  • Associations between fluctuations in daytime sleepiness and motor and non-motor symptoms in Parkinson's disease

    • Abstract Background Non-motor fluctuations (NMF) are a major concern in Parkinson's disease (PD), and they have been categorised into neuropsychiatric, autonomic and sensory fluctuations. However, this categorisation does not include sleep and sleep-related features, and the association between daytime sleepiness and other motor and/or non-motor fluctuations in PD remains to be elucidated. Objective To investigate the relationship between daytime sleepiness and other non-motor and motor fluctuations in people with PD. Methods A three-day home diary recording daytime sleepiness, mood, anxiety, and motor symptoms was used along with the Karolinska Sleepiness Scale (KSS) and six days of accelerometer (Parkinson's KinetiGraph?; PKG?) registration to detect motor fluctuations among people with a DaTSCAN verified clinical PD diagnosis (32 men; mean PD duration, 8.2?years). Participants were categorised as motor fluctuators or non-fluctuators according to the UPDRS part IV and/or the presence of motor and non-motor fluctuations. Results Fifty-two people with PD participated. Daytime sleepiness correlated significantly with motor symptoms, mood and anxiety among those classified as motor fluctuators (n = 28). Motor fluctuators showed stronger correlations between the individual mean level of all diary variables (daytime sleepiness, anxiety, mood and motor symptoms) when compared to the non-fluctuators (n = 24). Stronger positive within-individual correlations were found among fluctuators in comparison to non-fluctuators. In general, PKG data did not correlate with diary data. Conclusion Episodes of daytime sleepiness, as reported by home diaries, were associated with other self-reported non-motor and motor fluctuations, but were not supported by PKG data.

    • A thorough investigation of quantum-dots-in-a-well structures for infrared photodetector applications has been performed employing different experimental techniques. The electronic structure of self-assembled InAs quantum dots embedded in an In0.15Ga0.85As/GaAs quantum well (QW) was deduced from photoluminescence (PL) and PL excitation (PLE) spectroscopy. From polarization-dependent PL it was revealed that the quantum dots hold two electron energy levels and two heavy-hole levels. Tunnel capacitance spectroscopy confirmed an electron energy level separation of about 50 meV, and additionally, that the conduction-band ground state and excited state of the dots are twofold and fourfold degenerates, respectively. Intersubband photocurrent spectroscopy, combined with simultaneous interband pumping of the dots, revealed a dominant transition at 150 meV (8.5 mu m) between the ground state of the quantum dots and the excited state of the QW. Results from detailed full three-dimensional calculations of the electronic structure, including effects of composition intermixing and interdot interactions, confirm the experimentally unravelled energy level scheme of the dots and well.

  • Self-esteem in a clinical sample of morbidly obese children and adolescents

    • Aim: To study self-esteem in clinical sample of obese children and adolescents.Methods: Obese children and adolescents aged 8–19 years (n = 107, mean age 13.2 years, mean BMI 32.5 [range 22.3–50.6], mean BMI z-score 3.22 [range 2.19–4.79]; 50 boys and 57 girls) were referred for treatment of primary obesity. Self-esteem was measured with a validated psychological test with five subscales: physical characteristics, talents and skills, psychological well-being, relations with the family and relations with others. A linear mixed effect model used the factors gender and adolescence group, and the continuous covariates: BMI z-scores, and BMI for the parents as fixed effects and subjects as random effects.Results: Age and gender, but neither the child’s BMI z-score nor the BMI of the parents were significant covariates. Self-esteem decreased (p < 0.01) with age on the global scale as well as on the subscales, and was below the normal level in higher ages in both genders. Girls had significantly lower self-esteem on the global scale (p = 0.04) and on the two subscales physical characteristics (p < 0.01) and psychological well-being (p < 0.01).Conclusion: Self-esteem is lower in girls and decreases with age. In treatment settings special attention should be paid to adolescent girls.

    • Quantum dots-in-a-well (DWELL) infrared photodetectors is a new class of nanophotonic devices with the potential of significantly increasing the performance and reducing the cost of infrared detectors. Here we present a comprehensive study of DWELL photodetector structures using a variety of optical techniques (PL, PLE, and PC). Complementary tunnel capacitance measurements support the electronic structure obtained from the optical measurements. A detailed energy level scheme based on the experimental findings is presented and compared to theoretical modeling. The presented work show the importance of combining different electrical and optical techniques to obtain a consistent model of complicated quantum structures which is crucial for the development of future nanophotonic devices.

  • Prediction of Outcome After Endovascular Embolectomy in Anterior Circulation Stroke Using Biomarkers.

    • Stroke is a major public health problem that can cause a long-term disability or death due to brain damage. Serious stroke is frequently caused by a large vessel occlusion in the anterior circulation, which should be treated by endovascular embolectomy if possible. In this study, we investigated the use of the brain damage biomarkers tau, NFL, NSE, GFAp, and S100B to understand the progression of nervous tissue damage and their relationship to outcome in such stroke after endovascular treatment. Blood samples were taken from 90 patients pre-treatment and 2 h, 24 h, 48 h, 72 h and 3 months after endovascular treatment. Stroke-related neurological deficit was estimated using the National Institute of Health Stroke Scale (NIHSS) at admission and at 24 h. Neurological outcome was evaluated at 3 months. After stroke, tau, NFL, GFAp and S100B increased in a time dependent manner, while NSE remained constant over time. At 3 months, tau and GFAp levels were back to normal whereas NFL was still high. Tau, NFL and GFAp correlated well to outcome, as well as to infarct volume and NIHSS at 24 h. The best time for prediction of poor outcome was different for each biomarker. However, the combination of NIHSS at 24 h with either tau, NFL or GFAp at 48 h gave the best prediction. The use of biomarkers in the early setting after endovascular treatment of stroke will lead to a simplified and standardized way to estimate the nervous tissue damage and possibly complement the clinical judgement in foreseeing the need of rehabilitation measures.

  • Evolution in boron-based GEM detectors for diffraction measurements : From planar to 3D converters

    • The so-called '3He-crisis' has motivated the neutron detector community to undertake an intense R&D programme in order to develop technologies alternative to standard 3He tubes and suitable for neutron detection systems in future spallation sources such as the European spallation source (ESS). Boron-based GEM (gas electron multiplier) detectors are a promising '3He-free' technology for thermal neutron detection in neutron scattering experiments. In this paper the evolution of boron-based GEM detectors from planar to 3D converters with an application in diffraction measurements is presented. The use of 3D converters coupled with GEMs allows for an optimization of the detector performances. Three different detectors were used for diffraction measurements on the INES instrument at the ISIS spallation source. The performances of the GEM-detectors are compared with those of conventional 3He tubes installed on the INES instrument. The conceptual detector with the 3D converter used in this paper reached a count rate per unit area of about 25% relative to the currently installed 3He tube. Its timing resolution is similar and the signal-to-background ratio (S/B) is 2 times lower.

    • At the center of Alzheimer's disease pathogenesis is the aberrant aggregation of amyloid-β (Aβ) into oligomers, fibrils and plaques. Effective monitoring of Aβ deposition directly in patients is essential to assist anti-Aβ therapeutics in target engagement and participant selection. In the advent of approved anti-Aβ therapeutics, biomarkers will become of fundamental importance in initiating treatments having disease modifying effects at the earliest stage. Two well-established Aβ biomarkers are widely utilized: Aβ-binding ligands for positron emission tomography and immunoassays to measure Aβ42 in cerebrospinal fluid. In this review, we will discuss the current clinical, diagnostic and research state of biomarkers for Aβ pathology. Furthermore, we will explore the current application of blood-based markers to assess Aβ pathology.

In the first part, the influence of shaft speed, grease type, and temperatures on the flow of lubricating greases in a narrow double restriction sealing pocket is evaluated. Such geometries can be found in, for example, labyrinth-type seals. In a wide pocket the velocity profile is one-dimensional and the Herschel-Bulkley model is used. In a narrow pocket, it is shown by the experimental results that the side walls have a significant influence on the grease flow, implying that the grease velocity profile is two-dimensional. In this area, a single empirical grease parameter for the rheology is sufficient to describe the velocity profile.In the second part, the radial migration of contaminant particles through the grease is evaluated. Centrifugal forces acting on a solid spherical particle are calculated from the grease velocity profile. Consequently, particles migrate to a larger radius and finally settle when the grease viscosity becomes large due to the low shear rate. This behavior is important for the sealing function of the grease in the pocket and relubrication

    • The essential amino acid L-tryptophan (Trp) is the immediate precursor of the neurotransmitter serotonin (5-HT). Supplementing Trp through diet has been shown to suppress the neuroendocrine stress response in vertebrates including teleosts. In salmonid fish, adjusting to the social environment as well as habituation to seawater involves the neuroendocrine stress response, suggesting that such environmental factors may modulate the stress-reducing effects of Trp. To date, studies that have investigated the neuroendocrine effects of dietary Trp have only been conducted in rainbow trout (Oncorhynchus mykiss), a salmonid species, under conditions featuring social isolation in the freshwater environment. Here, we address the effects of dietary Trp on post-stress plasma cortisol and hypothalamic monoamines in seawater-adapted Atlantic salmon (Salmo salar), reared at densities relevant for aquaculture. Fish were given feed containing 1, 2, 3 or 4 times the Trp content in normal feed for one week. Subsequently, the fish were reintroduced to feed containing the lowest Trp level, corresponding to standard commercial feed for a number of days prior to exposure to an acute confinement stressor. Basal plasma cortisol levels were lower among non-stressed fish at 1 and 10 days post dietary Trp supplementation. By comparison, stressed fish displayed stimulatory post-stress plasma cortisol responses at 1 and 2 days after the Trp regimen was terminated. However, a reversed pattern was observed among these fish at 10 days after Trp treatment. The overall effects of dietary Trp were more pronounced in dopamine (DA) neurochemistry compared to 5-HT in the hypothalamus. The results demonstrate both short-and long-term effects of elevated dietary Trp on the neuroendocrine stress response. These findings suggest that hypothalamic DA may be more involved than 5-HT in the stress reducing effects of Trp in seawater-adapted Atlantic salmon, reared at densities relevant for aquaculture.

  • Dynamic ambulance relocation : a scoping review

    • Objectives Dynamic ambulance relocation means that the operators at a dispatch centre place an ambulance in a temporary location, with the goal of optimising coverage and response times in future medical emergencies. This study aimed to scope the current research on dynamic ambulance relocation.Design A scoping review was conducted using a structured search in PubMed, Scopus and Web of Science. In total, 21 papers were included.Results Most papers described research with experimental designs involving the use of mathematical models to calculate the optimal use and temporary relocations of ambulances. The models relied on several variables, including distances, locations of hospitals, demographic-geological data, estimation of new emergencies, emergency medical services (EMSs) working hours and other data. Some studies used historic ambulance dispatching data to develop models. Only one study reported a prospective, real-time evaluation of the models and the development of technical systems. No study reported on either positive or negative patient outcomes or real-life chain effects from the dynamic relocation of ambulances.Conclusions Current knowledge on dynamic relocation of ambulances is dominated by mathematical and technical support data that have calculated optimal locations of ambulance services based on response times and not patient outcomes. Conversely, knowledge of how patient outcomes and the working environment are affected by dynamic ambulance dispatching is lacking. This review has highlighted several gaps in the scientific coverage of the topic. The primary concern is the lack of studies reporting on patient outcomes, and the limited knowledge regarding several key factors, including the optimal use of ambulances in rural areas, turnaround times, domino effects and aspects of working environment for EMS personnel. Therefore, addressing these knowledge gaps is important in future studies.

    • Delayed diagnosis and misdiagnosis are frequent in people with amyotrophic lateral sclerosis (ALS), the most common form of motor neuron disease (MND). Neurofilament light chain (NFL) and phosphorylated neurofilament heavy chain (pNFH) are elevated in ALS patients. We retrospectively quantified cerebrospinal fluid (CSF) NFL, CSF pNFH and plasma NFL in stored samples that were collected at the diagnostic work-up of ALS patients (n=234), ALS mimics (n=44) and controls (n=9). We assessed the diagnostic performance, prognostication value and relationship to the site of onset and genotype. CSF NFL, CSF pNFH and plasma NFL levels were significantly increased in ALS patients compared to patients with neuropathies & myelopathies, patients with myopathies and controls. Furthermore, CSF pNFH and plasma NFL levels were significantly higher in ALS patients than in patients with other MNDs. Bulbar onset ALS patients had significantly higher plasma NFL levels than spinal onset ALS patients. ALS patients with C9orf72HREmutations had significantly higher plasma NFL levels than patients withSOD1mutations. Survival was negatively correlated with all three biomarkers. Receiver operating characteristics showed the highest area under the curve for CSF pNFH for differentiating ALS from ALS mimics and for plasma NFL for estimating ALS short and long survival. Allthree biomarkers have diagnostic value in differentiating ALS from clinically relevant ALS mimics. Plasma NFL levels can be used to differentiate between clinical and genetic ALS subgroups.

  • Cerebrospinal fluid tau fragment correlates with tau PET : a candidate biomarker for tangle pathology

    • To date, there is no validated fluid biomarker for tau pathology in Alzheimer's disease, with contradictory results from studies evaluating the correlation between phosphorylated tau in CSF with tau PET imaging. Tau protein is subjected to proteolytic processing into fragments before being secreted to the CSF. A recent study suggested that tau cleavage after amino acid 368 by asparagine endopeptidase (AEP) is upregulated in Alzheimer's disease. We used immunoprecipitation followed by mass spectrometric analyses to evaluate the presence of tau368 species in CSF. A novel Simoa® assay for quantification of tau368 in CSF was developed, while total tau (t-tau) was measured by ELISA and the presence of tau368 in tangles was evaluated using immunohistochemistry. The diagnostic utility of tau368 was first evaluated in a pilot study (Alzheimer's disease = 20, control = 20), then in a second cohort where the IWG-2 biomarker criteria were applied (Alzheimer's disease = 37, control = 45), and finally in a third cohort where the correlation with 18F-GTP1 tau PET was evaluated (Alzheimer's disease = 38, control = 11). The tau368/t-tau ratio was significantly decreased in Alzheimer's disease (P < 0.001) in all cohorts. Immunohistochemical staining demonstrated that tau fragments ending at 368 are present in tangles. There was a strong negative correlation between the CSF tau368/t-tau ratio and 18F-GTP1 retention. Our data suggest that tau368 is a tangle-enriched fragment and that the CSF ratio tau368/t-tau reflects tangle pathology. This novel tau biomarker could be used to improve diagnosis of Alzheimer's disease and to facilitate the development of drug candidates targeting tau pathology. Furthermore, future longitudinal studies will increase our understanding of tau pathophysiology in Alzheimer's disease and other tauopathies.

    • Type I interferons constitute a family of pleiotropic cytokines that have a key role in both adaptive and innate immunity. The interferon signalling pathways mediate transcriptional regulation of hundreds of genes, which result in mRNA degradation, decreased protein synthesis, cell cycle inhibition and induction of apoptosis. To elucidate regulatory networks important for interferon induced cell death, we generated interferon resistant U937 cells by selection in progressively increasing concentrations of interferon-α (IFN-α). The results show that IFN-α activates the death receptor signalling pathway and that IFN resistance was associated with cross-resistance to several death receptor ligands in a manner similar to previously described Fas resistant U937 cell lines. Increased expression of the long splice variant of the cellular FLICE-like inhibitor protein (cFLIP-L) was associated with the resistance to death receptor and IFN-α stimulation. Accordingly, inhibition of cFLIP-L expression with cycloheximide or through cFLIP short harpin RNA interference restored sensitivity to Fas and/or IFN-α. Thus, we now show that selection for interferon resistance can generate cells with increased expression of cFLIP, which protects the cells from both IFN-α and death receptor mediated apoptosis.

    • Susceptibility to cell death is a prerequisite for the elimination of tumour cells by cytotoxic immune cells, chemotherapy or irradiation. Activation of the death receptor Fas is critical for the regulation of immune cell homeostasis and efficient killing of tumour cells by apoptosis. To define the molecular changes that occur during selection for insensitivity to Fas-induced apoptosis, a resistant variant of the U937 cell line was established. Individual resistant clones were isolated and characterized. The most frequently observed defect in the resistant cells was reduced Fas expression, which correlated with decreased FAS transcription. Clones with such reduced Fas expression also displayed partial cross-resistance to tumour necrosis factor-alpha stimulation, but the mRNA expression of tumour necrosis factor receptors was not decreased. Reintroduction of Fas conferred susceptibility to Fas but not to tumour necrosis factor-alpha stimulation, suggesting that several alterations could be present in the clones. The reduced Fas expression could not be explained by mutations in the FAS coding sequence or promoter region, or by silencing through methylations. Protein kinase B and extracellular signal-regulated kinase, components of signalling pathways downstream of Ras, were shown to be activated in some of the resistant clones, but none of the three RAS genes was mutated, and experiments using chemical inhibitors could not establish that the activation of these proteins was the cause of Fas resistance as described in other systems. Taken together, the data illustrate that Fas resistance can be caused by reduced Fas expression, which is a result of an unidentified mode of regulation.

    • The results from the analysis of the course evaluations from FELASA category Ccompulsory courses in laboratory animal science at Uppsala University showed that thestudents realize that theoretical knowledge of the subject and practical skills are of great importance to the success of their future research involving animal experiments. All the subjects and elements of the course, in particular the practicals on animal handling and procedures using live anaesthetized animals, were fully appreciated by the students.

    • Tau is an axonal microtubule-binding protein. Tau pathology in brain and increased tau concentration in the cerebrospinal fluid (CSF) are hallmarks of Alzheimer’s disease (AD). Most of tau in CSF is present as fragments. We immunoprecipitated tau from CSF and identified several endogenous peptides ending at amino acid (aa) 123 or 224 using high-resolution mass spectrometry. We raised neo-epitope-specific antibodies against tau fragments specifically ending at aa 123 and 224, respectively. With these antibodies, we performed immunohistochemistry on brain tissue and designed immunoassays measuring N-123, N-224, and x-224 tau. Immunoassays were applied to soluble brain fractions from pathologically confirmed subjects (81 AD patients, 33 controls), CSF from three cross-sectional and two longitudinal cohorts (a total of 133 AD, 38 MCI, 20 MCI-AD, 31 PSP, 15 CBS patients, and 91 controls), and neuronally- and peripherally-derived extracellular vesicles (NDEVs and PDEVs, respectively) in serum from four AD patients and four controls. Anti-tau 224 antibody stained neurofibrillary tangles and neuropil threads, while anti-tau 123 only showed weak cytoplasmic staining in AD. N-224 tau was lower in the AD soluble brain fraction compared to controls, while N-123 tau showed similar levels. N-224 tau was higher in AD compared to controls in all CSF cohorts (p < 0.001), but not N-123 tau. Decrease in cognitive performance and conversion from MCI to AD were associated with increased baseline CSF levels of N-224 tau (p < 0.0001). N-224 tau concentrations in PSP and CBS were significantly lower than in AD (p < 0.0001) and did not correlate to t-tau and p-tau. In a longitudinal cohort, CSF N-224 tau levels were stable over 6 months, with no significant effect of treatment with AChE inhibitors. N-224 tau was present in NDEVs, while N-123 tau showed comparable concentrations in both vesicle types. We suggest that N-123 tau is produced both in CNS and PNS and represents a general marker of tau metabolism, while N-224 tau is neuron-specific, present in the tangles, secreted in CSF, and upregulated in AD, suggesting a link between tau cleavage and propagation, tangle pathology, and cognitive decline.

    • new position-sensitive thermal neutron detector based on boron-coated converters has been developed as an alternative to today's standard He-3-based technology for application to thermal neutron scattering. The key element of the development is a novel 3D (B4C)-B-10 converter which has been ad hoc designed and realized with the aim of combining a high neutron conversion probability via the B-10(n, alpha)(7) Li reaction together with an efficient collection of the produced charged particles. The developed 3D converter is composed of thin aluminium grids made by a micro-waterjet technique and coated on both sides with a thin layer of( 10)B(4)C. When coupled to a GEM detector this converter allows reaching neutron detection efficiencies close to 50% at neutron wavelengths equal to 4 angstrom. In addition, the new detector features a spatial resolution of about 5 min and can sustain counting rates well in excess of 1 MHz/cm(2). The newly developed neutron detector will enable time-resolved measurements of different kind of samples in neutron scattering experiments at high flux spallation sources and can find a use in applications where large areas and custom geometries of thermal neutron detectors are foreseen.

  • Genetic and Ecological Consequences of Fish Releases : With Focus on Supportive Breeding of Brown Trout Salmo trutta and Translocation of European Eel Anguilla anguilla

    • Although the practice of releasing fish into the wild is common in the management and conservation of fish populations, the success of release programmes and the potential harmful genetic and ecological effects that may follow are rarely considered. This thesis focuses on genetic and ecological consequences of fish releases, exemplified by supportive breeding of brown trout (Salmo trutta) and translocation of European eel (Anguilla anguilla). Specific questions addressed include: What is the relative performance of hatchery produced fish released to support wild populations, and do released hatchery fish contribute to the natural productivity? What is the variation in reproductive success in the wild, and how does it affect the genetic consequences of a supportive breeding programme? Is there a spatial genetic structure in the European eel that must be considered in the management of this rapidly declining species?Experiments conducted under natural and near-natural conditions in the River Dalälven, Sweden, suggest that hatchery produced trout can reproduce in the wild. In fact, when the pronounced variation between individual breeders was accounted for, there were no detectable differences between hatchery produced and wild born trout in reproductive success or offspring survival. These results were supported by molecular genetic data suggesting a pronounced gene flow from hatchery to wild trout in the river. Hatchery reared trout were, however, found to exhibit reduced survival rates immediately following release into the wild, an effect that was most likely due to phenotypic responses to the hatchery environment during ontogeny and a lack of experience of the wild.In sharp contrast to recently published studies, the present genetic analyses of European eels sampled across the whole distribution range suggest no spatial genetic structure but a subtle temporal genetic heterogeneity within sampled locations. These results emphasise the need to consider temporal replication when assessing population structure of marine species.The results obtained have general implications for the management and conservation of fish populations. First, supportive breeding of threatened salmonid populations might be successful, not only for boosting the census size and thereby reducing the short-term probability of extinction, but also for reducing the risks of inbreeding depression and loss of adaptive potential in future generations. However, the results also highlight the need to restore the natural productivity of a population under supportive breeding to avoid a potential reduction in fitness due to hatchery selection. Further, the lack of a detectable spatial genetic structure in the European eel suggests that the management strategy of translocating juvenile eels from locations were they are overabundant to other suitable freshwater habitats does not necessarily have to include genetic considerations with respect to the geographical origin of the translocated eels.

    • In this study, the flow of lubricating greases in a labyrinth seal geometry is studied using microparticle image velocimetry (µPIV). The aim is to evaluate the grease velocity distribution inside the gap of a labyrinth seal and to find a relationship between the grease consistency and the transferred speed from the rotating ring in order to choose the correct grease as a sealing medium. In addition, the grease flow characteristics are important for the understanding of fracture due to grease layer displacement. For these purposes, four greases with different rheological properties were used in µPIV experiments. It was found that the grease consistency plays a crucial role in speed development as well as the grease composition and presence of a slip effect at the grease–rotating wall interface.

    • Introduction: Osteolysis induced by wear particles in metal-on-polyethylene hip implants has been the key motivation to look for alternative bearings and in fact emergence and development of new metal-on-metal (MOM) implant materials for joint replacement. However, while the volume of wear particles produced in metal-on-metal articulations is lower the number of particles produced is higher per volume of wear, due to the reduced size of wear particles. Although various surface and interface characterization methods have been applied to study the physical wear, corrosion and implant surface interactions with biological environments, presently the local and systematic effects of metal debris are poorly understood. Materials and Methods: Cobalt-chromium-molybdenium (CoCr) alloys have been used in MOM implants extensively. Metallic samples were cut and mirror polished. In the present study The samples were immersed in four different biological lubricants (Human serum, synovial fluid, MEM and Milli-Q water) for 10 min, 1 hr, and 5 days of immersion and then studied by X-ray Photoelectron Spectroscopy (XPS) and time-of-flight secondary ion mass spectroscopy (ToF-SIMS). XPS determined the chemistry of elements located whitin the top few nanometers of materials. Significant differences in the absorbed layers and differences in the corrosive nature of Ti and CoCr implant substrates and wear particles were found. Results and discussion: Spectra from P 2p3/2, O1s, Ca2p3/2, C1s and N1s were collected. Metallic substrates behaved differently when immersed in the same lubricant. The four lubricants reacted different with metallic surfaces. Larger calcium deposits occurred in supersaturated physiological solutions. Deposition of calcium phosphate was different on CoCr alloys depending on the lubricant and the immersion period. Specimens immersed into synovial fluid gave thinner oxide layers and lower calcium phosphate deposits. For all specimens, water immersion resulted in thicker oxide layer. For many reactive metals, dissolution of ions from the metal surface takes place along with thickening of the metal oxide during passivation, or surface corrosion.Conclusion: Glycoaminoglycans (GAG) and related proteins may hinder calcium phosphate deposition on samples immersed in synovial fluid. ToF-SIMS measurements showed that the resulting corrosion products depend upon the nature of the environment. The thickness of the calcium phosphate deposits was different for different metal substrate.

    • Purpose: The purpose of this study was to assess the incidence of acute symptomatic seizures and poststroke epilepsy (PSE) in a well-characterized cohort of patients treated with mechanical thrombectomy. In addition, we aimed to describe the dynamics of blood markers of brain injury in patients that developed PSE. Methods: Participants of the prospective AnStroke Trial of anesthesia method during mechanical thrombectomy were included and acute symptomatic seizures and PSE ascertained by medical records review. Blood markers neurofilament light (NFL), tau, glial fibrillary acidic protein (GFAP), S100 calcium-binding protein B (S100B), and neuron-specific enolase (NSE) were assessed. Results: A total of 90 patients with acute anterior ischemic stroke were included. Median National Institutes of Health Stroke Scale (NIHSS) at admission to hospital was 18 (IQR 15–22). Recanalization was achieved in 90%. No patients had epilepsy prior to the ischemic stroke. Four patients (4.4%) had acute symptomatic seizures and four patients (4.4%) developed PSE during the follow-up time (to death or last medical records review) of 0–4.5 years (median follow-up 1070 days IQR 777–1306), resulting in a two-year estimated PSE risk of 5.3% (95%CI: 0.2–10.4%). Blood markers of brain injury (NFL, tau, GFAP, S100B, and NSE) were generally above the cohort median in patients that developed PSE. Conclusions: The incidence of PSE after mechanical thrombectomy was low in our cohort. All blood biomarkers displayed interesting sensitivity and specificity. However, the number of PSE cases was small and more studies are needed on risk factors for PSE after mechanical thrombectomy. The potential of blood markers of brain injury markers to contribute to assessment of PSE risk should be explored further. This article is part of the Special Issue "Seizures & Stroke".

    • Objective Our aim was to analyse the chromogranin A-derived peptides vasostatin and catestatin in serum from wild boar (Sus scrofa) captured in a corral trap. Acute capture-related stress quickly leads to a release of adrenalin and noradrenalin, but these hormones have a short half-life in blood and are difficult to measure. Chromogranin A (CgA), a glycoprotein which is co-released with noradrenalin and adrenalin, is relatively stable in circulation and the CgA-derived peptides catestatin and vasostatin have been measured in domestic species, but not yet in wildlife. Results Vasostatin and catestatin could be measured and the median (range) serum concentrations were 0.91 (0.54-2.86) and 0.65 (0.35-2.62) nmol/L, respectively. We conclude that the CgA-derived peptides vasostatin and catestatin can be measured in wild boar serum and may thus be useful as biomarkers of psychophysical stress.

    • Background Advances in the treatment of brain tumours have increased the number of long-term survivors, but at the cost of side effects following cranial radiotherapy ranging from neurocognitive deficits to outright tissue necrosis. At present, there are no tools reflecting the molecular mechanisms underlying such side effects, and thus no means to evaluate interventional effects after cranial radiotherapy. Therefore, fluid biomarkers are of great clinical interest. Objective Cerebrospinal fluid (CSF) levels of proteins involved in inflammatory signalling, synaptic plasticity and extracellular matrix (ECM) integrity were investigated following radiotherapy to the brain. Methods Patients with small-cell lung cancer (SCLC) eligible for prophylactic cranial irradiation (PCI) were asked to participate in the study. PCI was prescribed either as 2 Gy/fraction to a total dose of 30 Gy (limited disease) or 4 Gy/fraction to 20 Gy (extensive disease). CSF was collected by lumbar puncture at baseline, 3 months and 1 year following PCI. Protein concentrations were measured using immunobased assays or mass spectrometry. Results The inflammatory markers IL-15, IL-16 and MCP-1/CCL2 were elevated in CSF 3 months following PCI compared to baseline. The plasticity marker GAP-43 was elevated 3 months following PCI, and the same trend was seen for SNAP-25, but not for SYT1. The investigated ECM proteins, brevican and neurocan, showed a decline following PCI. There was a strong correlation between the progressive decline of soluble APP and brevican levels. Conclusion To our knowledge, this is the first time ECM-related proteins have been shown to be affected by cranial radiotherapy in patients with cancer. These findings may help us to get a better understanding of the mechanisms behind side effects following radiotherapy.

    • Tannins are polymeric, phenolic constituents found in the bark of pine and spruce. When reacting with iron ions, tannins form strongly coloured complexes. Thus, the presence of bark in the mechanical pulping process leads to decreased brightness of the pulp. In order to evaluate the effects of the presence of iron on the properties of pulp, we have impregnated thermomechanical pulp (TMP) with 30 parts per million (ppm i.e. mg/kg) iron either as Fe3+ or as tannin-iron complexes and studied how such treatments affect bleachability and heat-induced brightness reversion. The bleaching agents studied are hydrogen peroxide and sodium dithionite. Treatment of the tannin-iron impregnated pulp with 1% by weight of diethylenetriaminepentaacetic acid (DTPA) before bleaching with 4% hydrogen peroxide almost eliminated the brightness loss caused by the impregnation. Such a treatment also removed all of the added iron from both the tannin-iron and FeCl3 impregnated pulps. Approximately 5% more of the added peroxide was required for oxidation of the tannins in the tannin-iron impregnated pulp. Contrary to what was observed with peroxide bleaching, dithionite bleaching did not reduce the amount of iron in the pulps. Instead, the added iron and tannin-iron negatively affected the dithionite bleaching, even if the pulps were extracted with DTPA before bleaching. It should therefore be advantageous to first bleach with peroxide, which removes most of the iron, and then with dithionite. Compared with dithionite, peroxide yields a more efficient bleaching. The reason for this is that the former reduces the light absorption coefficient, the k-value, more efficiently in the whole visible spectrum, whereas dithionite reduces it mainly at shorter wavelengths. In our experiments, the addition of tannin-iron or FeCl3 to the untreated pulp did not increase heat-induced brightness reversion. This is Supported by the fact that although extraction of the samples with DTPA before bleaching lowered the iron content slightly, it-did not affect the brightness reversion. The initial brightness reversion of the dithionite bleached pulps was larger than that observed for the peroxide bleached pulps.

    • Tannins are polyphenolic compounds found mainly in bark. When reacting with iron, they form strongly coloured complexes, which through contamination from the bark may induce a brightness decrease of mechanical pulps. Wood itself contains phenolic compounds, which can form coloured complexes with iron. We have investigated gallotannin as a model for metal-binding sites in the pulp. The behaviour of tannin-iron complexes in solution and in pulp has been studied. In aqueous solution, the tannin-iron complexes can be decolourised by the addition of diethylenetriaminepentaacetic acid (DTPA). The colour of the tannin-iron complexes was very pH-dependent. Thus in solution, these were decolourised at low pH and at high pH the spectral characteristics were changed substantially. We have studied the effects on brightness and heatinduced brightness loss of the impregnation of thermomechanical pulp (TMP) with 30 parts per million iron (ppm i.e. mg/kg) either as iron or tannin-iron as well as the possibility to decrease such effects by using various solvent extractions. The tannin-iron impregnation causes a decrease in ISO-brightness of approximately 3% and an increase in the light absorption coefficient (k) by approximately 2 m(2)/kg at the tannin-iron absorbance maximum. 565 run. These effects are approximately ten times higher than those observed for the Pulp only impregnated with iron. Extraction with 1% by weight of DTPA provides a way to reduce the brightness decrease induced by the tanniniron complexes and the observed decrease can be attributed to removal of iron from the pulp. Acid extraction was the most efficient way to reduce the iron content in the pulps and to decoulorise the tannin-iron impregnated pulp. However, after acid extraction of iron impregnated Pulps, new chromophores were evidently formed. Addition of the reducing agent, sulphite, to extractions had no effect on the iron removal or the brightness of the impregnated pulps. The heat-induced brightness loss is not influenced by the addition of tannin-iron or iron. The brightness loss caused by heat was lower for pulps extracted with DTPA.

    • There are indications that some environmentally friendly oils provide performance advantages in hydrodynamic bearings compared to mineral oils. To verify and to quantify the advantages of the synthetic oils in hydrodynamic lubrication in terms of operating film thickness, bearing power loss and operating temperature, a test rig for laboratory experiments has been designed and manufactured. At the first stage of the project temperature and pressure distributions, power loss and oil film thickness for ISO VG46 and VG68 mineral and synthetic oils will be measured at different loads and rotational speeds. At the next stage several other lubricants will be tested. Among them rape seed oil and pine tree oil can be mentioned.

    • A constant trend towards more compact mechanical systems with higher power densities and increased thermo-mechanical loads emphasises the importance of the development of new design approaches and novel tribological systems. Ignoring this may cause a significant slow down in technological and industrial development. Tribotronics or active tribology based on adaptive performance is thought of as being critical in the implementation of smart machine concepts. Recognition of the importance of tribotronics, or active control of system loss outputs, such as those through friction and wear will have significant beneficial economic consequences as a result of the associated accelerated rate of technological progress. These smart tribotronic systems can be embedded in a great variety of machines and mechanisms. If this integration is made at the design stage, products that are more flexible, efficient and reliable can be produced. The concept of tribotronics is presented and discussed in this paper. Some illustrativeexamples that show the feasibility of an “active” approach are given. In addition, various possibilities already reported in literature are discussed.

    • A new method to visualize and quantify grease flow in between two sealing lips or, in general, a double restriction seal is presented. Two setups were designed to mimic different types of seals; that is, a radial and an axial shaft seal. The flow of the grease inside and in between the sealing restrictions was measured using microparticle image velocimetry. The results show that grease flow due to a pressure difference mainly takes place close to the rotating shaft surface with an exponentially decaying velocity profile in the radial direction. Consequently, contaminants may be captured in the stationary grease at the outer radius, which explains the sealing function of the grease.

    • Determining lubricant film thickness between contacting bodies under elastohydrodynamic (EHD) conditions is often simulated by using a ball/cylinder and transparent disc apparatus together with an interferometry technique. The simulated contact will have a point or elliptic shape and the light used can be white or monochromatic. The interference pattern is normally photographed with a regular camera or a video camera and the pictures are then evaluated by the naked eye of the observer. In most cases, only central or minimum thicknesses are evaluated.In this paper an image processing method for the analysis of film thickness is presented. This method makes it possible to extract considerably more information about film thickness fluctuations than is achievable by the naked eye. The method primarily matches hue (but also saturation and intensity values) from digitized colour interferometric images of the unknown film shapes with calibration values obtained with known geometric shapes.The method is shown to work well in the range from 95 up to 700 nm with white light and makes the results unbiased by the observer. Furthermore, absolute film thickness can be evaluated without prior knowledge about the fringe order in the interferogram.

    • Biomarkers of neurodegenerative disorders are needed to assist in diagnosis, to monitor disease progression and therapeutic interventions, and to provide insight into disease mechanisms. One route to identify such biomarkers is by proteomic and peptidomic analysis of cerebrospinal fluid (CSF). In the current study, we performed an in-depth analysis of the human CSF endopeptidome to establish an inventory thatmay serve as a basis for future targeted biomarker studies. High-pH RP HPLC was employed for off-line sample prefractionation followed by low-pH nano-LC-MS analysis. Different software programs and scoring algorithms for peptide identification were employed and compared. A total of 18 031 endogenous peptides were identified at a FDR of 1%, increasing the number of known endogenous CSF peptides 10fold compared to previous studies. The peptides were derived from 2 053 proteins of which more than 60 have been linked to neurodegeneration. Notably, among the findings were six peptides derived from microtubule-associated protein tau, three of which span the diagnostically interesting threonine-181 (Tau-F isoform). Also, 213 peptides from amyloid precursor protein were identified, 58 of which were partially or completely within the sequence of amyloid beta 1-40/42, as well as 109 peptides from apolipoprotein E, spanning sequences that discriminate between the E2/E3/E4 isoforms of the protein.

    • Patients with secondary acute myeloid leukemia (AML) often escape inclusion in clinical trials and thus, population-based studies are crucial for its accurate characterization. In this first large population-based study on secondary AML, we studied AML with an antecedent hematological disease (AHD-AML) or therapy-related AML (t-AML) in the population-based Swedish Acute Leukemia Registry. The study included 3,363 adult patients of which 2,474 (73.6%) had de novo AML, 630 (18.7%) AHD-AML, and 259 (7.7%) t-AML. Secondary AML differed significantly compared to de novo AML with respect to age, gender, and cytogenetic risk. Complete remission (CR) rates were significantly lower but early death rates similar in secondary AML. In a multivariable analysis, AHD-AML (HR 1.51; 95% CI 1.26-1.79) and t-AML (1.72; 1.38-2.15) were independent risk factors for poor survival. The negative impact of AHD-AML and t-AML on survival was highly age dependent with a considerable impact in younger patients, but without independent prognostic value in the elderly. Although patients with secondary leukemia did poorly with intensive treatment, early death rates and survival were significantly worse with palliative treatment. We conclude that secondary AML in a population-based setting has a striking impact on survival in younger AML patients, whereas it lacks prognostic value among the elderly patients. Am. J.     • Introduction. The aim of this prospective study was to investigate excessive daytime sleepiness (EDS) over time and in relation to other PD symptoms among people with Parkinson's disease (PD). Methods. Thirty participants younger than 65 years with PD were randomly selected. At inclusion, mean (SD) disease duration was 6.2 (4.8) years and median (min-max) severity of PD was classified as stage II (stages I-III) according to Hoehn and Yahr. Participants were followed annually for 10 years with clinical assessments of their PD status, medications, comorbidities, and a standardized interview about their sleep habits and occurrence of daytime sleepiness. EDS was assessed by the self-reported Epworth Sleepiness Scale (ESS). Seventeen participants completed the 10-year longitudinal follow-up. Results. Fifteen of 30 persons were classified to suffer from EDS (ESS > 10) at baseline. At the group level, EDS remained stable over 10 years and did not deteriorate in parallel with worsening of motor symptoms. Furthermore,EDS was associated with sleep quality, fatigue, anxiety, depression, and axial/postural/gait impairments. Conclusions. EDS did not worsen over 10 years, although other PD aspects did. EDS in PD seems to be a complex nonmotor symptom that is unrelated to deterioration of motor symptoms in PD.
