    • The European Union (EU) initiative on the Digital Transformation of Health and Care (Digicare) aims to provide the conditions necessary for building a secure, flexible, and decentralized digital health infrastructure. Creating a European Health Research and Innovation Cloud (HRIC) within this environment should enable data sharing and analysis for health research across the EU, in compliance with data protection legislation while preserving the full trust of the participants. Such a HRIC should learn from and build on existing data infrastructures, integrate best practices, and focus on the concrete needs of the community in terms of technologies, governance, management, regulation, and ethics requirements. Here, we describe the vision and expected benefits of digital data sharing in health research activities and present a roadmap that fosters the opportunities while answering the challenges of implementing a HRIC. For this, we put forward five specific recommendations and action points to ensure that a European HRIC: i) is built on established standards and guidelines, providing cloud technologies through an open and decentralized infrastructure; ii) is developed and certified to the highest standards of interoperability and data security that can be trusted by all stakeholders; iii) is supported by a robust ethical and legal framework that is compliant with the EU General Data Protection Regulation (GDPR); iv) establishes a proper environment for the training of new generations of data and medical scientists; and v) stimulates research and innovation in transnational collaborations through public and private initiatives and partnerships funded by the EU through Horizon 2020 and Horizon Europe.

  • EMMA - the electric and magnetic monitor of the aurora on Astrid-2

    • The Astrid-2 mission has dual primary objectives. First, it is an orbiting instrument platform for studying auroral electrodynamics. Second, it is a technology demonstration of the feasibility of using micro-satellites for innovative space plasma physics research. The EMMA instrument, which we discuss in the present paper, is designed to provide simultaneous sampling of two electric and three magnetic field components up to about 1 kHz. The spin plane components of the electric field are measured by two pairs of opposing probes extended by wire booms with a separation distance of 6.7 m. The probes have titanium nitride (TiN) surfaces. which has proved to be a material with excellent properties for providing good electrical contact between probe and plasma. The wire booms are of a new design in which the booms in the stowed position are wound around the exterior of the spacecraft body. The boom system was flown for the first time on this mission and worked flawlessly. The magnetic field is measured by a tri-axial fluxgate sensor located at the tip of a rigid. hinged boom extended along the spacecraft spin axis and facing away from the Sun. The new advanced-design fluxgate magnetometer uses digital signal processors for detection and feedback, thereby reducing the analogue circuitry to a minimum. The instrument characteristics as well as a brief review of the science accomplished and planned are presented.

  • MEFISTO - An electric field instrument for BepiColombo/MMO

    • MEFISTO, together with the companion instrument WPT, are planning the first-ever in situ measurements of the electric field in the magnetosphere of planet Mercury. The instruments have been selected by JAXA for inclusion in the BepiColombo/MMO payload, as part of the Plasma Wave Investigation coordinated by Kyoto University. The magnetosphere of Mercury was discovered by Mariner 10 in 1974 and will be studied further by Messenger starting in 2011. However, neither spacecraft did or will measure the electric field. Electric fields are crucial in the dynamics of a magnetosphere and for the energy and plasma transport between different regions within the magnetosphere as well as between the magnetosphere and the surrounding regions. The MEFISTO instrument will be capable of measuring electric fields from DC to 3 MHz, and will thus also allow diagnostics of waves at all frequencies of relevance to the Hermean magnetosphere. MEFISTO is a double-probe electric field instrument. The double-probe technique has strong heritage and is well proven on missions such as Viking, Polar, and Cluster. For BepiColombo, a newly developed deployment mechanism is planned which reduces the mass by a factor of about 5 compared to conventional mechanisms for 15 in long booms. We describe the basic characteristics of the instrument and briefly discuss the new developments made to tailor the instrument to flight in Mercury orbit.

    • The BepiColombo Mercury Magnetospheric Orbiter (MMO) spacecraft includes the plasma and radio wave observation system called Plasma Wave Investigation (PWI). Since the receivers for electric field, plasma waves, and radio waves are not installed in any of the preceding spacecraft to Mercury, the PWI will provide the first opportunity for conducting in-situ and remote-sensing observations of electric fields, plasma waves, and radio waves in the Hermean magnetosphere and exosphere. These observations are valuable in studying structure, dynamics, and energy exchange processes in the unique magnetosphere of Mercury. They are characterized by the key words of the non-MHD environment and the peculiar interaction between the relatively large planet without ionosphere and the solar wind with high dynamic pressure. The PWI consists of three sets of receivers (EWO, SORBET, and AM(2)P), connected to two sets of electric field sensors (MEFISTO and WPT) and two kinds of magnetic field sensors (LF-SC and DB-SC). The PWI will observe both waveforms and frequency spectra in the frequency range from DC to 10 MHz for the electric field and from 0.3 Hz to 640kHz for the magnetic field. From 2008, we will start the development of the engineering model, which is conceptually consistent with the flight model design. The present paper discusses the significance and objectives of plasma/radio wave observations in the Hermean magnetosphere, and describes the PWI sensors, receivers and their performance as well as the onboard data processing.

  • Interplanetary magnetic field control of theta aurora development

    • [1] We ascertain the influence of the B-y component of the interplanetary magnetic field (IMF) on theta aurora evolution. During most cases where a transpolar arc is observed to move across the polar region, and form a theta aurora, there are brief (minutes) southward excursions of IMF B-z, however northward IMF is required prior to theta aurora formation. Observations show that theta aurora can form during strictly northward IMF with its motion consistent with a change in sign of IMF B-y. It is important to note that since transpolar arcs can persist for 20-30 min after the IMF turns southward, errors will occur in assigning instantaneous IMF conditions to snapshots'' of particular auroral patterns. We consider the entire evolution of the theta aurora and the changing IMF conditions. The influence of IMF B-y is best illustrated by examples which occur during steady northward IMF as compared to times when the IMF is northward on average. We show examples, provided by the Polar UV imager, when the IMF is steady northward. For one case, DMSP F13 and F14 provide in situ measurements of precipitating particles, ionospheric plasma flows and ion density. This unique data set enables us to analyze in detail the evolution of a theta aurora, in one case crossing the entire polar region. No sign change in B-z is needed for theta aurora formation.

    • Cardiopulmonary diseases are major causes of death worldwide, but currently recommended strategies for diagnosis and prevention may be outdated because of recent changes in risk factor patterns. The Swedish CArdioPulmonarybioImage Study (SCAPIS) combines the use of new imaging technologies, advances in large-scale 'omics' and epidemiological analyses to extensively characterize a Swedish cohort of 30 000 men and women aged between 50 and 64 years. The information obtained will be used to improve risk prediction of cardiopulmonary diseases and optimize the ability to study disease mechanisms. A comprehensive pilot study in 1111 individuals, which was completed in 2012, demonstrated the feasibility and financial and ethical consequences of SCAPIS. Recruitment to the national, multicentre study has recently started.

    • The Rosetta dual Langmuir probe instrument, LAP, utilizes the multiple powers of a pair of spherical Langmuir probes for measurements of basic plasma parameters with the aim of providing detailed knowledge of the outgassing, ionization, and subsequent plasma processes around the Rosetta target comet. The fundamental plasma properties to be studied are the plasma density, the electron temperature, and the plasma flow velocity. However, study of electric fields up to 8 kHz, plasma density fluctuations, spacecraft potential, integrated UV flux, and dust impacts is also possible. LAP is fully integrated in the Rosetta Plasma Consortium (RPC), the instruments of which together provide a comprehensive characterization of the cometary plasma.

    • The mutual location of high-latitude electrojets, typical regions of the auroral luminosity and regions of auroral energy particle participations into the upper atmosphere under substorm conditions are considered. Three electrojets exist at high latitudes during substorm intervals: WE - westward electrojet, EE - eastward electrojet and PE – polar electrojet. Geomagnetic latitudes of the WE/EE and PE location vary depend on local time and magnetic activity level, respectively. It is shown that the WE is located within the limits of the auroral oval precipitation (AOP), the EE in the evening sector is located within the diffuse auroral zone (DAZ) and the PE near noon is located at the poleward AOP boundary shifting poleward with decreasing the magnetic activity level. The relationship of electrojets with different plasma domains in the magnetosphere is discussed.

  • Structure of the auroral precipitation region in the dawn sector : relationship to convection reversal boundaries and field-aligned currents

    • Simultaneous DMSP F7 and Viking satellite measurements of the dawnside high-latitude auroral energy electron and ion precipitation show that the region of the low and middle altitude auroral precipitation consists of three characteristic plasma regimes. The recommendation of the IAGA Working Group IIF/III4 at the IAGA Assembly in Boulder, July 1995 to decouple the nomenclature of ionospheric populations from magnetospheric population is used for their notation. The most equatorial regime is the Diffuse Auroral Zone (DAZ) of diffuse spatially unstructured precipitating electrons. It is generated by the plasma injection to the inner magnetosphere in the nightside and the subsequent drift plasma to the dawnside around the Earth. Precipitating par tides have a hard spectrum with typical energies of electrons and ions of more than 3 keV. In the DAZ, the ion pitch-angle distribution is anisotropic. with the peak near 90 degrees. The next part is the Auroral Oval (AO), a structured electron regime which closely resembles the poleward portion of the nightside auroral oval. The typical electron energy is several keV, and the ion energy is up to 10 keV. Ion distributions are predominantly isotropic. In some cases, this plasma regime may be absent in the prenoon sector. Poleward of the Auroral Oval, there is the Soft Small Scale Luminosity (SSSL) regime. It is caused by structured electron and ion precipitation with typical electron energy of about 0.3 keV and ion energy of about 1 keV. The connection of these low-altitude regimes with plasma domains of the distant magnetosphere is discussed. For mapping of the plasma regimes to the equatorial plane of the magnetosphere, the empirical model by Tsyganenko (1995) and the conceptual model by Alexeev et al. (1996) are used. The DAZ is mapped along the magnetic field lines to the Remnant Layer (RL), which is located in the outer radiation belt region: the zone of structured electrons and isotropic ion precipitation (AO) is mapped to the dawn periphery of the Central Plasma Sheet (CPS); the soft small scale structured precipitation (SSSL) is mapped to the outer magnetosphere close to the magnetopause, i.e. the Low Latitude Boundary Layer (LLBL). In the near-noon sector, earthward fluxes of soft electrons, which cause the Diffuse Red Aurora (DRA), are observed. The ion energies decrease with increasing latitude, The plasma spectra of the DRA regime are analogous to the spectra of the Plasma Mantle (PM). In the dawn sector, the large-scale field-aligned currents flow into the ionosphere at the SSSL latitudes (Region 1) and flow out at the AO or DAZ latitudes (Region 2). In the dawn and dusk sectors, the large-scale Region 1 and Region 2 FAC generation occurs in different plasma domains of the distant magnetosphere. The dawn and dusk FAC connection to the traditional Region 1 and Region 2 has only formal character, as FAC generating in various magnetospheric plasma domains integrate in the same region (Region 1 or Region 2). In the SSSL, there is anti-sunward convection in the DAZ and the AO, there is the sunward convection. At PM latitudes, the convection is controlled by the azimuthal IMF component (By) It is suggested to extend the notation of the plasma pattern boundaries, as proposed by Newell et al. (1996), for the nightside sector of the auroral oval to the dawn sector.

  • The double probe electric field experiment on Freja : Experiment description and first results

    • A description is given of the Freja double-probe electric field instrument. Its capability to perform high-resolution measurements of the aurora and its fine-structure as well as collect information on sub-auroral and low-latitude phenomena is illustrated by selected results from the first six months of operation. The instrument is highly flexible and possible to operate in a number of different modes. It is also equipped with a 4-Megabyte burst memory for high data sampling rate and temporary storage of data. It has been fully operational since October 1992, and delivers data from ≈22 hr day-1 including about 5-6 auroral crossings of the northern and southern auroral ionosphere. New and important information on the auroral fine structure and electrodynamics is obtained by means of burst resolution data (6144 samples s-1) and normal resolution data (768 samples s-1). Common burst data collection triggered by the electric field event detector has turned out to be very useful for the selection of scientifically interesting events. This is illustrated by high-resolution data of a pair of extremely intense and narrow electric field structures (1 V m-1) which are associated with a total absence of precipitating particles, depletions of the thermal plasma and with an intense wave activity. The low inclination of the Freja orbit provides a new perspective for studying largescale phenomena associated with east-west gradients as is exemplified by electric field data from a satellite crossing over north-south oriented auroral structures presumably resulting from rotational distortions of east-west aligned auroral arcs. The different plasma regimes encountered by Freja are continuously monitored by means of current sweeps applied to the probes and by the satellite potential. In addition, overview data (8 samples s-1) are collected from full orbits and stored in the on-board memory and have proved to be extremely valuable, providing new information on global electric field phenomena at subauroral and lower latitudes, such as the intense poleward electric fields and Pc-1 observations that have been made near the plasmapause during substorm activity. © 1994 Kluwer Academic Publishers.

    • The present study examines a sawtooth injection event that took place around 0800 UT on 18 April 2002 when the Cluster spacecraft were located in the inner magnetosphere in the premidnight sector. In association with this injection, Cluster, at a radial distance of 4.6 RE, observed that the local magnetic field became more dipolar and that both ion and electron fluxes increased without notable energy dispersion. These features were accompanied by intensifications of the equatorward component of a double- oval structure and also by an enhancement of the ring- current oxygen ENA flux. The event was also accompanied by large magnetic field ( a few tens of nT) and electric field ( a few tens of mV/ m) fluctuations with characteristic timescales of a few tens of seconds. These observations strongly suggest that this sawtooth injection extended not only widely in local time but also deeply into the inner magnetosphere. Interestingly, Cluster repeatedly observed dipolarization- like signatures afterward, which, however, were not associated with enhancements of local energetic ion flux or with geosynchronous dipolarization or injection signatures. Instead, these magnetic signatures were accompanied by oscillatory plasma motion in the radial direction with a characteristic timescale of about 10 min, which appears to be related to the westward propagation of a spatially periodic auroral structure. The associated azimuthal electric field component was well correlated with the time derivative of the north- south magnetic field component, suggesting that the observed electric field is inductive. These findings suggest that electromagnetic processes far inside geosynchronous orbit play an important role in energization of energetic ions and auroral dynamics during magnetospheric storms.

    • The thermal component of Mercury's electron population has never been measured. One scientific objective of the Plasma Wave Investigation consortium, PWI, is to determine the influence of the thermal plasma upon the formation and dynamics of the planetary magnetosphere, as a function of solar activity. The Active Measurement of Mercury's Plasma experiment, AM(2)p, has been proposed as part of PWI, to monitor the density and temperature of the thermal electron population, during the whole mission of the Mercury Magnetospheric Orbiter of BepiColombo. These two physical parameters will be deduced from the measurements of the self- and mutual-impedances of the MEFISTO (Mercury Electric Field In Situ Tool) double-sphere antenna, in a frequency range comprising the expected plasma frequency. The in situ measurement of the antenna impedance is also essential for calibrating the electric antenna which measures the natural waves; it will allow, in particular, the effective length of the antenna to be calculated as a function of frequency and plasma conditions. The purpose of this paper is to define the scientific objectives of AM(2)p, to explain the principle of the measurement, to describe the electronic device, and to show the ability of AM 2p to make reliable and accurate measurements of the thermal plasma density and temperature in the Hermean magnetosphere, as well as in the solar wind at heliocentric distances of 0.31-0.47 AU. The potential performance of this instrument has been evaluated using both an analytical approach and numerical simulations.

  • Nonlinear kinetic Alfven wave with Poisson equation correction in the low aurora

    • Nonlinear kinetic Alfven waves where beta much less than m(e)/m(i), have been solved both with and without the Poisson equation correction. It is found that the ratio of the perpendicular electric field and magnetic field, and the ratio of parallel and perpendicular electric field increase with deepening of the depressive density soliton. The former ratio may be larger than the Alfven velocity in the case of a large amplitude solitary kinetic Alfven wave. The Poisson equation correction is important for the nonlinear kinetic Alfven wave propagating along the magnetic field, which solves a puzzle of Sagdeev potential to approach infinity in the limit of K-x --> 0. This correction causes the solitary KAW possessing an electrostatic character along the direction of wave moving frame. These results have been compared with the observations from the Freja satellite in the low aurora.

    • The excellent performance of a new sample preparation method, microextraction in packed syringe (MEPS), was recently illustrated by online LC‐MS and GS‐MS assays of local anaesthetics in plasma samples. In the method, approximately 1 mg of solid packing material was inserted into a syringe (100–250 µL) as a plug. Sample preparation took place on the packed bed. The new method was easy to use, fully automated, of low cost, and rapid in comparison with previously used methods. This paper presents the use of molecularly imprinted polymers (MIPs) as packing material for higher extraction selectivity. Development and validation of a method for MIP‐MEPS online with LC‐MS‐MS using ropivacaine in plasma as model compound were investigated. A bupivacaine imprinted polymer was used. The method was validated and the standard curves were evaluated by means of quadratic regression and weighted by inverse of the concentration: 1/x for the calibration range 2–2000 nM. The applied polymer could be used more than 100 times before the syringe was discarded. The extraction recovery was 60%. The results showed high correlation coefficients (R 2 >0.999) for all runs. The accuracy, given as a percentage deviation from the nominal concentration values, ranged from -6% to 3%. The precision, given as the relative standard deviation, at three different concentrations (QC samples) was consistently about 3% to 10%. The limit of quantification was 2 nM.

    • We study the topological structure of the magnetosphere for northward IMF. Using a magnetospheric magnetic field model we study the high-latitude response to prolonged periods of northward IMF. For forced solar wind conditions we investigate the location of the polar cap region, the polar cap potential drop, and the field-aligned acceleration potentials, depending on the solar wind pressure and IMF B-y and B-x changes. The open field line bundles, which connect the Earth's polar ionosphere with interplanetary space, are calculated. The locations of the magnetospheric plasma domains relative to the polar ionosphere are studied. The specific features of the open field line regions arising when IMF is northward are demonstrated.   • New Techniques for Sample Preparation in Analytical Chemistry : Microextraction in Packed Syringe (MEPS) and Methacrylate Based Monolithic Pipette Tips

    • Sample preparation is often a bottleneck in systems for chemical analysis. The aim of this work was to investigate and develop new techniques to address some of the shortcomings of current sample preparation methods. The goal has been to provide full automation, on-line coupling to detection systems, short sample preparation times and high-throughput. In this work a new technique for sample preparation that can be connected on-line to liquid chromatography (LC) and gas chromatography (GC) has been developed. Microextraction in packed syringe (MEPS) is a new solid-phase extraction (SPE) technique that is miniaturized and can be fully automated. In MEPS approximately 1 mg of sorbent material is inserted into a gas tight syringe (100-250 μL) as a plug. Sample preparation takes place on the packed bed. Evaluation of the technique was done by the determination of local anaesthetics in human plasma samples using MEPS on-line with LC and tandem mass spectrometry (MS-MS). MEPS connected to an autosampler was fully automated and clean-up of the samples took about one minute. In addition, in the case of plasma samples the same plug of sorbent could be used for about 100 extractions before it was discarded. A further aim of this work was to increase sample preparation throughput. To do that disposable pipette tips were packed with a plug of porous polymer monoliths as sample adsorbent and were then used in connection with 96-well plates and LC-MS-MS. The evaluation of the methods was done by the analysis of local anaesthetics lidocaine and ropivacaine, and anti-cancer drug roscovitine in plasma samples. When roscovitine and lidocaine in human plasma and water samples were used as model substances, a 96-plate was handled in about two minutes. Further, disposable pipette tips may be produced at low cost and because they are used only once, carry-over is eliminated.

    • There is a need for potent invertebrate systems for assessing the impacts of environmental contaminants on marine ecosystems. Balanus improvisus, a marine athropod, has a number of promising characteristics that make it a good candidate in such efforts. We have conducted sequencing of a cDNA library from the cyprid larval stage and identified several detoxification systems as well as novel B. improvisus specific genes. To investigate the toxicological gene expression response in this organism, we performed a short-term exposure experiment of the cyprid larvae to two different biofouling substances. From a natural population of B. improvisus, 300 - 1000 cyprids were treated for 23 hours with 390nM CuO or with two different concentrations (10nM or 10μM) of meditomidine. Protein expression changes were studied by 2D-PAGE analysis after DIGE labelling. For gene expression analysis by DNA miroarrays total RNA was extracted and used for cDNA and cRNA/aRNA templates. Roughly 2000 B. improvisus genes were studied represented by 3000 different probes on the arrays (each in duplicates). Candidate genes were confirmed by qPCR. A number of protein expression changes were detected on the 2D gels as a result of the different treatments. Interestingly, the response to the different treatments clearly formed distinct groups in principle component analysis. The DNA microarray analysis revealed several genes as toxicity indicators, e.g. various heat shock proteins, some proteases and factors involved in regulatory processes (transcription factors). Our data indicate that B. improvisus may serve as a tool to evaluate the impacts of marine pollution, and thus to fill the niche as an important invertebrate marine model organism for ecotoxicology and environmental genomics.
