  • Longitudinal infusion of a complex of insulin-like growth factor-I and IGF-binding protein-3 in five preterm infants : pharmacokinetics and short-term safety.
  • 2013
  • In: Pediatric Research. - : Springer Science and Business Media LLC. - 0031-3998 .- 1530-0447. ; 73:1, s. 68-74
  • Journal article (peer-reviewed)abstract
    • Background:In preterm infants, low levels of insulin-like growth factor-I (IGF-I) and IGF binding protein 3 (IGFBP-3) are associated with impaired brain growth and retinopathy of prematurity (ROP). Treatment with IGF-I/IGFBP-3 may be beneficial for brain development and may decrease the prevalence of ROP.Methods:In a phase II pharmacokinetics and safety study, five infants (three girls) with a median (range) gestational age (GA) of 26 wk + 6 d (26 wk + 0 d to 27 wk + 2 d) and birth weight of 990 (900-1,212) g received continuous intravenous infusion of recombinant human (rh)IGF-I/rhIGFBP-3. Treatment was initiated during the first postnatal day and continued for a median (range) duration of 168 (47-168) h in dosages between 21 and 111 µg/kg/24 h.Results:Treatment with rhIGF-I/rhIGFBP-3 was associated with higher serum IGF-I and IGFBP-3 concentrations (P < 0.001) than model-predicted endogenous levels. Of 74 IGF-I samples measured during study drug infusion, 37 (50%) were within the target range, 4 (5%) were above, and 33 (45%) were below. The predicted dose of rhIGF-I/rhIGFBP-3 required to establish circulating levels of IGF-I within the intrauterine range in a 1,000 g infant was 75-100 µg/kg/24 h. No hypoglycemia or other adverse effects were recorded. Conclusion:In this study, continuous intravenous infusion of rhIGF-I/rhIGFBP-3 was effective in increasing serum concentrations of IGF-I and IGFBP-3, and was found to be safe.

  • ‘I want to go home’: a comparative study of unsafe and safe spaces in two Swedish school-age educare institutions
  • 2024
  • In: Ethnography and Education. - 1745-7823 .- 1745-7831.
  • Journal article (peer-reviewed)abstract
    • In Sweden, many children between the ages of 6 and 9 years attend Swedish school-age educare (SAEC) institutions before and after school, when their parents work or study. This study aimed to explore and compare safe and unsafe spaces in children’s everyday lives at SAEC institutions. Ethnographic fieldwork, including observations and interviews with the staff and children, was conducted at two different SAECs. Using space, agency, and violence, this study explores how the staff relates to safe and unsafe spaces in children’s everyday lives. Findings revealed differences in how everyday life plays out in the two SAECs, where unsafe spaces, with a risk of violence, more frequently exist in one SAEC than the other. The manner in which staff members perceived safe and unsafe spaces also impacted how children’s agency emerged in different spaces. The study findings have important implications for politicians and decision-makers.

  • "Jag ser att det här kan va' min framtida stad" : unga vuxans upplevelser av att leva och bo i Västervik
  • 2020
  • Reports (other academic/artistic)abstract

  • “Sadly, we just let it go, it is so normalized”: Staff descriptions of violence among children within organized leisure
  • 2023
  • In: Journal of Leisure Research. - 0022-2216.
  • Journal article (peer-reviewed)abstract
    • The aim of this study was to investigate how staff describe violence among children within organized leisure in Sweden, named school-age EduCare. The study is based on ten semi-structured interviews with staff, and analyzed with theoretical concepts of Lefebvre’s space theory, and Galtung’s theory of violence. The results reveal three types of violence—quarrel for fun, violations, and physical violence—and three ways of reacting and managing it—stress and resignation, material adaption, and adaption through guarding. In conclusion, this study shows an everyday life filled with violent spaces, but limited resources for staff to cope with violence and create safe spaces for children and staff. The study pinpoints the complexity in everyday life where violence in different ways occurs between children, and staff are unable to manage this in an appropriate way. Instead, staff adjust to time and place, where spaces of violence between children are normalized.

  • ‘This place does not feel safe’: safe and unsafe spaces in Swedish school-age educare
  • 2023
  • In: Children's Geographies. - : Informa UK Limited. - 1473-3285 .- 1473-3277. ; 21:6, s. 1044-1057
  • Journal article (peer-reviewed)abstract
    • This study explores the occurrence of violence in children’s everyday lives in different places in school and school-age educare. School-age educare is a Swedish institution providing care before and after school. The study is based on interviews, observations, and walk-and-talk sessions with 11 children in a school-age educare institution. A thematic analysis informed by the theoretical concepts of Doreen Massey (space and place) and Johan Galtung (direct, structural, and cultural violence) was conducted. Three themes were developed: spaces of tense waiting; pop-up yard spaces; and ignored spaces in the woods. The results show that physical and psychological violence arises in the schoolyard, gym, and canteen. The staff did not always pay attention to the violence that occurred, and the children found ways to navigate and handle violence by avoiding certain places. They also used relationships with children and adults for their support. This study examines the everyday life in which direct and cultural violence occur.

  • IGF-1 as a Drug for Preterm Infants : A Step-Wise Clinical Development
  • 2017
  • In: Current Pharmaceutical Design. - : Bentham Science Publishers Ltd.. - 1381-6128 .- 1873-4286. ; 23:38, s. 5964-5970
  • Research review (peer-reviewed)abstract
    • BACKGROUND: Insulin-like growth factor 1 (IGF-1) is a mitogenic hormone involved in many processes such as growth, metabolism, angiogenesis and differentiation. After very preterm birth, energy demands increase while maternal supplies of nutrients and other factors are lost and the infant may become dependent on parenteral nutrition for weeks. Low postnatal IGF-1 concentrations in preterm infants are associated with poor weight gain, retinopathy of prematurity (ROP) and other morbidities. We will describe the process by which we aim to develop supplementation with recombinant human (rh) IGF-1 and its binding protein rhIGFBP-3 as a possible therapy to promote growth and maturation and reduce morbidities in extremely preterm infants.METHODS: In order to calculate a dose of IGF-1 tolerated by neonates, a pharmacokinetic study of transfusion with fresh frozen plasma was performed, which provided a relatively low dose of IGF-1, (on average 1.4 µg/kg), that increased serum IGF-1 to levels close to those observed in fetuses and preterm infants of similar GAs. Thereafter, a Phase I 3 hours IV infusion of rhIGF-1/rhIGFBP-3 was conducted in 5 infants, followed by a Phase II study with four sections (A-D). In the Phase II, sections A-D studies, time on infusion increased and younger gestational ages were included.RESULTS: IV infusion increased IGF-1 but with short half-life (0.5h) implying a need for continuous infusion. In order to obtain in utero levels of IGF-I, the dose was increased from 100 to 250 µg/kg/24 h and the infusion was prolonged from 3 weeks postnatal age until a postmenstrual age of 29 weeks and 6 days.CONCLUSION: The purpose has been to ensure high-quality research into the development of a new drug for preterm infants. We hope that our work will help to establish a new standard for the testing of medications for preterm infants.

  • The spatial RNA integrity number assay for in situ evaluation of transcriptome quality
  • 2021
  • In: Communications Biology. - : Springer Nature. - 2399-3642. ; 4:1
  • Journal article (peer-reviewed)abstract
    • The RNA integrity number (RIN) is a frequently used quality metric to assess the completeness of rRNA, as a proxy for the corresponding mRNA in a tissue. Current methods operate at bulk resolution and provide a single average estimate for the whole sample. Spatial transcriptomics technologies have emerged and shown their value by placing gene expression into a tissue context, resulting in transcriptional information from all tissue regions. Thus, the ability to estimate RNA quality in situ has become of utmost importance to overcome the limitation with a bulk rRNA measurement. Here we show a new tool, the spatial RNA integrity number (sRIN) assay, to assess the rRNA completeness in a tissue wide manner at cellular resolution. We demonstrate the use of sRIN to identify spatial variation in tissue quality prior to more comprehensive spatial transcriptomics workflows. Kvastad et al. develop the spatial RNA Integrity Number (sRIN) assay that evaluates the RNA integrity at cellular resolution. This method improves the resolution of a similar method called the RNA Integrity Number (RIN), demonstrating spatial variation in the quality of RNA samples.

  • Gene expression profiling spares early breast cancer patients from adjuvant therapy : derived and validated in two population-based cohorts
  • 2005
  • In: Breast cancer research : BCR. - : Springer Science and Business Media LLC. - 1465-5411 .- 1465-542X. ; 7:6, s. R953-64
  • Journal article (peer-reviewed)abstract
    • INTRODUCTION: Adjuvant breast cancer therapy significantly improves survival, but overtreatment and undertreatment are major problems. Breast cancer expression profiling has so far mainly been used to identify women with a poor prognosis as candidates for adjuvant therapy but without demonstrated value for therapy prediction. METHODS: We obtained the gene expression profiles of 159 population-derived breast cancer patients, and used hierarchical clustering to identify the signature associated with prognosis and impact of adjuvant therapies, defined as distant metastasis or death within 5 years. Independent datasets of 76 treated population-derived Swedish patients, 135 untreated population-derived Swedish patients and 78 Dutch patients were used for validation. The inclusion and exclusion criteria for the studies of population-derived Swedish patients were defined. RESULTS: Among the 159 patients, a subset of 64 genes was found to give an optimal separation of patients with good and poor outcomes. Hierarchical clustering revealed three subgroups: patients who did well with therapy, patients who did well without therapy, and patients that failed to benefit from given therapy. The expression profile gave significantly better prognostication (odds ratio, 4.19; P = 0.007) (breast cancer end-points odds ratio, 10.64) compared with the Elston-Ellis histological grading (odds ratio of grade 2 vs 1 and grade 3 vs 1, 2.81 and 3.32 respectively; P = 0.24 and 0.16), tumor stage (odds ratio of stage 2 vs 1 and stage 3 vs 1, 1.11 and 1.28; P = 0.83 and 0.68) and age (odds ratio, 0.11; P = 0.55). The risk groups were consistent and validated in the independent Swedish and Dutch data sets used with 211 and 78 patients, respectively. CONCLUSION: We have identified discriminatory gene expression signatures working both on untreated and systematically treated primary breast cancer patients with the potential to spare them from adjuvant therapy.

  • A survey on incidence and factors that may influence first postoperative urination.
  • 2003
  • In: Urologic nursing : official journal of the American Urological Association Allied. - 1053-816X. ; 23:5
  • Journal article (peer-reviewed)abstract
    • A prospective study was conducted to investigate factors that might influence first postoperative urination. In 174 patients, 39% were not able to void spontaneously after surgery and required catheterization. Identified factors included males over 55 years of age, extended time in surgery, extended anesthesia time, and previous history of voiding problems. It is important for nurses to take a more active role in prescreening for voiding problems as well as be knowledgeable about the pharmacologic action of drugs causing bladder overdistention.
