    • OBJECTIVE: Infantile malignant osteopetrosis is a fatal disease caused by lack of functional osteoclasts. In most of patients, TCIRG1, encoding a subunit of a proton pump essential for bone resorption, is mutated. Osteopetrosis leads to bone marrow failure and blindness due to optic nerve compression. Oc/oc mice have a deletion in Tcirg1 and die around 3 to 4 weeks, but can be rescued by neonatal stem cell transplantation (SCT) after irradiation conditioning. However, as irradiation of neonatal mice results in retinal degeneration, we wanted to investigate whether conditioning with busulphan prior to SCT can lead to preservation of vision and reversal of osteopetrosis in the oc/oc mouse model. MATERIALS AND METHODS: Pregnant dams were conditioned with busulphan and their litters transplanted with 1 x 10(6) normal lineage-depleted bone marrow cells intravenously or intraperitoneally. Mice were followed in terms of survival and engraftment level, as well as with peripheral blood lineage analysis, bone and eye histopathology and a visual-tracking drum test to assess vision. RESULTS: Busulphan at 15 mg/kg was toxic to oc/oc mice. However, six of seven oc/oc mice conditioned with busulphan 7.5 mg/kg survived past the normal lifespan with 10% engraftment, correction of the skeletal phenotype, and normalization of peripheral blood lineages. Busulphan, in contrast to irradiation, did not have adverse effects on the retina as determined by histopathology, and 8 weeks after transplantation control and oc/oc mice retained their vision. CONCLUSION: Low-dose busulphan conditioning and neonatal SCT leads to prolonged survival of oc/oc mice, reverses osteopetrosis and prevents blindness even at low engraftment levels.

  • Prospects for gene therapy of osteopetrosis.

    • Dysfunction in or lack of osteoclasts result in osteopetrosis, a group of rare but often severe, genetic disorders characterized by an increase in bone mass, skeletal malformations and bone marrow failure that may be fatal. Several of the underlying defects have lately been characterized in humans and in animal disease models. In humans, these defects often involve mutations in genes expressing proteins involved in the acidification of the osteoclast sub-cellular compartment, a process necessary for proper bone resorption. So far, the only cure for children with severe osteopetrosis is allogeneic hematopoietic stem cell transplantation (SCT). However, the characterization of the genetic defects opens up the possibility for gene replacement therapy as an alternative to SCT. Recently, gene therapy targeting hematopoietic stem cells (HSC) in a mouse model of infantile malignant osteopetrosis was shown to correct many aspects of the disease. Here we review important aspects of this group of diseases and discuss the prospects for development of gene therapy of osteopetrosis.

    • A multilevel theory of agility for C2 is absent. This paper uses an organizing paradigm to reinterpret the existing body ofresearch in order to explore the multilevel self-organizing dynamics that underpin the emergence of organizationalagility. Following social theory’s practice-turn, we outline in section 2 the essence of the organizing paradigm and mapsome of its implications. We reflect on the routines, creativity and complexity inherent in human organizing, and howorganizing practices manifest new collective C2 capabilities for unanticipated, unexpected and unfamiliar circumstances.Then we consider the role and limits of individual agency and rationality within the process of organizing, placing theemphasis on a collective process of structuring that imposes change and order in structure. In section 3 we present anillustration of some organizing principles and practices that may feature within a multi-level theory of agility. We explorehow collective sensemaking is intertwined with collective organizing, and briefly reflect on a conceptual model of agilityin the organizing paradigm. In the concluding section 4 we discuss the usefulness of an organizing paradigm for C2 agilityresearch and invite collaboration in researching cross-level organizing processes and practices.

    • The explosive development of the telecom industry and in particular wireless and mobile communications in recent years has lead to a rapid development of new component and fabrication technologies to continually satisfy the mutually exclusive requirements for better performance and miniaturization on the one hand and low cost on the other. A fundamental element in radio communications is time and frequency control, which in turn is achieved by high performance electro-acoustic components made on piezoelectric single crystalline substrates. The latter, however, reach their practical limits in terms of performance and cost as the frequency of operation reaches the microwave range. Thus, the thin film electro-acoustic technology, which uses thin piezoelectric films instead, has been recently developed to alleviate these deficiencies. This thesis explores and addresses a number of issues related to thin film synthesis on the one hand as well as component design and fabrication on other. Specifically, the growth of highly c-axis textured AlN thin films has been studied and optimized for achieving high device performance. Perhaps, one of the biggest achievements of the work is the development of a unique process for the deposition of AlN films with a mean c-axis tilt, which is of vital importance for the fabrication of resonators operating in contact with liquids, i.e. biochemical sensors. This opens the way for the development of a whole range of sensors and bio-analytical tools. Further, high frequency Lamb wave resonators have been designed, fabricated and evaluated. Performance enhancement of FBAR devices is also addressed, e.g. spurious mode suppression, temperature compensation, etc. It has been demonstrated, that even without temperature compensation, shear mode resonators operating in a liquid still exhibit an excellent performance in terms of Q (200) and coupling (~1.8%) at 1.2 GHz, resulting in a mass resolution better than 2 ng cm-2 in water, which excels that of today’s quartz sensors.

    • A method for the deposition of thin piezoelectric aluminum nitride (AlN) films with a nonzero c-axis mean tilt has been developed. The deposition is done in a standard reactive magnetron sputter deposition system without any hardware modifications. In essence, the method consists of a two-stage deposition process. The resulting film has a distinct tilted texture with the mean tilt of the c-axis varying roughly in the interval 28 to 32 degrees over the radius of the wafer excluding a small exclusion zone at the center of the latter. The mean tilt angle distribution over the wafer has a circular symmetry. A membrane-type shear mode thickness-excited thin film bulk acoustic resonator together with a micro-fluidic transport system has been subsequently fabricated using the two stage AIN deposition as well as standard bulk micro machining of Si. The resonator consisted of a 2-mu m-thick AlN film with 200-nm-thick A1 top and bottom electrodes. The resonator was characterized with a network analyzer when operating in both air and water. The shear mode resonance frequency was about 1.6 GHz, the extracted device Q around 350, and the electromechanical coupling k(t)(2) 2% when the resonator was operated in air, whereas the latter two dropped down to 150 and 1.8%, respectively, when the resonator was operated in pure water.

  • Temperature compensation of liquid FBAR sensors

    • In this work we demonstrate a practically complete temperature compensation of the second harmonic shear mode in a composite Al/AlN/Al/SiO2 thin film bulk acoustic resonator (FBAR) in the temperature range 25 °C–95 °C. The main advantages of this mode are its higher Q value in liquids as well as its higher frequency and hence higher resolution for sensor applications. For comparative reasons the non-compensated fundamental shear mode is also included in these studies. Both modes have been characterized when operated both in air and in pure water. Properties such as Q value, electromechanical coupling, dissipation and sensitivity are studied. An almost complete temperature compensation of the second harmonic shear mode was observed for an oxide thickness of 1.22 µm for an FBAR consisting of 2 µm thick AlN and 200 nm thick Al electrodes. Thus, the measured temperature coefficient of frequency (TCF) in air for the non-compensated fundamental shear mode (1.25 GHz) varied between −31 and −36 ppm °C−1 over the above temperature range while that of the compensated second harmonic shear mode (1.32 GHz) varied between +2 ppm °C−1 and −2 ppm °C−1 over the same temperature interval. When operated in pure water the former type shows a Q value and coupling coefficient, k2t, around 180 and 2%, respectively, whereas for the second harmonic these are 230 and 1.4%, respectively.

  • Thin film Lamb wave resonant structures - The first approach

    • Resonant Lamb wave based geometries on c-oriented aluminum nitride (AlN) thin film membranes are studied. Attention has been focused on the lowest order symmetric Lamb mode because of its extremely high velocity approaching 10,600 m/s. The Lamb waves have been excited by means of both inter-digital transducers (IDT) and longitudinal wave (LW) transducers. Two basic reflector geometries have been used along with the conventional one-port resonator topology. The experimental results obtained demonstrate that Lamb waves can be successfully used as an alternative to high velocity surface acoustic waves. A discussion regarding the ways for improving the device performance is provided, and hence future research aspects in the area are identified.

  • Tunable spring balanced magnetic energy harvester for low frequencies and small displacements

    • In this paper we present a novel concept to efficiently harvest vibrational energy at low frequencies and very small displacement. We describe and evaluate an electromagnetic energy harvester which generates power from a magnetic circuit with motion induced variations of an air gap. External vibrations induce oscillations of the gap length around an equilibrium point, due to a linear spring counteracting the magnetic force. The relative position of the spring can be adjusted to optimize the harvester output for excitation amplitude and frequency. A simulation model is built in COMSOL and verified by comparison with lab measurements. The simulation model is used to determine the potential performance of the proposed concept under both harmonic and non-harmonic excitation. Under harmonic excitation, we achieve a simulated RMS load power of 26.5 μW at 22 Hz and 0.028 g acceleration amplitude. From a set of comparable EH we achieve the highest theoretical power metric of 1712.2 µW/cm3/g2 while maintaining the largest relative bandwidth of 81.8%. Using measured non-harmonic vibration data, with a mean acceleration of 0.039 g, resulted in a mean power of 52 μW. Moreover, the simplicity and robustness of our design makes it a competitive alternative for use in practical situations.

  • Unified Modeling and Analysis of Vibration Energy Harvesters under Inertial Loads and Prescribed Displacements

    • In this paper, we extend the optimization analysis found in the current literature for single-degree-of-freedom vibrational energy harvesters. We numerically derive and analyze the optimization conditions based on unified expressions for piezoelectric and electromagnetic energy harvesters. Our contribution lies in the detailed analysis and comparison of both resonant and anti-resonant states while fully including the effect of intrinsic resistance. We include both the case of excitation by inertial load and prescribed displacement, as the latter has not been elaborated on in the previous literature and provides new insights. We perform a general analysis but also consider typical values of applied piezoelectric and electromagnetic energy harvesters. Our results improve upon previous similar comparative studies by providing new and useful insights regarding optimal load, load power and power input to output efficiency. Our analysis shows an exponential increase in the critical mechanical quality factor due to the resistive loss coefficient. We find that the ratio of mechanical quality factor to resistive loss coefficient, at resonance, increases drastically close to the theoretical maximum for load power. Under the same optimization conditions, an equivalent conclusion can be drawn regarding efficiency. We find that the efficiency at anti-resonance behaves differently and is equal to or larger than the efficiency at resonance. We also show that the optimal load coefficient at resonance has a significant dependence on the mechanical quality factor only when the resistive loss coefficient is large. Our comparison of excitation types supports the previous literature, in a simple and intuitive way, regarding optimal load by impedance matching and power output efficiency. Our modeling and exploration of new parameter spaces provide an improved tool to aid the development of new harvester prototypes.

  • Vibrational energy harvesting for sensors in vehicles

    • The miniaturization of semiconductor technology and reduction in power requirements have begun to enable wireless self-sufficient devices, powered by ambient energy. To date the primary application lies in generating and transmitting sensory data. The number of sensors and their applications in automotive vehicles has grown drastically in the last decade, a trend that seems to continue still. Wireless self-powered sensors can facilitate current sensor systems by removing the need for cabling and may enable additional applications. These systems have the potential to provide new avenues of optimization in safety and performance. This thesis delves into the topic of vibrations as ambient energy source, primarily for sensors in automotive vehicles. The transduction of small amounts of vibrational, or kinetic, energy to electrical power, also known as vibrational energy harvesting, is an extensive field of research with a plethora of inventions. A short review is given for energy harvesters, in an automotive context, utilizing transduction through either the piezoelectric effect or magnetic induction. Two practical examples, for ambient vibration harvesting in vehicles, are described in more detail. The first is a piezoelectric beam for powering a strain sensor on the engines rotating flexplate. It makes combined use of centrifugal force, gravitational pull and random vibrations to enhance performance and reduce required system size. The simulated power output is 370 µW at a rotation frequency of 10.5 Hz, with a bandwidth of 2.44 Hz. The second example is an energy harvesting unit placed on a belt buckle. It implements magnetic induction by the novel concept of a spring balance air gap of a magnetic circuit, to efficiently harvest minute vibrations. Simulations show the potential to achieve 52 µW under normal road conditions driving at 70 km/h. Theoretical modeling of these systems is also addressed. Fundamental descriptions of the lumped and distributed models are given. Based on the lumped models of the piezoelectric energy harvester (PEH) and the electromagnetic energy harvester (EMEH), a unified model is described and analyzed. New insights are gained regarding the pros and cons of the two types of energy harvester run at either resonance or anti-resonance. A numerical solution is given for the exact boundary of dimensionless quality factor and dimensionless intrinsic resistance, at which the system begins to exhibit anti-resonance. Regarding the maximum achievable power, the typical PEH is favored when running the system in anti-resonance and the typical EMEH is favored at resonance. The described modeling considers all parameters of the lumped model and thus provides a useful tool for developing vibrational energy harvester prototypes.

    • A novel microsomal prostaglandin E synthase 1 (mPGES-1) inhibitor induced kidney injury at exposures representing less than 4 times the anticipated efficacious exposure in man during a 7-day toxicity study in rats. The findings consisted mainly of tubular lesions and the presence of crystalline material and increases in plasma urea and creatinine. In vitro and in vivo metabolic profiling generated a working hypothesis that a bis-sulfonamide metabolite (determined M1) formed by amide hydrolysis caused this toxicity. To test this hypothesis, rats were subjected to a 7-day study and were administered the suspected metabolite and two low-potency mPGES-1 inhibitor analogs, where amide hydrolysis was undetectable in rat hepatocyte experiments. The results suggested that compounds with a reduced propensity to undergo amide hydrolysis, thus having less ability to form M1, reduced the risk of inducing kidney toxicity. Rats treated with M1 alone showed no histopathologic change in the kidney, which was likely related to underexposure to M1. To circumvent rat kidney toxicity, we identified a potent mPGES-1 inhibitor with a low propensity for amide hydrolysis and superior rat pharmacokinetic properties. A subsequent 14-day rat toxicity study showed that this compound was associated with kidney toxicity at 42, but not 21, times the anticipated efficacious exposure in humans. In conclusion, by including metabolic profiling and exploratory rat toxicity studies, a new and active mPGES-1 inhibitor with improved margins to chemically induced kidney toxicity in rats has been identified.

    • The thesis analyses the continuously changing discourse of cultural diversity. The focus of this study is the ways in which this discourse is shaped within contemporary Swedish cultural politics in general, and how it unfolds and further changes through the specific activities performed by a cultural institution in Stockholm – Södra teatern.Where as the cultural diversity inspired cultural politics seems to be caught in the tension between defending the autonomous position of culture in society and arguing for the supposed positive integrative consequences of such a perspective the Södra teatern mirrors a similar complexity in their ways of performing cultural diversity. This complexity is expressed through the three aspects of cultural diversity the thesis analyses – the real, the good and the cool cultural diversity. Each of the three aspects expresses a search for authenticity as well as a resistance towards the very same notion; a belief in the enlightenment of knowledge at the same time as the content of the very same knowledge is questioned; and finally that ethnic differences some times is enforced through utterances that stresses tradition where as the supposed hybrid forms in other situations is celebrated.The three aspects of cultural diversity analysed here all express how cultural diversity works as a hyper complex concept within cultural policy where no single possible meaning erases other potential meanings. On the contrary: different and even contradictory meanings of the concept exist side by side. Thus the thesis argues that the discourse of cultural diversity expresses a hyper complex problem.

    • Infantile malignant osteopetrosis (IMO) is caused by lack of functional osteoclasts leading to skeletal abnormalities, blindness due to compression of the optic nerves, bone marrow (BM) failure and early death. In most patients TCIRG1, a proton pump subunit essential for bone resorption, is mutated. Oc/oc mice represent a model for IMO due to a deletion in Tcirg1 and die around 4 weeks. To determine if hematopoietic stem cell transplantation without prior conditioning can reverse osteopetrosis, neonatal mice were transplanted iv with lineage depleted BM cells. More than 85% of oc/oc mice transplanted with 5 x 10(6) cells survived long term with an engraftment of 3-5% in peripheral blood (PB). At 3 w engraftment in the BM was 1-2% but the cellularity had increased 60-fold compared to non-treated oc/oc and RANKL and M-CSF expression in the BM was normalized. Histopathology and micro-CT revealed almost complete reversal of osteopetrosis after 4 weeks. In vitro studies showed that bone resorption by osteoclasts from transplanted oc/oc mice was 14% of transplanted controls and immunofluorescence microscopy revealed that resorption was mainly associated with osteoclasts of donor origin. Lineage analysis of BM, PB and spleen did not provide any evidence for selective recruitment of cells to the osteoclastic lineage. The vision was also preserved in transplanted oc/oc mice as determined by a visual tracking drum test. In summary, nonablative neonatal transplantation leading to engraftment of only a small fraction of normal cells rapidly reverses severe osteopetrosis in the oc/oc mouse model.     • The osteoclast is vital for establishment of normal hematopoiesis in the developing animal. However, its role for maintenance of hematopoiesis in adulthood is more controversial. To shed more light on this process, we transplanted hematopoietic stem cells from two osteopetrotic mouse models, with lack of osteoclasts or defective osteoclast function, to normal adult mice and examined the bone phenotype and hematopoiesis in the recipients. B6SJL mice were lethally irradiated and subsequently transplanted with oc/oc, Receptor Activator of Nuclear Factor Kappa B knockout or control fetal liver cells. Osteoclasts derived from the recipient animals were tested in vitro for osteoclastogenesis and resorptive function. Bone remodeling changes were assessed using biomarkers of bone tumrnover and micro-CT. Hematopoiesis was assessed by flow cytometry and colony formation, and hematopoietic stem cell function by secondary competitive transplantations and cell cycle analysis. After transplantation, a donor chimerism of 97-98% was obtained, and by 15 weeks mild osteopetrosis had developed in recipients of cells from osteopetrotic mice. There were no alterations in the number of bone marrow cells. Colony formation was slightly reduced in Receptor Activator of Nuclear Factor Kappa B knock-out recipients but unchanged in oc/oc recipients. Phenotypically, stem cells were marginally reduced in recipients of cells from osteopetrotic mice, but no significant difference was seen in cell cycle status and in competitive secondary transplantations all three groups performed equally well. Our results indicate that osteoclast function is not crucial for hematopoietic stem cell maintenance in adult mice.

The book is organized into ten chapters. The first two chapters introduce key ideas and theories. In the next four chapters, four large media circuits are scrutinized. The final four chapters discuss the results in terms of intermedial relations, time, space and power. The book thus offers a new foundation for understanding media use, consumption and the fate of urban public space in late modernity

    • This report discusses ideas of a radical increase in the significance and scope of culture in modern society, where industrialism is said to have been superseded by a society of information, knowledge and experiences. Issues of cultural identity and community are increasingly focal concerns, the virtual worlds of media culture seem omnipotent, and the cultural sector is growing with each of its recurrent redefinitions. Politics and the economy are supposedly aestheticised, culture and design boost regional development, creativity is a core value. The theses of revolutionary -culturalisation- of society and everyday life have a long history through the various cultural turns proclaimed by scholars as well as in society at large. The report scrutinises some main arguments and their implications, pointing at a need to distinguish historical trends, material processes and ideological discourses. A differentiation is made between five main dimensions of the concept of culture that are associated with specific sets of culturalisation ideas and corresponding trans-formations of boundaries around and within the cultural field. This discussion also touches upon the position and role of cultural research itself. Examples are given from five focal areas for these processes: academic work, regional politics, public spaces, history constructions and border struggles.

    • Osteopetrosis caused by defective acid secretion by the osteoclast, is characterized by defective bone resorption, increased osteoclast numbers, while bone formation is normal or increased. In contrast the bones are of poor quality, despite this uncoupling of formation from resorption. To shed light on the effect of uncoupling in adult mice with respect to bone strength, we transplanted irradiated three-month old normal mice with hematopoietic stem cells from control or oc/oc mice, which have defective acid secretion, and followed them for 12 to 28 weeks. Engraftment levels were assessed by flow cytometry of peripheral blood. Serum samples were collected every six weeks for measurement of bone turnover markers. At termination bones were collected for mu CT and mechanical testing. An engraftment level of 98% was obtained. From week 6 until termination bone resorption was significantly reduced, while the osteoclast number was increased when comparing oc/oc to controls. Bone formation was elevated at week 6, normalized at week 12, and reduced onwards. mu CT and mechanical analyses of femurs and vertebrae showed increased bone volume and bone strength of cortical and trabecular bone. In conclusion, these data show that attenuation of acid secretion in adult mice leads to uncoupling and improves bone strength.

    • Piezoelectric c-textured Al(1-x)ScxN thin films, where the Sc relative concentration, x, varies in the range 0-0.15 have been studied in view of radio frequency (RF) electro-acoustic applications. Thin film bulk acoustic wave resonators (FBARs) employing these films were fabricated and characterized as a function of the Sc concentration for the first time. The measured electromechanical coupling is found to increase by as much as 100% in the above concentration range. The results from this work underline the potential of the c-textured Al(1-x)ScxN based FBARs for wide band RF applications.

    • Wurtzite AlN is a piezoelectric material with excellent electro-acoustic properties and is used for the fabrication of high frequency thin film micro-acoustic components, most notably filters, duplexers, resonators, etc. Its moderate electromechanical coupling coefficient (k(t)(2)) of 6%-7% is insufficient for applications requiring larger bandwidths. Recent theoretical and experimental studies indicate that AlN alloyed with Sc exhibits a substantially higher piezoelectric constant than pure AlN. This study aims at determining the main electro-acoustic parameters of Al(1-x)Sc-(x) N in view of large bandwidth applications. To this end, highly c-textured Al(1-x)Sc(x) N thin films have been synthesized with relative Sc concentrations of up to 0,15. Subsequently, FBAR resonators were fabricated and characterized as a function of the Sc content. It is seen that k(t)(2) increases linearly with the latter to a value of 12% for a Sc concentration of x=0,15, while the Q value decreases from 739 to about 348 in the same concentration range. Likewise, the TCF varies from -35,9ppm/degrees C to -39,8ppm/degrees C, while the dielectric constant increases from epsilon=10 to a value of 14,1 for x=0,15. Finally, the relative dielectric losses are seen to increase by approximately a factor of two.
