  • The unbounded body of the law of rape : the intrusive criterion of non-consent
  • 2018
  • Ingår i: Responsible selves. - : Routledge. - 9781315193656 - 075462160X - 9780815376514 ; , s. 331-352
  • Bokkapitel (refereegranskat)abstract
    • In the judicial context, the law of rape is plied chiefly around two central concepts: force and non-consent. These concepts are analogues to male and female sexuality and lie at the heart of what may be described as the dichotomy of rape law. When force is part of the legislative definition of rape, non-consent seems to be used in case law as an instrument to decide whether force may be proven. On the contrary, when the legislative definition contains non-consent, force is generally looked at in order to prove non-consent.

  • Environmental Impacts of ICT: Present and Future
  • 2016
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • ICT is developing rapidly and is playing an increasingly important role in society. High expectations are placed on ICT in relation to sustainable development. In order to provide basis for decision-making and ensure that ICT is used in the best possible way for enabling sustainable development, the sustainability impacts of ICT need to be studied. This thesis aims to provide new knowledge on the environmental impacts related to ICT, to explore the potential of ICT to contribute to sustainability, and discuss ways of assessing environmental impacts of ICT. In order to fulfill the aim a literature review of existing LCA studies of ICT was done, an LCA case study of printed and online media was performed, a methodological framework for sustainability assessment of scenarios was developed and then applied for environmental assessment of future ICT societies. The results show that manufacturing and use phase are the life cycle stages contributing the most to the ICT environmental impacts. For online newspapers online distribution and content production may give significant contribution to the overall impact. User behavior was observed to be crucial for the results of comparisons of ICT solutions with their traditional counterparts. The following key issues were concluded to influence the environmental risks and opportunities in future ICT societies: energy mix, economic conditions, life styles, technology, and environmental ambitions, incentives and regulation. The potential of ICT for sustainability is affected by these key issues.A new methodological framework (SAFS) was developed for the assessment of future scenarios (societal level). Life cycle assessment (LCA) was used for assessment on a product level. Application of both methods, their benefits, drawbacks, and challenges of assessment were discussed. Both types of assessments were concluded to be important to support decision-making.

Men hur fungerar miljöbedömning i planeringen?

  • Peripheral Monocyte Expression of the Chemokine Receptors CCR2, CCR5 and CXCR3 is Altered at Parturition in Healthy Women and in Women with Systemic Lupus Erythematosus
  • 2013
  • Ingår i: Scandinavian Journal of Immunology. - : Wiley. - 0300-9475 .- 1365-3083. ; 77:3, s. 200-212
  • Tidskriftsartikel (refereegranskat)abstract
    • Monocytes are precursors of macrophages and recruited to the uterus throughout pregnancy to perform important immunological functions. In this study, we hypothesized that pregnant women have reduced peripheral monocyte expression of chemokine receptors and alterations in PBMC responses to microbial stimuli as an adaption to pregnancy and that these changes are less pronounced in women with autoimmunity. We therefore investigated the chemokine receptor expression, migratory behaviour and responses to microbial stimulation of peripheral monocytes from pregnant women at parturition (n=13) and from non-pregnant women (n=9). In addition, we compared healthy pregnant women with women suffering from SLE (n=5), a condition with pronounced systemic inflammation increasing the risk for pregnancy complications. We demonstrate that peripheral monocytes are affected by pregnancy with reduced percentages of CCR2+, CCR5+ and CXCR3+ monocytes of both classical (CD16) and inflammatory (CD16+) subsets and that the trophoblast-secreted chemokine CCL2/MCP-1 recruited monocytes of both subsets in vitro. Further, PBMCs from pregnant women had a divergent response to microbial stimulation with lower CCL5/RANTES and higher CCL2/MCP-1 secretion compared with non-pregnant women. In addition, pregnant women had lower basal PBMC-secretion of CCL5/RANTES and higher basal secretion of IL-10 and CCL2/MCP-1. Interestingly, the women with SLE responded similar to pregnancy as did healthy women with lower percentages of CCR2+, CCR5+ and CXCR3+ monocytes. However, they had increased expression of CCR5 on CD16+ monocytes and heightened PBMC-secretion of CCL5/RANTES. In conclusion, our data indicate that monocyte chemokine receptor expression and the chemokine milieu during pregnancy are tightly regulated to support pregnancy.

  • Connexin 26 (GJB2) mutations in two Swedish patients with atypical Vohwinkel (mutilating keratoderma plus deafness) and KID syndrome both extensively treated with acitretin
  • 2006
  • Ingår i: Acta Dermato-Venereologica. - : Medical Journals Sweden AB. - 0001-5555 .- 1651-2057. ; 86:6, s. 503-508
  • Tidskriftsartikel (refereegranskat)abstract
    • Neuroectodermal syndromes involving the skin and inner ear may be associated with mutations in connexin proteins, which form gap junctions important for intercellular communication. Vohwinkel syndrome (keratodermia mutilans with hearing loss) and keratitis-ichthyosis-deafness (KID) syndrome are rare ectodermal dysplasias associated with dominant mutations in the GJB2 gene encoding connexin 26. We report here two patients, one with KID and one with Vohwinkel syndrome. Both displayed unusual clinical features and responded well to long-term treatment with oral retinoid. Mutation analysis revealed a novel GJB2 mutation p.Gly59Ser in the patient with Vohwinkel syndrome, whereas a recurrent mutation (p.Asp50Asn) was found in the patient with KID syndrome. The clinical features, particularly a proneness to skin cancer in the patient with Vohwinkel syndrome, are discussed in relation to the identified genotypes.

  • Exploring environmental justice in Sweden : How to improve planning for environmental sustainability and social equity in an “eco-friendly” context
  • 2008
  • Ingår i: Projections. - 1934-9688 .- 1934-9696. ; 8, s. 68-81
  • Tidskriftsartikel (refereegranskat)abstract
    • Environmental challenges, especially climate change, are highly discussed topics in the Swedish public debate, but questions about who is causing the problems and who is affected by them are seldom asked. This also applies to questions of who defines what should be regarded as acute environmental problems and what constitutes high-quality environments. This paper explores how environmental (in)justice issues can be framed in a Swedish social context, drawing from three cases: municipal promotion of eco-friendly lifestyles, large-scale infrastructure planning, and planners’ attitudes towards justice. The three cases deal not only with distributional, procedural, and substantive aspects of justice, as is common within the US environmental justice framework, but also with discursive dimensions of justice. We argue that elucidating such examples of environmental (in)justices is crucial to nuance the mainstream, consensus-oriented sustainability discourse in Sweden.

  • Miljörättvisa : ett nytt perspektiv i svensk planering
  • 2007
  • Ingår i: PLAN – Tidskrift för Samhällplanering. - 0032-0560. ; 3

  • 2007
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Men’s physical and psychological violence against women in intimate relationships is a serious social problem and contravenes the political goal of gender equality. Over the last few decades men’s violence against women has become a central issue in Swedish gender equality politics. Criminal law reforms have been carried through with the objective of enhancing criminal legal protection for women and promoting gender equality. The starting point of this thesis, however, is that the ability of criminal law to promote gender equality must be questioned. Instead criminal law is at risk of producing and reproducing hindrances to gender equality and criminal legal protection for women. It is such processes in criminal law that are explored and analysed in this thesis, inspired by feminist legal theory and social constructionist theory, and through the application of discourse analysis. The main purpose of the thesis is to explore and analyze how violence, perpetrator and victim are constructed in the criminal legal discourse of men’s physical and psychological violence against women in intimate relationships and what legal or social consequences these constructions might have. A second aim of the thesis is to throw a light on and analyze two major criminal legal aspects of men’s physical and psychological violence against women in intimate relationships. Firstly, how criminal law deals with issues of responsibility for the violence. Secondly, how questions about the use of criminal law are linked to possibilities and problems from a crime victim-perspective. The analysis indicates two major problem areas. In the first, the victim is treated in relation to stereotypical conceptions of women exposed to violence. A male-gendered discourse on responsibility limits the possibilities for women to behave as actors in relation to men and still being offered the subject position of victim. In addition, a focus on questions such as “Why doesn’t she leave?” and “Why doesn’t she cooperate with the criminal justice system?” produces an understanding of the main problem of violence as being related to the victim. The second problem area concerns a tension between how criminal legal reforms, aimed at enhancing criminal legal protection for women and promoting gender equality, have been justified by the legislator and how mainstream criminal legal scholarship argues in the issue of use of criminal law.

  • Low occurrence of extended-spectrum cephalosporinase producing Enterobacteriaceae and no detection of methicillin-resistant coagulase-positive staphylococci in healthy dogs in Sweden
  • 2020
  • Ingår i: Acta Veterinaria Scandinavica. - : BioMed Central (BMC). - 0044-605X .- 1751-0147. ; 62
  • Tidskriftsartikel (refereegranskat)abstract
    • Sweden has a long tradition of monitoring occurrence of antibiotic resistant bacteria in both animals and humans, but there currently is no organised and harmonized monitoring on carriage of Enterobacteriaceae producing extended-spectrum beta-lactamase (ESBL), plasmid-mediated AmpC beta-lactamase (pAmpC), or methicillin-resistant coagulase positive staphylococci e.g. methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-resistant Staphylococcus pseudintermedius (MRSP) in dogs. The aim of the current study was therefore to determine the prevalence of ESBL/pAmpC producing Enterobacteriaceae and methicillin-resistant coagulase positive staphylococci in healthy dogs in Sweden, and to phenotypically and genotypically characterize any identified isolates. It was shown that 0.9% (95% confident interval 0.3-2.7%) of the dogs (n = 325) carried multi-resistant ESBL-producing Escherichia coli, but that no methicillin-resistant coagulase positive staphylococci could be detected. In conclusion, the occurrence of multi-drug resistant bacteria remains rare among healthy dogs in Sweden. In addition, the ESBL-producing E. coli identified showed genetic characteristics related to those reported from humans.

  • Advancing Sustainable Transport Systems : Strengthening environmental considerations and sustainability perspectives in Strategic Choice of Measures
  • 2024
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Addressing cross-cutting issues such as climate change, urbanization, andenvironmental degradation in policy and planning is key for transitioning toa sustainable society. In the work of societal and transport systemdevelopment, the integration of environmental and sustainabilityperspectives has never been more crucial. In urban areas, the interconnection between transport and land use is evident, and integrated transport and land use planning plays an important role in promotingsustainable transport systems. However, both the integration ofenvironmental and sustainability considerations in transport planning and the integration of land use and transport planning are complex challenges. This thesis explores the consideration of environmental aspects andsustainability perspectives in strategic transport planning, with a focus onthe Swedish Strategic Choice of Measures (SCM) approach and lessonslearnt from planning approaches in the Nordic countries. The thesis is primarily based on case studies of SCM processes in the Stockholm region. Empirical material has been collected through observations of SCMs involving public officials from the Swedish Transport Administration, municipalities in Stockholm Region, and Stockholm's public transportauthority, as well as through semi-structured interviews, focus groupinterviews, informal interviews and document studies, with a qualitative research approach. The thesis is a compilation of four scientific papers. Three of the papers (1,2, and 4) discuss the SCM process, and in two of the papers (2 and 4), SCM processes in the Stockholm region are used as empirical cases of howstrategic choices of measures have been made in transport and land useplanning. Paper 3 explores and draws lessons from Nordic planning instruments to coordinate transport and land use planning with the aim of promoting sustainable development. The results highlight the difference between engaging and non-engagingenvironmental aspects and advocate for a systems perspective in planning to understand connections and promote holistic solutions. Planning approaches from different Nordic countries, such as the Finnish MAL and the Norwegian Urban Growth Agreements, demonstrate the effectiveness of integrating transport and land use planning through instruments based oncollaboration between transport and land use actors. The results also identify challenges related to fragmented transport and land use planning, and that actors' vary in their engagement when addressing environmental and sustainability considerations in the SCM planning context. Effective collaboration and sustainable choices of measures relies on knowledgeable professionals being capable of interdisciplinary collaborations and innovative thinking, being part of and coordinating the planning process, i.e.SCM. The use of Integrated Landscape Character Assessment (ILCA) can provide a framework for cross-actor collaboration and has the potential to identify multifunctional solutions contributing to a sustainable societal transition. Overall, the results of the thesis emphasize the importance of integrating environmental and sustainability perspectives into transport planning to promote sustainable development.

  • Challenges and Opportunities in Early Stage Planning of Transport Infrastructure Projects : Environmental Aspects in the Strategic Choice of Measures Approach
  • 2021
  • Ingår i: Sustainability. - : MDPI AG. - 2071-1050. ; 13:3
  • Tidskriftsartikel (refereegranskat)abstract
    • The Strategic Choice of Measures (SCM) approach aims to integrate different perspectives and identify measures to adapt new infrastructure projects to their local context at an early stage of Swedish transport planning. SCM is a loosely structured framework for collaboration between actors from, e.g., municipalities and the Swedish Transport Administration, in order to facilitate the coordination of transport planning and land use planning. This paper aims to explore the consideration of environmental aspects in early-stage transport planning by analyzing the SCM approach. An explorative research approach is applied based on literature studies, semi-structured interviews, and a focus group interview. The result shows that in the SCM process, environmental aspects such as noise and air pollution generated by road traffic in urban areas, engage the actors, whereas aspects related to landscape and water were perceived as poorly addressed and received less attention. The consideration of environmental aspects in the SCM process is affected by the local and national authorities' different interests and the competences involved. To consolidate environmental aspects in early transport planning, these aspects need to be explicitly addressed in the SCM guidelines and the link between the SCM and preceding and following planning stages needs to be strengthened.

  • New perspectives on Strategic Choice of Measures in Swedish Transport Planning
  • 2022
  • Licentiatavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Rapid changes in the global climate and declining biodiversity pronounces the need for adapting the transport system and transport infrastructure, which influence several sustainability issues and cause direct environmental impact through their use of land, as well as indirect impacts, for example, emissions of pollutants to air and water. The Swedish Transport Administration (STA) is responsible for and conducts a large part of the transport planning in Sweden and the STA endeavor to increase collaboration with other societal actors in transport planning. In 2013, the Strategic Choice of Measure (SCM) process was introduced by the STA to facilitate collaboration among multiple societal actors in the early stages of transport planning. In the SCM process many of the choices are made that will affect the development of the transport system as well as the future transport system's impact on the environment. Integrating environmental aspects at an early planning stage, such as in the SCM process, can help reduce and avoid the environmental impact of the transport system. Therefore, this licentiate thesis aims to explore the consideration of environmental aspects in the SCM process (Paper 1), and to analyze the SCM process from a systems perspective in order to illuminate the consequences of system boundaries on how environmental aspects are considered and what aspects are included or left out (Paper 2). In addition, proposals are being developed to strengthen practice (Papers 1 and 2).To achieve the aims of the thesis, interview studies, document studies and observational studies have been performed. Also, a case study of two SCM processes was conducted. This dissertation shows that there are several challenges associated with the consideration of environmental aspects in SCM processes. For example, the increased element of collaboration in the planning process implies that the coordination of the process will affect which environmental aspects are considered and in what way. Furthermore, land-use and transport planning are coordinated in the SCM process, which often leads to conflicting ideas about how an area should be developed. By finding solutions to transport and land-use related problems with synergetic effects, conflicts may be solved, and transport, land-use and environmental needs may be met simultaneously. To identify this type of measure and packages of measures in an SCM process, tools that enable the identification of links between transport, land-use and environmental aspects can be used as support.

  • 2008
  • Ingår i: PLoS Genetics. - : Public Library of Science (PLoS). - 1553-7390 .- 1553-7404. ; 4:2, s. e1000010-
  • Tidskriftsartikel (refereegranskat)abstract
    • Yellow skin is an abundant phenotype among domestic chickens and is caused by a recessive allele (W*Y) that allows deposition of yellow carotenoids in the skin. Here we show that yellow skin is caused by one or more cis-acting and tissue-specific regulatory mutation(s) that inhibit expression of BCDO2 (beta-carotene dioxygenase 2) in skin. Our data imply that carotenoids are taken up from the circulation in both genotypes but are degraded by BCDO2 in skin from animals carrying the white skin allele (W*W). Surprisingly, our results demonstrate that yellow skin does not originate from the red junglefowl (Gallus gallus), the presumed sole wild ancestor of the domestic chicken, but most likely from the closely related grey junglefowl (Gallus sonneratii). This is the first conclusive evidence for a hybrid origin of the domestic chicken, and it has important implications for our views of the domestication process.

  • 2019
  • Ingår i: Journal of Cleaner Production. - : ELSEVIER SCI LTD. - 0959-6526 .- 1879-1786. ; 236
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper describes how different backcasting scenarios for developments beyond traditional GDP growth 2050, in Sweden may fulfil a climate goal corresponding to keeping global warming to a maximum 1.5 degrees C with 50% likelihood. This corresponds to a 92% decrease of greenhouse gas emissions from Swedish consumption from today's level. The four scenarios illustrate different strategies: 1) collaborative economy, 2) local self-sufficiency, 3) automation for quality of life and 4) circular economy in the welfare state. The aim is to further hone and quantify the scenario narratives with a focus on greenhouse gas emissions occurring as a result of Swedish consumption, both private and public. The results show that the climate target can be met in all scenarios but this requires radical sector-specific as well as general changes, including decarbonisation, technology development, increased efficiencies, innovative practices and reduced demand. The mix of these strategies varies for different sectors and different scenarios, but all are needed to reach the climate goals. As we assume that Sweden is fossil-free 2050, particular areas of attention are diets, travel, emission intensities in other countries and the level of imports. Potential implications for other environmental goals, land use and biodiversity as well as the potential magnitude of negative emissions technologies, although uncertain and limited, that could offset some additional greenhouse gas emissions are discussed.

Preliminary results show that, as we assume that Sweden is fossil-free 2050, particular areas of attention are diets, air travel, emission intensities in other countries and the level of imports.. Potential implications for other environmental goals are discussed.

  • Sharing the doughnut : Exploring sustainable and just futures
  • 2018
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Despite decades of international discussions or summits on the need to radically reduce e.g. increasing concentrations of greenhouse gases (GHGs) or biodiversity loss, these are still rising. While these negative environmental trends continue, it is important to discuss alternative futures in an attempt to redirect society on a more sustainable and just path. The overall aim of the thesis is to develop images of the future and explore what sustainable and just futures might look like. The current environmental impact of Swedish consumption, both in Sweden and abroad, is shown using eight indicators of environmental pressures and resource use – illustrating where in the world the pressures or resource use occur and for which product groups. This gives us a starting point as to where we are today regarding some of the environmental challenges facing Sweden.Alternative futures that can challenge existing unsustainable trends are explored using four images of the future – so-called backcasting or long-term transformative scenarios. All of these need to fulfil two environmental and two social sustainability goals and do not rely on continued GDP growth. These images represent different strategies to reach the four selected goals.Such strategies may however have different consequences not just for these four specific goals but also for other sustainability issues and may have different implications for various groups in society. Therefore, they need to be evaluated accordingly. Existing methods to evaluate future scenarios regarding sustainability aspects are discussed in this thesis as well as the need to develop new methods to encompass all issues.A combination of methods and data is used to evaluate what it would actually mean if the scenarios were to fulfil a climate target for Swedish consumption in line with the 1.5°C. trajectory suggested as the target to strive for in the Paris Climate Agreement and in the latest IPCC report (IPCC, 2018) as regards reduction of goods consumption and altered consumption patterns in Sweden. This thesis stresses the need to clarify the assumptions made when formulating goals such as whether a perspective on intergenerational (between different generations) and intragenerational justice (within the current generation) is considered. It also underlines the need to identify and discuss potential goal conflicts that necessarily occur when considering several sustainability goals, whether they can be avoided or require potential trade-offs. It highlights the importance of making the underlying values embedded in assessment methods more visible. The intention in revealing goal conflicts, contradictions or hidden values is not to reach consensus but to ensure that the decisions are informed and made in a transparent manner.Indeed, these considerations imply moving from a first and rather vague level of meaning of the concept of sustainability where everyone can agree on a definition but no concrete and practical guidance can be gained to a second level where conflicts arise and values differ.

  • Sustainability goals combining social and environmental aspects
  • 2016
  • Licentiatavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis examines how to take into account both environmental and social sustainability goals to be used in scenarios or in policymaking. In paper I, we select four sustainability goals that have to be fulfilled by 2050 in normative future scenarios for Sweden in a degrowth context. Two goals address ecological challenges, climate change and land use issues specifically. The other two goals address social issues and deal with participation and influence in society as well as resource security and distribution. The environmental goals will require significant reductions in greenhouse gas (GHG) emissions and land use compared to today's levels. The social goals are within reach today, although the degree of fulfillment differs across different groups in society. In paper II, we review existing and suggested climate or energy targets at a global, national and local scale and search for justice perspectives or for proposals for such perspectives. We find that the justice aspect is not explicitly formulated in existing climate and energy targets and that, the community of justice i.e. the receivers of benefits or burdens, in our reviewed examples, is limited to human beings, thereby excluding all other living beings. In paper III, we assess how four different backcasting scenarios for land use in a Swedish context, all of which fulfil a climate target of zero CO2 emissions in 2060, impact on other sustainability goals. We conduct a goal conflict analysis between the chosen climate goal and the other Swedish environmental goals, the gender equity goals and the public health goal. We find that there are more potential goal conflicts in scenarios with no global climate agreement. From the results of all three papers, I then discuss several aspects that have to be taken into account when setting goals, such as the major uncertainties associated with long-term goals, the elusiveness, the normativity of goals and the need to separate goals from the means to achieve the goals.

  • Policy instruments towards a sustainable waste management
  • 2013
  • Ingår i: Sustainability. - Basel : MDPI AG. - 2071-1050. ; 5:3, s. 841-881
  • Tidskriftsartikel (refereegranskat)abstract
    • The aim of this paper is to suggest and discuss policy instruments that could lead towards a more sustainable waste management. The paper is based on evaluations from a large scale multi-disciplinary Swedish research program. The evaluations focus on environmental and economic impacts as well as social acceptance. The focus is on the Swedish waste management system but the results should be relevant also for other countries. Through the assessments and lessons learned during the research program we conclude that several policy instruments can be effective and possible to implement. Particularly, we put forward the following policy instruments: “Information”; “Compulsory recycling of recyclable materials”; “Weight-based waste fee in combination with information and developed recycling systems”; “Mandatory labeling of products containing hazardous chemicals”, “Advertisements on request only and other waste minimization measures”; and “Differentiated VAT and subsidies for some services”. Compulsory recycling of recyclable materials is the policy instrument that has the largest potential for decreasing the environmental impacts with the configurations studied here. The effects of the other policy instruments studied may be more limited and they typically need to be implemented in combination in order to have more significant impacts. Furthermore, policy makers need to take into account market and international aspects when implementing new instruments. In the more long term perspective, the above set of policy instruments may also need to be complemented with more transformational policy instruments that can significantly decrease the generation of waste.

    • The aim was to explore informal carers' perceptions of supporting the everyday life of a relative who has a psychiatric disability and resides in supported housing (SH). A qualitative study based on interviews with 12 informal carers was performed, and the data was analyzed with qualitative content analysis. The theme "Navigating in a misty landscape when striving to support a relative with a psychiatric disability" was identified, encompassing four categories pertaining to residents' needs, collaboration, environmental issues and the carer's situation. SH services can be enhanced by addressing informal carers' experiences and developing greater collaboration involving informal carers, residents and staff.

    • In this work we take the point of departure from two thoroughly conducted casestudies, in which we have analysed certain knowledge work activities with followingimplications for design. Both studies clearly stated the call for a ICT-support whensearching information and sharing knowledge. For example, sharing bookmarksamong knowledge workers seemed to be a necessary and useful activity. Importantand valuable bookmarks often mirror what kind of information and knowledgepeople are looking for, and also what kind of forum they usually go to on the worldwide web. In this work we have developed a prototype, KnowMan, which managesbookmarks on the world wide web. The prototype is designed as a user-friendly"pad-interface" in which bookmarks can be collaboratively collected, shared andfurther explored and annotated by other knowledge workers. We end up with ageneral discussion about the possible effects of using KnowMan and what furtherdesign aspects that should be considered.

  • Screening for copy-number alterations and loss of heterozygosity in chronic lymphocytic leukemia-A comparative study of four differently designed, high resolution microarray platforms

  • Tidskriftsartikel (refereegranskat)abstract
    • Screening for gene copy-number alterations (CNAs) has improved by applying genome-wide microarrays, where SNP arrays also allow analysis of loss of heterozygozity (LOH). We here analyzed 10 chronic lymphocytic leukemia (CLL) samples using four different high-resolution platforms: BAC arrays (32K), oligonucleotide arrays (185K, Agilent), and two SNP arrays (250K, Affymetrix and 317K, Illumina). Cross-platform comparison revealed 29 concordantly detected CNAs, including known recurrent alterations, which confirmed that all platforms are powerful tools when screening for large aberrations. However, detection of 32 additional regions present in 2-3 platforms illustrated a discrepancy in detection of small CNAs, which often involved reported copy-number variations. LOH analysis using dChip revealed concordance of mainly large regions, but showed numerous, small nonoverlapping regions and LOH escaping detection. Evaluation of baseline variation and copy-number ratio response showed the best performance for the Agilent platform and confirmed the robustness of BAC arrays. Accordingly, these platforms demonstrated a higher degree of platform-specific CNAs. The SNP arrays displayed higher technical variation, although this was compensated by high density of elements. Affymetrix detected a higher degree of CNAs compared to Illumina, while the latter showed a lower noise level and higher detection rate in the LOH analysis. Large-scale studies of genomic aberrations are now feasible, but new tools for LOH analysis are requested.

  • Genetic Studies of Pigmentation in Chicken
  • 2009
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Domestic animals have been selected by humans for thousands of years, which have drastically altered their genetic constitution and phenotypes. In this thesis, several of the most important genes causing pigmentation differences between the wild red junglefowl (Gallus gallus) and domestic chickens have been identified. Pigmentation phenotypes are easily scored, and the genes underlying these phenotypes are valuable models to study gene function and gene interaction. Dominant white colour is widespread among domestic chickens. The Dominant white allele specifically inhibits the expression of black (eumelanin) pigment and we identified several insertion/deletion mutations in the PMEL17 gene causing the different phenotypes controlled by this locus. The Silver allele on the other hand inhibits the expression of red (pheomelanin) colour and is a genetic variant of the SLC45A2 gene. Silver is the first pheomelanin-specific mutation(s) reported for this gene. An 8 kb deletion, including a conserved enhancer element, 14 kb upstream of the transcription factor SOX10 is causing the Dark brown phenotype. This phenotype restricts the expression of eumelanin and enhances red pheomelanin in specific parts of the plumage. These three gene identifications have extended the knowledge about genes affecting melanocyte function. Carotenoid-based pigmentation is of utmost importance in birds and other animals. The yellow skin allele in chicken allows deposition of carotenoids in skin and explains why most domestic chickens have yellow legs. We demonstrated that the yellow skin phenotype is caused by a tissue specific regulatory mutation in the gene for the enzyme beta-caroten dioxygenase 2 (BCDO2). This was the first identification of a specific gene underlying carotenoid-based pigmentation. Interestingly, the yellow skin haplotype was shown to originate from the grey junglefowl (Gallus sonneratii) and not the red junglefowl as expected, thus presenting the first conclusive evidence for a hybrid origin of the domestic chicken.

  • Mutations in SLC45A2 Cause Plumage Color Variation in Chicken and Japanese Quail

  • Ingår i: Genetics. - : Oxford University Press (OUP). - 0016-6731 .- 1943-2631. ; 175:2, s. 867-877
  • Tidskriftsartikel (refereegranskat)abstract
    • S*S (Silver), S*N (wild type/gold), and S*AL (sex-linked imperfect albinism) form a series of alleles at the S (Silver) locus on chicken (Gallus gallus) chromosome Z. Similarly, sex-linked imperfect albinism (AL*A) is the bottom recessive allele at the orthologous AL locus in Japanese quail (Coturnix japonica). The solute carrier family 45, member 2, protein (SLC45A2), previously denoted membrane-associated transporter protein (MATP), has an important role in vesicle sorting in the melanocytes. Here we report five SLC45A2 mutations. The 106delT mutation in the chicken S*AL allele results in a frameshift and a premature stop codon and the corresponding mRNA appears to be degraded by nonsense-mediated mRNA decay. A splice-site mutation in the Japanese quail AL*A allele causes in-frame skipping of exon 4. Two independent missense mutations (Tyr277Cys and Leu347Met) were associated with the Silver allele in chicken. The functional significance of the former mutation, associated only with Silver in White Leghorn, is unclear. Ala72Asp was associated with the cinnamon allele (AL*C) in the Japanese quail. The most interesting feature concerning the SLC45A2 variants documented in this study is the specific inhibition of expression of red pheomelanin in Silver chickens. This phenotypic effect cannot be explained on the basis of the current, incomplete, understanding of SLC45A2 function. It is an enigma why recessive null mutations at this locus cause an almost complete absence of both eumelanin and pheomelanin whereas some missense mutations are dominant and cause a specific inhibition of pheomelanin production.

  • The Dark brown plumage color in chickens is caused by an 8.3 kb deletion upstream of SOX10.
  • 2011
  • Ingår i: Pigment cell & melanoma research. - 1755-148X .- 1755-1471.
  • Tidskriftsartikel (refereegranskat)abstract
    • The Dark brown mutation in chickens reduces expression of black eumelanin and enhances expression of red pheomelanin but only in certain parts of the plumage. Here we present genetic evidence that an 8.3 kb deletion upstream of the SOX10 transcription start site is the causal mutation underlying the Dark brown phenotype. The SOX10 transcription factor has a well-established role in melanocyte biology and is essential for melanocyte migration and survival. Previous studies have demonstrated that the mouse homolog of a highly conserved element within the deleted region is a SOX10 enhancer. The mechanism of action of this mutation remains to be established but one possible scenario is that the deletion leads to reduced SOX10 expression which in turn down-regulates expression of key enzymes in pigment synthesis such as tyrosinase. Lower tyrosinase activity leads to a shift towards a more pheomelanistic (reddish) plumage color, which is the characteristic feature of the Dark brown phenotype.

  • Alternativa resepraktiker? : Potentiella beteendeförändringar ispåren av COVID-19
  • 2021

  • Climate change scenarios and behavioural change : Navigating between heuristics of deliberative planning processes and astroturfing
  • 2008
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • In constructing normative scenarios images of the future are generated illustrating potential ways of living, travelling and consuming products and services where certain goals such as a reduced climate impact are fulfilled (Börjeson et al, 2006; Myers and Kitsuse, 2000). This paper analyses the sustainability framing of behavioural changes in such normative scenarios (Larsen and Höjer, 2007) and how it relates to the participatory processes used to generate the scenarios. We discuss this building on concepts of deliberative planning processes as a means to achieve legitimate, effective and sustainable futures (Connelly and Richardson, forthcoming; Hendriks et al., 2007). The theoretical arguments are combined with examples from environmental scenario construction in practice (Carlsson-Kanyama et al., 2003, Dreborg et al., forthcoming). This illustrates fields of tension arising when either sustainability or process values are veneering goals of a scenario making process. When focus is on content values such as reduced climate impact, the process values might be depreciated. On the other hand, when focus is on process values such as legitimacy, content sustainability and possibilities for actual change might be downgraded.

    • This paper reviews and discusses papers related to women's studies, gender or feminist perspectives, published in the scientific journal Futures. The aim is to provide new understandings and remapping of futures studies by capturing how gender is created and understood in this field. The gender/feminist criticism of futures studies mainly relates to the field being male-dominated and male biased, which means that the future is seen as already colonised by men. When synthesising the insights from all 78 papers focusing on futures studies and feminism, gender or women, four conclusions are especially striking: (1) Women and non-Westerners are generally excluded from professional futures studies activities and so are feminist issues or issues of particular relevance for women. (2) Futures studies usually make no attempts to reveal underlying assumptions, i.e. often lack a critical and reflexive perspective, which is needed in order to add a critical feminist perspective and envision feminist futures. (3) Feminist futures are needed as a contrast to hegemonic male and Western technology-orientated futures. Feminist futures are diverse, but focus the well-being of all humans. (4) Futures studies often view women as victims, rather than as drivers for change, which means that their alternative futures are often ignored.
