    • The establishment of the bond of sessile marine organisms such as barnacles, mussels, and algae in the marine environment starts with the secretion and the adsorption of the adhesive biopolymers to the substrate. Subsequently, this is followed by the formation of cohesive interactions with the next layer of adhesive biopolymers that are deposited/adsorbed on top of the first layer. These two fundamental processes for the adhesive plaque buildup have been subjected to several investigations in recent years using model molecules, especially Mefp-1 extracted from the blue mussel Mytilus edulis. With the introduction of optical surface-sensitive methods such as ellipsometry, surface plasmon resonance (SPR), and infrared spectroscopy (IR), it has been possible to elucidate both the kinetics of adsorption and structure of the Mefp-1 film. In contrast to adsorption, the cohesive interactions or the cross-linking are not easily followed with these optical methods and new approaches and techniques are required. One such technique that has been useful is the quartz-crystal microbalance with dissipation monitoring (QCM-D), which has been used for cross-linking studies of a variety of biopolymers including bioadhesives from mussel and algae.   • Complexation Chemistry for Tuning Release from Polymer Coatings

    • The strategy of metal ion complexation is employed to design a delivery system for an antifouling agent (AFA) in marine paints. A poly(1-vinylimidazole- co-methyl methacrylate) copolymer (PVM), together with Cu 2+ or Zn 2+ formed a PVM-M 2+ complex. The AFA, Medetomidine, was then coordinated into the complex. The coordination strength was investigated in solution by 1 H NMR and on solid surfaces by using the Quartz Crystal Microbalance with Dissipation monitoring technique (QCM-D) and Surface Plasmon Resonance (SPR). From the 1 H NMR experiments strong interactions were observed between Cu 2+ and the PVM-polymer and between Medetomidine and the PYM-Cu 2+ complex. From the QCM-D and SPR measurements it was shown that Cu 2+ , compared to Zn 2+ , exhibited a larger affinity for the PVM-copolymer surface that resulted in higher degree of swelling of the polymer film. Large amounts of Medetomidine were adsorbed to the PVM-Cu 2+ complex resulting in low desorption rates. However, the adsorbed amount of Medetomidine was lowerto the Zn 2+ doped polymer and a higher desorption rate was observed. These results indicate the possibility of tuning the release of Medetomidine by altering the coordinating metal ion, which may prove to be favorable in a paint formulation. © 2006 American Chemical Society.

  • Investigation of Adsorption and Cross-Linking of a Mussel Adhesive Protein Using Attenuated Total Internal Reflection Fourier Transform Infrared Spectroscopy (ATR-FTIR)

    • Mytilus edulis foot protein 1 (Mefp-1) contains the redox-functional amino acid 3,4-dihydroxyphenylalanine (DOPA), which is a typical feature of most mefp proteins. We have previously shown, using combined optic (ellipsometry) and acoustic (QCM-D) measurements, that the oxidizing agent sodium periodate (NaIO4) and the transition metal ion Cu2+ promote cross-linking of Mefp-1. However, different chemical reaction mechanisms can not be distinguished using these methods. In the present study, we have complemented our previous investigations using Attenuated Total Internal Reflection Fourier Transform Infrared spectroscopy (ATR-FTIR), allowing a spectroscopic analysis of NaIO4 and Cu2+-induced cross-linking of Mefp-1 adsorbed on a ZnSe surface. In aqueous solution, adsorbed Mefp-1 displays absorption bands at 1570, 1472, 1260, and 973 cm(-1). Upon addition of NaIO4 and Cu2+, the absorptions at 1570, 1472, and 973 cm(-1) increase by approximately a factor of two. In contrast, the band at 1260 cm(-1) disappears upon cross-linking using NaIO4, but remains unchanged upon addition of Cu2+. This demonstrates that the band at 1260cm(-1) is attributed to the C O stretching vibration of the side chain hydroxyl groups in DOPA and that Cu2+ forms complexes with DOPA rather than transform it into an o-quinone. Moreover, upon addition of NaIO4 after cross-linking using Cu2+, the band at 1260cm(-1) disappears, indicating that the complex formation between DOPA and Cu2+ is reversed when DOPA is transformed into the o-quinone. These results demonstrate that NaIO4, which initiates a similar reaction to the naturally occurring enzyme catechol oxidase, contributes to the formation of di-DOPA cross-links. In contrast, the dominating contribution to the cross-linking from Cu2+, which is accumulated at high concentrations in the byssus thread of the blue mussel, is via complex formation between the metal and DOPA residues.

  • Antifouling Agent Release from Marine Coatings - Ion Pair Formation/Dissolution for Controlled Release

    • In marine coatings, the ability to sustain constant release of antifouling agents (AFA(s)) over a long period of time has become increasingly important. One efficient approach to prevent that diffusion depletes the paint film's antifouling activity is to adsorb the AFA strongly to a specie with low translational mobility, such as a high molecular weight polymer. Therefore, the AFA, Medetomidine, was adsorbed onto a sulfonated polystyrene-block-poly(ethylene-ran-butylene)-block-polystyrene (SDPS) generating a Medetomidine-SDPS ion pair in an organic solvent. The interaction was investigated by 1 H NMR in butanol (BuOH-d 10 ) and on solid surfaces by the quartz crystal microbalance with dissipation monitoring technique (QCM-D) in two different solvents, seawater and o-xylene. From the NMR studies in butanol (BuOH-d 10 ), a strong interaction between Medetomidine and SDPS was observed. From the QCM-D measurements, differences in affinity between Medetomidine and the SDPS was observed when changing from seawater to o-xylene. In seawater, the interaction was weak and displayed a large degree of reversibility compared to in o-xylene, where the interaction was strong and almost irreversible. Different swelling behaviour was also observed at the solid surfaces depending on the solvent used with o-xylene exhibiting the largest swelling of the polymer layer. © 2006 Elsevier B.V.     • This dissertation examines mealtime conversations between parents and children in eight Swedish and eight Italian middle class, dual-earner households, exploring the ways in which children are engaged in the cooperative construction of social order. The study is part of an international project (cf. Aronsson & Pontecorvo, 2002), coordinated with prior work in the US (cf. Ochs & Kremer-Sadlik, 2013).Study I explores how children’s accounts work during family dinner conversations. So called proto-accounts (laments, multiple repeats, want-statements) and varied verbal accounts are analyzed in relation to age class or prior language socialization experiences.Study II focuses on the use of endearment terms in directive sequences between parents and children. The findings show an asymmetrical distribution of endearment terms, in that only parents make use of them when interactional problems – children’s non-compliance with parental requests in particular – arise.  Study III examines the ways in which Italian parents deploy the discourse marker dai (‘come on’) in directive sequences. This is a flexible linguistic resource that is employed by parents as a cajoling token when children fail to comply with parental requests, hindering the advancement of the in-progress activity.This thesis describes family mealtimes as parent-directed activities where sociality, morality and local understandings of the world (Ochs & Shohet, 2006) are collaboratively re-created and enacted. This confirms the crucial role of everyday family meals as rich cultural sites (Ochs & Shohet, 2006) for reasserting moral attitudes of the family: participants learn moment by moment how to be competent actors that are able to choose between alternative courses of action and that can therefore be held accountable for their actions (Bergmann, 1998: 284). From this point of view, a dinner is paradigmatic of the deep moral sense that permeates the making of a family.

    • Hemoglobin (Hb) in single erythrocytes (red blood cells), adsorbed on polylysine-coated glass surfaces, was studied using resonance Raman spectroscopy and global Raman imaging. The erythrocytes were found to be sensitive to both surface adsorption and laser illumination. Substrate-dependent changes of the cell membrane shape were observed immediately after cell adsorption, while a photo-induced increase of fluorescence was observed for visible excitation (λ=514.5 nm). Concurrent changes in Raman spectra revealed a conversion of oxy-Hb (2+) to the inactive met-Hb state (3+). These effects severely complicate the interpretation of Raman images. However, at a low accumulated photon dose, the preparation method enabled the recording of Raman spectra during the oxygenation cycle of a single erythrocyte in buffer, which illustrates the feasibility of Raman investigations of functional cells in in-vivo environments.

The erythrocytes were found to be sensitive to both surface adsorption and to the laser light. Topological changes of the cell membrane were observed immediately after cell adsorption in Raman images. We observed a photo-induced increase of the fluorescence background occurring simultaneously with a decrease in the Hb Raman signal. Concurrent changes in Raman spectra revealed a conversion of oxy-Hb to the met-Hb state. However, at a low accumulated photon dose, the preparation method enabled the recording of Raman spectra during the oxygenation cycle of a single red blood cell in buffer, which shows that Hb was in an in-vivo environment. Thus, Raman spectroscopy of functional Hb in isolated red blood cells is feasible.

  • Adsorption of antifouling booster biocides on metal oxide nanoparticles: Effect of different metal oxides and solvents

    • Controlling the release rate of biocides (antifouling agents) from a paint coating is a key issue for the development of multi-season antifouling marine coatings. One promising approach is the use of nanoparticles onto which biocides are adsorbed to prevent premature depletion of the biocide. Adsorption of one novel (Medetomidine) and six commercially available and widely used antifouling biocides (Chlorothalonile, Dichlofluanid, Diuron, Irgarol, Seanine, Tolylfluanid) onto oxide nanoparticles (Al2O3, CuO, MgO, SiO2, TiO2, ZnO) was investigated by HPLC and NMR in different organic solvents. Large differences in adsorption strength depending on the type of nanoparticle and solvent employed were observed. It was shown that nanoparticles coordinate preferentially with the imidazole moiety of Medetomidine. Independent of the type of particle this interaction was considerably stronger in comparison to the other biocides. However, the interaction strength was strongly dependant on the type of solvent, where the largest strongest interaction was achieved in o-xylene. In addition field tests were performed where a considerable decrease in release rate was displayed from coatings containing Medetomidine adsorbed to nanoparticles compared to coatings containing Medetomidine as single additive.

    • The aim of the present study is to examine the interactional functions of code-switching in plurilingual conversation, with a particular focus on those related to the elaboration of the social identities of the interlocutors, i. e. their identity work. The adopted analytical model was initiated by Auer, and further developed by Gafaranga, who combined it with the Membership Categorization Analysis elaborated by Sacks. The study is based on a corpus consisting of the audio recorded dinner talk of an Italian-Swedish family whose members use Swedish, Italian and a Ligurian dialect, and focuses on the sequential progression of the talk in interaction, thus based on an approach proper of conversation analysis. The participants create and display their different roles (identities), which are related to both group membership and individuality, a subsequent distinction made by Fant. Code-switching is used as a communicative strategy among the speakers and is used to make relevant identities, as well as a tool for the management of the interaction. The analysis is divided in two distinctive parts: one related to the code-switching connected to the organisational management of the conversation, the other connected to the identity work of the interlocutors. The linguistic codes used within the group are either inclusive (Swedish, Italian) or selective (Ligurian) and the analysis indicates a general inclusion rather than exclusion of interlocutors, as the speakers accommodating to the linguistic preferences and competences of the interlocutor. The speakers, when selecting a code, thus make use of recipient design, a concept worked out by Sacks, Schegloff & Jefferson. They choose code in order to include a particular interlocutor, adjusting to the language preferences of the other. There are some instances of asymmetrical plurilingual conversation, in which the interlocutors maintain different codes, but since the code choice appears to be anticipated by the addressee the interaction runs smoothly between them. As for the direction of the code switch, two different tendencies emerge: one in the switches connected to the identity work of the participants in which the direction of the switches seems to be highly significant. The other in the switches connected to the management of the conversation in which the direction proves to be reversible.
