    • The role of Ribonuclease H1 (RNase H1) during primer removal and ligation at the mitochondrial origin of light-strand DNA synthesis (OriL) is a key, yet poorly understood, step in mitochondrial DNA maintenance. Here, we reconstitute the replication cycle of L-strand synthesis in vitro using recombinant mitochondrial proteins and model OriL substrates. The process begins with initiation of DNA replication at OriL and ends with primer removal and ligation. We find that RNase H1 partially removes the primer, leaving behind the last one to three ribonucleotides. These 5'-end ribonucleotides disturb ligation, a conclusion which is supported by analysis of RNase H1-deficient patient cells. A second nuclease is therefore required to remove the last ribonucleotides and we demonstrate that Flap endonuclease 1 (FEN1) can execute this function in vitro. Removal of RNA primers at OriL thus depends on a two-nuclease model, which in addition to RNase H1 requires FEN1 or a FEN1-like activity. These findings define the role of RNase H1 at OriL and help to explain the pathogenic consequences of disease causing mutations in RNase H1.

  • Zika among international travellers presenting to GeoSentinel sites, 2012-2019 : implications for clinical practice

    • Introduction: International travellers contribute to the rapid spread of Zika virus (ZIKV) and its sentinel identification globally. We describe ZIKV infections among international travellers seen at GeoSentinel sites with a focus on ZIKV acquired in the Americas and the Caribbean, describe countries of exposure and traveller characteristics, and assess ZIKV diagnostic testing by site. Methods: Records with an international travel-related diagnosis of confirmed or probable ZIKV from January 2012 through December 2019 reported to GeoSentinel with a recorded illness onset date were included to show reported cases over time. Records from March 2016 through December 2019 with an exposure region of the Americas or the Caribbean were included in the descriptive analysis. A survey was conducted to assess the availability, accessibility and utilization of ZIKV diagnostic tests at GeoSentinel sites. Results: GeoSentinel sites reported 525 ZIKV cases from 2012 through 2019. Between 2012 and 2014, eight cases were reported, and all were acquired in Asia or Oceania. After 2014, most cases were acquired in the Americas or the Caribbean, a large decline in ZIKV cases occurred in 2018-19. Between March 2016 and December 2019, 423 patients acquired ZIKV in the Americas or the Caribbean, peak reporting to these regions occurred in 2016 [330 cases (78%)]. The median age was 36 years (range: 3-92); 63% were female. The most frequent region of exposure was the Caribbean (60%). Thirteen travellers were pregnant during or after travel; one had a sexually acquired ZIKV infection. There was one case of fetal anomaly and two travellers with Guillain-Barre syndrome. GeoSentinel sites reported various challenges to diagnose ZIKV effectively. Conclusion: ZIKV should remain a consideration for travellers returning from areas with risk of ZIKV transmission. Travellers should discuss their travel plans with their healthcare providers to ensure ZIKV prevention measures are taken.

    • For the first time in serveral years, the El Nino-Southern Oscillation did not dominate regional climate conditions around the globe. A weak La Ni a dissipated to ENSOneutral conditions by spring, and while El Nino appeared to be emerging during summer, this phase never fully developed as sea surface temperatures in the eastern conditions. Nevertheless, other large-scale climate patterns and extreme weather events impacted various regions during the year. A negative phase of the Arctic Oscillation from mid-January to early February contributed to frigid conditions in parts of northern Africa, eastern Europe, and western Asia. A lack of rain during the 2012 wet season led to the worst drought in at least the past three decades for northeastern Brazil. Central North America also experienced one of its most severe droughts on record. The Caribbean observed a very wet dry season and it was the Sahel's wettest rainy season in 50 years. Overall, the 2012 average temperature across global land and ocean surfaces ranked among the 10 warmest years on record. The global land surface temperature alone was also among the 10 warmest on record. In the upper atmosphere, the average stratospheric temperature was record or near-record cold, depending on the dataset. After a 30-year warming trend from 1970 to 1999 for global sea surface temperatures, the period 2000-12 had little further trend. This may be linked to the prevalence of La Ni a-like conditions during the 21st century. Heat content in the upper 700 m of the ocean remained near record high levels in 2012. Net increases from 2011 to 2012 were observed at 700-m to 2000-m depth and even in the abyssal ocean below. Following sharp decreases in to the effects of La Ni a, sea levels rebounded to reach records highs in 2012. The increased hydrological cycle seen in recent years continued, with more evaporation in drier locations and more precipitation in rainy areas. In a pattern that has held since 2004, salty areas of the ocean surfaces and subsurfaces were anomalously salty on average, while fresher areas were anomalously fresh. Global tropical cyclone activity during 2012 was near average, with a total of 84 storms compared with the 1981-2010 average of 89. Similar to 2010 and 2011, the North Atlantic was the only hurricane basin that experienced above-normal activity. In this basin, Sandy brought devastation to Cuba and parts of the eastern North American seaboard. All other basins experienced either near-or below-normal tropical cyclone activity. Only three tropical cyclones reached Category 5 intensity-all in Bopha became the only storm in the historical record to produce winds greater than 130 kt south of 7 N. It was also the costliest storm to affect the Philippines and killed more than 1000 residents. Minimum Arctic sea ice extent in September and Northern Hemisphere snow cover extent in June both reached new record lows. June snow cover extent is now declining at a faster rate (-17.6% per decade) than September sea ice extent (-13.0% per decade). Permafrost temperatures reached record high values in northernmost Alaska. A new melt extent record occurred on 11-12 July on the Greenland ice sheet; 97% of the ice sheet showed some form of melt, four times greater than the average melt for this time of year. The climate in Antarctica was relatively stable overall. The largest maximum sea ice extent since records begain in 1978 was observed in September 2012. In the stratosphere, warm air led to the second smallest ozone hole in the past two decades. Even so, the springtime ozone layer above Antarctica likely will not return to its early 1980s state until about 2060. Following a slight decline associated with the global 2 emissions from fossil fuel combustion and cement production reached a record 9.5 +/- 0.5 Pg C in 2011 and a new record of 9.7 +/- 0.5 Pg C is estimated for 2012. Atmospheric CO2 concentrations increased by 2.1 ppm in 2012, to 392.6 ppm. In spring 2012, 2 concentration exceeded 400 ppm at 7 of the 13 Arctic observation sites. Globally, other greenhouse gases including methane and nitrous oxide also continued to rise in concentration and the combined effect now represents a 32% increase in radiative forcing over a 1990 baseline. Concentrations of most ozone depleting substances continued to fall.

    • Current calls for deep societal transformation stress the need to go beyond green consumption and aim for the reduction of material consumption. Such a shift is enacted by grassroots initiatives around reuse, repair and sharing. However, the possibility of postconsumerist perspectives and practices to reach a broader audience is affected by discursive opportunity structures (DOS) formed in public debate. To understand the DOS it is relevant to pay attention both to the continuous normalisation of consumption and to the ways in which alternatives are represented. To develop new analytical tools for examining what postconsumerist initiatives are up against, we introduce the concept of banal consumerism: mundane, habitual expressions that reproduce consumer culture. Through an empirical study of Swedish daily newspapers, we construct the basis for a typology of different expressions of banal consumerism. We find several expressions, of which the massive advertising of consumer goods is the most common but editorial material also plays an important role. This largely disabling DOS is then put in relation to the potentially enabling opportunities entailed in the existing media coverage of postconsumerist initiatives, practices and perspectives. The results show that postconsumerist initiatives and practices are newsworthy and presented as commendable. The fact that high levels of material consumption negatively impact the environment and life on the planet is widely accepted. To argue for degrowth or criticise consumerism is, however, controversial. Thus, support for postconsumerist practices coexist with massive expressions of banal consumerism, creating a complex set of DOS for the postconsumerist initiatives to navigate.

    • There is currently a marked interest in do-it-yourself culture and sharing of skills, tools and spaces, manifesting in maker spaces, tool libraries and open workshops for remaking furniture, electronics, bicycles or clothing. This paper explores the phenomenon of 'Bike Kitchens', do-it-yourself bicycle repair studios run on a non-profit basis. The Bike Kitchen in Malmo, Sweden, is used as a case study involving interviews with key persons and users of that Bicycle Kitchen and on-site observations. The exploration of the Bike Kitchen is situated in a wider theoretical discussion around technology in relation to degrowth. Two theoretical perspectives are used, firstly, lllich's (1973) notion of tools for conviviality, meaning tools that enable citizens to reconquer practical knowledge for autonomy and creativity rather than being confined to commercial relations, and secondly, forms of non-capitalist relations. It is argued that the Bike Kitchen is an example of democratisation of technology in practice a social innovation to make low-cost technology, tools and know-how easily available to anyone. The concept of the Bike Kitchen is a way to develop and cultivate conviviality, i.e. a social and spatial infrastructure - a space for convivial tools.

  • Community repair in the circular economy : Fixing more than stuff

    • In the circular economy discourse it is stressed that products ought to be repairable and that repair work is assumed to be growing. However, repair can be organised and performed in different ways – by corporate entities, independent repairers, laypersons and communities. Some corporations are integrating repair and maintenance into their offering, while simultaneously restricting consumers to open, repair or modify their products. In opposition to such developments, there is a movement for “right to repair”, which works for consumers’ legal rights to repair and modify products, pushing for the free availability of spare parts and manuals. Recent years have also seen a growth of repair cafés and other forms of DIY community repair spaces. This paper explores the discourses of DIY community repair through two Swedish case studies – an NGO-led nationwide repair campaign and a local government initiative of open DIY repair spaces. Our case studies show how DIY community repair works towards enabling all, particularly marginalised groups, to participate and live well in a low-impact future. In contrast to the mainstream circular economy discourse, the purpose of community repair is not only about repairing broken stuff and reducing waste, but about building social relations and practicing non-consumerist forms of citizenship. By elucidating these different perspectives on repair – who is to perform it, with what skills and for what purposes – we highlight how the transition to future, more circular economies, can be enacted and steered in ways that allow for different roles and powers for citizen-consumers.

    • This contribution takes its starting point from a Nordic study on planning for sustainable development, which highlights that the debatein this area seems to have settled around a uniform picture of thesustainable society, and that this is therefore something that needs to becritically examined. The mainstream way of planning for sustainabledevelopment does not adequately address the issue of dependency orrelationships with development in distant locations. In this chapter it isargued that an environmental justice perspective could add new depth tothe debate on planning for sustainable development. This is done throughhighlighting just how different social groups, mainly based on income,ethnicity or gender, are exposed to, generate and interpret environmental qualities and risks. In the Nordic debate on planning for sustainable development such social justice and power-related aspects have in general not been studied or taken into account. This contribution is thus acall for research in this field, particularly as the populations of the Nordic countries are becoming increasingly heterogeneous.

    • Environmental challenges, especially climate change, are highly discussed topics in the Swedish public debate, but questions about who is causing the problems and who is affected by them are seldom asked. This also applies to questions of who defines what should be regarded as acute environmental problems and what constitutes high-quality environments. This paper explores how environmental (in)justice issues can be framed in a Swedish social context, drawing from three cases: municipal promotion of eco-friendly lifestyles, large-scale infrastructure planning, and planners’ attitudes towards justice. The three cases deal not only with distributional, procedural, and substantive aspects of justice, as is common within the US environmental justice framework, but also with discursive dimensions of justice. We argue that elucidating such examples of environmental (in)justices is crucial to nuance the mainstream, consensus-oriented sustainability discourse in Sweden.

    • Samhällsplaneringen är på många sätt en produkt av modernismen och intimt förknippad med välfärdsstaten och folkhemsbygget. Därmed också sagt att den är i kris. Förutsättningarna för planering har förändrats så radikalt under de senaste decennierna att det inte längre går att hålla fast vid gamla mål och medel. Planeringen ändras visserligen i praxis men vi som forskar i ämnet och har i uppdrag att utbilda nästa generation planerare måste också försöka dra upp riktlinjer för en fungerande och angelägen samhällsplanering för i morgon. I denna artikel efterlyser vi en planerare som – till skillnad från gängse praxis – är både proaktiv och inriktad på att värna de eller det marginaliserade.

  • Just Environments : Politicising Sustainable Urban Development

    •   European cities are becoming increasingly multicultural and diverse in terms of lifestyles and socioeconomic conditions. However, in planning for sustainable urban development, implications of this increased diversity and possibly conflicting perspectives are seldom considered. The aim of this thesis is to explore dimensions of justice and politics in sustainable urban development by studying inclusionary/exclusionary effects of discursive power of official strategies for eco-friendly living on the one hand and everyday lifestyles on the other, in ethnically and socially diverse areas. Two case studies have been conducted, one in a city district of Stockholm, Sweden, and one in an area of Sheffield, England. The empirical material consists of interviews with residents, interviews with planners and officials and an analysis of strategic planning documents. The case study in Stockholm illustrated the prevalence of a dominant discourse among residents in which Swedishness is connected with environmental responsibility in the form of tidiness, recycling and familiarity with nature. In Sheffield there are more competing and parallel environmental discourses. The mainstream British environmental discourse and sustainability strategies are being criticised from Muslim as well as green radical perspectives. The mainstream discourse is criticised for being tokenistic in its focus on gardening, tidiness, recycling and eco-consumption, and hence ignoring deeper unsustainable societal structures. This can be interpreted as a postpolitical condition, in which there is a consensus around “what needs to be done,” such as more recycling, but in which difficult societal problems and conflicting perspectives on these are not highlighted. In the thesis it is argued that the strategies for urban sustainability are underpinned by Swedish/British middle-class norms, entailing processes of (self-)disciplining and normalisation of the Other into well-behaving citizens. It is argued that an appreciation of the multiple and others’ ways of saving natural resources would make the sustainability strategies more attuned to social and cultural diversity as well as more environmentally progressive. Finally, the importance of asserting the political in sustainability strategies is stressed, highlighting the organisation of society and possible alternative socioenvironmental futures.

    • Within contemporary architecture and urbanism there is marked interest in urban commons. This paper explores the creation of temporary urban commons, or, more specifically, what can be called ‘open-source urbanism’. Citing two practices – urban commons initiated by Atelier d’architecture autogérée in Paris, and Park(ing) Day initiated by San Francisco-based Rebar – I argue that these practices can be understood as open-source urbanism since their initiators act as open-source programmers, constructing practice manuals to be freely copied, used, developed and shared, thus producing self-managed commons. Although this tradition of ‘commoning’ is not new, it is currently being reinvented with the use of digital technologies. Combining Elinor Ostrom’s analysis of self-managed natural resource commons with Yochai Benkler’s assertion that commons-based peer production constitutes a ‘third mode of production’ that lies beyond capitalism, socialism and their blends, I argue that open-source urbanism critiques both government and privately-led urban development by advancing a form of postcapitalist urbanism.

    • Swedish cities are becoming increasingly multicultural and diverse in terms of lifestylesand socioeconomic conditions. However, cultural and social diversity is seldomconsidered when planning for sustainable urban development. This paper examinesplanning for more eco-friendly living in the increasingly diverse population of a citydistrict of Stockholm. The study reveals the prevalence of a discourse in which aSwedish identity carries environmental responsibility in the form of tidiness, recyclingand familiarity with nature. It is argued that planning for urban sustainability isunderpinned by Swedish middle-class norms, indirectly entailing processes of (self-)disciplining and transforming the other (foreign and/or troublesome dwellers) intowell-behaving Swedes. A clearer definition of the environmental improvementintended, its goals and target groups is needed. Finally, an appreciation of the multipleways we can save natural resources would make urban planning policies more attunedto social and cultural diversity as well as more environmentally progressive.

    • This article aims to make a contribution to the debate on how contemporary collaborative commons, as part of the wider sharing economy, can be understood and supported. Three cases of contemporary commons are analysed: a DIY bike repair studio, a pop-up home office concept and Wikipedia. The article shows how the design principles developed for governing natural resource commons are only partly applicable to these contemporary commons. It also illustrates the differences in these types of commons in terms of the nature of the resource being shared, scarcity, barriers to entry and how rules are formulated and upheld.

    • During the spring of this year numerous reports appeared in the Swedish media concerning women being attacked and raped in public places. Does planning have anything to do with this? Around the world, women often avoid walking alone in cities at night or feel insecure when they do so. The mere presence of an unaccompanied woman in the city is, in certain situations, seen as an invitation to sexual harassment, robbery or rape. In some cities, such as Mexico City or Bombay, this has led to the establishment of train cars restricted to women only. In this way our gender determines our movements, our fears, whether we can walk alone at night, whether we can take the shortcut through the woods or whether we are restricted to the home and to private spaces.

    • In transitions toward more sustainable and just societies, there is an urgent need to address overconsumption and to include a sufficiency perspective. This article contributes to previous research by exploring what a framework for a politics of sufficiency might entail and how such a framework can be used to analyze existing public policy. Our case analysis is the policy field of sustainable consumption and waste prevention in the context of Sweden. Based on interviews with public officials and civil society representatives, we identify key areas to address when aiming for a sufficiency orientation. Our results suggest that local and regional governments that strive for a commitment to sufficiency should formulate clear goals that serve to set environmental limits, for instance, in the form of carbon budgets, and then steer toward well-being for the inhabitants within these limits. Efforts should be made to secure stable funding for work within sustainable consumption and waste prevention, especially for projects with synergies in terms of reducing consumption and strengthening non-market relations. Using public procurement is another key tool. In the absence of an overall national politics of sufficiency, the above-mentioned strategies can be and already are to some degree, used by authorities in Sweden.
