    • The transferability and clinical value of genetic risk scores (GRSs) across populations remain limited due to an imbalance in genetic studies across ancestrally diverse populations. Here we conducted a multi-ancestry genome-wide association study of 156,319 prostate cancer cases and 788,443 controls of European, African, Asian and Hispanic men, reflecting a 57% increase in the number of non-European cases over previous prostate cancer genome-wide association studies. We identified 187 novel risk variants for prostate cancer, increasing the total number of risk variants to 451. An externally replicated multi-ancestry GRS was associated with risk that ranged from 1.8 (per standard deviation) in African ancestry men to 2.2 in European ancestry men. The GRS was associated with a greater risk of aggressive versus non-aggressive disease in men of African ancestry (P = 0.03). Our study presents novel prostate cancer susceptibility loci and a GRS with effective risk stratification across ancestry groups.

    • Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction. A meta-analysis of genome-wide association studies across different populations highlights new risk loci and provides a genetic risk score that can stratify prostate cancer risk across ancestries.

  • Echinoids as hard substrates : Varied examples from the Oligocene of Antigua, Lesser Antilles

    • A collection of unremarkably preserved fossil irregular echinoids from the Upper Oligocene (Chattian) Antigua Formation of Antigua, Lesser Antilles, nonetheless provides evidence of a range of palaeoecological interactions. A dead test of the heart urchin Eupatagus sp. formed a hard substrate for the attachment of gregarious Thecidellina? sp., a thecidoid brachiopod. Although obligate encrusters, these brachiopods more commonly occur as disarticulated valves free of the substrate in the Antillean fossil record. Elongate pits in test fragments were formed, variously, before and after the death of the host echinoids. These depressions on the external surface were formed either by invertebrates excavating domiciles or by claws or teeth; the echinoid later reclaimed the pits and grew new tubercles in the base. Post-mortem pits lack such new tuberculation. A test of Eupatagus sp. bears the boring Oichnus isp., formed either by a predator (gastropod?) or after the death of the echinoid (domicile), and a serpulid worm tube which grew on the test subsequent to the echinoid's death. The echinoid fauna of the Antigua Formation has been easy to collect and specimens are to be found in many museums; they now await re-examination to reveal palaeosynecological data analogous to that determined from the fragments discussed herein.

  • Observational and genetic associations between cardiorespiratory fitness and cancer : a UK Biobank and international consortia study

    • Background: The association of fitness with cancer risk is not clear.Methods: We used Cox proportional hazards models to estimate hazard ratios (HRs) and 95% confidence intervals (CIs) for risk of lung, colorectal, endometrial, breast, and prostate cancer in a subset of UK Biobank participants who completed a submaximal fitness test in 2009-12 (N = 72,572). We also investigated relationships using two-sample Mendelian randomisation (MR), odds ratios (ORs) were estimated using the inverse-variance weighted method.Results: After a median of 11 years of follow-up, 4290 cancers of interest were diagnosed. A 3.5 ml O2⋅min−1⋅kg−1 total-body mass increase in fitness (equivalent to 1 metabolic equivalent of task (MET), approximately 0.5 standard deviation (SD)) was associated with lower risks of endometrial (HR = 0.81, 95% CI: 0.73–0.89), colorectal (0.94, 0.90–0.99), and breast cancer (0.96, 0.92–0.99). In MR analyses, a 0.5 SD increase in genetically predicted O2⋅min−1⋅kg−1 fat-free mass was associated with a lower risk of breast cancer (OR = 0.92, 95% CI: 0.86–0.98). After adjusting for adiposity, both the observational and genetic associations were attenuated.Discussion: Higher fitness levels may reduce risks of endometrial, colorectal, and breast cancer, though relationships with adiposity are complex and may mediate these relationships. Increasing fitness, including via changes in body composition, may be an effective strategy for cancer prevention.

    • Insulin‐like growth factors (IGFs) and insulin‐like growth factor binding proteins (IGFBPs) have been implicated in the aetiology of several cancers. To better understand whether anthropometric, behavioural and sociodemographic factors may play a role in cancer risk via IGF signalling, we examined the cross‐sectional associations of these exposures with circulating concentrations of IGFs (IGF‐I and IGF‐II) and IGFBPs (IGFBP‐1, IGFBP‐2 and IGFBP‐3). The Endogenous Hormones, Nutritional Biomarkers and Prostate Cancer Collaborative Group dataset includes individual participant data from 16,024 male controls (i.e. without prostate cancer) aged 22–89 years from 22 prospective studies. Geometric means of protein concentrations were estimated using analysis of variance, adjusted for relevant covariates. Older age was associated with higher concentrations of IGFBP‐1 and IGFBP‐2 and lower concentrations of IGF‐I, IGF‐II and IGFBP‐3. Higher body mass index was associated with lower concentrations of IGFBP‐1 and IGFBP‐2. Taller height was associated with higher concentrations of IGF‐I and IGFBP‐3 and lower concentrations of IGFBP‐1. Smokers had higher concentrations of IGFBP‐1 and IGFBP‐2 and lower concentrations of IGFBP‐3 than nonsmokers. Higher alcohol consumption was associated with higher concentrations of IGF‐II and lower concentrations of IGF‐I and IGFBP‐2. African Americans had lower concentrations of IGF‐II, IGFBP‐1, IGFBP‐2 and IGFBP‐3 and Hispanics had lower IGF‐I, IGF‐II and IGFBP‐3 than non‐Hispanic whites. These findings indicate that a range of anthropometric, behavioural and sociodemographic factors are associated with circulating concentrations of IGFs and IGFBPs in men, which will lead to a greater understanding of the mechanisms through which these factors influence cancer risk.

  • A new silurian ophiuroid from the west of Ireland

    • Silurian echinoderms from Ireland are poorly known; hitherto, only three nominal species have been described, all crinoids and all from the Telychian (Upper Llandovery, Lower Silurian) Kilbride Formation in County Galway. A new species from this formation, Crepidosoma doylei, is the first recorded Irish Silurian ophiuroid (brittle star). The new species is described from five articulated specimens; all are mouldic and none retains a counterpart. The disc and immediately adjacent arm lateral ossicles of the new species differ in shape from those of the type species in that these ossicles are comparatively flat rather than angular or ridged, and the series is marked by a well-defined channelway that appears to have extended from within the disc for the full length of the free arm. Ambital framework ossicles are smooth, and not S- or Z-shaped and ridged as in the type species. The new specimens of C. doylei were recovered from distal storm deposits that are thought to be deep-water equivalents of Benthic Assemblage Zone 5 (Clorinda Community).

    • Crinoids and brachiopods are described from the Silurian Uzyan Formation of the Zilair Zone in the southern Urals. The occurrence of the graptolites Coronograptus praedeubeli suggests a late Homerian (Wenlock) age for the strata. A new disparid crinoid, Cicerocrinus gracilis Donovan sp. nov., is the oldest known member of this genus. It has a long, flexible and homeomorphic column, and a tall bryozoan palaeontology terminology (IBr2) (second primibrachial) axillary. All species of Cicerocrinus described previously have been limited to the Ludlow of the British Isles, Sweden and Estonia, and the Pridoli of Estonia. The poorly preserved brachiopod fauna is represented by small atrypid (Atrypa? sp.) and dalmanellid brachiopods (Levenea? sp.). The reported assemblage generally inhabited deep-water environments.

    • The monobathrid camerate crinoid Macrostylocrinus bornholmensis Laursen has an unfortunate history. It was published in a journal not commonly consulted by echinoderm workers and, worse, in time of war; written in a language not in common use for crinoid studies; and described by a stratigrapher, not an expert on pelmatozoans. These and other factors combined to ensure that M. bornholmensis has not been reassessed since it was first described almost 80 years ago, despite belonging to a genus well-known from the Lower Palaeozoic. Macrostylocrinus bornholmensis is Llandovery (Telychian) and not Wenlock as has been reported elsewhere. Diagnostic features include a column that does not bear radices close beneath the cup; a heteromorphic mesistele with five orders of regularly inserted internodals; three basal plates; smooth thecal plates with central folds following rays; and ten pinnulate free arms that are biserial distally. Macrostylocrinus bornholmensis is distinctly different in morphology from well-known, congeneric species described from the Lower Palaeozoic of northern Europe.

    • BACKGROUND: Polygenic risk scores comprising established susceptibility variants have shown to be informative classifiers for several complex diseases including prostate cancer. For prostate cancer it is unknown if inclusion of genetic markers that have so far not been associated with prostate cancer risk at a genome-wide significant level will improve disease prediction.METHODS: We built polygenic risk scores in a large training set comprising over 25,000 individuals. Initially 65 established prostate cancer susceptibility variants were selected. After LD pruning additional variants were prioritized based on their association with prostate cancer. Six-fold cross validation was performed to assess genetic risk scores and optimize the number of additional variants to be included. The final model was evaluated in an independent study population including 1,370 cases and 1,239 controls.RESULTS: The polygenic risk score with 65 established susceptibility variants provided an area under the curve (AUC) of 0.67. Adding an additional 68 novel variants significantly increased the AUC to 0.68 (P = 0.0012) and the net reclassification index with 0.21 (P = 8.5E-08). All novel variants were located in genomic regions established as associated with prostate cancer risk.CONCLUSIONS: Inclusion of additional genetic variants from established prostate cancer susceptibility regions improves disease prediction.
