  • Horizontal Cell Biology: Monitoring Global Changes of Protein Interaction States with the Proteome-Wide Cellular Thermal Shift Assay (CETSA)
  • 2019
  • Ingår i: Annual review of biochemistry. - : Annual Reviews. - 1545-4509 .- 0066-4154. ; 88, s. 383-408
  • Tidskriftsartikel (refereegranskat)abstract
    • The cellular thermal shift assay (CETSA) is a biophysical technique allowing direct studies of ligand binding to proteins in cells and tissues. The proteome-wide implementation of CETSA with mass spectrometry detection (MS-CETSA) has now been successfully applied to discover targets for orphan clinical drugs and hits from phenotypic screens, to identify off-targets, and to explain poly-pharmacology and drug toxicity. Highly sensitive multidimensional MS-CETSA implementations can now also access binding of physiological ligands to proteins, such as metabolites, nucleic acids, and other proteins. MS-CETSA can thereby provide comprehensive information on modulations of protein interaction states in cellular processes, including downstream effects of drugs and transitions between different physiological cell states. Such horizontal information on ligandmodulation in cells is largely orthogonal to vertical information on the levels of different proteins and therefore opens novel opportunities to understand operational aspects of cellular proteomes.

  • A Genome-wide Drosophila Screen for Heat Nociception Identifies alpha 2 delta 3 as an Evolutionarily Conserved Pain Gene
  • 2010
  • Ingår i: Cell. - : Elsevier Science B.V., Amsterdam.. - 0092-8674 .- 1097-4172. ; 143:4, s. 628-638
  • Tidskriftsartikel (refereegranskat)abstract
    • Worldwide, acute, and chronic pain affects 20% of the adult population and represents an enormous financial and emotional burden. Using genome-wide neuronal-specific RNAi knockdown in Drosophila, we report a global screen for an innate behavior and identify hundreds of genes implicated in heat nociception, including the alpha 2 delta family calcium channel subunit straightjacket (stj). Mice mutant for the stj ortholog CACNA2D3 (alpha 2 delta 3) also exhibit impaired behavioral heat pain sensitivity. In addition, in humans, alpha 2 delta 3 SNP variants associate with reduced sensitivity to acute noxious heat and chronic back pain. Functional imaging in alpha 2 delta 3 mutant mice revealed impaired transmission of thermal pain-evoked signals from the thalamus to higher-order pain centers. Intriguingly, in alpha 2 delta 3 mutant mice, thermal pain and tactile stimulation triggered strong cross-activation, or synesthesia, of brain regions involved in vision, olfaction, and hearing.

  • HV1 mtDNA Reveals the High Genetic Diversity and the Ancient Origin of Vietnamese Dogs
  • 2023
  • Ingår i: Animals. - : MDPI AG. - 2076-2615. ; 13:6, s. 1036-
  • Tidskriftsartikel (refereegranskat)abstract
    • Simple Summary This study evaluated the genetic diversity and investigated the origin of Vietnamese domestic dogs on the basis of genetic data. A total of 56 haplotypes (group of genes inherited together from a single parent), including 14 indigenous ones (two of them have not been previously reported) were observed, with some of them implying that Australian dingo and Polynesian dogs originated from Southeast Asian ones, reinforcing the theory of domestication of dogs to the south of the Yangtze River. In this study, samples from 429 dog individuals across three main regions of Vietnam (Southern Vietnam (SVN), Central Vietnam (CVN), and Northern Vietnam (NVN)) were collected to analyze the 582 bp region mtDNA HVI, so as to study the genetic diversity and to screen the rare haplotype E in the Vietnamese village dog population. Nine new haplotypes A, two new haplotypes B, and three haplotypes C were unique to Vietnam dogs, in which the new haplotypes An3, An7, Cn1, and Cn3 concerned mutations at new polymorphism sites (15,517, 15,505, 15,479, and 15,933, respectively) which have not been previously reported. The detection of haplotypes A9 and A29, and the appearance of haplotype A200 in the two individual dogs sampled support that the Southeast Asian dog is the ancestor of today's Australian dingo and Polynesian dog. The two rare haplotypes E (E1 and E4) were reconfirmed in Vietnamese dogs and discussed. This study also contributes to strengthening the theory of domestication of dogs to the south of the Yangtze River and the Southeast Asian origin of the dingo.

  • Enhancement of selective monoaromatic hydrocarbon and syngas products from fast pyrolysis of cassava stalks over Co, Mo promoted Ni catalysts
  • 2024
  • Ingår i: Journal of the Energy Institute. - 1743-9671. ; 115
  • Tidskriftsartikel (refereegranskat)abstract
    • Catalytic fast pyrolysis of cassava stalks has been intriguing as a promising, feasible chemical refinery method in terms of waste-to-energy conversion from cassava cultivation. This study aims to determine the synergetic influence of Mo and Co on Ni catalysts (Ni, Ni–Mo, and Ni–Co) prepared via ultrasonic-assisted incipient wetness impregnation method for catalytic pyrolysis vapor upgrading. The results indicated that Co and Mo enhance the reducibility of NiO and promote weak-to-medium acidity, thereby enhancing Ni's performance. Mono-Ni favors cracking deoxygenation, whereas oxophilicity promoters particularly enhance the oxygen absorbability efficiency to selective deoxygenated production. A fine Ni dispersion on CoNi/SiO2 catalyst was observed to positively affect the production of monoaromatic hydrocarbons (46.10 %), in contrast to those without promoters (21.63 %). This is attributed to an optimal Ni-metallic distribution with a small crystal size of 7.01 nm and a high surface area of 342.6 m2 g−1, highlighting the effectiveness of the promoter-assisted Ni catalyst. Additionally, a high yield of H2 (66.9 %) in non-condensable gas was recorded at 550 °C. The study suggests that a simplified approach to developing Ni-based catalysts could enhance the eco-friendliness of commercial catalysts, thereby facilitating the scale-up of biomass pyrolysis applications.
