    • Reliable taxonomy underpins communication in all of biology, not least nature conservation and sustainable use of ecosystem resources. The flexibility of taxonomic interpretations, however, presents a serious challenge for end-users of taxonomic concepts. Users need standardised and continuously harmonised taxonomic reference systems, as well as high-quality and complete taxonomic data sets, but these are generally lacking for non-specialists. The solution is in dynamic, expertly curated web-based taxonomic tools.The Pan-European Species-directories Infrastructure (PESI) worked to solve this key issue by providing a taxonomic e-infrastructure for Europe. It strengthened the relevant social (expertise) and information (standards, data and technical) capacities of five major community networks on taxonomic indexing in Europe, which is essential for proper biodiversity assessment and monitoring activities. The key objectives of PESI were: 1) standardisation in taxonomic reference systems, 2) enhancement of the quality and completeness of taxonomic data sets and 3) creation of integrated access to taxonomic information.This paper describes the results of PESI and its future prospects, including the involvement in major European biodiversity informatics initiatives and programs.

  • Importance of Thomas single-electron transfer in fast p-He collisions

    • We report experimental angular differential cross sections for nonradiative single-electron capture in p-He collisions (p + He -> H + He+) with a separate peak at the 0.47 mrad Thomas scattering angle for energies in the 1.3-12.5 MeV range. We find that the intensity of this peak scales with the projectile velocity as v(P)(-11). This constitutes the first experimental test of the prediction from 1927 by L. H. Thomas [Proc. R. Soc. 114, 561 (1927)]. At our highest energy, the peak at the Thomas angle contributes with 13.5% to the total integrated nonradiative single-electron capture cross section.

  • Two-Center Double-Capture Interference in Fast He2++H2 Collisions

    • We report the first observation of Young-type interference effects in a two-electron transfer process. These effects change strongly as the projectile velocity changes in fast (1.2 and 2.0 MeV) He^{2+}-H_2 collisions as manifested in strong variations of the double-electron capture rates with the H_2 orientation. This is consistent with fully quantum mechanical calculations, which ignore sequential electron transfer, and a simple projectile de Broglie wave picture assuming that two-electron transfer probabilities are higher in collisions where the projectile passes close to either one of the H_2 nuclei.

    • We report the direct observation of interference effects in a Young's double-slit experiment where the interfering waves are two spatially separated components of the de Broglie wave of single 1.3 MeV hydrogen atoms formed close to either target nucleus in H+ + H2 electron-transfer collisions. Quantum interference strongly influences the results even though the hydrogen atoms have a de Broglie wavelength, \lambda_dB, as small as 25 fm.

    • With the increasing incidence of prostate cancer, identifying common genetic variants that confer risk of the disease is important. Here we report such a variant on chromosome 8q24, a region initially identified through a study of Icelandic families. Allele -8 of the microsatellite DG8S737 was associated with prostate cancer in three case-control series of European ancestry from Iceland, Sweden and the US. The estimated odds ratio (OR) of the allele is 1.62 (P = 2.7 x 10(-11)). About 19% of affected men and 13% of the general population carry at least one copy, yielding a population attributable risk (PAR) of approximately 8%. The association was also replicated in an African American case-control group with a similar OR, in which 41% of affected individuals and 30% of the population are carriers. This leads to a greater estimated PAR (16%) that may contribute to higher incidence of prostate cancer in African American men than in men of European ancestry.

    • This following special issue of the European Journal of Transport Infrastructure Research (EJTIR) containing 4 scientific papers is the result of the work conducted under the research project 'National Transport Planning - Sustainability, Institutions and Tools' (SUSTAIN) (20122017) financed by the Danish Innovation Fund. SUSTAIN was coordinated by first the Department of Transport of the Technical University of Denmark (DTU Transport) and later as the result of a reorganisation of the transport research at DTU by the Department of Management Engineering (DTU Management Engineering). The project was carried out in cooperation with several Danish and international partners. In SUSTAIN the following definition of national sustainable transport planning (NSTP) was adopted: deliberate, knowledge-based, and strategic endeavours to integrate sustainability principles, criteria and goals in the development, management, regulation and assessment of nationally significant transport systems and services. This paper presents the research outcome by reviewing some of the major findings and seeing these collectively as a basis for promoting sustainability through the formulated research topic of NSTP. This basis includes defining criteria and indicators for use in NSTP combining evidence-based and performance oriented planning approaches. Furthermore, it contains a flexible evaluation framework that includes a decision support model that when informed by criteria and indicators can structure and assist an NSTP practice. This practice can support a planning process aiming at realising a sustainable transport development. Finally, the basis comprises a review of the complex political and administrative fabric in which NSTP is embedded, and thereby it can condition the ability to promote sustainability in practice.

    • Implementation is a fairly old concept but as relevant as ever. We have witnessed a general change of the public sector in the direction of governance, networks, contracting out, Public-Private Partnerships etc. This brings to the fore the following question: What has happened since the early 1970’s in theory and practice that might lead us to reconsider the ways we understand implementation in theory but also the advice we could offer to decision makers? To narrow down this broad question we choose to focus on the transport sector and more specifically on sustainable transport. We argue that sustainable transport is a particularly good case to discuss possible changes in implementation patterns.

    • Environmental manipulation studies are integral to determining biological consequences of climate warming. Open Top Chambers (OTCs) have been widely used to assess summer warming effects on terrestrial biota, with their effects during other seasons normally being given less attention even though chambers are often deployed year-round. In addition, their effects on temperature extremes and freeze-thaw events are poorly documented. To provide robust documentation of the microclimatic influences of OTCs throughout the year, we analysed temperature data from 20 studies distributed across polar and alpine regions. The effects of OTCs on mean temperature showed a large range (-0.9 to 2.1 degrees C) throughout the year, but did not differ significantly between studies. Increases in mean monthly and diurnal temperature were strongly related (R-2 = 0.70) with irradiance, indicating that PAR can be used to predict the mean warming effect of OTCs. Deeper snow trapped in OTCs also induced higher temperatures at soil/vegetation level. OTC-induced changes in the frequency of freeze-thaw events included an increase in autumn and decreases in spring and summer. Frequency of high-temperature events in OTCs increased in spring, summer and autumn compared with non-manipulated control plots. Frequency of low-temperature events was reduced by deeper snow accumulation and higher mean temperatures. The strong interactions identified between aspects of ambient environmental conditions and effects of OTCs suggest that a detailed knowledge of snow depth, temperature and irradiance levels enables us to predict how OTCs will modify the microclimate at a particular site and season. Such predictive power allows a better mechanistic understanding of observed biotic response to experimental warming studies and for more informed design of future experiments. However, a need remains to quantify OTC effects on water availability and wind speed (affecting, for example, drying rates and water stress) in combination with microclimate measurements at organism level.

    • This document represents the conceptual foundations of the EU-FP7 OPTIC project. As such, it seeks to provide a range of theoretical resources with which to develop an informed and pragmatic understanding of the complex causal processes involved in contemporary transport policy-making at the European level. Specifically, this deliverable aims to further methodological advancement with respect to the identification, classification, ex-ante prevention and ex-post mitigation of policies' unintended effects, and the systematic manner in which individual policy measures may be combined so as to improve their effectiveness, acceptability and feasibility. Overall, we argue that policy packaging can offer a far greater potential for achieving policy targets and objectives than single policy measures deployed in isolation. Yet, a careful and relatively well designed process must be undertaken for such packages to be effective.

  • Metastatic potential in renal cell carcinomas <= 7 cm : swedish kidney cancer quality register data

    • Background: Renal cell carcinoma(RCC) represents 2-3% of all malignancies and accounts for approximately 90% of all kidney malignancies. An increasing proportion of RCCs are discovered incidentally, and the average tumor diameter at diagnosis has decreased over the last few decades. Small RCCs have often been regarded by many as relatively harmless. Objective: The objective was to evaluate the incidence of local T-category distribution and lymph node and distant metastases in relation to tumor size in RCCs <= 7 cm in a nationally based patient population. Design, setting, and participants: Data were extracted from the National Swedish Kidney Cancer Register containing 3489 RCCs diagnosed between 2005 and 2008. This is a population-based registry including 99% of all RCCs diagnosed nationwide. The study included 2033 patients having a tumor <= 7 cm in diameter. Measurements: The size of the tumors was compared with sex, age, cause of diagnosis, Fuhrman grade, RCC type, and TNM category. Results and limitations: Most RCCs were discovered incidentally and incidence correlated inversely to tumor size. There were 887 (43%) patients with category T1a tumors, 836 (40%) with category T1b, 174 (8%) with T3a, 131 (6%) with T3b/c, and 12 (1%) patients had invasion of adjacent organs (T4). A total of 309 (15%) patients had lymph node and/or distant metastases. Of the 177 1- to 2-cm RCCs, category T3 tumors were identified in three patients and lymph node and/or distant metastases were identified in 8 (5%). Only for tumors <= 1 cm was there neither advanced stage nor metastasis. The occurrence of locally advanced growth, lymph node and distant metastases, and high tumor grade correlated to tumor size. Patients with Fuhrman grade III or IV had a fourfold greater risk of metastases than grades I or II. Conclusions: Lymph node and distant metastases occur even in small RCCs. Risk of metastases increases with tumor size. The data clearly show that small RCCs also have a malignant potential and should be properly evaluated and adequately treated.     • In the 2011 Transport Policy White Paper, the European Commission introduced ten goals for a more competitive and resource efficient transport system. One of the goal concerns urban transport, with the dual targets: “To halve the use of ‘conventionally-fuelled’ cars in urban transport by 2030; and to phase them out by 2050; and “To achieve ‘essentially CO2- free city logistics’ in major urban centres by 2030.” This paper describes how the EU FP7 project TRANSFORUM conducted a stakeholder-driven process to produce a roadmap for the urban transport goal.A main conclusion was that the implementation of a roadmap for urban transport needs to adopt a broad and open approach, given the diversity among member states and cities across Europe. Also it was found that replacing conventional vehicles and fuels is an important but not sufficient strategy to reach the goal. A roadmap to reach towards the ‘halving’ goal would need to engage many cities in shared political commitments to generate a stronger momentum for change. The notion of a ‘Covenant of Halflings’ would be one way to express this need.

    • In its 2011 Transport Policy White Paper, the European Commission introduced ten targets to be met in order to reach a more competitive and resource efficient transport system. The following dual goal focused on urban transport and commuting: “To halve the use of ‘conventionally-fuelled’ cars in urban transport by 2030; and to phase them out by 2050; and to achieve “essentially CO2-free city logistics in major urban centres by 2030.” The question driving the research behind this paper is how to realise this dual urban transport goal for 2030, taking into consideration existing trends, available policy options, the diverse interests of stakeholders involved in urban mobility in Europe.The paper draws on data gathered in the EU FP7 TRANSFORuM project, including workshop dialogues with stakeholders representing different interests in and perspectives on urban transport and mobility. The main result is a roadmap that proposes a broad strategy to answer the question “Who has to do what, by when” to achieve the urban transport goal. The roadmap includes detailed proposals for action and milestones at different levels of decision making. This paper will describe the process of consultations and the outcomes of the roadmap process and will also discuss the perspectives for implementation.

  • Framing the role of Decision Support in the case of Stockholm Congestion Charging Trial

    • Congestion charging was - as a trial - introduced in Stockholm from January 3rd to July 31st 2006. After the referendum in September 2006, the charging system was finally introduced as permanent from August 2007 with some adjustments to the Trial design. The idea of congestion charging is unique in a Swedish context. and the introduction of the Stockholm system has been highly controversial. Considerable efforts have therefore been undertaken to provide information that could serve as 'Decision Support' along the way. This has included e.g. modelling and forecasts before the Trial, a comprehensive evaluation programme during the Trial, extensive stakeholder consultations throughout, and various information and communication strategies. But what difference did this information input make, and what was its role in the process from initiating the system, to its final adoption? In this paper we pave the way for investigating the use and role of 'Decision Support' in the Stockholm Congestion Charging experiment. We adopt a definition of Decision Support as the systematic application of externally produced knowledge in transport planning and policy making processes. We then derive an analytical framework from the research literature on 'knowledge utilization' in policy making. This research has generally found that both 'technical', 'communicative', and 'institutional' aspects of the Decision Support matter for its influence on actual policy making processes and results. In our analysis we find a similar pattern. This high technical quality of the monitoring and evaluation programmes provided for solid verified results, while the institutional arrangements and the communication strategies helped to ensure the credibility and legitimacy of the information for the decision makers. The availability of rich contents coupled with strategies for the timely and targeted information delivery suggest that direct 'instrumental' use could have taken place. At a more general level the Trial represents an advanced form of 'Decision Support' that goes beyond the mere application of calculated results to encompass a process where the decision parameters themselves become part of the change process.     • A wide variety of policy measures and interventions can be applied in order to promote efficient, accessible and sustainable transport systems. However it is widely recognized that transport systems are complex and their performance depends on multiple factors. It is therefore a significant challenge to ensure that policies actually lead towards their intended objectives, and to avoid unanticipated negative side-effects within and beyond the transport sector. One way to approach this challenge is to adopt integrated policy packages where measures supplement each other and compensate for expected negative effects of individual measures. A key element in a systematic approach for policy packaging must be to enable the identification in advance of non-intentional effects of individual or joint policy measures. This paper develops a typology of non-intentional effects of transport policy measures as part of a methodology for policy packaging in the OPTIC research project. The paper first establishes the need to address systematically unintended effects and then proceeds to unfold the dimensions of the typology with concrete examples along the way. Ways to use the typology is discussed in the final section of the paper.

  • Some use - Little influence? : On the roles of indicators in European sustainable transport policy

    • The paper focuses on the use and influence of indicators in European sustainable transport policy from a ‘knowledge utilization’ research point of view. The starting point is the contrast between the widely held idea that indicators are important tools for sustainable transport policy making, versus ‘knowledge utilization’ oriented research, which has often demonstrated that formal technical knowledge is used much less, or at least differently, than expected in policy and decision making.The paper looks at two cases of indicators applied for strategic policy making within the sustainable transport agenda. The first case concerns indicators tracking the fulfillment of national transport policy objectives in Sweden. The case explores the use and influence of an annual monitoring and evaluation report produced for this purpose, within a general ‘Management-by-Objectives’ regime. The second case deals with indicators applied for the Mid-Term Review of the European Commission's transport policy White Paper ‘Time to Decide’ (issued in 2001). The focus in this case is on a particular indicator based study conducted in 2005 by a group of consultants in close collaboration with European Commission staff.It is found that several indicators in both cases are actually used in policy processes, with evidence of use found in documents as well as interviews. However, ‘use’ does not automatically mean ‘influence’ on policies or processes in more than a superficial manner. Indicators seem to play a very limited direct instrumental role, while some signs of symbolic, conceptual and process roles are found. In the EU case we identify what is called a rationalization role of indicators, meaning that indicators inform and help to rationalize a change in the position towards key objectives. Several factors that characterize the indicators, the users, and policy context are found to contribute to understand the type of use and influence observed in both cases.

Vi arbejder med operationalisering af bæredygtighedsbegrebet, opstilling af værktøjer til at analysere transportpolitiske projekter og strategier, samt analyser af den faktiske transportpolitik. I artiklen beskrives et par af de emner vi i øjeblikket arbejder med i projektet, nemlig indikatorer og beslutningsstøtte. Vi kommenterer også kort på Transportministeriets ”strategiske analyser”, som bidrager med spændende resultater, men ikke rigtig besvarer om der opnås bæredygtige fremskridt. Projektet løber til 2016.

  • Sustainable development principles and their implications for transport

    • 'Sustainable mobility' has been adopted as an overall objective for European transport policy, and similar intentions are expressed in other parts of the world. However, little has been done to define what 'sustainable mobility' would actually imply for our understanding and assessment of transport. We suspect that this much used term could merely end up acting as a lubricant to the very development it was meant to challenge: The ever increasing movement of people and goods. In this article we attempt to place transport in the context of sustainable development principles. We emphasise the need to include principles of development (increasing well-being and equity) as well as sustainability (preserving natural and man-made capital). Four such principles are suggested, taking inspiration from Herman Daly and others. We then turn to explore the main features of transport, establishing a comprehensive transport concept. The concept combines a systems perspective with a service perspective. Omitting either one would disable a complete consideration of sustainable development. Our confrontation of the four principles with the reality of current transport trends gives rise to several critical implications. The most important of which relate to the contribution of transport to depleting natural capital and quality of life. Moreover, we are also forced to challenge the value of increasing mobility itself, if other forms of access may provide relevant substitutes.     • The paper focuses on the use and influence of indicators in European sustainable transport policy from a knowledge utilization research point of view, using a qualitative approach. The starting point is the contrast between a general proposition that indicators are important tools for sustainable transport policy making, and utilization oriented research literature, which has often demonstrated that formal technical knowledge is used much less, or at least differently, than expected in policy and decision making.The paper looks at two cases of indicators applied for strategic policy making within the sustainable transport agenda. The first case concerns indicators tracking the fulfillment of national transport policy objectives in Sweden. The case explores the use and influence of an annual monitoring and evaluation report produced for this purpose. The second case deals with indicators applied for the Mid-Term Review of the European Commission.s transport policy White Paper „Time to Decide. We focus on use and influence of the indicator based study ASSESS conducted in 2005.It is found that several indicators are in fact used in policy processes, as evidenced in policy documents as well as through interviews. However, use does not automatically mean influence on policies or processes in more than the most superficial manner. Indicators seem to play an – at most - limited instrumental role in the studied policy cases while some signs of symbolic, conceptual and process roles are found. In the EU case some evidence of what we call a rationalization role of indicators is found, meaning that indicators inform and help to rationalize a change in the European Commissions position towards key objectives, and gain influence in this way. Factors that characterize the indicators, the users, the policy setting and the economic and political background may all contribute to explaining the type of use and influence observed in both cases.

  • Angular scattering in fast ion-atom electron transfer collisions : projectile wave diffraction and Thomas mechanisms

    • We report experimental angular differential cross sections for double-electron capture in He2+ + He collisions and single-electron capture in H+ + He collisions for the 1.3-12.5 MeV kinetic energy range. In all cases, the total cross sections are dominated by forward scattering peaks in d sigma/d Omega. The shapes and widths (but not the magnitudes) of these peaks are very similar for all energies and for capture of one or two electrons corresponding also to our measured linear increases in the transverse momentum transfers with increasing projectile velocities. These observations may be ascribed to diffraction limitations which are connected to electron transfer probabilities P(b) which are significant in limited regions of b only. For the H+ + He single-electron capture we observe two additional maxima in the angular differential cross sections. We conclude that while the secondary maxima at similar to 0.5 mrad probably have large contributions from the Thomas proton-electron-nucleus scattering mechanism, the third maxima at similar to 0.75 mrad are most likely mainly due to projectile de Broglie wave diffraction.

  • Heavy particle interference and diffraction in fast electron transfer collisions

    • This thesis presents experimental results from the synchrotron cooler and storage ring CRYRING on charge transfer processes in fast electron transfer collisions using high-resolution cold target recoil-ion momentum spectroscopy. The main focus of these studies was to investigate a key concept of quantum mechanics: the wave-particle duality. One series of experiments has been dedicated to the study of heavy particle de Broglie wave interference due to scattering on a molecular ’double slit’. This is a fundamental manifestation of the wave properties of matter. Quantum interference oscillations were observed in the target orientation dependent cross section of single- and double-electron capture from H2 to 1.3 MeV protons and to 1.2 and 2.0 MeV He2+ ions. Another study, included in this work, is a series of angular differential cross section measurements for single-electron capture to 1.3-12.5 MeV kinetic energy protons from He that enabled us to systematically investigate the classically allowed non-radiative electron capture process in fast collisions predicted by L. H. Thomas in 1927. The cross section for this process is expected to have a nonrelativistic, asymptotic dependence on the projectile velocity, vp, of vp−11. This prediction (from 1927) was verified experimentally for the first time through the present measurements. Using the above mentioned experimental data in addition to measurements of double electron capture by 6.0 MeV He2+ from He, we have also studied the dominating, central part of the angular differential cross section, dσ/dΩ, where the peak shapes and widths surprisingly are very similar regardless of projectile energy and the number of captured electrons. We explain this with a diffraction model for the electron capture and calculate the corresponding diffracting electron capture ‘apertures’ from the shapes and widths of the measured cross sections and the projectile de Broglie wavelengths. We have on one hand established very strong experimental support for the picture suggested by Thomas in 1927 in which electrons and protons are described as classical particles. On the other hand, the diffraction picture describes the shapes of the central peaks in dσ/dΩ quite well, and nicely explains appearances of second and a third maxima in the angular differential cross section. It is hard to see how these seemingly contradicting results can be explained through complementary classical and quantum descriptions of the same underlying physical processes.

  • Kinetic and molecular dynamics study of inhibition and transglycosylation in Hypocrea jecorina family 3 beta-glucosidases

    • -Glucosidases enhance enzymatic biomass conversion by relieving cellobiose inhibition of endoglucanases and cellobiohydrolases. However, the susceptibility of these enzymes to inhibition and transglycosylation at high glucose or cellobiose concentrations severely limits their activity and, consequently, the overall efficiency of enzyme mixtures. We determined the impact of these two processes on the hydrolytic activity of the industrially relevant family 3 -glucosidases from Hypocrea jecorina, HjCel3A and HjCel3B, and investigated the underlying molecular mechanisms through kinetic studies, binding free energy calculations, and molecular dynamics (MD) simulations. HjCel3B had a 7-fold higher specificity for cellobiose than HjCel3A but greater tendency for glucose inhibition. Energy decomposition analysis indicated that cellobiose has relatively weak electrostatic interactions with binding site residues, allowing it to be easily displaced by glucose and free to inhibit other hydrolytic enzymes. HjCel3A is, thus, preferable as an industrial -glucosidase despite its lower activity caused by transglycosylation. This competing pathway to hydrolysis arises from binding of glucose or cellobiose at the product site after formation of the glycosyl-enzyme intermediate. MD simulations revealed that binding is facilitated by hydrophobic interactions with Trp-37, Phe-260, and Tyr-443. Targeting these aromatic residues for mutation to reduce substrate affinity at the product site would therefore potentially mitigate transglycosidic activity. Engineering improved variants of HjCel3A and other structurally similar -glucosidases would have a significant economic effect on enzymatic biomass conversion in terms of yield and production cost as the process can be consequently conducted at higher substrate loadings.

    • The filamentous fungus Hypocrea jecorina produces a number of cellulases and hemicellulases that act in a concerted fashion on biomass and degrade it into monomeric or oligomeric sugars. beta-Glucosidases are involved in the last step of the degradation of cellulosic biomass and hydrolyse the beta-glycosidic linkage between two adjacent molecules in dimers and oligomers of glucose. In this study, it is shown that substituting the beta-glucosidase from H. jecorina (HjCel3A) with the beta-glucosidase Cel3A from the thermophilic fungus Rasamsonia emersonii (ReCel3A) in enzyme mixtures results in increased efficiency in the saccharification of lignocellulosic materials. Biochemical characterization of ReCel3A, heterologously produced in H. jecorina, reveals a preference for disaccharide substrates over longer gluco-oligosaccharides. Crystallographic studies of ReCel3A revealed a highly N-glycosylated three-domain dimeric protein, as has been observed previously for glycoside hydrolase family 3 beta-glucosidases. The increased thermal stability and saccharification yield and the superior biochemical characteristics of ReCel3A compared with HjCel3A and mixtures containing HjCel3A make ReCel3A an excellent candidate for addition to enzyme mixtures designed to operate at higher temperatures.

    • We report on an experimental investigation of 1.04 MeV H++N2 electron transfer collisions. The fast protons were stored in the electron-cooler ion-storage ring, CRYRING and the molecular nitrogen target was provided with a supersonic gas jet. We report momentum distributions of atomic nitrogen dissociation products Nq+ with charge states q+ (q=1, 2, 3) which are detected in coincidence with neutralized projectiles. Further, we investigate the influence of the angle between the direction of the incoming projectile beam and the target molecular axis. The orientation of the latter is determined a posteriorly from the momentum vector of one emitted atomic nitrogen fragment ion. We find signicantly higher total yields, dominated by N+, of charged atomic dissociation products when the N2 molecular axis is perpendicular to the incoming H+-beam. The relative contributions from N2+- and N3+- fragments, however, are strongest when the N2 axis is aligned -or close to aligned- with the ion beam. This, we suggest, is due to increased probabilities for formation of two-site double-core-hole states.

    • In light of our previous observation of higher levels of cerebrospinal fluid (CSF) amyloid beta-42 (Aβ42) and CSF/serum albumin ratio in major depressive disorder (MDD), we analyzed two additional CSF biomarkers reflecting neurodegeneration—neurofilament protein light (NFL) and glial fibrillary acidic protein (GFAp)—in relationship to prevalent geriatric depression. Neuropsychiatric, physical, and lumbar puncture examinations, with DSM-III-R-based depression diagnoses and measurement of CSF levels of NFL and GFAp, were evaluated among a population-based sample of 78 elderly women (mean age, 73.9±3.2 years) without dementia for at least 10 years after CSF collection. Eleven (13.1%) women had MDD, and higher levels of NFL compared with women without depression. A multivariate model including age, NFL, Aβ42 and the CSF/serum albumin ratio showed that each biomarker was independently and positively associated with MDD, and that this biomarker profile explained more variation in the model compared with single or combined biomarkers. A CSF profile with higher levels of NFL, Aβ42, and CSF/serum albumin ratio may indicate neuropathological and vascular events in depression etiology. This contrasts with the well-characterized pattern of low Aβ42, higher CSF/serum albumin ratio, and higher NFL in Alzheimer's disease.

The conditional distribution of the underlying process given that the rare event occurs has the probability of the rare event as its normalizing constant. Using the MCMC methodology, a Markov chain is simulated, with the aforementioned conditional distribution as its invariant distribution, and information about the normalizing constant is extracted from its trajectory. The algorithm is described in full generality and applied to the problem of computing the probability that a heavy-tailed random walk exceeds a high threshold. An unbiased estimator of the reciprocal probability is constructed whose normalized variance vanishes asymptotically. The algorithm is extended to random sums and its performance is illustrated numerically and compared to existing importance sampling algorithms.

It is well known that standard methods of simulation are inefficient for computing rare-event probabilities and there- fore more advanced methods are needed to those problems.This thesis presents a new method based on Markov chain Monte Carlo (MCMC) algorithm to effectively compute the probability of a rare event. The assumption of heavy-tails allows us to propose distributions which approximate the conditional distribution conditioned on the rare event. The first problem considers a random walk Y1 + · · · + Yn exceeding a high threshold, where the increments Y are independent and identically distributed and heavy-tailed. The second problem is an extension of the first one to a heavy-tailed random sum Y1+···+YN exceeding a high threshold,where the number of increments N is random and independent of Y1 , Y2 , . . .. The third problem considers the solution Xm to a stochastic recurrence equation, Xm = AmXm−1 + Bm, exceeding a high threshold, where the innovations B are independent and identically distributed and heavy-tailed and the multipliers A satisfy a moment condition. The fourth problem is closely related to the third and considers the ruin probability for an insurance company with risky investments.In last two papers of this thesis, the algorithm is extended to the context of light- tailed distributions and applied to four problems. The light-tail assumption ensures the existence of a large deviation principle or Laplace principle, which in turn allows us to propose distributions which approximate the conditional distribution conditioned on the rare event. The first problem considers a random walk Y1 + · · · + Yn exceeding a high threshold, where the increments Y are independent and identically distributed and light-tailed. The second problem considers a discrete-time Markov chains and the computation of general expectation, of its sample path, related to rare-events. The third problem extends the the discrete-time setting to Markov chains in continuous- time. The fourth problem is closely related to the third and considers a birth-and-death process with spatial intensities and the computation of first passage probabilities.An unbiased estimator of the reciprocal probability for each corresponding prob- lem is constructed with efficient rare-event properties. The algorithms are illustrated numerically and compared to existing importance sampling algorithms.

    • During the past 15 – 20 years large parts of the public sector in the OECD countries but also outside the OECD have experienced so-called New Public Management (NPM) reforms. Not least the transport sector, and in particular public transport have been objects of these reforms. Important elements have been: Division of previously horizontal integrated organisations, corporatisation, privatisation, public-private partnerships, tendering and management by goals and results. The main purpose of the NPM reforms has been to increase the efficiency, effectiveness and accountability of public management. But taking a broader view on public transport policy, these are not the only things that matter. In our present work we are looking into the consequences of the NPM reforms in public transport for the realisation of Sustainable Transport objectives.Our basic assumption is that ‘organisation matters’ and that the reforms may have changed significantly the institutional conditions for the implementation of sustainable transport policies and easures. We have focused the research on public transport in larger cities and how their organisation has changed due to the reforms. We will address two different components of the NPM inspired transport reforms in the UK. The first component regards the organisation of public bus transport at the local level. This involves the deregulation and privatisation reforms introduced by the Conservative Government from 1986 onwards, as well as the ‘partnerships’ and other new instruments introduced by the Labour Government in the Transport Act 2000, with the aim to modify some extreme effects of the initial deregulations. The second component includes reforms of how central government governs transport performance at the local level more generally through systems of Management by Objectives, performance control and economic incentives. Key elements here are national policies such as the ’10 year Plan for Transport’ with associated targets, and the ‘Local Transport Plan’ regime with annual funding settlements, etc.The objective of the work carried out is to analyse which conditions the reforms create for attracting travellers from cars and for greening the bus fleets and fuels. Methodology and work carried out The empirical material includes literature on the bus industry, NPM reforms, and governance structures at the local level in the United Kingdom, as well as policy documents and reports on these topics. Moreover, 13 qualitative research interviews have been carried out in autumn 2007. The interviews involved management and staff at the Greater Manchester Passenger Transport Executive, GMPTE (eight persons); Greater Manchester Passenger Transport Authority, GMPTA (one officer, one politician); as well as operators and an association of operators (three persons).

    • I de senere år er der i dansk transportpolitik sendt signaler om en mere sammenhængende og langsigtet planlægning. Der opereres også med en række principper, som har karakter af målsætninger. Sverige og Norge kan måske inspirere, da de to lande i en årrække har arbejdet med store, nationale transportplaner baseret på transportpolitiske mål og tilhørende indikatorer. Her analyseres de svenske og norske erfaringer, og vi spørger: Hvilke elementer kunne med fordel anvendes i Danmark? Vi skelner i artiklen mellem ”målorienteret politik”, ”målorienteret forvaltning” samt ”målstyring”, som betragtes som en instrumentel kobling mellem de to første fænomener. Konklusionen er, at de transportpolitiske mål i politik og forvaltning er dekoblede i de to nabolande, og at der ikke er tale om målstyring, hvilket er meget vanskeligt at realisere. Ikke desto mindre er der elementer i målorienteret politik og målorienteret forvaltning, som kunne have betydning og værdi for dansk transportpolitik.

A definition is provided and it is suggested that three interlinked dimensions are of importance for transitions, thus a normative, an analytic and a governance dimension.The definition of national sustainable transport planning is confronted with current national transport planning practices in Sweden and Norway, which are somewhat advanced and have long traditions of recurrent, comprehensive, cross modal planning processes and integrated documents. Nevertheless, it is found that the Swedish and Norwegian planning efforts do not qualify to the label of ‘national sustainable transport planning.Finally three research topics for future research in national sustainable transport planning are proposed, which all link to the above mentioned dimensions and their interlinkages.

    • “Sustainable transport” has become a headline for transport planning and policy making around the world. While there are many different definitions of what Sustainable Transport means, it commonly includes efforts to improve the environmental performance of transport systems, and to promote alternative modes of transport to private cars, such as public transport. In this paper we focus only on the potential of bus transport services in cities for attracting travellers from cars. It is a basic assumption behind this paper that the ability of public bus transport to attract travellers from cars depends on how the bus sector is organised. By organisation we adopt an institutional point of view and consider basic modes of governance. Basic modes of governance include the forms “market”, “hierarchy” and “network” (Powell 1990). So-called New Public Management (NPM) reforms have changed the modes of governance of bus transport in several countries, including Denmark and the United Kingdom. Our focus is on how New Public Mangement Reforms have changed the mix of governance modes and thus may have influenced the ability of urban bus transport to attract travellers from cars. We focus on a particular case: Public bus transport in England, exemplified by Greater Manchester. England represents a situation where - compared to continental Europe including Denmark - the NPM reforms have been particularly radical. In no other European country the market forces has achieved a role as dominant as in England (outside London). We therefore assume that it is relevant to investigate the influence from the NPM reforms in this case, in order to discuss how the change in governance modes more generally enable or constrain the delivery of more sustainable transport patterns.

    • 'Sustainable transport' has become a priority for transport planning and policy making around the world. Sustainable transport plans often promote efforts to shift passengers from private cars to other modes such as public transport. However, the actual success of such efforts is likely to depend on how the transport sector is organised and governed. In this paper, we study the impacts of new public management (NPM) reforms in the British local transport sector on the attraction of passengers to buses. Britain is an interesting example since high level sustainable transport policies have been pursued in a deregulated context. We focus on bus transport in Greater Manchester as the case in point. First, we study the effects of the NPM reforms on modal shift. We find that the reforms generally have contributed to a decline in bus passengers, while some reform elements have made positive contributions. Second, we apply theoretical notions of 'governance modes', to examine whether the strengths and failures of 'market', 'hierarchy' and 'network' governance respectively can help to explain the results we observe. We find that these concepts are particularly useful to clarify the conditions under which public transport can attracts travellers.
