  • Characterizing prostate cancer risk through multi-ancestry genome-wide discovery of 187 novel risk variants

    • The transferability and clinical value of genetic risk scores (GRSs) across populations remain limited due to an imbalance in genetic studies across ancestrally diverse populations. Here we conducted a multi-ancestry genome-wide association study of 156,319 prostate cancer cases and 788,443 controls of European, African, Asian and Hispanic men, reflecting a 57% increase in the number of non-European cases over previous prostate cancer genome-wide association studies. We identified 187 novel risk variants for prostate cancer, increasing the total number of risk variants to 451. An externally replicated multi-ancestry GRS was associated with risk that ranged from 1.8 (per standard deviation) in African ancestry men to 2.2 in European ancestry men. The GRS was associated with a greater risk of aggressive versus non-aggressive disease in men of African ancestry (P = 0.03). Our study presents novel prostate cancer susceptibility loci and a GRS with effective risk stratification across ancestry groups.

    • The Cancer Genome Atlas (TCGA) Research Network has profiled and analyzed large numbers of human tumors to discover molecular aberrations at the DNA, RNA, protein and epigenetic levels. The resulting rich data provide a major opportunity to develop an integrated picture of commonalities, differences and emergent themes across tumor lineages. The Pan-Cancer initiative compares the first 12 tumor types profiled by TCGA. Analysis of the molecular aberrations and their functional roles across tumor types will teach us how to extend therapies effective in one cancer type to others with a similar genomic profile. © 2013 Nature America, Inc.     • We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • In search and rescue scenarios local information on victims and other finds needs to be disseminated rapidly toother rescue workers and team leaders. However, post disaster scenarios may imply the collapse of informationinfrastructure including cellular communication and Internet connectivity. Even if we consider wireless ad hoccommunication as a means of information dissemination we should count on frequent loss of connectivity in thenetwork due to unpredictable mobility and sparse network topologies. In this paper we present the realization ofan existing manycast protocol (random walk gossip) on commodity handheld devices running the Androidplatform. This communication mode is used to demonstrate the potential for distributed informationdissemination on victims and finds. The application layer is an adaptation of an existing surveying informationtool (POSIT) which is now fully decentralized and relies on text communication to achieve energy efficiency.

  • DMO processing on the Ketzin 3D seismic data

    • The Dip-moveout (DMO) correction is a process which attempts to make the finite offset data closer to zero offset data after the normal-moveout (NMO) correction. The NMO correction is then dip independent and reflections with different dips will stack coherently. DMO plays a critical role in seismic processing by enhancing the final image quality of the seismic data. In this study, we apply 3D Squeezing DMO (Hale and Artley, 1993) to seismic data from the Ketzin pilot CO2 site after NMO to study the impact of DMO on time-lapse seismic imaging and to investigate if it enhances the CO2 seismic monitoring technique. This 3D DMO method is based on an integral approach and incorporates Hale and Artley’s (1993) modifications for variable velocity with time. A constant velocity algorithm is used with a gamma correction function which depends on the velocity function. An anti-alias velocity of 3000 m/s is used for the DMO. After DMO the data are stacked and F-XY deconvolution is applied. Finally, 3D finite-difference migration using the final smoothed NMO velocities is performed for each data set.We then apply a time-lapse analysis to the 3D seismic data sets and compare the results with and without DMO processing. The most important aspect of the DMO processing is determining the velocity field for the NMO step. This is done by using the initial smoothed velocity field obtained from the conventional velocity analysis before DMO as a first estimate. The data are input into the DMO process and then inverse NMO is applied. These data are then subjected to a new velocity analysis and the velocity field is updated and used as input for the NMO process. A number of iterations are generally required until the velocity field does not need further updating. In this study velocities were picked at every 20th CDP in the inline and crossline directions. Compared to the velocity spectrum without DMO processing, the velocity trend is improved and the ambiguity in the velocity picks is eliminated after DMO correction. The improved accuracy of velocity picking makes it easier to interpret the velocity spectrum and obtain correct interval velocities. Considering the stacked section, DMO suppresses the random noise to a greater extent and thus the signal-to-noise ratio is enhanced. From the comparison of the amplitude difference horizon at the reservoir level, the shape of the anomaly observed in the data with DMO processing is similar to that observed in the data without DMO processing. However, the amplitude anomalies of the former are stronger than those of the latter, especially close to the injection well. In addition, one stronger amplitude anomaly in the DMO time-lapse horizon indicates a preferred trend of the CO2 migration in WNW direction due to the reservoir heterogeneity.

  • Integration of Reservoir Simulation with 3D Reflection Seismic Time-lapse Data at Ketzin

    • In this study we integrate reservoir simulation with 3D reflection seismic time-lapse data from the Ketzin CO2 storage pilot site using density and velocity models, a porosity model and a fluid flow simulation model. To simulate the seismic response corresponding to the real data, detailed density and velocity models are properly generated from the stratigraphic model and available borehole logging data. The reservoir simulation model providing the spatial CO2 saturation in the reservoir based on numerical simulations history-matched to the site monitoring data is used to calculate the bulk densities and compressional wave velocities of the rock after CO2 injection using a fluid substitution method. 3D synthetic seismic data are generated by convolving the density and velocity models with two different sources to study the change in seismic response after CO2 injection and to investigate the impact of the different source types on CO2 monitoring. Our results provide estimates of the time-lapse anomalies caused by CO2 injection and demonstrate that the choice of source type is a critical factor for time-lapse seismic monitoring to assess the distribution of injected CO2. The same source type should be used in the baseline and repeat surveys for site operation.

    • There is limited convergence in neuroimaging investigations into volumes of subcortical brain regions in social anxiety disorder (SAD). The inconsistent findings may arise from variations in methodological approaches across studies, including sample selection based on age and clinical characteristics. The ENIGMA-Anxiety Working Group initiated a global mega-analysis to determine whether differences in subcortical volumes can be detected in adults and adolescents with SAD relative to healthy controls. Volumetric data from 37 international samples with 1115 SAD patients and 2775 controls were obtained from ENIGMA-standardized protocols for image segmentation and quality assurance. Linear mixed-effects analyses were adjusted for comparisons across seven subcortical regions in each hemisphere using family-wise error (FWE)-correction. Mixed-effects d effect sizes were calculated. In the full sample, SAD patients showed smaller bilateral putamen volume than controls (left: d = −0.077, pFWE = 0.037; right: d = −0.104, pFWE = 0.001), and a significant interaction between SAD and age was found for the left putamen (r = −0.034, pFWE = 0.045). Smaller bilateral putamen volumes (left: d = −0.141, pFWE < 0.001; right: d = −0.158, pFWE < 0.001) and larger bilateral pallidum volumes (left: d = 0.129, pFWE = 0.006; right: d = 0.099, pFWE = 0.046) were detected in adult SAD patients relative to controls, but no volumetric differences were apparent in adolescent SAD patients relative to controls. Comorbid anxiety disorders and age of SAD onset were additional determinants of SAD-related volumetric differences in subcortical regions. To conclude, subtle volumetric alterations in subcortical regions in SAD were detected. Heterogeneity in age and clinical characteristics may partly explain inconsistencies in previous findings. The association between alterations in subcortical volumes and SAD illness progression deserves further investigation, especially from adolescence into adulthood.

  • 3D Time-lapse Analysis of Seismic Reflection Data to Characterize the Reservoir at the Ketzin CO2 Storage Pilot Site

    • 3D time-lapse seismics, also known as 4D seismics, have great potential for monitoring the migration of CO2 at underground storage sites. This thesis focuses on time-lapse analysis of 3D seismic reflection data acquired at the Ketzin CO2 geological storage site in order to improve understanding of the reservoir and how CO2 migrates within it.Four 3D seismic surveys have been acquired to date at the site, one baseline survey in 2005 prior to injection, two repeat surveys in 2009 and 2012 during the injection period, and one post-injection survey in 2015. To accurately simulate time-lapse seismic signatures in the subsurface, detailed 3D seismic property models for the baseline and repeat surveys were constructed by integrating borehole data and the 3D seismic data. Pseudo-boreholes between and beyond well control were built. A zero-offset convolution seismic modeling approach was used to generate synthetic time-lapse seismograms. This allowed simulations to be performed quickly and limited the introduction of artifacts in the seismic responses.Conventional seismic data have two limitations, uncertainty in detecting the CO2 plume in the reservoir and limited temporal resolution. In order to overcome these limitations, complex spectral decomposition was applied to the 3D time-lapse seismic data. Monochromatic wavelet phase and reflectivity amplitude components were decomposed from the 3D time-lapse seismic data. Wavelet phase anomalies associated with the CO2 plume were observed in the time-lapse data and verified by a series of seismic modeling studies. Tuning frequencies were determined from the balanced amplitude spectra in an attempt to discriminate between pressure effects and CO2 saturation. Quantitative assessment of the reservoir thickness and CO2 mass were performed.Time-lapse analysis on the post-injection survey was carried out and the results showed a consistent tendency with the previous repeat surveys in the CO2 migration, but with a decrease in the size of the amplitude anomaly. No systematic anomalies above the caprock were detected. Analysis of the signal to noise ratio and seismic simulations using the detailed 3D property models were performed to explain the observations. Estimation of the CO2 mass and uncertainties in it were investigated using two different approaches based on different velocity-saturation models.

  • Application of seismic complex decomposition on thin layer detection of the CO2 plume at Ketzin, Germany

    • The seismic complex decomposition technique is a spectral decomposition method using inversion strategies to decompose a seismic trace into its constituent frequencies and corresponding complex coefficients. This method has high time-frequency resolution and it is not necessary to select a signal window in comparison to conventional spectral decomposition methods. The thickness of the reservoir at the Ketzin pilot site is relatively thin, making it difficult to resolve seismically due to the band-limited seismic spectrum. This study presents an application of seismic complex decomposition to the time-lapse 3D seismic datasets at the Ketzin pilot site for estimating the temporal thickness of the injected CO2 within the thin reservoir via frequency tuning. Quantitative analysis for CO2 thickness and mass is investigated. Comparison between the real recorded data and the estimates shows that our results are reliable in assessing the amount of the CO2 in the plume at the Ketzin pilot site.

    • Spectral decomposition is a powerful tool that can provide geological details dependent upon discrete frequencies. Complex spectral decomposition using inversion strategies differs from conventional spectral decomposition methods in that it produces not only frequency information but also wavelet phase information. This method was applied to a time‐lapse three‐dimensional seismic dataset in order to test the feasibility of using wavelet phase changes to detect and map injected carbon dioxide within the reservoir at the Ketzin carbon dioxide storage site, Germany. Simplified zero‐offset forward modelling was used to help verify the effectiveness of this technique and to better understand the wavelet phase response from the highly heterogeneous storage reservoir and carbon dioxide plume. Ambient noise and signal‐to‐noise ratios were calculated from the raw data to determine the extracted wavelet phase. Strong noise caused by rainfall and the assumed spatial distribution of sandstone channels in the reservoir could be correlated with phase anomalies. Qualitative and quantitative results indicate that the wavelet phase extracted by the complex spectral decomposition technique has great potential as a practical and feasible tool for carbon dioxide detection at the Ketzin pilot site.

    • In order to simulate the 3D time-lapse seismic response from real data in a consistent manner detailed 3D property models at the Ketzin pilot site were constructed by robustly integrating borehole and 3D reflection seismic data. The spatial CO2 distribution and the detailed CO2 density in the reservoir were derived from dynamic flow simulations that had been history-matched to the site monitoring data. Changes in velocity and density after CO2 injection were estimated utilizing the CO2 saturation distributions at two repeat times and fluid substitution models. 4D seismic data were generated by convolving the property models at different times with an extracted wavelet. Time-lapse analysis was performed to qualitatively and quantitatively investigate the changes in reflection amplitude after CO2 injection. Comparison between the synthetic and real data at the corresponding time indicates that the 3D property models were built successfully and model the 3D time-lapse seismic response induced by CO2 injection. A synthetic experiment with two different source wavelets was implemented to investigate the impact of source non-repeatability on the seismic amplitude anomaly. Analysis shows that the same or similar sources should be used in time-lapse seismic monitoring to minimize the impact of source non-repeatability on the monitoring.

  • Preliminary Seismic Time-lapse Results from the First Post-injection Survey at the Ketzin Pilot Site

    • Since the CO2SINK project start in April 2004, different methods involving seismics, geoelectrics andpressure-temperature monitoring have been applied at the Ketzin pilot site to map geological structuresand assess CO2 distribution and movement. One 3D baseline seismic survey prior to CO2 injection andtwo 3D repeat seismic surveys during CO2 injection have proven that the 3D time-lapse surface seismicmethod is an effective tool for providing good quality images of the CO2 plume. In the autumn of 2015, athird 3D repeat seismic survey, serving as the first post-injection survey, was acquired. The preliminaryresults show that the predominant westward trend of the CO2 migration is in agreement with the previousresults. No CO2 migration upwards through the caprock was detected. The decrease in the size of CO2plume may be related to ongoing dissolution into the saline formation water.

    • Determining thin layer thickness is very important for reservoir characterization and CO2 quantification. Given its high time-frequency resolution and robustness, the complex spectral decomposition method was applied on time-lapse 3D seismic data from the Ketzin pilot site for CO2 storage to evaluate the frequency-dependent characteristics of thin layers at the injection level. Higher temporal resolution and more stratigraphic details are seen in the all-frequency and monochromatic reflectivity amplitude sections obtained by complex spectral decomposition compared to the stacked sections. The mapped geologic discontinuities within the reservoir are consistent with the preferred orientation of CO2 propagation. Tuning frequency mapping shows the thicknesses of the reservoir sandstone and gaseous CO2 is consistent with the measured thickness of the sandstone unit from well logging. An attempt to discriminate between pressure effects and CO2 saturation using the extracted tuning frequency indicates that CO2 saturation is the main contributor to the amplitude anomaly at the Ketzin site. On the basis of determined thickness of gaseous CO2 in the reservoir, quantitative analysis of the amount of CO2 was performed and shows a discrepancy between the injected and calculated CO2 mass. This may be explained by several uncertainties, like structural reservoir heterogeneity, a limited understanding of the complex subsurface conditions, error of determined tuning frequency, the presence of ambient noise and ongoing CO2 dissolution.

  • The first post-injection seismic monitor survey at the Ketzin pilot CO2 storage site : results from time-lapse analysis

    • The injection of CO2 at the Ketzin pilot CO2 storage site started in June 2008 and ended in August 2013. During the 62 months of injection, a total amount of about 67 kt of CO2 was injected into a saline aquifer. A third repeat 3D seismic survey, serving as the first post-injection survey was acquired in 2015, aiming to investigate the recent movement of the injected CO2. Consistent with the previous two time-lapse surveys, a predominantly WNW migration of the gaseous CO2 plume in the up-dip direction within the reservoir is inferred in this first post-injection survey. No systematic anomalies are detected through the reservoir overburden. The extent of the CO2 plume west of the injection site is almost identical to that found in the 2012 second repeat survey (after injection of 61 kt), however there is a significant decrease in its size east of the injection site. Assessment of the CO2 plume distribution suggests that the decrease in the size of the anomaly may be due to multiple factors, such as limited vertical resolution, CO2 dissolution and CO2 diffusion, in addition to the effects of ambient noise. 4D seismic modelling based on dynamic flow simulations indicates that a dynamic balance between the newly injected CO2 after the second repeat survey and the CO2 being dissolved and diffused was reached by the time of the first post-injection survey. Considering the considerable uncertainties in CO2 mass estimation, both patchy and non-patchy saturation models for the Ketzin site were taken into consideration.

    • The injection of CO2 at the Ketzin pilot site commenced in June 2008 and was terminated in August 2013 after 67 kT had been injected into a saline formation at a depth of 630-650 m. As part of the site monitoring program, four 3D surface seismic surveys have been acquired to date, one baseline and three repeats, of which two were conducted during the injection period, and one during the post-injection phase. The surveys have provided the most comprehensive images of the spreading CO2 plume within the reservoir layer. Both petrophysical experiments on core samples from the Ketzin reservoir and spectral decomposition of the 3D time-lapse seismic data show that the reservoir pore pressure change due to CO2 injection has a rather minor impact on the seismic amplitudes. Therefore, the observed amplitude anomaly is interpreted to be mainly due to CO2 saturation. In this study, amplitude versus offset analysis has been applied to investigate the amplitude versus offset response from the top of the sandstone reservoir during the injection and post-injection phases, and utilize it to obtain a more quantitative assessment of the CO2 gaseous saturation changes. Based on the amplitude versus offset modelling, a prominent decrease in the intercept values imaged at the top of the reservoir around the injection well is indeed associated solely with the CO2 saturation increase. Any change in the gradient values, which would, in case it was positive, be the only signature induced by the reservoir pressure variations, has not been observed. The amplitude versus offset intercept change is, therefore, entirely ascribed to CO2 saturation and used for its quantitative assessment. The estimated CO2 saturation values around the injection area in the range of 40%-60% are similar to those obtained earlier from pulsed neutron-gamma logging. The highest values of 80% are found in the second seismic repeat in close vicinity to the injection and observation wells.

To identify common susceptibility alleles, we performed the largest pancreatic cancer GWAS to date, including 9040 patients and 12,496 controls of European ancestry from the Pancreatic Cancer Cohort Consortium (PanScan) and the Pancreatic Cancer Case-Control Consortium (PanC4). Here, we find significant evidence of a novel association at rs78417682 (7p12/TNS3, P = 4.35 x 10(-8)). Replication of 10 promising signals in up to 2737 patients and 4752 controls from the PANcreatic Disease ReseArch (PAN-DoRA) consortium yields new genome-wide significant loci: rs13303010 at 1p36.33 (NOC2L, P = 8.36 x 10(-14)), rs2941471 at 8q21.11 (HNF4G, P = 6.60 x 10(-10)), rs4795218 at 17q12 (HNF1B, P = 1.32 x 10(-8)), and rs1517037 at 18q21.32 (GRP, P = 3.28 x 10(-8)). rs78417682 is not statistically significantly associated with pancreatic cancer in PANDoRA. Expression quantitative trait locus analysis in three independent pancreatic data sets provides molecular support of NOC2L as a pancreatic cancer susceptibility gene.

    • Smart photovoltaic windows represent a promising green technology featuring tunable transparency and electrical power generation under external stimuli to control the light transmission and manage the solar energy. Here, we demonstrate a thermochromic solar cell for smart photovoltaic window applications utilizing the structural phase transitions in inorganic halide perovskite caesium lead iodide/bromide. The solar cells undergo thermally-driven, moisture-mediated reversible transitions between a transparent non-perovskite phase (81.7% visible transparency) with low power output and a deeply coloured perovskite phase (35.4% visible transparency) with high power output. The inorganic perovskites exhibit tunable colours and transparencies, a peak device efficiency above 7%, and a phase transition temperature as low as 105 degrees C. We demonstrate excellent device stability over repeated phase transition cycles without colour fade or performance degradation. The photovoltaic windows showing both photoactivity and thermochromic features represent key stepping-stones for integration with buildings, automobiles, information displays, and potentially many other technologies.

  • The Child as Econometrician : A Rational Model of Preference Understanding in Children

    • Recent work has shown that young children can learn about preferences by observing the choices and emotional reactions of other people, but there is no unified account of how this learning occurs. We show that a rational model, built on ideas from economics and computer science, explains the behavior of children in several experiments, and offers new predictions as well. First, we demonstrate that when children use statistical information to learn about preferences, their inferences match the predictions of a simple econometric model. Next, we show that this same model can explain children's ability to learn that other people have preferences similar to or different from their own and use that knowledge to reason about the desirability of hidden objects. Finally, we use the model to explain a developmental shift in preference understanding.

  • 4D Seismic Monitoring of CO2 Storage During Injection and Post-closure at the Ketzin Pilot Site

    • At the Ketzin pilot site for geological CO2 storage, about 67,000 tons of CO2 were injected during the period June 2008 – August 2013. Since August 2013, the site is in its post-closure phase. Before and during the injection phase, a comprehensive monitoring programme was established. In the early post-injection phase, a majority of the monitoring activities have continued. The stepwise abandonment of the pilot site, which is planned to be accomplished in 2018, marks also the termination of most monitoring activities. Four 3D seismic surveys were acquired between 2005 and 2015 for characterizing the reservoir structure and its overburden and for monitoring the propagation of the injected CO2 in the storage formation. The first and second repeat surveys revealed the lateral extension of the CO2 plume after injecting 22 and 61 ktons, respectively. In autumn 2015, the third 3D repeat seismic survey, serving as the first post-injection survey, was acquired. The survey was acquired using the same acquisition geometry as for previous surveys, consisting of 33 templates with five receiver lines and twelve source profiles perpendicular to the receiver lines. Seismic processing of the recently acquired data has resulted in preliminary observations which can be summarized as follows: As in previous seismic repeat surveys, a clear CO2 signature is observed at the top of the storage formation. No systematic amplitude changes are observed above the reservoir which might indicate leakage. Compared to the second repeat survey acquired in 2012, the lateral extent of the CO2 plume seems to have been reduced, which may be an indication for ongoing (and relatively fast) dissolution of the CO2 in the formation brine and diffusion into very thin layers indicating pressure release.

  • Epithelioid and Spindle Cell Hemangioma : Clinicopathologic Analysis of 18 Primary Bone and Soft Tissue Tumors Highlighting a Predilection for the Hands and Feet, Frequent Multicentricity, and Benign Behavior

    • Epithelioid and spindle cell hemangioma was initially described in 1999 in a series of primary bone tumors and was subsequently suggested by some to represent a variant of epithelioid hemangioma. Here, we studied 18 epithelioid and spindle cell hemangiomas. Nine patients (50%) were male. Age at presentation ranged from 12 to 78 years (median: 38.5 y). Nine patients (50%) had tumor(s) limited to bone, 5 (28%) had tumor(s) limited to soft tissue, and 4 (22%) had tumor(s) involving bone and soft tissue. Nine patients (50%) had multiple tumors, all in a unilateral anatomic region involving the wrist, hand, ankle, or foot. Seventeen tumors (94%) occurred in an extremity, including 12 (67%) in the hands and feet, and 1 occurred in a vertebra. In imaging studies, primary bone tumors were lobulated, expansile, and lytic, and 7 bone tumors with available imaging (58%) showed cortical breakthrough. Tumor sizes were 0.8 to 7.2 cm (median: 2.2 cm). Epithelioid and spindle cell hemangioma is composed of lobules of epithelioid and spindled endothelial cells with bland, vesicular nuclei. Neoplastic cells show orderly vasoformative growth, with hemorrhagic stroma and no endothelial atypia or multilayering. Immunohistochemistry demonstrated uniform positivity for CD31 and ERG. Where positive, SMA highlighted pericytes (11/13 tumors). FOSB was strongly positive in 4 of 16 tumors (25%), and FOS was strongly positive in 5 of 10 stained tumors (50%). Break-apart fluorescence in situ hybridization confirmed the presence of FOS split signals in 4 tumors positive for FOS by immunohistochemistry and FOSB split signals in 2 FOSB-positive tumors. DNA sequencing demonstrated a GATA6::FOXO1 fusion in 1 of 3 sequenced tumors. Clinical follow-up was available for 15 patients (83%; range: 5 mo to 11 y; median: 3.5 y). Seven patients (47%) had no evidence of disease at most recent follow-up. Seven of 13 patients (54%) who underwent surgery experienced local recurrence at the primary tumor site: 5 patients within a year, 1 at 2.4 years, and 1 thrice at 2, 3, and 5 years. Six patients were alive with multifocal disease (median: 3.5 y; range: 5 mo to 6 y). No tumors gave rise to distant metastases. The clinicopathologic and genetic findings in this study support the notion that epithelioid and spindle cell hemangioma is a morphologic variant of epithelioid hemangioma that can occur in soft tissue as well as bone and that shows a striking predilection for the extremities. Given that most recurrences and primary tumors behaved indolently, watchful waiting would be reasonable for patients with multicentric disease that is not readily amenable to surgery.

    • Recent developments in compact object astrophysics, especially the discovery of merging neutron stars by LIGO, the imaging of the black hole in M87 by the Event Horizon Telescope, and high- precision astrometry of the Galactic Center at close to the event horizon scale by the GRAVITY experiment motivate the development of numerical source models that solve the equations of general relativistic magnetohydrodynamics (GRMHD). Here we compare GRMHD solutions for the evolution of a magnetized accretion flow where turbulence is promoted by the magnetorotational instability from a set of nine GRMHD codes: Athena++, BHAC, Cosmos++, ECHO, H-AMR, iharm3D, HARM-Noble, IllinoisGRMHD, and KORAL. Agreement among the codes improves as resolution increases, as measured by a consistently applied, specially developed set of code performance metrics. We conclude that the community of GRMHD codes is mature, capable, and consistent on these test problems.

    • The Colorado potato beetle is one of the most challenging agricultural pests to manage. It has shown a spectacular ability to adapt to a variety of solanaceaeous plants and variable climates during its global invasion, and, notably, to rapidly evolve insecticide resistance. To examine evidence of rapid evolutionary change, and to understand the genetic basis of herbivory and insecticide resistance, we tested for structural and functional genomic changes relative to other arthropod species using genome sequencing, transcriptomics, and community annotation. Two factors that might facilitate rapid evolutionary change include transposable elements, which comprise at least 17% of the genome and are rapidly evolving compared to other Coleoptera, and high levels of nucleotide diversity in rapidly growing pest populations. Adaptations to plant feeding are evident in gene expansions and differential expression of digestive enzymes in gut tissues, as well as expansions of gustatory receptors for bitter tasting. Surprisingly, the suite of genes involved in insecticide resistance is similar to other beetles. Finally, duplications in the RNAi pathway might explain why Leptinotarsa decemlineata has high sensitivity to dsRNA. The L. decemlineata genome provides opportunities to investigate a broad range of phenotypes and to develop sustainable methods to control this widely successful pest.

  • A comparison of small, affordable seismic sources at the Ketzin CO2 storage site, Germany

    • Seismic methods have proven to be effective for monitoring the movement and location of injected CO2 within deep saline aquifers. However, a disadvantage of seismic monitoring is the high costs associated with many repeat seismic surveys as part of a long term monitoring strategy of a CO2 storage site. As the cost for the use of the seismic source is often a significant part of the overall survey cost, affordable, smaller sources would increase the potential feasibility of a long term seismic monitoring strategy. A comparison of three land seismic sources is performed at the Ketzin CO2 storage site, Germany. Two of these sources (Vibsist 500 and Bobcat drop hammer) can be considered to be smaller and more affordable sources than those conventionally used in the seismic monitor surveys at Ketzin. In this study these smaller sources are compared to a larger more conventional Vibsist 3000 source. The subsurface target for the three sources in this comparison is the CO2 storage reservoir for the Ketzin site, located within the Triassic Stuttgart formation, which lies at a depth of approximately 600m/500ms. Two of the sources are Swept Impact (SIST) type courses (Vibsist 500 and 3000) which use hydraulic concrete breaking hammers. The third source uses a concrete breaking drop hammer tool mounted on a Bobcat loader. Data were collected along a 984m long profile with 24m receiver spacing and 12m shot spacing in 2011, 2012 and 2013 using the three different sources. A quantitative and qualitative comparison of the raw data from the three sources was performed in order to assess their relative performance. Frequency content, signal to noise ratio and penetration depth curves were calculated for the raw data. Data from the three sources was also processed using a conventional workflow to produce stacked sections which were compared. Based on the results from this study the Bobcat drop hammer source appears to perform better than the Vibsist 500 source. However both of the smaller sources were capable of producing good images of the target CO2 storage reservoir. Hence, both provide viable options as small affordable seismic sources for long term monitoring at the Ketzin site, or other shallow CO2 storage sites.

    • We apply a range of quantitative pre-stack analysis techniques to assess the feasibility of using smaller and cheaper seismic sources, than those currently used at the Ketzin CO2 storage site. Results from two smaller land sources are presented alongside those from a larger, more powerful source, typically utilized for seismic acquisition at the Ketzin. The geological target for the study is the Triassic Stuttgart Formation which contains a saline aquifer currently used for CO2 storage. The reservoir lies at a depth of approximately 630 m, equivalent to a travel time of 500 ms along the study profile. The three sources discussed in the study are the Vibsist 3000, Vibsist 500 (using industrial hydraulic driven concrete breaking hammers) and a drop hammer source. Data were collected for the comparison using the three sources in 2011, 2012 and 2013 along a 984 m long line with 24m receiver spacing and 12 m shot spacing. Initially a quantitative analysis is performed of the noise levels between the 3 surveys. The raw shot gathers are then analyzed quantitatively to investigate the relative energy output, signal to noise ratio, penetration depth, repeatability and frequency content for the different sources. The performance of the sources is also assessed based on stacked seismic sections. Based on the results from this study it appears that both of the smaller sources are capable of producing good images of the target reservoir and can both be considered suitable as lower cost, less invasive sources for use at the Ketzin site or other shallow CO2 storage projects. Finally, the results from the various pre-stack analysis techniques are discussed in terms of how representative they are of the final stacked sections.

  • The TESS-Keck Survey. XI. Mass Measurements for Four Transiting Sub-Neptunes Orbiting K Dwarf TOI-1246

    • Multiplanet systems are valuable arenas for investigating exoplanet architectures and comparing planetary siblings. TOI-1246 is one such system, with a moderately bright K dwarf (V = 11.6, K = 9.9) and four transiting sub-Neptunes identified by TESS with orbital periods of 4.31, 5.90, 18.66, and 37.92 days. We collected 130 radial velocity observations with Keck/HIRES and TNG/HARPS-N to measure planet masses. We refit the 14 sectors of TESS photometry to refine planet radii (2.97 +/- 0.06 R (circle plus), 2.47 +/- 0.08 R (circle plus), 3.46 +/- 0.09 R (circle plus), and 3.72 +/- 0.16 R (circle plus)) and confirm the four planets. We find that TOI-1246 e is substantially more massive than the three inner planets (8.1 +/- 1.1 M (circle plus), 8.8 +/- 1.2 M (circle plus), 5.3 +/- 1.7 M (circle plus), and 14.8 +/- 2.3 M (circle plus)). The two outer planets, TOI-1246 d and TOI-1246 e, lie near to the 2:1 resonance (P (e)/P ( d ) = 2.03) and exhibit transit-timing variations. TOI-1246 is one of the brightest four-planet systems, making it amenable for continued observations. It is one of only five systems with measured masses and radii for all four transiting planets. The planet densities range from 0.70 +/- 0.24 to 3.21 +/- 0.44 g cm(-3), implying a range of bulk and atmospheric compositions. We also report a fifth planet candidate found in the RV data with a minimum mass of 25.6 +/- 3.6 M (circle plus). This planet candidate is exterior to TOI-1246 e, with a candidate period of 93.8 days, and we discuss the implications if it is confirmed to be planetary in nature.

    • Monitoring of the CO2 distribution at depth is imperative for onshore geological storage of CO2. Seismic methods are effective monitoring tools during and after the injection process, but are generally expensive and time consuming to perform. In this paper we perform a series of synthetic experiments in order to compare the seismic waveform inversion method with conventional seismic monitoring methods for time-lapse monitoring at CO2 geological storage sites. We mainly focus our study on seismic data sets with typical reflection acquisition geometries that is far offset limited. Two synthetic seismic data sets (consisting of one baseline and repeat) were generated by a third party 2D forward modeling code to compare the waveform inversion method with a conventional time-lapse procedure. The velocity models are based on a simplified structure of the Heletz site in Israel. The area into which CO2 is injected is on the order of a few hundred meters wide and a few 10 s of meters high. We compare the waveform inversion results and conventional time-lapse results to determine how sparse spatial sampling affects the results by increasing both the shot and receiver spacing. We test the waveform inversion method for a synthetic time-lapse data set with different inversion strategies. First, we invert each data set independently by using both phase and amplitude information. Then we invert them by using only phase information. We also test the influence of the starting model. Finally, we test the double difference inversion method which is equivalent to only inverting the difference between the baseline and repeat data sets. During the test a relative coarse starting model and higher starting frequency were used in order to better represent real seismic reflection data. Our results show that, under certain noise conditions, it may be possible to use a combination of sparse spatial sampling geometries and seismic waveform inversion to monitor the velocity changes caused by CO2 injection. We conclude that seismic waveform inversion may be a good complement to standard CDP processing when monitoring CO2 injection.
