  • First genome-wide association study of esophageal atresia identifies three genetic risk loci at CTNNA3, FOXF1/FOXC2/FOXL1, and HNF1B

    • Esophageal atresia with or without tracheoesophageal fistula (EA/TEF) is the most common congenital malformation of the upper digestive tract. This study represents the first genome-wide association study (GWAS) to identify risk loci for EA/TEF. We used a European case-control sample comprising 764 EA/TEF patients and 5,778 controls and observed genome-wide significant associations at three loci. On chromosome 10q21 within the gene CTNNA3 (p = 2.11 × 10−8; odds ratio [OR] = 3.94; 95% confidence interval [CI], 3.10–5.00), on chromosome 16q24 next to the FOX gene cluster (p = 2.25 × 10−10; OR = 1.47; 95% CI, 1.38–1.55) and on chromosome 17q12 next to the gene HNF1B (p = 3.35 × 10−16; OR = 1.75; 95% CI, 1.64–1.87). We next carried out an esophageal/tracheal transcriptome profiling in rat embryos at four selected embryonic time points. Based on these data and on already published data, the implicated genes at all three GWAS loci are promising candidates for EA/TEF development. We also analyzed the genetic EA/TEF architecture beyond the single marker level, which revealed an estimated single-nucleotide polymorphism (SNP)-based heritability of around 37% ± 14% standard deviation. In addition, we examined the polygenicity of EA/TEF and found that EA/TEF is less polygenic than other complex genetic diseases. In conclusion, the results of our study contribute to a better understanding on the underlying genetic architecture of ET/TEF with the identification of three risk loci and candidate genes. © 2022 The Authors

  • Interactions between a 30% charged polyelectrolyte and an anionic surfactant in bulk and at a solid-liquid interface

    • The association between a 30% charged cationic polyelectrolyte and an anionic surfactant, sodium dodecyl sulfate (SDS), in 10 mM 1:1 electrolyte was investigated using surface force measurements and dynamic light scattering. The polyelectrolyte employed was a random copolymer of the neutral acrylamide and cationic [3-(2-methylpropionamide)propyl]trimethylammonium chloride (AM-MAPTAC-31). Light scattering measurements show that upon progressive addition of SDS to an AM-MAPTAC-31 solution the single coil size decreases until precipitation occurs at an SDS/MAPTAC ratio of just above 0.4. At SDS/MAPTAC ratios at or above 2, redispersion of the aggregates takes place. The interfacial behavior of AM-MAPTAC-31/SDS complexes was investigated in two ways. In one set of experiments a droplet containing a mixture of SDS and AM-MAPTAC-31 was placed between the surfaces and adsorption was allowed to occur from the aqueous mixture. It was found that the range of the steric force decreased when the SDS/MAPTAC ratio was increased from 0 to 0.4, indicating adsorption in a less extended conformation due to a decreased repulsion between the polyelectrolyte segments. At a ratio of 0.6 a compact interfacial complex was formed and the measured force was attractive over a small distance regime. A further increase in SDS/MAPTAC ratio resulted in precipitation of large aggregates at the surface, and reproducible force data could not be obtained. At an even higher SDS/AM-MAPTAC ratio of 4, individual aggregates were once again adsorbed at the surface. Hence, we find a good correspondence between association in bulk and at the solid surface. In another set of experiments the polyelectrolyte was first preadsorbed to mica surfaces and then SDS was added to the polyelectrolyte-free solution surrounding the surfaces. In this way precipitation of large SDS-polyelectrolyte aggregates onto the surfaces was avoided. Addition of SDS up to a concentration of 0.1 mM hardly affected the long-range interaction but gave an increased compressed layer thickness. A further increase in SDS concentrations to 1 mM results in a dramatic increase in the range of the force, suggesting formation of strongly negatively charged polyelectrolyte-surfactant complexes.

    • The effects of complex formation between sodium dodecylsulfate (SDS) and the positively charged (3-(2-methylpropionamide)propyl)trimethyl-ammonium chloride-acrylamide (MAPTAC-AM) copolymer have been studied in dilute and semidilute aqueous solution in the presence of 10 mM NaCl. Two different charge densities of the copolymer have been used in the study: 0.31 and 0.66, corresponding to the proportion of MAPTAC units. Dynamic light scattering (DLS) and rheometry (static low-shear and capillary viscometry) have been performed on the systems at different charge ratios, i.e., SDS/MAPTAC molar ratios, r. Regarding the phase behavior, the maximum binding ratio prior to precipitation differs between the copolymers. A 1.0% w/v solution of SDS/31% MAPTAC-AM is soluble at r = 0.4 , while an SDS/66% MAPTAC-AM solution of 1.0% w/v shows phase separation at this ratio. With excess surfactant, the complex in the former system is resolubilized at r=2.0, whereas the latter system is still phase-separated at r=5.0. DLS results show that, for both copolymers, the hydrodynamic radius, Rh, of the single-chain copolymer-surfactant complex decreases as a function of r, but then increases slightly prior to phase separation. The corresponding hydrodynamic virial coefficient, kD, changes in the same manner as Rh. The light-scattering data also show that the formation of larger structures is promoted as the polymer concentration is increased from 0.2 to 1.0% w/v. This is shown by the increase in the relative aggregate-to-single coil peak areas in the relaxation time distributions. Both systems have this common trend. The results from rheological measurements support the results from DLS. A reduction in intrinsic viscosity, [], is observed on increasing r up to phase separation. The major part of the static low-shear measurements showed Newtonian behavior for both systems at different copolymer concentrations (27.6-138 mM), and at different r. These systems, partially ionic polymer/oppositely charged surfactant, present very interesting rheological behavior at relatively high polymer concentrations and at low r values. Their behavior is similar to those of hydrophobically modified polyelectrolytes.

  • The Genetic Architecture of Obsessive-Compulsive Disorder: Contribution of Liability to OCD From Alleles Across the Frequency Spectrum.

    • Obsessive-compulsive disorder (OCD) is known to be substantially heritable; however, the contribution of genetic variation across the allele frequency spectrum to this heritability remains uncertain. The authors used two new homogeneous cohorts to estimate the heritability of OCD from inherited genetic variation and contrasted the results with those of previous studies.The sample consisted of 2,090 Swedish-born individuals diagnosed with OCD and 4,567 control subjects, all genotyped for common genetic variants, specifically >400,000 single-nucleotide polymorphisms (SNPs) with minor allele frequency (MAF) ≥0.01. Using genotypes of these SNPs to estimate distant familial relationships among individuals, the authors estimated the heritability of OCD, both overall and partitioned according to MAF bins.Narrow-sense heritability of OCD was estimated at 29% (SE=4%). The estimate was robust, varying only modestly under different models. Contrary to an earlier study, however, SNPs with MAF between 0.01 and 0.05 accounted for 10% of heritability, and estimated heritability per MAF bin roughly followed expectations based on a simple model for SNP-based heritability.These results indicate that common inherited risk variation (MAF ≥0.01) accounts for most of the heritable variation in OCD. SNPs with low MAF contribute meaningfully to the heritability of OCD, and the results are consistent with expectation under the "infinitesimal model" (also referred to as the "polygenic model"), where risk is influenced by a large number of loci across the genome and across MAF bins.
