    • Implementing information and communications technology (ICT) at scale requires evaluation processes to capture the impact on users as well as the infrastructure into which it is being introduced. For older adults living with cognitive impairment, this requires evaluation that can accommodate different levels of cognitive impairment, alongside input from family and formal caregivers, plus stakeholder organisations. The European Horizon 2020 project INdependent LIving support Functions for the Elderly (IN LIFE) set out to integrate 17 technologies into a single digital platform for older people living with cognitive impairment plus their families, care providers and stakeholders. The IN LIFE evaluation took place across six national pilot sites to examine a number of variables including impact on the users, user acceptance of the individual services and the overall platform, plus the economic case for the IN LIFE platform. The results confirmed the interest and need among older adults, family caregivers, formal caregivers and stakeholders, for information and communications technology (ICT). Relative to the baseline, quality of life improved and cognition stabilised; however, there was an overall reluctance to pay for the platform. The findings provide insights into existing barriers and challenges for adoption of ICT for older people living with cognitive impairment.

    • In this paper, we present the European H2020 project INLIFE (INdependent LIving support Functions for the Elderly). The project brought together 20 partners from nine countries with the goal of integrating into a common ICT platform a range of technologies intended to assist community-dwelling older people with cognitive impairment. The majority of technologies existed prior to INLIFE and a key goal was to bring them together in one place along with a number of new applications to provide a comprehensive set of services. The range of INLIFE services fell into four broad areas: Independent Living Support, Travel Support, Socialization and Communication Support and Caregiver Support. These included security applications, services to facilitate interactions with formal and informal caregivers, multilingual conversation support, web-based physical exercises, teleconsultations, and support for transport navigation. In total, over 2900 people participated in the project; they included elderly adults with cognitive impairment, informal caregivers, healthcare professionals, and other stakeholders. The aim of the study was to assess whether there was improvement/stabilization of cognitive/emotional/physical functioning, as well as overall well-being and quality of life of those using the INLIFE services, and to assess user acceptance of the platform and individual services. The results confirm there is a huge interest and appetite for technological services to support older adults living with cognitive impairment in the community. Different services attracted different amounts of use and evaluation with some proving extremely popular while others less so. The findings provide useful information on the ways in which older adults and their families, health and social care services and other stakeholders wish to access technological services, what sort of services they are seeking, what sort of support they need to access services, and how these services might be funded.

  • ANGPTL4 gene E40K variation protects against obesity-associated dyslipidemia in participants with obesity

    • Objective ANGPTL4 inhibits lipoprotein lipase in adipose tissue, regulating plasma triglycerides levels. In persons with obesity plasma ANGPTL4 levels have been positively correlated with body fat mass, TG levels and low HDL. A loss-of-function E40K mutation in ANGPTL4 prevents LPL inhibition, resulting in lower TGs and higher HDLc in the general population. Since obesity determines metabolic alterations and consequently is a major risk factor for cardiovascular disease, the aim was to explore if obesity-related metabolic abnormalities are modified by the ANGPTL4-E40K mutation. Methods ANGPTL4-E40K was screened in 1206 Italian participants, of which 863 (71.5%) with obesity. All subjects without diabetes underwent OGTT with calculation of indices of insulin-sensitivity. Results Participants with obesity carrying the E40K variant had significantly lower TG (p = 0.001) and higher HDLc levels (p = 0.024). Also in the whole population low TGs and high HDLc were confirmed in E40K carriers. In the obese subpopulation it was observed that almost all E40K carriers were within the lowest quartile of TGs (p = 1.1 x 10(-9)). E40K had no substantial effect of on glucose metabolism. Finally, none of the obese E40K carriers had T2D, and together with the favourable lipid profile, they resemble a metabolically healthy obese (MHO) phenotype, compared to 38% of E40E wild-type obese that had diabetes and/or dyslipidaemia (p = 0.0106). Conclusions In participants with obesity the ANGPTL4-E40K variant protects against dyslipidemia. The phenotype of obese E40K carriers is that of a patient with obesity without metabolic alterations, similar to the phenotype described as metabolic healthy obesity.

  • Magnetism between magnetic adatoms on monolayer NbSe2

    • In this work, we report on an ab-initio computational study of the electronic and magnetic properties of transition metal adatoms on a monolayer of NbSe2. We demonstrate that Cr, Mn, Fe and Co prefer all to sit above the Nb atom, where the d states experience a substantial hybridization. The inter-atomic exchange coupling is shown to have an oscillatory nature accompanied by an exponential decay, in accordance with what theory predicts for a damped Ruderman-Kittel-Kasuya-Yosida interaction. Our results indicate that the qualitative features of the magnetic coupling for the four investigated adatoms can be connected to the fine details of their Fermi surface. In particular, the oscillations of the exchange in Fe and Co are found to be related to a single nesting vector, connecting large electrons and hole pockets. Most interestingly, this behavior is found to be unaffected by changes induced on the height of the impurity, which makes the magnetism robust to external perturbations. Considering that NbSe2 is a superconductor down to a single layer, our research might open the path for further research into the interplay between magnetic and superconducting characteristics, which could lead to novel superconductivity engineering.

  • Emergent half-metal with mixed structural order in (111)-oriented (LaMnO3)2n|(SrMnO3)n superlattices

    • Using first-principles techniques, we study the structural, magnetic, and electronic properties of (111)-oriented (LaMnO3)2n |(SrMnO3)n superlattices of varying thickness (n = 2, 4, 6). We find that the properties of the thinnest superlattice (n = 2) are similar to the celebrated half-metallic ferromagnetic alloy La2/3Sr1/3MnO3, with quenched Jahn-Teller distortions. At intermediate thickness (n = 4), the a-a-a- tilting pattern transitions to the a-a-c+ tilting pattern, driven by the lattice degrees of freedom in the LaMnO3 region. The emergence of the Jahn-Teller modes and the spatial extent needed for their development play a key role in this structural transition. For the largest thickness considered (n = 6), we unveil an emergent separation of Jahn-Teller and volume-breathing orders in the ground-state structure with the a-a-c+ tilting pattern, whereas it vanishes in the antiferromagnetic configurations. The ground state of all superlattices is half-metallic ferromagnetic, not affected by the underlying series of structural transitions. Overall, these results outline a thickness-induced crossover between the physical properties of bulk La2/3Sr1/3MnO3 and bulk LaMnO3.

  • Unveiling hidden charge density waves in single-layer NbSe2 by impurities

    • We employ ab initio calculations to investigate the charge density waves in single-layer NbSe2, and we explore how they are affected by transition metal atoms. Our calculations reproduce the observed orthorhombic phase in single-layer NbSe2 in the clean limit, establishing the energy order between three different distorted structures, two consisting of triangular Nb-Nb clusters and a third, energetically unfavored, consisting of hexagonal Nb-Nb clusters. Such energy order, in agreement with known experimental work, is reversed by the adsorption of Co and Mn, which favor the formation of hexagonal Nb-Nb clusters; this CDW structure is indeed allowed from a symmetry point of view but hidden in pure single layers because it is at a higher energy. The other adsorbates, K and Ga, still favor one of the triangular Nb-Nb cluster, while suppressing the other. We report how the energy difference between such distorted structure varies with these adsorbates. Furthermore, transition metals induce magnetism and favor the reduction of the symmetry of the charge density distribution.

  • Delaying transition to turbulence by a passive mechanism

    • Reducing skin friction is important in nature and in many technological applications. This reduction may be achieved by reducing stresses in turbulent boundary layers, for instance tailoring biomimetic rough skins. Here we take a second approach consisting of keeping the boundary layer laminar as long as possible by forcing small optimal perturbations. Because of the highly non-normal nature of the underlying linearized operator, these perturbations are highly amplified and able to modify the mean velocity profiles at leading order. We report results of wind-tunnel experiments in which we implement this concept by using suitably designed roughness elements placed on the skin to enforce nearly optimal perturbations. We show that by using this passive control technique it is possible to sensibly delay transition to turbulence.

    • An experimental activity aimed at the development of a passive method for viscous drag reduction in an aerodynamic body is presented. The reduction is achieved by delaying the laminar-turbulent transition of the boundary layer. The research is motivated by previous linear stability analysis where it was demonstrated that stable streaks in a boundary layer are capable to stabilize the growth of Tollmien-Schlichting waves. In this paper the results of a series of experiments on a flat plate are presented. Firstly, it is shown that by means of suitable roughness elements it is possible to generate stable steady streaks of amplitudes up to 12% of the free stream velocity. Secondly, it is demonstrated that the generated streaks are really effective in reducing the growth of the Tollmien-Schlichting waves as predicted by the theory. Finally, it is shown that using this passive control technique it is possible to sensibly delay transition to turbulence.

    • It has recently been found by using temporal and spatial numerical simulations that steady optimal streaks of moderate amplitude, i.e., sufficiently large but not exceeding the critical amplitude for the inflectional instability, are able to reduce the growth of Tollmien-Schlichting (TS) waves up to their complete suppression. This investigation aims at experimentally verifying this stabilizing effect by generating stable and symmetric, close to sinusoidal, streaks of moderate amplitudes (similar to 12% of the free-stream velocity) by means of a spanwise array of cylindrical roughness elements. The three-dimensional (3D) streaky base flow is then subjected to a secondary instability generated through a spanwise slot in the plate by means of regulated blowing and suction. In this study the stabilizing role of the streaks on TS waves is unambiguously confirmed and by increasing the height of the roughness elements, thus inducing larger amplitude streaks, we are also able to show that the stabilizing action on the TS waves increases with the streak amplitude. These results are the first to confirm the numerical predictions reported in earlier works. The full cross-stream plane has been measured at different downstream positions allowing a complete evaluation and comparison of the different amplitude measures used in previous experimental works. Furthermore, theoretical impulse response analysis and stability calculations are applied to the present experimental streaky base flow enabling a qualitative comparison of the 3D modulated TS wave distribution.

  • Subcellular distribution of melatonin receptors in human parotid glands

    • The hormone melatonin influences oral health through a variety of actions, such as anti-inflammatory, anti-oxidant, immunomodulatory and antitumour. Many of these melatonin functions are mediated by a family of membrane receptors expressed in the oral epithelium and salivary glands. Using immunoblotting and immunohistochemistry, recent studies have shown that the melatonin membrane receptors, MT1 and MT2, are present in rat and human salivary glands. To date, no investigation has dealt with the ultrastructural distribution of the melatonin receptors. This was the aim of the present study, using the immunogold method applied to the human parotid gland. Reactivity to MT1 and, with less intensity, to MT2 appeared in the secretory granules of acinar cells and in the cytoplasmic vesicles of both acinar and ductal cells. Plasma membranes were also stained, albeit slightly. The peculiar intracytoplasmic distribution of these receptors may indicate that there is an uptake/transport system for melatonin from the circulation into the saliva.

    • Human stem cells have the potential to transform medicine. However, hurdles remain to ensure that manufacturing processes produce safe and effective products. A thorough understanding of the biological processes occurring during manufacture is fundamental to assuring these qualities and thus, their acceptability to regulators and clinicians. Leaders in both human pluripotent and somatic stem cells, were brought together with experts in clinical translation, biomanufacturing and regulation, to discuss key issues in assuring appropriate manufacturing conditions for delivery of effective and safe products from these cell types. This report summarizes the key issues discussed and records consensus reached by delegates and emphasizes the need for accurate language and nomenclature in the scientific discourse around stem cells.
