  • The Analgesic Effect of Oxygen in Suspected Acute Myocardial Infarction : A Substudy of the DETO2X-AMI Trial
  • 2018
  • Ingår i: JACC: Cardiovascular Interventions. - : Elsevier BV. - 1936-8798 .- 1876-7605. ; 39, s. 546-546
  • Tidskriftsartikel (refereegranskat)abstract
    • Objectives: In this substudy of the DETO2X-AMI (An Efficacy and Outcome Study of Supplemental Oxygen Treatment in Patients With Suspected Myocardial Infarction) trial, the authors aimed to assess the analgesic effect of moderate-flow oxygen supplementation in patients with suspected acute myocardial infarction (AMI) treated with percutaneous coronary intervention (PCI) and to study the effect of oxygen supplementation on the use of opiates and sedatives during PCI. Background: Routine oxygen in normoxemic patients with AMI does not provide clinical benefit. However, oxygen may relieve ischemic pain. Methods: Patients were randomly allocated to oxygen or ambient air according to the main study protocol. After PCI, peak level of pain during PCI was measured by the Visual Analogue Scale. The total amount of opiates and sedatives was reported. Results: A total of 622 patients were enrolled: 330 in the oxygen group and 292 in the ambient air group. There was no significant difference in peak level of pain (oxygen 4.0 [1.0 to 6.0] vs. air 3.0 [0.6 to 6.0]; p = 0.37), use of opiates (mg) (oxygen 0.0 [0.0 to 3.0] vs. air 0.0 [0.0 to 3.0]; p = 0.31), or use of sedatives between the groups (median [interquartile range]) (oxygen 2.5 [0.0 to 2.5] vs. air 2.5 [0.0 to 2.5]; p = 0.74). Conclusions: In the present study, the authors did not find any analgesic effect of routine oxygen as compared with ambient air, and no differences in the use of sedatives and opiates during PCI. Our results indicate that moderate-flow oxygen supplementation does not relieve pain in normoxemic patients with suspected AMI undergoing treatment with PCI and should thus not be used for this purpose.

  • Mapping and Evaluating Marine Protected Areas and Ecosystem Services: A Transdisciplinary Delphi Forecasting Process Framework
  • 2021
  • Ingår i: Frontiers in Ecology and Evolution. - : Frontiers Media SA. - 2296-701X. ; 9
  • Tidskriftsartikel (refereegranskat)abstract
    • Marine Protected Areas (MPAs) are an important tool for management and conservation and play an increasingly recognised role in societal and human well-being. However, the assessment of MPAs often lacks a simultaneous consideration of ecological and socio-economic outcomes, and this can lead to misconceptions on the effectiveness of MPAs. In this perspective, we present a transdisciplinary approach based on the Delphi method for mapping and evaluating Marine Protected Areas for their ability to protect biodiversity while providing Ecosystem Services (ES) and related human well-being benefits – i.e., the ecosystem outputs from which people benefit. We highlight the need to include the human dimensions of marine protection in such assessments, given that the effectiveness of MPAs over time is conditional on the social, cultural and institutional contexts in which MPAs evolve. Our approach supports Ecosystem-Based Management and highlights the importance of MPAs in achieving restoration, conservation, and sustainable development objectives in relation to EU Directives such as the Marine Strategy Framework Directive (MSFD), the Maritime Spatial Planning Directive (MSPD), and the Common Fisheries Policy (CFP).

  • Internship as a Mean for Integration. A Critical Study
  • 2019
  • Ingår i: Journal of International Migration and Integration. - : Springer Science and Business Media LLC. - 1488-3473 .- 1874-6365. ; 20:2, s. 323-340
  • Tidskriftsartikel (refereegranskat)abstract
    • Creating internships for newly arrived refugees and immigrants is a well-established part of the Swedish national integration program and is seen as a strategy to speed up immigrants’ establishment process. One common belief is that the workplace is expected to give the newly arrived trainee necessary language training, as well as contextual and cultural knowledge. Here, the transferring direction of knowledge is from the Swedish workplace to the receiving newly arrived trainee. However, it is rarely discussed whether the trainee will bring valuable knowledge to the organization, or what the relational aspects of the internship should look like. In this paper, we develop the analysis of internships as a mean for integration in Sweden, by analyzing what trainees and their supervisors experience in terms of knowledge transfer and development. By interviewing trainees (4) and supervisors (5), the aim of this paper is to discuss ideas and practices in an internship program for refugees that The University of Gothenburg started in 2015. The interviews focus on the structural and organizational level, though individual experiences also play a central role. Theoretically, the analysis is interdisciplinary and a cross-cut between organizational theory and cultural studies, especially the work of Sara Ahmed. Our results show that the trainee is seen both as a resource and as someone who represents multiculturalism, hospitality, and the altruism of the university and its employees. The trainee is expected not only to integrate in a productive way, but also to stand out in non-productive areas, such as the social context.

  • Interannual variation and trends in air pollution over Europe due to climate variability during 1958–2001 simulated with a regional CTM coupled to the ERA40 reanalysis
  • 2007
  • Ingår i: Tellus. Series B, Chemical and physical meteorology. - Stockholm : Tellus. - 0280-6509 .- 1600-0889. ; 59, s. 77-98
  • Tidskriftsartikel (refereegranskat)abstract
    • A three-dimensional Chemistry Transport Model was used to study the meteorologically induced interannual variability and trends in deposition of sulphur and nitrogen as well as concentrations of surface ozone (O3), nitrogen dioxide (NO2) and particulate matter (PM) and its constituents over Europe during 1958–2001. The model was coupled to the meteorological reanalysis ERA40, produced at the European Centre for Medium-range Weather Forecasts. Emissions and boundary conditions of chemical compounds and PM were kept constant at present levels. The average European interannual variation, due to meteorological variability, ranges from 3% for O3, 5%for NO2, 9% for PM, 6–9% for dry deposition, to about 20% for wet deposition of sulphur and nitrogen. For the period 1979–2001 the trend in ozone, due to climate variability is increasing in central and southwestern Europe and decreasing in northeastern Europe, the trend in NO2 is approximately opposite. The trend in PM is positive in eastern Europe. There are negative trends in wet deposition in southwestern and central Europe and positive trends in dry deposition overall. A bias in ERA40 precipitation could be partly responsible for the trends. The variation and trends need to be considered when interpreting measurements and designing measurement campaigns.

  • Population exposure and mortality due to regional background PM in Europe – longterm simulations of source-region and shipping contributions
  • 2009
  • Ingår i: Atmospheric Environment. - : Elsevier BV. - 1352-2310 .- 1873-2844. ; 43:22-23, s. 3614-3620
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper presents the contribution to population exposure (PE) of regional background fine primary (PPM2.5) and secondary inorganic (SIA) particulate matter and its impact on mortality in Europe during 1997–2003 calculated with a chemistry transport model. Contributions to concentrations and PE due to emissions from shipping, Western (WEU), Eastern (EEU), and Northern Europe are compared. WEU contributes about 40% to both PPM2.5 and SIA concentrations, whereas the EEU contribution to PPM2.5 is much higher (43% of total PPM2.5) than to SIA (29% of total SIA). The population weighted average concentration (PWC) of PPM2.5 is a factor of 2.3 higher than average (non-weighted) concentrations, whereas for SIA the PWC is only a factor 1.6 higher. This is due to PPM2.5 concentrations having larger gradients and being relatively high over densely populated areas, whereas SIA is formed outside populated areas. WEU emissions contribute relatively more than EEU to PWC and mortality due to both PPM2.5 and SIA in Europe. The number of premature deaths in Europe is estimated to 301 000 per year due to PPM2.5 exposure and 245 000 due to SIA, despite 3.3 times higher average SIA concentrations. This is due to population weighting and assumed (and uncertain) higher relative risk of mortality for PPM2.5 components (2.8 times higher RR for PPM2.5). This study indicates that it might be more efficient, for the health of the European population, to decrease primary PM emissions (especially in WEU) than to decrease precursors of SIA, but more knowledge on the toxicity of different PM constituents is needed before firm conclusions can be drawn.

  • Occurrence of methicillin-resistant Staphylococci in surgically treated dogs and the environment in a Swedish animal hospital
  • 2012
  • Ingår i: Journal of Small Animal Practice. - : Wiley. - 0022-4510 .- 1748-5827. ; 53, s. 404-410
  • Tidskriftsartikel (refereegranskat)abstract
    • ObjectivesTo investigate whether hospitalised dogs treated surgically may become culture positive for methicillin-resistant Staphylococcus pseudintermedius or methicillin-resistant Staphylococcus aureus.MethodsSurgically treated dogs (n=45) were sampled for methicillin-resistant Staphylococcus pseudintermedius or methicillin-resistant Staphylococcus aureus on admission, before and after surgery and at the time of removal of surgical stitches. The hospital environment (n=57), including healthy dogs in the veterinary hospital environment (n=34), were sampled for methicillin-resistant Staphylococcus pseudintermedius or methicillin-resistant Staphylococcus aureus. Genetic variations among methicillin-resistant Staphylococcus pseudintermedius or methicillin-resistant Staphylococcus aureus isolates were identified through detection of restriction fragment polymorphisms.ResultsNo dogs developed a wound infection due to methicillin-resistant Staphylococcus pseudintermedius or methicillin-resistant Staphylococcus aureus. However, there was a significant increase in the number of dogs carrying methicillin-resistant Staphylococcus pseudintermedius after hospitalisation compared to admission (P<0.001). No methicillin-resistant Staphylococcus aureus was isolated from dogs, but was present in the environment. Methicillin-resistant Staphylococcus pseudintermedius isolates were recovered from environmental surfaces and hospitalised animals, but not from healthy dogs. Methicillin-resistant Staphylococcus pseudintermedius isolates representing nine different restriction endonuclease digestion patterns were found, with two of these occurring in both the environment and on dogs.Clinical SignificanceDogs may contract methicillin-resistant Staphylococcus pseudintermedius in association with surgery and hospitalisation. Resistant bacteria may be transmitted between dogs, staff and the environment. Dogs colonised with methicillin-resistant Staphylococcus pseudintermedius may be a source for hospital- and community-acquired infections.

  • Trends of inorganic and organic aerosols and precursor gases in Europe: Insights from the EURODELTA multi-model experiment over the 1990-2010 period
  • 2019
  • Ingår i: Geoscientific Model Development. - : Copernicus GmbH. - 1991-959X .- 1991-9603. ; 12:12, s. 4923-4954
  • Tidskriftsartikel (refereegranskat)abstract
    • In the framework of the EURODELTA-Trends (EDT) modeling experiment, several chemical transport models (CTMs) were applied for the 1990-2010 period to investigate air quality changes in Europe as well as the capability of the models to reproduce observed long-term air quality trends. Five CTMs have provided modeled air quality data for 21 continuous years in Europe using emission scenarios prepared by the International Institute for Applied Systems Analysis/Greenhouse Gas - Air Pollution Interactions and Synergies (IIASA/GAINS) and corresponding year-by-year meteorology derived from ERA-Interim global reanalysis. For this study, long-term observations of particle sulfate (SO2 4-), total nitrate (TNO3), total ammonium (TNHx) as well as sulfur dioxide (SO2) and nitrogen dioxide (NO2) for multiple sites in Europe were used to evaluate the model results. The trend analysis was performed for the full 21 years (referred to as PT) but also for two 11-year subperiods: 1990-2000 (referred to as P1) and 2000-2010 (referred to as P2). The experiment revealed that the models were able to reproduce the faster decline in observed SO2 concentrations during the first decade, i.e., 1990-2000, with a 64%-76% mean relative reduction in SO2 concentrations indicated by the EDT experiment (range of all the models) versus an 82% mean relative reduction in observed concentrations. During the second decade (P2), the models estimated a mean relative reduction in SO2 concentrations of about 34%-54%, which was also in line with that observed (47%). Comparisons of observed and modeled NO2 trends revealed a mean relative decrease of 25% and between 19% and 23% (range of all the models) during the P1 period, and 12% and between 22% and 26% (range of all the models) during the P2 period, respectively. Comparisons of observed and modeled trends in SO4 2- concentrations during the P1 period indicated that the models were able to reproduce the observed trends at most of the sites, with a 42%-54% mean relative reduction indicated by the EDT experiment (range of all models) versus a 57% mean relative reduction in observed concentrations and with good performance also during the P2 and PT periods, even though all the models overpredicted the number of statistically significant decreasing trends during the P2 period. Moreover, especially during the P1 period, both modeled and observational data indicated smaller reductions in SO42- concentrations compared with their gas-phase precursor (i.e., SO2), which could be mainly attributed to increased oxidant levels and pH-dependent cloud chemistry. An analysis of the trends in TNO3 concentrations indicated a 28%-39% and 29% mean relative reduction in TNO3 concentrations for the full period for model data (range of all the models) and observations, respectively. Further analysis of the trends in modeled HNO3 and particle nitrate (NO-3 ) concentrations revealed that the relative reduction in HNO3 was larger than that for NO-3 during the P1 period, which was mainly attributed to an increased availability of "free ammonia". By contrast, trends in modeled HNO3 and NO-3 concentrations were more comparable during the P2 period. Also, trends of TNHx concentrations were, in general, underpredicted by all models, with worse performance for the P1 period than for P2. Trends in modeled anthropogenic and biogenic secondary organic aerosol (ASOA and BSOA) concentrations together with the trends in available emissions of biogenic volatile organic compounds (BVOCs) were also investigated. A strong decrease in ASOA was indicated by all the models, following the reduction in anthropogenic non-methane VOC (NMVOC) precursors. Biogenic emission data provided by the modeling teams indicated a few areas with statistically significant increase in isoprene emissions and monoterpene emissions during the 1990-2010 period over Fennoscandia and eastern European regions (i.e., around 14 %-27 %), which was mainly attributed to the increase of surface temperature. However, the modeled BSOA concentrations did not linearly follow the increase in biogenic emissions. Finally, a comprehensive evaluation against positive matrix factorization (PMF) data, available during the second period (P2) at various European sites, revealed a systematic underestimation of the modeled SOA fractions of a factor of 3 to 11, on average, most likely because of missing SOA precursors and formation pathways, with reduced biases for the models that accounted for chemical aging of semi-volatile SOA components in the atmosphere.

  • EURODELTA-Trends, a multi-model experiment of air quality hindcast in Europe over 1990–2010
  • 2017
  • Ingår i: Geoscientific Model Development. - : Copernicus GmbH. - 1991-959X .- 1991-9603. ; 10:9, s. 3255-3276
  • Tidskriftsartikel (refereegranskat)abstract
    • The EURODELTA-Trends multi-model chemistry-transport experiment has been designed to facilitate a better understanding of the evolution of air pollution and its drivers for the period 1990–2010 in Europe. The main objective of the experiment is to assess the efficiency of air pollutant emissions mitigation measures in improving regional-scale air quality.  The present paper formulates the main scientific questions and policy issues being addressed by the EURODELTA-Trends modelling experiment with an emphasis on how the design and technical features of the modelling experiment answer these questions.  The experiment is designed in three tiers, with increasing degrees of computational demand in order to facilitate the participation of as many modelling teams as possible. The basic experiment consists of simulations for the years 1990, 2000, and 2010. Sensitivity analysis for the same three years using various combinations of (i) anthropogenic emissions, (ii) chemical boundary conditions, and (iii) meteorology complements it. The most demanding tier consists of two complete time series from 1990 to 2010, simulated using either time-varying emissions for corresponding years or constant emissions. Eight chemistry-transport models have contributed with calculation results to at least one experiment tier, and five models have – to date – completed the full set of simulations (and 21-year trend calculations have been performed by four models). The modelling results are publicly available for further use by the scientific community.  The main expected outcomes are (i) an evaluation of the models' performances for the three reference years, (ii) an evaluation of the skill of the models in capturing observed air pollution trends for the 1990–2010 time period, (iii) attribution analyses of the respective role of driving factors (e.g. emissions, boundary conditions, meteorology), (iv) a dataset based on a multi-model approach, to provide more robust model results for use in impact studies related to human health, ecosystem, and radiative forcing.

  • Antibodies to SARS-CoV-2 and risk of past or future sick leave
  • 2021
  • Ingår i: Scientific Reports. - : Springer Nature. - 2045-2322. ; 11:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The extent that antibodies to SARS-CoV-2 may protect against future virus-associated disease is unknown. We invited all employees (n=15,300) at work at the Karolinska University Hospital, Stockholm, Sweden to participate in a study examining SARS-Cov-2 antibodies in relation to registered sick leave. For consenting 12,928 healthy hospital employees antibodies to SARS-CoV-2 could be determined and compared to participant sick leave records. Subjects with viral serum antibodies were not at excess risk for future sick leave (adjusted odds ratio (OR) controlling for age and sex: 0.85 [95% confidence interval (CI) (0.85 (0.43-1.68)]. By contrast, subjects with antibodies had an excess risk for sick leave in the weeks prior to testing [adjusted OR in multivariate analysis: 3.34 (2.98-3.74)]. Thus, presence of viral antibodies marks past disease and protection against excess risk of future disease. Knowledge of whether exposed subjects have had disease in the past or are at risk for future disease is essential for planning of control measures.Trial registration: First registered on 02/06/20, ClinicalTrials.gov NCT04411576.

  • A reduced level of consciousness affects non-conscious processes
  • 2021
  • Ingår i: NeuroImage. - : Elsevier. - 1053-8119 .- 1095-9572. ; 244
  • Tidskriftsartikel (refereegranskat)abstract
    • Being conscious is a profound aspect of human existence, and understanding its function and its inception is considered one of the truly grand scientific challenges. However, the nature of consciousness remains enigmatic, to a large part because “being conscious” can refer to both the content (phenomenology) and the level (arousal) of consciousness, and how these different aspects are related remains unclear. To empirically assess the relation between level and content of consciousness, we manipulated these two aspects by presenting stimuli consciously or non-consciously and by using Propofol sedation, while brain activity was measured using fMRI. We observed that sedation affected both conscious and non-conscious processes but at different hierarchical levels; while conscious processing was altered in higher-order regions (the intraparietal sulcus) and spared sensory areas, the opposite effect was observed for non-conscious processing. The observation that Propofol affected non-conscious processing calls for a reconsideration of what kind of information one can gain on “consciousness” from recording neural responses to sedation without considering both (content) conscious and (content) non-conscious processing.

  • Secretion and uptake of α-synuclein via extracellular vesicles in cultured cells
  • 2018
  • Ingår i: Cellular and molecular neurobiology. - : Springer Science and Business Media LLC. - 0272-4340 .- 1573-6830. ; 38:8, s. 1539-1550
  • Tidskriftsartikel (refereegranskat)abstract
    • In Parkinson’s disease and other Lewy body disorders, the propagation of pathology has been accredited to the spreading of extracellular α-synuclein (α-syn). Although the pathogenic mechanisms are not fully understood, cell-to-cell transfer of α-syn via exosomes and other extracellular vesicles (EVs) has been reported. Here, we investigated whether altered molecular properties of α-syn can influence the distribution and secretion of α-syn in human neuroblastoma cells. Different α-syn variants, including α-syn:hemi-Venus and disease-causing mutants, were overexpressed and EVs were isolated from the conditioned medium. Of the secreted α-syn, 0.1–2% was associated with vesicles. The major part of EV α-syn was attached to the outer membrane of vesicles, whereas a smaller fraction was found in their lumen. For α-syn expressed with N-terminal hemi-Venus, the relative levels associated with EVs were higher than for WT α-syn. Moreover, such EV-associated α-syn:hemi-Venus species were internalized in recipient cells to a higher degree than the corresponding free-floating forms. Among the disease-causing mutants, A53T α-syn displayed an increased association with EVs. Taken together, our data suggest that α-syn species with presumably lost physiological functions or altered aggregation properties may shift the cellular processing towards vesicular secretion. Our findings thus lend further support to the tenet that EVs can mediate spreading of harmful α-syn species and thereby contribute to the pathology in α-synucleinopathies.

  • Systematic evaluation of SARS-CoV-2 antigens enables a highly specific and sensitive multiplex serological COVID-19 assay
  • 2021
  • Ingår i: Clinical & Translational Immunology. - : Wiley. - 2050-0068. ; 10:7
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective. The COVID-19 pandemic poses an immense need for accurate, sensitive and high-throughput clinical tests, and serological assays are needed for both overarching epidemiological studies and evaluating vaccines. Here, we present the development and validation of a high-throughput multiplex bead-based serological assay. Methods. More than 100 representations of SARS-CoV-2 proteins were included for initial evaluation, including antigens produced in bacterial and mammalian hosts as well as synthetic peptides. The five best-performing antigens, three representing the spike glycoprotein and two representing the nucleocapsid protein, were further evaluated for detection of IgG antibodies in samples from 331 COVID-19 patients and convalescents, and in 2090 negative controls sampled before 2020. Results. Three antigens were finally selected, represented by a soluble trimeric form and the S1-domain of the spike glycoprotein as well as by the C-terminal domain of the nucleocapsid. The sensitivity for these three antigens individually was found to be 99.7%, 99.1% and 99.7%, and the specificity was found to be 98.1%, 98.7% and 95.7%. The best assay performance was although achieved when utilising two antigens in combination, enabling a sensitivity of up to 99.7% combined with a specificity of 100%. Requiring any two of the three antigens resulted in a sensitivity of 99.7% and a specificity of 99.4%. Conclusion. These observations demonstrate that a serological test based on a combination of several SARS-CoV-2 antigens enables a highly specific and sensitive multiplex serological COVID-19 assay.

    • The effect of insulin on blood pressure (BP) is debated, and an involvement of an activated renin-angiotensin aldosterone system (RAAS) has been suggested. We studied the effect of chronic insulin infusion on telemetry BP and assessed sympathetic activity and dependence of the RAAS. Female Sprague-Dawley rats received insulin (2 units/day, INS group, n = 12) or insulin combined with losartan (30 mg.kg(-1).day(-1), INS+LOS group, n = 10), the angiotensin II receptor antagonist, for 6 wk. Losartan-treated (LOS group, n = 10) and untreated rats served as controls (n = 11). We used telemetry to measure BP and heart rate (HR), and acute ganglion blockade and air-jet stress to investigate possible control of BP by the sympathetic nervous system. In addition, we used myograph technique to study vascular function ex vivo. The INS and INS+LOS groups developed euglycemic hyperinsulinemia. Insulin did not affect BP but increased HR (27 beats/min on average). Ganglion blockade reduced mean arterial pressure (MAP) similarly in all groups. Air-jet stress did not increase sympathetic reactivity but rather revealed possible blunting of the stress response in hyperinsulinemia. Chronic losartan markedly reduced 24-h-MAP in the INS+LOS group (-38 +/- 1 mmHg P < 0.001) compared with the LOS group (-18 +/- 1 mmHg, P

  • 2019

  • Konferensbidrag (refereegranskat)abstract
    • We present an interactive visual environment for linked analysis of brain imaging and clinical measurements. The environment is developed in an iterative participatory design process involving neuroscientists investigating the causes of brain-related complex diseases. The hypotheses formation process about correlations between active brain regions and physiological or psychological factors in studies with hundreds of subjects is a central part of the investigation. Observing the reasoning patterns during hypotheses formation, we concluded that while existing tools provide powerful analysis options, they lack effective interactive exploration, thus limiting the scientific scope and preventing extraction of knowledge from available data.Based on these observations, we designed methods that support neuroscientists by integrating their existing statistical analysis of multivariate subject data with interactive visual explorationto enable them to better understand differences between patient groups and the complex bidirectional interplay between clinical measurement and the brain. These exploration concepts enable neuroscientists, for the first time during their investigations, to interactively move between and reason about questions such as ‘which clinical measurements are correlated with a specific brain region?’ or ‘are there differences in brain activity between depressed young and old subjects?’. The environment uses parallel coordinates for effective overview and selection of subject groups, Welch's t-test to filter out brain regions with statistically significant differences, and multiple visualizations of Pearson correlations between brain regions and clinical parameters to facilitate correlation analysis. A qualitative user study was performed with three neuroscientists from different domains. The study shows that the developed environment supports simultaneous analysis of more parameters, provides rapid pathways to insights, and is an effective support tool for hypothesis formation.

    • We present an application, and its development process, for interactive visual analysis of brain imaging data and clinical measurements. The application targets neuroscientists interested in understanding the correlations between active brain regions and physiological or psychological factors. The application has been developed in a participatory design process and has subsequently been released as the free software VisualNeuro. From initial observations of the neuroscientists workflow, we concluded that while existing tools provide powerful analysis options, they lack effective interactive exploration requiring the use of many tools side by side. Consequently, our application has been designed to simplify the workflow combining statistical analysis with interactive visual exploration. The resulting environment comprises parallel coordinates for effective overview and selection, Welchs t-test to filter out brain regions with statistically significant differences and multiple visualizations for comparison between brain regions and clinical parameters. These exploration concepts enable neuroscientists to interactively explore the complex bidirectional interplay between clinical and brain measurements and easily compare different patient groups. A qualitative user study has been performed with three neuroscientists from different domains. The study shows that the developed environment supports simultaneous analysis of more parameters, provides rapid pathways to insights and is an effective tool for hypothesis formation.

  • The vulnerability of northern European vegetation to ozone damage in a changing climate An assessment based on current knowledge
  • 2021
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • The potential vulnerability of vegetation at northern latitudes to ozone damage was assessed based on current knowledge with regard to air ozone concentrations and leaf ozone uptake as well as to plant traits affecting ozone tolerance. The focus was on the northern European arctic, alpine and northern boreal vegetation zones, with a special focus on high-altitude vegetation. In particular, we analysed if there are increasing risks for ozone impacts on northern vegetation due to high spring ozone concentrations in relation to climate change induced shifts such as e.g. an earlier start of the growing season. The ozone concentrations in these regions are characterized by the influence of a combination of conditions caused by high latitudes and high altitudes. Ozone concentrations increase with altitude and the difference in ozone concentrations between day and night are smaller at high-altitude and high-latitude sites. Summer periods with long daylight conditions potentially promote the leaf ozone uptake through the open stomata. The aims of this report were: • To assess the current state of knowledge regarding the potential vulnerability of far northern vegetation to ozone damage, today and in the future • To provide advice for policy implications regarding necessary ozone precursor emission abatement • To provide advice for future research and monitoring of ozone impacts on the vegetation at northern latitudes Ongoing environmental changes affecting far northern latitude ecosystems were reviewed. Current and novel methods were described for how to estimate the time of year during which the ozone exposure for vegetation should be accumulated. Time trends for ozone concentrations at northern latitudes were analysed. Ozone episodes with high concentrations at far northern latitudes were described. Source attributions of northern ozone concentrations were analysed. Environmental conditions at far northern latitudes that might be important for ozone damage were evaluated. Plant traits that can influence the ozone vulnerability were discussed. Current experimental results for ozone injury on northern plant species were evaluated. Future scenarios for ozone impact on northern vegetation were discussed. Some important results from the analyses are described below. At high altitudes and high latitudes, the ozone concentrations are relatively similar during day- and night-time. Furthermore, at high latitudes, the long daylight duration during the summer has the potential to increase the duration of the daily period with plant gas exchange and leaf ozone uptake. Therefore, the absorption of ozone through the stomata may potentially be higher at northern latitudes. However, measurements of light intensity and quality at northern sites in combination with a simple calculation example illustrated that this probably was not the case, since the potential added ozone uptake in the early morning and late evening at northern sites may be cancelled out by a lower ozone uptake in the middle of the day, as compared to southern sites. Both data on budburst and data on ecosystem CO2 exchange as well as meteorological observations show that there has been a development towards an earlier start of the growing season during the year, with approximately 0.5 – 1 day per year. Thus, there is clear evidence for an earlier start of the growing season, which is likely to continue. However, the timing of the spring ozone maximum is also shifted towards earlier in the year. There is presently no evidence for an increasing overlap between the growing season and the ozone peak. Despite this, there is a potential for increased ozone uptake to vegetation in spring due to the earlier growing start of vegetation and increased uptake of ozone to vegetation in May. The impact of this on the accumulated phytotoxic ozone dose for northern vegetation needs to be investigated further. The overall conclusions about the present and near future ozone vulnerability of northern vegetation were: • There remain uncertainties regarding to what extent northern vegetation is affected by ozone exposure. • According to current knowledge, we could not find evidence that expected changes in ozone concentrations and climate would make the northern arctic, alpine and subalpine vegetation substantially more vulnerable to ozone than other types of European vegetation. • The risk of significant and lasting negative impact of the current exposure to ozone on northern boreal forests is most likely not greater than for boreonemoral and nemoral forests in southern Fennoscandia. • However, peak ozone concentrations occurring in spring and early summer may affect vegetation at northern latitudes in Fennoscandia since the start of the growing season in the future may occur earlier during the year. The policy implications that can be derived from these conclusions were: • The current state of knowledge implies that ecosystems in the far north are not more susceptible to ozone than vegetation in other parts of Europe. Hence, we cannot advocate for a stronger reduction of ozone precursors emissions based exclusively on the ozone sensitivity of vegetation in the far north. • Policies designed to reduce emissions of ozone precursors to protect vegetation in other parts of Europe as well as in the entire northern hemisphere are likely to suffice to protect vegetation in northern Fennoscandia. There are important remaining knowledge gaps. Our conclusions are based on important, but limited observations. Experimental evidence from investigations specifically designed to study ozone sensitivity of high-altitude vegetation in northern Europe are to a large extent lacking. It is recommended that further experimental research is undertaken to directly compare the ozone sensitivity of plants of high-latitude/high-altitude origin with that of plants (species, genotypes) representative of regions of the southern part of the Nordic region. This research should include the characteristics of the high-latitude climate and other conditions. A specific research question is if the new ozone critical levels for European vegetation based on PODYSPEC (Mapping Manual, 2017) are correct, both regarding calculation methodology as well as impact assessments? In particular, there is a lack of information about the degree of stomata closure during nights in high-latitude area plants. This is important for the modelling of ozone uptake (dry deposition) in these areas and requires coordinated measurement campaigns in close cooperation with modelers. Further research questions may be related to the future development of the northern regions – e.g. oil and gas extraction including flaring, shipping, more tourism and climate change – how will that affect the ozone exposure of in the northern vegetation? Do future ozone precursor emission scenarios describe this correctly? Will warm and dry summers like 2018 become more frequent in connection with climate change, and how will this affect ozone impacts on vegetation? There are currently very few, long term ozone monitoring stations in the arctic and alpine vegetation zones, in particular at high altitudes. Given the expected increase in anthropogenic activities in these areas in combination with climate change, it is strongly recommended to increase the number of high-altitude ozone monitoring sites in these regions.

  • Intake of Dairy Products as a Risk Factor for Rheumatoid Arthritis; A Nested Case-Control Study
  • 2022
  • Ingår i: Arthritis & Rheumatology. - 2326-5205. ; 74:Suppl. 9
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • Background/Purpose: There has been increasing interest in diet as a factor that may contribute to the development of rheumatoid arthritis (RA). There is limited and somewhat contradictory information on the impact of dairy products in this context. The purpose of this study was to investigate the relation between intake of various dairy products and the risk of RA in a nested case-control study. Methods: Participants in a population-based survey conducted in 1991-1996 who were subsequently diagnosed with RA (from inclusion until December 2016) were identified through register linkage and validated in a structured review of case records. Four controls for each validated case, matched for sex, year of birth, and year of inclusion, were selected from the study cohort. The controls were alive and free of RA when the index person was diagnosed with RA. At inclusion in the survey, diet was assessed using a modified diet history method, consisting of a seven-day food record, a food questionnaire, and a structured interview. Reported intakes of dairy products were divided into groups based on quartiles, with the lowest quartile set as the reference in all analyses. Based on conditional logistical regression, including adjustments for total energy intake and for potential confounders that have been associated with diet and RA (i.e. current smoking, physical activity and alcohol intake), odds ratios (ORs) for RA were estimated, with 95% confidence intervals (CI). Potential misreporters of total energy intake were excluded. Assessed types of dairy products included regular (non-fermented) milk, fermented milk, cream, cheese, and butter. Results: There were 305 incident cases of RA (76 % females, 67 % anti-citrullinated protein antibody and/or rheumatoid factor positive, mean age 68.9 years at onset and mean duration of 12 years from screening to RA diagnosis). The group with highest intake of regular milk ( >398 g/day) had a significantly increased risk of RA (multi-adjusted OR 1.86; 95% CI 1.08-3.22). High intake of cheese with >11% fat ( >56 g/day) was inversely associated with risk of RA (adjusted OR 0.53; 95% CI 0.31-0.92) and a trend of lower risk across quartiles of cheese intake was observed. Intake of cream, fermented milk or butter did not have a significant impact on the risk of developing RA. Associations for intakes of cheese and regular milk with the risk of RA remained significant in multivariable analysis, including both exposures (Table 1).Conclusion: High intake of regular milk appears to increase the risk of RA, whereas high intake of cheese may reduce the risk. Potential explanations for these patterns include differential effects of dairy product depending on the extent of processing and fermentation, that may affect the gut microbiota and modulate the risk of developing RA.

    • Sustainability is aimed at addressing environmental and socio-economic issues of this generation and future ones. Sustainability requires the engagement of stakeholders, as highlighted by Agenda 21 where stakeholder engagement and participation in economic, environmental and social change as one of its most important themes. The purchasing power of public-sector organizations can increase considerable the demand for sustainable products and services can be promoted, set a trend for other organizations and enlarge the market for sustainable products or services. Collaboration is a sine qua non of sustainable public procurement, since it harvests its benefits from differences in perspectives, knowledge and approaches, solving problems while at the same time offering benefits to all those involved in the process. Most sustainable public procurement discussion have focused on the interactions between procurer and suppliers. The triple helix helps to explain to the relations between university-industry-government. It also represents social innovation. The quadruple helix model is an extension of the triple helix, where the fourth helix of civil society is added. The quintuple helix model is based on the earlier helix models but are also take natural environments of society into account. This includes stakeholders such as local communities, employees, shareholders, business partners, suppliers, customers, public authorities and NGOs. The aim of this chapter is to contribute to SPP by proposing to move from engaging only supplier and procurer to a collaborative approach of the quadruple helix. The chapter is empirically based on a function procurement project (of lightening) in Bollnäs, Sweden, focusing foremost on economic sustainability and aiming to develop a quadruple helix model of public procurement. The Bollnäs municipality had a need for changing old roof and lightning and makes the schools and preschools local more flexible for different activities and individual needs. The procurement was done in a collaborative quintuple helix process and focusing on innovation within a holistic approach. The procurement was designed to achieve an effect on flexibility/modularity, sustainability in environment aspect and increased knowledge. The smallest company/supplier was able to win the contact because the process focused on new solutions/innovation. To work in collaborative quintuple helix process give a higher effect and outcome than just the contract. The stakeholders who participate in the process increased their knowledge and changed their mind set to a more holistic point of view, sustainable thinking. © 2020 selection and editorial matter, Marta Andhov, Roberto Caranta and Anja Wiesbrock; individual chapters, the contributors. • During 2016 to 2018 the research node “Science, validation, partial perspectives: Knowledge production beyond the norms,” create spaces for transdisciplinary meetings at Uppsala university. In spring of 2017, the node arranged a journey to California in the USA, for MA candidates, PhD candidates, lecturers, and researchers. The aim was to meet with academics, activists, and artists to discuss the conditions for knowledge production in relation to the current political situation, in which ideas of “post truth” and “alternative facts” have surfaced. We visited the Scandinavian Studies and the Gender and Women’s Studies departments at UC Berkeley, and the Science and Justice Research Center at UC Santa Cruz. This multilingual essay, written in the months following the trip, is a collective, rhizomatic reflection over the relationship between narratives, cultural identities and truths; privilege and politics; language and reality; art and science; potential and risk in boundarycrossing encounters.

  • A multi-model comparison of meteorological drivers of surface ozone over Europe

  • Ingår i: Atmospheric Chemistry and Physics. - : Copernicus GmbH. - 1680-7316 .- 1680-7324. ; 18:16, s. 12269-12288
  • Tidskriftsartikel (refereegranskat)abstract
    • The implementation of European emission abatement strategies has led to a significant reduction in the emissions of ozone precursors during the last decade. Ground-level ozone is also influenced by meteorological factors such as temperature, which exhibit interannual variability and are expected to change in the future. The impacts of climate change on air quality are usually investigated through air-quality models that simulate interactions between emissions, meteorology and chemistry. Within a multi-model assessment, this study aims to better understand how air-quality models represent the relationship between meteorological variables and surface ozone concentrations over Europe. A multiple linear regression (MLR) approach is applied to observed and modelled time series across 10 European regions in springtime and summertime for the period of 2000-2010 for both models and observations. Overall, the air-quality models are in better agreement with observations in summertime than in springtime and particularly in certain regions, such as France, central Europe or eastern Europe, where local meteorological variables show a strong influence on surface ozone concentrations. Larger discrepancies are found for the southern regions, such as the Balkans, the Iberian Peninsula and the Mediterranean basin, especially in springtime. We show that the air-quality models do not properly reproduce the sensitivity of surface ozone to some of the main meteorological drivers, such as maximum temperature, relative humidity and surface solar radiation. Specifically, all air-quality models show more limitations in capturing the strength of the ozone-relative-humidity relationship detected in the observed time series in most of the regions, for both seasons. Here, we speculate that dry-deposition schemes in the air-quality models might play an essential role in capturing this relationship. We further quantify the relationship between ozone and maximum temperature (m(o3-T), climate penalty) in observations and air-quality models. In summertime, most of the air-quality models are able to reproduce the observed climate penalty reasonably well in certain regions such as France, central Europe and northern Italy. However, larger discrepancies are found in springtime, where air-quality models tend to overestimate the magnitude of the observed climate penalty.

    • Ten cats were treated for distal femoral Salter-Harris fractures types I and II using a stainless steel wire in a figure-of-eight configuration. Healing was uneventful in 8 of the cats. Early fixation failure occurred in one cat, which required revisional surgery. In one cat, lameness recurred after fracture healing, which was related to breakage of the wire. The lameness was resolved after removing the wire. At the long-term follow-up, 1 out of 6 cats was lame. This report indicates that the figure-of-eight stainless steel wire technique is an alternative method for the repair of distal femoral Salter-Harris fractures types I and II in cats.

  • An evaluation of European nitrogen and sulfur wet deposition and their trends estimated by six chemistry transport models for the period 1990–2010
  • 2019
  • Ingår i: Atmospheric Chemistry and Physics. - : Copernicus GmbH. - 1680-7316 .- 1680-7324. ; 19:1, s. 379-405
  • Tidskriftsartikel (refereegranskat)abstract
    • The wet deposition of nitrogen and sulfur in Europe for the period 1990–2010 was estimated by six atmospheric chemistry transport models (CHIMERE, CMAQ, EMEP MSC-W, LOTOS-EUROS, MATCH and MINNI) within the framework of the EURODELTA-Trends model intercomparison. The simulated wet deposition and its trends for two 11-year periods (1990–2000 and 2000–2010) were evaluated using data from observations from the EMEP European monitoring network. For annual wet deposition of oxidised nitrogen (WNOx), model bias was within 30 % of the average of the observations for most models. There was a tendency for most models to underestimate annual wet deposition of reduced nitrogen (WNHx), although the model bias was within 40 % of the average of the observations. Model bias for WNHx was inversely correlated with model bias for atmospheric concentrations of NH3 + NH4+, suggesting that an underestimation of wet deposition partially contributed to an overestimation of atmospheric concentrations. Model bias was also within about 40 % of the average of the observations for the annual wet deposition of sulfur (WSOx) for most models. Decreasing trends in WNOx were observed at most sites for both 11-year periods, with larger trends, on average, for the second period. The models also estimated predominantly decreasing trends at the monitoring sites and all but one of the models estimated larger trends, on average, for the second period. Decreasing trends were also observed at most sites for WNHx, although larger trends, on average, were observed for the first period. This pattern was not reproduced by the models, which estimated smaller decreasing trends, on average, than those observed or even small increasing trends. The largest observed trends were for WSOx, with decreasing trends at more than 80 % of the sites. On average, the observed trends were larger for the first period. All models were able to reproduce this pattern, although some models underestimated the trends (by up to a factor of 4) and others overestimated them (by up to 40 %), on average. These biases in modelled trends were directly related to the tendency of the models to under- or overestimate annual wet deposition and were smaller for the relative trends (expressed as % yr−1 relative to the deposition at the start of the period). The fact that model biases were fairly constant throughout the time series makes it possible to improve the predictions of wet deposition for future scenarios by adjusting the model estimates using a bias correction calculated from past observations. An analysis of the contributions of various factors to the modelled trends suggests that the predominantly decreasing trends in wet deposition are mostly due to reductions in emissions of the precursors NOx, NH3 and SOx. However, changes in meteorology (e.g. precipitation) and other (non-linear) interactions partially offset the decreasing trends due to emission reductions during the first period but not the second. This suggests that the emission reduction measures had a relatively larger effect on wet deposition during the second period, at least for the sites with observations.

  • Training and support for the role of facilitator in implementation of innovations in health and community care : a scoping review protocol
  • 2023

  • Forskningsöversikt (refereegranskat)abstract
    • BackgroundImplementing and sustaining innovations in clinical practice, such as evidence-based practices, programmes, and policies, is frequently described as challenging. Facilitation as a strategy for supporting implementation requires a facilitator, i.e. an individual with a designated role to support the implementation process. A growing number of studies report that facilitation can help tackle the challenges in implementation efforts. To optimise the potential contribution of facilitation as a strategy to improve the implementation of new practices, there is a need to enhance understanding about what training and support is required for individuals in the facilitator role. The objective of this scoping review is to map how facilitators have been trained for, and supported in, the facilitator role in implementation studies in health and community care. Specifically, the review aims to examine what is reported on training and support of facilitators in terms of learning outcomes, content, dose, mode of delivery, learning activities, and qualifications of the trainers and how the facilitators perceive training and support.MethodsThis scoping review will follow the guidance of the Joanna Briggs Institute and the PRISMA Extension for Scoping Review checklist. We will include articles in which (a) facilitation is deployed as an implementation strategy, with identified facilitator roles targeting staff and managers, to support the implementation of specified innovations in health or community care, and (b) training and/or support of facilitators is reported. We will exclude articles where facilitation is directed to education or training in specific clinical procedures or if facilitation supports the implementation of general quality improvement systems. All types of peer-reviewed studies and study protocols published in English will be included. A systematic search will be performed in MEDLINE (Ovid), Embase (embase.com), Web of Science Core Collection, and CINAHL (Ebsco).DiscussionThe proposed scoping review will provide a systematic mapping of the literature on the training and support of implementation facilitators and contribute useful knowledge within the field of implementation science to inform future facilitation initiatives.

  • Cardiac concentric remodelling induced by non-aromatizable (dihydro-)testosterone is antagonized by oestradiol in ovariectomized rats
  • 2006
  • Ingår i: J Endocrinol. ; 189:3, s. 485-91
  • Tidskriftsartikel (refereegranskat)abstract
    • Previous studies on the cardiovascular effects of androgens in females, most of them using testosterone treatment, have yielded conflicting results. Testosterone is metabolized into oestradiol (E2) and dihydrotestosterone (DHT) within cardiovascular tissues. The aim of the present study was to explore the cardiovascular effects exerted by E2 and the non-aromatizable androgen DHT and to study possible interactions between these in female rats. Ovariectomized rats were treated with DHT, E2, or DHT+E2 for 6 weeks. DHT increased left-ventricular posterior wall thickness, assessed by echocardiography, whereas left-ventricular dimension, as well as total heart weight and calculated left-ventricular mass, were unchanged. DHT also increased the levels of insulin-like growth factor-I mRNA in the left ventricle. E2 abolished the effect of DHT on left-ventricular remodelling and insulin-like growth factor-I mRNA when the two treatments were given in combination. E2 also reduced androgen receptor mRNA levels in the heart. Neither E2 nor DHT changed blood pressure measured by telemetry. In conclusion, treatment with the endogenous non-aromatizable androgen DHT causes cardiac concentric remodelling in ovariectomized rats, possibly mediated by increased local levels of insulin-like growth factor-I. The effect of DHT on cardiac wall thickness was antagonized by E2, possibly through downregulation of cardiac androgen receptors. These mechanisms may be of importance for the concentric left-ventricular geometric pattern developing in women after menopause.

  • Learning aspect in Italian as a third language : Transfer patterns among multilingual learners in the Swedish context
  • 2023
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis explores the impact of previously acquired or learned background languages (BL) on the learning of Italian as a third language (L3) among undergraduate students in the Swedish context. Focusing on the learning of past tense-aspect (TA) inflectional categories in Italian, the thesis investigates the influence of Swedish, English and Romance languages, French or Spanish, in light of four factors. The typology factor entails the linguistic similarity between the L3 and the BLs. The language status factor examines the different roles played by first (L1) and second (L2) languages. The proficiency factor refers to proficiency in both the L2s and the L3. The prototype factor deals with different combinations of grammatical and lexical aspect, defined as prototypical and non-prototypical. Eight Italian native speakers and 36 undergraduate students learning Italian in Sweden were included in the studies making up the thesis. The learner group had knowledge of Swedish and English. A majority had additional knowledge of a Romance language, which allowed for a distinction to be made between a non-Romance and a Romance group. The participants completed a background questionnaire and a test battery of seven tests consisting of three C-tests (French, Spanish, and Italian), three interpretation tests of aspectual contrasts (English, French, and Italian), and an oral retelling story test in Italian. The data were analyzed quantitatively, by means of descriptive and inferential statistics, and qualitatively, through content analysis. Study I probed the impact of typology, L3 proficiency and prototypes on oral production data. At low-proficiency levels, prior L2 knowledge of a Romance language favored the emergence of imperfective morphology in L3 Italian while learners at high-proficiency levels were sensitive to prototypes. Based on interpretation data, Study II went further into the role of typology, L2 proficiency and prototypes, revealing a beneficial effect of English and Romance languages, except with non-prototypical associations. Study III looked into typology and L2/L3 proficiency. Four categories, differently distributed depending on L2/L3 proficiency, unfolded from written introspection data submitted to qualitative content analysis: explicit rules, intuition, other languages and uncertainty/unknown. Study IV, closely examining three participants with different acquisition orders of Swedish, English and French, showed an interaction of the language status with L3 proficiency, typology and prototypes. Overall, the results indicate that all BLs impact the learning of prototypical associations, while the L1 alone influences learning of non-prototypical associations. Further, the typology factor and the L2 proficiency factor are crucial for transfer of forms and transfer of meanings, respectively. The influence of the BLs is prevalent at early acquisition stages while advanced stages are uniformly shaped by prototypes. Also, learners gradually add different layers of information to their explicit knowledge of tenses. The results align with L3 accounts arguing for property-by-property hybrid transfer. The thesis further connects L3 transfer research focusing on linguistic and conceptual transfer to research on learning TA morphology. In light of the above contributions, the thesis recommends explicit crosslinguistic instruction aiming to foster positive transfer through tailor-made use of the learners’ BLs.

  • Tidskriftsartikel (refereegranskat)abstract
    • The study examines the role played by English and Romance languages (L2s) when learning grammatical aspect in Italian as additional language (Ln). Swedish university students of Italian (n = 34), divided according to knowledge of a Romance L2 and English aspectual knowledge, completed an interpretation task of aspectual contrast in Italian. Eight native speakers served as a control group. The findings showed that knowledge of a Romance language as L2 and high English aspectual knowledge exerted a differential influence on learning aspect in Italian. This outcome is discussed in the light of a consistent form-meaning relationship between the L2s and Italian. Yet, with a mismatch between grammatical and lexical aspect, the learners' judgments differed from the native speakers' judgments. Thus, our findings also support the idea of the existence of differential learning paths sustained by the L2s when learning complex aspectual configurations.

    • Noninvasive tracking of biomaterials is vital for determining the fate and degradation of an implant in vivo, and to show its role in tissue regeneration. Current biomaterials have no inherent capacity to enable tracing but require labeling with, for example, fluorescent dyes, or nanoparticles. Here a novel biocompatible fully conjugated electrospun scaffold is described, based on a semiconducting luminescent polymer that can be visualized in situ after implantation using fluorescence imaging. The polymer, poly [2,3-bis-(3-octyloxyphenyl)quinoxaline-5,8-diyl-alt -thiophene-2,5-diyl] (TQ1), is electrospun to form a fibrous mat. The fibers display fluorescence emission in the near-infrared region with lifetimes in the sub-nanosecond range, optimal for in situ imaging. The material shows no cytotoxic behaviors for embryonic chicken cardiomyocytes and mouse myoblasts, and cells migrate onto the TQ1 fibers even in the presence of a collagen substrate. Subcutaneous implantations of the material in rats show incorporation of the TQ1 fibers within the tissue, with limited inflammation and a preponderance of small capillaries around the fibers. The fluorescent properties of the TQ1 fibers are fully retained for up to 90 d following implantation and they can be clearly visualized in tissue using fluorescence and lifetime imaging, thus making it both a pro-angiogenic and traceable biomaterial.

    • Apex predators may affect mesopredators through intraguild predation and/or supply of carrion from their prey, causing a trade-off between avoidance and attractiveness. We used wildlife triangle snow-tracking data to investigate the abundance of red fox (Vulpes vulpes) in relation to lynx (Lynx lynx) and wolf (Canis lupus) occurrence as well as land composition and vole (Microtus spp.) density. Data from the Swedish wolf-monitoring system and VHF/GPS-collared wolves were used to study the effect of wolf pack size and time since wolf territory establishment on fox abundance. Bottom-up processes were more influential than top-down effects as the proportion of arable land was the key indicator of fox abundance at the landscape level. At this spatial scale, there was no effect of wolf abundance on fox abundance, whereas lynx abundance had a positive effect. In contrast, at the wolf territory level there was a negative effect of wolves on fox abundance when including detailed information of pack size and time since territory establishment, whereas there was no effect of lynx abundance. This study shows that different apex predator species may affect mesopredator abundance in different ways and that the results may be dependent on the spatiotemporal scale and resolution of the data. © 2017 The Author(s).

  • Response of Moose Hunters to Predation following Wolf Return in Sweden
  • 2015
  • Ingår i: PLoS ONE. - : Public Library of Science (PLoS). - 1932-6203. ; 10
  • Tidskriftsartikel (refereegranskat)abstract
    • BackgroundPredation and hunter harvest constitute the main mortality factors affecting the size and dynamics of many exploited populations. The re-colonization by wolves (Canis lupus) of the Scandinavian Peninsula may therefore substantially reduce hunter harvest of moose (Alces alces), the main prey of wolves.Methodology/Principal findingsWe examined possible effects of wolf presence on hunter harvest in areas where we had data before and after wolf establishment (n = 25), and in additional areas that had been continuously exposed to wolf predation during at least ten years (n = 43). There was a general reduction in the total number of moose harvested (n = 31,827) during the ten year study period in all areas irrespective of presence of wolves or not. However, the reduction in hunter harvest was stronger within wolf territories compared to control areas without wolves. The reduction in harvest was larger in small (500-800 km(2)) compared to large (1,200-1,800 km(2)) wolf territories. In areas with newly established wolf territories moose management appeared to be adaptive with regard to both managers (hunting quotas) and to hunters (actual harvest). In these areas an instant reduction in moose harvest over-compensated the estimated number of moose killed annually by wolves and the composition of the hunted animals changed towards a lower proportion of adult females.Conclusions/SignificanceWe show that the re-colonization of wolves may result in an almost instant functional response by another large predator-humans-that reduced the potential for a direct numerical effect on the density of wolves' main prey, the moose. Because most of the worlds' habitat that will be available for future colonization by large predators are likely to be strongly influenced by humans, human behavioural responses may constitute a key trait that govern the impact of large predators on their prey.

  • Epithelial and stromal characteristics of primary tumors predict the bone metastatic subtype of prostate cancer and patient survival after androgen-deprivation therapy

    • Prostate cancer (PC) bone metastases can be divided into transcriptomic subtypes, by us termed MetA-C. The MetB subtype, constituting about 20% of the cases, is characterized by high cell cycle activity, low androgen receptor (AR) activity, and a limited response to standard androgen deprivation therapy (ADT). Complementary treatments should preferably be introduced early on if the risk of developing metastases of the MetB subtype is predicted to behigh. In this study, we therefore examined if the bone metastatic subtype and patient outcome after ADT could be predicted by immunohistochemical analysis of epithelial and stromal cell markers in primary tumor biopsies obtained at diagnosis (n = 98). In this advanced patient group, primary tumor International Society of Urological Pathology (ISUP) grade was not associated with outcome or metastasis subtype. In contrast, high tumor cell Ki67 labeling (proliferation) in combination with low tumor cell immunoreactivity for PSA, and a low fraction of AR positive stroma cells in the primary tumors were prognostic for poor survival after ADT. Accordingly, the same tissue markers were associated with developing metastases enriched for the aggressive MetB subtype. The development of the contrasting MetA subtype, showing the best response to ADT, could be predicted by the opposite staining pattern. We conclude that outcome after ADT and metastasis subtype can, at least to some extent, be predicted by analysis of primary tumor characteristics, such as tumor cell proliferation and PSA expression, and AR expression in stromal cells.

  • High monocyte count and expression of s100a9 and s100a12 in peripheral blood mononuclear cells are associated with poor outcome in patients with metastatic prostate cancer

    • Increasing evidence indicates calcium-binding S100 protein involvement in inflammation and tumor progression. In this prospective study, we evaluated the mRNA levels of two members of this family, S100A9 and S100A12, in peripheral blood mononuclear cells (PBMCs) in a cohort of 121 prostate cancer patients using RT-PCR. Furthermore, monocyte count was determined by flow cytometry. By stratifying patients into different risk groups, according to TNM stage, Gleason score and PSA concentration at diagnosis, expression of S100A9 and S100A12 was found to be significantly higher in patients with metastases compared to patients without clinically detectable metastases. In line with this, we observed that the protein levels of S100A9 and S100A12 in plasma were higher in patients with advanced disease. Importantly, in patients with metastases at diagnosis, high monocyte count and high levels of S100A9 and S100A12 were significantly associated with short progression free survival (PFS) after androgen deprivation therapy (ADT). High monocyte count and S100A9 levels were also associated with short cancer-specific survival, with monocyte count providing independent prognostic information. These findings indicate that circulating levels of monocytes, as well as S100A9 and S100A12, could be biomarkers for metastatic prostate cancer associated with particularly poor prognosis.
