    • The Large Observatory For x-ray Timing (LOFT) was studied within ESA M3 Cosmic Vision framework and participated in the final downselection for a launch slot in 2022-2024. Thanks to the unprecedented combination of effective area and spectral resolution of its main instrument, LOFT will study the behaviour of matter under extreme conditions, such as the strong gravitational field in the innermost regions of accretion flows close to black holes and neutron stars, and the supranuclear densities in the interior of neutron stars. The science payload is based on a Large Area Detector (LAD, 10 m2 effective area, 2-30 keV, 240 eV spectral resolution, 1° collimated field of view) and a Wide Field Monitor (WFM, 2-50 keV, 4 steradian field of view, 1 arcmin source location accuracy, 300 eV spectral resolution). The WFM is equipped with an on-board system for bright events (e.g. GRB) localization. The trigger time and position of these events are broadcast to the ground within 30 s from discovery. In this paper we present the status of the mission at the end of its Phase A study.

    • The LOFT mission concept is one of four candidates selected by ESA for the M3 launch opportunity as Medium Size missions of the Cosmic Vision programme. The launch window is currently planned for between 2022 and 2024. LOFT is designed to exploit the diagnostics of rapid X-ray flux and spectral variability that directly probe the motion of matter down to distances very close to black holes and neutron stars, as well as the physical state of ultradense matter. These primary science goals will be addressed by a payload composed of a Large Area Detector (LAD) and a Wide Field Monitor (WFM). The LAD is a collimated (<1 degree field of view) experiment operating in the energy range 2-50 keV, with a 10 m2 peak effective area and an energy resolution of 260 eV at 6 keV. The WFM will operate in the same energy range as the LAD, enabling simultaneous monitoring of a few-steradian wide field of view, with an angular resolution of <5 arcmin. The LAD and WFM experiments will allow us to investigate variability from submillisecond QPO's to yearlong transient outbursts. In this paper we report the current status of the project.

  • Precarious employment and occupational accidents and injuries - a systematic review

    • Objectives Precarious employment conditions have become more common in many countries over the last decades, and have been linked to various adverse health outcomes. The objective of this review was to collect and summarize existing scientific research of the relationship between dimensions of precarious employment and the rate of occupational injuries. Methods A protocol was developed in accordance with the PRISMA-P checklist for systematic literature reviews. We searched PubMed, Web of Science and Scopus for articles on observational studies from North America, Europe, Australia and New Zealand published in peer-reviewed journals 1990-2017. A minimum of two independent reviewers assessed each article with respect to quality and eligibility criteria. Articles of high/moderate quality meeting all specified inclusion criteria were included in the review. Results The literature search resulted in 471 original titles, of which 17 articles met all the inclusion criteria. The most common exposures were in descending order; temporary employment, multiple jobs, working for a subcontractor at the same worksite/temp agency, part-time, self-employment, hourly pay, union membership, insurance benefits, flexible versus fixed work schedule, wages, job insecurity, work-time control and precarious career trajectories. Ten studies reported a positive association between precarious employment and occupational injuries. Four studies reported a negative association, and three studies did not show any significant association. Conclusions This review supports an association between some of the dimensions of precarious employment and occupational injuries; most notably for multiple jobholders and employees of temp agencies or subcontractors at the same worksite. However, results for temporary employment are inconclusive. There is a need for more prospective studies of high quality, designed to measure effect sizes as well as causality.

  • Precarious employment and mental health: a systematic review and meta-analysis of longitudinal studies

    • Objectives Precarious employment (PE) is a term used to describe non-standard employment forms characterized by low security that may have negative effects on mental health. The objective of this review was to systematically review the evidence for effects of PE on mental health and identify important areas for further research. Methods A protocol was developed following PRISMA-P guidelines. Web of Science, PubMed and PsycINFO were searched up to 4 September 2017. All unique records were assessed for eligibility and quality by at least two reviewers. Data from included studies were summarized in forest plots and meta-analyses using a random-effects model. Evidence quality was rated using the GRADE method. Results We obtained 3328 unique records, of which 16 studies of sufficient quality met the inclusion criteria. Moderate quality evidence (GRADE score 3 of 4) was found for an adverse effect of job insecurity on mental health; summary odds ratio (OR) 1.52 [95% confidence interval (CI) 1.35-1.70]. There was very low quality (GRADE 1 of 4) evidence for effects of temporary employment or unpredictable work hours on mental health. Five studies on multidimensional exposures all showed adverse effects, weighted average OR 2.01 (95% CI 1.60-2.53). Conclusions Research on PE and mental health is growing, but high-quality prospective studies are still scarce. Job insecurity likely has an adverse effect on mental health. A clear multi-dimensional definition of PE is lacking, and harmonization efforts are needed. Further single-variable observational studies on job insecurity or temporary employment should not be prioritized.

    • Complex networks of relationships among and between people and nature (social-ecological networks) play an important role in sustainability; yet, we have limited empirical understanding of their temporal dynamics.We empirically examine the evolution of a social-ecological network in a common-pool resource system faced with escalating social and environmental change over the past two decades.We first draw on quantitative and qualitative data collected between 2002 and 2018 in a Papua New Guinean reef fishing community to provide contextual evidence regarding the extent of social and environmental change being experienced. We then develop a temporal multilevel exponential random graph model using complete social-ecological network data, collected in 2016 and 2018, to test key hypotheses regarding how fishing households have adapted their social ties in this context of change given their relationships with reef resources (i.e. social-ecological ties). Specifically, we hypothesized that households will increasingly form tight-knit, bonding social and social-ecological network structures (H1 and H3, respectively) with similar others (H2), and that they will seek out resourceful actors with specialized knowledge that can promote learning and spur innovation (H4).Our results depict a community that is largely ‘bunkering down’ and looking inward in response to mounting risk to resource-dependent livelihoods and a breakdown in the collaborative processes that traditionally sustained them. Community members are increasingly choosing to interact with others more like themselves (H2), with friends of friends (H1), and with those connected to interdependent ecological resources (H3)—in other words, they are showing a strong, increasing preference for forming bonding social-ecological network structures and interacting with like-minded, similar others. We did not find strong support for H4.Bonding network structures may decrease the risk associated with unmonitored behaviour and help to build trust, thereby increasing the probability of sustaining cooperation over time. Yet, increasing homophily and bonding ties can stifle innovation, reducing the ability to adapt to changing conditions. It can also lead to clustering, creating fault lines in the network, which can negatively impact the community's ability to mobilize and agree on/enforce social norms, which are key for managing common resources.

  • Sexual and gender harassment in Swedish workplaces: A prospective cohort study on implications for long-term sickness absence

    • Objectives This prospective cohort study aimed to investigate gender harassment and sexual harassment as risk factors for prospective long-term sickness absence (LTSA, >= 21 days). Furthermore, support from colleagues was investigated as a moderating factor of this association. Methods Information on gender harassment, sexual harassment and support by colleagues were derived from the biannual Swedish Work Environment Survey 1999-2013, a representative sample of the Swedish working population (N=64 297). Information on LTSA as well as demographic and workplace variables were added from register data. Relative rates of LTSA the year following the exposure were determined using modified Poisson regression. Results Monthly to daily exposure to gender harassment was a risk factor for prospective LTSA among women [rate ratio (RR) 1.04, 95% confidence interval (CI) 1.02-1.05] and men (RR 1.07, 95% CI 1.04-1.10). Monthly to daily exposure to sexual harassment was also a risk factor for LTSA among women (RR 1.05, 95% CI 1.01-1.10) and men (RR 1.07, 95% CI 1.02-1.13). Exposure to sexual or gender harassment once in the last 12 months was not associated with LTSA. There was no support for an interaction between either of the exposures and support from colleagues in relation to LTSA. Conclusions Sexual harassment and gender harassment appear to contribute to a small excess risk for LTSA among women and men. For both kinds of offensive behaviors, the pervasiveness appears to be important for the outcome. The role of support by colleagues was inconclusive and needs further investigation.

  • Nasal hyperresponders and atopic subjects report different symptom intensity to air quality : a climate chamber study

    • Short-term exposure to dust and dust added with beta-(1,3)-d-glucan or aldehydes may cause sensory reactions. In random order, we exposed 36 volunteers in a climate chamber to clean air, office dust, dust with glucan, and dust with aldehydes. Three groups of subjects were exposed, eleven were non-atopic with nasal histamine hyperreactivity, 13 were non-atopic, and 12 were atopic. Subjective ratings of symptoms and general health were registered four times during four 6-h exposure sessions. Six symptom intensity indices were constructed. The nasal hyperreactive group had a high and time-dependent increase of mucous membrane irritations, whereas the atopic group had a low and stable rate of irritations with exposure time, close to the reference group (P = 0.02 for differences between the groups with respect to time under exposure for Weak Inflammatory Responses and P = 0.05 for Irritative Body Perception, significance mainly because of the nasal hyperreactive group). Exposure to dust, with or without glucan or aldehydes, showed increased discomfort measured by the index for Constant Indoor Climate, and dust with glucan had a similar effect for the index for Lower Respiratory Effects. For Psychological and Neurological Effects these were dependent on group affiliation, thus preventing a uniform statement of exposure effects for all three investigated groups.Opportunities for identifying persons with high or low sensitivity to low-level exposures are important in preventive medicine and will reduce intra-group variability and thus increase the power of experimental and epidemiological studies searching for correlations between exposures and health effects. The contrast between nasal hyperreactive on one side and atopic and reference subjects on the other side is particularly important. The atopic group indicated a non-homogenous reaction depending on their hyperreactive status, a finding that could be important but needs further confirmation.

  • Accounting for interannual variability in agricultural intensification : The potential of crop selection in Sub-Saharan Africa

    • Providing sufficient food for a growing global population is one of the fundamental global challenges today. Crop production needs not only to be increased, but also remain stable over the years, in order to limit the vulnerability of producers and consumers to inter-annual weather variability, especially in areas of the world where the food consumed is mainly produced locally (e.g. Sub Saharan Africa (SSA)). For subsistence agriculture, stable yields form a crucial contribution to food security. At a regional to global scale dynamical crop models can be used to study the impact of future changes in climate on food production. However, simulations of future crop production, for instance in response to climate change, often do not take into account either changes in the sown areas of crops or yield interannual variability. Here, we explore the response of simulated crop production to assumptions of crop selection, also taking into account interannual variability in yields and considering the response of agricultural productivity to climate change. We apply the dynamic global vegetation model LPJ-GUESS, which is designed to simulate yield over large regions under a changing environment. Model output provides the basis for selecting the relative fractions of sown areas of a range of crops, either by selecting the highest yielding crop, or by using an optimization approach in which crop production is maximized while the standard deviation in crop production is kept at below current levels. Maximizing simulated crop production for current climate while keeping interannual variability in crop production constant at today's level generates rather similar simulated geographical distributions of crops compared to observations. Even so, the optimization results suggest that it is possible to increase crop production regionally by adjusting crop selection, both for current and future climate, assuming the same cropland cover as today. For future climates modelled production increase is > 25% in more than 15% of the grid cells. For a small number of grid cells it is possible to both increase crop production while at the same time decreasing its interannual variability. Selecting the highest yielding crop for any location will lead to a large potential increase in mean food production, but at the cost of a very large increase in variability.

    • Background Chronic heat stress and dehydration from strenuous work in hot environments is considered an essential component of the epidemic of chronic kidney disease in Central America. Objective (1) To assess feasibility of providing an intervention modelled on OSHA's Water. Rest. Shade programme (WRS) during sugarcane cutting and (2) to prevent heat stress and dehydration without decreasing productivity. Methods Midway through the 6-month harvest, the intervention introduced WRS practices. A 60-person cutting group was provided water supplied in individual backpacks, mobile shaded rest areas and scheduled rest periods. Ergonomically improved machetes and efficiency strategies were also implemented. Health data (anthropometric, blood, urine, questionnaires) were collected preharvest, preintervention, mid-intervention and at the end of harvest. A subsample participated in focus group discussions. Daily wet bulb globe temperatures (WBGT) were recorded. The employer provided individual production records. Results Over the harvest WBGT was >26 degrees C from 9:00 onwards reaching average maximum of 29.3 +/- 1.7 degrees C, around 13:00. Postintervention self-reported water consumption increased 25%. Symptoms associated with heat stress and with dehydration decreased. Individual daily production increased from 5.1 to a high of 7.3 tons/person/day postintervention. This increase was greater than in other cutting groups at the company. Focus groups reported a positive perception of components of the WRS, and the new machete and cutting programmes. Conclusions A WRS intervention is feasible in sugarcane fields, and appears to markedly reduce the impact of the heat stress conditions for the workforce. With proper attention to work practices, production can be maintained with less impact on worker health.

  • Improving network approaches to the study of complex social–ecological interdependencies

    • Achieving effective, sustainable environmental governance requires a better understanding of the causes and consequences of the complex patterns of interdependencies connecting people and ecosystems within and across scales. Network approaches for conceptualizing and analysing these interdependencies offer one promising solution. Here, we present two advances we argue are needed to further this area of research: (i) a typology of causal assumptions explicating the causal aims of any given network-centric study of social–ecological interdependencies; (ii) unifying research design considerations that facilitate conceptualizing exactly what is interdependent, through what types of relationships and in relation to what kinds of environmental problems. The latter builds on the appreciation that many environmental problems draw from a set of core challenges that re-occur across contexts. We demonstrate how these advances combine into a comparative heuristic that facilitates leveraging case-specific findings of social–ecological interdependencies to generalizable, yet context-sensitive, theories based on explicit assumptions of causal relationships.

    • Objectives Organic dust is associated with adverse effects on human airways. This study was done to investigate whether the addition of β-(1,3)-D glucan or aldehydes to office dust causes enhanced inflammation in human airways. Methods Thirty-six volunteers were exposed randomly to clean air, office dust, dust spiked with glucan, and dust spiked with aldehydes. The three dust exposures contained between 332 and 379 µg dust/m3. Spiking with 1 gram of dust was done with 10 milligrams of glucan or 0.1 microliters of aldehydes. Acoustic rhinometry, rhinostereometry, nasal lavage, and lung function tests were applied. Results After the exposures to dust spiked with the glucan and aldehydes, the nasal volume decreased (–1.33 and –1.39 cm3 (mean), respectively) when compared with the –0.9 cm3 after clean air or office dust (P=0.036 for a difference in decrease between exposures). After 2–3 hours the aldehyde-spiked dust caused a 0.6-mm swelling of the inferior turbinate, and glucan-spiked dust produced a 0.7-mm swelling (P=0.039 for a difference in the swelling between the four exposures). The preexposure nasal lavage cleaned off the mucosa, and lower cytokine concentrations were found after all of the exposures. For interleukin-8, this decrease in concentration was smaller after the dust exposures spiked with glucan and aldehydes (–2.9 and –25.8 pg/ml, respectively) than after office dust or clean air (–65.9 and –74.1 pg/ml, respectively) (P=0.042). The nasal eosinophil cell concentration increased after exposure to dust spiked with glucan (P=0.045). Conclusions β-(1,3)-D glucan and aldehydes in office dust enhance the inflammatory effects of dust on the upper airways

    • Coral reefs support the livelihood of millions of people especially those engaged in marine fisheries activities. Coral reefs are highly vulnerable to climate change induced stresses that have led to substantial coral mortality over large spatial scales. Such climate change impacts have the potential to lead to declines in marine fish production and compromise the livelihoods of fisheries dependent communities. Yet few studies have examined social vulnerability in the context of changes specific to coral reef ecosystems. In this paper, we examine three dimensions of vulnerability (exposure, sensitivity, and adaptive capacity) of 29 coastal communities across five western Indian Ocean countries to the impacts of coral bleaching on fishery returns. A key contribution is the development of a novel, network-based approach to examining sensitivity to changes in the fishery that incorporates linkages between fishery and non-fishery occupations. We find that key sources of vulnerability differ considerably within and between the five countries. Our approach allows the visualization of how these dimensions of vulnerability differ from site to site, providing important insights into the types of nuanced policy interventions that may help to reduce vulnerability at a specific location. To complement this, we develop framework of policy actions thought to reduce different aspects of vulnerability at varying spatial and temporal scales. Although our results are specific to reef fisheries impacts from coral bleaching, this approach provides a framework for other types of threats and different social-ecological systems more broadly.

    • Institutions are vital to the sustainability of social-ecologicalsystems, balancing individual and group interests andcoordinating responses to change. Ecological decline andsocial conflict in many places, however, indicate that ourunderstanding and fostering of effective institutions for naturalresource management is still lacking. We assess theoreticaland methodological challenges facing positivist institutionalanalysis, focusing on natural resource governance according toOstrom’s social-ecological systems (SES) framework. Ratherthan adding more variables, progress requires a clearer, moreconsistent approach to selecting, defining and measuringinstitutional elements; stronger links between theory andempirical research; a greater focus on mechanisms andcausality; and the development and application of newmethods, including quantitative approaches. Strengthening theconnections between theory, models, and data suggestsseveral promising avenues for advancing institutional analysisthrough the study of relationships between institutionalstructure, process, function, context, and outcomes.

    • In the current study we investigate the antifouling potential of three polyphenolic resveratrol multimers (-)-hopeaphenol, vaticanol B and vatalbinoside A, isolated from two species of Anisoptera found in the Papua New Guinean rainforest. The compounds were evaluated against the growth and settlement of eight marine microfoulers and against the settlement and metamorphosis of Amphibalanus improvisus barnacle cyprids. The two isomeric compounds (-)-hopeaphenol and vaticanol B displayed a high inhibitory potential against the cyprid larvae metamorphosis at 2.8 and 1.1 mu M. (-)-Hopeaphenol was also shown to be a strong inhibitor of both microalgal and bacterial adhesion at submicromolar concentrations with low toxicity. Resveratrol displayed a lower antifouling activity compared to the multimers and had higher off target toxicity against MCR-5 fibroblasts. This study illustrates the potential of natural products as a valuable source for the discovery of novel antifouling leads with low toxicity.

  • Can web crawlers revolutionize ecological monitoring?

    • Despite recent advances, ecosystem service monitoring is limited by insufficient data, the complexity of social–ecological systems, and poor integration of information that tracks changes in ecosystems and economic activities. However, new information and communication technologies are revolutionizing the generation of, and access to, such data. Can researchers who are interested in ecological monitoring tap into these increased flows of information by “mining” the internet to detect “early-warning” signs that may signal abrupt ecological changes? Here, we explore the possibility of using web crawlers and internet-based information to complement conventional ecological monitoring, with a special emphasis on the prospects for avoiding “late warnings”, that is, when ecosystems have already shifted to less desirable states. Using examples from coral reef ecosystems, we explore the untapped potential, as well as the limitations, of relying on web-based information to monitor ecosystem services and forewarn us of negative ecological shifts.

    • We present a method to quantify abrupt changes (or changepoints) in data series, represented as a function of depth or time. These changes are often the result of climatic or environmental variations and can be manifested in multiple datasets as different responses, but all datasets can have the same changepoint locations/timings. The method we present uses transdimensional Markov chain Monte Carlo to infer probability distributions on the number and locations (in depth or time) of changepoints, the mean values between changepoints and, if required, the noise variance associated with each dataset being considered. This latter point is important as we generally will have limited information on the noise, such as estimates only of measurement uncertainty, and in most cases it is not practical to make repeat sampling/measurement to assess other contributions to the variation in the data. We describe the main features of the approach (and describe the mathematical formulation in supplementary material), and demonstrate its validity using synthetic datasets, with known changepoint structure (number and locations of changepoints) and distribution of noise variance for each dataset. We show that when using multiple data, we expect to achieve better resolution of the changepoint structure than when we use each dataset individually. This is conditional on the validity of the assumption of common changepoints between different datasets. We then apply the method to two sets of real geochemical data, both from peat cores, taken from NE Australia and eastern Tibet. Under the assumption that changes occur at the same time for all datasets, we recover solutions consistent with those previously inferred qualitatively from independent data and interpretations. However, our approach provides a quantitative estimate of the relative probability of the inferred changepoints, allowing an objective assessment of the significance of each change.

  • Labour market strategies addressing precarious employment and its impacts : A systematic review

    • BackgroundPrecarious employment (PE), characterized by reduced worker rights, and employment and income insecurity, has complex public health implications including negative impacts on workers’ mental and physical health, occupational health and safety, wellbeing, and inequities in access to health and social protections. There is, however, a knowledge gap regarding effectiveness of interventions. We describe findings from a review of evaluated interventions with potential to address PE.MethodsOur systematic review followed the 2020 PRISMA framework and covered PubMed, Scopus, Web of Science, and sources of grey literature. We included qualitative, quantitative, or mixed-methods studies evaluating initiatives to reduce workers’ PE published from 2000 to 2021 and focused on adult workers.ResultsThe 23 eligible studies from across the world evaluated diverse strategies addressing PE including tax and trade reforms, industrial disputes legislation, business registration, and use of incentives to stimulate permanent contracts. Also included were union strategies to reach precarious workers, the provision of social benefits, and youth apprenticeships. Generally, while most initiatives had the potential to tackle certain PE aspects, they usually acted only on one or two PE dimensions. Additionally, the evaluation components were missing key details, thus, limiting the generalizability of findings, as did the heterogeneity of study designs, initiative purposes, economic and political context, and diverse populations targeted.ConclusionsThe increase in PE prevalence and its complex health implications requires sustainable upstream public health solutions. Multidisciplinary collaborations among public health and occupational health practitioners along with researchers, evaluation specialists, economists, and politicians could facilitate the implementation and evaluation of policies and standards regulating and monitoring PE and its health impacts.Key messages• Precarious employment has complex public health implications.• Sustainable solutions to address precarious employment must be upstream and multidisciplinary.

    • Objective This study investigated time trends in occupational exposure to various chemicals in Sweden and the distribution across demographic and labor market sectors. Methods Exposure to six chemicals was investigated from 1980 to 2013 by application of a job exposure matrix to national population registers. Respirable crystalline silica (RCS), diesel engine exhaust, welding fumes, wood dust, chlorinated hydrocarbon solvents, and lead were selected to represent different groups of chemicals. Trends in exposure prevalence were investigated by linear regression and compared to the occupationally active popula-tion. Confidence intervals for the rate of change over time were obtained by bootstrapping. Results The proportion of workers born outside the Nordic countries increased over time in those exposed to RCS, diesel exhaust and wood dust. There was a shift of exposed jobs to small companies (<50 employees), especially for RCS, welding fumes, wood dust, and chlorinated hydrocarbon solvents. For RCS and welding fumes, there was a marked drop in exposure levels from 1980 to 1990 but small changes thereafter. Exposure to lead diminished, both in terms of prevalence and intensity. Conclusions Over time, several exposures tended to shift to small companies, the construction sector, and migrant workers, all factors being indicative of less well-controlled working conditions. Occupational exposure to chlorinated organic solvents and lead diminished, while exposure levels to RCS and welding fumes have changed little since 1990. In view of the serious and well-established negative health effects, increased efforts to reduce exposure to RCS and welding fumes are needed.

    • BACKGROUND AND AIMS: An increased occurrence of anti-Saccharomyces cerevisiae antibodies (ASCA) is reported in unaffected members of families with Crohn's disease. Whether ASCA is a familial trait due to genetic factors or is caused by exposure to environmental factors is unknown. To assess the genetic influence of ASCA we studied its occurrence in a twin population.PATIENTS AND METHODS: ASCA were analysed in 98 twin pairs with inflammatory bowel disease and were related to clinical phenotype and CARD15/NOD2 genotype.RESULTS: ASCA were more common in Crohn's disease than in ulcerative colitis (40/70 (57%) twins v 5/43 (12%) twins). Associations with ileal Crohn's disease, stricturing/penetrating behaviour, and young age, but not CARD15/NOD2 were confirmed. ASCA were found in 1/20 (5%) healthy siblings in discordant monozygotic pairs with Crohn's disease compared with 7/27 (26%) in discordant dizygotic pairs. Using the intraclass correlation coefficient (ICC), no agreement in ASCA titres was observed in discordant twin pairs with Crohn's disease, in monozygotic (ICC = -0.02) or dizygotic (ICC = -0.26) pairs. In contrast, strong agreement was seen within concordant monozygotic twin pairs with Crohn's disease (ICC = 0.76).CONCLUSIONS: These findings question the concept of ASCA as a marker of genetic susceptibility for Crohn's disease. The agreement in ASCA titres within concordant monozygotic twin pairs with Crohn's disease, suggests that the level of increase is genetically determined. We propose that ASCA are a marker of a response to an environmental antigen and that a specific gene(s) other than CARD15/NOD2 determines the level of response and perhaps also specific phenotypic characteristics.

    • Gastrointestinal (GI) problems constitute an important comorbidity in many patients with autism. Multiple mutations in the neuroligin family of synaptic adhesion molecules are implicated in autism, however whether they are expressed and impact GI function via changes in the enteric nervous system is unknown. We report the GI symptoms of two brothers with autism and an R451C mutation in Nlgn3 encoding the synaptic adhesion protein, neuroligin-3. We confirm the presence of an array of synaptic genes in the murine GI tract and investigate the impact of impaired synaptic protein expression in mice carrying the human neuroligin-3 R451C missense mutation (NL3(R451C)). Assessing in vivo gut dysfunction, we report faster small intestinal transit in NL3(R451C) compared to wild-type mice. Using an ex vivo colonic motility assay, we show increased sensitivity to GABA(A) receptor modulation in NL3(R451C) mice, a well-established Central Nervous System (CNS) feature associated with this mutation. We further show increased numbers of small intestine myenteric neurons in NL3(R451C) mice. Although we observed altered sensitivity to GABA(A) receptor modulators in the colon, there was no change in colonic neuronal numbers including the number of GABA-immunoreactive myenteric neurons. We further identified altered fecal microbial communities in NL3(R451C) mice. These results suggest that the R451C mutation affects small intestinal and colonic function and alter neuronal numbers in the small intestine as well as impact fecal microbes. Our findings identify a novel GI phenotype associated with the R451C mutation and highlight NL3(R451C) mice as a useful preclinical model of GI dysfunction in autism. Autism Res 2019, 12: 1043-1056. We report gut symptoms in patients with the autism-associated R451C mutation encoding the neuroligin-3 protein. We show that many of the genes implicated in autism are expressed in mouse gut. The neuroligin-3 R451C mutation alters the enteric nervous system, causes gastrointestinal dysfunction, and disrupts gut microbe populations in mice. Gut dysfunction in autism could be due to mutations that affect neuronal communication.
