sexual selection and mate choice operate, how predictors like potential reproductive rates, operational sex ratios work and how male parental care is prominent in influencing selection. This research, that started at Klubban, has broadened our understanding of the ecological importance of shallow marine areas. The evolutionary understanding of how males and females can behave and how adaptive traits are selected in interaction with social and an increasingly changing ambient environment is in focus in our continued scientific endeavors. We have happily compiled this report illustrating how science and scientist can stimulate each other at a wonderful place like Klubban Biological Station, with the access to amazing organisms like pipefishes, gobies, sticklebacks and nudibranchs.

    • Denna rapport redovisar resultat från projektet Cirkulära plastgolv, som pågick från november 2019 till februari 2020. Syftet med projektet var att identifiera en möjlig avsättning för det plastmaterial som samlas in genom systemet GBR Golvåtervinning, men inte återvinns av golvtillverkare. Ett annat syfte har varit att öka medvetenheten bland offentliga aktörer om insamlingssystemet och hur de kan bidra till att det används i större utsträckning.

    • Rapporten redovisar arbeten från kursen Restaureringskonst på Konsthögskolan som under läsåret 2015-2016 har arbetat med tema Museum och Iscensättning. Hur kulturmiljöer påverkas när de blir museum, hur man restaurerar för museum, hur man bevarar och utvecklar en plats för kunskapsutbyte, bildning och upplevelser. Mälsåkers slott, utanför Mariefred, förvaltat av Statens Fastighetsverk, var studieobjekt för 20 studenter, (yrkesverksamma arkitekter, antikvarier, ingenjörer, konservatorer m fl) där utbildningens olika studiemoment som uppmätning, inventering, dokumentation har tillämpats. Fältarbetet har följts av studier i historik, kulturhistorisk värdering och analyser som gett visioner och gestaltningsförslag på ny verksamhet i slottet. Rapporten presenterar förslagen, med text, skisser, foton och ritningar. Fyra olika typer av museum med varierande grad av åtgärder, förändringar och utveckling av slottet. Restaureringsexperiment redovisas utifrån traditionella och digitala dokumentationsmetoder. Dessutom finns arbeten om barockens ljus, bladguld, brandskydd, pod-radio, kraftstation, engelska parken, dekorationsmålade tak, Gustavianum, Julius Kronbergs ateljé och barockträdgård m m. Förslagen visar att det är möjligt att transformera Mälsåkers barockslott till ett museum av idag med bibehållen historik och synliga tidslager.

  • The mixed blessing of responsibility relief: An application to household recycling and curbside waste collection

    • This paper addresses the role of personal norms and warm glow in influencing households' waste recycling preferences. The purpose is to explore inter-household differences in the preferences toward the introduction of curbside recycling, which implies that households are relieved from the responsibility of transporting sorted waste to assigned drop-off stations. The main theoretical point of departure for the analysis is an existing model that integrates norm-motivated behavior into neoclassical utility theory. This builds on the assumption that the household members have preferences for upholding a self-image as responsible—norm-compliant—persons, and it also contains a warm-glow component. The empirical investigation relies on a postal survey to households in a Swedish municipality, and this asks households about their willingness-to-pay (WTP) for the introduction of a curbside recycling scheme, as well as about time use and the presence of personal norms and warm glow motives. The results are based on a Heckman selection specification and show that individuals expressing a strong personal norm for recycling are more likely to be willing to pay for curbside recycling, while those with strong warm glow motives are less likely to do so. This suggests the existence of a mixed blessing of responsibility relief. Curbside recycling implies that households are relieved from a moral responsibility that takes time away from leisure activities, but they also experience a loss in warm glow as such a scheme removes the possibility to pursue something that they have learned to appreciate. There could then exist ‘motivational inertia' making it difficult for policy makers to activate personal norms for new pro-environmental household activities in replacement of existing ones.

  • Identification of molecular targets associated with transformed diffuse large B cell lymphoma using highly purified tumor cells

    • Follicular lymphoma (FL) frequently transforms into the more aggressive diffuse large B cell lymphoma (DLBCL-tr), but no protein biomarkers have been identified for predictive or early diagnosis. Gene expression analyses have identified genes changing on transformation but have failed to be reproducible in different studies, reflecting the heterogeneity within the tumor tissue and between tumor samples. Gene expression analyses on Affymetrix Human Genome U133 Plus 2.0 arrays were performed, using flow cytometry sorted tumor cells derived from FL and transformed DLBCL. To identify molecular targets associated with the transformation, subsequent immunohistochemistry (IHC) analyses of the corresponding proteins were performed. Using highly purified cells, this study identified 163 genes, which were significantly deregulated during the transformation in a majority of cases. Among the upregulated transcripts, 13 genes were selected for validation using IHC, based on the availability of commercial antibodies, and galectin-3 and NEK2 proteins specifically identify DLBCL-tr, when compared with FL. We demonstrate that by purifying tumor cells through cell sorting, thereby reducing the heterogeneity due to infiltrating cells, it was possible to identify distinct differences between tumor entities rather than variations due to cellular composition. Galectin-3 and NEK2 both identified a subgroup of DLBCL-tr, and the function of these protein markers also suggests a biological role in the transformation process.

    • Transcription factors (TFs) are critical for B-cell differentiation, affecting gene expression both by repression and transcriptional activation. Still, this information is not used for classification of B-cell lymphomas (BCLs). Traditionally, BCLs are diagnosed based on a phenotypic resemblance to normal B-cells; assessed by immunohistochemistry or flow cytometry, by using a handful of phenotypic markers. In the last decade, diagnostic and prognostic evaluation has been facilitated by global gene expression profiling (GEP), providing a new powerful means for the classification, prediction of survival, and response to treatment of lymphomas. However, most GEP studies have typically been performed on whole tissue samples, containing varying degrees of tumor cell content, which results in uncertainties in data analysis. In this study, global GEP analyses were performed on highly purified, flow-cytometry sorted tumor-cells from eight subgroups of BCLs. This enabled identification of TFs that can be uniquely associated to the tumor cells of chronic lymphocytic leukemia (CLL), diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), hairy cell leukemia (HCL), and mantle cell lymphoma (MCL). The identified transcription factors influence both the global and specific gene expression of the BCLs and have possible implications for diagnosis and treatment.

    • BACKGROUND & AIMS: The signaling mechanisms that regulate trypsinogen activation and inflammation in acute pancreatitis (AP) are unclear. We explored the involvement of the calcium- and calcineurin-dependent transcription factor nuclear factor of activated T-cells (NFAT) in development of AP in mice. METHODS: We measured levels of myeloperoxidase and macrophage inflammatory protein-2 (CXCL2), trypsinogen activation, and tissue damage in the pancreas 24 h after induction of AP by retrograde infusion of taurocholate into the pancreatic ducts of wild-type, NFAT luciferase reporter (NFAT-luc), and NFATc3-deficient mice. We isolated acinar cells and measured NFAT nuclear accumulation, trypsin activity, and expression of NFAT-regulated genes. RESULTS: Infusion of taurocholate increased the transcriptional activity of NFAT in the pancreas, aorta, lung, and spleen of NFAT-luc mice. Inhibition of NFAT with A-285222 blocked taurocholate-induced activation of NFAT in all organs. A-285222 also reduced taurocholate-induced increases in levels of amylase, myeloperoxidase and CXCL2; activation of trypsinogen; necrosis of acinar cells; edema; leukocyte infiltration; and hemorrhage in the pancreas. NFATc3-deficient mice were protected from these effects of taurocholate. Similar results were obtained using an L-arginine-induced model of AP. Reverse transcriptase PCR and confocal immunofluorescence analyses showed that NFATc3 is expressed by acinar cells. NFATc3 expression was activated by stimuli that increase intracellular calcium; activation was prevented by the calcineurin blocker cyclosporine A or A-285222. Activation of trypsinogen by secretagogues in acinar cells was prevented by pharmacologic inhibition of NFAT signaling or lack of NFATc3. A-285222 also reduced expression of inflammatory cytokines such as CXCL2 in acinar cells. CONCLUSIONS: NFATc3 regulates trypsinogen activation, inflammation, and pancreatic tissue damage during development of AP in mice, and might be a therapeutic target.

MAES is one of the building-block initiatives supporting the EU Biodiversity Strategy to 2020.

    • Introduction: Clinical trials have demonstrated a positive effect of nusinersen therapy on survival of infants with SMA type 1. However, there is a lack of data outside clinical trials on how the introduction of nusinersen has affected the survival of patients with SMA. We therefore set out to analyse survival in patients diagnosed at less than 24 months before, during and after the introduction of nusinersen in a nationwide population-based real-world setting. Methods: SMA patients diagnosed before the age of 24 months in the time period between February 21, 2000 and December 19, 2019 were identified using ICD-codes, and medical procedures for identification of treatment utilizing information from the public available National Patient Registry held by the National Board of Health and Welfare. Data was divided into 3 different calendar periods (before, during, and after introduction of nusinersen treatment in Sweden). Time to Event analysis was then applied. Results: A total of 155 patients were enrolled in the study, and median follow-up was 1.14 years (inter-quartile range (IQR): 0.27-8.37 years). Data did not provide conclusive evidence that survival differed between the calendar periods (P-value from the log-rank test = 0.419) and while hazards were lower in the middle period, HR 0.70 (95% CI: 0.34-1.47), and 3, HR 0.71 (95% CI: 0.28-1.77) compared to the first period, all confidence in-tervals were wide., However, nusinersen treatment was associated with a decreased mortality rate, HR 0.05 (95% CI: 0.01-0.37). Conclusion: SMA patients receiving nusinersen therapy had a dramatically increased overall survival compared to patients not receiving therapy. This indicates that nusinersen treatment has an effect on survival, in patients diagnosed with SMA, in a nationwide real-world setting. Larger studies are warranted.

    • PurposeMolecular radiotherapy is a treatment modality that is highly suitable for targeting micrometastases and [177Lu]Lu-DOTATATE is currently being explored as a potential novel treatment option for high-risk neuroblastoma. p53 is a key player in the proapoptotic signalling in response to radiation-induced DNA damage and is therefore a potential target for radiosensitisation.MethodsThis study investigated the use of the p53 stabilising peptide VIP116 and [177Lu]Lu-DOTATATE, either alone or in combination, for treatment of neuroblastoma tumour xenografts in mice. Initially, the uptake of [177Lu]Lu-DOTATATE in the tumours was confirmed, and the efficacy of VIP116 as a monotherapy was evaluated. Subsequently, mice with neuroblastoma tumour xenografts were treated with placebo, VIP116, [177Lu]Lu-DOTATATE or a combination of both agents.ResultsThe results demonstrated that monotherapy with either VIP116 or [177Lu]Lu-DOTATATE significantly prolonged median survival compared to the placebo group (90 and 96.5 days vs. 50.5 days, respectively). Notably, the combination treatment further improved median survival to over 120 days. Furthermore, the combination group exhibited the highest percentage of complete remission, corresponding to a twofold increase compared to the placebo group. Importantly, none of the treatments induced significant nephrotoxicity. Additionally, the therapies affected various molecular targets involved in critical processes such as apoptosis, hypoxia and angiogenesis.ConclusionIn conclusion, the combination of VIP116 and [177Lu]Lu-DOTATATE presents a promising novel treatment approach for neuroblastoma. These findings hold potential to advance research efforts towards a potential cure for this vulnerable patient population.

  • Modelling Subscription‑Based Streaming Services for Books

    • By departing from the Nordic countries, the purpose is to explain and conceptualize how subscription-based streaming services for books operate in the contemporary book trade, and furthermore to discuss what effects these new business models are causing regarding the production, distribution, and consumption of books. A model is proposed that outlines the aspects of streaming services and the relationships to other actors involved. The model illustrates how streaming services affect the entire chain of publishing, and influence the constitution of books in contemporary digital culture. Since audiobooks are by far most popular, the production get altered to suit this format. Streaming services themselves also act as publishers, which makes them powerful. Distributional aspects are both more complex and more important in streaming services than in traditional book retailing. When consumption is transferred to digital platforms, the distributional frame and the final product converge, and consumer data becomes a key source of information.

    • This paper presents a lumped thermal model of a dual-axis laser micromirror device for beam steering in a free-space optical (FSO) communication system, designed for fractionated spacecraft. An FSO communication system provides several advantages, such as larger bandwidth, smaller size and weight of the communication payload and less power consumption. A dual-axis mirror device is designed and realized using microelectromechanical systems technology. The fabrication is based on a double-sided, bulk micromachining process, where the mirror actuates thermally by joints consisting of v-grooves filled with the SU-8 polymer. The size of the device, consisting of a mirror, which is deflectable versus its frame in one direction, and through deflection of the frame in the other, is 15.4 × 10.4 × 0.3 mm3. In order to further characterize and understand the micromirror device, a Simulink state-space model of the actuator is set up using thermal and mechanical properties from a realized actuator. A deviation of less than 2% between the modelled and measured devices was obtained in an actuating temperature range of 20–200 °C. The model of the physical device was examined by evaluating its performance in vacuum, and by changing physical parameters, such as thickness and material composition. By this, design parameters were evaluated for performance gain and usability. For example, the crosstalk between the two actuators deflecting the mirror along its two axes in atmospheric pressure is projected to go down from 97% to 6% when changing the frame material from silicon to silicon dioxide. A feedback control system was also designed around the model in order to examine the possibility to make a robust control system for the physical device. In conclusion, the model of the actuator presented in this paper can be used for further understanding and development of the actuator system.

  • Hearing outcome after myringoplasty in Sweden: A nationwide registry-based cohort study

    • Objectives To present hearing results after successful primary myringoplasty surgeries registered in the Swedish Quality Registry for Myringoplasty and to evaluate the chance of hearing improvement and the risk of hearing loss. Design A retrospective nationwide cohort study based on prospectively collected registry data between 2002 and 2012. Settings Registry data from secondary and tertiary hospitals performing myringoplasty. Participants Patients with healed tympanic membrane after primary myringoplasty surgery performed from 2002 to 2012 in Sweden. Main outcome measures Postoperative hearing results, hearing gain and air-bone gap (ABG). Results In 2226 myringoplasties, air conduction audiograms were recorded, and the average preoperative pure tone average (PTA(4)) of the group was 28.5 dB, which improved postoperatively to 19.6 dB with an average of 8.8 dB improvement. Bone conduction was measured for 1476 procedures. Closure of the ABG to 10 dB or less was achieved in 51% of the ears and to less than 20 dB in 89% of the ears. Sixty-one percent of patients with preoperatively deteriorated hearing experienced improved hearing, but 3% of all patients experienced deteriorated hearing. After the surgery, 93% of the patients were satisfied. Conclusions Hearing results after successful myringoplasty surgery are often favourable, but although the tympanic membrane is healed, hearing improvement is not guaranteed, and hearing deterioration can also occur.

    • BACKGROUND: The Swedish Quality Register for Ear Surgery (SwedEar) is a national register monitoring surgical procedures and outcomes of ear surgery to facilitate quality improvement. The value of the register is dependent on the quality of its data. SwedEar has never been validated regarding data quality or missing entries. Therefor, the purpose of this study was to assess coverage, completeness and response rate in the register and validate the physicians' reported data accuracy.METHODS: In this validation study, the completeness, response rate and missing registrations were analysed. Data in SwedEar were compared with the yearly collected statistics of otosurgical procedures in The Swedish Otosurgical Society and the comparison of rates between groups was calculated with Fisher's exact test. Validation of registered data accuracy was performed on every 20th registered case during a five-year period. Data were reabstracted from medical records and compared with the original registration. Interrater agreement, reliability measures, Cohen's kappa, Gwet's AC1 and positive predictive value were calculated.RESULTS: SwedEar has a coverage of 100%. The completeness of registered cases was 84% and the response rate was 74%. The validation of data accuracy assessed 13 530 variables, including audiograms. Less than 3% of incorrect or missing variables were identified. For most of the pre- and postoperative variables the Kappa and Gwet´s AC1 results show an almost perfect agreement (> 0.80). For audiogram data the ICC shows an excellent reliability (> 0.9) for all but one value.CONCLUSION: This validation shows that SwedEar has excellent coverage, high completeness, and that the data in the register have almost perfect reliability. The data are suitable for both clinical and research purposes. Further efforts to improve completeness are warranted.

    • Background: We studied the association between cardiorenal function and survival, neurological outcome and trends in survival after in-hospital Methods: We included cases aged 18 years in the Swedish Cardiopulmonary Resuscitation Registry during 2008 to 2020. The CKD-EPI equation was used to calculate estimated glomerular filtration rate (eGFR). A history of heart failure was defined according to contemporary guideline criteria. Logistic regression was used to study survival. Neurological outcome was assessed using cerebral performance category (CPC). Results: We studied 22,819 patients with IHCA. The 30-day survival was 19.3%, 16.6%, 22.5%, 28.8%, 39.3%, 44.8% and 38.4% in cases with eGFR < 15, 15-29, 30-44, 45-59, 60-89, 90-130 and 130-150 ml/min/1.73 m2, respectively. All eGFR levels below and above 90 ml/min/1.73 m2 were associated with increased mortality. Probability of survival at 30 days was 62% lower in cases with eGFR < 15 ml/min/1.73 m2, compared with normal kidney function. At every level of eGFR, presence of heart failure increased mortality markedly; patients without heart failure displayed higher mortality only at eGFR below 30 ml/min/1.73 m2. Among survivors with eGFR < 15 ml/min/1.73 m2, good neurological outcome was noted in 87.2%. Survival increased in most groups over time, but most for those with eGFR < 15 ml/min/1.73 m2, and least for those with normal eGFR. Conclusions: All eGFR levels below and above normal range are associated with increased mortality and this association is modified by the presence of heart failure. Neurological outcome is good in the majority of cases, across kidney function levels and survival is increasing.

    • The thermohaline stream function has previously been used to describe the ocean circulation in temperature and salinity space. In the present study, the Lagrangian thermohaline stream function is introduced and computed for northward flowing water masses in the Atlantic Ocean, using Lagrangian trajectories. The stream function shows the water-mass transformations in the Atlantic Ocean, where warm and saline water is converted to cold and fresh as it flows from 17 degrees S to 58 degrees N. By analysing the Lagrangian divergence of heat and salt flux, the conversion of temperature is found to take place in the Gulf Stream, the upper flank of the North Atlantic subtropical gyre and in the North Atlantic Drift, whereas the conversion of salinity rather occurs over a narrower band in the same regions. Thus, conversions of temperature and salinity as shown by the Lagrangian thermohaline stream function are confined to the same regions in the domain. The study of a specific, representative trajectory shows that, in the absence of air-sea interactions, a mixing process leads to the conversion of temperature and salinity from warm and saline to cold and fresh, and that this process is confined to the North Atlantic subtropical gyre. However, to define and to understand this process, further investigation is needed.

Lagrangian trajectories have here been used to divide the North Atlantic Ocean circulation into four different pathways. In the boundary between the Subpolar and Subtropical Gyres, we show that the northward flowing waters exchange heat and salt with the water originating from the subpolar regions. This subsurface water mass exchange takes place in the first 1,000 m and is a key piece of the puzzle of how the Atlantic Meridional Overturning Circulation transports heat and salt. Between 30 & DEG; and 60 & DEG;N the northward flowing water loses 8.8 Gg/s salt to the Subpolar Gyre and an equivalent loss of only 1.7 Gg/s to the atmosphere due to the net fresh water influx.

  • Patients' Experiences After Attempted Suicide : A Literature Review

    • The aim of this study has been to synthesize research on suicidal patients' experiences of the suicide process. A literature search was performed in CINAHL, PubMed, and PsycINFO, and the analysis of the 15 articles covered was based on meta-synthesis. Patients experience a wide variety of feelings regarding their situation during the suicide process, and these exist on two levels: they relate to the different aspects of care that the patients receive and the patients' need to communicate with others and regain hope. The patients in this study described the struggle to maintain hope when life became too difficult and their suffering despite a sense of security, and they sought to achieve emotional balance. A good understanding of how suicidal individuals live with and manage suicidal ideation, while maintaining hope is important for planning effective nursing care. Further research from the patient perspective is needed to further develop psychiatric care for people at risk of suicide.

    • The meridional transport of mass, heat and salt in the North Atlantic Ocean is often described for separate regions and parts, but rarely are all components of the circulation followed in the same study. In the present study we use Lagrangian trajectories to divide the North Atlantic Ocean Circulation into four different pathways, all contributing to the total circulation and its appurtenant heat and salt changes. In the boundary between the Subpolar and Subtropical Gyres, we show that the northward flowing waters in the North Atlantic Ocean lose heat and salt through exchange with the water originating from the subpolar North Atlantic Ocean. This in turn means that the subpolar waters gain this amounts of heat and salt.Water leaving the subpolar region as North Atlantic Deep Water are clearly distinguishable from this subpolar water, both geographically and in temperature and salinity. The southward flowing North Atlantic Deep Water meets the colder and fresher Antarctic Bottom Water in the interior, where they exchange heat and salt before returning southwards as a unified flow. The separation of the overturning circulation by Lagrangian trajectories thus reveals how the components of the North Atlantic Ocean Circulation exchange heat and salt and thus have contrasting roles.

  • The Water Mass Transformation in the Upper Limb of the Overturning Circulation in the Southern Hemisphere

    • The warming and salinification of the northwards flowing water masses from the Southern Ocean to the tropics are studied with Lagrangian trajectories simulated using fields from an Earth System Model. The trajectories are used to trace the geographical distribution of the water mass transformation and connect it with the pathways of the upper limb of the overturning circulation in the Southern Hemisphere. In the Antarctic Circumpolar Current water gains heat just below the mixed layer, mainly when the layer is thin during Austral spring and summer. This gain is therefore suggested to be a consequence of heat flux from the atmosphere and mixing processes at the base of the mixed layer. In the Southern Hemispheric subtropical gyres on the other hand, a large warming and salinification of the northwards flowing water results from internal mixing with other warmer and more saline water masses. Close to the Antarctic shelf waters are getting fresher as a result of ice melting, whereas further north, in the Antarctic Circumpolar current, waters are getting more saline as a result of evaporation. Our results show that it is not only the heat and freshwater fluxes through the sea surface that control the heat and salt changes of the upper limb of the overturning circulation in the Southern Hemisphere. In fact, internal mixing accounts for 25% of the heat change, and 22% of the salinity change.

  • Tracing pathways in the ocean circulation : A temperature and salinity perspective

    • The ocean circulation plays an essential role in our climate system. It redistributes heat, salt, carbon and other tracers across the globe, making the climate of Earth more moderate. This thesis targets density differences that are driving the ocean circulation. These differences are caused by changes in temperature and salinity. The analysis is based on the usage of Lagrangian trajectories simulated with velocity fields from an Earth System Model. The Lagrangian approach opens up for the possibility to follow specific water paths and water masses. The results herein provide a new insight to specific circulation patterns in the ocean, and which regions that play an important role in controlling temperature and salinity changes.In the first two articles, the Lagrangian divergence is introduced. It shows the geographical distribution of heat and salt changes of a simulated water mass. Using this, we are able to show that the northward flowing water in the Atlantic Ocean cools and freshens in the North Atlantic Subtropical Gyre, the Gulf Stream and the North Atlantic Current. Similarly, we show that the water flowing from the Drake Passage, following the Antarctic Circumpolar Current (ACC) and moving northwards into the Atlantic, Pacific and Indian oceans, transforms from cold and fresh to warm and saline. This warming and salinification are a result of, not only air-sea fluxes, but also interior mixing.In the third study, we show that 70% of the water flowing northwards as part of the Atlantic Meridional Overturning Circulation circuits the North Atlantic Subtropical Gyre at least once before it continues northwards. In the gyre, the water spirals downwards as it gets denser, due to a combination of air-sea fluxes and interior mixing. These results bring a new perspective on the Subtropical Gyre's role to the circulation patterns of the Atlantic Meridional Overturning Circulation.In the last part of this thesis, the circulation in the North Atlantic Ocean is traced into four different pathways. The pathways are visualised geographically together with their change in temperature, salinity and density. With this, we are able to show that the northward flowing water in the Atlantic Ocean exchanges heat and salt with the colder and fresher waters circulating the Subpolar Gyre.

    • The way we understand and treat our horses, like the purpose for which they are bred, is culturally conditioned. The transition between differing social formations involves not only new life circumstances for people and animals, but also changed social modes of relating between them. Certain of the animals? previous functions and types of use come to an end at the same time new ones arise in their place. In this dissertation, the focus is on harness racing, where the meaning and function of the horse over the years have been subject to renegotiation. The purpose is to examine how the trotting horse is made, in relation to the conceptions, expectations and requirements of the sport. The making of the trotting horse is both social and physical; that is, it is manifested in the understanding of the animal as well as in its physicality. This process coincided with the organisation of the sport. Then as now, control was exercised over when, how and which individuals were allowed to reproduce. The work of breeding has fashioned trotting horses that are faster as well as built more for man's specific purposes than their predecessors. Physically and mentally they have been adapted to the racing form and the high speeds. Horses that enter race courses today differ in physiognomy and gait pattern not only from trotting horses of older times, but also, for example, from riding- and race horses. When the purpose of keeping horses changed, new groups of people became associated with the enterprise, such as gamblers, professional trainers and drivers, all of whom in their own ways affect the way the trotting horse is understood and made.

    • Persistent organic pollutants (POPs) are industrial chemicals resistant to degradation and have been shown to have adverse effects on reproductive health in wildlife and humans. Although regulations have reduced their levels, they are still ubiquitously present and pose a global concern. Here, we studied a cohort of 185 women aged 21-43 years with a median of 2 years of infertility who were seeking assisted reproductive technology (ART) treatment at the Carl von Linne Clinic in Uppsala, Sweden. We analyzed the levels of 9 organochlorine pesticides (OCPs), 10 polychlorinated biphenyls (PCBs), 3 polybrominated diphenyl ethers (PBDEs), and 8 perfluoroalkyl substances (PFASs) in the blood and follicular fluid (FF) samples collected during ovum pick-up. Impact of age on chemical transfer from blood to FF was analyzed. Associations of chemicals, both individually and as a mixture, to 10 ART endpoints were investigated using linear, logistic, and weighted quantile sum regression, adjusted for age, body mass index, parity, fatty fish intake and cause of infertility. Out of the 30 chemicals, 20 were detected in more than half of the blood samples and 15 in FF. Chemical transfer from blood to FF increased with age. Chemical groups in blood crossed the blood-follicle barrier at different rates: OCPs > PCBs > PFASs. Hexachlorobenzene, an OCP, was associated with lower anti-Miillerian hormone, clinical pregnancy, and live birth. PCBs and PFASs were associated with higher antral follicle count and ovarian response as measured by ovarian sensitivity index, but also with lower embryo quality. As a mixture, similar findings were seen for the sum of PCBs and PFASs. Our results suggest that age plays a role in the chemical transfer from blood to FF and that exposure to POPs significantly associates with ART outcomes. We strongly encourage further studies to elucidate the underlying mechanisms of reproductive effects of POPs in humans.

    • The folate metabolism enzyme MTHFD2 (methylenetetrahydrofolate dehydrogenase/cyclohydrolase) is consistently overexpressed in cancer but its roles are not fully characterized, and current candidate inhibitors have limited potency for clinical development. In the present study, we demonstrate a role for MTHFD2 in DNA replication and genomic stability in cancer cells, and perform a drug screen to identify potent and selective nanomolar MTHFD2 inhibitors; protein cocrystal structures demonstrated binding to the active site of MTHFD2 and target engagement. MTHFD2 inhibitors reduced replication fork speed and induced replication stress followed by S-phase arrest and apoptosis of acute myeloid leukemia cells in vitro and in vivo, with a therapeutic window spanning four orders of magnitude compared with nontumorigenic cells. Mechanistically, MTHFD2 inhibitors prevented thymidine production leading to misincorporation of uracil into DNA and replication stress. Overall, these results demonstrate a functional link between MTHFD2-dependent cancer metabolism and replication stress that can be exploited therapeutically with this new class of inhibitors. Helleday and colleagues describe a nanomolar MTHFD2 inhibitor that causes replication stress and DNA damage accumulation in cancer cells via thymidine depletion, demonstrating a potential therapeutic strategy in AML tumors in vivo.

    • BACKGROUND: The infection caused by SARS CoV-2 has been postulated to induce a cytokine storm syndrome that results in organ failure and even death in a considerable number of patients. However, the inflammatory response in Corona virus disease-19 (Covid-19) and its potential to cause collateral organ damage has not been fully elucidated to date. This study aims to characterize the acute cytokine response in a cohort of critically ill Covid-19 patients.METHOD: 24 adults with PCR-confirmed Covid-19 were included at time of admission to intensive care a median of eleven days after initial symptoms. Eleven adult patients admitted for elective abdominal surgery with preoperative plasma samples served as controls. All patients were included after informed consent was obtained. 27 cytokines were quantified in plasma. The expression of inflammatory mediators was then related to routine inflammatory markers, SAPS3, SOFA score, organ failure and 30-day mortality.RESULTS: A general increase in cytokine expression was observed in all Covid-19 patients. A strong correlation between respiratory failure and IL-1ra, IL-4, IL-6, IL-8 and IP-10 expression was observed. Acute kidney injury development correlated well with increased levels of IL-1ra, IL-6, IL-8, IL-17a, IP-10 and MCP-1. Generally, the cohort demonstrated weaker correlations between cytokine expression and 30-day mortality out of which IL-8 showed the strongest signal in terms of mortality.CONCLUSION: The present study found that respiratory failure, acute kidney injury and 30-day mortality in critically ill Covid-19 patients are associated with moderate increases of a broad range of inflammatory mediators at time of admission.
