  • Blood cell DNA methylation biomarkers in preclinical malignant pleural mesothelioma : the EPIC prospective cohort

    • Malignant pleural mesothelioma (MPM) is a rare and aggressive cancer mainly caused by asbestos exposure. Specific and sensitive noninvasive biomarkers may facilitate and enhance screening programs for the early detection of cancer. We investigated DNA methylation (DNAm) profiles in MPM prediagnostic blood samples in a case-control study nested in the European Prospective Investigation into Cancer and nutrition (EPIC) cohort, aiming to characterise DNAm biomarkers associated with MPM. From the EPIC cohort, we included samples from 135 participants who developed MPM during 20 years of follow-up and from 135 matched, cancer-free, controls. For the discovery phase we selected EPIC participants who developed MPM within 5 years from enrolment (n = 36) with matched controls. We identified nine differentially methylated CpGs, selected by 10-fold cross-validation and correlation analyses: cg25755428 (MRI1), cg20389709 (KLF11), cg23870316, cg13862711 (LHX6), cg06417478 (HOOK2), cg00667948, cg01879420 (AMD1), cg25317025 (RPL17) and cg06205333 (RAP1A). Receiver operating characteristic (ROC) analysis showed that the model including baseline characteristics (age, sex and PC1wbc) along with the nine MPM-related CpGs has a better predictive value for MPM occurrence than the baseline model alone, maintaining some performance also at more than 5 years before diagnosis (area under the curve [AUC] < 5 years = 0.89; AUC 5-10 years = 0.80; AUC >10 years = 0.75; baseline AUC range = 0.63-0.67). DNAm changes as noninvasive biomarkers in prediagnostic blood samples of MPM cases were investigated for the first time. Their application can improve the identification of asbestos-exposed individuals at higher MPM risk to possibly adopt more intensive monitoring for early disease identification.

    • In this paper, we will explore the theme of the documentation of 3D cultural heritage assets, not only as entire artefacts but also including the interesting features of the object from an archaeological perspective. Indeed, the goal is supporting archaeological research and curation, providing a different approach to enrich the documentation of digital resources and their components with corresponding measurements, combining semantic and geometric techniques. A documentation scheme based on CIDOC, where measurements on digital data have been included extending CIDOC CRMdig, is discussed. To annotate accurately the components and features of the artefacts, a controlled vocabulary named Cultural Heritage Artefact Partonomy (CHAP) has been defined and integrated into the scheme as a SKOS taxonomy to showcase the proposed methodology. CHAP concerns Coroplastic, which is the study of ancient terracotta figurines and in particular the Cypriot production. Two case studies have been considered: the terracotta statues from the port of Salamis and the small clay statuettes from the Ayia Irini sanctuary. Focussing both on the artefacts and their digital counterparts, the proposed methodology supports effectively typical operations within digital libraries and repositories (e.g. search, part-based annotation), and more specific objectives such as the archaeological interpretation and digitally assisted classification, as proved in a real archaeological scenario. The proposed approach is general and applies to different contexts, since it is able to support any archaeological research where the goal is an extensive digital documentation of tangible findings including quantitative attributes.

  • Towards an Autonomous Unwrapping System for Intralogistics

    • Warehouse logistics is a rapidly growing market for robots. However, one key procedure that has not received much attention is the unwrapping of pallets to prepare them for objects picking. In fact, to prevent the goods from falling and to protect them, pallets are normally wrapped in plastic when they enter the warehouse. Currently, unwrapping is mainly performed by human operators, due to the complexity of its planning and control phases. Autonomous solutions exist, but usually they are designed for specific situations, require a large footprint and are characterized by low flexibility. In this work, we propose a novel integrated robotic solution for autonomous plastic film removal relying on an impedance-controlled robot. The main contribution is twofold: on one side, a strategy to plan Cartesian impedance and trajectory to execute the cut without damaging the goods is discussed; on the other side, we present a cutting device that we designed for this purpose. The proposed solution presents the characteristics of high versatility and the need for a reduced footprint, due to the adopted technologies and the integration with a mobile base. Experimental results are shown to validate the proposed approach.

  • Bortezomib, melphalan, prednisone (VMP) versus melphalan, prednisone, thalidomide (MPT) in elderly newly diagnosed multiple myeloma patients: A retrospective case-matched study

    • Novel agents in combination with melphalan and prednisone (MP) significantly improved progression-free survival (PFS) and overall survival (OS) in multiple myeloma (MM). Randomized trials comparing MP plus bortezomib (VMP) versus MP plus thalidomide (MPT) are lacking. Nine hundred and fifty-six elderly (>65 years) newly diagnosed MM patients from six European randomized trials were retrospectively analyzed and matched for age, albumin, and beta2-microglobulin at diagnosis, 296 patients were selected from the VMP groups, and 294 from MPT. Complete response rate was 21% in the VMP patients and 13% in the MPT patients (P=0.007). After a median follow-up of 34 months (range, 1-92), VMP significantly prolonged both PFS (median 32.5 vs. 22.9 months, HR 0.65; 95% CI 0.52-0.82; P<0.001) and OS (median 79.7 vs. 45.1 months, HR 0.44; 95% CI 0.32-0.59; P<0.001) in comparison with MPT. The benefit in terms of OS of the VMP group was quite similar among patients with different risk factors defined by sex, ISS, ECOG performance status, or serum creatinine but not among patients 75 years. Multivariate analysis confirmed that VMP was an independent predictor of longer PFS and OS. In a control-case matched analysis, PFS and OS were prolonged in patients who received VMP in comparison with those treated with MPT. Am. J. Hematol. 89:355-362, 2014.
