  • Structure-activity relationships for lipoprotein lipase agonists that lower plasma triglycerides in vivo
  • 2015
  • Ingår i: European Journal of Medicinal Chemistry. - : Elsevier. - 0223-5234 .- 1768-3254. ; 103, s. 191-209
  • Tidskriftsartikel (refereegranskat)abstract
    • The risk of cardiovascular events increases in individuals with elevated plasma triglyceride (TG) levels, therefore advocating the need for efficient TG-lowering drugs. In the blood circulation, TG levels are regulated by lipoprotein lipase (LPL), an unstable enzyme that is only active as a non-covalently associated homodimer. We recently reported on a N-phenylphthalimide derivative (1) that stabilizes LPL in vitro, and moderately lowers triglycerides in vivo (Biochem. Biophys. Res. Common. 2014, 450, 1063). Herein, we establish structure activity relationships of 51 N-phenylphthalimide analogs of the screening hit 1. In vitro evaluation highlighted that modifications on the phthalimide moiety were not tolerated and that lipophilic substituents on the central phenyl ring were functionally essential. The substitution pattern on the central phenyl ring also proved important to stabilize LPL However, in vitro testing demonstrated rapid degradation of the phthalimide fragment in plasma which was addressed by replacing the phthalimide scaffold with other heterocyclic fragments. The in vitro potency was retained or improved and substance 80 proved stable in plasma and efficiently lowered plasma TGs in vivo. 2015 The Authors. Published by Elsevier Masson SAS.

  • Identification of a small molecule that stabilizes lipoprotein lipase in vitro and lowers triglycerides in vivo
  • 2014
  • Ingår i: Biochemical and Biophysical Research Communications - BBRC. - : Elsevier. - 0006-291X .- 1090-2104. ; 450:2, s. 1063-1069
  • Tidskriftsartikel (refereegranskat)abstract
    • Patients at increased cardiovascular risk commonly display high levels of plasma triglycerides (TGs) levels, elevated LDL cholesterol, small dense LDL particles and low levels of HDL-cholesterol. Many remain at high risk even after successful statin therapy, presumably because TG levels remain high. Lipoprotein lipase (LPL) maintains TG homeostasis in blood by hydrolysis of TG-rich lipoproteins. Efficient clearance of TGs is accompanied by increased levels of HDL-cholesterol and decreased levels of small dense LDL. Given the central role of LPL in lipid metabolism we sought to find small molecules that could increase LPL activity and serve as starting points for drug development efforts against cardiovascular disease. Using a small molecule screening approach we have identified small molecules that can protect LPL from inactivation by the controller protein angiopoietin-like protein 4 during incubations in vitro. One of the selected compounds, 50F10, was directly shown to preserve the active homodimer structure of LPL, as demonstrated by heparin-Sepharose chromatography. This compound tended to reduce fasting TG levels in normal rats. On injection to hypertriglyceridemic apolipoprotein A-V deficient mice the compound ameliorated the postprandial response after an olive oil gavage. This compound is a potential lead compound for the development of drugs that could reduce the residual risk associated with elevated TGs in dyslipidemia.

  • Weld Joint Tracking System in an Automatic Inspection Cell by Using Emissivity Variation
  • 2014
  • Ingår i: Proceedings of the 6th International Swedish Production Symposium 2014. - 9789198097412 ; , s. 1-7
  • Konferensbidrag (refereegranskat)abstract
    • Thermography has proven to be a suitable nondestructive testing method for automatic crack inspection of welds. However automatic weld inspection raises challenges. E.g. the position of the weld might not be exactly as the predefined weld seam, and a weld joint tracking system is needed. To reduce the number of equipment used, a solution is presented in this papers. The infrared camera in the thermography system is a carrier of information of the weld path. This is used for the weld joint tracking system. It is shown that the weld joint tracker is fast enough for an on-line automatic inspection.

  • Personality- and size-related metabolic performance in invasive round goby (Neogobius melanostomus)
  • 2020
  • Ingår i: Physiology and Behavior. - : Elsevier. - 0031-9384 .- 1873-507X. ; 215, s. 1-6
  • Tidskriftsartikel (refereegranskat)abstract
    • Differences between individuals in behavioral type (i.e. animal personality) are ecologically and evolutionarily important because they can have significant effects on fitness components such as growth and predation risk. In the present study we are used the invasive round goby (Neogobius melanostomus) from an established population in controlled experiments to examine the relationships among personality, metabolic performance, and growth rate (inferred as size-at-age). Boldness was measured as the time to return to normal behavior after a simulated predator attack, where fish with shorter freezing times were categorized as "bold" and fish with longer times were categorized as "shy." We show that bold fish have significantly higher standard metabolic rate (SMR) than their shy conspecifics, whereas there was no difference between personality types in their maximum metabolic rate (MMR) or aerobic scope (AS). Bold fish furthermore had a smaller size-at-age as compared to shy fish. Together this provides evidence of a metabolic underpinning of personality where the high-SMR bold fish require more resources to sustain basic life functions than their low-SMR shy conspecifics, indicating that bold round goby from established populations with high densities (and high competition for food) pay a price of reduced growth rate.

  • A High Precision Method for Quantitative Measurements of Reactive Oxygen Species in Frozen Biopsies
  • 2014
  • Ingår i: PLOS ONE. - San Francisco : Public Library of Science. - 1932-6203. ; 9:3, s. e90964-
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: An electron paramagnetic resonance (EPR) technique using the spin probe cyclic hydroxylamine 1-hydroxy-3-methoxycarbonyl-2,2,5,5-tetramethylpyrrolidine (CMH) was introduced as a versatile method for high precision quantification of reactive oxygen species, including the superoxide radical in frozen biological samples such as cell suspensions, blood or biopsies. Materials and Methods: Loss of measurement precision and accuracy due to variations in sample size and shape were minimized by assembling the sample in a well-defined volume. Measurement was carried out at low temperature (150 K) using a nitrogen flow Dewar. The signal intensity was measured from the EPR 1st derivative amplitude, and related to a sample, 3-carboxy-proxyl (CPN) with known spin concentration. Results: The absolute spin concentration could be quantified with a precision and accuracy better than +/- 10 mu M (k = 1). The spin concentration of samples stored at -80 degrees C could be reproduced after 6 months of storage well within the same error estimate. Conclusion: The absolute spin concentration in wet biological samples such as biopsies, water solutions and cell cultures could be quantified with higher precision and accuracy than normally achievable using common techniques such as flat cells, tissue cells and various capillary tubes. In addition; biological samples could be collected and stored for future incubation with spin probe, and also further stored up to at least six months before EPR analysis, without loss of signal intensity. This opens for the possibility to store and transport incubated biological samples with known accuracy of the spin concentration over time.

  • Scaling Up Data-Driven Pilot Projects
  • 2020
  • Ingår i: The AI Magazine. - : Association for the Advancement of Artificial Intelligence. - 0738-4602 .- 2371-9621. ; 41:3, s. 94-102
  • Tidskriftsartikel (refereegranskat)abstract
    • Conducting pilot projects are a common approach among organizations to test and evaluate new technology. A pilot project is often conducted to remove uncertainties from a large-scale project and should be limited in time and scope. Nowadays, several organizations are testing and evaluating artificial intelligence techniques and more advanced forms of analytics via pilot projects. Unfortunately, many organizations are experiencing problems in scaling-up the findings from pilot projects to the rest of the organization. Hence, results from pilot projects become siloed with limited business value. In this article, we present an overview of barriers for conducting and scaling-up data-driven pilot projects. Lack of senior management support is a frequently mentioned top barrier in the literature. In response to this, we present our recommendations on what type of activities can be performed, to increase the chances of getting a positive response from senior management regarding scaling-up the usage of artificial intelligence and advanced analytics within an organization.

  • Cytosolic lipid droplets increase in size by microtubule-dependent complex formation
  • 2005
  • Ingår i: Arterioscler Thromb Vasc Biol. - 1524-4636. ; 25:9, s. 1945-51
  • Tidskriftsartikel (refereegranskat)abstract
    • OBJECTIVE: Adipocyte differentiation-related protein (ADRP)-containing lipid droplets have an essential role in the development of insulin resistance and atherosclerosis. Such droplets form in a cell-free system with a diameter of 0.1 to 0.4 microm, while the droplets present in cells vary in size, from small to very large, suggesting that the droplets can increase in size after being assembled. We have addressed this possibility. METHODS AND RESULTS: Experiments in NIH 3T3 cells demonstrated that the lipid droplets could increase in size independently of triglyceride biosynthesis. NIH 3T3 cells were either microinjected with ADRP-GFP (green fluorescent protein) or stained with Nile Red and followed by confocal microscopy and time-lapse recordings. The results showed that lipid droplets formed complexes with each other, with a volume equal to the sum of the merging particles. The formation of complexes could be inhibited by the nocodazole-induced depolymerization of the microtubules; thus, the process is dependent on microtubules. The presence of dynein on ADRP-containing droplets supports a role for this motor protein. CONCLUSIONS: Lipid droplets can grow after they have been assembled. This increase in size is independent of triglyceride biosynthesis and involves formation of complexes, which requires intact microtubules.

  • Systems perspectives on combined production of advanced biojet fuel and biofuels in existing industrial infrastructure in Sweden
  • 2023
  • Ingår i: Energy Conversion and Management: X. - 2590-1745. ; 19
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper assesses the opportunities for resource- and energy-efficient biojet fuel production from forest-based biomass utilising existing industrial infrastructure in Sweden. Two categories of production pathways are considered: one including technologies that are certified for the production of biojet fuel, and the other uncertified technologies under development. Aspects included are production potential, regional balance between biomass feedstock and eligible host industries, potential demand and supply of biogas-based hydrogen for upgrading, economic incentives for reducing greenhouse gas (GHG) emissions, and future markets for the co-products generated. The overall conclusion is that energy integration through the use of combined heat and power plants, sawmills and pulp mills to produce biofuel intermediates leads to a lower net demand for biomass feedstock, which can be met by the long-term sustainable potential of forestry residues and sawdust. The regional balance between the long-term, sustainable availability of forestry residues and sawdust, and the demand at eligible host industries is good where over 80% of this demand can be met by regionally produced biomass feedstock. The biofuel intermediates can then be upgraded to biojet fuel in centralised refineries connected to the gas grid in the southwest of Sweden, and with current Swedish policy tools it is economically motivated to use biogas-based hydrogen. Co-product demand will not limit commercial development since the future markets are predicted to grow when replacing fossil counterparts. Thus, current policy tools that promote biojet fuel with low life-cycle GHG emissions provide important incentives for the commercial development of resource- and energy-efficient combined biojet fuel and biofuel production systems based on biomass residues and existing industrial infrastructure.

  • Revenue Maximization in Resource Allocation : Applications in Wireless Communication Networks
  • 2004
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Revenue maximization for network operators is considered as a criterion for resource allocation in wireless cellular networks. A business model encompassing service level agreements between network operators and service providers is presented. Admission control, through price model aware admission policing and service level control, is critical for the provisioning of useful services over a general purpose wireless network. A technical solution consisting of a fast resource scheduler taking into account service requirements and wireless channel properties, a service level controller that provides the scheduler with a reasonable load, and an admission policy to uphold the service level agreements and maximize revenue, is presented. Two different types of service level controllers are presented and implemented. One is based on a scalar PID controller, that adjusts the admitted data rates for all active clients. The other one is obtained with linear programming methods, that optimally assign data rates to clients, given their channel qualities and price models. Two new scheduling criteria, and algorithms based on them, are presented and evaluated in a simulated wireless environment. One is based on a quadratic criterion, and is implemented through approximative algorithms, encompassing a search based algorithm and two different linearizations of the criterion. The second one is based on statistical measures of the service rates and channel states, and is implemented as an approximation of the joint probability of achieving the delay limits while utilizing the available resources efficiently. Two scheduling algorithms, one based on each criterion, are tested in combination with each of the service level controllers, and evaluated in terms of throughput, delay, and computational complexity, using a target test system. Results show that both schedulers can, when feasible, meet explicit throughput and delay requirements, while at the same time allowing the service level controller to maximize revenue by allocating the surplus resources to less demanding services.

  • Magnetocaloric effect in Fe2 P: Magnetic and phonon degrees of freedom
  • 2019
  • Ingår i: Physical Review B. - 2469-9969 .- 2469-9950. ; 99:17
  • Tidskriftsartikel (refereegranskat)abstract
    • Devices based on magnetocaloric materials provide great hope for environmentally friendly and energy efficient cooling that does not rely on the use of harmful gasses. Fe2P based compounds are alloys that have shown great potential for magnetocaloric devices. The magnetic behavior in Fe2P is characterized by a strong magnetocaloric effect that coexists with a first-order magnetic transition (FOMT). Neutron diffraction and inelastic scattering, Mossbauer spectroscopy, and first-principles calculations have been used to determine the structural and magnetic state of Fe2P around the FOMT. The results reveal that ferromagnetic moments in the ordered phase are perturbed at the FOMT such that the moments cant away from the principle direction within a small temperature region. The acoustic-phonon modes reveal a temperature-dependent nonzero energy gap in the magnetically ordered phase that falls to zero at the FOMT. The interplay between the FOMT and the phonon energy gap indicates hybridization between magnetic modes strongly affected by spin-orbit coupling and phonon modes leading to magnon-phonon quasiparticles that drive the magnetocaloric effect.

  • Teknikenkät - enskilda avlopp 2009
  • 2011

  • Analytical Methods For Sports Drugs: Challenges and Approaches
  • 2019
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Drugs used to enhance human performance in sport competitions are prohibited by the world anti-doping association (WADA). Biological samples from athletes are continuously tested for adverse analytical findings regarding the identity and/or quantity of the banned substances. The current thesis deals with the development of new analytical methods to determine the concentrations of certain drugs used by athletes and even by regular users for therapeutic purposes. The developed methods aim to analyze the contents of these drugs in the biological matrices; plasma, serum and saliva to provide a successful approach towards either doping detection or therapeutic monitoring. β-adrenergic blockers such as propranolol and metoprolol are used in sports to relief stress and as therapeutic agents in the treatment of hypertension. Both drugs are in chiral forms and available only as racemic mixtures. The different pharmacology of each enantiomer necessitates the monitoring of each enantiomer by stereoselective analytical technique such as chiral liquid chromatography for separation and mass spectrometry for selective detection. The Endogenous anabolic androgenic steroids (EAAS) on the other hand are only notoriously used in sports to increase muscle mass and strength. A method utilizing high-resolution mass spectrometry (HRMS) coupled to ultra-high performance liquid chromatography (UHPLC) was developed for the simultaneous determination of EAAS and their conjugated metabolites to provide a better insight into the steroidal module of the athlete biological passport (ABP). Moreover, the steroidal profile was assessed in serum using the proposed method after the administration of Growth hormone injection as an approach toward the implementation of a new endocrinological module based on steroids biomarkers to hormone doping.  Biological samples contain many components that may interfere with the analytical measurements. Therefore, sample preparation methods were developed using solid phase extraction (SPE) and miniaturized techniques such as microextraction by packed sorbents (MEPS) for the purification and pre-concentration of analytes prior to LC/MS analysis.

    • Master data management programs are large by nature since the aim is to provide the entire enterprise with a shared trusted view of the organisation’s most critical data assets. In this paper, we present what dimensions and activities a master data management program in a large organisation should consider and how to monitor such a program once it is up and running. A heatmap approach is used to visualize the inherent complexity of a master data management program. Our approach is derived from participating in four different master data management programs in four different global organisations during 2007-2020.

  • Konferensbidrag (refereegranskat)abstract
    • Many applications need to detect and respond to occurring events and combine these event occurrences into new events with a higher level of abstraction. Specifying how events can be combined is often supported by design tools specific to the current event processing engine. However, the issue of ensuring that the combinations of events provide the system with the correct combination of information is often left to the developer to analyze. We argue that analyzing correctness of event composition is a complex task that needs tool support. In this paper we present a novel development tool for specifying composition of events with time constraints. One key feature of our tool is to automatically transform composite events for real-time systems into a timed automaton representation. The timed automaton representation allow us to check for design errors, for example, whether the outcome of combining events with different operators in different consumption policies is consistent with the requirement specification

    • The chemokine CCL25 is selectively and constitutively expressed in the small intestinal epithelium and plays an important role in mediating lymphocyte recruitment to this site. In this study, we demonstrate that CCL25 expression in murine small intestinal epithelial cells is independent of signaling through the lymphotoxin 0 receptor and is not enhanced by inflammatory stimuli, pathways involved in driving the expression of most other chemokines. We define a transcriptional start site in the CCL25 gene and a region -141 to -5 proximal of exon 1 that is required for minimal promoter activity in the small intestinal epithelial cell lines, MODE-K and mICc12. These cell lines expressed far less CCL25 mRNA than freshly isolated small intestinal epithelial cells indicating that they are missing important factors driving CCL25 expression. The CCL25 promoter contained putative binding sites for,the intestinal epithelial-associated Caudal-related homeobox (Cdx) transcription factors Cdx-1 and Cdx-2, and small intestinal epithelial cells but not MODE-K and mICc12 cells expressed Cdx-1 and Cdx-2. EMSA analysis demonstrated that Cdx proteins were present in nuclear extracts from freshly isolated small intestinal epithelial cells but not in MODE-K or mICcl2 cells, and bound to putative Cdx sites within the CCL25 promoter. Finally, cotransfection of MODE-K cells with Cdx transcription factors significantly increased CCL25 promoter activity as well as endogenous CCL25 mRNA levels. Together these results demonstrate a unique pattern of regulation for CCL25 and suggest a role for Cdx proteins in regulating CCL25 transcription.

    • Complex Event Processing (CEP) is a technology with support for matching patterns in a cloud or streams of events in order to support detection of specific combinations of event occurrences. A clever specification of event patterns may, for example, detect fraud attempts in a banking system, fire an alarm in response to hazardous situations in a control system or report suspicious customer behavior.Several CEP engines have support for graphically modelling applications as well as perform tests and provide execution traces to verify the application behavior. We argue that it is beneficial to complement testing with formal verification in order to detect errors in early stages of development. In this paper, we present the research prototype tool REX. REX is built as a loosely coupled front end to the timed-automata CASE tool UPPAAL. CEP applications and application specific properties can be specified in REX. To support formal verification, REX seamlessly transforms the CEP application together with the specified properties to the timed automata CASE tool UPPAAL where the properties are verified by the model-checker provided by UPPAAL.

    • Despite proven successful in previous projects, the use of formal methods for enhancing quality of software is still not used in its full potential in industry. We argue that seamless support for formal verification in a high-level specification tool enhances the attractiveness of using a formal approach for increasing software quality.Commercial Complex Event Processing (CEP) engines often have support for modelling, debugging and testing CEP applications. However, the possibility of utilizing formal analysis is not considered. We argue that using a formal approach for verifying a CEP system can be performed without expertise in formal methods. In this paper, a prototype tool REX is presented with support for specifying both CEP systems and correctness properties of the same application in a high-level graphical language. The specified CEP applications are seamlessly transformed into a timed automata representation together with the high-level properties for automatic verification in the model-checker UPPAAL.

    • Formal methods are not used in their full potential for enhancing software quality in industry. We argue that seamless support in a high-level specification tool is a viable way to provide system designers with powerful and paradigm specific formal verification techniques. Event condition action (ECA) rules can be used to model and implement reactive behavior in, for example, the semantic web. Independently of target system, the behavior of rule-based systems are known to be hard to analyze. The REX tool is a rule-based front-end to the timed automata CASE-tool Uppaal. The model-checker in Uppaal is used by REX enabling seamless support for model-checking rule-based specifications in REX.This paper presents experiences from modeling and verifying a system of industrial complexity as interacting rules using EX. We conclude that repeatedly performing normal analysis when constructing a system with interacting rules is a viable way of coping with the complexity of the model. Additionally, we present an implemented algorithm for optimizing the model to reduce the effect of state-space explosion.

  • 2007
  • Ingår i: Welding Journal. - 0043-2296. ; 86:8, s. 238-244
  • Tidskriftsartikel (refereegranskat)abstract
    • Simulation tools to search for optimal process parameters are of great interest to reduce the number of experiments and thereby reduce cost and production time. In this paper, robot simulation has been used in combination with finite element simulations to optimize robot speed in order to minimize distortion while keeping complete joint penetration. In an earlier work performed by the authors, a finite element model was developed to predict heat transfer and residual stresses of parts with complex shapes. An interface between a robot simulation model and a finite element analysis model was also constructed. In this paper, an iterative method for robot speed optimization has been developed using MATLAB. The algorithm is designed to maintain complete joint penetration while maximizing productivity by utilizing the fastest weld speed. The method makes it possible to optimize the heat input to the component and thereby minimize component deformation for parts with complex shapes. The system was evaluated on stainless steel plates with varying thicknesses. Robot weld paths were defined off line and automatically downloaded to the finite element software where the optimization was performed. Simulations and experimental validations are presented.

    • The theory and applications of Smart Factories and Industry 4.0 are increasing the entry into the industry. It is common in industry to start converting exclusive parts, of their production, into this new paradigm rather than converting whole production lines all at once. In Europe and Sweden, recent political decisions are taken to reach the target of greenhouse gas emission reduction. One possible solution is to replace concrete in buildings with Cross Laminated Timber. In the last years, equipment and software that have been custom made for a certain task, are now cheaper and can be adapted to fit more processes than earlier possible. This in combination, with lessons learned from the automotive industry, makes it possible to take the necessary steps and start redesigning and building tomorrows automated and flexible production systems in the wood industry. This paper presents a proof of concept of an automated inspection system, for wood surfaces, where concepts found in Industry 4.0, such as industrial Internet of things (IIoT), smart factory, flexible automation, artificial intelligence (AI), and cyber physical systems, are utilized. The inspection system encompasses, among other things, of the shelf software and hardware, open source software, and standardized, modular, and mobile process modules. The design of the system is conducted with future expansion in mind, where new parts and functions can be added as well as removed.

  • Konferensbidrag (refereegranskat)abstract

  • 2002
  • Ingår i: 6th International Conference on Trends in Welding Research. - Materials Park : ASM International. ; , s. 930-935
  • Konferensbidrag (refereegranskat)abstract
    • This paper describes the application of infrared (IR) thermal imaging and temperature measurements in welding applications, both on single plane plates and on an aero engine turbine component with complex geometry. Temperature profiles were measured on the plates using thermocouples (T/C) in combination with an IR camera system, and the results were compared. The IR camera was used both in line scan mode (270 Hz scan frequency) and in full frame mode (1 Hz frame rate). Different methods of surface treatments have been tested to handle the problem of the surface emissivity variations due to oxidation during welding. Results from measurements using thermocouples and IR camera is presented in the paper as well as temperature measurements using the IR camera on an turbine exhaust case (TEC) engine component.

    • Metal Deposition (MD) is a rapid prototyping technique to build parts by depositing metal in a required fashion. When a complex-shaped part is to be built, a simulation tool is needed to define robot trajectories. Three different simulation-based methods for robot trajectory generation are introduced and compared in this study. The methods are; reversed milling, adapted rapid prototyping and application programming in a computer aided robotics software. All methods were shown capable of creating robot paths for complex shapes, with the CAR software approach being the most flexible. Using this method, the geometry to be built is automatically sliced into layers and a robot path is automatically generated. The method was tentatively evaluated and appears to provide a powerful technique in the design and optimisation of robot paths for MD. Experiments showed that it is possible to manufacture fully dense parts using an Nd-Yag laser.

  • Simulation of robot paths and heat effects in welding
  • 2006
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • The objective of this thesis is to develop a simulation tool and a method by which robot trajectories, temperature histories, residual stresses and distortion can be analysed and optimized off-line. This was performed by integrating robot simulation software with finite element analysis software. A special interface was created which facilitated information exchange between the two software programs. To validate the method comparisons were made between simulation results and measurements during real welding. The method was used to program welding trajectories, both for planar plates and for parts with complex shapes. The welding trajectories were downloaded to the finite element analysis software where temperature and residual stress prediction were performed. Good agreement was found between the programmed robot trajectory, and the actual trajectory, necessitating only minor adjustments. Temperature measurements were performed using both thermocouples and infrared imaging. Good agreement was also found between the results using these two methods, as well as between predicted and measured temperatures. Predicted residual stress distributions were compared with neutron diffraction measurements and fair agreement was found. A specified software architecture was developed which allowed full time synchronization between different simulation systems. Finally, weld velocity optimization was performed through a developed algorithm making it possible to minimize distortion. The research conducted in the present work indicates that the models and computer programs that were developed could be combined to create powerful tools for the evaluation and optimisation of welding processes.

  • Supporting the use of design knowledge : an assessment of commenting agents
  • 1999
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis contributes to an understanding of the usefulness of and effects from using commenting agents for supporting the use of design knowledge in user interface design. In two empirical studies, we have explored and investigated commenting agents from the aspects of usefulness, appropriateness of different tool behaviour and forms of comments. Our results show a potential value of the commenting approach, but also rais,es several questions concerning the cost and actual effects. The use of formalized design is considered valuable, yet problematic. Such knowledge is valuable in order to achieve reuse, quality assurance, and design training, but hard to use due to the large volumes, complex structures and weak reference to the design context. The use of knowledge-based tools, capable of generating comments on an evolving design, has been seen as a promising approach to providing user interface designers with formalized design knowledge in the design situation. However, there is a lack of empirical explorations of the idea.In our research, we have conducted a three-part study of the usefulness of commenting tools. First, a Wizard-of-Oz study with 16 subjects was performed to investigate designers' perceptions of the usefulness of a commenting tool, along with the appropriateness of different tool behaviors and forms of comment. We focus on tool mode (active/passive support) and mood (imperative/declarative comments). Secondly, eight professional designers participated in an interview about support needs. Thirdly, a conceptual design prototype was tested by 7 designers, using cooperative evaluation. A broad set of qualitative and quantitative methods have been used to collect and analyse data. Our results show that a commenting tool is seen as disturbing but useful (since it affects the user's work situation). Using a commenting tool affects the designer's evaluation behaviour, i.e., there is an indication of some form of knowledge transfer. The short-term result is an increased consciousness in terms of design reflection and guideline usage. In terms of preferred tool behaviour, our results show that imperative presentation, i.e. pointing out ways of overcoming identified design problems, is the easiest to understand. A high perceived mental workload relates to problems detecting comments when using a commenting tool; this means that comments from an active agent risk being overlooked. In addition, a large part of this thesis can be described as a report of our experiences from using Wizard of Oz techniques to study user interface design support tools. We present our experience and advice for future research.

    • The application of commercial software (OLP) packages for robot simulation, and programming, use interactive computer graphics, provide powerful tools for creating welding paths off-line. By the use of such software, problems of robot reach, accessibility, collision and timing can be eliminated during the planning stage. This paper describes how such software can be integrated with a numerical model that predicts temperature-time histories in the solid material. The objective of this integration is to develop a tool for the engineer where robot trajectories and process parameters can be optimized on parts with complex geometry. Such a tool would decrease the number of weld trials, increase productivity and reduce costs. Assumptions and principles behind the modeling techniques are presented together with experimental evaluation of the correlation between modeled and measured temperatures.

  • Konferensbidrag (refereegranskat)abstract
    • New aero engine design puts new demands on the manufacturing methods with increased automation level. Therefore, the use of vision sensors for control and guiding of industrial robots is being increasingly used. In such system, it is need to customise the machine vision system with real components in the real environment which is normally done close to the start-up of the production. This paper addresses a new concept for designing, programming, analysing, testing and verifying a machine vision application early in the design phase, called Virtual Machine Vision. It is based on a robot simulation software where the real machine vision application is simulated before the implementation in the production line. To verify the Virtual Machine Vision concept an advanced stereo vision application was used. Using two captured images from the robot simulated environment, camera calibration, image analysis and stereo vision algorithms are applied to determine a desired welding joint. The information of the weld joint, i.e. robot position and orientation for the weld path, are sent from the machine vision system to the robot control system in the simulation environment and the weld path is updated. The validation of the Virtual Machine Vision concept using the stereo vision application is promising for industrial use, and it is emphasised that the same programs are used in the virtual and real word.

  • 2021
  • Ingår i: Journal of Fish Biology. - : Wiley-Blackwell. - 0022-1112 .- 1095-8649. ; 98:5, s. 1234-1241
  • Tidskriftsartikel (refereegranskat)abstract
    • This study examines the impact of boldness on foraging competition of the highly invasive round goby Neogobius melanostomus Pallas 1815. Individual risk tolerance, or boldness, was measured as the time to resume movement after a simulated predation strike. Fish that resumed movement faster were categorized as "bold," fish that took more time to resume movement were categorized as "shy" and those that fell in between these two categories were determined to have "intermediate" boldness. Competitive impacts of boldness in N. melanostomus were determined in a laboratory foraging experiment in which interspecific (juvenile Atlantic cod Gadus morhua Linnaeus 1758) and intraspecific (intermediate N. melanostomus) individuals were exposed to either bold or shy N. melanostomus competitors. G. morhua consumed fewer prey when competing with bold N. melanostomus than when competing with shy N. melanostomus, whereas intermediately bold N. melanostomus foraging was not affected by competitor boldness. Bold and shy N. melanostomus consumed similar amounts of prey, and the number of interactions between paired fish did not vary depending on the personality of N. melanostomus individuals. Therefore, intraspecific foraging competition was not found to be personality dependent. This study provides evidence that individual differences in boldness can mediate competitive interactions in N. melanostomus; nonetheless, results also show that competition is also governed by other mechanisms that require further study.
