  • Tillämpning av multivariata metoder för övervakning och optimering av skogsindustriella biologiska reningsanläggningar

    • I takt med att utsläppskraven på skogsindustrin ökat under 90-talet byggde många luftade dammar om till långtidsluftad aktivslamanläggning (LAS). Med en LAS anläggning ökar möjligheterna till styrning väsentligt i jämförelse med en traditionell luftad damm. I detta projekt som initierats av IVL Svenska Miljöinstitutet AB och Å-forsk var målet att skapa ett multivariat övervakningssystem för att ta fram viktig processinformation för övervakning och styrning av en LAS-anläggning. Inledningsvis installerades en del kompletterande givare för mätning av suspenderat material och TOC vid olika positioner i processen. Därefter implementerades en dynamisk databas för att produktionsanpassa datainsamlingen. Databasmodellen kompenserar för tidsförskjutningarna i processen vilket gör att tidsseriemodellering inte är nödvändig. Ett viktigt resultat av databasmodellen är att data loggas snabbt när produktionen är snabb (högt flöde) och med lägre frekvens vid låg produktion (lågt flöde). Detta medför att man minimerar 'data overload'. Parallellt med framtagning av databasmodellen beräknades multivariata modeller på laboratoriedata för att få fram indikationer på viktiga processparametrar och på modelleringskonceptet. Resultaten för modelleringen på laboratoriedata föll mycket väl ut, bl.a. hittades interaktioner mellan variabler som tidigare var okända, t.ex. korrelerade fastsittande ciliater, filament i anoxisk miljö och frisimmande bakterier i olika utsträckning mot kvalitetsvariablerna utgående COD, utgående suspenderade ämnen samt utgående total fosfor. Detta har medfört en större processkunskap och bättre styrningsmöjligheter för processoperatörerna. I slutfasen av projektet utvärderades realtidsdata med en multivariat modell baserad på data från den dynamiska databasen. Modeller för processövervakning (PCA) och för prediktion av utgående kvalitétsvärden (PLS) genomfördes med goda resultat. Systemet från insamlat mätvärde till databasmodell och multivariat prediktion fungerar väl. Förseningar i projektet gör dock att dataomfattningen i kalibreringsmodellerna är för liten för att modellera alla verkliga driftscenarior. Implementeringen av systemet var mycket lyckad och modeller predikterar och övervakar idag i realtid. I den närmaste framtiden kommer detta system fortfarande att vara i drift för att ta fram bättre och mer omfattande modeller. Projektet har framförallt utvecklat ett verktyg som visar på styrkan av att tillämpa multivariata realtidssystem för processövervakning. Eftersom verktyget är generellt och kan tillämpas på andra processer är det givetvis intressant för t.ex. skogsindustrin att tillämpa dessa metoder på andra processer så som massa- och pappersframställning

  • Energi och miljöintegrering genom utnyttjande av lågvärdigt värme från kraftvärmeverk och fjärrvärmeanläggningar för effektivisering av avloppsvattenrening

    • Kväve i utgående vatten från avloppsreningsverk ger upphov till övergödning av många havsområden och sjöar med syrebrist och döda bottnar som följd. EU:s direktiv för avloppsvatten ställer högre krav på rening av kväve vid Svenska avloppsvattenrening. Att höja temperaturen på avloppsvattnet under de kalla perioderna kan vara en kostnadseffektiv lösning på kvävereningen. Värmeavtappning från fjärrvärmenätet till reningsverken leder till lägre temperaturer på returvattnet och detta medför att tillvaratagandet av energi kan optimeras. En lägre returtemperatur ger bättre förutsättningar för rökgaskondensering och därmed en förbättrad rening av de utgående rökgaserna. De praktiska möjligheterna att generera el i kraftvärmeanläggningar begränsas av fjärrvärmereturens temperatur. Det innebär att fjärrvärmeproducenterna får avsättning för lågvärdig värme och kan optimera sin elproduktion. För reningsverken innebär det miljömässiga och ekonomiska vinster genom att de får en effektivare och stabilare reningsprocess och kan uppfylla de hårdare kraven på kvävereningen. I en pilotstudie under verkliga förutsättningar har man inom projektet utrett den verkliga vinsten och studerat de problematiska stegen i den tekniska lösningen.

  • Lifetime Prevalence of Verbal, Physical, and Sexual Abuses in Young Elite Athletics Athletes

    • To examine prevalence of verbal, physical, and sexual abuses in young elite athletes, a cross sectional questionnaire-based survey was conducted during the World Athletics under 20 World Championships. This questionnaire aimed at distinguishing between abuses perpetrated in the context of Athletics from those which were unrelated to Athletics. Four hundred and eighty athletes (52.3%, male) from North America, South America, Europe, Africa, Asia, and Oceania took part in the electronic anonymous survey. Outside Athletics setting, no gender difference was found for the prevalence of verbal, physical, and sexual abuses. However, 45 males (18% of the male population) and 34 females (15% of the female population) athletes reported sexual abuse. Asian athletes reported a slightly higher rate of sexual abuse; three quarters of them being non-touching abuses. Inside Athletics setting, no gender difference was found for the prevalence of verbal, physical, and non-touching sexual abuses. However, 58 males (23%) and 47 females (21%) reported verbal abuses. Thirty-one males (12%) and 20 females (9%) reported physical abuses, whereas 30 males (12%) and 17 females (7%) reported sexual abuses. Physical abuses were slightly more frequent in Asia and in Africa and less frequent in South America. Sexual abuses inside Athletics also differed over regions, and were unexpectedly twice more frequent than expected in Asia and slightly less frequent than expected in Europe. Friends and partners were identified as the more frequent (> 50%) abusers outside or inside the Athletics settings, whereas outside Athletics and inside Athletics, coaches were identified as sexual abuse perpetrators in 8 and 25% of cases, respectively. The prevalence of verbal, physical, or sexual abuses is high but consistent with what has been reported in United Kingdom, Norway, Canada, and Sweden at national level in recreational or elite athletes. Sexual abuse, including touching or penetrative abuses, occurred significantly more often in male athletes when compared to female athletes. This finding invites healthcare and social workers, and policymakers to also consider the risk of sexual abuse of young male athletes in Athletics. These results also call for longitudinal studies on young elite athletes.

  • Economic Project Risk Assessment in Remediation Projects Prior to Construction: Methodology Development and Case Study Application

    • Probabilistic economic analysis, including uncertainty of probabilities and consequences of project risks, is not widely used in remediation projects. This article presents a project risk assessment (PRA) method to identify, quantify, and analyze risks in remediation projects. The suggested method is probabilistic and includes uncertainty analysis of input variables based on expert judgment. It was originally developed as a part of a sustainability assessment tool, but is viable as a stand-alone tool for remediation projects. The method is applied to a case study: a former paint factory that is being redeveloped into a residential area. The PRA method is used for analyzing and comparing the project risks associated with four remediation options, all including excavation but with different degrees of onsite treatment. The result of the case study application shows which alternative has the lowest mean risk cost, the highest probability to have the lowest risk cost, and how the risk costs are distributed, but also, importantly, helps the user to prioritize between risk-reduction measures.

  • A Structural Investigation into the OCV Hysteresis of the Ni(OH)2 Electrode

    • Powder X-ray diffraction (XRPD), X-ray absorption spectroscopy (EXAFS and XANES) and Raman spectroscopy were used to study chemical changes in the polycrystalline nickel hydroxide positive electrode material of a NiMH battery at four states of charge: 0%, 50% and 100% charged, and 50% discharged. The two 50% samples were at the same state of charge but in different hysteresis states, manifested by differences in the open circuit potential. The nickel hydroxide electrodes consist of particles in the µm size range, and all measurements were performed ex situ. The material studied was taken from commercial batteries and as such contained both metallic nickel particles, cobalt, and zinc dopants in the active Ni(OH)2 material as well as a cobalt oxide surface layer. Combining the results from all the characterization methods was necessary to better understand the chemistry behind the physio-chemical hysteresis behavior in this complex system. Our results show that there are structural differences between the two 50% samples. Comparison of the XRPD results and the EXAFS results on the nickel edge indicate a presence of the kinetically favored TP2-NiOOH phase in the transition between β-Ni(OH)2 and β-NiOOH and that the amount differs between the two hysteresis states. The measurements on the zinc edge using EXAFS and XANES suggest short range differences in the active material bulk that stems from disorder. Raman spectroscopy measurements show a difference in degree of lithium intercalation in the LiCoO2 surface layer between the hysteresis states. As electrochemistry takes place on the surface of the particles, it is likely that differences in the surface structure are responsible for the open circuit voltage hysteresis. However, due to the coherence of the structure differences detected, it is probable that they are all connected and have a part in the observed behavior.

  • Training in spikes and number of training hours correlate to injury incidence in youth athletics (track and field) : A prospective 52-week study

    • Objectives: The aim was to describe the annual incidence and types of musculoskeletal injuries, and to examine factors associated with injury risk. Design: A 52-week prospective study in Swedish youth athletics aged 12-15 years. Methods: Data on exposure to training and injury were collected from parents/caregivers and youth athletes using a web-survey system. Results: A total of 101 (86%) youth athletes participated. Fifty-four (53%) of the athletes reported one new injury. Girls were at higher risk of sustaining an injury than boys (p = 0.048). Ninety-one percent of the new injuries were non-traumatic and 85% occurred in the lower extremities. Injuries to the front thigh represented 20% of the injuries. Cox proportional hazard regression analyses showed a six-fold increased risk for a first injury for athletes reporting use of spikes and training <6 h every two weeks (hazard ratio, 6.1; 95% confidence interval, 1.2-31.3) compared to athletes training <6 h using no spikes. Athletes training 6 h or more reporting use or no use of spikes had an eight-fold increase injury risk (p < 0.01). Conclusions: Almost half of the youth athletes experienced a new injury and girls had a higher risk compared to boys. Nine out of ten injuries were related to overuse. An interesting observation was the high incidence of injuries to the quadriceps muscle complex. The study identified a correlation with training hours and an interaction with track spikes and risk of injury that needs further attention. (c) 2021 The Author(s).   • Resultat från nationella provet i matematik 1c : höstterminen 2021

    • De nationella proven i matematik 1a, 1b och 1c konstrueras och utvecklas, på uppdrag av Skolverket, av PRIM-gruppen vid Stockholms universitet. I denna rapport presenteras en sammanställning av resultaten för det nationella provet i matematik 1c som gavs höstterminen 2021. Det huvudsakliga syftet med rapporten är att redovisa och diskutera resultaten från genomförandet av detta prov.Resultatredovisningen från höstterminens nationella prov i matematik 1c kommer från PRIM-gruppens urvalsinsamling och bestod av lärarenkäter och inrapporterade resultat på uppgiftsnivå från ett slumpmässigt urval elever. PRIM-gruppens urvalsinsamling för matematik 1c omfattade höstterminen 2021 elever från naturvetenskapsprogrammet och teknikprogrammet. Cirka 60 % av eleverna i insamlingen läste naturvetenskapsprogrammet och cirka 40 % läste teknikprogrammet. Skolverket genomför enbart insamling av provbetyg på nationella prov för elevers avslutande matematikkurs i gymnasieskolan.Resultaten från lärarenkäterna och elevresultaten på uppgiftsnivå är viktiga källor för utvecklingen av de nationella proven. För matematik 1c bestod insamlingen från höstterminen 2021 av resultat från ca 220 lärare och ca 1400 elever (ca 550 flickor respektive ca 850 pojkar).Det nationella provet från höstterminen 2021 omfattas av sekretess vilket innebär att ingen av dessa uppgifter kan publiceras. Nationella prov som inte längre omfattas av sekretess finns tillgängliga på PRIM-gruppens webbplats (www.su.se/primgruppen).

    • De nationella proven i matematik i årskurs 9 konstrueras och utvecklas, på uppdrag av Skolverket, av PRIM-gruppen vid Stockholms universitet. Syftet med de nationella proven i årskurs 9 är att stödja en likvärdig och rättvis betygssättning. De nationella proven kan också bidra till att stärka skolornas kvalitetsarbete genom analyser av provresultaten i relation till uppnådda kunskapskrav på skolnivå, huvudmannanivå och på nationell nivå. I denna rapport presenteras en sammanställning av resultaten för det nationella provet i matematik i årskurs 9 läsåret 2021/2022. Det huvudsakliga syftet med rapporten är att redovisa och diskutera resultaten från genomförandet av detta prov. Jämförelser görs även med resultat från tidigare års nationella prov. Då både 2020 och 2021 års nationella prov ställdes in på grund av pandemin görs jämförelser med resultat från de nationella proven som gavs 2017–2019.

    • För drygt tio år sedan startades det s.k. mellanstadieprojektet – numera ”femteklassprojektet". Det hela går ut på att teknologer besöker mellanstadieklasser, i första hand femteklasser. Besöket inleds med ett antal spännande demonstrationsförsök ("teknik-, kemi- och fysikshow"). Teknologerna berättar om sig själva, om vad man gör på den tekniska högskolan och vad LTH är för någonting. Ett viktigt moment vid besöket är att barnen själva får göra egna experiment.. Två teknologer (manlig + kvinnlig teknolog) besöker varje klass. Som stöd finns en experimentbank som också finns tillgänglig på webben. Projektet har divits av teknologerna själva och har varit mycket lyckosamt. Responsen från skolorna är överväldigande. Så långt låter detta som ett rekryteringsprojekt. Det finns emellertid ytterligare ett element som kanske är ännu viktigare: teknologerna utvecklar en omedveten kompetens som organisatörer, som kommunikatörer av naturvetenskap och teknik till icke-initierade, etc. Teknologerna har själva det fulla ansvaret för resultatet. De får ett gott självförtroende. Allt detta är förmågor och egenskaper som borde utvecklas under utbildningen men som ofta kommer i andra hand i de vanliga kurserna.

    • Till en ren och homogen sand sattes 3-metylbensoesyra, p-kresol, m-xylenol, karbazol och fluoranten, vardera till en halt kring 100 mg/kg fuktig sand. Föreningarna valdes för att de ofta förekommer i mark förorenad med kreosot och petroleumprodukter. Den förorenade sanden ympades med slam från ett kommunalt avloppsreningsverk och fördelades på 10 olika tunnor för komposteringsförsök. Fukthalt (33-59% av fältkapacitet), temperatur (25-35°C) och till-sats av extra substrat och närsalter varierades i ett faktorförsök. Prover togs ut från sanden under nedbrytningsförloppet, och utgående luft från tunnorna analyserades också. Som jämförelse behandlades samma förorenade sand i slurryform, med och utan en inhibitor för biologisk aktivitet. De mest svårlösliga föreningarna, karbazol och fluoranten, blev så ojämnt fördelade i sanden att det inte gick att säga något om deras eventuella nedbrytning. Av de övriga föreningarna avlägsnades i de flesta fall 60-100% under 16 veckors försök. Både hastigheten och sluthalten påverkades av fukthalt och temperatur, däremot hade extra tillsats av substrat och närsalter ingen inverkan. Fukthalt och temperatur påverkade förhållandet mellan avgång till luft och biologisk (kemisk) omvandling. De kemiska analyserna kompletterades med biologiska test med växter och maskar. Den hämmande effekten av den ursprungliga förorenade sanden och ett par behandlade prover var ungefär den som man kunde vänta sig från tidigare försök med rena substanser. I de fall den toxiska effekten var större än den förväntade, kan skillnaden bero på synergistiska effekter eller på toxiska metaboliter

    • Jenny Hettne arbetar innovativt och gärna i nära samarbete med musiker där hon utforskar musikens klangliga möjligheter. Hon komponerar för såväl orkester som mindre besättningar, ibland i kombination med elektronik. Hennes akustiska musik är starkt influerad av elektroakustisk musik och vice versa. Stycket As they flutter about för basklarinett och elektronik är komponerat i nära samarbete med och tillägnat klarinettisten Robert Ek. Musiken startar i basklarinettens djupaste register, i ett statiskt ljudlandskap av knappt skönjbara variationer, lätt böljande eller försiktigt skorrande. I styckets andra del expanderar rörelserna; vibrerar, fladdrar och skimrar. Solisten och den elektroniska stämman fungerar som en duo där de ibland byter funktion med varandra: speciella tekniker på instrumentet skapar en närmast elektronisk klang medan obearbetade klarinettinspelningar hörs i den elektroniska stämman.

    • Auditing is a key tool for governing performance. Informed by the ‘audit trail logic’ this article takes a micro-perspective on auditing in which the auditees play an active role. Using the case of eldercare, it explores what actors do as they take part in audits and how they make sense of the underlying logic. Findings reveal that the roles of auditor and auditee may not only shift between audits, but also blur during audit activities. Therefore, this article contributes to public performance literature and practice by empirically showing how actors involved in an audit negotiate a shared understanding of performance.

    • In the last decade, many initiatives have been taken to develop antibodies for proteome-wide studies, as well as characterization and validation of clinically relevant disease biomarkers. Phage display offers many advantages compared to conventional antibody generation by immunization and hybridoma technology, since it is an unlimited resource of affinity reagents without batch-to-batch variation and is amendable for high throughput. One of the major bottlenecks to proteome-wide binder selection is the limited supply of suitable target antigens representative of the human proteome. Here, we provide proof of principle of using easily accessible, cancer-associated protein epitope signature tags (PrESTs), routinely generated within the Human Protein Atlas project, as surrogate antigens in phage selectionsfor the retrieval of target specific binders. These binders were subsequently tested in western blot, immunohistochemistry and protein microarray application to demonstrate their functionality.

    • The rapid increase of antibiotic resistance has created an urgent need to develop novel antimicrobial agents. Here we describe the crystal structure of the promising bacterial target phospho-N-acetylmuramoyl-pentapeptide translocase (MraY) in complex with the nucleoside antibiotic tunicamycin. The structure not only reveals the mode of action of several related natural-product antibiotics but also gives an indication on the binding mode of the MraY UDP-MurNAc-pentapeptide and undecaprenyl-phosphate substrates.

    • The alarming growth of antibiotic resistance that is currently ongoing is a serious threat to human health. One of the most promising novel antibiotic targets is MraY (phospho-MurNAc-pentapeptide-transferase), an essential enzyme in bacterial cell wall synthesis. Through recent advances in biochemical research, there is now structural information available for MraY, and for its human homologue GPT (GlcNAc-1-P-transferase), that opens up exciting possibilities for structure-based drug design. The antibiotic compound tunicamycin is a natural product inhibitor of MraY that is also toxic to eukaryotes through its binding to GPT. In this work, we have used tunicamycin and modified versions of tunicamycin as tool compounds to explore the active site of MraY and to gain further insight into what determines inhibitor potency. We have investigated tunicamycin variants where the following motifs have been modified: the length and branching of the tunicamycin fatty acyl chain, the saturation of the fatty acyl chain, the 6″-hydroxyl group of the GlcNAc ring, and the ring structure of the uracil motif. The compounds are analyzed in terms of how potently they bind to MraY, inhibit the activity of the enzyme, and affect the protein thermal stability. Finally, we rationalize these results in the context of the protein structures of MraY and GPT.

    • The rapid growth of antibiotic-resistant bacterial infections is of major concern for human health. Therefore, it is of great importance to characterize novel targets for the development of antibacterial drugs. One promising protein target is MraY (UDP-N-acetylmuramyl-pentapeptide: undecaprenyl phosphate N-acetylmuramyl-pentapeptide-1-phosphate transferase or MurNAc-1-P-transferase), which is essential for bacterial cell wall synthesis. Here, we summarize recent breakthroughs in structural studies of bacterial MraYs and the closely related human GPT (UDP-N-acetylglucosamine: dolichyl phosphate N-acetylglucosamine-1-phosphate transferase or GlcNAc-1-P-transferase). We present a detailed comparison of interaction modes with the natural product inhibitors tunicamycin and muraymycin D2. Finally, we speculate on possible routes to design an antibacterial agent in the form of a potent and selective inhibitor against MraY.

    • Overproduction and purification of membrane proteins are generally challenging and time-consuming procedures due to low expression levels, misfolding, and low stability once extracted from the membrane. Reducing processing steps and shortening the timespan for purification represent attractive approaches to overcome some of these challenges. We have therefore compared a fast "teabag" purification method with conventional purification for five different membrane proteins (MraY, AQP10, CIC-1, PAR2 and KCC2). Notably, this new approach reduces the purification time significantly, and the quality of the purified membrane proteins is equal to or exceeds conventional methods as assessed by size exclusion chromatography, SDS-PAGE and downstream applications such as ITC, crystallization and cryo-EM. Furthermore, the method is scalable, applicable to a range of affinity resins and allows for parallelization. Consequently, the technique has the potential to substantially simplify purification efforts of membrane proteins in basic and applied sciences.

    • Streptococcus spp. are important causes of infective endocarditis but challenging in species identification. This study compared identification based on sequence determination of the rnpB gene with 2 systems of matrix-assisted laser desorption ionization-time of flight mass spectrometry, MALDI Biotyper (Bruker) and VITEK MS IVD (bioMerieux). Blood culture isolates of viridans streptococci from 63 patients with infective endocarditis were tested. The 3 methods showed full agreement for all 36 isolates identified in the Anginosus, Bovis, and Mutans groups or identified as Streptococcus cristatus, Streptococcus gordonii, or Streptococcus sanguinis. None of the methods could reliably identify the 23 isolates to the species level when designated as Streptococcus mitis, Streptococcus oralis, or Streptococcus tigurinus. In 7 isolates classified to the Mitis group, the rnpB sequences deviated strikingly from all reference sequences, and additional analysis of sodA and groEL genes indicated the occurrence of yet unidentified Streptococcus spp. (C) 2015 Elsevier Inc.   • Identification of Novel Downstream Molecules of Tissue Factor Activation by Comparative Proteomic Analysis

    • Tissue factor (TF) is both an initiator of blood coagulation and a signaling receptor. Using a proteomic approach, we investigated the role of TF in cell signaling when stimulated by its ligand, activated factor VII (FVIIa). From a 2-D difference gel electrophoresis (DIGE) study we found forty one spots that were differentially regulated over time in FVIIa stimulated cells or in comparison to nonstimulated cells. Mass spectrometry identifies 23 out of these as 13 different proteins. One of them, elongation factor 2 (EF-2), was investigated in greater detail by Western blot, a protein synthesis assay and cell cycle analysis. When tissue factor was stimulated by FVIIa, the phosphorylation of EF-2 increased which inactivates this protein. Analyzing the effect using site inactivated FVIIa (FVIIai), as well as the protease activated receptor 2 (PAR-2) agonist SLIGKV, indicated that the inactivation was not PAR-2 dependent. A panel of tissue factor mutants was analyzed further to try to pinpoint what part of the cytoplasmic domain that is needed for this effect. Performing a protein synthesis assay in two different cell lines we could confirm that protein synthesis decreased upon stimulation by FVIIa. Cell cycle analysis showed that FVIIa also promotes a higher degree of cell proliferation.

    • Objective. To describe the prevalence, awareness, and control of hypertension in a Swedish population during the early 2000s to address implications for care and prevention. Design. A cross-sectional population survey. Setting. Primary health care in Skaraborg, a rural part of western Sweden. Subjects. Participants (n =2816) in a population survey of a random sample of men and women between 30 and 75 years of age in the municipalities of Vara (81% participation rate) and Skövde (70%), in western Sweden during 2001–2005. Main outcome measures. Anthropometric measures, blood pressure, leisure-time physical activity, current smoking, fasting glucose, and cholesterol. Hypertension was defined as ongoing treatment for hypertension, or three consecutive blood pressure readings ≥140 systolic and/or ≥90 mmHg diastolic. Hypertension was considered controlled when the blood pressure was <140/90 mm Hg (both). Results. The prevalence of hypertension was 20% in both men and women with a steep increase by age. Among hypertensive subjects, 33% were unaware, 36% aware but uncontrolled, and 31% aware and controlled, with no statistically significant differences between men and women. Patients with diabetes had a higher awareness (87% vs. 64%, p <0.001), but the same control rate (56% vs. 44%, p =0.133), when compared with those without diabetes. Conclusion. A large proportion of subjects with hypertension are still unaware of their condition, or aware but not controlled. It is important to emphasize population-based prevention to reduce the prevalence of hypertension, to perform screening to increase awareness, and to improve implementation of expert guidelines in clinical practice to improve control.

    • Development of antibody drugs against novel targets and pathways offers great opportunities to improve current cancer treatment. We here describe a phenotypic discovery platform enabling efficient identification of therapeutic antibody-target combinations. The platform utilizes primary patient cells throughout the discovery process and includes methods for differential phage display cell panning, high-throughput cell-based specificity screening, phenotypic in vitro screening, target deconvolution, and confirmatory in vivo screening. In this study the platform was applied on cancer cells from patients with Chronic Lymphocytic Leukemia resulting in discovery of antibodies with improved cytotoxicity in vitro compared to the standard of care, the CD20-specific monoclonal antibody rituximab. Isolated antibodies were found to target six different receptors on Chronic Lymphocytic Leukemia cells; CD21, CD23, CD32, CD72, CD200, and HLA-DR of which CD32, CD200, and HLA-DR appeared as the most potent targets for antibody-based cytotoxicity treatment. Enhanced antibody efficacy was confirmed in vivo using a patient-derived xenograft model.

  • Diminishing benefits of urban living for children and adolescents' growth and development

    • Optimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was <1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.

    • BACKGROUND: CRIM1 is a plasma membrane bound protein containing six cysteine-rich repeats (CRR). Through these, CRIM1 has been shown to interact with a subgroup of the TGF-beta superfamily, the bone morphogenic proteins (BMP) isoforms 2, 4 and 7. The probable action is to modulate the signalling properties of these factors. CRIM1 has also been shown to regulate the release of VEGFA by podocytes during renal organogenesis. Knock-out studies in mice have shown that CRIM1 is critically involved in the development of the central nervous system, eye and kidney. Replacement of CRIM1 with a defective version leads to renal dysgenesis and perinatal death. We have analysed the distribution of CRIM1 in adult human renal tissue. METHODS: To this end, we have used immunofluorescence, immunohistochemistry and immunoelectron microscopy. We performed western blotting for the CRIM1 protein, using lysates from isolated glomerular podocytes and human renal tissue homogenate. By using quantitative PCR, we compared the CRIM1 mRNA levels in podocytes, human renal tissue homogenate, primary human renal proximal tubular epithelial cells and primary human pulmonary artery smooth muscle cells. RESULTS: The results show that in the human adult kidney, CRIM1 is mainly expressed in the glomerular podocytes and is associated with the insertional region of the filtration slit diaphragm (SD) of the podocyte pedicles. CONCLUSIONS: CRIM1 is a protein that should be added to the list of proteins associated with the podocyte filtration SD and with the probable action of modulating BMP and VEGFA signalling.

    • For decades, alternative (to carbon) sources of energy in Sweden have been linked to hydro- and nuclear power. However, this is set to change as the Swedish government’s agenda has placed extraordinary emphasis on renewables. The implementation of renewables in Sweden poses several challenges. Literature shows that two main aspects deter local communities from embracing large renewable projects: lack of acceptance (of the impacts) and lack of participation (in the making and benefits). Sweden has a long tradition of stakeholder engagement in state-funded projects in the form of participatory meetings and written feedbacks. However, other participatory techniques are less established. Since 2014, we have engaged in research projects dealing with energy landscapes, design thinking, and what we have recently named “resourceful communities”. The aim of this chapter is to report on the results of our recent projects that engage with the above-mentioned concepts/strategies to foster collaboration and understanding between end-users and other stakeholders.

  • Synthesis of functional degradable polymers by radical ring-opening polymerization

    • The increased demand for understanding cell-material interactions and subsequently create personalized implants for tissue engineering has resulted in an increased interest in developing new monomers with functional groups. The focus of the work presented in this thesis has been on the synthesis of functional monomers and the polymerization of these into functional amorphous aliphatic polyesters. The cyclic ketene acetal 2-methylene-1,3,6-trioxocane (MTC) was synthesized in a two-step reaction and subsequently polymerized at different temperatures using either 2,2’-azoisobutyronitrile (AIBN) or dicumyl peroxide as initiator. It was shown that the polymerization mechanism was not temperature-dependent and the polymerization proceeded with 100% ring-opening at all evaluated temperatures. The polyester-ether was then successfully copolymerized with another cyclic ketene acetal 2-methylene-1,3-dioxepane (MDO) and a functional, hydrophilic and  amorphous copolymer was obtained. The feed ratios were varied between 90 to 25 mol% of MTC and it was shown that the proportion of MTC in the copolymers was in general higher than the proportion of MTC in the feed, confirming that the reactivity ratio was higher for MTC. A number average molecular weight of 6500 g/mol was obtained after 2 days at 70 °C in bulk. The high polydispersity index as well as the glass transition temperatures indicated a significant amount of branching. MDO was also copolymerized with vinyl acetate (VAc). A series of copolymers with various combinations of MDO and VAc were synthesized by using AIBN as radical initiator. It was shown that the proportion of MDO in the copolymers was strictly controlled by the feed ratios and that the composition of the copolymers, the thermal properties and the molecular weights could be predetermined. The reactivity ratios for MDO and VAc were determined to be rMDO = 0.93 and rVAc = 1.71. The copolymerization resulted in a high degree of conversions and a high number average molecular weights, where both the molecular weight and the glass transition temperature increased with increased proportion of VAc in the feed. It was also confirmed that the copolymer were susceptible to both alkali and enzymatic hydrolysis and it was apparent that it is possible to synthesize various degradable materials based on VAc and MDO with predetermined polymer compositions and high number average molecular weights.
