    • Using a sample of 1:06 X 10(8) psi(3686) events collected with the BESIII detector, we present the first observation of the decays of psi(3686) -> Lambda(Sigma) over bar (+) pi(-) + c.c. and psi(3686) -> Lambda(Sigma) over bar (-) pi(+) + c.c. The branching fractions are measured to be B(psi(3686) -> Lambda(Sigma) over bar (+) pi(-) + c.c.) = (1.40 +/- 0.03 +/- 0.13) X 10(-4) and B(psi(3686) -> Lambda (Sigma) over bar (-) pi(+) + c.c.) = (1.54 +/- 0.04 +/- 0.13) X 10(-4) where the first errors are statistical and the second ones systematic.

  • Search for eta(c)(2S)h(c) -> p(p)over-bar decays and measurements of the chi(cJ) -> p(p)over-bar branching fractions

    • Using a sample of 1.06 x 10(8)psi(3686) events collected with the BESIII detector at BEPCII, the decays eta(c)(2S) -> p (p) over bar and h(c) -> p (p) over bar are searched for, where eta(c)(2S) and h(c) are reconstructed in the decay chains psi(3686) -> gamma eta(c)(2S), eta(c)(2S) -> p (p) over bar and psi(3686) -> pi(0)h(c), h(c) -> p (p) over bar, respectively. No significant signals are observed. The upper limits of the product branching fractions are determined to be B(psi(3686) -> gamma eta(c)(2S)) x B(eta(c)(2S) -> p (p) over bar) < 1.4 x 10(-6) and B(psi(3686) -> pi(0)h(c)) x B(h(c) -> p<(p)over bar>) < 1.3 x 10(-7) at the 90% C.L.. The branching fractions for chi(cJ) -> p<(p)over bar> (J = 0, 1, 2) are also measured to be (24.5 +/- 0.8 +/- 1.3, 8.6 +/- 0.5 +/- 0.5, 8.4 +/- 0.5 +/- 0.5) x 10(-5), which are the world's most precise measurements.

The new families, Ascocylindricaceae, Caryosporaceae and Wicklowiaceae (Ascomycota) are introduced based on their distinct lineages and unique morphology.   • The Faces of Fungi database: fungal names linked with morphology, phylogeny and human impacts

    • Taxonomic names are key links between various databases that store information on different organisms. Several global fungal nomenclural and taxonomic databases (notably Index Fungorum, Species Fungorum and MycoBank) can be sourced to find taxonomic details about fungi, while DNA sequence data can be sourced from NCBI, EBI and UNITE databases. Although the sequence data may be linked to a name, the quality of the metadata is variable and generally there is no corresponding link to images, descriptions or herbarium material. There is generally no way to establish the accuracy of the names in these genomic databases, other than whether the submission is from a reputable source. To tackle this problem, a new database (FacesofFungi), accessible at www.facesoffungi.org (FoF) has been established. This fungal database allows deposition of taxonomic data, phenotypic details and other useful data, which will enhance our current taxonomic understanding and ultimately enable mycologists to gain better and updated insights into the current fungal classification system. In addition, the database will also allow access to comprehensive metadata including descriptions of voucher and type specimens. This database is user-friendly, providing links and easy access between taxonomic ranks, with the classification system based primarily on molecular data (from the literature and via updated web-based phylogenetic trees), and to a lesser extent on morphological data when molecular data are unavailable. In FoF species are not only linked to the closest phylogenetic representatives, but also relevant data is provided, wherever available, on various applied aspects, such as ecological, industrial, quarantine and chemical uses. The data include the three main fungal groups (Ascomycota, Basidiomycota, Basal fungi) and fungus-like organisms. The FoF webpage is an output funded by the Mushroom Research Foundation which is an NGO with seven directors with mycological expertise. The webpage has 76 curators, and with the help of these specialists, FoF will provide an updated natural classification of the fungi, with illustrated accounts of species linked to molecular data. The present paper introduces the FoF database to the scientific community and briefly reviews some of the problems associated with classification and identification of the main fungal groups. The structure and use of the database is then explained. We would like to invite all mycologists to contribute to these web pages.

    • Twenty years have passed since first light for the Sloan Digital Sky Survey (SDSS). Here, we release data taken by the fourth phase of SDSS (SDSS-IV) across its first three years of operation (2014 July-2017 July). This is the third data release for SDSS-IV, and the 15th from SDSS (Data Release Fifteen; DR15). New data come from MaNGA-we release 4824 data cubes, as well as the first stellar spectra in the MaNGA Stellar Library (MaStar), the first set of survey-supported analysis products (e.g., stellar and gas kinematics, emission-line and other maps) from the MaNGA Data Analysis Pipeline, and a new data visualization and access tool we call "Marvin." The next data release, DR16, will include new data from both APOGEE-2 and eBOSS; those surveys release no new data here, but we document updates and corrections to their data processing pipelines. The release is cumulative; it also includes the most recent reductions and calibrations of all data taken by SDSS since first light. In this paper, we describe the location and format of the data and tools and cite technical references describing how it was obtained and processed. The SDSS website (www.sdss.org) has also been updated, providing links to data downloads, tutorials, and examples of data use. Although SDSS-IV will continue to collect astronomical data until 2020, and will be followed by SDSS-V (2020-2025), we end this paper by describing plans to ensure the sustainability of the SDSS data archive for many years beyond the collection of data.

  • Bidirectional Mendelian Randomisation Analysis Provides Evidence for the Causal Involvement of Dysregulation of CXCL9, CCL11 and CASP8 in the Pathogenesis of Ulcerative Colitis

    • Background and Aims Systemic inflammation is well recognised to be associated with ulcerative colitis [UC], but whether these effects are causal or consequential remains unclear. We aimed to define potential causal relationship of cytokine dysregulation with different tiers of evidence. Methods We first synthesised serum proteomic profiling data from two multicentred observational studies, in which a panel of systemic inflammatory proteins was analysed to examine their associations with UC risk. To further dissect observed associations, we then performed a bidirectional two-sample Mendelian randomisation [TSMR] analysis from both forward and reverse directions using five genome-wide association study [GWAS] summary level data for serum proteomic profiles and the largest GWAS of 28 738 European-ancestry individuals for UC risk. Results Pooled analysis of serum proteomic data identified 14 proteins to be associated with the risk of UC. Forward MR analysis using only cis-acting protein quantitative trait loci [cis-pQTLs] or trans-pQTLs further validated causal associations of two chemokines and the increased risk of UC: C-X-C motif chemokine ligand 9 [CXCL9] [OR 1.45, 95% CI 1.08, 1.95, p = 0.012] and C-C motif chemokine ligand 11 [CCL11] [OR 1.14, 95% CI 1.09, 1.18, p = 3.89 x 10(-10)]. Using both cis- and trans-acting pQTLs, an association of caspase-8 [CASP8] [OR 1.04, 95% CI 1.03, 1.05, p = 7.63 x 10(-19)] was additionally identified. Reverse MR did not find any influence of genetic predisposition to UC on any of these three inflammation proteins. Conclusion Pre-existing elevated levels of CXCL9, CCL11 and CASP8 may play a role in the pathogenesis of UC.

  • Interfacial Property Tuning Enables Copper Electrodes in High-Performance n-i-p Perovskite Solar Cells

    • Developing cost-effective metal electrodes is essential for reducing the overall cost of perovskite solar cells (PSCs). Although copper is highly conductive and economical, it is rarely used as a positive electrode in efficient n-i-p PSCs due to its unmatched Fermi level and low oxidation threshold. We report herein that modification for the inner surface of electrodes using mercaptopyridine-based molecules readily tunes the electronic and chemical properties of copper, which has been achieved by fine-tuning the substituents of mercaptopyridines. The systematic adjustment for the Fermi level and oxidation potential of copper facilitates interfacial hole extraction and enhances the oxidation resistance of copper electrodes, which enables pure copper electrodes to be used in high-performance n-i-p PSCs with different hole transport materials. The resulting PSCs with copper electrodes display excellent power conversion efficiency and long-term stability, even comparable to those of the gold electrodes, showing great potential in the manufacturing and commercialization of PSCs.

  • Adsorption, aggregation and phase separation in colloidal systems

    • The thesis presents work regarding amphiphilic molecules associated in aqueous solution or at the liquid/solid interface. Two main topics are included: the temperature-dependent behavior of micelles and the adsorption of dispersants on carbon nanotube (CNT) surfaces. Various NMR methods were used to analyze those systems, such as chemical shift detection, spectral intensity measurements, spin relaxation and, in particular, self-diffusion experiments. Besides this, small angle X-ray scattering (SAXS) was also applied for structural characterization. A particular form of phase transition, core freezing, was detected as a function of temperature in micelles composed by a single sort of Brij-type surfactants. In mixed micelles, that phase transition still occurs accompanied by a reversible segregation of different surfactants into distinct aggregates. Adding a hydrophobic solubilizate shifts the core freezing point to a lower temperature. Upon lowering the temperature to the core freezing point, the solubilizate is released. The temperature course of the release curves with different initial solubilizate loadings is rationalized in terms of a temperature-dependent loading capacity. The behavior of amphiphilic dispersant molecules in aqueous dispersions of carbon nanotubes (CNTs) has been investigated with a Pluronic-type block copolymer as frequent model dispersant. Detailed dispersion curves were recorded and the distribution of the dispersant among different available environments was analyzed. The amount of dispersed CNT was shown to be defined by a complex interplay of several factors during the dispersion process such as dispersant concentration, sonication time, centrifugation and CNT loading. In the dispersion process, high amphiphilic concentration is required because the pristine CNT surfaces made available by sonication must be rapidly covered by dispersants to avoid their re-attachment. In the prepared dispersions, the competitive adsorption of possible dispersants was investigated that provided information about the relative strength of the interaction of those with the nanotube surfaces. Anionic surfactants were found to have a strong tendency to replace Pluronics, which indicates a strong binding of those surfactants. CNTs were dispersed in an epoxy resin to prepare nanotube-polymer composites. The molecular mobility of epoxy was investigated and the results demonstrated the presence of loosely associated CNT aggregates within which the molecular transport of epoxy is slow because of strong attractive intermolecular interactions between epoxy and the CNT surface. The rheological behavior is dominated by aggregate-aggregate jamming.

    • Debundling and dispersing single-walled carbon nanotubes (SWNTs) is essential for applications, but the process is not well understood. In this work, aqueous SWNT dispersions were produced by sonicating pristine SWNT powder in the presence of an amphiphilic triblock copolymer (Pluronic F127) as dispersant. Upon centrifugation, one obtains a supernatant with suspended individual tubes and thin bundles and a precipitate with large bundles (and impurities). In the supernatant, that constitutes the final dispersion, we determined the dispersed SWNT concentration by thermogravi-metric analysis (TGA) and UV-vis spectroscopy, and the dispersant concentration by NMR The fraction of dispersant adsorbed at the SWNT surface was obtained by H-1 diffusion NMR Sigmoidal dispersion curves recording the concentration of dispersed SWNTs as a function of supernatant dispersant concentration were obtained at different SWNT loadings and sonication times. As SWNT bundles are debundled into smaller and smaller ones, the essential role of the dispersant is to sufficiently quickly cover the freshly exposed surfaces created by shear forces induced during sonication. Primarily kinetic reasons are behind the need for dispersant concentrations required to reach a substantial SWNT concentration. Centrifugation sets the size threshold below which SWNT particles are retained in the dispersion and consequently determines the SWNT concentration as a function of sonication time.

  • Molecular and Segmental Orientational Order in a Smectic Mesophase of a Thermotropic Ionic Liquid Crystal

    • We investigate conformational dynamics in the smectic A phase formed by the mesogenic ionic liquid 1-tetradecyl-3-methylimidazolium nitrate. Solid-state high-resolution C-13 nuclear magnetic resonance (NMR) spectra are recorded in the sample with the mesophase director aligned in the magnetic field of the NMR spectrometer. The applied NMR method, proton encoded local field spectroscopy, delivers heteronuclear dipolar couplings of each C-13 spin to its H-1 neighbours. From the analysis of the dipolar couplings, orientational order parameters of the C-H bonds along the hydrocarbon chain were determined. The estimated value of the molecular order parameter S is significantly lower compared to that in smectic phases of conventional non-ionic liquid crystals.

  • NMR Spectroscopic Study of Orientational Order in Imidazolium-Based Ionic Liquid Crystals

    • We report on molecular and local orientational order of a series of imidazolium-based ionic liquid crystals exhibiting layered smectic A mesophase. Materials constituting of 1-dodecyl-3-methylimidazolium cation, and different counter-ions, were investigated. We apply two-dimensional C-13-H-1 dipolar NMR spectroscopy to quantify orientational order of C-H bonds of the organic cation. The experimental data supported the structural model of the interdigitated chains aligned with the smectic layer normal. Molecular order parameter S was found to increase in the anion sequence BF4- < I- < Br- < Cl-. This trend correlates well with ionic radius, negative charge delocalization, and hydrogen-bonding properties of the anions.

  • Release of Solubilizate from Micelle upon Core Freezing

    • By combining NMR (yielding H-1 chemical shift, spin relaxation, and self-diffusion data) and small-angle X-ray scattering experiments, we investigate the complex temperature dependence of the molecular and aggregate states in aqueous solutions of the surfactant [CH3(CH2)(17)(OCH2CH2)(20)OH], abbreviated as C18E20, and.hexamethyldisiloxane, HMDSO. The latter molecule serves as a model for hydrophobic solubilizates. Previously, the pure micellar solution was demonstrated to exhibit core freezing at approximately 7-8 degrees C. At room temperature, we find that HMDSO solubilizes at a volume fraction of approximately 10% in the core of the C18E20 micelles, which consists of molten and thereby highly mobile alkyl chains. Upon lowering the temperature, core freezing is found, just like in pure micelles, but at a temperature shifted significantly to 3 degrees C. The frozen cores contain immobile alkyl chains and exhibit a higher density but are essentially devoid (volume fraction below 1%) of the solubilizate. The latter molecules are released, first gradually and then rather steeply, from the core in the temperature range that is roughly delimited by the two core freezing temperatures, one for pure micelles and one for micelles with solubilizates. The release behavior of systems with different initial HMDSO loading follows the same master curve. This feature is rationalized in terms of loading capacity being strongly temperature dependent: upon lowering the temperature, release commences once the loading capacity descends below the actual solubilizate content. The sharp release curves and the actual release mechanism with its molecular features shown in rich detail have some bearing on a diverse class of possible applications.

  • The dispersion process of carbon nanotubes sonicated in aqueous solutions of a dispersant

    • Single-walled carbon nanotube (SWNT) dispersions are created by sonicating pristine SWNT powders added to aqueous solutions of the dispersant block copolymer Pluronic F127. In those dispersions, the amount of the dispersed SWNT is determined by the combination of TGA and UV-Vis methods, while the dispersant concentration is estimated by 1H NMR spectroscopy. In addition, the amount of dispersant adsorbed at the SWNT surface is obtained by 1H NMR diffusion experiments. A part of the dispersant is taken up by non-dispersed and precipitated particles. Dispersion curves recording the amount of the dispersed SWNT as a function of either the initial dispersant concentration or the final dispersant concentration are obtained at different initial SWNT loadings and sonication times. The results show in detail the way the original SWNT particles are divided into smaller and smaller sizes thereby increasing the available SWNT surface to be covered by dispersant. Centrifugation sets the size-threshold above which SWNT particles are retained in the dispersion which determined the SWNT content as a function of sonication time.

    • Submerged aquatic vegetation (SAV) is one of the main producers in inland lakes. Tracking the temporal and spatial changes in SAV is crucial for the identification of state changes in lacustrine ecosystems, such as changes in light, nutrients, and temperature. However, the available SAV classification algorithms based on remote sensing are highly dependent on field survey data and/or human interventions, prohibiting the extraction of large-scale and/or long-term patterns. Here, we developed an automatic SAV classification algorithm using Landsat imagery, where the thresholds of two key parameters (the floating algae index (FAI) and reflectance in the shortwave-infrared (SWIR) band) are automatically determined. The algorithm was applied to eight Landsat images of four Yangtze Plain lakes and obtained a mean producer accuracy of 82.9% when gauged against field-surveyed datasets. The algorithm was further employed to obtain long-term SAV areal data from Changdang Lake on the Yangtze Plain from 1984 to 2018, and the result was highly consistent with lake transparency data. Numerical simulations indicated that our developed algorithm is insensitive to the Chl-a concentration of the water column. Yet, it has a detection limit of ~0.35 m below the water surface, and such a limit changes with different fractions of vegetation coverage within a pixel. The automatic classification algorithm proposed in this study has the potential to obtain the temporal and spatial distribution patterns of SAV in other shallow lakes where SAV grows in lakes sharing similar hydrological characteristics as the lakes in the Yangtze Plain.

    • Viscosity is one of the most important fundamental properties of fluids. However, accurate acquisition of viscosity for ionic liquids (ILs) remains a critical challenge. In this study, an approach integrating prior physical knowledge into the machine learning (ML) model was proposed to predict the viscosity reliably. The method was based on 16 quantum chemical descriptors determined from the first principles calculations and used as the input of the ML models to represent the size, structure, and interactions of the ILs. Three strategies based on the residuals of the COSMO-RS model were created as the output of ML, where the strategy directly using experimental data was also studied for comparison. The performance of six ML algorithms was compared in all strategies, and the CatBoost model was identified as the optimal one. The strategies employing the relative deviations were superior to that using the absolute deviation, and the relative ratio revealed the systematic prediction error of the COSMO-RS model. The CatBoost model based on the relative ratio achieved the highest prediction accuracy on the test set (R2 = 0.9999, MAE = 0.0325), reducing the average absolute relative deviation (AARD) in modeling from 52.45 % to 1.54 %. Features importance analysis indicated the average energy correction, solvation-free energy, and polarity moment were the key influencing the systematic deviation.

    • The displacement of a nonionic polymeric dispersant, Pluronic F127, adsorbed at the surface of single-walled carbon nanotubes, by low molecular-weight ionic dispersants (surfactants) is studied in aqueous dispersion. The method applied is diffusion NMR spectroscopy that can accurately measure the fraction of F127 molecules adsorbed at the tube surface because of the slow exchange (over the experimental time scale) of F127 between bulk and surface. In a series of surfactants with varying chain length and headgroups, we find that anionic surfactants replace in general more nonionic F127 than do cationic surfactants. The data collected show a strong correlation with the critical dispersibility concentration of the different surfactants, a parameter that signifies the concentration at which one obtains significant dispersed nanotube concentration by ultrasonication. We posit that this finding indicates the existence of a threshold surface coverage for dispersants that constitutes a necessary condition for de-bundling by ultrasonication. The results are discussed in relation to previous findings in the literature.

    • 1H NMR peak intensities, 1H NMR diffusion measurements and TGA experiments were used to clarify the fate of the dispersing molecules, block copolymer Pluronic F127, during preparation of single-walled carbon nanotube (CNT) dispersions and their state in the created dispersions. In the dispersions, a fraction of the F127 molecules is adsorbed to the CNT surface. The mode of adsorption is the attachment and significant immobilization of the hydrophobic polypropylene oxide (PPO) block to the CNT surface and, as a result, the 1H NMR signal from the attached PPO blocks is lost. On the other hand, the hydrophilic polyethylene oxide (PEO) blocks remain highly mobile and thereby detectable by NMR. The F127 is revealed to exhibit significant block polydispersity. Molecules with large PPO blocks become enriched upon the surface of that fraction of the initial CNT powder that does not become dispersed. The molecular motions involved in creating the observed NMR features are clarified.

    • When using amphiphilic polymers to exfoliate and disperse carbon nanotubes in water, the balance between the hydrophobic and hydrophilic moieties is critical and nontrivial. Here, we investigate the mode of surface attachment of a triblock copolymer, Pluronics F127, composed of a central hydrophobic polypropylene oxide block flanked by hydrophilic polyethylene oxide blocks, onto single-walled carbon nanotubes (SWNTs). Crucially, we analyze the composition in dispersant of both the as-obtained dispersion (the supernatant) and the precipitate-containing undispersed materials. For this, we combine the carefully obtained data from H-1 NMR peak intensities and self-diffusion and thermogravimetric analysis. The molecular motions behind the observed NMR features are clarified. We find that the hydrophobic blocks attach to the dispersed SWNT surface and remain significantly immobilized leading to H-1 NMR signal loss. On the other hand, the hydrophilic blocks remain highly mobile and thus readily detectable by NMR. The dispersant is shown to possess significant block polydispersity that has a large effect on dispersibility. Polymers with large hydrophobic blocks adsorb on the surface of the carbonaceous particles that precipitate, indicating that although a larger hydrophobic block is good for enhancing adsorption, it may be less effective in dispersing the tubes. A model is also proposed that consistently explains our observations in SWNT dispersions and some contradicting findings obtained previously in carbon nanohorn dispersions. Overall, our findings help elucidating the molecular picture of the dispersion process for SWNTs and are of interest when looking for more effective (i.e., well-balanced) polymeric dispersants.

    • Lithium-ion-free tris(2,2′-bipyridine) Co(ii/iii)-mediated electrolytes have previously been proposed for long-term stable dye-sensitized solar cells (DSSCs). Such redox systems also offer an impressive DSSC performance improvement under light soaking exposure, manifested by an increase in photocurrent and fill factor without the expense of decreasing photovoltage. Kinetic studies show that charge transfer and ion diffusion at the electrode/electrolyte interface are improved due to the light exposure. Control experiments reveal that the light effect is unambiguously associated with electrolyte components, [Co(bpy)3]3+ and the Lewis-base additive tert-butylpyridine (TBP). Electrochemical and spectroscopic investigation of the [Co(bpy)3]3+/TBP mixtures points out that the presence of TBP, which retards the electrolyte diffusion, however causes an irreversible redox reaction of [Co(bpy)3]3+ upon light exposure that improves the overall conductivity. This discovery not only provides a new strategy to mitigate the typical Jsc-Voc trade-off in Co(ii/iii)-mediated DSSCs but also highlights the importance of investigating the photochemistry of a photoelectrochemical system.

    • The first inter-calibration study of the stable silicon isotope composition of dissolved silicic acid in seawater, delta Si-30(OH)(4), is presented as a contribution to the international GEOTRACES program. Eleven laboratories from seven countries analyzed two seawater samples from the North Pacific subtropical gyre (Station ALOHA) collected at 300 m and at 1000 m water depth. Sampling depths were chosen to obtain samples with a relatively low (9 mmol L-1, 300 m) and a relatively high (113 mmol L-1, 1000 m) silicic acid concentration as sample preparation differs for low- and highconcentration samples. Data for the 1000 m water sample were not normally distributed so the median is used to represent the central tendency for the two samples. Median delta Si-30(OH)(4) values of +1.66& for the low-concentration sample and +1.25& for the high-concentration sample were obtained. Agreement among laboratories is overall considered very good; however, small but statistically significant differences among the mean isotope values obtained by different laboratories were detected, likely reflecting inter-laboratory differences in chemical preparation including pre-concentration and purification methods together with different volumes of seawater analyzed, and the use of different mass spectrometers including the Neptune MC-ICP-MS (Thermo Fisher (TM), Germany), the Nu Plasma MC-ICP-MS (Nu Instruments (TM), Wrexham, UK), and the Finnigan (TM) (now Thermo Fisher (TM), Germany) MAT 252 IRMS. Future studies analyzing delta Si-30(OH)(4) in seawater should also analyze and report values for these same two reference waters in order to facilitate comparison of data generated among and within laboratories over time.

    • BackgroundHistone deacetylase (HDAC) plays an important role in the deacetylation of histone, which can alter gene expression patterns and affect cell behavior associated with malignant transformation. The aims of this study were to investigate the relationships between HDAC1, HDAC2, clinicopathologic characteristics, patient prognosis and apoptosis, to clarify the mechanism of upregulation of the Axis inhibitor Axin (an important regulator of the Wnt pathway) by X-radiation and to elucidate the effect of siRNA on radiation therapy of non-small cell lung cancer (NSCLC).MethodsHDAC1 and HDAC2 expression levels were measured by immunohistochemistry and reverse transcription PCR. Apoptosis was determined by terminal deoxynucleotidyl transferase-mediated dUTP-nick end labeling and fluorescence activated cell sorting. BE1 cells expressing Axin were exposed to 2 Gy of X-radiation.ResultsExpression of HDAC1 and that of HDAC2 were correlated, and significantly higher in NSCLC tissues than in normal lung tissues (P < 0.05). HDAC1 and HDAC2 expression was correlated with pTNM stage and negatively correlated with differentiation of NSCLC and apoptotic index (P < 0.05). The prognosis of patients with low expression of HDAC1 and HDAC2 was better than that of those with high expression. X-radiation and siRNA inhibited HDAC1 and HDAC2 expression in NSCLC cells and Axin levels were significantly higher in BE1 cells.ConclusionsX-radiation and siRNA inhibit expression of HDAC1 and HDAC2, weaken the inhibitory effect of HDAC on Axin, upregulate Axin expression and induce apoptosis of lung cancer cells. Inhibition of HDAC1 and HDAC2 is a means of enhancing the radiosensitivity of NSCLC.

    • The human islet amyloid polypeptide (hIAPP) co-operates with insulin to maintain glycemic balance. It also constitutes the amyloid plaques that aggregate in the pancreas of type-II diabetic patients. We have performed extensive in silico investigations to analyse the structural landscape of monomeric hIAPP, which is presumed to be intrinsically disordered. For this, we construct from first principles a highly predictive energy function that describes a monomeric hIAPP observed in a nuclear magnetic resonance experiment, as a local energy minimum. We subject our theoretical model of hIAPP to repeated heating and cooling simulations, back and forth between a high temperature regime where the conformation resembles a random walker and a low temperature limit where no thermal motions prevail. We find that the final low temperature conformations display a high level of degeneracy, in a manner which is fully in line with the presumed intrinsically disordered character of hIAPP. In particular, we identify an isolated family of a-helical conformations that might cause the transition to amyloidosis, by nucleation.

    • The indoor environment influences occupants' health. From March 1, 2018, to February 28, 2019, we continuously monitored indoor temperature (T), relative humidity (RH), and CO2 concentration in bedrooms via an online system in 165 residences that covered all five climate zones of China. Meanwhile, we asked one specific occupant in each home to complete questionnaires about perceived air quality and sick building syndrome (SBS) symptoms at the end of each month. Higher CO2 concentration was significantly associated with a higher percentage of perceived stuffy odor and skin SBS symptoms. Higher relative humidity was associated with higher percentage of perceived moldy odor and humid air, while lower RH was associated with a higher percentage of perceived dry air. Occupants who lived in residences with high RH were less likely to have mucosal and skin SBS symptoms (adjusted odds ratio (AOR): 0.73-0.78). However, the benefit of high humidity for perceived dry air and skin dryness symptoms is weaker if there is a high CO2 concentration level.

    • Over the past half century, the Yangtze Plain of China has experienced rapid economic development. Lake reclamation (i.e., conversion of natural lake/wetland areas to agricultural/urban land or aquaculture, thereby reducing the area of natural waters) in particular has greatly contributed to meeting the increasing demands for food and urban development. However, until now, a comprehensive quantification and understanding of historical anthropogenic lacustrine exploitation in this region has been lacking, prohibiting assessment of the impacts of these activities. We used Landsat observations from 1973 to 2018 to track reclamation-induced changes in 112 large lakes (97.8% of the total lake area) in the Yangtze Plain. We show that 41.6% (6056.9 km2) of the total lake area has been reclaimed since the 1970s. The expansion of agricultural and built-up lands dominated the reclamation activities in the 1970s, while the increase of aquaculture zones has prevailed since the mid-1980s. Reclamation activities were closely connected to government policies and major socio-economic events and had strong impacts on lake hydrology, flood risk mitigation capacity, and water quality as revealed by satellite and in situ observations. This new quantitative understanding of anthropogenic reclamation and its associated impacts on Yangtze Plain freshwater lakes can underpin the development of strategies to reduce the impacts of lake reclamation on environmental quality. The study has also demonstrated the unique strength of using long-term series satellite images in tracking historical environmental changes in a substantial region of the world, and the methods used here are potentially extendable to other inland and coastal areas to understand similar human-environment interaction problems.

    • Algal blooms constitute an emerging threat to global inland water quality, yet their spatial and temporal distribution at the global scale remains largely unknown. Here we establish a global bloom database, using 2.91 million Landsat satellite images from 1982 to 2019 to characterize algal blooms in 248,243 freshwater lakes, representing 57.1% of the global lake area. We show that 21,878 lakes (8.8%) spread across six continents have experienced algal blooms. The median bloom occurrence of affected lakes was 4.6%, but this frequency is increasing; we found increased bloom risks in the 2010s, globally (except for Oceania). The most pronounced increases were found in Asia and Africa, mostly in developing countries that remain reliant on agricultural fertilizer. As algal blooms continue to expand in scale and magnitude, this baseline census will be vital towards future risk assessments and mitigation efforts.

    • Concerted activation of different voltage-gated Ca2+ channel isoforms may determine the kinetics of insulin release from pancreatic islets. Here we have elucidated the role of R-type CaV2.3 channels in that process. A 20% reduction in glucose-evoked insulin secretion was observed in CaV2.3-knockout (CaV2.3–/–) islets, close to the 17% inhibition by the R-type blocker SNX482 but much less than the 77% inhibition produced by the L-type Ca2+ channel antagonist isradipine. Dynamic insulin-release measurements revealed that genetic or pharmacological CaV2.3 ablation strongly suppressed second-phase secretion, whereas first-phase secretion was unaffected, a result also observed in vivo. Suppression of the second phase coincided with an 18% reduction in oscillatory Ca2+ signaling and a 25% reduction in granule recruitment after completion of the initial exocytotic burst in single CaV2.3–/– ß cells. CaV2.3 ablation also impaired glucose-mediated suppression of glucagon secretion in isolated islets (27% versus 58% in WT), an effect associated with coexpression of insulin and glucagon in a fraction of the islet cells in the CaV2.3–/– mouse. We propose a specific role for CaV2.3 Ca2+ channels in second-phase insulin release, that of mediating the Ca2+ entry needed for replenishment of the releasable pool of granules as well as islet cell differentiation.

    • Background: 2,3-butanediol is an important platform compound which has a wide range of applications, involving in medicine, chemical industry, food and other fields. Especially the optically pure (2R,3R)-2,3-butanediol can be employed as an antifreeze agent and as the precursor for producing chiral compounds. However, some (2R,3R)-2,3-butanediol overproducing strains are pathogenic such as Enterobacter cloacae and Klebsiella oxytoca. Results: In this study, a (3R)-acetoin overproducing C. glutamicum strain, CGS9, was engineered to produce optically pure (2R,3R)-2,3-butanediol efficiently. Firstly, the gene bdhA from B. subtilis 168 was integrated into strain CGS9 and its expression level was further enhanced by using a strong promoter Psod and ribosome binding site (RBS) with high translation initiation rate, and the (2R,3R)-2,3-butanediol titer of the resulting strain was increased by 33.9%. Then the transhydrogenase gene udhA from E. coli was expressed to provide more NADH for 2,3-butanediol synthesis, which reduced the accumulation of the main byproduct acetoin by 57.2%. Next, a mutant atpG was integrated into strain CGK3, which increased the glucose consumption rate by 10.5% and the 2,3-butanediol productivity by 10.9% in shake-flask fermentation. Through fermentation engineering, the most promising strain CGK4 produced a titer of 144.9 g/L (2R,3R)-2,3-butanediol with a yield of 0.429 g/g glucose and a productivity of 1.10 g/L/h in fed-batch fermentation. The optical purity of the resulting (2R,3R)-2,3-butanediol surpassed 98%. Conclusions: To the best of our knowledge, this is the highest titer of optically pure (2R,3R)-2,3-butanediol achieved by GRAS strains, and the result has demonstrated that C. glutamicum is a competitive candidate for (2R,3R)-2,3-butanediol production.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • Priming of insulin secretory granules for release requires intragranular acidification and depends on vesicular Cl(-)-fluxes, but the identity of the chloride transporter/ion channel involved is unknown. We tested the hypothesis that the chloride transport protein ClC-3 fulfills these actions in pancreatic beta cells. In ClC-3(-/-) mice, insulin secretion evoked by membrane depolarization (high extracellular K(+), sulfonylureas), or glucose was >60% reduced compared to WT animals. This effect was mirrored by a approximately 80% reduction in depolarization-evoked beta cell exocytosis (monitored as increases in cell capacitance) in single ClC-3(-/-) beta cells, as well as a 44% reduction in proton transport across the granule membrane. ClC-3 expression in the insulin granule was demonstrated by immunoblotting, immunostaining, and negative immuno-EM in a high-purification fraction of large dense-core vesicles (LDCVs) obtained by phogrin-EGFP labeling. The data establish the importance of granular Cl(-) fluxes in granule priming and provide direct evidence for the involvement of ClC-3 in the process.

We therefore initiated a large-scale international study to reanalyze and delineate the transcriptome landscape of 1,223 BCP ALL cases using RNA sequencing. Fourteen BCP ALL gene expression subgroups (G1 to G14) were identified. Apart from extending eight previously described subgroups (G1 to G8 associated with MEF2D fusions, TCF3–PBX1 fusions, ETV6–RUNX1–positive/ETV6–RUNX1–like, DUX4 fusions, ZNF384 fusions, BCR–ABL1/Ph–like, high hyperdiploidy, and KMT2A fusions), we defined six additional gene expression subgroups: G9 was associated with both PAX5 and CRLF2 fusions; G10 and G11 with mutations in PAX5 (p.P80R) and IKZF1 (p.N159Y), respectively; G12 with IGH–CEBPE fusion and mutations in ZEB2 (p.H1038R); and G13 and G14 with TCF3/4–HLF and NUTM1 fusions, respectively. In pediatric BCP ALL, subgroups G2 to G5 and G7 (51 to 65/67 chromosomes) were associated with low-risk, G7 (with ≤50 chromosomes) and G9 were intermediate-risk, whereas G1, G6, and G8 were defined as high-risk subgroups. In adult BCP ALL, G1, G2, G6, and G8 were associated with high risk, while G4, G5, and G7 had relatively favorable outcomes. This large-scale transcriptome sequence analysis of BCP ALL revealed distinct molecular subgroups that reflect discrete pathways of BCP ALL, informing disease classification and prognostic stratification. The combined results strongly advocate that RNA sequencing be introduced into the clinical diagnostic workup of BCP ALL. four decades, most of the recurring chromosomal abnormalities, including aneuploidy, chromosomal rearrangements/gene fusions (e.g., ETV6–RUNX1, BCR–ABL1, and TCF3–PBX1), and rearrangements of KMT2A (previously MLL), were identified by.

    • Background: Epidemiological studies observed gender differences in COVID-19 outcomes, however, whether sex hormone plays a causal in COVID-19 risk remains unclear. This study aimed to examine associations of sex hormone, sex hormones-binding globulin (SHBG), insulin-like growth factor-1 (IGF-1), and COVID-19 risk. Methods: Two-sample Mendelian randomization (TSMR) study was performed to explore the causal associations between testosterone, estrogen, SHBG, IGF-1, and the risk of COVID-19 (susceptibility, hospitalization, and severity) using genome-wide association study (GWAS) summary level data from the COVID-19 Host Genetics Initiative (N=1,348,701). Random-effects inverse variance weighted (IVW) MR approach was used as the primary MR method and the weighted median, MR-Egger, and MR Pleiotropy RESidual Sum and Outlier (MR-PRESSO) test were conducted as sensitivity analyses. Results: Higher genetically predicted IGF-1 levels have nominally significant association with reduced risk of COVID-19 susceptibility and hospitalization. For one standard deviation increase in genetically predicted IGF-1 levels, the odds ratio was 0.77 (95% confidence interval [CI], 0.61-0.97, p=0.027) for COVID-19 susceptibility, 0.62 (95% CI: 0.25-0.51, p=0.018) for COVID-19 hospitalization, and 0.85 (95% CI: 0.52-1.38, p=0.513) for COVID-19 severity. There was no evidence that testosterone, estrogen, and SHBG are associated with the risk of COVID-19 susceptibility, hospitalization, and severity in either overall or sex-stratified TSMR analysis. Conclusions: Our study indicated that genetically predicted high IGF-1 levels were associated with decrease the risk of COVID-19 susceptibility and hospitalization, but these associations did not survive the Bonferroni correction of multiple testing. Further studies are needed to validate the findings and explore whether IGF-1 could be a potential intervention target to reduce COVID-19 risk.

    • The hydrogen-bonding interaction is studied in imidazolium-based mesogenic ionic liquids in their isotropic, smectic, and solid phases and in a nanoconfined state by proton solid-state nuclear magnetic resonance (NMR). In the smectic phase, the more basic anions form stronger hydrogen bonds. A small decrease of H-bonding in the mesophase with respect to that in the isotropic phase is associated with the presence of a layered assembly with high orientational order and limited conformational freedom. Hydrogen bond strength is not sensitive to the cation structural modification as long as the aprotic nature of the material is preserved. The strong cation-anion hydrogen bonding observed in the smectic phases provides direct support for the presence of ionic sublayers which form in ionic liquid crystals regardless of the location and alignment of the charged group in the cation, particularly irrespective of whether the charged group occupies a terminal or central position in the cation structure. A comparison of the results obtained in isotropic, liquid-crystalline, and solid states shows that in the bulk materials the dynamic state of ions ranging from high reorientational and translational freedom to partial orientation and positional order to full immobilization, respectively, has no strong impact on the cation-anion hydrogen bond strength. On the other hand, nanoconfinement of ionic liquid crystals led to hydrogen bond disruption due to competing interactions of anions with a solid interface.

    • The correlation between the water contribution to hydrogen bonding within ionic sublayer, mesophase order parameter, and ion translational self-diffusion in the layered ionic liquid crystalline phase is investigated. Changes in hydrogen bonding, conformational and translational dynamics, and orientational order upon hydration were followed by solid-state NMR combined with density functional theory (DFT) analysis. We observed that the smectic mesophase of monohydrated imidazolium-based ionic liquids, which was stabilized in a wider temperature range compared to that of anhydrous materials, counterintuitively exhibited a lower orientational order of organic cations. Thus the role of anisotropic alignment of cations and contribution of dispersion forces in the mesophase stability decreased upon hydration. The local dynamics of cations is controlled by the alignment of the bulky methyl-imidazolium ring, experiencing strong electrostatic and H-bond interactions in the ionic sublayer. Anisotropy of translational diffusion increased in the hydrated samples, thus supporting the layer-stabilizing effect of water. The effect of decreasing molecular order is outweighed by the contribution of water hydrogen bonding to the overall interaction energy within the ionic sublayer.

    • Despite advances in cancer therapeutics, pancreatic cancer remains difficult to treat and often develops resistance to chemotherapies. We have evaluated a bioavailable genistein analogue, AXP107-11 which has completed phase Ib clinical trial, as an approach to sensitize tumor cells to chemotherapy. Using organotypic cultures of 14 patient-derived xenografts (PDX) of pancreatic ductal adenocarcinoma, we found that addition of AXP107-11 indeed sensitized 57% of cases to gemcitabine treatment. Results were validated using PDX models in vivo. Further, RNA-Seq from responsive and unresponsive tumors proposed a 41-gene treatment-predictive signature. Functional and molecular assays were performed in cell lines and demonstrated that the effect was synergistic. Transcriptome analysis indicated activation of G-protein-coupled estrogen receptor (GPER1) as the main underlying mechanism of action, which was corroborated using GPER1-selective agonists and antagonists. GPER1 expression in pancreatic tumors was indicative of survival, and our study proposes that activation of GPER1 may constitute a new avenue for pancreatic cancer therapeutics.

The majority of the research in this topic has focused on bulk rheological properties, while here we employ NMR diffusion experiments to explore the mobility (diffusivity) of epoxy molecules when loaded with CNTs. Rheology and light microscopy indicate percolation, or jamming events of CNT aggregates, caging a substantial amount of epoxy molecules while small angle X-ray scattering indicates rearrangements of epoxy molecules in the vicinity of the nanotubes. NMR diffusion experiments distinguish between the diffusion of the caged molecules and that of the free ones, and relate the fraction of the former to the system viscosity. These findings demonstrate the utility of NMR diffusion experiments as an additional method applied to the rheological behavior of polymer mixtures.

Here we present the results of propofol adsorption at the air/water interface studied by means of vibrational sum frequency spectroscopy (VSFS), neutron reflectometry (NR), and surface tensiometry. VSFS was utilized to elucidate the orientation change of propofol at the surface as a function of concentration, and the water of hydration was studied by probing the OH vibrational region of the spectrum in two different polarisation combinations. Data show that propofol adsorbs at the air/water interface in an ordered fashion showing strong interactions with the water of hydration, as well as weak interactions with water in the proximity of the hydrocarbon parts of the molecule. In the concentration range studied (0 – 0.89 mM) there is almost no change in the orientation adopted at the interface. NR shows that propofol forms a dense monolayer with a thickness of 4.8 Å, and this result is consistent with a limiting area per molecule equivalent to a close-packed monolayer as demonstrated by surface tensiometry. The possibility that islands or multilayers of propofol form at the air/water interface is therefore excluded. Additionally, the ability of propofol to form associations/multimeric structures in water was studied using nuclear magnetic resonance (NMR). The 1H NMR chemical shifts recorded indicate that propofol does not form dimers or multimers in bulk water (D2O).

    • 5-methyl-2'-deoxycytidine (5mdC) and 5-hydroxymethyl-2'-deoxycytidine (5hmdC), products of DNA methylation and hydroxymethylation processes, have been detected previously in human urine, but their associations with environmental chemicals or healthy outcomes are unclear. The present investigation explored the associations between urinary 5mdC and 5hmdC with phthalate exposure and semen quality. We assessed semen parameters including sperm concentration, motility, and morphology, before measuring urinary 5mdC, 5hmdC and 13 phthalate metabolites among 562 subfertile men from Nanjing, China. Urinary 5mdC and 5hmdC were positively associated with the levels of low molecular weight phthalate metabolites (Low-MWP), high molecular weight phthalate metabolites (High-MWP), and the sum of all phthalate metabolites (ΣPAEs), respectively. Urinary 5mdC was associated with below-reference sperm concentration (odds ratios for increasing quartiles=1.0, 2.2, 3.0, 2.0; p for trend =0.02), sperm motility (1.0, 1.1, 1.9, 1.3; p for trend =0.05), and sperm morphology (1.0, 1.4, 2.3, 1.5; p for trend =0.05). Sperm concentration was associated with the highest quartile of urinary 5hmdC [odds ratio=1.9 (95% CI: 1.1, 3.6)]. Our findings showed significant associations between urinary 5mdC and 5hmdC with phthalate metabolites and semen parameters, which suggested urinary 5mdC and 5hmdC may be promising biomarkers in future epidemiological studies.

    • Nonionic surfactants containing poly(ethylene oxide) are chemically simple and biocompatible and form core-shell micelles at a wide range of conditions. For those reasons, they and their aggregates have been widely investigated. Recently, irregularities that were observed in the low-temperature behavior of surfactants of the kind [CH3(CH2)(n)O-(CH2CH2O)(m)H], (abbreviated CnEm) were assigned to a freezing-melting phase transition in the micellar core. In this work we expand the focus from the case of single component systems to binary surfactant systems at temperatures between 1 and 15 degrees C. By applying small-angle X-ray scattering (SAXS), differential scanning calorimetry (DSC), nuclear magnetic resonance (NMR), and density measurements in pure C18E20 and C18E100 solutions and their mixtures, we show that core freezing/melting is also present in mixtures. Additionally, comparing SAXS data obtained from the mixture with those from the single components, it was possible to demonstrate that the phase transition leads to a reversible segregation of the surfactants from mixed micelles to distinct kinds of micelles of the two components.

    • Nicotinamide adenine dinucleotide phosphate (NADPH) enhances Ca(2+)-induced exocytosis in pancreatic beta-cells, an effect suggested to involve the cytosolic redox protein glutaredoxin-1 (GRX-1). We here detail the role of GRX-1 in NADPH-stimulated beta-cell exocytosis and glucose-stimulated insulin secretion. Silencing of GRX-1 by RNA interference reduced glucose-stimulated insulin secretion in both clonal INS-1 832/13 cells and primary rat islets. GRX-1 silencing did not affect cell viability or the intracellular redox environment, suggesting that GRX-1 regulates the exocytotic machinery by a local action. By contrast, knockdown of the related protein thioredoxin-1 (TRX-1) was ineffective. Confocal immunocytochemistry revealed that GRX-1 locates to the cell periphery, whereas TRX-1 expression is uniform. These data suggest that the distinct subcellular localizations of TRX-1 and GRX-1 result in differences in substrate specificities and actions on insulin secretion. Single-cell exocytosis was likewise suppressed by GRX-1 knockdown in both rat beta-cells and clonal 832/13 cells, whereas after overexpression exocytosis increased by approximately 40%. Intracellular addition of NADPH (0.1 mm) stimulated Ca(2+)-evoked exocytosis in both cell types. Interestingly, the stimulatory action of NADPH on the exocytotic machinery coincided with an approximately 30% inhibition in whole-cell Ca(2+) currents. After GRX-1 silencing, NADPH failed to amplify insulin release but still inhibited Ca(2+) currents in 832/13 cells. In conclusion, NADPH stimulates the exocytotic machinery in pancreatic beta-cells. This effect is mediated by the NADPH acceptor protein GRX-1 by a local redox reaction that accelerates beta-cell exocytosis and, in turn, insulin secretion.

    • India has pledged to reduce the carbon intensity of GDP by 33-35% in the year 2030 compared to 2005 level in its Intended Nationally-Determined Contributions (INDC). The goal of limiting the global average temperature rise well below 2 °C was reaffirmed in the Paris Agreement adopted under UNFCCC. This chapter assesses three scenarios for India spanning till 2030. First, the reference scenario assumes continuation of the ongoing policies along the conventional path. Second, INDC scenario assumes the successful implementation of INDC decarbonization target. Two-degree (2 °C) scenario assumes an emission constraint aligned with the global of 2 °C stabilization target. The modelling assessment is carried out using a top-down computable general equilibrium AIM/CGE (Asia-Pacific Integrated Model/Computable General Equilibrium) model. The results show that even after full implementation of the INDCs, the emissions would still be 25 billion tonnes of CO2 equivalent higher than 2 °C stabilization target over the period 2016-2030. Enhanced actions like penetration of renewable technologies, end-use demand management and improvement in energy efficiency could help to close this emission gap.

    • Bile salt-stimulated lipase (BSSL) is a lipolytic digestive enzyme with broad substrate specificity secreted from exocrine pancreas into the intestinal lumen in all species and from the lactating mammary gland into the milk of some species, notably humans but not cows. BSSL in breast milk facilitates digestion and absorption of milk fat and promotes growth of small for gestational age preterm infants. Thus, purified recombinant human BSSL (rhBSSL) can be used for treatment of patients with fat malabsorption and expressing rhBSSL in the milk of transgenic cloned cows would therefore be a mean to meet a medical need. In the present study, a vector pBAC-hLF-hBSSL was constructed, which efficiently expressed active rhBSSL in milk of transgenic cloned cows to a concentration of 9.8 mg/ml. The rhBSSL purified from cow milk had the same enzymatic activity, N-terminal amino acid sequence, amino acid composition and isoelectric point and similar physicochemical characteristics as human native BSSL. Our study supports the use of transgenic cattle for the cost-competitive, large-scale production of therapeutic rhBSSL.
