  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Cryptococcal Hsf3 controls intramitochondrial ROS homeostasis by regulating the respiratory process

    • Mitochondrial quality control prevents accumulation of intramitochondrial-derived reactive oxygen species (mtROS), thereby protecting cells against DNA damage, genome instability, and programmed cell death. However, underlying mechanisms are incompletely understood, particularly in fungal species. Here, we show that Cryptococcus neoformans heat shock factor 3 (CnHsf3) exhibits an atypical function in regulating mtROS independent of the unfolded protein response. CnHsf3 acts in nuclei and mitochondria, and nuclear- and mitochondrial-targeting signals are required for its organelle-specific functions. It represses the expression of genes involved in the tricarboxylic acid cycle while promoting expression of genes involved in electron transfer chain. In addition, CnHsf3 responds to multiple intramitochondrial stresses; this response is mediated by oxidation of the cysteine residue on its DNA binding domain, which enhances DNA binding. Our results reveal a function of HSF proteins in regulating mtROS homeostasis that is independent of the unfolded protein response.

  • Physicochemical characterization of a polysaccharide from Agrocybe aegirita and its anti-ageing activity

    • The aim of the present study is to characterize the structure of a novel natural polysaccharide from Agrocybe aegirita (AAPS) and evaluate its anti-aging activity. The MALLS and GC-MS analysis indicated that the AAPS with molecular weights of 1.81 x 10(4) Da was mainly composed by rhamnose, fucose, mannose, and glucose in a molar ratio of 2.90:10.25:3.70:38.27. The FT-IR and NMR analysis showed that the backbone of AAPS was alpha-L-Rhap-(1 -> 6)-beta-D-Glcp-(1 -> 2)-alpha-L-Fucp-(1 -> 6)-alpha-D-Glcp-(1 -> 5)-alpha-L-Araf-(1 -> 4)-beta-D-GlcpA-(1 -> 5)-alpha-L-Araf-(1 -> 6)-alpha-D-Manp-(1 -> 6)-alpha-D-Manp-(1 -> 2)-alpha-L-Fucp-(1 -> 6)-beta-D-Glap-(1 -> 2)-alpha-L-Rhap-(1 -> 6)-beta-D-Galp-(1 -> which linked with two side chains alpha-L-Fucp-(1 -> 6)-beta-D-Glcp-(1 -> 6)-beta-D-Manp-(1 -> and alpha-D-Xylp-(1 -> 2)-alpha-L-Fucp-(1 -> 5) -alpha-D-Araf-(1.6)-beta-D-Galp-(1 -> at O-H2 at H-4-arabinose and the terminal Galp residues, respectively. The MRC-5 cells induced by H2O2 were used to explore the anti-ageing effect and its underlying mechanism of AAPS. It showed a potent anti-ageing activity, representing by the increased cell viability and beta-Gal viability, prevented G1-phase cell-cycle arrest, and decreased mitochondrial membrane potential. The polysaccharides extracted from A. aegirita might be applied in functional food as anti-ageing ingredient.

    • Oligosaccharides from green algae Ulva lactuca (ULO) and Enteromorpha prolifera (EPO) were used for investigation of anti-ageing effects and the underlying mechanism in SAMP8 mice. The structural properties of ULO and EPO were analyzed by fourier-transform infrared spectroscopy, gas chromatography-mass spectrometry, and agarose gel electrophoresis. These oligosaccharides enhanced the glutathione, superoxide dismutase, catalase, and telomerase levels and total antioxidant capicity, and decreased the levels of malondialdehyde and advanced glycation end products. After ULO and EPO treatment, the levels of inflammatory factors, including IFN-gamma, TNF-alpha, and IL-6, decreased; the BDNF and ChAT levels increased; and hippocampal neurons were protected. Downregulation of the p53 and FOXO1 genes and upregulation of the Sirt1 gene indicated that ULO and EPO have potential therapeutic effects in the prevention of ageing in SAMP8 mice. By 16S rRNA gene high-throughput sequencing, the abundance of Desulfovibrio was discovered to be markedly different in mice treated with ULO and EPO. The abundances of Verrucomicrobiaceae, Odoribacteraceae, Mogibacteriaceae, Planococcaceae, and Coriobacteriaceae were positively correlated with age-related indicators. These results demonstrated that oligosaccharides from U. lactuca and E. prolifera are ideal candidate compounds that can be used in functional foods and pharmaceuticals to prevent ageing.

  • Analysis of Heritability and Shared Heritability Based on Genome-Wide Association Studies for 13 Cancer Types

    • Background: Studies of related individuals have consistently demonstrated notable familial aggregation of cancer. We aim to estimate the heritability and genetic correlation attributable to the additive effects of common single-nucleotide polymorphisms (SNPs) for cancer at 13 anatomical sites. Methods: Between 2007 and 2014, the US National Cancer Institute has generated data from genome-wide association studies (GWAS) for 49 492 cancer case patients and 34 131 control patients. We apply novel mixed model methodology (GCTA) to this GWAS data to estimate the heritability of individual cancers, as well as the proportion of heritability attributable to cigarette smoking in smoking-related cancers, and the genetic correlation between pairs of cancers. Results: GWAS heritability was statistically significant at nearly all sites, with the estimates of array-based heritability, h(l)(2), on the liability threshold (LT) scale ranging from 0.05 to 0.38. Estimating the combined heritability of multiple smoking characteristics, we calculate that at least 24% (95% confidence interval [CI] = 14% to 37%) and 7% (95% CI = 4% to 11%) of the heritability for lung and bladder cancer, respectively, can be attributed to genetic determinants of smoking. Most pairs of cancers studied did not show evidence of strong genetic correlation. We found only four pairs of cancers with marginally statistically significant correlations, specifically kidney and testes (rho = 0.73, SE = 0.28), diffuse large B-cell lymphoma (DLBCL) and pediatric osteosarcoma (rho = 0.53, SE = 0.21), DLBCL and chronic lymphocytic leukemia (CLL) (rho = 0.51, SE = 0.18), and bladder and lung (rho = 0.35, SE = 0.14). Correlation analysis also indicates that the genetic architecture of lung cancer differs between a smoking population of European ancestry and a nonsmoking Asian population, allowing for the possibility that the genetic etiology for the same disease can vary by population and environmental exposures. Conclusion: Our results provide important insights into the genetic architecture of cancers and suggest new avenues for investigation.

    • Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UN's Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

  • Spatial-temporal differentiation and influencing factors of carbon emission trajectory in Chinese cities - A case study of 247 prefecture-level cities

    • Cities, where human energy activities and greenhouse gas emissions are concentrated, contribute significantly to alleviating the impacts of global climate change. Utilizing the China Carbon Emissions Accounting Database (CEADs) to provide carbon dioxide emission inventories for urban areas in China at the prefecture level, this study closely examines the historical evolution trajectories of carbon emissions across 247 urban units from 2005 to 2019. The logarithmic cubic function model was employed to simulate these trajectories, evaluating urban emission peaks and classifying the different carbon emission trajectories. Further, the Geographical and Temporal Weighted Regression model was employed to explore spatiotemporal traits and essential variables that impact the variations in carbon emissions among four identified trajectory types. Our results showed that Chinese urban carbon emission trajectories can be classified into four categories: a) peaking emissions, b) fluctuating growth, c) continuous growth, and d) passive decline. Specifically, 43 cities, primarily in North China, proactively attained their emission peak post-2010, driven by the reduction in secondary industry and energy intensity. 90 cities, largely industrial hubs in the southeast coast and inland, reached an emission plateau around 2015, exhibiting fluctuating growth due to dependencies on secondary industries. 101 cities, predominantly located in western and central regions, demonstrated a clear upward trend in carbon emissions, propelled by rapid urbanization and heavy industry-oriented economic development. Lastly, 13 cities, typically in the northeastern and southwestern regions, experienced a passive decline in carbon emissions, attributable to resource depletion or economic downturns. It is evident that China's city-level carbon peaking has demonstrated some effectiveness, yet considerable progress is still required.

  • The Simons Observatory : science goals and forecasts

    • The Simons Observatory (SO) is a new cosmic microwave background experiment being built on Cerro Toco in Chile, due to begin observations in the early 2020s. We describe the scientific goals of the experiment, motivate the design, and forecast its performance. SO will measure the temperature and polarization anisotropy of the cosmic microwave background in six frequency bands centered at: 27, 39, 93, 145, 225 and 280 GHz. The initial con figuration of SO will have three small-aperture 0.5-m telescopes and one large-aperture 6-m telescope, with a total of 60,000 cryogenic bolometers. Our key science goals are to characterize the primordial perturbations, measure the number of relativistic species and the mass of neutrinos, test for deviations from a cosmological constant, improve our understanding of galaxy evolution, and constrain the duration of reionization. The small aperture telescopes will target the largest angular scales observable from Chile, mapping approximate to 10% of the sky to a white noise level of 2 mu K-arcmin in combined 93 and 145 GHz bands, to measure the primordial tensor-to-scalar ratio, r, at a target level of sigma(r) = 0.003. The large aperture telescope will map approximate to 40% of the sky at arcminute angular resolution to an expected white noise level of 6 mu K-arcmin in combined 93 and 145 GHz bands, overlapping with the majority of the Large Synoptic Survey Telescope sky region and partially with the Dark Energy Spectroscopic Instrument. With up to an order of magnitude lower polarization noise than maps from the Planck satellite, the high-resolution sky maps will constrain cosmological parameters derived from the damping tail, gravitational lensing of the microwave background, the primordial bispectrum, and the thermal and kinematic Sunyaev-Zel'dovich effects, and will aid in delensing the large-angle polarization signal to measure the tensor-to-scalar ratio. The survey will also provide a legacy catalog of 16,000 galaxy clusters and more than 20,000 extragalactic sources.

    • The antioxidant activity of Spirulina platensis polysaccharide (SPP) was investigated in Caenorhabditis elegans. Nuclearmagnetic resonance, Fourier-transform infrared, multi-angle laser light scattering, and GC-MS were used to identify the structural characteristics of SPP. It was composed of the -> 2)-alpha-L-Rhap-(1 ->, -> 4)-beta-D-Manp-(1 ->, -> 6)-beta-D-Glcp-(1 ->, -> 4)-beta-Xylp-(1 ->, -> 3)-beta-L-Araf-(1 ->, and -> 2)-beta-L-Fucp-(1 ->, respectively. The modulation of gut microbiota in C. elegans was determined using 16S rRNA gene high-throughput sequencing. The malondialdehyde (MDA) content was significantly decreased, while the total superoxide dismutase (T-SOD) and reactive oxygen species (ROS) levels were improved after SPP supplementation. The cellular mitochondrial content and apoptosis were significantly down-regulated. The obvious increased levels of the DAF-16 and SKN-1 mRNAs were observed in the SPP-treated group, while the levels of miR-48 and miR-51 were significantly reduced. Moreover, SPP administration significantly increased the abundance of Flavobacterium, Achromobacter, Empedobacter, Anaerolinea, and Pseudoalteromonas of the intestinal flora. Based on these results, S. platensis polysaccharides may be used as a functional food to ameliorate diseases related to oxidative stress.

    • The aim of this study is to construct a new model to measure the supply and demand non-equilibrium of transportation system, which is more accuracy and effect compared to the traditional model. Based on the non-equilibrium problem of supply greater or less than demand, which is common in current city agglomeration transportation system, the supply and demand non-equilibrium depth of city agglomeration transportation system is proposed. From the perspective of the connotation of supply and demand non-equilibrium depth of city agglomeration transportation system, the category is definitely divided, and then the corresponding functions of supply and demand of the city agglomeration transportation system are established. According to the interaction mechanism of the supply and demand of the city agglomeration transportation system, the non equilibrium depth in a narrow and general sense can be calculated respectively, also the total non-equilibrium depth is obtained, containing its three-dimensional structure and the fitting surface. In the case study, it is indicated that the research conclusion meets the actual requirements completely.

    • Genome-wide association studies (GWAS) have identified more than 200 common genetic variants independently associated with colorectal cancer (CRC) risk, but the causal variants and target genes are mostly unknown. We sought to fine-map all known CRC risk loci using GWAS data from 100,204 cases and 154,587 controls of East Asian and European ancestry. Our stepwise conditional analyses revealed 238 independent association signals of CRC risk, each with a set of credible causal variants (CCVs), of which 28 signals had a single CCV. Our cis-eQTL/mQTL and colocalization analyses using colorectal tissue-specific transcriptome and methylome data separately from 1299 and 321 individuals, along with functional genomic investigation, uncovered 136 putative CRC susceptibility genes, including 56 genes not previously reported. Analyses of single-cell RNA-seq data from colorectal tissues revealed 17 putative CRC susceptibility genes with distinct expression patterns in specific cell types. Analyses of whole exome sequencing data provided additional support for several target genes identified in this study as CRC susceptibility genes. Enrichment analyses of the 136 genes uncover pathways not previously linked to CRC risk. Our study substantially expanded association signals for CRC and provided additional insight into the biological mechanisms underlying CRC development.

  • Truck platooning reshapes greenhouse gas emissions of the integrated vehicle-road infrastructure system

    • Reducing greenhouse gas emissions has turned into a pillar of climate change mitigation. Truck platooning is proposed as a strategy to lower emissions from vehicles on roads. However, the potential interactive impacts of this technology on road infrastructure emissions remain unclear. Here, we evaluate the decarbonization effects of truck platooning on the integrated vehicle-road system at a large-scale road network level, spanning 1457 road sections across North America. We show that truck platooning decreases emissions induced by truck operations, but it degrades faster the durability of road infrastructure and leads to a 27.9% rise in road emissions due to more frequent maintenance work. Overall, truck platooning results in a 5.1% emission reduction of the integrated vehicle-road system. In contrast to the benefits of emission reduction, truck platooning leads to additional financial burdens on car users and transportation agencies, calling for the consideration of tradeoffs between emissions and costs and between agencies and users. Our research provides insights into the potential applications of truck platooning to mitigate climate change.

    • Ginger (Zingiber officinale) is one of the most valued spice plants worldwide; it is prized for its culinary and folk medicinal applications and is therefore of high economic and cultural importance. Here, we present a haplotype-resolved, chromosome-scale assembly for diploid ginger anchored to 11 pseudochromosome pairs with a total length of 3.1 Gb. Remarkable structural variation was identified between haplotypes, and two inversions larger than 15 Mb on chromosome 4 may be associated with ginger infertility. We performed a comprehensive, spatiotemporal, genome-wide analysis of allelic expression patterns, revealing that most alleles are coordinately expressed. The alleles that exhibited the largest differences in expression showed closer proximity to transposable elements, greater coding sequence divergence, more relaxed selection pressure, and more transcription factor binding site differences. We also predicted the transcription factors potentially regulating 6-gingerol biosynthesis. Our allele-aware assembly provides a powerful platform for future functional genomics, molecular breeding, and genome editing in ginger.

    • Triglycerides are transported in plasma by specific triglyceride-rich lipoproteins; in epidemiological studies, increased triglyceride levels correlate with higher risk for coronary artery disease (CAD). However, it is unclear whether this association reflects causal processes. We used 185 common variants recently mapped for plasma lipids (P < 5 x 10(-8) for each) to examine the role of triglycerides in risk for CAD. First, we highlight loci associated with both low-density lipoprotein cholesterol (LDL-C) and triglyceride levels, and we show that the direction and magnitude of the associations with both traits are factors in determining CAD risk. Second, we consider loci with only a strong association with triglycerides and show that these loci are also associated with CAD. Finally, in a model accounting for effects on LDL-C and/or high-density lipoprotein cholesterol (HDL-C) levels, the strength of a polymorphism's effect on triglyceride levels is correlated with the magnitude of its effect on CAD risk. These results suggest that triglyceride-rich lipoproteins causally influence risk for CAD.

    • Background: According to the World Health Organization, two billion people will attain the age of 60 years or more by 2050. Ageing is a major risk factor for a number of neurodegenerative disorders, which currently possess challenge to the global health status, carrying economic and social consequences. Therefore, attention has been dedicated towards the development of neuroprotective agents derived from natural sources. Honeybee products, such as honey, bee pollen, bee bread, propolis, royal jelly, beeswax, and bee venom have been used for therapeutic purposes since ancient times in Egypt, Greece, and China. Despite the emergence of modern medicine, bee products remain clinically relevant owing to their potential as anti-inflammatory, anti-oxidant, and neuroprotective agents.Scope and approach: This review demonstrates the potential of bee products against neurological disorders in the light of the current literature.Key findings and conclusions: Bee products and individual isolated components have enormous therapeutic potential for multiple neurological disorders. The different studies show overall neuroprotective and nerve-tonic characteristics of bee products, mainly due to their anti-oxidant, anti-inflammatory and anti-apoptotic features. However, some limitations such as allergic reactions and the cytotoxic effect of some bee products warrant a special care in its development as drug leads in future studies.

    • Individualised treatment through therapeutic drug monitoring (TDM) may improve tuberculosis (TB) treatment outcomes but is not routinely implemented. Prospective clinical studies of drug exposure and minimum inhibitory concentrations (MICs) in multidrug-resistant TB (MDR-TB) are scarce. This translational study aims to characterise the area under the concentration-time curve of individual MDR-TB drugs, divided by the MIC for Mycobacterium tuberculosis isolates, to explore associations with markers of treatment progress and to develop useful strategies for clinical implementation of TDM in MDR-TB.Methods and analysis: Adult patients with pulmonary MDR-TB treated in Xiamen, China, are included. Plasma samples for measure of drug exposure are obtained at 0, 1, 2, 4, 6, 8 and 10 hours after drug intake at week 2 and at 0, 4 and 6 hours during weeks 4 and 8. Sputum samples for evaluating time to culture positivity and MIC determination are collected at days 0, 2 and 7 and at weeks 2, 4, 8 and 12 after treatment initiation. Disease severity are assessed with a clinical scoring tool (TBscore II) and quality of life evaluated using EQ-5D-5L. Drug concentrations of pyrazinamide, ethambutol, levofloxacin, moxifloxacin, cycloserine, prothionamide and para-aminosalicylate are measured by liquid chromatography tandem-mass spectrometry and the levels of amikacin measured by immunoassay. Dried blood spot on filter paper, to facilitate blood sampling for analysis of drug concentrations, is also evaluated. The MICs of the drugs listed above are determined using custom-made broth microdilution plates and MYCOTB plates with Middlebrook 7H9 media. MIC determination of pyrazinamide is performed in BACTEC MGIT 960.Ethics and dissemination: This study has been approved by the ethical review boards of Karolinska Institutet, Sweden and Fudan University, China.     • Herein, we reported the designed synthesis of three isostructural three-dimensional covalent organic frameworks (3D COFs) with -H, -Me, or -F substituents, which have similar crystallinity and topology. Their crystal structures were determined by continuous rotation electron diffraction (cRED), and all three 3D COFs were found to adopt a fivefold interpenetrated pts topology. More importantly, the resolution of these cRED datasets reached up to 0.9-1.0 angstrom, enabling the localization of all non-hydrogen atomic positions in a COF framework directly by 3D ED techniques for the first time. In addition, the precise control of the pore environments through the use of different functional groups led to different selectivities for CO2 over N-2. We have thus confirmed that polycrystalline COFs can be definitely studied to the atomic level as other materials, and this study should also inspire the design and synthesis of 3D COFs with tailored pore environments for interesting applications.

    • The tuning of molecular switches in solid state toward stimuli-responsive materials has attracted more and more attention in recent years. Herein, we report a switchable three-dimensional covalent organic framework (3D COF), which can undergo a reversible transformation through a hydroquinone/quinone redox reaction while retaining the crystallinity and porosity. Our results clearly show that the switching process gradually happened through the COF framework, with an almost quantitative conversion yield. In addition, the redox-triggered transformation will form different functional groups on the pore surface and modify the shape of pore channel, which can result in tunable gas separation property. This study strongly demonstrates 3D COFs can provide robust platforms for efficient tuning of molecular switches in solid state. More importantly, switching of these moieties in 3D COFs can remarkably modify the internal pore environment, which will thus enable the resulting materials with interesting stimuli-responsive properties. Tuning of molecular switches in solid state toward stimuli-responsive materials attracted attention in recent years but has not yet been realized in three-dimensional (3D) covalent organic frameworks (COFs). Herein, the authors demonstrate a stable and switchable 3D COF which undergoes reversible transformation through a hydroquinone/quinone redox reaction.

    • Rational construction of covalent organic frameworks (COFs) with novel structures still remains a challenge. Herein, we report the designed synthesis of two COFs, 2D-BPTA-COF and 3D-BMTA-COF, starting from biphenyl-based precursors but with different groups at the ortho positions. Both COFs exhibited high crystallinity and large surface area, and interestingly, 2D-BPTA-COF crystallizes into 2D sheets with AB stacking mode while 3D-BMTA-COF adopts a 7-fold interpenetrated structure with pts topology. This structural difference could be ascribed to the introduction of methyl groups in the building blocks, as the dihedral angle of biphenyl rings in 2D-BPTA-COF is similar to 0 degrees while in 3D-BMTA-COF it is similar to 60 degrees. Therefore, it is possible to synthesize COFs with different structures by twisting building blocks from planar to tetrahedral with steric hindrance. We believe this result represents a general and straightforward way to expand the diversity of tetrahedral nodes for constructing 3D COFs in the future, and moreover, a new tetrahedral node for constructing 3D COFs is now available.

    • Noninvasive measurement of blood pressure by optical methods receives considerable interest, but the complexity of the measurement and the difficulty of adjusting parameters restrict applications. We develop a method for estimating the systolic and diastolic blood pressure using a single-point optical recording of a photoplethysmographic (PPG) signal. The estimation is data-driven, we use automated machine learning algorithms instead of mathematical models. Combining supervised learning with a discrete wavelet transform, the method is insensitive to minor irregularities in the PPG waveform, hence both pulse oximeters and smartphone cameras can record the signal. We evaluate the accuracy of the estimation on 78 samples from 65 subjects (40 male, 25 female, age 29±7) with no history of cardiovascular disease. The estimate for systolic blood pressure has a mean error 4.9±4.9 mm Hg, and 4.3±3.7 mm Hg for diastolic blood pressure when using the oximeter-obtained PPG. The same values are 5.1±4.3 mm Hg and 4.6±4.3 mm Hg when using the phone-obtained PPG, comparing with A&D UA-767PBT result as gold standard. The simplicity of the method encourages ambulatory measurement, and given the ease of sharing the measured data, we expect a shift to data-oriented approaches deriving insight from ubiquitous mobile devices that will yield more accurate machine learning models in monitoring blood pressure.

    • Feeding is essential for animal survival and reproduction and is regulated by both internal states and external stimuli. However, little is known about how internal states influence the perception of external sensory cues that regulate feeding behavior. Here, we investigated the neuronal and molecular mechanisms behind nutritional state-mediated regulation of gustatory perception in control of feeding behavior in the brown planthopper and Drosophila. We found that feeding increases the expression of the cholecystokinin-like peptide, sulfakinin (SK), and the activity of a set of SK-expressing neurons. Starvation elevates the transcription of the sugar receptor Gr64f and SK negatively regulates the expression of Gr64f in both insects. Interestingly, we found that one of the two known SK receptors, CCKLR-17D3, is expressed by some of Gr64f-expressing neurons in the proboscis and proleg tarsi. Thus, we have identified SK as a neuropeptide signal in a neuronal circuitry that responds to food intake, and regulates feeding behavior by diminishing gustatory receptor gene expression and activity of sweet sensing GRNs. Our findings demonstrate one nutritional state-dependent pathway that modulates sweet perception and thereby feeding behavior, but our experiments cannot exclude further parallel pathways. Importantly, we show that the underlying mechanisms are conserved in the two distantly related insect species.

    • At the beginning of the new century, since nuclear energy received widespread attention due to its economy and environmental protection, the nuclear industry has developed rapidly. However, there are still some issues challenging the safety of nuclear power plants (NPPs) especially at extreme conditions. During a hypothetical severe accident in a light water reactor, the core fuel will be disrupted and melt-down resulting from the absence of coolant, and finally forms a high-temperature molten pool at the lower head of reactor pressure vessel (RPV), which threaten the integrity of RPV, one of the most important barriers to prevent radioactive fission product releasing to the environment. In this regard, the decay heat removal of the molten pool plays an essential role in the strategy of in-vessel retention (IVR)-external reactor vessel cooling (ERVC) to arrest the molten corium inside the RPV, as shown in Fig. 1.This paper examines the research work about experimental investigation, theoretical analysis, and numerical simulation conducted for ERVC and summarizes the different approaches proposed by scholars to strengthen ERVC capabilities. Those approaches are the replacement of the cooling work materials, the application of nanofluids, surface modification, and other methods. Finally, it concludes the deficiencies of the current study and the direction of future efforts.

    • The occurrence of porous deposits on the core surface during the operation of a pressurized water reactor can seriously affect its heat transfer characteristics and increase the surface temperature of the cladding. Continued heat transfer deterioration causes subcooled boiling and boron enrichment within porous deposits, leading to axial power anomalies in the core, and the study of porous deposits is beneficial to improving the economics and safety of reactor operation. In this paper, a two-dimensional boiling unit model of a porous deposits was established using COMSOL software to couple heat conduction, darcy flow, solute transport and chemical reactions in a multiphysics field to reasonably predict the temperature field, velocity field, boric acid concentration distribution, and PH distribution within the porous deposits. On this basis, the influence of different parameters (porosity, boiling unit radius of porous deposits, heat flow density) on the maximum effective thermal conductivity, the pattern of thermal parameters (temperature, superheat, percentage of subcooled boiling, and saturation temperature) on the chimney boundary are analyzed. The results show that localized superheating, maximum boric acid concentration and subcooled boiling occur at the bottom of the porous deposits. There is a negative correlation between the porosity and the percentage of subcooled boiling area, and a positive correlation between the radius of the porous deposits and the heat flow density and the percentage of subcooled boiling area.

    • IVR-ERVC (In-Vessel Retention – External Reactor Vessel Cooling) is a critical accident management method for ensuring the integrity of the reactor pressure vessel (RPV) lower head. One of the most crucial aspects within this method is to enhance the CHF (Critical Heat Flux) on the outer surface of the reactor pressure vessel (RPV) lower head. This paper explores the application of graphene nanofluids in IVR-ERVC. This paper uses computational fluid dynamics to numerically simulate the CHF characteristics of graphene nanofluids under different undercooling, mass flow rate, concentration, tilt angle, and gap size conditions and analyzes the impact of different factors on CHF and the coupling effects between different factors. The undercooling range is 5 K–100 K, the mass flow rate range is 150 kg/(m2∙s) to 3000 kg/(m2∙s), the concentration range is 0–1%, the inclination angle range is 0°–90°, and the gap sizes are 10 mm and 20 mm. The results show that the CHF can be effectively improved by adding nano-graphene into the base solution, and the CHF increases with the increase of subcooling degree, mass flow rate, concentration, dip Angle and gap size. Within the simulation range, the strengthening effect on CHF weakens when raising the undercooling and mass flow rate. And the larger the concentration and inclination angle within the simulation range, the better the enhancement effect on CHF. The maximum increase was 290%, while the average increase was 75.6%. This article studies the coupling effects between different parameters through numerical simulation. It can be concluded that increasing the mass flow rate weakens the enhancement of subcooling and concentration on CHF, increasing the subcooling weakens the enhancement effect of mass flow rate and concentration on CHF, and increasing the concentration enhances the enhancement effect of subcooling and mass flow rate on CHF. Enhance the mass flow rate will weaken tilt angle effect, while increasing the concentration will enhance the strengthening effect of tilt angle on CHF.

    • Corrosion of pressurized water reactors (PWR) in nuclear power plants can lead to serious safety hazards. This study aims to analyze the deposition of corrosion products using FLUENT software. Deposition models and thermal resistance models were developed, and the effects of deposits on the reactor's thermal–hydraulic characteristics were evaluated. Additionally, the impact of various parameters on deposition and thermal–hydraulic characteristics was examined. Results show that deposits accumulate extensively in the inlet section of the fuel cladding, while appearing as spot deposits in the outlet section. For deposit thicknesses below 30 μm, the surface temperature of the cladding gradually increases. However, when the thickness exceeds 30 μm, the surface temperature rapidly rises. Furthermore, the study reveals that the deposition amount decreases with increasing inlet flow velocity, exhibits an upward trend with higher inlet temperature, and increases with a higher wall heat flux density. This research provides important insights for understanding core deposition and thermal–hydraulic characteristics in nuclear reactor systems. It offers valuable guidance for enhancing safety and operational efficiency in nuclear power plants.

  • Adaptive Thresholding with Inverted Triangular Area for Real-Time Detection of the Heart Rate from Photoplethysmogram Traces on a Smartphone

    • Photoplethysmogram (PPG) signals acquired by smartphone cameras are weaker than those acquired by dedicated pulse oximeters. Furthermore, the signals have lower sampling rates, have notches in the waveform and are more severely affected by baseline drift, leading to specific morphological characteristics. This paper introduces a new feature, the inverted triangular area, to address these specific characteristics. The new feature enables real-time adaptive waveform detection using an algorithm of linear time complexity. It can also recognize notches in the waveform and it is inherently robust to baseline drift. An implementation of the algorithm on Android is available for free download. We collected data from 24 volunteers and compared our algorithm in peak detection with two competing algorithms designed for PPG signals, Incremental-Merge Segmentation (IMS) and Adaptive Thresholding (ADT). A sensitivity of 98.0 % and a positive predictive value of 98.8 % were obtained, which were 7.7 % higher than the IMS algorithm in sensitivity, and 8.3 % higher than the ADT algorithm in positive predictive value. The experimental results confirmed the applicability of the proposed method.

  • Real-time Quantifying Heart Beat Rate from Facial Video Recording on a Smart Phone using Kalman Filters

    • Photoplethysmography (PPG) can be carried out through facial video recording by a smart phone camera in ambient light. The main challenge is to eliminate motion artifacts and ambient noise. We describe a real-time algorithm to quantify the heart beat rate from facial video recording captured by the camera of a smart phone. We extract the green channel from the video. Then we normalize it and use a Kalman filter with a particular structure to eliminate ambient noise. This filter also enhances the heart pulse component in the signal distorted by Gaussian noise and white noise. After that we employ a band-pass FIR filter to remove the remaining motion artifacts. This is followed by peak detection or Lomb periodogram to estimate heart rate. The algorithm has low computational overhead, low delay and high robustness, making it suitable for real-time interaction on a smart phone. Finally we describe an Android application based on this study.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

    • 3C 279 is an archetypal blazar with a prominent radio jet that show broadband flux density variability across the entire electromagnetic spectrum. We use an ultra-high angular resolution technique - global Very Long Baseline Interferometry (VLBI) at 1.3mm (230 GHz) - to resolve the innermost jet of 3C 279 in order to study its fine-scale morphology close to the jet base where highly variable-ray emission is thought to originate, according to various models. The source was observed during four days in April 2017 with the Event Horizon Telescope at 230 GHz, including the phased Atacama Large Millimeter/submillimeter Array, at an angular resolution of ∼20 μas (at a redshift of z = 0:536 this corresponds to ∼0:13 pc ∼ 1700 Schwarzschild radii with a black hole mass MBH = 8 × 108 M⊙). Imaging and model-fitting techniques were applied to the data to parameterize the fine-scale source structure and its variation.We find a multicomponent inner jet morphology with the northernmost component elongated perpendicular to the direction of the jet, as imaged at longer wavelengths. The elongated nuclear structure is consistent on all four observing days and across diffierent imaging methods and model-fitting techniques, and therefore appears robust. Owing to its compactness and brightness, we associate the northern nuclear structure as the VLBI "core". This morphology can be interpreted as either a broad resolved jet base or a spatially bent jet.We also find significant day-to-day variations in the closure phases, which appear most pronounced on the triangles with the longest baselines. Our analysis shows that this variation is related to a systematic change of the source structure. Two inner jet components move non-radially at apparent speeds of ∼15 c and ∼20 c (∼1:3 and ∼1:7 μas day-1, respectively), which more strongly supports the scenario of traveling shocks or instabilities in a bent, possibly rotating jet. The observed apparent speeds are also coincident with the 3C 279 large-scale jet kinematics observed at longer (cm) wavelengths, suggesting no significant jet acceleration between the 1.3mm core and the outer jet. The intrinsic brightness temperature of the jet components are ≤1010 K, a magnitude or more lower than typical values seen at ≥7mm wavelengths. The low brightness temperature and morphological complexity suggest that the core region of 3C 279 becomes optically thin at short (mm) wavelengths.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • Molecular design of acceptor and donor molecules has enabled major progress in organic photovoltaics. Li et al. show that branched alkyl chains in non-fullerene acceptors allow favourable morphology in the active layer, enabling a certified device efficiency of 17.9%. Molecular design of non-fullerene acceptors is of vital importance for high-efficiency organic solar cells. The branched alkyl chain modification is often regarded as a counter-intuitive approach, as it may introduce an undesirable steric hindrance that reduces charge transport in non-fullerene acceptors. Here we show the design and synthesis of a highly efficient non-fullerene acceptor family by substituting the beta position of the thiophene unit on a Y6-based dithienothiophen[3,2-b]-pyrrolobenzothiadiazole core with branched alkyl chains. It was found that such a modification to a different alkyl chain length could completely change the molecular packing behaviour of non-fullerene acceptors, leading to improved structural order and charge transport in thin films. An unprecedented efficiency of 18.32% (certified value of 17.9%) with a fill factor of 81.5% is achieved for single-junction organic solar cells. This work reveals the importance of the branched alkyl chain topology in tuning the molecular packing and blend morphology, which leads to improved organic photovoltaic performance.

    • Glycan microarrays have played important roles in detection and specificity assignment of glycan recognition by proteins. However, the size and diversity of glycan libraries in current microarray systems are small compared to estimated glycomes, and these may lead to missed detection or incomplete assignment. For microarray construction, covalent and noncovalent immobilization are the two types of methods used, but a direct comparison of results from the two platforms is required. Here we develop a chemical strategy to prepare lipid-linked probes from both naturally derived aldehyde-terminating and synthetic amino-terminating glycans that addresses the two aspects: expansion of sequence-defined glycan libraries and comparison of the two platforms. We demonstrate the specific recognition by plant and mammalian lectins, carbohydrate-binding modules and antibodies and the overall similarities from the two platforms. Our results provide new knowledge on unique glycan-binding specificities for the immune receptor Dectin-1 toward beta-glucans and the interaction of rotavirus P[19] adhesive protein with mucin O-glycan cores.

  • Effect of Initial Oriented Columnar Grains on the Texture Evolution and Magnetostriction in Fe-Ga Rolled Sheets

    • The effects of initial oriented columnar grains on the texture evolution and magnetostriction in (Fe83Ga17)(99.9)(NbC)(0.1) rolled sheets were investigated. The recrystallization texture evolution exhibited the heredity of initial orientations, concerning the formation of cube and Goss textures in the primary recrystallized sheet for the columnar-grained sample. Moreover, the growth advantage of Goss grains was more obvious than that of cube grains during the secondary recrystallization process. Because of the combined effect of this and Nb-rich precipitates as inhibitors, a sharp Goss texture and very large Goss grains were achieved in the secondary recrystallized sheet for the columnar-grained sample. For comparison, the secondary recrystallization in the equiaxed-grained sample was not fully developed although there were Nb-rich precipitates as inhibitors. We think this could be ascribed to the large particle size and premature coarsening of precipitates. Magnetostriction of the secondary recrystallized columnar-grained sheet was up to 232 ppm owing to the ideal Goss texture and quite large grain size. As for the equiaxed-grained sample, the magnetostriction was only 163 ppm in the secondary recrystallized sheet.

    • Waste Electrical and Electronic Equipment (WEEE) are one of the most significant waste streams in modern societies. Full disassembly of WEEE is rarely an ideal solution due to high disassembly costs. Selective disassembly, which prioritizes operations for partial disassembly according to the legislative and economic considerations of specific stakeholders, is becoming an important yet still challenging research topic in recent years. In this paper, a Particle Swarm Optimization (PSO)-based selective disassembly planning method embedded with customizable decision-making models has been developed. The developed method is flexible to handle WEEE to meet the various requirements of stakeholders, and is capable to achieve optimized selective plans. Practical cases on Liquid Crystal Display (LCD) televisions have been used to verify and demonstrate the effectiveness of the research in application scenarios.

    • A novel polysaccharide from Ulva lactuca (ULP) was purified using a Sepharose CL-4B column. Fourier transform infrared spectroscopy, high-performance liquid chromatography, and nuclear magnetic resonance spectroscopy were employed to analyze the structure of ULP. It consisted of rhamnose (Rha), glucuronic acid (GluA), galactose (Gal), and xylose (Xyl) at a molar ratio of 32.75:22.83:1.07:6.46 with the molecular weight of 2.24 x 10(5) Da. The four major glycosidic residues found in ULP were -> 2,3)-alpha-L-Rhap-(1 ->, -> 4)-beta-D-GlcpA-(1 ->, -> 2,6)-beta-D-Galp-(1 ->, and -> 4)-beta-D-Xylp-(1 ->. The antihyperuricemic activity of ULP was exhibited by detecting related biochemical indexes, urate transporter gene expressions, renal histopathology, and intestinal microbiota shifts. ULP obviously decreased the levels of serum uric acid (UA), blood urea nitrogen, and creatinine, while inhibited serum and hepatic xanthine oxidase activities as well as improved renal injury in hyperuricemic mice. Furthermore, the upregulation of UA excretion genes ABCG2/OAT1 and downregulation of UA resorption genes URAT1 and GLUT9 were detected. In addition, ULP exerted its antihyperuricemic effect through regulating the intestinal microbiome, characterized by elevating the helpful microbial abundance, meanwhile declining the harmful bacterial abundance and restoring the gut microbiome homeostasis. This study demonstrates the antihyperuricemic activity of ULP and its potential effect for the treatment of hyperuricemia-related diseases.

    • The Schottky barrier height (SBH) of an ultrathin epitaxial NiSi(2-y) film grown on Si(100) is modified significantly by means of dopant segregation (DS). The DS process begins with the NiSi(2-y) formation and is followed by dopant implantation and drive-in annealing. The rapid lattice restoration and superior morphological stability upon heat treatment up to 800 degrees C allow the epitaxial NiSi(2-y) film to take full advantage of the DS process. For drive-in annealing below 750 degrees C, the effective SBH is altered to similar to 0.9-1 eV for both electrons and holes by B-DS and As-DS, respectively, without deteriorating the integrity of the NiSi(2-y) film.

    • Analyses of genome-wide association study (GWAS) data have revealed that detectable genetic mosaicism involving large (>2 Mb) structural autosomal alterations occurs in a fraction of individuals. We present results for a set of 24,849 genotyped individuals (total GWAS set II [TGSII]) in whom 341 large autosomal abnormalities were observed in 168 (0.68%) individuals. Merging data from the new TGSII set with data from two prior reports (the Gene-Environment Association Studies and the total GWAS set I) generated a large dataset of 127,179 individuals; we then conducted a meta-analysis to investigate the patterns of detectable autosomal mosaicism (n = 1,315 events in 925 [0.73%] individuals). Restricting to events >2 Mb in size, we observed an increase in event frequency as event size decreased. The combined results underscore that the rate of detectable mosaicism increases with age (p value = 5.5 x 3 10(-31)) and is higher in men (p value = 0.002) but lower in participants of African ancestry (p value = 0.003). In a subset of 47 individuals from whom serial samples were collected up to 6 years apart, complex changes were noted over time and showed an overall increase in the proportion of mosaic cells as age increased. Our large combined sample allowed for a unique ability to characterize detectable genetic mosaicism involving large structural events and strengthens the emerging evidence of non-random erosion of the genome in the aging population.

    • To investigate large structural clonal mosaicism of chromosome X, we analysed the SNP microarray intensity data of 38,303 women from cancer genome-wide association studies (20,878 cases and 17,425 controls) and detected 124 mosaic X events >2 Mb in 97 (0.25%) women. Here we show rates for X-chromosome mosaicism are four times higher than mean autosomal rates; X mosaic events more often include the entire chromosome and participants with X events more likely harbour autosomal mosaic events. X mosaicism frequency increases with age (0.11% in 50-year olds; 0.45% in 75-year olds), as reported for Y and autosomes. Methylation array analyses of 33 women with X mosaicism indicate events preferentially involve the inactive X chromosome. Our results provide further evidence that the sex chromosomes undergo mosaic events more frequently than autosomes, which could have implications for understanding the underlying mechanisms of mosaic events and their possible contribution to risk for chronic diseases.

    • A cost-effective technique to dissipate the energy in hydropower systems is the formation of a swirling flow within a tunnel. In such flows, an air core with a significant cross section usually occurs. To reveal the air-core features in the horizontal tunnel of a high-head shaft spillway, laboratory tests, numerical modeling, and prototype observations are performed, to examine issues such as the formation of the air core, the interjacent air motion, the air-carrying capacity, and the scale effects. It is shown that the shape of the air core varies greatly in the axial and radial directions along the tunnel and that the center of the core deviates from the axis of the tunnel. The motion of the air within the core is caused by the combined action of the water entrainment on the inner surface of the swirling flow and the axial pressure difference in the air core. The aeration process can be divided into five processes with respect to the changes of the gate openings. A theoretical expression is established for the air-carrying capacity of the swirling flow. The vacuum degree is the similarity condition of the air-carrying capacity of the swirling flow between the model and prototype tests based on the Froude law of the similitude, and this similarity condition is verified by both the model and prototype results. This work provides a reference for the application of the swirling flows in horizontal hydropower tunnels.

  • Galectin-3 Decreases 4-1BBL Bioactivity by Crosslinking Soluble and Membrane Expressed 4-1BB

    • 4-1BB is a T cell costimulatory receptor and a member of the tumor necrosis factor receptor superfamily. Here, we show that Galectin-3 (Gal-3) decreases the cellular response to its ligand (4-1BBL). Gal-3 binds to both soluble 4-1BB (s4-1BB) and membrane-bound 4-1BB (mem4-1BB), without blocking co-binding of 4-1BBL. In plasma, we detected complexes composed of 4-1BB and Gal-3 larger than 100 nm in size; these complexes were reduced in synovial fluid from rheumatoid arthritis. Both activated 4-1BB+ T cells and 4-1BB-transfected HEK293 cells depleted these complexes from plasma, followed by increased expression of 4-1BB and Gal-3 on the cell surface. The increase was accompanied by a 4-fold decrease in TNFα production by the 4-1BBhighGal-3+ T cells, after exposure to 4-1BB/Gal-3 complexes. In RA patients, complexes containing 4-1BB/Gal-3 were dramatically reduced in both plasma and SF compared with healthy plasma. These results support that Gal-3 binds to 4-1BB without blocking the co-binding of 4-1BBL. Instead, Gal-3 leads to formation of large soluble 4-1BB/Gal-3 complexes that attach to mem4-1BB on the cell surfaces, resulting in suppression of 4-1BBL’s bioactivity.
