    • The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process flow and r-process beta-decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will offer unique, unprecedented opportunities to investigate many of the important reactions. The high yield of radioactive isotopes, even far away from the valley of stability, allows the investigation of isotopes involved in processes as exotic as the r or rp processes.

  • Scientific program of DERICA-prospective accelerator and storage ring facility for radioactive ion beam research

    • Studies of radioactive ions (RIs) are the most thriving field of low-energy nuclear physics. In this paper, the concept and the scientific agenda of the prospective accelerator and storage ring facility for RI beam (RIB) research are proposed for a large-scale international project based at the Flerov Laboratory of Nuclear Reactions of the Joint Institute for Nuclear Research. The motivation for the new facility is discussed and its characteristics are briefly presented and shown to be comparable to those of advanced world centers, the so-called "RIB factories". In the project, the emphasis is made on studies with short-lived RIBs in storage rings. A unique feature of the project is the possibility of studying electron-RI interactions in a collider experiment to determine the fundamental properties of nuclear matter, in particular, electromagnetic form factors of exotic nuclei.

    • The project of a new and more powerful in-flight fragment separator ACCULINNA-2 at U-400M cyclotron in FLNR, JINR planned to build in addition to the existing separator ACCULINNA is presented. The new separator will provide high intensity RIBs in the lowest energy range (5÷50 MeV/nucleon) which is attainable for in-flight separators. The possibilities for the astrophysics studies at the proposed device are presented. ACCULINNA-2 separator is planned to be constructed in the years 2010-2015. The current status of the project is reported.

    • The new facility fragment separator ACCULINNA-2 will be put into operation at the beginning of 2015 in FLNR JINR. The new separator is destined to add considerably to the studies of drip-line nuclei performed with the use of variety of direct reactions known to be distinctive to the 15 - 50 MeV/amu exotic secondary RIBs. Intense beams provided by the U-400M cyclotron will ensure the achievement of this objective. In addition to the RIB separation accomplished by means of the dipole-wedge-dipole selection, the addition of a zero-degree dipole magnet setup is foreseen. A long (13 m) straight section will provide precise time-of-flight measurements. © Owned by the authors, published by EDP Sciences, 2014.

    • A program for upgrade of existing radioactive ion beams facilities at Flerov Laboratory of Nuclear Reactions, JINR Dubna is presented. A project of a new in-flight fragment separator ACCULINNA-2 is proposed. It is expected the new instrument will be more universal and powerful than the existing nowadays. The beam intensity should be increased by factor 10-15, its optical quality greatly improved and the range of the accessible secondary radioactive beams broadened up to Z∼20. Main ion-optical characteristics, operating principles and a tentative plan for the project realization are included. An extensive research program based on local experiments made so far and international proposals for these equipments is discussed.

  • Recent results related to excited states of 6Be and 10He

    • The 1H(6Li,6Be)n charge-exchange reaction and the 3H(8He, p)10He two-neutron transfer reaction were recently carried out at the ACCULINNA fragment separator (FLNR, Dubna) to populate the ground and excited states of 6Be and 10He nuclei, respectively. The 6Be ET spectrum (ET is the 6Be energy above its 4He+p+p decay threshold) was obtained with high statistics and described by the well-known 0+ ground state of 6Be at ET = 1.37 MeV, the 2+ state at ET = 3.05 MeV and a broad structure extending from 4 to 16 MeV which could be interpreted as the isovector soft dipole mode associated with the 6Li ground state. In the 10He case the 0+ ground state was found at about 2.1(2) MeV (T ∼ 2 MeV) above the 8He+n+n breakup threshold. Angular correlations observed for 10He decay products show prominent interference patterns allowing to draw conclusions about the structure of low-energy excited states: 1- (E T ∼ 5 MeV) and 2+ (ET > 6 MeV).

  • Evidence for the First Excited State of H 7

    • The H7 system was populated in the H2(He8,He3)H7 reaction with a 26 AMeV He8 beam. The H7 missing mass energy spectrum, the H3 energy and angular distributions in the H7 decay frame were reconstructed. The H7 missing mass spectrum shows a peak, which can be interpreted either as unresolved 5/2+ and 3/2+ doublet or one of these states at 6.5(5) MeV. The data also provide indications of the 1/2+ ground state of H7 located at 1.8(5) MeV with quite a low population cross section of ∼25 μb/sr within angular range θc.m.≃(17°-27°).

    • The R3B experiment (Reactions with Relativistic Radioactive Beams) at FAIR (Facility for Antiproton and Ion Research) is a versatile setup dedicated to the study of reactions induced by high-energy radioactive beams. It will provide kinematically complete measurements with high efficiency, acceptance and resolution, making possible a broad physics program with rare-isotopes. CALIFA (CALorimeter for In-Flight detection of gamma-rays and high energy charged pArticles), is a complex detector based on scintillation crystals, that will surround the target of the R3B experiment. CALIFA will act as a total absorption gamma-calorimeter and spectrometer, as well as identifier of charged particles from target residues. This versatility is its most challenging requirement, demanding a huge dynamic range, to cover from low energy gamma-rays up to 300 MeV protons. This fact, along with the high-energy of the beams determine the conceptual design of the detector, presented in this paper, together with the technical solutions proposed for its construction.

Namely, ACCULINNA-2 should provide high intensity RIBs in the lowest energy range attainable for in-flight separators.

  • Deep excursion beyond the proton dripline. I. Argon and chlorine isotope chains

    • The proton-unbound argon and chlorine isotopes have been studied by measuring trajectories of their decay-in-flight products by using a tracking technique with microstrip detectors. The proton (1p) and two-proton (2p) emission processes have been detected in the measured angular correlations "heavy-fragment"+p and "heavy-fragment"+p+p, respectively. The ground states of the previously unknown isotopes Cl30 and Cl28 have been observed for the first time, providing the 1p-separation energies Sp of -0.48(2) and -1.60(8), MeV, respectively. The relevant systematics of 1p- and 2p-separation energies have been studied theoretically in the core+p and core+p+p cluster models. The first-time observed excited states of Ar31 allow one to infer the 2p-separation energy S2p of 6(34) keV for its ground state. The first-time observed state in Ar29 with S2p=-5.50(18) MeV can be identified as either a ground state or an excited state according to different systematics.

  • Resonant states in H 7: Experimental studies of the H 2 (He 8, He 3) reaction

    • The extremely neutron-rich system H7 was studied in the direct H2(He8,He3)H7 transfer reaction with a 26 AMeV secondary He8 beam [Bezbakh et al., Phys. Rev. Lett. 124, 022502 (2020)PRLTAO0031-900710.1103/PhysRevLett.124.022502]. The missing mass spectrum and center-of-mass angular distributions of H7, as well as the momentum distribution of the H3 fragment in the H7 frame, were constructed. In addition, we carried out another experiment with the same beam but a modified setup, which was cross-checked by the study of the H2(Be10,He3)Li9 reaction. A solid experimental evidence is provided that two resonant states of H7 are located in its spectrum at 2.2(5) and 5.5(3)MeV relative to the H3+4n decay threshold. Also, there are indications that the resonant states at 7.5(3) and 11.0(3)MeV are present in the measured H7 spectrum. Based on the energy and angular distributions, obtained for the studied H2(He8,He3)H7 reaction, the weakly populated 2.2(5)-MeV peak is ascribed to the H7 ground state. It is highly plausible that the firmly ascertained 5.5(3)-MeV state is the 5/2+ member of the H7 excitation 5/2+-3/2+ doublet, built on the 2+ configuration of valence neutrons. The supposed 7.5-MeV state can be another member of this doublet, which could not be resolved in Bezbakh et al. [Phys. Rev. Lett. 124, 022502 (2020)PRLTAO0031-900710.1103/PhysRevLett.124.022502]. Consequently, the two doublet members appeared in the spectrum of H7 in the work mentioned above as a single broad 6.5-MeV peak.

  • H 6 states studied in the H 2 (He 8, He 4) reaction and evidence of an extremely correlated character of the H 5 ground state

    • The extremely neutron-rich system H6 was studied in the direct H2(He8,He4)H6 transfer reaction with a 26A MeV secondary He8 beam. The measured missing mass spectrum shows a broad bump at ≈4-8 MeV above the H3+3n decay threshold. This bump can be interpreted as a broad resonant state in H6 at 6.8(5) MeV. The population cross section of such a presumably p-wave state (or it may be few overlapping states) in the energy range from 4 to 8 MeV is dσ/dωc.m.≃190-80+40μb/sr in the angular range 5°<θc.m.<16°. The obtained missing mass spectrum is practically free of H6 events below 3.5 MeV (dσ/dωc.m. 5μb/sr in the same angular range). The steep rise of the H6 missing mass spectrum at ≈3 MeV allows us to derive the lower limit for the possible resonant-state energy in H6 to be 4.5(3) MeV. According to the paring energy estimates, such a 4.5(3) MeV resonance is a realistic candidate for the H6 ground state (g.s.). The obtained results confirm that the decay mechanism of the H7 g.s. (located at 2.2 MeV above the H3+4n threshold) is the "true"(or simultaneous) 4n emission. The resonance energy profiles and the momentum distributions of fragments of the sequential H6→H5(g.s.)+n→H3+3n decay were analyzed by the theoretically updated direct four-body-decay and sequential-emission mechanisms. The measured momentum distributions of the H3 fragments in the H6 rest frame indicate very strong "dineutron-type"correlations in the H5 ground state decay.

    • The extremely neutron-rich systems 7H, 6H were studied in the 2H(8He, 3He)7H and 2H(8He, 4He)6H proton and deuteron pickup reactions with a 26 AMeV secondary 8He beam produced at the new ACCULINNA-2 fragment separator. In addition, the same proton and deuteron pickup reactions were generated using the 42 AMeV 10Be beam, and the population of low-lying 9Li and 8Li states was measured in reactions 2H(10Be,3He)9Li and 2H(10Be,4He)8Li, respectively. The latter were used as reference measurements in order to check the setup calibration over the excitation energy of 7,6H and to determine the real experimental energy resolution which was compared with Monte Carlo calculations. The corresponding results obtained for the superheavy hydrogen systems 7H, 6H are presented and discussed. Typical excitation spectra of the 9Li and 8Li nuclei are also shown.

  • Three-body correlations in direct reactions: Example of Be 6 populated in the (p,n) reaction

    • The Be6 continuum states were populated in the charge-exchange reaction H1(Li6,Be6)n collecting very high statistics data (∼5×106 events) on the three-body α+p+p correlations. The Be6 excitation energy region below ∼3MeV is considered, where the data are dominated by contributions from the 0+ and 2+ states. It is demonstrated how the high-statistics few-body correlation data can be used to extract detailed information on the reaction mechanism. Such a derivation is based on the fact that highly spin-aligned states are typically populated in the direct reactions.

  • Lifetime of 26S and a Limit for its 2p Decay Energy

    • The unknown isotope (26)S, expected to decay by two-proton (2p) emission, was studied theoretically and searched experimentally. The structure of this nucleus was examined within the relativistic mean field (RMF) approach. A method for taking into account the many-body structure in the three-body decay calculations was developed. The results of the RMF calculations were used as an input for the three-cluster decay model optimized for the study of a possible 2p decay branch of this nucleus. The experimental search for (26)S was performed by fragmentation of a 50.3 A MeV (32)S beam. No events of a particles table (26)S or (25)P (a presumably proton-unstable subsystem of (26)S) were observed. Based on the obtained production systematics, an upper half-life limit of T(1/2) 640 keV for (26)S. Analogous limits for (25)P are found as T(1/2) 110 keV. In the case that the one-proton emission is the main branch of the (26)S decay, a limit Q(2p) > 230 keV would follow for this nucleus. According to these limits, it is likely that (26)S resides in the picosecond life time range

  • Deep excursion beyond the proton dripline. II. Toward the limits of existence of nuclear structure

    • Prospects of experimental studies of argon and chlorine isotopes located far beyond the proton dripline are studied by using systematics and cluster models. The deviations from the widespread systematics observed in Cl-28,Cl-29 and Ar-29,Ar-30 have been theoretically substantiated, and analogous deviations have been predicted for the lighter chlorine and argon isotopes. The limits of nuclear structure existence are predicted for Ar and Cl isotopic chains, with Ar-26 and Cl-25 found to be the lightest sufficiently long-living nuclear systems. By simultaneous measurements of protons and gamma rays following decays of such systems as well as their beta-delayed emission, an interesting synergy effect may be achieved, which is demonstrated by the example of Cl-30 and Ar-31 ground-state studies. Such a synergy effect may be provided by the new EXPERT setup (EXotic Particle Emission and Radioactivity by Tracking) being operated inside the fragment separator and spectrometer facility at GSI, Darmstadt.

  • Towards the Limits of Existence of Nuclear Structure: Observation and First Spectroscopy of the Isotope K-31 by Measuring Its Three-Proton Decay

    • The most remote isotope from the proton dripline (by 4 atomic mass units) has been observed: K-31. It is unbound with respect to three-proton (3p) emission, and its decays have been detected in flight by measuring the trajectories of all decay products using microstrip detectors. The 3p emission processes have been studied by the means of angular correlations of S-28 + 3p and the respective decay vertices. The energies of the previously unknown ground and excited states of K-31 have been determined. This provides its 3p separation energy value S-3p of -4.6(2) MeV. Upper half-life limits of 10 ps of the observed K-31 states have been derived from distributions of the measured decay vertices.

  • Observation and Spectroscopy of New Proton-Unbound Isotopes Ar-30 and Cl-29: An Interplay of Prompt Two-Proton and Sequential Decay

    • Previously unknown isotopes Ar-30 and Cl-29 have been identified by measurement of the trajectories of their in-flight decay products S-28 + p + p and S-28 + p, respectively. The analysis of angular correlations of the fragments provided information on decay energies and the structure of the parent states. The ground states of Ar-30 and Cl-29 were found at 2.25(-0.10)(+0.15) and 1.8 +/- 0.1 MeV above the two-and one-proton thresholds, respectively. The lowest states in Ar-30 and Cl-29 point to a violation of isobaric symmetry in the structure of these unbound nuclei. The two-proton decay has been identified in a transition region between simultaneous two-proton and sequential proton emissions from the Ar-30 ground state, which is characterized by an interplay of three-body and two-body decay mechanisms. The first hint of a fine structure of the two-proton decay of Ar-30*(2(+)) has been obtained by detecting two decay branches into the ground and first-excited states of the S-28 fragment.

  • Isovector soft dipole mode in Be-6

    • By using the H-1(Li-6,Be-6)n charge-exchange reaction, population of continuum states in Be-6 was observed up to E-T = 16 MeV, E-T being the Be-6 energy above its three-body decay threshold. In kinematically complete measurements performed by detecting alpha + p + p coincidences, an E-T spectrum of high statistics was obtained, containing approximately similar to 5 x 10(6) events. The spectrum provides detailed correlation information about the well-known 0(+) ground state of Be-6 at E-T = 1.37 MeV and its 2(+) state at E-T = 3.05 MeV. Moreover, a broad structure extending from 4 to 16 MeV was observed. It contains negative parity states populated by Delta L = 1 angular momentum transfer without other significant contributions. This structure can be interpreted as a novel phenomenon, i.e. the isovector soft dipole mode associated with the Li-6 ground state. The population of this mode in the charge-exchange reaction is a dominant phenomenon for this reaction, being responsible for about 60% of the cross-section obtained in the measured energy range.

This versatility is its most challenging requirement, demanding a huge dynamic range, to cover from low energy gamma-rays up to 300 MeV protons. This fact, along with the high-energy of the beams determine the conceptual design of the detector, presented in this paper, together with the technical solutions proposed for its construction.

    • Database search is the most popular approach used for the identification of peptides in contemporary shotgun proteomics; it utilizes only mass spectrometric data. In this work, we introduce three criteria for the verification of peptide identification; these are based on the analysis of data orthogonal to tandem mass spectra. The first one utilizes chromatographic retention times of peptides. The development of such approaches has been hindered by the relatively low accuracy of retention time prediction algorithms. In this work, we suggest the use of two independent models of the liquid chromatography of peptides, which increase the reliability of the results. The second criterion utilizes the mean number of missed tryptic cleavages per peptide. The third one results from the analysis of the difference between theoretical and experimentally measured peptide masses. The proposed criteria were applied to the tandem mass spectra of tryptic peptides from rat kidney tissue, which were processed by the Mascot search engine. All the criteria showed that Mascot significantly overestimated the reliability of an identification. This conclusion was supported by the PeptideProphet algorithm.

    • Delayed emission of protons following β decay of neutron deficient nuclei P26 and S27 was investigated at the ACCULINNA separator in the Flerov Laboratory of Nuclear Reactions at Dubna. Ions of interest, identified in flight, were implanted into the active volume of the gaseous optical time projection chamber, which allowed us to record tracks of charged particles emitted in the decay. Total branching ratios for β-delayed proton emission and for β-delayed two-proton emission were determined. In addition, energy spectra for delayed protons below 2 MeV were established. Our findings for P26 agree with results of previous experiments. In the case of S27, however, the observed probability of delayed proton emission is an order of magnitude larger than reported in literature. Two new strong proton transitions were identified representing decays of the first two excited states of P27 to the ground state of Si26. The probability ratio of γ-to-proton emission from these states is discussed.

    • Experimental data on the separation of synthetic and natural peptides are presented as treated in terms of the separation model proposed by the authors, which allows for the chain connectivity of amino acid residues and the cooperative character of their interaction with the surface. It was shown that the model accurately predicts the separation of peptides with identical amino acid contents and different sequences of units in the chain. The differences in the sequence may be permutation of amino acid residues and the presence of terminal groups, amino acid isomers, or mirror sequences in the chain. The separation model was used to predict the retention times of peptides prepared via the enzymatic hydrolysis of E. coli proteins and bovine serum albumin with trypsin. It was shown that in general the model accurately explains the array of experimental data on the separation of such peptides, thus being the first successful attempt to relate the chain sequence to the retention volume.

    • The fusion-evaporation reaction 244Pu(48Ca, 3-4n)288,289114 was studied at the new gas-filled recoil separator TASCA. Thirteen correlated decay chains were observed and assigned to the production and decay of 288,289114. At a compound nucleus excitation energy of E* = 39.8-43.9 MeV, the 4n evaporation channel cross section was 9.8(-3.1)(+3.9) pb. At E* = 36.1-39.5 MeV, that of the 3n evaporation channel was 8.0-(+7.4)(4.5) pb. In one of the 3n evaporation channel decay chains, a previously unobserved alpha branch in 281Ds was observed ( probability to be of random origin from background: 0.1%). This alpha decay populated the new nucleus 277Hs, which decayed by spontaneous fission after a lifetime of 4.5 ms.

    • Experiments with the new recoil separator, Transactinide Separator and Chemistry Apparatus (TASCA), at the GSI were performed by using beams of Ca-48 to irradiate targets of Pb206-208, which led to the production of No252-254 isotopes. These studies allowed for evaluation of the performance of TASCA when coupled to a new detector and electronics system. By following these studies, the isotopes of element 114 ((288-291)114) were produced in irradiations of Pu-244 targets with Ca-48 beams at compound nucleus excitation energies around 41.7 and 37.5 MeV, demonstrating TASCA's ability to perform experiments with picobarn-level cross sections. A total of 15 decay chains were observed and were assigned to the decay of 288-291114. A new a-decay branch in (281)Ds was observed, leading to the new nucleus (277)Hs.

    • An approach to sequence-dependent retention time prediction of peptides based on the concept of liquid chromatography at critical conditions (LCCC) is presented. Within the LCCC approach applied to biopolymers (BioLCCC), the specific retention time corresponds to a particular sequence. In combination with mass spectrometry, this approach provides an efficient tool to solve problems wherein the protein sequencing is essential. In this work, we present a theoretical background of the BioLCCC concept and demonstrate experimentally its feasibility for sequence-dependent LC retention time prediction for peptides. BioLCCC model is based on three notions: ( a) a random walk model for a macromolecule chain; (b) an entropy and energy compensation for the macromolecules within the adsorbent pore; and ( c) a set of phenomenological parameters for the effective interaction energies of interactions between the amino acid residues and the adsorbent surface. In this work, the phenomenological parameters have been obtained for C-18 reversed-phase HPLC. Note, that contrary to alternative additive models for retention time prediction based on summation of the so-called "retention coefficients", the BioLCCC approach takes into account the location of amino acids within the primary structure of a peptide and, thus, allows the identification of the peptides having the same composition of amino acids but differing by their arrangement. As a result, this new approach allows prediction of retention time for any possible amino acid sequence in particular HPLC experiments. In addition, the BioLCCC model lacks of main drawbacks of additive approaches that predict retention time for sequences of limited chain lengths and provide information about amino acid composition only. The proposed BioLCCC approach was characterized experimentally using LTQ FT LC-MS and LC-MS/MS data obtained earlier for Escherichia coli. The HPLC system calibration was performed using peptide retention standards. The results received show a linear correlation between predicted and experimental retention times, with a correlation coefficient, R-2, of 0.97 for a peptide standard mixture and 0.9 for E. coli data, respectively, with the standard error below 1 min. The work presents the first description of a BioLCCC approach for high-throughput peptide characterization and preliminary results of its feasibility tests.

    • The deformed doubly magic nucleus (270)Hs has so far only been observed as the four-neutron (4n) evaporation residue of the reaction Mg-26+Cm-248, where a maximum cross section of 3 pb was measured. Theoretical studies on the formation of (270)Hs in the 4n evaporation channel of fusion reactions with different entrance channel asymmetry in the framework of a two-parameter Smoluchowski equation predict that the reactions Ca-48+Ra-226 and S-36+U-238 result in higher cross sections due to lower reaction Q values, in contrast to simple arguments based on the reaction asymmetry, which predict opposite trends. Calculations using HIVAP predict cross sections for the reaction S-36+U-238 that are similar to those of the Mg-26+Cm-248 reaction. Here, we report on the first measurement of evaporation residues formed in the complete nuclear fusion reaction S-36+U-238 and the observation of (270)Hs, which is produced in the 4n evaporation channel, with a measured cross section of 0.8(-0.7)(+2.6) pb at 51-MeV excitation energy. The one-event cross-section limits (68% confidence level) for the 3n, 4n, and 5n evaporation channels at 39-MeV excitation energy are 2.9 pb, while the cross-section limits of the 3n and 5n channel at 51 MeV are 1.5 pb. This is significantly lower than the 5n cross section of the Mg-26+Cm-248 reaction at similar excitation energy.

    • Generation of a complex proteome database requires use of powerful analytical methods capable of following rapid changes in the proteome due to changing physiological and pathological states of the organism under study. One of the promising technologies with this regard is the use of so-called Accurate Mass and Time (AMT) tag peptide databases. Generation of an AMT database for a complex proteome requires combined efforts by many research groups and laboratories, but the chromatography data resulting from these efforts are tied to the particular experimental conditions and, in general, are not transferable from one platform to another. In this work, we consider an approach to solve this problem that is based on the generation of a universal scale for the chromatography data using a multiple-point normalization method. The method follows from the concept of linear correlation between chromatography data obtained over a wide range of separation parameters. The method is further tested for tryptic peptide mixtures with experimental data collected from mutual studies by different independent research groups using different separation protocols and mass spectrometry data processing tools.

    • Amphibian skin secretion represents a cerain scientific interest as a source of biologically active natural peptides. In the present research skin peptidome of wide-spread European frog Rana lessonae (Camerano, 1882) was studied for the first time ever. Peptide sequencing was accomplished with Fourier transform ion cyclotron resonance mass spectrometer in collision-induced and electron capture dissociation modes. A portion of amphibian peptides contains intramolecular C-terminal disulfide cycle which obstructs mass spectrometric sequencing. Two methods were utilized to overcome this difficulty: reduction with dithiotreithol followed by thiol group alkylation and oxidation into sulfonic acid groups with performic acid. Integrated approach employed in the present study allowed the identification of 49 peptides (of 6 to 37 amino acid residues), including 19 novel species.

    • Skin secretions of amphibian are an interesting source of biologically active peptides. The present study provides the profile of the skin secretions of the brown frog Rana temporaria from Zvenigorod population (Russia). Sequencing of the skin secretion components has been carried out on an ion cyclotron resonance instrument with electrospray ionization and two methods of fragmentation activation, collisional activation and electron capture. For sequencing of the peptides containing intermolecular C-terminal disulfide cycle two methods of disulfide bond opening have been used: reduction with subsequent alkylation of the free thiol groups and oxidation with performic acid with the formation of sulfo-acid groups. The peptide profile of Rana temporaria studied by a complex mass spectral method has been compared with the data for the frogs of other European populations of this species. For the first time we have revealed ornithokinin-antagonist of the ornithokinin receptor-in skin secretions of amphibians.

    • Hyla arborea schelkownikowi is one of the leaf frog species inhabiting the southern territories of Russia and the former USSR. This frog species is a member of the Hylidae Rafinesque, 1815 batrachians family. The present study deals with the previously uninvestigated peptidome of the Hyla arborea schelkownikowi skin secretion. Nano-electrospray ionization Fourier transform mass spectrometry (nanoESI-FTMS) of the skin secretion, in the intact form and after acetylation, was selected as the general method of analysis. Electron-capture dissociation (ECD) and collision-induced dissociation (CID) fragmentation were both employed, while de novo sequencing was performed by manual interpretation of the MS data. The suppression of the cyclization of b-ions in the mass spectrometer by the acetylation reaction proved to be very efficient for the de novo sequencing of short peptides. Ten skin peptides were found and all of them, except for bradykinin, had not previously been reported. Six of the peptides belong to the tryptophyllins and related peptides, while three peptides are similar to the aureins.

    • Estimation of false discovery rate (FDR) for identified peptides is an important step in large-scale proteomic studies. We introduced an empirical approach to the problem that is based on the FDR-like functions of sets of peptide spectral matches (PSMs). These functions have close values for equal-sized sets with the same FDR and depend monotonically on the FDR of a set. We have found three of them, based on three complementary sources of data: chromatography, mass spectrometry, and sequences of identified peptides. Using a calibration on a set of putative correct PSMs these functions were converted into the FDR scale. The approach was tested on a set of similar to 2800 PSMs obtained from rat kidney tissue. The estimates based on all three data sources were rather consistent with each other as well as with one made using the target-decoy strategy.

    • The average charge states of Pb-188 and No-252,No-254 ions in dilute helium gas were measured at the gas-filled recoil separator TASCA. Hydrogen gas was also used as a filling gas for measurements of the average charge state of No-254. Helium and hydrogen gases at pressures from 0.2 mbar to 2.0 mbar were used. A strong dependence of the average charge state on the pressure of the filling gases was observed for both, helium and hydrogen. The influence of this dependence, classically attributed to the so-called "density effect", on the performance of TASCA was investigated. The average charge states of No-254 ions were also measured in mixtures of helium and hydrogen gases at low gas pressures around 1.0 mbar. From the experimental results simple expressions for the prediction of average charge states of heavy ions moving in rarefied helium gas, hydrogen gas, and in their mixture were derived.     • Identification of species constituting Rana esculenta complex represents a certain problem as two parental species Rana ridibunda and Rana lessonae form their hybrid R. esculenta, while external signs and sizes of the members of this complex are intersected. However the composition of skin secretion consisting mainly of peptides is different for the species of the complex. LC-MS/MS is an ideal analytical tool for the quantitative and qualitative analysis of these peptides. The results covering elemental composition of these peptides, their levels in the secretion, as well as their belonging to a certain family of peptides may be visualized by means of 2D mass maps. The proposed approach proved itself to be a perspective tool for the reliable identification of all 3 species constituting R. esculenta complex. Easy distinguishing between the species may be achieved using 2D maps as fingerprints. Besides this approach may be used to study hybridogenesis and mechanisms of hemiclonal transfer of genetic information, when rapid and reliable identification of species involved in the process is required.

Among the participants were scientists as well as developers working in the field of computational mass spectrometry (MS) and proteomics. The 4-day program was split between introductory keynote lectures and parallel hackathon sessions. During the latter, the participants developed bioinformatics tools and resources addressing outstanding needs in the community. The hackathons allowed less experienced participants to learn from more advanced computational MS experts, and to actively contribute to highly relevant research projects. We successfully produced several new tools that will be useful to the proteomics community by improving data analysis as well as facilitating future research.     • Amphibian skin secretion is known to contain biologically active peptides. Bradykinins and related peptides (BRPs) can be found in these animals, while frogs from the genus Rana are considered to be leaders in the levels and variety of these peptides. A reasonable rationalization of this fact is that bradykinins are efficient defense compounds against predators. Forty-four various BRPs have been identified in the skin secretions of five ranid frog species (R. ridibunda, R. lessonae, R. esculenta, R. temporaria, R. arvalis) from the Zvenigorod region (Moscow district, Russia). Some of these peptides are already known, but the novel ones constitute a significant portion. An interesting group of novel peptides was isolated from R. lessonae. These are bradykinin analogues bearing a tyrosine residue in the 5th or 8th position. [Arg(0), Trp(5), Leu(8)] bradykinin and [Thr(6), Leu(8)] bradykinin that had been isolated from fish and avian species, respectively, were also detected in the frog secretion, supporting the predator defense hypothesis. Furthermore, a novel group of BRPs named 'lessonakinins' was discovered in R. lessonae and R. esculenta. All of them include the [Arg(0), Trp(5), Leu(8)] bradykinin sequence and have some structural resemblance to the precursor of this peptide cloned by Chen and coworkers recently. However, the C-terminal part of the lessonakinins does not match the sequence predicted by Chen, demonstrating possible incompleteness of information obtained by cDNA cloning.

    • Amphibian skin glands are known to secrete various types of bioactive peptides. The array of these peptides is specific for every frog species. The present research deals with the identification of peptides isolated from the skin secretion of the Marsh frog R. ridibunda inhabiting the Kolkhida Canyon of the Caucasian region. The research is based on comprehensive high-performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS) analysis of intact and chemically modified peptides. In particular, an oxidation procedure was applied directly to the crude skin secretion to open S-S loops whereas N-terminal acetylation was additionally carried out for one individual peptide. Sequences were determined by manual interpretation of electron capture dissociation (ECD) and collisionally induced dissociation (CID) tandem mass spectra. A total of 29 peptides were identified in the skin secretion of the Caucasian Marsh frog. The peptide profile is represented with disulfide-containing peptides belonging to the brevinin, esculentin and ranatuerin families, neuropeptides of the bradykinin and bombesin families. Two identified peptides belonging to the ranatuerins are the first peptides of this family discovered in the skin secretions of European frogs. Ten of the identified peptides coincide with those reported earlier for the European Edible frog. Another ten are identical to those found in R. ridubunda from the Moscow region. This fact verifies the described method as being art efficient analytical tool to compare intra- and interspecific variabilities.

    • Tandem mass spectrometry sequencing, as well as Edman sequencing of peptides belonging to the Rana genus, represents a difficult task due to the presence of a disulfide bridge at the C-terminus and their rather high molecular masses (over 2000Da). The present study throws light upon the sequence of three rather long peptides (more than 20 amino acid residues each) isolated from the skin secretion of Russian frogs, Rana ridibunda and Rana arvalis. This novel aspect involves the fact that the sequences (including two sequences established de novo) were determined exclusively by means of mass spectrometry. A combination of electron capture dissociation (ECD) and collision-induced dissociaiton (CID) data accompanied by exact mass measurements (LTQ Fourier transform ion cyclotron resonance mass spectrometer) facilitated reaching the goal. To overcome the difficulty dealing with disulphide bridges ("Rana box"), reduction of the S-S bond with dithiotreitol followed by derivatization of Cys residues with iodoacetamide was used. The sequence was determined using combined spectral data on y and b series of fragment ions. A multiple mass spectrometry (MS') experiment was also used to elucidate the sequence inside the "Rana box" after cysteine derivatization. Exact mass measurements were used to differentiate between Lys and Gln residues, while characteristic losses of 29 and 43 Da (d and w fragment ions) in CID and ECD experiments allowed us to distinguish between Ile and Leu isomeric acids.

  • N-Terminal tagging strategy for De Novo sequencing of short peptides by ESI-MS/MS and MALDI-MS/MS

    • The major portion of skin secretory peptidome of the European Tree frog Hyla arborea consists of short peptides from tryptophyllin family. It is known that b-ions of these peptides undergo head-to-tail cyclization, forming a ring that can open, resulting in several linear forms. As a result, the spectrum contains multiple ion series, thus complicating de novo sequencing. This was observed in the Q-TOF spectrum of one of the tryptophyllins isolated from Hyla arborea; the sequence FLPFFP-NH2 was established by Edman degradation and counter-synthesis. Though no rearrangements were observed in FTICR-MS and MALDI-TOF/TOF spectra, both of them were not suitable for mass-spectrometry sequencing due to the low sequence coverage. To obtain full amino acid sequence by mass spectrometry, three chemical modifications to N-terminal amino moiety were applied. They include acetylation and sulfobenzoylation of N-amino group and its transformation to 2,4,6-trimethylpyridinium by interaction with 2,4,6-trimethylpyrillium tetrafluoroborate. All three reagents block scrambling and provide spectra better than the intact peptide. Unfortunately, all of them also readily react with lysine side chain. Hence, all investigated procedures can be used to improve sequencing of short peptides, while acetylation is the recommended one. It shows excellent results, and it is plain and simple to perform. This is the procedure of choice for MS-sequencing of short peptides by manual or automatic algorithms.
