  • Five-year prospective patient evaluation of bladder and bowel symptoms after dose-escalated radiotherapy for prostate cancer with the Beamcath (R) technique

    • Purpose: Late side effects were prospectively evaluated up to 5 years after dose-escalated external beam radiotherapy (EBRT) and were compared with a previously treated series with conventional conformal technique.Methods and Materials: Bladder and bowel symptoms were prospectively evaluated with the Prostate Cancer Symptom Scale (PCSS) questionnaire up to 5 years posttreatment. In all, 257 patients completed the questionnaire 5 years posttreatment. A total of 168 patients were treated with the conformal technique at doses <71 Gy, and 195 were treated with the dose-escalated stereotactic BeamCath® technique comprising three dose levels: 74 Gy (n = 68), 76 Gy (n = 74), and 78 Gy (n = 53).Results: For all dose groups analyzed together, 5 years after treatment, urinary starting problems decreased and urinary incontinence increased in comparison to baseline values. No increase in other bladder symptoms or frequency was detected. When comparing dose groups after 5 years, both the 74-Gy and 78-Gy groups reported increased urinary starting problems compared with patients given the conventional dose (<71 Gy). No increased incontinence was seen in the 76-Gy or the 78-Gy groups. Bowel symptoms were slightly increased during the follow-up period in comparison to baseline. Dose escalation with stereotactic EBRT (74–78 Gy) did not increase gastrointestinal late side effects after 5 years in comparison to doses <71 Gy.Conclusion: Dose-escalated EBRT with the BeamCath® technique with doses up to 78 Gy is tolerable, and the toxicity profile is similar to that observed with conventional doses <71 Gy.

    • © 2019 The Authors Eutrophication is a serious threat to aquatic ecosystems globally with pronounced negative effects in the Baltic and other semi-enclosed estuaries and regional seas, where algal growth associated with excess nutrients causes widespread oxygen free “dead zones” and other threats to sustainability. Decades of policy initiatives to reduce external (land-based and atmospheric) nutrient loads have so far failed to control Baltic Sea eutrophication, which is compounded by significant internal release of legacy phosphorus (P) and biological nitrogen (N) fixation. Farming and harvesting of the native mussel species (Mytilus edulis/trossulus) is a promising internal measure for eutrophication control in the brackish Baltic Sea. Mussels from the more saline outer Baltic had higher N and P content than those from either the inner or central Baltic. Despite their relatively low nutrient content, harvesting farmed mussels from the central Baltic can be a cost-effective complement to land-based measures needed to reach eutrophication status targets and is an important contributor to circularity. Cost effectiveness of nutrient removal is more dependent on farm type than mussel nutrient content, suggesting the need for additional development of farm technology. Furthermore, current regulations are not sufficiently conducive to implementation of internal measures, and may constitute a bottleneck for reaching eutrophication status targets in the Baltic Sea and elsewhere.

  • Grön infrastruktur i havet : landskapsperspektiv i förvaltningen av Sveriges marina områden

    • En kartläggning av havets naturkvaliteter har påbörjats genom arbetet med grön infrastruktur. Rapporten undersöker olika scenarier för hur den marina gröna infrastrukturen påverkas av mänskliga aktiviteter i förhållande till olika strategier för förvaltning. Projektet har genomfört tre fallstudier med olika fokus: strandexploatering, trålfiske och klimatförändring. Genom scenariobaserade analyser visar forskarna vilken effekt olika beslut och åtgärder kan få på naturskydds- och miljömål.Det tvärvetenskapliga projektet är ett samarbete mellan experter inom ekologi och juridik. Forskarna beskriver hur lagstiftning, planering och förvaltningsstrategier bör utformas och fungera ihop, och presenterar förslag för att stärka skyddet av den gröna infrastrukturen i Sveriges havsområden.Forskningen har finansierats av Naturvårdsverkets miljöforskningsanslag till stöd för Naturvårdsverket och Havs- och vattenmyndighetens verksamhet.

  • Exceptional longevity in northern peripheral populations of Wels catfish (Siluris glanis)

    • Studies of life-history variation across a species range are crucial for ecological understanding and successful conservation. Here, we examined the growth and age of Wels catfish (Silurus glanis) in Sweden, which represent the northernmost populations in Europe. A total of 1183 individuals were captured, marked and released between 2006 and 2020. Mark-recapture data from 162 individuals (size range: 13-195 cm) were used to estimate von Bertalanffy growth curve parameters which revealed very slow growth rates compared to catfish within the core distribution area (central Europe). The fitted von Bertalanffy growth curve predicted a 150 cm catfish to be around 40 years old, while the largest recaptured individual (length 195 cm) was estimated to be 70 (95% CI 50-112) years old. This was substantially older than the previously documented maximum age of a catfish. The weight at length relationships in these northern peripheral populations were similar to those documented for catfish in central Europe indicating that resources did not constrain growth. This indicates that the slow growth and exceptional high age in the northern catfish populations are the result of lower temperatures and/or local adaptations.

  • Seasonal and Daily Movement Patterns by Wels Catfish (Silurus glanis) at the Northern Fringe of Its Distribution Range

    • Fish behavior often varies across a species' distribution range. Documenting how behaviors vary at fringes in comparison to core habitats is key to understanding the impact of environmental variation and the evolution of local adaptations. Here, we studied the behavior of Wels catfish (Silurus glanis) in Lake M & ouml;ckeln, Sweden, which represent a European northern fringe population. Adult individuals (101-195 cm, N = 55) were caught and externally marked with data storage tags (DSTs). Fifteen DSTs were recovered a year after tagging, of which 11 tags contained long-term high-resolution behavioral data on the use of vertical (depth) and thermal habitats. This showed that the catfish already became active in late winter (<5 degrees C) and displayed nocturnal activity primarily during summer and late autumn. The latter included a transition from the bottom to the surface layer at dusk, continuous and high activity close to the surface during the night, and then descent back to deeper water at dawn. During the daytime, the catfish were mainly inactive in the bottom layer. These behaviors contrast with what is documented in conspecifics from the core distribution area, perhaps reflecting adaptive strategies to cope with lower temperatures and shorter summers.

  • Ekologisk kompensation i kustmiljön : Hur kan man uppväga förluster av biologisk mångfald och ekosystemtjänster i samband med mänsklig verksamhet i kustområdet?

    • Sveriges kustmiljöer är utsatta för olika typer av mänsklig påverkan, med negativa effekter på arter, livsmiljöer och ekosystemtjänster. Enligt skadelindringshierarkin ska negativa effekter från mänsklig verksamhet i första hand undvikas, minimeras eller avhjälpas. I de kvarstående fall där skada trots dessa åtgärder är oundviklig, kan ekologisk kompensation vara ett sätt att mildra effekterna i syfte att bibehålla eller öka biologisk mångfald och ekosystemtjänster.I projektet ECOCOA har vi undersökt om och hur ekologisk kompensation skulle kunna användas i förvaltningen av kustområden. Vår målsättning har varit att bidra till en vetenskapligt grundad syn på hur ekologisk kompensation skulle kunna medverka till att stoppa nettoförluster av biologisk mångfald och ekosystemtjänster. Eftersom kompensation kan beröra en mängd aktörer och sakägare med olika intressen och perspektiv är det viktigt med en transparent process som hanteras på ett jämförbart sätt mellan olika områden och fall. En viktig flaskhals idag är brist på praktisk erfarenhet hos aktörer om hur kompensationen ska kunna tillämpas. Vi har utvecklat ett ramverk utifrån de steg som man behöver beakta i en bedömning av skada och kompensationsbehov, med fokus på att öka förutsättningarna för en transparent bedömningsprocess. Det föreslagna ramverket bygger på fyra steg: 1) skadebedömning; 2) bedömning av kompensationsbehov; 3) val av kompensationsåtgärder; och 4) utvärdering av resultat. Ramverket baserar sig på att tydliggöra samband mellan ekosystemets strukturer och de nyttigheter dessa kan medföra för människan, där nyttigheter är beroende av ekosystemtjänster, ekosystemtjänster av funktioner, och funktioner av strukturer. Vi kopplar ramverket till en utvärdering av aktuellt kunskapsläge kring åtgärders effektivitet i kustområdet. Viktiga livsmiljöer som ingår är ålgräs, grunda vegetationsklädda mjukbottnar, naturligt vegetationsfria mjukbottnar, tångbälten, musselbankar, stenrev och kustnära våtmarker.Vi har med hjälp av ramverket utvärderat nuvarande och möjliga tillämpningar av ekologisk kompensation. Utvärderingen har gjorts dels genom ett fullständigt exempel som beskriver förlust av ålgräsängar och dels genom att undersöka befintliga fall och sammanhang där kompensation är eller skulle kunna bli relevant. Fallen bygger på i) domslut gällande tillståndspliktig vattenverksamhet, ii) dagens hantering av småskalig exploatering i kustområden, samt iii) tillämpningen av särskild fiskeavgift ur ett kompensationsperspektiv. Slutligen har vi utvecklat förslag på hur liknande tillämpningar skulle kunna se ut för ytterligare livsmiljöer i tillägg till ålgräs.Våra resultat visar det finns fortsatt stor osäkerhet kring hur kompensationsprocesser och kompensationsåtgärder fungerar i praktiken. Samtidigt är det en realitet att man idag i de allra flesta fall exploaterar utan kompensation, eller med otillräcklig kompensation. Detta innebär i praktiken att förluster värderas till noll, det vill säga att man accepterar förluster på biologisk mångfald, ekosystemtjänster och nyttigheter, och att de skador som uppstår inte uppmärksammas i form av åtgärdsbehov som ska täckas av den som orsakar skadan. Eftersom tillämpningen av vårt ramverk synliggör värdet av att förvalta biologisk mångfald och ekosystemtjänster tillsammans kan det även stödja förvaltningen av kustområden i enlighet med ekosystemansatsen på en mer generell nivå.Våra resultat synliggör vikten av att värna biologisk mångfald. Vår genomgång av potentiella kompensationsåtgärder, som redogörs för i rapporten, visar med tydlighet att det är kostsamt att först förstöra och sedan restaurera, i stället för att skydda och undvika skada i första hand. Vi ser dock ett utrymme och ett behov av att förbättra användningen av ekologisk kompensation som ett av många verktyg för att värna biologisk mångfald och ekosystemtjänster i svenska kustmiljöer i enlighet med miljöbalken. ECOCOAs ramverk tillsammans med kaskadmodeller för viktiga livsmiljöer och information om potentiella åtgärder, kunde här fungera som en gemensam utgångspunkt vid diskussioner, samrådsprocesser och bedömningar.

  • Shape verification using dual-wavelength holographic interferometry

    • In automotive industry there is an interest of controlling the shape of a large number of identical components on-line in the manufacturing process. We propose a method to do this by capturing a digital hologram of the object and then using information from its computer aided design (CAD) model to calculate the shape and determine the agreement between the manufactured object and the CAD-model. The holographic recording of the object is done using dual wavelengths with a synthetic wavelength of approximately 400 μm. The optical measurement results in a wrapped phase map with the phase values in the interval [−π, π]. Each phase interval represents a depth distance on the object of about 0.2 mm. The phase unwrapping is done iteratively using information from the CADmodel. This implies that it is possible to measure large discontinuities on the surface of the measured object. The method also gives a point-to-point correspondence between the measurement and the CAD-model which is vital for tolerance control.

    • RATIONALE: Chronic obstructive pulmonary disease (COPD) includes respiratory symptoms and chronic airflow limitation (CAL). In some cases, emphysema and impaired diffusing capacity for carbon monoxide (DLCO) are present, but characteristics and symptoms vary with smoking exposure.OBJECTIVES: To study the prevalence of CAL, emphysema and impaired DLCO in relation to smoking and respiratory symptoms in a middle-aged population.METHODS: We investigated 28,746 randomly invited individuals (52% women) aged 50-64 years across six Swedish sites. We performed spirometry, DLCO, high-resolution computed tomography (HRCT) and asked for smoking habits and respiratory symptoms. CAL was defined as post-bronchodilator forced expiratory volume in 1 second divided by forced expiratory volume (FEV1/FVC)<0.7.RESULTS: The overall prevalence was for CAL 8.8%, for impaired DLCO (DLCOCONCLUSIONS: In this large population-based study of middle-aged people, CAL and impaired DLCO were associated with common respiratory symptoms. Self-reported asthma was not associated with CAL in never-smokers. Our findings suggest that CAL in never-smokers signifies a separate clinical phenotype that may be monitored and, possibly, treated differently from smoking-related COPD.   • Environmental compensation for biodiversity and ecosystem services : A flexible framework that addresses human wellbeing

    • Environmental compensation should address negative impacts from human activities on nature, including loss of biodiversity and ecosystem services. However, successful compensation, achieving no net loss, requires broad quantitative information on different types of losses and gains. We find that the scope of compensatory schemes varies in what is considered compensable, which makes it challenging to apply a conceptual approach consistently across schemes with different needs. We propose a flexible yet structured framework for determining which values should be compensated and how. Our framework focuses specifically on habitat deterioration and is illustrated with a case study involving loss of eelgrass habitat. The framework helps identify compensation needs and selects among suitable compensation options, merging science-based information with normative issues and local concerns. By integrating the ecosystem services cascade model, it encompasses aspects from biodiversity structure to human wellbeing. The framework prefers in-kind compensation because this targets the structure level and thus meets compensation needs in all subsequent levels of the cascade model; further, it is more likely to capture non-instrumental values (i.e. in nature) and reduce exposure to uncertainty. We highlight the importance of spatial aspects of ecosystem functions, services and their subsequent impacts on wellbeing. Although our selection hierarchy assumes a similar and nearby principle for habitat restoration (preference for in-kind/on-site), this criterion is not universal. We underscore the hierarchy's implicit normative assumptions and suggest that apparent disagreement about who should benefit may be traced to an unresolved conflict between egalitarianism and utilitarianism.

  • The effects of small-scale coastal development on the eelgrass (Zostera marina L.) distribution along the Swedish west coast – Ecological impact and legal challenges

    • Anthropogenic impacts on coastal areas have led to an increased degradation of marine environments globally. Eelgrass ecosystems are particularly susceptible to human induced stressors as they are sensitive to low light conditions and usually grow in shallow protected areas where pressure from coastal development is high. The extensive decline in coverage of eelgrass along the Swedish Northwest coast since the 1980s has largely been attributed to the effects of coastal eutrophication and overfishing. However, the impact on eelgrass from small-scale coastal development (docks and marinas) has never been investigated in this area. The aim of this study was to assess the local and large-scale effect of shading by docks and marinas on eelgrass habitats along the Swedish NW coast and to investigate the decision process behind small-scale exploitation to identify problems with the current legislation, which allows for continued exploitation of eelgrass. Through field assessments of eelgrass around docks and analysis of available data on eelgrass and dock distribution along the coast, the present study demonstrates that shading from docks reduced eelgrass coverage with on average 42e64% under and adjacent to the docks, and that floating docks affected larger areas and caused a much stronger reduction in eelgrass coverage (up to 100% loss) compared to docks elevated on poles (up to 70% reduction in coverage). The total eelgrass area negatively affected by docks and marinas along the NW coast was estimated to approximately 480 ha, an area corresponding to over 7% of the present areal coverage of eelgrass in the region. The analysis of decisions for dock construction showed that eelgrass was generally not assessed or considered in the decision process and that 69e88% of the applications were approved also in areas where eelgrass was present. Furthermore, marine protected areas only marginally reduced the approval of applications in eelgrass habitats. The continued small-scale development along the Swedish NW coast constitutes a significant threat to the already decimated coverage of eelgrass along the coast and changes in the management practices are needed in order to achieve both national and international goals on environmental status.

  • Effect of Increased Radiotoxicity on Survival of Patients with Non-small Cell Lung Cancer Treated with Curatively Intended Radiotherapy

    • Aim: To elucidate the impact of different forms of radiation toxicities (esophagitis, radiation pneumonitis, mucositis and hoarseness), on the survival of patients treated with curatively intended radiotherapy for non-small cell lung cancer (NSCLC).Patients and Methods: Data were individually collected retrospectively for all patients diagnosed with NSCLC subjected to curatively intended radiotherapy (>= 50 Gy) in Sweden during the time period 1990 to 2000.Results: Esophagitis was the only radiation-induced toxicity with an impact on survival (hazard ratio=0.83, p=0.016). However, in a multivariate model, with clinical-and treatment-related factors taken into consideration, the impact of esophagitis on survival was no longer statistically significant (hazard ratio=0.88, p=0.17).Conclusion: The effect on survival seen in univariate analysis may be related to higher radiation dose and to the higher prevalence of chemotherapy in this group. The results do not suggest that the toxicities examined have any detrimental effect on overall survival.

    • Background and purpose. The variability in target delineation for similar cases between centres treating paediatric and adolescent patients, and the apparent differences in interpretation of radiotherapy guidelines in the treatment protocols encouraged us to perform a dummy-run study as a part of our quality assurance work. The aim was to identify and quantify differences in the segmentation of target volumes and organs at risk (OARs) and to analyse the treatment plans and dose distributions. Materials and methods. Four patient cases were selected: Wilms tumour, Hodgkins disease, rhabdomyosarcoma of the prostate and chordoma of the skull base. The five participating centres received the same patient-related material. They introduced the cases in their treatment planning system, delineated target volumes and OARs and created treatment plans. Dose-volume histograms were retrieved for relevant structures and volumes and dose metrics were derived and compared, e. g. target volumes and their concordance, dose homogeneity index (HI), treated and irradiated volumes, remaining volume at risk and relevant V x and D x values. Results. We found significant differences in target segmentation in the majority of the cases. The planning target volumes (PTVs) varied two-to four-fold and conformity indices were in the range of 0.3-0.6. This resulted in large variations in dose distributions to OARs as well as in treated and irradiated volumes even though the treatment plans showed good conformity to the PTVs. Potential reasons for the differences in target delineation were analysed. Conclusion. Considerations of the growing child and difficulties in interpretation of the radiotherapy information in the treatment protocols were identified as reasons for the variation. As a result, clarified translated detailed radiotherapy guidelines for paediatric/adolescent patients have been recognised as a way to reduce this variation.

    • Several platelet-derived growth factor (PDGF) and vascular endothelial growth factor (VEGF) family members display C-terminal protein motifs that confer retention of the secreted factors within the pericellular space. To address the role of PDGF-B retention in vivo, we deleted the retention motif by gene targeting in mice. This resulted in defective investment of pericytes in the microvessel wall and delayed formation of the renal glomerulus mesangium. Long-term effects of lack of PDGF-B retention included severe retinal deterioration, glomerulosclerosis, and proteinuria. We conclude that retention of PDGF-B in microvessels is essential for proper recruitment and organization of pericytes and for renal and retinal function in adult mice.

    • BACKGROUND: The standard treatment of solitary brain metastases previously has been tumour resection in combination with whole-brain radiation therapy (WBRT). Stereotactic radiotherapy has emerged as a non-invasive treatment option especially for small brain metastases. We now report our results on resection + WBRT or hypofractionated stereotactic irradiation (HCSRT) in the treatment of solitary brain metastases. METHODS: Between 1993 and 2004 patients with metastatic cancer and solitary brain metastases were selected for surgical resection + WBRT or HCSRT alone at the Umeå University Hospital. Fifty-nine patients were treated with surgical resection + WBRT (34 male, 25 female, mean age 63.3 years). Forty-seven patients were treated with HCSRT alone (15 male, 32 female, mean age 64.9 years). FINDINGS: In patients followed radiologically, 28% treated with resection + WBRT showed a local recurrence after a median time of 8.0 months, whereas there was a lack of local control in 16% in the HCSRT group after a median time of 3.0 months. There was a significantly longer survival time for patients treated with resection + WBRT (median 7.9, mean 12.9 months) compared to HCSRT (median 5.0, mean 7.6 months). Even in patients with a tumour volume <10 cc, there was a significantly longer survival in favour of resection + WBRT (median 8.4, mean 17.4 months) compared to HCSRT (median 5.0, mean 7.9 months). CONCLUSION: This retrospective and non-randomised study indicates that surgical resection in combination with WBRT may be an option even for small brain metastases suitable for treatment with HCSRT. Since survival and local control following resection + WBRT was at least as favourable as compared to HCSRT alone, tumour location and expected neurological outcome may be the strongest aspect when selecting treatment modality.

    • There are three major treatment options for cerebral AVMs; surgery, embolization and radiosurgery. Embolization may be effective to reduce the size and density but completely obliterates AVMs only in a minority of cases. Radiosurgery may be an alternative to resection, especially in smaller AVMs. Large AVMs have been considered difficult to treat safely and effectively with single fraction radiosurgery. Hypofractionated conformal stereotactic radiotherapy (HCSRT)alone or in combination with embolization may be an alternative treatment. Embolization may reduce the volume and density of AVMs, followed by HCSRT, allowing a safe delivery of a higher total dose of radiation than possible with a single fraction. Sixteen patients with AVMs were treated with embolization and HCSRT. Embolization was performed in 1-6(median 2) sessions. HCSRT was delivered in 5fractions with 6-7 Gy each to the total dose of30–35 Gy. Cerebral angiographies before and after embolization were digitally compared for calculation of volume reduction and luminescence as a measure of AVM density. The mean AVM volume in 15 patients was reduced from11.9 ± 2.1 (1-29, median 10.0) ml to 6.5 ± 2.0(0.5–28, median 3) ml by embolization. The luminescence for all AVMs was significantly higher after than before embolization, indicating that all AVMs were less dense after embolization. Thirteen out of 16 patients (13/16, 81%) treatedwith embolization and HCSRT have so farshown obliteration of their AVMs 2-9 (median4) years after HCSRT. Three patients experienced neurological sequele after embolization, and three patients developed radionecrosis after HCSRT. Using a new method to compare cerebral angiographies in AVMs we report reduction in density and volume after embolization. The obliteration rate of a combined treatment with embolization and HCSRT seems comparable with single fraction radiosurgery although the AVMs in our series are larger than reported in most series treated with single fraction radiosurgery.

  • Reproducibility and geometric accuracy of the Fixster system during hypofractionated stereotactic radiotherapy

    • BACKGROUND:Hypofractionated radiotherapy has been used for the treatment of AVMs and brain metastases. Hypofractionation necessitates the use of a relocatable stereotactic frame that has to be applied on several occasions. The stereotactic frame needs to have a high degree of reproducibility, and patient positioning is crucial to achieve a high accuracy of the treatment.METHODS:In this study we have, by radiological means, evaluated the reproducibility of the isocenter in consecutive treatment sessions using the Fixster frame. Deviations in the X, Y and Z-axis were measured in 10 patients treated with hypofractionated radiotherapy.RESULTS:The mean deviation in the X-axis was 0.4 mm (range -2.1 - 2.1, median 0.7 mm) and in the Y-axis -0.3 mm (range -1.4 - 0.7, median -0.2 mm). The mean deviation in the Z-axis was -0.6 (range -1.4 - 1.4, median 0.0 mm).CONCLUSION:There is a high degree of reproducibility of the isocenter during successive treatment sessions with HCSRT using the Fixster frame for stereotactic targeting. The high reducibility enables a safe treatment using hypofractionated stereotactic radiotherapy.

  • Neutrino Physics : Proceedings of Nobel Symposium 129

    • PREFACE Nobel Symposium 129 on Neutrino Physics was held at Haga Slott in Enköping, Sweden during August 19–24, 2004. Invited to the symposium were around 40 globally leading researchers in the field of neutrino physics, both experimental and theoretical. In addition to these participants, some 30 local researchers and graduate students participated in the symposium. The dominant theme of the lectures was neutrino oscillations, which after several years were recently verified by results from the Super-Kamiokande detector in Kamioka, Japan and the SNO detector in Sudbury, Canada. Discussion focused especially on effects of neutrino oscillations derived from the presence of matter and the fact that three different neutrinos exist. Since neutrino oscillations imply that neutrinos have mass, this is the first experimental observation that fundamentally deviates from the standard model of particle physics. This is a challenge to both theoretical and experimental physics. The various oscillation parameters will be determined with increased precision in new, specially designed experiments. Theoretical physics is working intensively to insert the knowledge that neutrinos have mass into the theoretical models that describe particle physics. It will probably turn out that the discovery of neutrino oscillations signifies a breakthrough in the description of the very smallest constituents of matter. The lectures provided a very good description of the intensive situation in the field right now. The topics discussed also included mass models for neutrinos, neutrinos in extra dimensions as well as the `seesaw mechanism', which provides a good description of why neutrino masses are so small. Also discussed, besides neutrino oscillations, was the new field of neutrino astronomy. Among the questions that neutrino astronomy hopes to answer are what the dark matter in the Universe consists of and where cosmic radiation at extremely high energies comes from. For this purpose, large neutrino telescopes are built deep in the Antarctic ice, in the Baikal Lake, and in the Mediterranean Sea. Among prominent unanswered questions, highlighted as one of the most important, was whether neutrinos are Dirac or Majorana particles. By studying neutrino double beta decay, researchers hope to answer this question, but it will put very large demands on detectors. The programme also included ample time for lively and valuable discussions, which cannot normally be held at ordinary conferences. The symposium concluded with a round-table discussion, where participants discussed the future of neutrino physics.Without a doubt, neutrino physics today is moving toward a very exciting and interesting period. An important contribution to the success of the symposium was the wonderful setting that the Haga Slott manor house hotel and conference center offered to the participants.

    • In the Skagerrak-Kattegat (eastern North Sea), the alien benthic foraminifer Nonionella sp. T1 (previously referred to as “Nonionella stella”) was reported for the first time in 2011 and 2012 in the Swedish and southern Norwegian fjords, respectively. Based on dated sediment cores its first occurrence can be traced back to the 1980s in the Gullmar Fjord, to the 2000s in the Öresund and to 2010 in the Oslofjord. Since then, Nonionella sp T1 has spread all over the Kattegat and coastal Skagerrak, according to sampling campaigns performed between 2016 and 2022. The species is now highly abundant in the entire Kattegat, including the Öresund, as well as in fjord mouths of the seasonally hypoxic Gullmar Fjord, the oxic Hakefjord and the long-term polluted Idefjord as demonstrated by molecular and morphospecies data. At the same time, Nonionella sp T1 is rare to absent in the Baltic Sea, Skagerrak deep basin and in deep fjords of western and northern Norway. This study shows some preliminary results on the species’ present distribution in the study area and raises questions about the driving factors and potential effects on the local biodiversity.

  • Dual-wavelength holographic shape measurement with iterative phase unwrapping

    • In order to measure the shape of a large number of identical components in a manufacturing industry we propose a method where digital holography is used to capture an image of the object and then the shape of the object is achieved by using information from the CAD-model. The holographic recording of the object is done using dual wavelengths giving a synthetic wavelength of about 400 μm. This gives a phase map where the phase intervals represent a depth distance on the object of about 0.2 mm. To find the shape of the object the phase map has to be unwrapped. Since the surface contains discontinuities we use information from the CAD-model of the measured object and unwrap the phase iteratively. The result becomes a digital point representation of the measured surface that can either be used just as a description of the object shape or as a way to describe how well the object has been manufactured compared to the CAD-model. The measurement process that is proposed is adapted for on-line purposes; hence it is fast and reliable.

  • Comparison between process simulation and deformation measured by defocused speckle photography

    • Defocused laser speckle photography is used as a tool to measure the heat responses in a titanium component during laser heating. The evolution of the response is compared with a set of preprocessed Finite Element Simulations of the corresponding process with the aim to verify the simulation model and to find the simulation settings that best resemble the experimental results. The titanium component consists of a 300 x 100 mm2 substrate of thickness 3.2 mm on which a 200 x 30 x 11 mm3 ridge is built up using the laser metal deposition by wire process. The component is heated on the top of the ridge by a 300 W laser for 10 s and the deformation of the subtrate is followed throughout the heating-cooling cycle. The simulated deformation gradient is shown to resemble the measured response, and the magnitude of the response indicates that about 70 % of the laser power transferres into heat in the metal.

    • An important aspect of precision medicine is to probe the stability in molecular profiles among healthy individuals over time. Here, we sample a longitudinal wellness cohort with 100 healthy individuals and analyze blood molecular profiles including proteomics, transcriptomics, lipidomics, metabolomics, autoantibodies andimmune cell profiling, complementedwith gut microbiota composition and routine clinical chemistry. Overall, our results show high variation between individuals across different molecular readouts, while the intra-individual baseline variation is low. The analyses show that each individual has a unique and stable plasma protein profile throughout the study period and that many individuals also show distinct profiles with regards to the other omics datasets, with strong underlying connections between the blood proteome and the clinical chemistry parameters. In conclusion, the results support an individual-based definition of health and show that comprehensive omics profiling in a longitudinal manner is a path forward for precision medicine.

  • In vitro and in silico prediction of drug-drug interactions with transport proteins

    • Drug transport across cells and cell membranes in the human body is crucial for the pharmacological effect of drugs. Active transport governed by transport proteins plays an important role in this process. A vast number of transport proteins with a wide tissue distribution have been identified during the last 15 years. Several important examples of their role in drug disposition and drug-drug interactions have been described to date. Investigation of drug-drug interactions at the transport protein level are therefore of increasing interest to the academic, industrial and regulatory research communities. The gene expression of transport proteins involved in drug transport was investigated in the jejunum, liver, kidney and colon to better understand their influence on the ADMET properties of drugs. In addition, the gene and protein expression of transport proteins in cell lines, widely used for predictions of drug transport and metabolism, was examined. The substrate and inhibitor heterogeneity of many transport proteins makes it difficult to foresee whether the transport proteins will cause drug-drug interactions. Therefore, in vitro assays for OCT1 and OATP1B1, among the highest expressed transport proteins in human liver, were developed to allow investigation of the inhibitory patterns of these proteins. These assays were used to investigate two data sets, consisting of 191 and 135 registered drugs and drug-like molecules for the inhibition of OCT1 and OATP1B1, respectively. Numerous new inhibitors of the transport proteins were identified in the data sets and the properties governing inhibition were determined. Further, antidepressant drugs and statins displayed strong inhibition of OCT1 and OATP1B1, respectively. The inhibition data was used to develop predictive in silico models for each of the two transport proteins. The highly polymorphic nature of some transport proteins has been shown to affect drug response and may lead to an increased risk of drug-drug interactions, and therefore, the OCT1 in vitro assay was used to study the effect of common genetic variants of OCT1 on drug inhibition and drug-drug interactions. The results indicated that OCT1 variants with reduced function were more susceptible to inhibition. Further, a drug-drug interaction of potential clinical significance in the genetic OCT1 variant M420del was proposed. In summary, gene expression of transport proteins was investigated in human tissues and cell lines. In vitro assays for two of the highest expressed liver transport proteins were used to identify previously unknown SLC transport protein inhibitors and to develop predictive in silico models, which may detect previously known drug-drug interactions and enable new ones to be identified at the transport protein level. In addition, the effect of genetic variation on inhibition of the OCT1 was investigated.

    • The liver-specific organic cation transport protein (OCT1; SLC22A1) transports several cationic drugs including the antidiabetic drug metformin and the anticancer agents oxaliplatin and imatinib. In this study, we explored the chemical space of registered oral drugs with the aim of studying the inhibition pattern of OCT1 and of developing predictive computational models of OCT1 inhibition. In total, 191 structurally diverse compounds were examined in HEK293-OCT1 cells. The assay identified 47 novel inhibitors and confirmed 15 previously known inhibitors. The enrichment of OCT1 inhibitors was seen in several drug classes including antidepressants. High lipophilicity and a positive net charge were found to be the key physicochemical properties for OCT1 inhibition, whereas a high molecular dipole moment and many hydrogen bonds were negatively correlated to OCT1 inhibition. The data were used to generate OPLS-DA models for OCT1 inhibitors; the final model correctly predicted 82% of the inhibitors and 88% of the noninhibitors of the test set.

    • A new non-hydrolytic, alkoxide-based route was developed to synthesize iron oxide nanocrystals. Surfactant-free thermal decomposition of the iron 2-methoxy-ethoxide precursors results in the formation of uniform iron oxide nanocrystals with an average size of 5.6 nm. Transmission electron microscope study shows that the nanocrystals are protected against aggregation by a repulsive surface layer, probably originating from the alkoxy-alkoxide ligands. Addition of oleic acid resulted in monodisperse nanocrystals with an average size of 4 nm. Mössbauer analysis confirmed that the nanocrystals mainly consisted of maghemite. Analysis of the magnetic hysteresis loop measurements and the zero field and field cooled measurements displayed an excellent fit to established theories for single-domain superparamagnetic nanocrystals and the size of the magnetic domains correlated well to the crystallite size obtained from transmission electron microscope.

  • Microgels as drug delivery vehicles : loading and release of amphiphilic drugs

    • Polyelectrolyte microgels are used as delivery vehicles for amphiphilic drugs in, e.g., treatments of liver cancer by a method called trans-arterial chemoembolization. The thesis deals with fundamental properties of such delivery systems related to the self-assembling properties of the drug molecules and their interaction with the charged polymer network of the microgel. The main objective was to establish mechanistic models describing the loading and release of drugs under relevant conditions. For that purpose experimental techniques providing thermodynamic, compositional and microstructural information were used to elucidate how the kinetics depend on the stability of the drug self-assemblies and the volume response of the microgels. Micromanipulator-assisted microscopy studies showed that negatively charged microgels phase separated during loading and release of cationic amphiphilic drugs. At intermediate loading levels the drug aggregates and part of the network formed a collapsed phase coexisting with a swollen, drug-lean phase. In particular, during release in a medium of physiological ionic strength, the drug-lean phase formed a depletion layer (shell) surrounding a drug-rich core. Investigations of a series of drugs with different molecular architectures showed that the drug release rate was determined mainly by the stability of the drug aggregates in the core and the diffusive mass transport of drug molecules through the shell. Detailed studies of polyacrylate microgels interacting with amitriptyline hydrochloride showed that swelling of the shell network greatly influenced the release rate. Furthermore, experiments with a specially constructed microscopy cell was used to establish that the collapsed and swollen phases could coexist in equilibrium, and that the swelling of the network in the swollen phase depended on the proportion between them when present in the same microgel. The latter effect was related to the elastic coupling between the phases. Confocal Raman microscopy was employed to demonstrate, for the first time, the related elastic effect, that the concentration of amitriptyline in the swollen phase decreased with increasing proportion of the collapsed phase. Small-angle X-ray scattering showed that the collapsed phase had a disordered microstructure of drug micelles with ellipsoidal shape. The aggregation number increased with increasing concentration of drug in the microgel, most likely by incorporating the uncharged base form. By providing detailed information about thermodynamic properties and microstructures, the results of the thesis provide a basis for rational design of microgel drug delivery systems.

    • This study examines primary school students’ perception of functionality in their spontaneous programminglanguage for controlling a simple robot. Classroom activities were designed in order to create opportunitiesfor the students (year 1 and year 4) to discuss and develop together with their teachers a sharedprogramming language for controlling a simple robot. The students spontaneously used (a) natural language,(b) images or (c) symbols when they created their programming language. The findings show thatthe students primarily perceived a code’s functionality as a question of readability, rather than how wellit fit the purpose of controlling the robot. Possible consequences of the findings for teaching in technologyeducation are discussed.

    • This article describes the development and empirical status of guided Internet-delivered self-help. The treatment approach combines the benefits of bibliotherapy with book-length text materials and the support given online via web pages and e-mail. Interactive features such as online registrations, tests, and online discussion forums are also included. Cognitive behavior therapy (CBT) guided the research and clinical implementations of this approach, as it lends itself more easily to the self-help format compared with other presently available psychotherapy approaches. We include an overview of the research, current issues and research in service delivery, lessons learned through a program of research, and directions for future investigations

    • BACKGROUND:Major depression can be treated by means of cognitive-behavioural therapy, but as skilled therapists are in short supply there is a need for self-help approaches. Many individuals with depression use the internet for discussion of symptoms and to share their experience.AIMS:To investigate the effects of an internet-administered self-help programme including participation in a monitored, web-based discussion group, compared with participation in web-based discussion group only.METHOD:A randomised controlled trial was conducted to compare the effects of internet-based cognitive-behavioural therapy with minimal therapist contact (plus participation in a discussion group) with the effects of participation in a discussion group only.RESULTS:Internet-based therapy with minimal therapist contact, combined with activity in a discussion group, resulted in greater reductions of depressive symptoms compared with activity in a discussion group only (waiting-list control group). At 6 months' follow-up, improvement was maintained to a large extent.CONCLUSIONS:Internet-delivered cognitive cognitive-behavioural therapy should be pursued further as a complement or treatment alternative for mild-to-moderate depression.
