  • Spectroscopy along flerovium decay chains: Discovery of 280Ds and an excited state in 282Cn

    • A nuclear spectroscopy experiment was conducted to study α-decay chains stemming from isotopes of flerovium (element Z=114). An upgraded TASISpec decay station was placed behind the gas-filled separator TASCA at the GSI Helmholtzzentrum für Schwerionenforschung in Darmstadt, Germany. The fusion-evaporation reactions 48Ca+242Pu and 48Ca+244Pu provided a total of 32 flerovium-candidate decay chains, of which two and eleven were firmly assigned to 286Fl and 288Fl, respectively. A prompt coincidence between a 9.60(1)-MeV α-particle event and a 0.36(1)-MeV conversion electron marked the first observation of an excited state in an even-even isotope of the heaviest man-made elements, namely 282Cn. Spectroscopy of 288Fl decay chains fixed Qα=10.06(1) MeV. In one case, a Qα=9.46(1)-MeV decay from 284Cn into 280Ds was observed, with 280Ds fissioning after only 518 μs. The impact of these findings, aggregated with existing data on decay chains of 286,288Fl, on the size of an anticipated shell gap at proton number Z=114 is discussed in light of predictions from two beyond-mean-field calculations, which take into account triaxial deformation.

  • Review on geophysical monitoring of CO2 injection at Ketzin, Germany

    • Geophysical monitoring activities were an important component of the CO2 injection program at the Ketzin site, Germany. Here we report on the seismic and electrical resistivity tomography (ERT) measurements performed during the period of the site development and CO2 injection. Details on the site geology, the history of the CO2 injection operation, and petrophysical models relevant for the interpretation of the geophysical data are presented. The seismic measurements comprise 2D and 3D surface seismic surveys, vertical seismic profilings, and crosshole measurements. Apart from the measurements, results from advanced processing methods, such as impedance inversion and full-waveform inversion are also presented. In addition, results from crosshole ERT and surface-downhole ERT are presented. If operational efforts are taken into consideration we conclude that a combination of several geophysical methods is preferable given the demands of a spatiotemporally comprehensive monitoring program. We base this conclusion on that the different imaging characteristics and petrophysical sensitivities of different methods can complement each other. An important finding is, based on signal quality and reduced operational costs, that the use of permanent installations is promising. Generally, specific monitoring layouts will depend on site-specific characteristics, such as reservoir depth, availability of wells, petrophysical characteristics, and accessibility of surface locations. Given the comprehensive range of methods applied, the reported results are not only relevant to the operation of CO2 storage sites, but are also of interest to other monitoring projects dealing with fluid injection or production.

  • Environmental refuges from disease in host–parasite interactions under global change

    • The physiological performance of organisms depends on their environmental context, resulting in performance–response curves along environmental gradients. Parasite performance–response curves are generally expected to be broader than those of their hosts due to shorter generation times and hence faster adaptation. However, certain environmental conditions may limit parasite performance more than that of the host, thereby providing an environmental refuge from disease. Thermal disease refuges have been extensively studied in response to climate warming, but other environmental factors may also provide environmental disease refuges which, in turn, respond to global change. Here, we (1) showcase laboratory and natural examples of refuges from parasites along various environmental gradients, and (2) provide hypotheses on how global environmental change may affect these refuges. We strive to synthesize knowledge on potential environmental disease refuges along different environmental gradients including salinity and nutrients, in both natural and food-production systems. Although scaling up from single host–parasite relationships along one environmental gradient to their interaction outcome in the full complexity of natural environments remains difficult, integrating host and parasite performance–response can serve to formulate testable hypotheses about the variability in parasitism outcomes and the occurrence of environmental disease refuges under current and future environmental conditions.

  • Preliminary Seismic Time-lapse Results from the First Post-injection Survey at the Ketzin Pilot Site

    • Since the CO2SINK project start in April 2004, different methods involving seismics, geoelectrics andpressure-temperature monitoring have been applied at the Ketzin pilot site to map geological structuresand assess CO2 distribution and movement. One 3D baseline seismic survey prior to CO2 injection andtwo 3D repeat seismic surveys during CO2 injection have proven that the 3D time-lapse surface seismicmethod is an effective tool for providing good quality images of the CO2 plume. In the autumn of 2015, athird 3D repeat seismic survey, serving as the first post-injection survey, was acquired. The preliminaryresults show that the predominant westward trend of the CO2 migration is in agreement with the previousresults. No CO2 migration upwards through the caprock was detected. The decrease in the size of CO2plume may be related to ongoing dissolution into the saline formation water.

    • The simulation models of wireless networks rapidly increase in complexity to accurately model wireless channel characteristics and the properties of advanced transmission technologies. Such detailed models typically lead to a high computational load per simulation event that accumulates to extensive simulation runtimes. Reducing runtimes through parallelization is challenging since it depends on detecting causally independent events that can execute concurrently. Most existing approaches base this detection on lookaheads derived from channel propagation latency or protocol characteristics. In wireless networks, these lookaheads are typically short, causing the potential for parallelization and the achievable speedup to remain small. This paper presents Horizon, which unlocks a substantial portion of a simulation model's workload for parallelization by going beyond the traditional lookahead. We show how to augment discrete events with durations to identify a much larger horizon of independent simulation events and efficiently schedule them on multi-core systems. Our evaluation shows that this approach can significantly cut down the runtime of simulations, in particular for complex and accurate models of wireless networks.

  • 4D Seismic Monitoring at the Ketzin Pilot Site during Five Years of Storage – Results and Quantitative Assessment

    • The 3D seismic and surface-downhole time-lapse monitoring at the Ketzin pilot site for CO, storage has provided the data base for a quantitative estimation of the amount of CO2 detected by the seismic surveys and of the CO2 plume thickness. The monitoring results have been compared to reservoir simulations, considering noise and thickness threshold values and indicating that conformity between monitoring and simulation results can be achieved when a thickness detection threshold of between 1 and 7 m is taken into account.

    • Ketzin is an European pilot site for onshore CO2 storage in a saline aquifer. Injection started in summer 2008 and to date, more than 37.000 tons of CO2 have been injected into the Stuttgart Formation at approximately 650 m depth. A wide range of monitoring methods are being applied at Ketzin, among which are active seismic observations at various scales. 3D reflection seismic, combined surface-downhole measurements and crosshole tomographic surveys were performed before injection and after the start of injection in order to image the reservoir and to track the CO2. Time-lapse signatures of the injected CO2 were observed by all active seismic methods. The CO2 could be detected by increased reflectivity at the top of the injection reservoir, by a change in the attenuation behaviour and also by reduced propagation velocity within the reservoir. The ongoing injection of CO2 during the next years will be followed by further repeat surveys. Current investigations focus on the still difficult problem of the quantification of the CO2 imaged by the geophysical measurements.

  • Staphylococcus carnosus : from starter culture to protein engineering platform

    • Since the 1950s, Staphylococcus carnosus is used as a starter culture for sausage fermentation where it contributes to food safety, flavor, and a controlled fermentation process. The long experience with S. carnosus has shown that it is a harmless and "food grade" species. This was confirmed by the genome sequence of S. carnosus TM300 that lacks genes involved in pathogenicity. Since the development of a cloning system in TM300, numerous genes have been cloned, expressed, and characterized and in particular, virulence genes that could be functionally validated in this non-pathogenic strain. A secretion system was developed for production and secretion of industrially important proteins and later modified to also enable display of heterologous proteins on the surface. The display system has been employed for various purposes, such as development of live bacterial delivery vehicles as well as microbial biocatalysts or bioadsorbents for potential environmental or biosensor applications. Recently, this surface display system has been utilized for display of peptide and protein libraries for profiling of protease substrates and for generation of various affinity proteins, e.g., Affibody molecules and scFv antibodies. In addition, by display of fragmented antigen-encoding genes, the surface expression system has been successfully used for epitope mapping of antibodies. Reviews on specific applications of S. carnosus have been published earlier, but here we provide a more extensive overview, covering a broad range of areas from food fermentation to sophisticated methods for protein-based drug discovery, which are all based on S. carnosus.

  • Global Impact of the COVID-19 Pandemic on Stroke Volumes and Cerebrovascular Events: A 1-Year Follow-up.

    • Declines in stroke admission, IV thrombolysis (IVT), and mechanical thrombectomy volumes were reported during the first wave of the COVID-19 pandemic. There is a paucity of data on the longer-term effect of the pandemic on stroke volumes over the course of a year and through the second wave of the pandemic. We sought to measure the effect of the COVID-19 pandemic on the volumes of stroke admissions, intracranial hemorrhage (ICH), IVT, and mechanical thrombectomy over a 1-year period at the onset of the pandemic (March 1, 2020, to February 28, 2021) compared with the immediately preceding year (March 1, 2019, to February 29, 2020).We conducted a longitudinal retrospective study across 6 continents, 56 countries, and 275 stroke centers. We collected volume data for COVID-19 admissions and 4 stroke metrics: ischemic stroke admissions, ICH admissions, IVT treatments, and mechanical thrombectomy procedures. Diagnoses were identified by their ICD-10 codes or classifications in stroke databases.There were 148,895 stroke admissions in the 1 year immediately before compared with 138,453 admissions during the 1-year pandemic, representing a 7% decline (95% CI [95% CI 7.1-6.9]; p < 0.0001). ICH volumes declined from 29,585 to 28,156 (4.8% [5.1-4.6]; p < 0.0001) and IVT volume from 24,584 to 23,077 (6.1% [6.4-5.8]; p < 0.0001). Larger declines were observed at high-volume compared with low-volume centers (all p < 0.0001). There was no significant change in mechanical thrombectomy volumes (0.7% [0.6-0.9]; p = 0.49). Stroke was diagnosed in 1.3% [1.31-1.38] of 406,792 COVID-19 hospitalizations. SARS-CoV-2 infection was present in 2.9% ([2.82-2.97], 5,656/195,539) of all stroke hospitalizations.There was a global decline and shift to lower-volume centers of stroke admission volumes, ICH volumes, and IVT volumes during the 1st year of the COVID-19 pandemic compared with the prior year. Mechanical thrombectomy volumes were preserved. These results suggest preservation in the stroke care of higher severity of disease through the first pandemic year.This study is registered under NCT04934020.

  • CRISPR-Mediated Induction of Neuron-Enriched Mitochondrial Proteins Boosts Direct Glia-to-Neuron Conversion

    • Astrocyte-to-neuron conversion is a promising avenue for neuronal replacement therapy. Neurons are particularly dependent on mitochondrial function, but how well mitochondria adapt to the new fate is unknown. Here, we determined the comprehensive mitochondrial proteome of cortical astrocytes and neurons, identifying about 150 significantly enriched mitochondrial proteins for each cell type, including transporters, metabolic enzymes, and cell-type-specific antioxidants. Monitoring their transition during reprogramming revealed late and only partial adaptation to the neuronal identity. Early dCas9-mediated activation of genes encoding mitochondrial proteins significantly improved conversion efficiency, particularly for neuron-enriched but not astrocyte-enriched antioxidant proteins. For example, Sod1 not only improves the survival of the converted neurons but also elicits a faster conversion pace, indicating that mitochondrial proteins act as enablers and drivers in this process. Transcriptional engineering of mitochondrial proteins with other functions improved reprogramming as well, demonstrating a broader role of mitochondrial proteins during fate conversion.
