    • Electron capture dissociation of the peptide Substance P is reported for the first time, with an unmodified, commercially available Fourier transform ion cyclotron resonance (FTICR) mass spectrometer. The fragmentation pattern is compared with that obtained with collisionally induced dissociation of the ions in the electrospray ion source, and note that electron capture dissociation gives a more easily interpreted spectrum, showing mainly C-fragments. With the exception of the proline residues, which require cleavage of two chemical bonds, we observe all C-fragmental we find the bias voltage of the electron gun not to be very critical.

    • A new method is presented to accurately determine the probability of having a deuterium or hydrogen atom on a specific amide position within a peptide after deuterium/hydrogen (D/H) exchange in solution. Amide hydrogen exchange has been proven to be a sensitive probe for studying protein structures and structural dynamics. At the same time, mass spectrometry in combination with physical fragmentation methods is commonly used to sequence proteins based on an amino acid residue specific mass analysis. In the present study it is demonstrated that the isotopic patterns of a series of peptide fragment ions obtained with capillary-skimmer dissociation, as observed with a 9.4 T Fourier transform ion cyclotron resonance (FTICR) mass spectrometer, can be used to calculate the isotopic state of specific amide hydrogens. This calculation is based on the experimentally observed isotopic patterns of two consecutive fragments and on the isotopic binomial distributions of the atoms in the residue constituting the difference between these two consecutive fragments. The applicability of the method is demonstrated by following the sequence-specific D/H exchange rate in solution of single amide hydrogens within some peptides.

    • 9.4 Tesla Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometers (Bruker BioAPEX-94e) have been installed at the Division of Ion Physics, Uppsala University, and at the Department of Chemistry, University of Warwick, The BioAPEX-94e FT-ICR instrument is built around a high-field, superconducting magnet and a platform with easily interchangeable ion sources [matrix-assisted laser desorption/ionisation (MALDI), secondary ion mass spectrometry (SIMS), electrospray ionisation (ESI) and electron impact/chemical ionisation (EI/CI)I. In this paper a technical description of the instrument is given. Outstanding performance characteristics are demonstrated, notably clear resolution of C59N+ and (C58C2+)-C-13 (mass difference 3.65 mDa) and mass measurement accuracy at the low ppm level. A wide range of applications in Warwick and Uppsala is described, demonstrating the versatility and high performance of the instrument.

    • This report presents a project at Lund University Libraries (LUB) during the fall of 2012, with the object to identify areas where the present research support services ought to be developed and strengthened. The project is three folded and contains a) a literature review focusing on definitions and examples of research support services as well as researchers’ needs and experiences of support services, b) a survey to get an overview of the support services offered today at LUB, the decentralized library network at Lund University, and c) focus group interviews with researchers, one group from each partaking faculty, to investigate key obstacles in their research processes. In all three parts a model of the research process schematically divided into four phases was applied; to start a project, to collect material, to process/analyze/write and to communicate the results. The first two stages of the project, the literature overview and the survey, form a background and both show that many support services are concentrated around the phases to collect material and to communicate results. The findings from the focus group interviews point to common needs and issues as well as special needs and issues related to different ways of doing and communicating research. Common for most researchers is limited time, a need for funding and an everyday fragmentation. They talk about the importance of networks, their efforts to navigate in an abundance of material and ways to find it and a need for overviews and help to understand new ways and demands to communicate. Most concern is expressed in the phases to start a project, to collect material and to communicate results. A major conclusion is that the research support services need to be available, visible and in close proximity to the researchers. Further, the development of support services needs to take into consideration both common traits as well as local variations in research practices. Therefore, discussions concerning LUB as a whole, need to be related to discussions at the single faculty libraries. Apart from knowledge about how to address the research support services at LUB, the project generates a model for how librarians can understand the research community that they are working with and with whom to create networks and partnerships for the future.

  • Oral Administration of Lactobacillus plantarum 299v Reduces Cortisol Levels in Human Saliva during Examination Induced Stress : A Randomized, Double-Blind Controlled Trial

    • Objective. To clarify the effect of Lactobacillus plantarum 299v on the salivary cortisol and salivary IgA levels in young adults under examination stress. Design. Forty-one students with an upcoming academic exam were included in a randomized double-blind, placebo-controlled study. The probiotic bacteria or the placebo product was administered in capsules once a day during 14 days. Saliva was collected and a perceived stress test was filled out at each sampling occasion. Saliva was collected for cortisol analysis by Electrochemiluminescence Immunoassay (ECLI) and salivary IgA was analysed by Enzyme-Linked Immunosorbent Assay (ELISA). Abundance of lactobacilli was evaluated by cultivation of saliva on selective medium and identification of L. plantarum 299v was done on randomly selected colonies by a random amplification of polymorphic DNA (RAPD) typing. Results. A significant difference in cortisol levels was found between the treatment group and the placebo group (P < 0.05), together with a significant increase in levels of lactobacilli in the treatment group compared with the placebo group (P < 0.001). No significant changes were found for salivary IgA. Conclusion. A probiotic bacterium with ability to reduce symptoms of irritable bowel syndrome (IBS) prohibited increased levels of the stress marker cortisol during the examination period. The registration number of the study is NCT02974894, and the study is registered at ClinicalTrials.gov.

    • A non-toxic hydrolytically fast-degradable antibacterial hydrogel is herein presented to preemptively treat surgical site infections during the first crucial 24 h period without relying on conventional antibiotics. The approach capitalizes on a two-component system that form antibacterial hydrogels within 1 min and consist of i) an amine functional linear-dendritic hybrid based on linear poly(ethylene glycol) and dendritic 2,2-bis(hydroxymethyl)propionic acid, and ii) a di-N-hydroxysuccinimide functional poly(ethylene glycol) cross-linker. Broad spectrum antibacterial effect is achieved by multivalent representation of catatonically charged β-alanine on the dendritic periphery of the linear dendritic component. The hydrogels can be applied readily in an in vivo setting using a two-component syringe delivery system and the mechanical properties can accurately be tuned in the range equivalent to fat tissue and cartilage (G' = 0.5-8 kPa). The antibacterial effect is demonstrated both in vitro toward a range of relevant bacterial strains and in an in vivo mouse model of surgical site infection.

    • In the Swedish labour market, recent research has revealed tendencies of both dualisation – implying growth in temporary employment – and polarisation – referring to increased employment in both low- and high-paid jobs, while middle-paid jobs decrease. This study explores the relationships between changes in the occupational structure and the distribution of temporary employment. Using the Labour Force Survey and comparing changes between 2000 and 2015, the study shows a main pattern of upgrading. However, splitting the analysis into permanent and temporary employment, tendencies of polarisation are revealed: growth at the low-paid end consists of temporary employment, while the increase at the high-paid end is mainly of openended contracts. Different kinds of temporary contracts matter – on-call employees are more likely to be found at the low end, and project workers at the high end. The study shows increasing precariousness in the lower end of the occupational structure.

    • The aim of this article is to describe and explain the development of temporary employment in Sweden between 1992 and 2010, and to investigate the effect of temporary employment for individuals' futurecareer on the labor market. The article analyzes temporary employees' status transitions on the labor market using Swedish Labour Force Survey (LFS) data for the period 1992-2010. Each cohort consists of 2-year panels and focuses on changes between the first and last measuring points. The findings indicate that the specific type of temporary employment is crucial as regards whether or not it constitutes a stepping-stone toward permanent employment. The chances are greater in the case of, for example, substitutes, but are considerably less in the case of on-call employment. Certain types of temporary employment thus seem to be used by the employer to screen the employability of the employee, while others are used for achieving flexibility.

    • Background: Opioid overdose (OD) and opioid OD death are major health threats to people with opioid use disorder (OUD). Socioeconomic factors are underexplored potential determinants of opioid OD. In this study, we assessed socioeconomic and other factors and their associations with incident and fatal opioid OD, in a cohort consisting of 22,079 individuals with OUD. Methods: We performed a retrospective, longitudinal study based on Swedish national register data for the period January 2005–December 2017. We used Cox proportional hazard models to investigate the risk of incident and fatal opioid OD as a function of several individual, parental and neighborhood covariates. Results: Univariate analysis showed that several covariates were associated with incident and fatal opioid OD. In the multivariate analysis, incident opioid OD was associated with educational attainment (Hazard ratio [HR] 0.96; 95% confidence interval [CI] 0.94–0.97), having received social welfare (HR 1.31; 95% CI 1.22–1.39), and criminal conviction (HR 1.53; 95% CI 1.42–1.65). Fatal opioid OD was also associated with criminal conviction (HR 1.93; 95% CI 1.61–2.32). Conclusion: Individuals with low education and receipt of social welfare had higher risks of incident opioid OD and individuals with criminal conviction were identified as a risk group for both incident and fatal opioid OD. Our findings should raise attention among health prevention policy makers in general, and among decision-makers within the criminal justice system and social services in particular.

  • Analysis of spatial variability in hyperspectral imagery of the uterine cervix in vivo

    • The use of fluorescence and reflectance spectroscopy in the analysis of cervical histopathology is a growing field of research. The majority of this research is performed with point-like probes. Typically, clinicians select probe sites visually, collecting a handful of spectral samples. An exception to this methodology is the Hyperspectral Diagnostic Imaging (HSDI®) instrument developed by Science and Technology International. This non-invasive device collects contiguous hyperspectral images across the entire cervical portio. The high spatial and spectral resolution of the HSDI instruments make them uniquely well suited for addressing the issues of coupled spatial and spectral variability of tissues in vivo. Analysis of HSDI data indicates that tissue spectra vary from point to point, even within histopathologically homogeneous regions. This spectral variability exhibits both random and patterned components, implying that point monitoring may be susceptible to significant sources of noise and clutter inherent in the tissue. We have analyzed HSDI images from clinical CIN (cervical intraepithelial neoplasia) patients to quantify the spatial variability of fluorescence and reflectance spectra. This analysis shows the spatial structure of images to be fractal in nature, in both intensity and spectrum. These fractal tissue textures will limit the performance of any point-monitoring technology.

    • Patterns of daily occupations (PDO) and occupational balance (OB) are recurring phenomena in the literature. Both are related with health and well-being, which makes them central in occupational therapy practice and occupational science. The aim was to review how PDO and OB are described in the literature, to propose a view of how the two constructs may be linked, and elaborate on how such a view may benefit occupational science and occupational therapy. The literature was analysed by latent and manifest content analysis and comparative analysis. The findings were summarized in a model, framing PDO as the more objective and OB as the more subjective result from an interaction between personal preferences and environmental influences. The proposed model does not assume a cause–effect relationship between the targeted constructs, rather a mutual influence and a joint reaction to influencing factors. Indicators of PDO and OB were identified, as well as tools for assessing PDO and OB. The authors propose that discerning PDO and OB as separate but interacting phenomena may be useful in developing a theoretical discourse in occupational science and enhancing occupational therapy practice. Although the scope of this study was limited, the proposed view may hopefully inspire further scrutiny of constructs.

This development entails that job transformations and future skills need attention from the perspective of industry 5.0, where human and machine find ways of working together to improve production performance. Facing this perspective, one challenge is a growing need for novel lifelong learning initiatives, to meet emerging and altering occupations for the fulfilment of future skill requirements. This challenge is addressed here by portraying a case where three Swedish universities have formed a distinctive collaboration to develop a flexible (i.e. blended) course for professionals, in the subject of Digitally-enabled production. The purpose is to develop a sustainable collaboration between the universities and create a course format on master level addressing lifelong learning for the increasingly digitalized production. The ambition is to increase the impact of the universities respective efforts by sharing resources and utilizing individual specialized expertise to develop a practical and relevant course that can reach a larger target group. The course encompasses industry 4.0 readiness on three levels of production systems; plant-, production cell-, and component level; to adopt a holistic view of digitalization in production. We followed an action research approach for continuously collecting and documenting our experiences during the course development, implementation, and dissemination of the course. Within the frame of action research, an explorative case study describes and analyzes the initiative. The results highlight challenges and opportunities for succeeding with this form of co-produced course. The joint course gives professionals possibilities to work on cases from their own companies with expert supervision from three manufacturing levels to address complex challenges in industry 4.0 implementation. To conclude, the importance of lifelong learning in relation to the human-centric approach of industry 5.0 is emphasized as a future direction.

    • New competences and knowledge needs arises as manufacturing industry evolves and becomes increasingly digitalized. Facing this transformation, one of the challenges is the continuous and growing need for novel initiatives for competence development.The case portrayed here stems from a Swedish cross-university project aiming to jointly develop and offer courses for competence development of professionals in the manufacturing and IT sectors. The ambition is to increase the impact of the universities' respective efforts of meeting industry competence needs, where the continuous digital transformation entails that employees must develop or even change their qualifications.The case outlined focus co-production of a joint course package, at master level, between three universities and their respective company networks. Participating universities have long traditions in working closely with companies in research and education, where approaches for co-production have evolved over time. We make use of our joint understanding of the manufacturing industry's specific competence needs and our experiences of sustainable course formats for participants working full time.The joint course covers aspects of a manufacturing company on three levels: plant level i.e. material and production flows, cell level e.g. robotic simulation and visualisation, and system level i.e. data acquisition and monitoring through sensors. Each university is developing a course module of 2.5 ECTS, addressing a level respectively of their specialist competence. Participants are to complete assignments for each course module, i.e. for all three manufacturing levels, where the previous assignment provides an input to the next level, enabling the participants to encompass a holistic view of a manufacturing system. Participants need to combine study and work and at the same time they wish to extend their network, hence we are adopting a blended learning approach, where virtual labs and web conferences are mixed with physical meetings.A variety of challenges arise when designing such novel approaches: combining company networks, course design including online learning, planning of physical course meetings,joint promotion, common admission and validation process, financial models and more. However, engaging in partnerships with industry for knowledge transformation and development has the potential to become rewarding for all parties.

    • How can you as a planner tackle messy realities without losing sight of possible problematic outcomes of what you put in practice? This study explores the value of everyday frictions as a resource for phronetic planning, i.e. the ability to make situated ethical judements of what is ’better’ or ’worse’ in a particular setting. The intent is to offer a situated gaze of frictions from the perspective of a civil servant of the well organised and innovative municipality of Botkyrka in the metropolitan area of Stockholm, Sweden. From this outset, an autoethnographical methodology from a poststructural approach, is used to explore the frictions that the author has experienced as a practicing planner. The study shows that frictions can be used as 'weak signals' to identify possible tricky consequences of the creation and the staging of planning 'simplifications'. Based on this insight a 'seismological' approach to planning is proposed. The argument is that planning practice should on the one hand utilize frictions when they arise and, on the other hand, actively challenge existing 'simplifications'. To achieve this, practical tools are offered to 'evoke', 'narrate' and 'diffract' on frictions and 'trickster-objects' within the constraints of the planner’s role as a municipal civil servant.

    • The prevalence of antibiotic resistance and their genetic determinants in colonizing group B streptococci (GBS) sampled in a Swedish nationwide survey was examined. In five GBS isolates (1.3%), kanamycin/amikacin resistance and the presence of the aphA-3 gene was identified. Three of these isolates carried the aad-6 gene and were streptomycin-resistant. Screening with kanamycin and streptomycin 1,000-μg disks enabled a rapid and easy detection of these isolates. In all, 312/396 (79%) GBS were tetracycline-resistant and 95% of the examined isolates harbored the tetM gene. Among the 22 (5.5%) GBS resistant to erythromycin and/or clindamycin, the ermB gene was detected in nine isolates (41%) and erm(A/TR) in ten isolates (45%). A high level of erythromycin and clindamycin resistance with minimum inhibitory concentrations (MICs) >256mg/L was found in four serotype V isolates that harbored ermB. The erythromycin/clindamycin resistance was distributed among all of the common serotypes Ia, Ib, II, III, IV, and V, but was not present in any of the 44 serotype III isolates associated to clonal complex 17. Screening for penicillin resistance with 1-μg oxacillin disks showed a homogenous population with a mean inhibition zone of 20mm. A change in the present oxacillin breakpoints for GBS is suggested.

    • The aim of this study was to explore older workers’ motivation for a full or extended working life. With particular focus on assistant nurses aged 55–64 years, working in the elderly care sector. Focus group interviews were conducted with five different groups of assistant nurses. Inductive thematic analysis was used to analyse the interviews and five main themes were developed from the data: ‘Organisational issues’, ‘Health-related problems’, ‘Private issues’, ‘Meaningfulness and appreciation’ and ‘Social support’. Several of the main themes concerned problems with too high work demands of the assistant nurses. These findings suggest that it is important to improve the working conditions of assistant nurses in order to create a more sustainable working life. Increasing the number of staff and improving recovery opportunities and work–life balance could be important steps to improving the working conditions for this group. Finally, upgrading the competency and professionalism of assistant nurses could help to increase the motivation for a full or extended working life.

    • Developments in the understanding of language disorders and the progression of clinical practice for children with developmental language disorder (DLD) in Sweden started with a focus on speech, with a gradual widening of focus towards other aspects of language skills — grammar, semantics, comprehension and pragmatics. Sweden is a multicultural country with about one-third of preschool children brought up in multilingual contexts. Speech and language therapy (SLT) education in Sweden was initiated by Gunnar Bjuggren at Karolinska Institute in Stockholm in 1964. From the establishment of the SLT profession, SLTs are well aware of the fact that children with DLD have difficulties with different aspects of language in addition to phonology, for example grammar. Irrespective of employer, SLTs are obligated to document data and interventions in patient files, in all their work undertaken within their clinical professional competence, that is, assessing, diagnosing and treating patients.

    • Background: Respiratory tract infections are the most common reason for antibiotic prescription in Sweden as in other countries. The prescription rates vary markedly in different countries, counties and municipalities. The reasons for these variations in prescription rate are not obvious. Aim: To find possible explanations for different antibiotic prescription rates in children. Design of study Prospective population based study. Setting All child health clinics in four municipalities in Sweden which, according to official statistics, had high antibiotic prescription rates, and all child health clinics in three municipalities which had low antibiotic prescription rates. Method: During one month, parents recorded all infectious symptoms, physician consultations and antibiotic treatments, from 848 18-month-old children in a log book. The parents also answered a questionnaire about socioeconomic factors and concern about infectious diseases. Results: Antibiotics were prescribed to 11.6% of the children in the high prescription area and 4.7% in the low prescription area during the study month (crude odds ratio [OR] = 2.67; 95% confidence interval [CI] = 1.45 to 4.93). After multiple logistic regression analyses taking account of socioeconomic factors, concern about infectious illness, number of symptom days and physician consultations, differences in antibiotic prescription rates remained (adjusted OR = 2.61; 95% CI = 1.14 to 5.98). The variable that impacted most on antibiotic prescription rates, although it was not relevant to the geographical differences, was a high level of concern about infectious illness in the family. Conclusions: The differences in antibiotic prescription rates could not be explained by socioeconomic factors, concern about infectious illness, number of symptom days and physician consultations. The differences may be attributable to different prescription behaviour.
