  • EPOXI: Comet 103P/Hartley 2 Observations from a Worldwide Campaign

    • Earth- and space-based observations provide synergistic information for space mission encounters by providing data over longer timescales, at different wavelengths and using techniques that are impossible with an in situ flyby. We report here such observations in support of the EPOXI spacecraft flyby of comet 103P/Hartley 2. The nucleus is small and dark, and exhibited a very rapidly changing rotation period. Prior to the onset of activity, the period was ~16.4?hr. Starting in 2010 August the period changed from 16.6?hr to near 19?hr in December. With respect to dust composition, most volatiles and carbon and nitrogen isotope ratios, the comet is similar to other Jupiter-family comets. What is unusual is the dominance of CO 2 -driven activity near perihelion, which likely persists out to aphelion. Near perihelion the comet nucleus was surrounded by a large halo of water-ice grains that contributed significantly to the total water production.

  • Determinants of morbidity and mortality following emergency abdominal surgery in children in low-income and middle-income countries

    • Background: Child health is a key priority on the global health agenda, yet the provision of essential and emergency surgery in children is patchy in resource-poor regions. This study was aimed to determine the mortality risk for emergency abdominal paediatric surgery in low-income countries globally.Methods: Multicentre, international, prospective, cohort study. Self-selected surgical units performing emergency abdominal surgery submitted prespecified data for consecutive children aged <16 years during a 2-week period between July and December 2014. The United Nation's Human Development Index (HDI) was used to stratify countries. The main outcome measure was 30-day postoperative mortality, analysed by multilevel logistic regression.Results: This study included 1409 patients from 253 centres in 43 countries; 282 children were under 2 years of age. Among them, 265 (18.8%) were from low-HDI, 450 (31.9%) from middle-HDI and 694 (49.3%) from high-HDI countries. The most common operations performed were appendectomy, small bowel resection, pyloromyotomy and correction of intussusception. After adjustment for patient and hospital risk factors, child mortality at 30 days was significantly higher in low-HDI (adjusted OR 7.14 (95% CI 2.52 to 20.23), p<0.001) and middle-HDI (4.42 (1.44 to 13.56), p=0.009) countries compared with high-HDI countries, translating to 40 excess deaths per 1000 procedures performed.Conclusions: Adjusted mortality in children following emergency abdominal surgery may be as high as 7 times greater in low-HDI and middle-HDI countries compared with high-HDI countries. Effective provision of emergency essential surgery should be a key priority for global child health agendas.

    • The Sub-millimetre and Millimetre Radiometer (SMR) is the main instrument on the Swedish, Canadian, Finnish and French spacecraft Odin. It consists of a 1.1 metre diameter telescope with four tuneable heterodyne receivers covering the ranges 486-504 GHz and 541-581 GHz, and one fixed at 118.75 GHz together with backends that provide spectral resolution from 150 kHz to 1 MHz. This Letter describes the Odin radiometer, its operation and performance with the data processing and calibration described in Paper II.

    • Key Odin operational and instrumental features and highlights from our sub-millimetre and millimetre wave observations of H2O, H218O, NH3, 15NH3 and O2 are presented, with some insights into accompanying Odin Letters in this A&A issue. We focus on new results where Odin's high angular resolution, high frequency resolution, large spectrometer bandwidths, high sensitivity or/and frequency tuning capability are crucial: H2O mapping of the Orion KL, W3, DR21, S140 regions, and four comets; H2O observations of Galactic Centre sources, of shock enhanced H2O towards the SNR IC443, and of the candidate infall source IRAS 16293-2422; H218O detections in Orion KL and in comet Ikeya-Zhang; sub-mm detections of NH3 in Orion KL (outflow, ambient cloud and bar) and ρ Oph, and very recently, of 15NH3 in~Orion KL. Simultaneous sensitive searches for the 119 GHz line of O2 have resulted in very low abundance limits, which are difficult to accomodate in chemical models. We also demonstrate, by means of a quantitative comparison of Orion KL H2O results, that the Odin and SWAS observational data sets are very consistently calibrated. Odin is a Swedish-led satellite project funded jointly by the Swedish National Space Board (SNSB), the Canadian Space Agency (CSA), the National Technology Agency of Finland (Tekes), and the Centre National d'études Spatiales (CNES, France). The Swedish Space Corporation (SSC) has been the prime industrial contractor, and is also responsible for the satellite operation from its Odin Mission Control Centre at SSC in Solna and its Odin Control Centre at ESRANGE near Kiruna in northern Sweden.     • Cytotoxic lymphocytes, encompassing cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, kill pathogen-infected, neoplastic, or certain hematopoietic cells through the release of perforin-containing lytic granules. In the present study, we first performed probability-state modeling of differentiation and lytic granule markers on CD8(+) T cells to enable the comparison of bona fide CTLs with NK cells. Analysis identified CD57(bright) expression as a reliable phenotype of granule marker-containing CTLs. We then compared CD3(+)CD8(+)CD57(bright) CTLs with NK cells. Healthy adult peripheral blood CD3(+)CD8(+)CD57(bright) CTLs expressed more granzyme B but less perforin than CD3(-)CD56(dim) NK cells. On stimulation, such CTLs degranulated more readily than other T-cell subsets, but had a propensity to degranulate that was similar to NK cells. Remarkably, the CTLs produced cytokines more rapidly and with greater frequency than NK cells. In patients with biallelic mutations in UNC13D, STX11, or STXBP2 associated with familial hemophagocytic lymphohistiocytosis, CTL and NK cell degranulation were similarly impaired. Therefore, cytotoxic lymphocyte subsets have similar requirements for Munc13-4, syntaxin-11, and Munc18-2 in lytic granule exocytosis. The present results provide a detailed comparison of human CD3(+)CD8(+)CD57(bright) CTLs and NK cells and suggest that analysis of CD57(bright) CTL function may prove useful in the diagnosis of primary immunodeficiencies including familial hemophagocytic lymphohistiocytosis.

    • Background Congenital dyserythropoietic anaemia type I (CDA-I) is a hereditary anaemia caused by biallelic mutations in the widely expressed genes CDAN1 and C15orf41. Little is understood about either protein and it is unclear in which cellular pathways they participate. Methods Genetic analysis of a cohort of patients with CDA-I identifies novel pathogenic variants in both known causative genes. We analyse the mutation distribution and the predicted structural positioning of amino acids affected in Codanin-1, the protein encoded by CDAN1. Using western blotting, immunoprecipitation and immunofluorescence, we determine the effect of particular mutations on both proteins and interrogate protein interaction, stability and subcellular localisation. Results We identify six novel CDAN1 mutations and one novel mutation in C15orf41 and uncover evidence of further genetic heterogeneity in CDA-I. Additionally, population genetics suggests that CDA-I is more common than currently predicted. Mutations are enriched in six clusters in Codanin-1 and tend to affect buried residues. Many missense and in-frame mutations do not destabilise the entire protein. Rather C15orf41 relies on Codanin-1 for stability and both proteins, which are enriched in the nucleolus, interact to form an obligate complex in cells. Conclusion Stability and interaction data suggest that C15orf41 may be the key determinant of CDA-I and offer insight into the mechanism underlying this disease. Both proteins share a common pathway likely to be present in a wide variety of cell types; however, nucleolar enrichment may provide a clue as to the erythroid specific nature of CDA-I. The surprisingly high predicted incidence of CDA-I suggests that better ascertainment would lead to improved patient care.

  • Low upper limits on the O2 abundance from the Odin satellite

    • For the first time, a search has been conducted in our Galaxy for the 119 GHz transition connecting to the ground state of O2, using the Odin satellite. Equipped with a sensitive 3 mm receiver (Tsys(SSB) = 600 K), Odin has reached unprecedented upper limits on the abundance of O2, especially in cold dark clouds where the excited state levels involved in the 487 GHz transition are not expected to be significantly populated. Here we report upper limits for a dozen sources. In cold dark clouds we improve upon the published SWAS upper limits by more than an order of magnitude, reaching N(O2)/N(H2) <= 10-7 in half of the sources. While standard chemical models are definitively ruled out by these new limits, our results are compatible with several recent studies that derive lower O2 abundances. Goldsmith et al. (\cite{SWAS2002}) recently reported a SWAS tentative detection of the 487 GHz transition of O2 in an outflow wing towards rho Oph A in a combination of 7 beams covering approximately 10arcmin x 14arcmin . In a brief (1.3 hour integration time) and partial covering of the SWAS region (~65% if we exclude their central position), we did not detect the corresponding 119 GHz line. Our 3 sigma upper limit on the O2 column density is 7.3x 1015 cm-2. We presently cannot exclude the possibility that the SWAS signal lies mostly outside of the 9\arcmin Odin beam and has escaped our sensitive detector. Based on observations with Odin, a Swedish-led satellite project funded jointly by the Swedish National Space Board (SNSB), the Canadian Space Agency (CSA), the National Technology Agency of Finland (Tekes) and Centre National d'Études Spatiales (CNES). The Swedish Space Corporation was the industrial prime contractor and is operating Odin.

    • New drill cores from the largest known impact structure in Europe, the relict of the Siljan meteorite crater, provide new possibilities to reconstruct Early Palaeozoic marine environments and ecosystems, and to document changes in sedimentary facies, sea level and palaeoclimate in Baltoscandia. The impact crater is an important target of the project "Concentric Impact Structures in the Palaeozoic" within the framework of the "Swedish Deep Drilling Program". Two core sections, Mora 001 and Solberga 1, have been analysed. The sedimentary successions of these core sections include strata of late Tremadocian through late Wenlock ages. Our preliminary studies show not only that several of the classical Palaeozoic units of Sweden are represented in the area, but also that other significantly different facies are preserved in the Siljan district. An erosional unconformity representing a substantial hiatus occurs between Middle Ordovician limestone and a Llandovery-Wenlock (Silurian) shale succession in the western part of the Siljan structure and suggests an extended period of uplift and erosion. This may be related to forebulge migration due to flexural loading by the Caledonian thrust sheet to the west. Thus, this part of Sweden, previously regarded as a stable cratonic area, presumably was affected by the Caledonian collision between Baltica and Laurentia.

    • The apparition of bright comets C/2012 F6 (Lemmon) and C/2014 Q2 (Lovejoy) in March-April 2013 and January 2015, combined with the improved observational capabilities of submillimeter facilities, offered an opportunity to carry out sensitive compositional and isotopic studies of the volatiles in their coma. We observed comet Lovejoy with the IRAM 30 m telescope between 13 and 26 January 2015, and with the Odin submillimeter space observatory on 29 January-3 February 2015. We detected 22 molecules and several isotopologues. The (H2O)-O-16 and (H2O)-O-18 production rates measured with Odin follow a periodic pattern with a period of 0.94 days and an amplitude of similar to 25%. The inferred isotope ratios in comet Lovejoy are O-16/O-18 = 499 +/- 24 and D/H = 1.4 +/- 0.4 x 10(-4) in water, S-32/S-34 = 24.7 +/- 3.5 in CS, all compatible with terrestrial values. The ratio C-12/C-13 = 109 +/- 14 in HCN is marginally higher than terrestrial and N-14/N-15 = 145 +/- 12 in HCN is half the Earth ratio. Several upper limits for D/H or C-12/C-13 in other molecules are reported. From our observation of HDO in comet C/2014 Q2 (Lovejoy), we report the first D/H ratio in an Oort Cloud comet that is not larger than the terrestrial value. On the other hand, the observation of the same HDO line in the other Oort-cloud comet, C/2012 F6 (Lemmon), suggests a D/H value four times higher. Given the previous measurements of D/H in cometary water, this illustrates that a diversity in the D/H ratio and in the chemical composition, is present even within the same dynamical group of comets, suggesting that current dynamical groups contain comets formed at very different places or times in the early solar system.

    • Comet 9P/Tempel 1 was the target of a multi-wavelength worldwide investigation in 2005. The NASA Deep Impact mission reached the comet on 4.24 July 2005, delivering a 370-kg impactor which hit the comet at 10.3 km s -1 . Following this impact, a cloud of gas and dust was excavated from the comet nucleus. The comet was observed in 2005 prior to and after the impact, at 18-cm wavelength with the Nançay radio telescope, in the millimeter range with the IRAM and CSO radio telescopes, and at 557 GHz with the Odin satellite. OH observations at Nançay provided a 4-month monitoring of the outgassing of the comet from March to June, followed by the observation of H 2 O with Odin from June to August 2005. The peak of outgassing was found to be around 1 × 10 28   molec. s -1 between May and July. Observations conducted with the IRAM 30-m radio telescope in May and July 2005 resulted in detections of HCN, CH 3 OH and H 2 S with classical abundances relative to water (0.12, 2.7 and 0.5%, respectively). In addition, a variation of the HCN production rate with a period of 1.73 ± 0.10 days was observed in May 2005, consistent with the 1.7-day rotation period of the nucleus. The phase of these variations, as well as those of CN seen in July by Jehin et al. [Jehin, E., Manfroid, J., Hutsemékers, D., Cochran, A.L., Arpigny, C., Jackson, W.M., Rauer, H., Schulz, R., Zucconi, J.-M., 2006. Astrophys. J. 641, L145-L148], is consistent with a rotation period of the nucleus of 1.715 days and a strong variation of the outgassing activity by a factor 3 from minimum to maximum. This also implies that the impact took place on the rising phase of the "natural" outgassing which reached its maximum ≈4 h after the impact. Post-impact observations at IRAM and CSO did not reveal a significant change of the outgassing rates and relative abundances, with the exception of CH 3 OH which may have been more abundant by up to one order of magnitude in the ejecta. Most other variations are linked to the intrinsic variability of the comet. The Odin satellite monitored nearly continuously the H 2 O line at 557 GHz during the 38 h following the impact on the 4th of July, in addition to weekly monitoring. Once the periodic variations related to the nucleus rotation are removed, a small increase of outgassing related to the impact is present, which corresponds to the release of ≈ 5000 ± 2000 tons of water.     • In 2005, as part of a world-wide multi-wavelength investigation of comet 9P/Tempel 1 in support to the Deep Impact mission, we conducted radio spectroscopic observations with the Nançay radio telescope, the Odin satellite, the CSO 10-m and the IRAM 30-m telescopes. We report here our results concerning the chemical composition of the comet. The relative abundances of the detected species (H2O, CH3OH, H2S, HCN) or their upper limits (CO, H2CO, CS) are comparable to the mean values observed in other comets. No significant changes of the outgassing rates (except possibly for CH3OH) or of the molecular abundances were observed following the impact.

    • We show, in the context of single-photon detection, that an atomic three-level model for a transmon in a transmission line does not support the predictions of the nonlinear polarizability model known as the cross-Kerr effect. We show that the induced displacement of a probe in the presence or absence of a single photon in the signal field, cannot be resolved above the quantum noise in the probe. This strongly suggests that cross-Kerr media are not suitable for photon counting or related single-photon applications. Our results are presented in the context of a transmon in a one-dimensional microwave waveguide, but the conclusions also apply to optical systems.

    • Aims/hypothesis: In type 1 diabetes (T1D), most insulin-producing beta cells are destroyed, but the trigger is unknown. One of the possible triggers is a virus infection and the aim of this study was to test if enterovirus infection affects glucose stimulated insulin secretion and the effect of virus replication on cellular macromolecules and organelles involved in insulin secretion. Methods: Isolated human islets were infected with different strains of coxsackievirus B (CVB) virus and the glucose-stimulated insulin release (GSIS) was measured in a dynamic perifusion system. Classical morphological electron microscopy, large-scale electron microscopy, so-called nanotomy, and immunohistochemistry were used to study to what extent virus-infected beta cells contained insulin, and real-time PCR was used to analyze virus induced changes of islet specific genes. Results: In islets infected with CVB, GSIS was reduced in correlation with the degree of virus-induced islet disintegration. The expression of the gene encoding insulin was decreased in infected islets, whereas the expression of glucagon was not affected. Also, in islets that were somewhat disintegrated, there were uninfected beta cells. Ultrastructural analysis revealed that virus particles and virus replication complexes were only present in beta cells. There was a significant number of insulin granules remaining in the virus-infected beta cells, despite decreased expression of insulin mRNA. In addition, no typical Golgi apparatus was detected in these cells. Exposure of islets to synthetic dsRNA potentiated glucose-stimulated insulin secretion. Conclusions/interpretation: Glucose-stimulated insulin secretion; organelles involved in insulin secretion and gene expression were all affected by CVB replication in beta cells.

    • Odin has successfully observed three regions in the Orion A cloud, i.e. Ori KL, Ori S and the Orion Bar, in the 572.5 GHz rotational ground state line of ammonia, ortho-NH3 (J,K) = (1,0) -> (0,0), and the result for the Orion Bar represents the first detection in an ammonia line. Several velocity components are present in the data. Specifically, the observed line profile from the Orion Bar can be decomposed into two components, which are in agreement with observations in high-J CO lines by Wilson et al. (\cite{wilson01}). Using the source model for the Orion Bar by these authors, our Odin observation implies a total ammonia abundance of NH3/H2 = 5x 10-9.     • Odin has successfully observed the molecular core rho Oph A in the 572.5 GHz rotational ground state line of ammonia, NH3 (JK = 10 -> 00). The interpretation of this result makes use of complementary molecular line data obtained from the ground (C17O and CH3OH) as part of the Odin preparatory work. Comparison of these observations with theoretical model calculations of line excitation and transfer yields a quite ordinary abundance of methanol, X(CH3OH)= 3 x 10-9. Unless NH3 is not entirely segregated from C17O and CH3OH, ammonia is found to be significantly underabundant with respect to typical dense core values, viz. X(NH3) = 8 x 10-10.     • New results from water mapping observations of the Orion KL region using the submm/mm wave satellite Odin (2.1\arcmin beam size at 557 GHz), are presented. The ortho-H2O \jkktrans{1}{1}{0}{1}{0}{1} ground state transition was observed in a 7arcminx 7arcmin rectangular grid with a spacing of 1\arcmin, while the same line of H218O was measured in two positions, Orion KL itself and 2\arcmin south of Orion KL. In the main water species, the KL molecular outflow is largely resolved from the ambient cloud and it is found to have an extension of 60\arcsec-110\arcsec. The H2O outflow profile exhibits a rather striking absorption-like asymmetry at the line centre. Self-absorption in the near (or ``blue'') part of the outflow (and possibly in foreground quiescent halo gas) is tentatively suggested to play a role here. We argue that the dominant part of the KL H218O outflow emission emanates from the compact (size ~ 15\arcsec) low-velocity flow and here estimate an H2O abundance of circa 10-5 compared to all H2 in the flow - an order of magnitude below earlier estimates of the H2O abundance in the shocked gas of the high-velocity flow. The narrow ambient cloud lines show weak velocity trends, both in the N-S and E-W directions. H218O is detected for the first time in the southern position at a level of ~ 0.15 K and we here estimate an H2O abundance of (1-8) x 10-8.     • The Malingen structure is an approximately 700m wide, rimmed, sediment-filled, circular depression in Precambrian crystalline basement approximately 16.2km from the concentric, marine-target Lockne crater (inner, basement crater diameter approximately 7.5km, total diameter in sedimentary strata approximately 13.5km). We present here results from geologic mapping, a 148.8m deep core drilling from the center of the structure, detailed biostratigraphic dating of the structure's formation and its age correlation with Lockne, chemostratigraphy of the sedimentary infill, and indication for shock metamorphism in quartz from breccias below the crater infill. The drill core reveals, from bottom to the top, approximately 33m of basement rocks with increased fracturing upward, approximately 10m of polymict crystalline breccia with shock features, approximately 97m of slumped Cambrian mudstone, approximately 4.7m of a normally graded, polymict sedimentary breccia that in its uppermost part grades into sandstone and siltstone (cf. resurge deposits), and approximately 1.6m of secular sediments. The combined data set shows that the Malingen structure formed in conjunction with the Lockne crater in the same marine setting. The shape and depth of the basement crater and the cored sequence of crystalline breccias with shocked quartz, slumped sediments, and resurge deposits support an impact origin. The stratigraphic and geographic relationship with Lockne suggests the Lockne and Malingen craters to be the first described doublet impact structure by a binary asteroid into a marine-target setting.

    • Aims/hypothesisEnteroviral infection has been implicated in the development of islet autoimmunity in type 1 diabetes and enteroviral antigen expression has been detected by immunohistochemistry in the pancreatic beta cells of patients with recent-onset type 1 diabetes. However, the immunohistochemical evidence relies heavily on the use of a monoclonal antibody, clone 5D8/1, raised against an enteroviral capsid protein, VP1. Recent data suggest that the clone 5D8/1 may also recognise non-viral antigens; in particular, a component of the mitochondrial ATP synthase (ATP5B) and an isoform of creatine kinase (CKB). Therefore, we evaluated the fidelity of immunolabelling by clone 5D8/1 in the islets of patients with type 1 diabetes.MethodsEnteroviral VP1, CKB and ATP5B expression were analysed by western blotting, RT-PCR and immunocytochemistry in a range of cultured cell lines, isolated human islets and human tissue.ResultsClone 5D8/1 labelled CKB, but not ATP5B, on western blots performed under denaturing conditions. In cultured human cell lines, isolated human islets and pancreas sections from patients with type 1 diabetes, the immunolabelling of ATP5B, CKB and VP1 by 5D8/1 was readily distinguishable. Moreover, in a human tissue microarray displaying more than 80 different cells and tissues, only two (stomach and colon; both of which are potential sites of enterovirus infection) were immunopositive when stained with clone 5D8/1.Conclusions/interpretationWhen used under carefully optimised conditions, the immunolabelling pattern detected in sections of human pancreas with clone 5D8/1 did not reflect cross-reactivity with either ATP5B or CKB. Rather, 5D8/1 is likely to be representative of enteroviral antigen expression.

    • The Odin satellite has been used to detect emission and absorption in the 557-GHz H216O line in the Galactic Centre towards the Sgr Astar Circumnuclear Disk (CND), and the Sgr A +20 km s-1 and +50 km s-1 molecular clouds. Strong broad H2O emission lines have been detected in all three objects. Narrow H2O absorption lines are present at all three positions and originate along the lines of sight in the 3-kpc Spiral Arm, the -30 km s-1 Spiral Arm and the Local Sgr Spiral Arm. Broad H2O absorption lines near -130 km s-1 are also observed, originating in the Expanding Molecular Ring. A new molecular feature (the ``High Positive Velocity Gas'' - HPVG) has been identified in the positive velocity range of ~+120 to +220 km s-1, seen definitely in absorption against the stronger dust continuum emission from the +20 km s-1 and +50 km s-1 clouds and possibly in emission towards the position of Sgr Astar CND. The 548-GHz H218O isotope line towards the CND is not detected at the 0.02 K (rms) level.     • Introduction Pediatric patients with Hodgkin lymphoma (HL) have several risk factors for venous thromboembolism (VTE). Although these patients are occasionally treated with thromboprophylaxis, no guidelines are implemented in Sweden. Scarce data from adult patients indicate an increased risk of VTE, but pediatric data is largely missing. Given the favorable overall survival of HL, there should reasonably be more focus on preventing complications. Materials and methods We conducted a retrospective cohort study, including all patients registered in the Childhood Cancer Registry under the age of 18 years diagnosed with HL between January 2005 and December 2015 in Sweden. Results Data was retrieved from the medical records of all 163 patients (100%) at six Swedish pediatric cancer centers. The incidence of VTE was 7.7% (symptomatic VTE 3.9%). The median follow-up was 3.4 years (range 0.3–10.5). Only five patients (3.1%) were treated with thromboprophylaxis. All VTE events occurred in the older age category (11–17 years) and all but one (92.7%) had a mediastinal mass. While the VTE did not significantly affect the treatment of HL, it caused increased morbidity and 2/12 developed a post-thrombotic syndrome. No significant risk factors for VTE were identified. Conclusions VTE is a relatively common complication of HL and its treatment, causing increased acute and long-term morbidity. However, due to limited number of events we could not demonstrate risk-factors for VTE that would identify patients who might benefit from thromboprophylaxis.

  • Odin/SMR limb observations of stratospheric trace gases: Validation of N2O

    • The Sub-Millimetre Radiometer (Odin/SMR) on board the Odin satellite, launched on 20 February 2001, performs regular measurements of the global distribution of stratospheric nitrous oxide (N2O) using spectral observations of the J = 20&RARR; 19 rotational transition centered at 502.296 GHz. We present a quality assessment for the retrieved N2O profiles (level 2 product) by comparison with independent balloonborne and aircraftborne validation measurements as well as by cross-comparing with preliminary results from other satellite instruments. An agreement with the airborne validation experiments within 28 ppbv in terms of the root mean square (RMS) deviation is found for all SMR data versions (v222, v223, and v1.2) under investigation. More precisely, the agreement is within 19 ppbv for N2O volume mixing ratios (VMR) lower than 200 ppbv and within 10% for mixing ratios larger than 150 ppbv. Given the uncertainties due to atmospheric variability inherent to such comparisons, these values should be interpreted as upper limits for the systematic error of the Odin/SMR N2O measurements. Odin/SMR N2O mixing ratios are systematically slightly higher than nonvalidated data obtained from the Improved Limb Atmospheric Spectrometer-II (ILAS-II) on board the Advanced Earth Observing Satellite-II (ADEOS-II). Root mean square deviations are generally within 23 ppbv (or 20% for VMR-N2O > 100 ppbv) for versions 222 and 223. The comparison with data obtained from the Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) on the Envisat satellite yields a good agreement within 9-17 ppbv (or 10% for VMR-N2O > 100 ppbv) for the same data versions. Odin/SMR version 1.2 data show somewhat larger RMS deviations and a higher positive bias.

The boron content was found to dominate the mechanical properties of the TFMGs. The similar to 10% smaller strength of Fe-Nb-B TFMGs compared to existing bulk metallic glass with similar composition may be attributed to the absence of a network-like structure based on (Fe,M)(23)B-6 phase due to the extreme quenching conditions employed.

    • Magnetic droplets are a type of non-topological magnetic soliton, which are stabilised and sustained by spin-transfer torques for instance. Without this, they would collapse. Here Ahlberg et al show that by decreasing the applied magnetic field, droplets can be frozen, forming a static nanobubble Magnetic droplets are non-topological magnetodynamical solitons displaying a wide range of complex dynamic phenomena with potential for microwave signal generation. Bubbles, on the other hand, are internally static cylindrical magnetic domains, stabilized by external fields and magnetostatic interactions. In its original theory, the droplet was described as an imminently collapsing bubble stabilized by spin transfer torque and, in its zero-frequency limit, as equivalent to a bubble. Without nanoscale lateral confinement, pinning, or an external applied field, such a nanobubble is unstable, and should collapse. Here, we show that we can freeze dynamic droplets into static nanobubbles by decreasing the magnetic field. While the bubble has virtually the same resistance as the droplet, all signs of low-frequency microwave noise disappear. The transition is fully reversible and the bubble can be thawed back into a droplet if the magnetic field is increased under current. Whereas the droplet collapses without a sustaining current, the bubble is highly stable and remains intact for days without external drive. Electrical measurements are complemented by direct observation using scanning transmission x-ray microscopy, which corroborates the analysis and confirms that the bubble is stabilized by pinning.

    • Purpose:Extracellular matrix metalloproteinase inducer (EMMPRIN) has been shown to stimulate fibroblasts to production of matrix metalloproteinases (MMPs). MMPs comprise a family of proteolytic enzymes implicated in the degradation of extracellular matrix which has been proposed to be one of the essential steps in tumor invasion and metastases. In the present study we investigated the expression and location of mRNAs forEMMPRIN, matrix metalloproteinase-2 (MMP-2), and membrane-type 1 matrix metalloproteinase (MT1-MMP) in mesenchymal tumors with different tendencies to recur or metastasize.Subjects:Eight malignant fibrous histiocytomas (MFH), seven aggressive fibromatosis (AF), and six benign fibrous tumors (BF).Method:The mRNA-expression ofEMMPRIN,MMP-2andMT1-MMPwere studied using mRNAin situhybridization technique.Results:The mRNA-expression ofEMMPRIN,MMP-2andMT1-MMPrespectively were found at varying frequency and level in all tumor types. The mRNAs corresponding toEMMPRINandMMP-2were seen in neoplastic cells as well as in endothelial cells both inside and outside the tumor pseudo-capsule, whereasMT1-MMPwas seen only within the tumors. The estimated mRNA levels ofEMMPRINandMMP-2covariated significantly. Overall, the highest expression was found in the MFH tumors and the lowest levels in the BF tumors.Discussion:These findings suggest that the MMP-inducerEMMPRINand the extracellular matrix degrading system involving the metalloproteinasesMMP-2andMT1-MMPis frequently activated in mesenchymal tumors. The covariation betweenEMMPRINandMMP-2support previous findings that EMMPRIN may be an inducer of MMP-2. The high levels ofMMP-2mRNA in MFH indicate a relationship between the proteolytic activity ofMMP-2and the tumor aggressiveness.

    • Type 1 diabetes mellitus is believed to be triggered, in part, by one or more environmental factors and human enteroviruses (HEVs) are among the candidates. Therefore, this study has examined whether two strains of HEV may differentially affect the induction of genes involved in pathways leading to the synthesis of islet hormones, chemokines and cytokines in isolated, highly purified, human islets. Isolated, purified human pancreatic islets were infected with strains of Coxsackievirus B1. Viral replication and the degree of CPE/islet dissociation were monitored. The expression of insulin, glucagon, CXCL10, TLR3, IF1H1, CCL5, OAS-1, IFN beta, and DDX58 was analyzed. Both strains replicated in islets but only one of strain caused rapid islet dissociation/CPE. Expression of the insulin gene was reduced during infection of islets with either viral strain but the gene encoding glucagon was unaffected. All genes analyzed which are involved in viral sensing and the development of innate immunity were induced by Coxsackie B viruses, with the notable exception of TLR3. There was no qualitative difference in the expression pattern between each strain but the magnitude of the response varied between donors. The lack of virus induced expression of TLR3, together with the differential regulation of IF1H1, OAS1 and IFN beta, (each of which has polymorphic variants influence the predisposition to type 1 diabetes), that might result in defective clearance of virus from islet cells. The reduced expression of the insulin gene and the unaffected expression of the gene encoding glucagon by Coxsackie B1 infection is consistent with the preferential beta-cell tropism of the virus.

    • Heart failure (HF) is a major cause of morbidity and mortality worldwide, highlighting an urgent need for novel treatment options, despite recent improvements. Aberrant Ca2+ handling is a key feature of HF pathophysiology. Restoring the Ca2+ regulating machinery is an attractive therapeutic strategy supported by genetic and pharmacological proof of concept studies. Here, we study antisense oligonucleotides (ASOs) as a therapeutic modality, interfering with the PLN/SERCA2a interaction by targeting Pln mRNA for downregulation in the heart of murine HF models. Mice harboring the PLN R14del pathogenic variant recapitulate the human dilated cardiomyopathy (DCM) phenotype; subcutaneous administration of PLN-ASO prevents PLN protein aggregation, cardiac dysfunction, and leads to a 3-fold increase in survival rate. In another genetic DCM mouse model, unrelated to PLN (Cspr3/Mlp−/−), PLN-ASO also reverses the HF phenotype. Finally, in rats with myocardial infarction, PLN-ASO treatment prevents progression of left ventricular dilatation and improves left ventricular contractility. Thus, our data establish that antisense inhibition of PLN is an effective strategy in preclinical models of genetic cardiomyopathy as well as ischemia driven HF.

    • Context: Comet C/2001 Q4 (NEAT) was extensively studied with the 1.1-m submillimetre telescope of the Odin satellite. The H2O line at 557 GHz was regularly observed from 6 March to 16 May 2004 and nearly continuously monitored during 3 periods between 26 April and 2 May 2004. Aims: This last set of data shows periodic variations in the line intensity, and we looked for characterising the long- and short-term behaviour of this comet. Methods: We used the variance ratio method and χ2 minimization to find the period of variation in the water production rate and simulations to infer its amplitude at the nucleus surface. Results: A 40% periodic variation in the water production rate is measured with a period of 0.816±0.004 day (19.58±0.1 h). The comet also exhibits a seasonal effect with a mean peak of outgassing around 2.7×1029 molec. s-1 taking place about 18 days before perihelion.

    • A new thermodynamic database has been combined with an existing kinetic database to perform coarsening simulations in ternary systems including MC and M7C3 carbides in an fee matrix. The kinetic database was revised taking into consideration the new experimental information on the Fe-Cr-V-C system obtained in the present work, and available experiments on the ternary Fe-Cr-C and Fe-V-C systems. After revision the agreement between experimental results and simulations was satisfactory. It was found that the interfacial energy of M7C3 was twice as large as that of the MC carbide. The calculations for commercial steels with 6 alloy elements gave results in satisfactory agreement with new experimental measurements. The present coarsening simulations use the calculated equilibrium state and the observed particle sizes as the state for the start of the simulations. All the simulations were performed with the DICTRA software. © 2008 Institute of Materials, Minerals and Mining.
