    • This final response to the two climate change denial papers by Shani and Arad further highlights the inaccuracies, misinformation and errors in their commentaries. The obfuscation of scientific research and the consensus on anthropogenic climate change may have significant long-term negative consequences for better understanding the implications of climate change and climate policy for tourism and create confusion and delay in developing and implementing tourism sector responses.

    • Shani and Arad (2014) claimed that tourism scholars tend to endorse the most pessimistic assessments regarding climate change, and that anthropogenic climate change was a "fashionable" and "highly controversial scientific topic". This brief rejoinder provides the balance that is missing from such climate change denial and skepticism studies on climate change and tourism. Recent research provides substantial evidence that reports on anthropogenic climate change are accurate, and that human-induced greenhouse gas emissions, including from the tourism industry, play a significant role in climate change. Some positive net effects may be experienced by some destinations in the short-term, but in the long-term all elements of the tourism system will be impacted. The expansion of tourism emissions at a rate greater than efficiency gains means that it is increasingly urgent that the tourism sector acknowledge, accept and respond to climate change. Debate on tourism-related adaptation and mitigation measures is to be encouraged and welcomed. Climate change denial is not.

  • A hot and fast ultra-stripped supernova that likely formed a compact neutron star binary

    • Compact neutron star binary systems are produced from binary massive stars through stellar evolution involving up to two supernova explosions. The final stages in the formation of these systems have not been directly observed. We report the discovery of iPTF 14gqr (SN 2014ft), a type Ic supernova with a fast-evolving light curve indicating an extremely low ejecta mass (approximate to 0.2 solar masses) and low kinetic energy (approximate to 2 x 10(50) ergs). Early photometry and spectroscopy reveal evidence of shock cooling of an extended helium-rich envelope, likely ejected in an intense pre-explosion mass-loss episode of the progenitor. Taken together, we interpret iPTF 14gqr as evidence for ultra-stripped supernovae that form neutron stars in compact binary systems.

  • COMMON ENVELOPE EJECTION FOR A LUMINOUS RED NOVA IN M101

    • We present the results of optical, near-infrared, and mid-infrared observations of M101 OT2015-1 (PSN J14021678+ 5426205), a luminous red transient in the Pinwheel galaxy (M101), spanning a total of 16 years. The light curve showed two distinct peaks with absolute magnitudes M-r <= -12.4 and M-r similar or equal to -12, on 2014 November 11 and 2015 February 17, respectively. The spectral energy distributions during the second maximum show a cool outburst temperature of approximate to 3700 K and low expansion velocities (approximate to -300 km s(-1)) for the H I, Ca II, Ba II, and K I lines. From archival data spanning 15-8 years before the outburst, we find a single source consistent with the optically discovered transient, which we attribute to being the progenitor; it has properties consistent with being an F-type yellow supergiant with L similar to 8.7 x 10(4) L-circle dot, T-eff approximate to 7000. K, and an estimated mass of M1= 18 +/- 1 M-circle dot. This star has likely just finished the H-burning phase in the core, started expanding, and is now crossing the Hertzsprung gap. Based on the combination of observed properties, we argue that the progenitor is a binary system, with the more evolved system overfilling the Roche lobe. Comparison with binary evolution models suggests that the outburst was an extremely rare phenomenon, likely associated with the ejection of the common envelope of a massive star. The initial mass of the primary fills the gap between the merger candidates V838 Mon (5-10 M-circle dot) and NGC. 4490-OT. (30M(circle dot)).

    • In this paper, we describe recent architectural and technological advances of the end to end optical network architecture proposed by the DISCUS project (the DIStributed Core for unlimited bandwidth supply for all Users and Services). The two main targets of DISCUS are the principle of equivalence in the access and the reduction of optical-to-electronic conversions in the metro-core network. Technological advances and techno-economic evaluation of Long-Reach Passive Optical Networks (LR-PON), as well as the optimal metro-core node architecture and the required network control plane framework are reported. Network infrastructure sharing challenges are also discussed.

    • This paper reports on the architectural, protocol, physical layer, and integrated testbed demonstrations carried out by the DISCUS FP7 consortium in the area of access-metro network convergence. Our architecture modeling results show the vast potential for cost and power savings that node consolidation can bring. The architecture, however, also recognizes the limits of long-reach transmission for low-latency 5G services and proposes ways to address such shortcomings in future projects. The testbed results, which have been conducted end-toend, across access-metro and core, and have targeted all the layers of the network from the application down to the physical layer, show the practical feasibility of the concepts proposed in the project.

    • Objectives To develop evidence-based European League Against Rheumatism (EULAR) recommendations for vaccination in patients with autoimmune inflammatory rheumatic diseases (AIIRD). Methods A EULAR task force was composed of experts representing 11 European countries, consisting of eight rheumatologists, four clinical immunologists, one rheumatologist/clinical immunologist, one infectious disease physician, one nephrologist, one paediatrician/rheumatologist and one clinical epidemiologist. Key questions were formulated and the eligible spectrum of AIIRD, immunosuppressive drugs and vaccines were defined in order to perform a systematic literature review. A search was made of Medline from 1966 to October 2009 as well as abstracts from the EULAR meetings of 2008 and 2009 and the American College of Rheumatology (ACR) meetings of 2007 and 2008. Evidence was graded in categories I-IV, the strength of recommendations was graded in categories A-D and Delphi voting was applied to determine the level of agreement between the experts of the task force. Results Eight key questions and 13 recommendations addressing vaccination in patients with AIIRD were formulated. The strength of each recommendation was determined. Delphi voting revealed a very high level of agreement with the recommendations among the experts of the task force. Finally, a research agenda was proposed. Conclusion Recommendations for vaccination in patients with AIIRD based on the currently available evidence and expert opinion were formulated. More research is needed, particularly regarding the incidence of vaccine-preventable infectious diseases and the safety of vaccination in patients with AIIRD.

    • Objectives: To present the systematic literature review (SLR), which formed the basis for the European League Against Rheumatism (EULAR) evidence-based recommendations for vaccination in adult patients with autoimmune inflammatory rheumatic diseases (AIIRD). Methods: AIIRD, vaccines and immunomodulating drugs, as well as eight key questions were defined by the multidisciplinary expert committee commissioned by EULAR for developing the recommendations. A SLR was performed using MedLine through October 2009 and including data from meta-analyses, systematic reviews, randomized trials, and observational studies, excluding case series with participants. Articles in English and regarding patients years of age, were eligible. Results: Several vaccine-preventable infections (VPI) occur more often in AIIRD-patients and most vaccines are efficacious in AIIRD-patients, even when treated with immunomodulating agents, except rituximab. There does not appear to be an increase in vaccination-related harms in vaccinated patients with AIIRD in comparison with unvaccinated patients with AIIRD. However, these studies are underpowered and therefore not conclusive. Conclusion: Based on the current evidence from the literature, recommendations for vaccination in patients with AIIRD were made. However, more research is needed in particular regarding incidence of VPI, harms of vaccination and the influence of (new and established) immunomodulating agents on vaccination efficacy.     • Context. Temporal variability of narrow absorption lines in high-resolution spectra of Type Ia supernovae (SNe Ia) is studied to search for circumstellar matter. Time series which resolve the profiles of absorption lines such as Na I D or Ca II H&K are expected to reveal variations due to photoionisation and subsequent recombination of the gases. The presence, composition, and geometry of circumstellar matter may hint at the elusive progenitor system of SNe Ia and could also affect the observed reddening law. Aims. To date, there are few known cases of time-varying Na I D absorption in SNe Ia, all of which occurred during relatively late phases of the supernova (SN) evolution. Photoionisation, however, is predicted to occur during the early phases of SNe Ia, when the supernovae peak in the ultraviolet. We attempt, therefore, to observe early-time absorption-line variations by obtaining high-resolution spectra of SNe before maximum light. Methods. We have obtained photometry and high-resolution spectroscopy of SNe Ia 2013gh and iPTF 13dge, to search for absorption-line variations. Furthermore, we study interstellar absorption features in relation to the observed photometric colours of the SNe. Results. Both SNe display deep Na I D and Ca II H&K absorption features. Furthermore, small but significant variations are detected in a feature of the Na I D profile of SN 2013gh. The variations are consistent with either geometric effects of rapidly moving or patchy gas clouds or photoionisation of Na I gas at R approximate to 10(19) cm from the explosion. Conclusions. Our analysis indicates that it is necessary to focus on early phases to detect photoionisation effects of gases in the circumstellar medium of SNe Ia. Different absorbers such as Na I and Ca II can be used to probe for matter at different distances from the SNe. The nondetection of variations during early phases makes it possible to put limits on the abundance of the species at those distances.

  • Expression, purification and crystallization of CTB-MPR, a candidate mucosal vaccine component against HIV-1

    • CTB-MPR is a fusion protein between the B subunit of cholera toxin (CTB) andthe membrane-proximal region of gp41 (MPR), the transmembrane envelopeprotein ofHuman immunodeficiency virus 1(HIV-1), and has previously beenshown to induce the production of anti-HIV-1 antibodies with antiviralfunctions. To further improve the design of this candidate vaccine, X-raycrystallography experiments were performed to obtain structural informationabout this fusion protein. Several variants of CTB-MPR were designed,constructed and recombinantly expressed inEscherichia coli. The first variantcontained a flexible GPGP linker between CTB and MPR, and yielded crystalsthat diffracted to a resolution of 2.3 A ̊, but only the CTB region was detectedin the electron-density map. A second variant, in which the CTB was directlyattached to MPR, was shown to destabilize pentamer formation. A thirdconstruct containing a polyalanine linker between CTB and MPR proved tostabilize the pentameric form of the protein during purification. The purificationprocedure was shown to produce a homogeneously pure and monodispersesample for crystallization. Initial crystallization experiments led to pseudo-crystals which were ordered in only two dimensions and were disordered inthe third dimension. Nanocrystals obtained using the same precipitant showedpromising X-ray diffraction to 5 A ̊resolution in femtosecond nanocrystallo-graphy experiments at the Linac Coherent Light Source at the SLAC NationalAccelerator Laboratory. The results demonstrate the utility of femtosecondX-ray crystallography to enable structural analysis based on nano/microcrystalsof a protein for which no macroscopic crystals ordered in three dimensions havebeen observed before.

    • We present the best 265 sampled R-band light curves of spectroscopically identified Type Ia supernovae (SNe) from the Palomar Transient Factory (PTF; 2009-2012) survey and the intermediate Palomar Transient Factory (iPTF; 2013-2017). A model-independent light-curve template is built from our data-set with the purpose to investigate average properties and diversity in our sample. We searched for multiple populations in the light-curve properties using machine learning tools. We also utilized the long history of our light curves, up to 4000 days, to exclude any significant pre- or post- supernova flares. From the shapes of light curves we found the average rise time in the R band to be 16.8−0.6+0.5'>16.8 +0.5 −0.6  16.8−0.6+0.5 days. Although PTF/iPTF were single-band surveys, by modelling the residuals of the SNe in the Hubble–Lemaître diagram, we estimate the average colour excess of our sample to be ⟨E(B − V)⟩ ≈ 0.05(2) mag and thus the mean corrected peak brightness to be MR = −19.02 ± 0.02 +5log⁡(H0[kms−1Mpc−1]/70)'>+5log(H 0 [kms −1 Mpc −1 ]/70) +5log⁡(H0[kms−1Mpc−1]/70) mag with only weak dependennce on light–curve shape. The intrinsic scatter is found to be σR = 0.186 ± 0.033 mag for the redshift range 0.05 < z < 0.1, without colour corrections of individual SNe. Our analysis shows that Malmquist bias becomes very significant at z = 0.13. A similar limitation is expected for the ongoing Zwicky Transient Facility (ZTF) survey using the same telescope, but new camera expressly designed for ZTF.

    • Fiber to the premises has promised to increase the capacity in telecommunications access networks for well over 30 years. While it is widely recognized that optical-fiber-based access networks will be a necessity in the shortto medium-term future, its large upfront cost and regulatory issues are pushing many operators to further postpone its deployment, while installing intermediate unambitious solutions such as fiber to the cabinet. Such high investment cost of both network access and core capacity upgrade often derives from poor planning strategies that do not consider the necessity to adequately modify the network architecture to fully exploit the cost benefit that a fiber-centric solution can bring. DISCUS is a European Framework 7 Integrated Project that, building on optical-centric solutions such as long-reach passive optical access and flat optical core, aims to deliver a cost-effective architecture for ubiquitous broadband services. DISCUS analyzes, designs, and demonstrates end-to-end architectures and technologies capable of saving cost and energy by reducing the number of electronic terminations in the network and sharing the deployment costs among a larger number of users compared to current fiber access systems. This article describes the network architecture and the supporting technologies behind DISCUS, giving an overview of the concepts and methodologies that will be used to deliver our end-to-end network solution.

    • Targeting the androgen receptor (AR) pathway prolongs survival in patients with prostate cancer, but resistance rapidly develops. Understanding this resistance is confounded by a lack of noninvasive means to assess AR activity in vivo. We report intracellular accumulation of a secreted antigen-targeted antibody (SATA) that can be used to characterize disease, guide therapy, and monitor response. AR-regulated human kallikrein-related peptidase 2 (free hK2) is a prostate tissue-specific antigen produced in prostate cancer and androgen-stimulated breast cancer cells. Fluorescent and radio conjugates of 11B6, an antibody targeting free hK2, are internalized and noninvasively report AR pathway activity in metastatic and genetically engineered models of cancer development and treatment. Uptake is mediated by a mechanism involving the neonatal Fc receptor. Humanized 11B6, which has undergone toxicological tests in nonhuman primates, has the potential to improve patient management in these cancers. Furthermore, cellspecific SATA uptake may have a broader use for molecularly guided diagnosis and therapy in other cancers.

    • Streptococcus agalactiae, also known as group B Streptococcus (GBS), is the major cause of neonatal sepsis in humans. A critical step to infection is adhesion of bacteria to epithelial surfaces. GBS adhesins have been identified to bind extracellular matrix components and cellular receptors. However, several putative adhesins have no host binding partner characterised. We report here that surface-expressed β protein of GBS binds to human CEACAM1 and CEACAM5 receptors. A crystal structure of the complex showed that an IgSF domain in β represents a novel Ig-fold subtype called IgI3, in which unique features allow binding to CEACAM1. Bioinformatic assessment revealed that this newly identified IgI3 fold is not exclusively present in GBS but is predicted to be present in adhesins from other clinically important human pathogens. In agreement with this prediction, we found that CEACAM1 binds to an IgI3 domain found in an adhesin from a different streptococcal species. Overall, our results indicate that the IgI3 fold could provide a broadly applied mechanism for bacteria to target CEACAMs.

  • iPTF 16asu : A Luminous, Rapidly Evolving, and High-velocity Supernova

    • Wide-field surveys are discovering a growing number of rare transients whose physical origin is not yet well understood. Here we present optical and UV data and analysis of intermediate Palomar Transient Factory (iPTF) 16asu, a luminous, rapidly evolving, high-velocity, stripped-envelope supernova ( SN). With a rest-frame rise time of just four. days and a peak absolute magnitude of M-g = -20.4 mag, the light curve of iPTF 16asu is faster and more luminous than that of previous rapid transients. The spectra of iPTF 16asu show a featureless blue continuum near peak that develops into an SN Ic-BL spectrum on the decline. We show that while the late-time light curve could plausibly be powered by Ni-56 decay, the early emission requires a different energy source. Nondetections in the X-ray and radio strongly constrain the energy coupled to relativistic ejecta to be at most comparable to the class of low-luminosity gamma-ray bursts (GRBs). We suggest that the early emission may have been powered by either a rapidly spinning-down magnetar or by shock breakout in an extended envelope of a very energetic explosion. In either scenario a central engine is required, making iPTF 16asu an intriguing transition object between superluminous SNe, SNe Ic-BL, and low-luminosity GRBs.
