    • Myoglobin, encoded by MB, is a small cytoplasmic globular hemoprotein highly expressed in cardiac myocytes and oxidative skeletal myofibers. Myoglobin binds O2, facilitates its intracellular transport and serves as a controller of nitric oxide and reactive oxygen species. Here, we identify a recurrent c.292C>T (p.His98Tyr) substitution in MB in fourteen members of six European families suffering from an autosomal dominant progressive myopathy with highly characteristic sarcoplasmic inclusions in skeletal and cardiac muscle. Myoglobinopathy manifests in adulthood with proximal and axial weakness that progresses to involve distal muscles and causes respiratory and cardiac failure. Biochemical characterization reveals that the mutant myoglobin has altered O2 binding, exhibits a faster heme dissociation rate and has a lower reduction potential compared to wild-type myoglobin. Preliminary studies show that mutant myoglobin may result in elevated superoxide levels at the cellular level. These data define a recognizable muscle disease associated with MB mutation.

  • ELIXIR and Toxicology : a community in development

    • Toxicology has been an active research field for many decades, with academic, industrial and government involvement. Modern omics and computational approaches are changing the field, from merely disease-specific observational models into target-specific predictive models. Traditionally, toxicology has strong links with other fields such as biology, chemistry, pharmacology and medicine. With the rise of synthetic and new engineered materials, alongside ongoing prioritisation needs in chemical risk assessment for existing chemicals, early predictive evaluations are becoming of utmost importance to both scientific and regulatory purposes. ELIXIR is an intergovernmental organisation that brings together life science resources from across Europe. To coordinate the linkage of various life science efforts around modern predictive toxicology, the establishment of a new ELIXIR Community is seen as instrumental. In the past few years, joint efforts, building on incidental overlap, have been piloted in the context of ELIXIR. For example, the EU-ToxRisk, diXa, HeCaToS, transQST, and the nanotoxicology community have worked with the ELIXIR TeSS, Bioschemas, and Compute Platforms and activities. In 2018, a core group of interested parties wrote a proposal, outlining a sketch of what this new ELIXIR Toxicology Community would look like. A recent workshop (held September 30th to October 1st, 2020) extended this into an ELIXIR Toxicology roadmap and a shortlist of limited investment-high gain collaborations to give body to this new community. This Whitepaper outlines the results of these efforts and defines our vision of the ELIXIR Toxicology Community and how it complements other ELIXIR activities.

    • Primary care physicians are uniquely placed to offer holistic, patient-centred care to patients with T2DM. While the recent FDA-mandated cardiovascular outcome trials offer a wealth of data to inform treatment discussions, they have also contributed to increasing complexity in treatment decisions, and in the guidelines that seek to assist in making these decisions. To assist physicians in avoiding treatment inertia, Primary Care Diabetes Europe has formulated a position statement that summarises our current understanding of the available T2DM treatment options in various patient populations. New data from recent outcomes trials is contextualised and summarised for the primary care physician. This consensus paper also proposes a unique and simple tool to stratify patients into 'very high' and 'high' cardiovascular risk categories and outlines treatment recommendations for patients with atherosclerotic cardiovascular disease, heart failure and chronic kidney disease. Special consideration is given to elderly/frail patients and those with obesity. A visual patient assessment tool is provided, and a comprehensive set of prescribing tips is presented for all available classes of glucose-lowering therapies. This position statement will complement the already available, often specialist-focused, T2DM treatment guidelines and provide greater direction in how the wealth of outcome trial data can be applied to everyday practice.

  • Positive Masculinities and Gender-Based Violence Educational Interventions Among Young People: A Systematic Review

    • Hegemonic masculinity has been recognized as contributing to the perpetration of different forms of gender-based violence (GBV). Abandoning hegemonic masculinities and promoting positive masculinities are both strategies used by interventions that foreground a “gender-transformative approach.” Preventing GBV among young people could be strengthened by engaging young men. In this article, we aim to systematically review the primary characteristics, methodological quality, and results of published evaluation studies of educational interventions that aim to prevent different forms of GBV through addressing hegemonic masculinities among young people. Main body: We conducted a systematic review of available literature (2008–2019) using Medline (PubMed), Scopus, Web of Science, PsycInfo, the CINAHL Complete Database, and ERIC as well as Google scholar. The Template for Intervention Description and Replication was used for data extraction, and the quality of the selected studies was analyzed using the Mixed Method Appraisal Tool. More than half of the studies were conducted in Africa ( n = 10/15) and many were randomized controlled trials ( n = 8/15). Most of the studies with quantitative and qualitative methodologies ( n = 12/15) reported a decrease in physical GBV and/or sexual violence perpetration/victimization ( n = 6/15). Longitudinal studies reported consistent results over time. Conclusions: Our results highlight the importance of using a gender-transformative approach in educational interventions to engage young people in critical thinking about hegemonic masculinity and to prevent GBV.

    • Despite public policies and legislative changes aiming to curtail men’s violence against women (VAW) around the world, women continue to be exposed to VAW throughout their life. One in three women in Europe has reported physical or sexual abuse. Men who display unequitable masculinities are more likely to be perpetrators. VAW is increasingly appearing at younger ages. The aims of the project are fourfold: (1) to explore and position the discourses that young people (men and women, 18–24 years) in Sweden, Spain, Ireland and Israel use in their understanding of masculinities, (2) to explore how these discourses influence young people’s attitudes, behaviours and responses to VAW, (3) to explore individual and societal factors supporting and promoting anti-VAW masculinities discourses and (4) to develop actions and guidelines to support and promote anti-VAW masculinities in these settings.Methods and analysisA participatory explorative mixed-method study will be used. In Phase 1, qualitative methods will be used to identify the discourses that young people and stakeholders use to conceptualise masculinities, VAW and the actions that are needed to support and promote antiviolence masculinities. In Phase 2, concept mapping will be used to quantify the coherence, relative importance and perceived relationship between the different actions to support and promote anti-VAW masculinities. Phase 3 is a knowledge creation and translation phase, based on findings from Phases 1 and 2, where actions and guidelines to promote and support anti-VAW masculinities will be developed.Ethics and disseminationEthical clearance has been obtained from ethics review boards in each country. Results will be disseminated through peer-reviewed publications, presentations at international conferences, policy briefs, social media and through the project online hub. With its multicountry approach, our project results seek to inform policies and interventions aimed at promoting discourses which challenge hegemonic masculinities.

This position statement offers a simple and patient-centred clinical decision-making model with practical treatment recommendations that can be widely implemented by primary care clinicians worldwide through shared-decision conversations with their patients. It highlights the importance of managing cardiovascular disease and elevated cardiovascular risk in people with type 2 diabetes and aims to provide innovative risk stratification and treatment strategies that connect patients with the most effective care.

  • Promoting Positive Masculinities to Address Violence Against Women: A Multicountry Concept Mapping Study

    • Interventions engaging men that challenge unequal gender norms have been shown to be effective in reducing violence against women (VAW). However, few studies have explored how to promote anti-VAW positive masculinity in young adults. This study aims to identify key multicountry strategies, as conceived by young adults and other stakeholders, for promoting positive masculinities to improve gender equity and prevent and target VAW. This study (2019–2021) involved young adults (aged 18–24 years) and stakeholders from Ireland, Israel, Spain, and Sweden. We applied concept mapping, a participatory mixed-method approach, in phases: (1) brainstorming, using semi-structured interviews with young adults ( n = 105) and stakeholders ( n = 60), plus focus group discussions ( n = 88), to collect ideas for promoting anti-VAW positive masculinity; (2) development of an online questionnaire for sorting ( n = 201) and rating ideas emerging from brainstorming by importance ( n = 406) and applicability ( n = 360); (3) based on sorting and rating data, creating rating maps for importance and applicability and clusters/strategies using multidimensional scaling and hierarchical cluster analysis with groupwisdom™ software; and (4) interpretation of results with multicountry stakeholders to reach agreement. The cluster map identified seven key strategies (41 actions) for promoting anti-VAW positive masculinities ranked from highest to lowest: Formal and informal education and training; Preventive education and activities in different settings/areas; Skills and knowledge; Empathy, reflection, and understanding; Media and public efforts; Policy, legislation, and the criminal justice system; and Organizational actions and interventions. Pattern matches indicated high agreement between young people and stakeholders in ranking importance ( r = 0.96), but low agreement for applicability ( r = 0.60). Agreement in the total sample on prioritizing statements by importance and applicability was also low ( r = 0.20); only 14 actions were prioritized as both important and applicable. Young people and stakeholders suggested seven comprehensive, multidimensional, multi-setting strategies to facilitate promoting positive masculinity to reduce VAW. Discrepancy between importance and applicability might indicate policy and implementation obstacles.

    • The mechanism of oleate interaction with fluorite is investigated through adsorption, electrokinetic, and diffuse reflectance FT-IR measurements coupled with theoretical thermodynamic chemical equilibrium calculations. The adsorption isotherms exhibit an infinite increase in the slope beyond an oleate density of 10 μmole m-2. This adsorption density corresponds to a bilayer formation of oleate by two-dimensional condensation with a reported molecular coverage area of 33Ao2 (liquid-crystal state). Comparison of the theoretical chemical equilibria with the experimental data shows that the infinite increase in the slope of the isotherms is due to the precipitation of calcium oleate. The solubility product of this three-dimensional phase is found to be in the pK range 14.4-14.5, and agrees with most of the values quoted in the literature. From IR studies, monocoordination of oleate through counter sodium and calcium ions is suggested for the monolayer filling. ζ potentials of fluorite are correlated to the adsorption of oleate up to bilayer formation, and formation on surface of calcium oleate (three-dimensional growth on substrate) at high oleate concentrations. Two-dimensional condensation of oleate on the fluorite surface followed by the association of hydrocarbon chains (tail-tail bond) for the second layer, prior to the precipitation of calcium oleate in the bulk solution, is proposed as the adsorption mechanism.

    • The mechanism of adsorption of oleate onto a synthetic fluorite sample at different solid/liquid ratios in alkaline solutions is elucidated through adsorption isotherms in conjunction with ζ-potential and diffuse reflectance Fourier transform-infrared spectroscopic studies. These studies show that the chemisorption of oleate on surface calcium species followed by the physical adsorption of hydrocarbon chains (bilayer formation) precede the precipitation of calcium oleate in bulk solution. With an increase in the solid/liquid ratio, the bulk precipitation of calcium oleate after bilayer formation is found to be reduced, and at a high solid concentration, the isotherm is seen to level off with bilayer formation. As the solid concentration increases, the concentration of calcium in bulk solution decreases because of its adsorption on the head group of the hydrophilic layer of oleate (bilayer), and hence the extent of bulk precipitation decreases. The adsorption density for a monolayer coverage corresponds to the condensed state of an alkyl chain with a cross-sectional area of 33Ao2 (liquid-crystal state). Application of the two-dimensional condensation model is considered and no evidence for the surface precipitation process is observed.

    • Objetivo: La violencia de compañero íntimo (VCI) contra las mujeres es un importante problema de salud pública y de derechos humanos. Los profesionales de atención primaria tienen un papel clave en su detección y abordaje. El objetivo de este estudio es determinar la frecuencia y los factores asociados a la realización de preguntas de detección de VCI en atención primaria, y describir las principales acciones desarrolladas en los casos identificados.Métodos: Estudio transversal en 15 centros de salud de cuatro comunidades autónomas, con 265 profesionales sanitarios. Recogida de información mediante el cuestionario PREMIS (Physician Readiness to Manage Intimate Partner Violence Survey) autocumplimentado, que incluye variables sobre realización de preguntas para detección, sociodemográficas, nivel de formación/conocimientos en VCI, percepciones y acciones. Se realizó un análisis descriptivo y analítico.Resultados: El 67,2% de los participantes señalan que preguntan sobre VCI en la consulta, siendo las acciones más frecuentes referir/derivar a otros servicios, consejo individual y entrega de información. Se asociaron a mayor probabilidad de indagar sobre VCI tener ≥21 de horas de formación, nivel formativo avanzado, conocer políticas y programas, y disponer de protocolo y recursos apropiados de referencia. La realización de preguntas aumenta a medida que los/las profesionales se sienten formados en VCI y provistos/as de un protocolo de manejo de casos.Conclusiones: Dos terceras partes de los/las profesionales sanitarios/as encuestados/as refirieron indagar sobre VCI. Dada la influencia de la formación en VCI y el conocimiento sobre los recursos para su abordaje, resulta imprescindible continuar invirtiendo en la formación en VCI del personal sanitario.

    • The dependence of shear-flocculation on surface coverage and zeta potential is elucidated using a model system of fluorite/scheelite/oleate. The experiments carried out were (1) shear-flocculation, (2) oleate adsorption/abstraction, (3) flotation of the shear flocs, (4) zeta potential, and (5) turbidity. At a sufficiently high constant shear both an increase in surface coverage and a decrease in zeta potential lead to an increase in the amount of shear flocculating. Manipulation of these two factors permits the turbidity to be reduced from as high as 450 NTU (Θ(surface coverage) = .5; ζ (zeta potential) = -40 mV) to as low as 40 NTU (Θ= 1.2; ζ = -29 mV) in the case of scheelite. With fluorite, turbidity drop is from 120 NTU (Θ= 1; ζ= -40 mV) to 20 NTU (Θ= 1.5; ζ= -19 mV). A close relationship exists between the turbidity measurements and the amount of shear flocculating in percentage. The total flotation of the shear flocs is possible at apparent surface coverages corresponding to a partly formed bilayer
