    • Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives.

  • Engineering of the Ultra-stable Cystine Knot Framework of Microproteins : Design, Chemical Synthesis and Structural Studies

    • Ultra-stable cystine knotted microproteins, in which two disulfides and their connecting backbones form a circle that is penetrated by the third disulfide bonds, have attracted high interest due to their resistance to degradation in vitro and potential for the development of peptide drugs. This thesis gives new insights into engineering of that framework of microproteins, including approaches to their chemical synthesis, backbone engineering, structural and biological evaluations. Synthetic and oxidative folding approaches for bracelet cyclotides, a family of cyclic cystine knotted microproteins, was developed using a model peptide, cycloviolacin O2. Following assembly of the peptide chain, protected peptide was generated by mild cleavage that was subsequently thioesterified and cyclized in solution. The cyclic peptide was oxidatively folded under optimized conditions containing co-solvent and non-ionic detergent affording native cycloviolacin O2 as a major product. To gain further insights into the heterogeneity, efficiency and kinetics of cyclotides’ oxidative folding, the intermediates that accumulate in oxidative refolding pathways of all cyclotide subfamilies: Möbius, bracelet and the hybrid cyclotides were quantitatively determined under four different folding conditions. The results were used for defining major folding pathways, which indicated that Möbius cyclotides might accumulate heterogeneous folding intermediates with one-, two- and three-disulfides, whereas bracelet tend to accumulate a homogenous intermediate with three-disulfides, depending on the buffer systems used. Furthermore, to probe the internal factors contributing to inefficiency of oxidative folding, as well as undesired bioactivities of bracelet cyclotides (e.g., cytotoxic activity), polymer-hybridized cyclotides were designed by replacing non-conserved residues with small isosteric polymers. The designed hybrid analogs in which hybridization involved replacement of loop 3 with isosteric polymers showed improved synthetic and oxidative folding properties. The cytoxicity of a model hybrid designed with replacement of loop 3 and 5 exhibited no cytotoxic activity at concentration of 128-fold relative to that of native peptide. Furthermore, 1D and 2D 1H NMR analysis of this hybrid showed that it had well structured fold.

    • A method for producing self-organised arrays of nanometric metallic dots is reported. It consists on developing first the nanodot pattern by ion erosion on a semiconductor cover film and transferring it to a previously buried metallic layer. This procedure has been applied to Co, and the ferromagnetic behaviour of the dots at room temperature is demonstrated.

  • Limited capacity of tree growth to mitigate the global greenhouse effect under predicted warming

    • It is generally accepted that animal heartbeat and lifespan are often inversely correlated, however, the relationship between productivity and longevity has not yet been described for trees growing under industrial and pre-industrial climates. Using 1768 annually resolved and absolutely dated ring width measurement series from living and dead conifers that grew in undisturbed, high-elevation sites in the Spanish Pyrenees and the Russian Altai over the past 2000 years, we test the hypothesis of grow fast-die young. We find maximum tree ages are significantly correlated with slow juvenile growth rates. We conclude, the interdependence between higher stem productivity, faster tree turnover, and shorter carbon residence time, reduces the capacity of forest ecosystems to store carbon under a climate warming-induced stimulation of tree growth at policy-relevant timescales.

  • New Tree-Ring Evidence from the Pyrenees Reveals Western Mediterranean Climate Variability since Medieval Times

    • Paleoclimatic evidence is necessary to place the current warming and drying of the western Mediterranean basin in a long-term perspective of natural climate variability. Annually resolved and absolutely dated temperature proxies south of the European Alps that extend back into medieval times are, however, mainly limited to measurements of maximum latewood density (MXD) from high-elevation conifers. Here, the authors present the world's best replicated MXD site chronology of 414 living and relict Pinus uncinata trees found >2200 m above mean sea level (MSL) in the Spanish central Pyrenees. This composite record correlates significantly (p <= 0.01) with May-June and August-September mean temperatures over most of the Iberian Peninsula and northern Africa (r = 0.72; 1950-2014). Spanning the period 1186-2014 of the Common Era (CE), the new reconstruction reveals overall warmer conditions around 1200 and 1400, and again after around 1850. The coldest reconstructed summer in 1258 (-4.4 degrees C compared to 1961-90) followed the largest known volcanic eruption of the CE. The twentieth century is characterized by pronounced summer cooling in the 1970s, subsequently rising temperatures until 2003, and a slowdown of warming afterward. Little agreement is found with climate model simulations that consistently overestimate recent summer warming and underestimate preindustrial temperature changes. Interannual-multidecadal covariability with regional hydroclimate includes summer pluvials after large volcanic eruptions. This study demonstrates the relevance of updating MXD-based temperature reconstructions, not only back in time but also toward the present, and emphasizes the importance of comparing temperature and hydroclimatic proxies, as well as model simulations for understanding regional climate dynamics.

    • The growth/climate relationship of theoretically temperature-controlled high-elevation forests has been demonstrated to weaken over recent decades. This is likely due to new tree growth limiting factors, such as an increasing drought risk for ecosystem functioning and productivity across the Mediterranean Basin. In addition, declining tree growth sensitivity to spring temperature may emerge in response to increasing drought stress. Here, we evaluate these ideas by assessing the growth/climate sensitivity of 1500 tree-ring width (TRW) and 102 maximum density (MXD) measurement series from 711 and 74 Pinus uncinata trees, respectively, sampled at 28 high-elevation forest sites across the Pyrenees and two relict populations of the Iberian System. Different dendroclimatological standardization and split period approaches were used to assess the high- to low-frequency behavior of 20th century tree growth in response to temperature means, precipitation totals and drought indices. Long-term variations in TRW track summer temperatures until about 1970 but diverge afterwards, whereas MXD captures the recent temperature increase in the low-frequency domain fairly well. On the other hand summer drought has increasingly driven TRW along the 20th century. Our results suggest fading temperature sensitivity of Iberian high-elevation P. uncinata forest growth, and reveal the importance of summer drought that is becoming the emergent limiting factor of tree ring width formation in many parts of the Mediterranean Basin.

  • The International Soil Moisture Network : Serving Earth system science for over a decade

    • In 2009, the International Soil Moisture Network (ISMN) was initiated as a community effort, funded by the European Space Agency, to serve as a centralised data hosting facility for globally available in situ soil moisture measurements . The ISMN brings together in situ soil moisture measurements collected and freely shared by a multitude of organisations, harmonises them in terms of units and sampling rates, applies advanced quality control, and stores them in a database. Users can freely retrieve the data from this database through an online web portal (https://ismn.earth/en/, last access: 28 October 2021). Meanwhile, the ISMN has evolved into the primary in situ soil moisture reference database worldwide, as evidenced by more than 3000 active users and over 1000 scientific publications referencing the data sets provided by the network. As of July 2021, the ISMN now contains the data of 71 networks and 2842 stations located all over the globe, with a time period spanning from 1952 to the present. The number of networks and stations covered by the ISMN is still growing, and approximately 70 % of the data sets contained in the database continue to be updated on a regular or irregular basis. The main scope of this paper is to inform readers about the evolution of the ISMN over the past decade, including a description of network and data set updates and quality control procedures. A comprehensive review of the existing literature making use of ISMN data is also provided in order to identify current limitations in functionality and data usage and to shape priorities for the next decade of operations of this unique community-based data repository.

    • Growth of old trees in cold-limited forests may benefit from recent climate warming and rising atmospheric CO2 concentrations (c(a)) if age-related constraints do not impair wood formation. To test this hypothesis, we studied old Mountain pine trees at three Pyrenean high-elevation forests subjected to cold-wet (ORD, AIG) or warmer drier (PED) conditions. We analyzed long-term trends (1450-2008) in growth (BAI, basal area increment), maximum (MXD) and minimum (MID) wood density, and tree-ring carbon (delta C-13) and oxygen (delta O-18) isotope composition, which were used as proxies for intrinsic water-use efficiency (iWUE) and stomatal conductance (g(s)), respectively. Old pines showed positive (AIG and ORD) or stable (PED) growth trends during the industrial period (since 1850) despite being older than 400 years. Growth and wood density covaried from 1850 onwards. In the cold-wet sites (AIG and ORD) enhanced photosynthesis through rising c(a) was likely responsible for the post-1850 iWUE improvement. However, uncoupling between BAI and iWUE indicated that increases in iWUE were not responsible for the higher growth but climate warming. A reduction in g(s) was inferred from increased delta O-18 for PED trees from 1960 onwards, the wannest site where the highest iWUE increase occurred (34%). This suggests that an emergent drought stress at warm-dry sites could trigger stomatal closure to avoid excessive transpiration. Overall, carbon acquisition as lasting woody pools is expected to be maintained in aged trees from cold and high-elevation sites where old forests constitute unique long-term carbon reservoirs.

    • Ring-width series are important for diverse fields of research such as the study of past climate, forest ecology, forest genetics, and the determination of origin (dendro-provenancing) or dating of archaeological objects. Recent research suggests diverging climate-growth relationships in tree-rings due to the cardinal direction of extracting the tree cores (i.e. direction-specific effect). This presents an understudied source of bias that potentially affects many data sets in tree-ring research. In this study, we investigated possible direction-specific growth variability based on an international (10 countries), multi-species (8 species) tree-ring width network encompassing 22 sites. To estimate the effect of direction-specific growth variability on climate-growth relationships, we applied a combination of three methods: An analysis of signal strength differences, a Principal Component Gradient Analysis and a test on the direction-specific differences in correlations between indexed ring-widths series and climate variables. We found no evidence for systematic direction-specific effects on tree radial growth variability in high-pass filtered ring-width series. In addition, direction-specific growth showed only marginal effects on climate-growth correlations. These findings therefore indicate that there is no consistent bias caused by coring direction in data sets used for diverse dendrochronological applications on relatively mesic sites within forests in flat terrain, as were studied here. However, in extremely dry, warm or cold environments, or on steep slopes, and for different life-forms such as shrubs, further research is advisable.

    • This paper analyzes the influence of the interannual variability of climatic drought on ecological and hydrological droughts for a basin in the central Spanish Pyrenees using variables derived from observations and hydro-ecological simulation in order to determine the possible connection between meteorological, ecological and hydrological drought considering a cascading approach and encompassing different variables that give insights into water availability in the basin (e.g., soil moisture, streamflow, reservoir storages and releases). Using different climatic, ecological and hydrological standardized drought indices, we show the greater role of meteorological droughts in hydrological systems than in ecological systems, and the small influence of vegetation activity and growth in explaining the interannual variability of water resources in the basin. By contrast,hydrological droughts are strongly affected by precipitation variability with relationships characterized by seasonal differences and the role of different time-scales in the standardized drought metrics.

    • A mutant laccase from the Ascomycete Myceliophthora thermophila has been submitted to iterative cycles of combinatorial satn. mutagenesis through in vivo overlap extension in Saccharomyces cerevisiae. Over 180,000 clones were explored, among which the S510G mutant revealed a direct interaction between the conserved 509VSG511 tripeptide, located in the neighborhood of the T1 site, and the C-terminal plug. The KmO2 value of the mutant increased 1.5-fold, and the electron transfer pathway between the reducing substrate and the T1 copper ion was altered, improving the catalytic efficiency towards non-phenolic and phenolic substrates by about 3- and 8-fold. Although the geometry at the T1 site was perturbed by the mutation, paradoxically the laccase redox potential was not significantly altered. Together, the results obtained in this study suggest that the 509VSG511 tripeptide may play a hitherto unrecognized role in regulating the traffic of oxygen through the C-terminal plug, the latter blocking access to the T2/T3 copper cluster in the native enzyme.
