    • Here, we describe the results of a genome-wide study conducted in 11 939 coronavirus disease 2019 (COVID-19) positive cases with an extensive clinical information that were recruited from 34 hospitals across Spain (SCOURGE consortium). In sex-disaggregated genome-wide association studies for COVID-19 hospitalization, genome-wide significance (P < 5 × 10−8) was crossed for variants in 3p21.31 and 21q22.11 loci only among males (P = 1.3 × 10−22 and P = 8.1 × 10−12, respectively), and for variants in 9q21.32 near TLE1 only among females (P = 4.4 × 10−8). In a second phase, results were combined with an independent Spanish cohort (1598 COVID-19 cases and 1068 population controls), revealing in the overall analysis two novel risk loci in 9p13.3 and 19q13.12, with fine-mapping prioritized variants functionally associated with AQP3 (P = 2.7 × 10−8) and ARHGAP33 (P = 1.3 × 10−8), respectively. The meta-analysis of both phases with four European studies stratified by sex from the Host Genetics Initiative (HGI) confirmed the association of the 3p21.31 and 21q22.11 loci predominantly in males and replicated a recently reported variant in 11p13 (ELF5, P = 4.1 × 10−8). Six of the COVID-19 HGI discovered loci were replicated and an HGI-based genetic risk score predicted the severity strata in SCOURGE. We also found more SNP-heritability and larger heritability differences by age (<60 or ≥60 years) among males than among females. Parallel genome-wide screening of inbreeding depression in SCOURGE also showed an effect of homozygosity in COVID-19 hospitalization and severity and this effect was stronger among older males. In summary, new candidate genes for COVID-19 severity and evidence supporting genetic disparities among sexes are provided.

  • Applying digital twins for the management of information in turnaround event operations in commercial airports

    • The aerospace sector is one of the many sectors in which large amounts of data are generated. Thanks to the evolution of technology, these data can be exploited in several ways to improve the operation and management of industrial processes. However, to achieve this goal, it is necessary to define architectures and data models that allow to manage and homogenise the heterogeneous data collected. In this paper, we present an Airport Digital Twin Reference Conceptualisation's and data model based on FIWARE Generic Enablers and the Next Generation Service Interfaces-Linked Data standard. Concretely, we particularise the Airport Digital Twin to improve the efficiency of flight turnaround events. The architecture proposed is validated in the Aberdeen International Airport with the aim of reducing delays in commercial flights. The implementation includes an application that shows the real state of the airport, combining two-dimensional and three-dimensional virtual reality representations of the stands, and a mobile application that helps ground operators to schedule departure and arrival flights.

  • APOE genotype and extent of bleeding and outcome in lobar intracerebral haemorrhage: a genetic association study

    • Background Carriers of APOE epsilon 2 and epsilon 4 have an increased risk of intracerebral haemorrhage (ICH) in lobar regions, presumably because of the effects of these gene variants on risk of cerebral amyloid angiopathy. We aimed to assess whether these variants also associate with severity of ICH, in terms of haematoma volume at presentation and subsequent outcome. Methods We investigated the association of APOE epsilon 2 and epsilon 4 with ICH volume and outcomes in patients with primary ICH in three phases: a discovery phase of 865 individuals of European ancestry from the Genetics of Cerebral Hemorrhage on Anticoagulation study, and replication phases of 946 Europeans (replication 1) and 214 African-Americans (replication 2) from an additional six studies. We also assessed the association of APOE variants with ICH volume and outcomes in meta-analyses of results from all three phases, and the association of APOE epsilon 4 with mortality in a further meta-analysis including data from previous reports. Admission ICH volume was quantified on CT scan. We assessed functional outcome (modified Rankin scale score 3-6) and mortality at 90 days. We used linear regression to establish the effect of genotype on haematoma volume and logistic regression to assess the effect on outcome from ICH. Findings For patients with lobar ICH, carriers of the APOE epsilon 2 allele had larger ICH volumes than did non-carriers in the discovery phase (p=2. 5x10(-5)), in both replication phases (p=0.008 in Europeans and p=0.016 in African-Americans), and in the meta-analysis (p=3.2x10(-8)). In the meta-analysis, each copy of APOE epsilon 2 increased haematoma size by a mean of 5.3 mL (95% CI 4.7-5.9; p=0.004). Carriers of APOE epsilon 2 had increased mortality (odds ratio [OR] 1.50, 95% CI 1.23-1.82; p=2.45x10(-5)) and poorer functional outcomes (modified Rankin scale score 3-6; 1-52, 1.25-1.85; p=1.74x10(-5)) compared with non-carriers after lobar ICH. APOE epsilon 4 was not associated with lobar ICH volume, functional outcome, or mortality in the discovery phase, replication phases, or meta-analysis of these three phases; in our further meta-analysis of 2194 patients, this variant did not increase risk of mortality (1.08,0.86-1.36; p=0.52). APOE allele variants were not associated with deep ICH volume, functional outcome, or mortality. Interpretation Vasculopathic changes associated with the APOE epsilon 2 allele might have a role in the severity and clinical course of lobar ICH. Screening of patients who have ICH to identify the epsilon 2 variant might allow identification of those at increased risk of mortality and poor functional outcomes.

    • CRIRES+ extended the capabilities of CRIRES, the CRyogenic InfraRed Echelle Spectrograph. It transformed this VLT instrument into a cross-dispersed spectrograph to increase the wavelength range that is covered simultaneously by a factor of ten. In addition, a new detector focal plane array of three Hawaii 2RG detectors with a 5.3 mu m cut-off wavelength replaced the existing detectors. Amongst many other improvements a new spectropolarimetric unit was added and the calibration system has been enhanced. The instrument was installed at the VLT on Unit Telescope 3 beginning of 2020 and successfully commissioned and verified for science operations during 2021, partly remote from Europe due to the pandemic. The instrument was subsequently offered to the community from October 2021 onwards. This article describes the performance and capabilities of this development and presents on sky results.

    • Ongoing efforts within synthetic and systems biology have been directed towards the building of artificial computational devices using engineered biological units as basic building blocks. Such efforts, inspired in the standard design of electronic circuits, are limited by the difficulties arising from wiring the basic computational units (logic gates) through the appropriate connections, each one to be implemented by a different molecule. Here, we show that there is a logically different form of implementing complex Boolean logic computations that reduces wiring constraints thanks to a redundant distribution of the desired output among engineered cells. A practical implementation is presented using a library of engineered yeast cells, which can be combined in multiple ways. Each construct defines a logic function and combining cells and their connections allow building more complex synthetic devices. As a proof of principle, we have implemented many logic functions by using just a few engineered cells. Of note, small modifications and combination of those cells allowed for implementing more complex circuits such as a multiplexer or a 1-bit adder with carry, showing the great potential for re-utilization of small parts of the circuit. Our results support the approach of using cellular consortia as an efficient way of engineering complex tasks not easily solvable using single-cell implementations.

  • The Effect of Hydrocarbon Impurities in the Hydrogen Fuel on the Anode Activity in PEMFC

    • The reformate fuel cell has recently gained increasing attention both for APUs in vehiclesoperating on diesel and in stationary applications such as micro-CHP operating on naturalor biogas. In addition to hydrogen gas, reformate contain considerable amounts of CO2,nitrogen, water vapour and traces of CO, sulphur species and hydrocarbons. CO and H2Sare well known poisons to the anode [1] but the influence of hydrocarbon species in thefuel cell has not been much investigated. We have previously investigated toluene [2] andethene [3] on the anode Pt/C catalyst in the PEM fuel cell. In this paper we will discuss theinfluences of alkenes and alkanes in the light of some novel results on the effect ofpropene, propane and methane in the PEM fuel cell. We have especially focused on theadsorption and deactivation phenomena of low concentrations of contaminant on a Pt/Ccatalyst. In the experiments, in situ stripping voltammetry and on-line mass spectrometerwere employed. The effects of adsorption potential and temperature are discussed. Weshow that propene is more poisonous to the Pt/C catalyst than ethene as it is adsorbed onthe catalyst surface within the Hupd region and forms an adlayer that can be oxidized in twosteps between 0.5-1 V (at 80°C, 90%RH) or be hydrogenated to propane in the Hupd regionand in the presence of hydrogen.
