  • Physical properties of sea ice cores from site MCS-SYI measured on legs 1 to 3 of the MOSAiC expedition

    • We present sea ice temperature and salinity data from first-year ice (FYI) and second-year ice (SYI) relevant to the temporal development of sea ice permeability and brine drainage efficiency from the early growth phase in October 2019 to the onset of spring warming in May 2020. Our dataset was collected in the central Arctic Ocean during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) Expedition in 2019 to 2020. MOSAiC was an international transpolar drift expedition in which the German icebreaker RV Polarstern anchored into an ice floe to gain new insights into Arctic climate over a full annual cycle. In October 2019, RV Polarstern moored to an ice floe in the Siberian sector of the Arctic at 85 degrees north and 137 degrees east to begin the drift towards the North Pole and the Fram Strait via the Transpolar Drift Stream. The data presented here were collected during the first three legs of the expedition, so all the coring activities took place on the same floe. The end dates of legs 1, 2, and 3 were 13 December, 24 February, and 4 June, respectively. The dataset contributed to a baseline study entitled, Deciphering the properties of different Arctic ice types during the growth phase of the MOSAiC floes: Implications for future studies. The study highlights downward directed gas pathways in FYI and SYI by inferring sea ice permeability and potential brine release from several time series of temperature and salinity measurements. The physical properties presented in this paper lay the foundation for subsequent analyses on actual gas contents measured in the ice cores, as well as air-ice and ice-ocean gas fluxes. Sea ice cores were collected with a Kovacs Mark II 9 cm diameter corer. To measure ice temperatures, about 4.5 cm deep holes were drilled into the core (intervals varied by site and leg) . The temperatures were measured by a digital thermometer within minutes after the cores were retrieved. The ice cores were placed into pre-labelled plastic sleeves sealed at the bottom end. The ice cores were transported to RV Polarstern and stored in a -20 degrees Celsius freezer. Each of the cores was sub-sampled, melted at room temperature, and processed for salinity within one or two days. The practical salinity was estimated by measuring the electrical conductivity and temperature of the melted samples using a WTW Cond 3151 salinometer equipped with a Tetra-Con 325 four-electrode conductivity cell. The practical salinity represents the the salinity estimated from the electrical conductivity of the solution. The dataset also contains derived variables, including sea ice density, brine volume fraction, and the Rayleigh number.

    • OBJECTIVE: To monitor the concentration of markers of cartilage and synovium metabolism in the knee (stifle) joint synovial fluid of young beagles subjected to immobilisation and subsequent remobilisation.METHODS: The right hind limb of 17 dogs was immobilised in flexion for 11 weeks. Simultaneously, the contralateral left knee was exposed to increased weight bearing. The remobilisation period lasted 50 weeks. Litter mates served as controls. The concentration in joint lavage fluid of interleukin 1 (IL1) was measured by immunoassay, the activity of phospholipase A2 (PLA2) was determined by an extraction method, chondroitin sulphate (CS) concentration by precipitation with Alcian blue, hyaluronan (HA) by an ELISA-like assay using biotinylated HA-binding complexes, matrix metalloproteinase 3 (MMP-3), and tissue inhibitor of metalloproteinases 1 (TIMP-1) by sandwich ELISA, and synovitis was scored by light microscopy.RESULTS: Synovitis or effusion was absent in all experimental and control groups. Immobilisation decreased the joint lavage fluid levels of IL1 (p<0.05), TIMP (p< 0.05), and the concentration of CS down to 38 (p<0.05) in comparison with untreated litter mates with normal weight bearing. Immobilisation did not affect the activity of PLA2, or the concentration of MMP-3 or HA in synovial fluid. Joint remobilisation restored the decreased concentrations of markers to control levels. Increased weight bearing did not change the concentrations of markers in comparison with the control joints with normal weight bearing.CONCLUSIONS: 11 weeks joint immobilisation decreased the concentration of markers of cartilage and synovium metabolism in the synovial fluid, and remobilisation restored the concentrations to control levels. The changes in joint metabolism induced by immobilisation, as reflected by the markers, are thus different from those found in osteoarthritis, where increased levels of these markers are associated with enhanced degradation and synthesis. These findings suggest that the change induced in joint metabolism by immobilisation is reversible in its early stages.

  • Characterization and antifungal properties against wood decaying fungi of hydrothermal liquefaction liquids from spent mushroom substrate and tomato residues

    • This study aimed to investigate the potential of converting bio-based residues from industrial production of mushrooms and tomatoes into more valuable chemicals with antifungal properties using hydrothermal liquefaction (HTL). Liquid fractions were obtained from HTL of spent substrate of Agaricus bisporus (Lange) Imbach and Pleurotus ostreatus (jacq.) P. kumm., recomposted Agaricus bisporus spent substrate, and tomato residues. The quantitative 1H NMR spectroscopy analysis revealed that the HTL liquids of all residues contained antifungal constituents like phenols and organic acids. The HTL liquids at dilutions of 10 % were able to inhibit the fungi by over 80 %. Interestingly, the fungus P. ostreatus showed tolerance to these constituents as its growth was promoted at the lowest concentration of all the HTL liquids. The HTL liquids had lower ecotoxicity than the commercial wood preservative. These results suggest that the tested residues could be a promising source of preservative chemical constituents for the wood industry.

  • The MOSAiC Distributed Network: Observing the coupled Arctic system with multidisciplinary, coordinated platforms

    • Central Arctic properties and processes are important to the regional and global coupled climate system. The Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) Distributed Network (DN) of autonomous ice-tethered systems aimed to bridge gaps in our understanding of temporal and spatial scales, in particular with respect to the resolution of Earth system models. By characterizing variability around local measurements made at a Central Observatory, the DN covers both the coupled system interactions involving the ocean-ice-atmosphere interfaces as well as three-dimensional processes in the ocean, sea ice, and atmosphere. The more than 200 autonomous instruments (“buoys”) were of varying complexity and set up at different sites mostly within 50 km of the Central Observatory. During an exemplary midwinter month, the DN observations captured the spatial variability of atmospheric processes on sub-monthly time scales, but less so for monthly means. They show significant variability in snow depth and ice thickness, and provide a temporally and spatially resolved characterization of ice motion and deformation, showing coherency at the DN scale but less at smaller spatial scales. Ocean data show the background gradient across the DN as well as spatially dependent time variability due to local mixed layer sub-mesoscale and mesoscale processes, influenced by a variable ice cover. The second case (May–June 2020) illustrates the utility of the DN during the absence of manually obtained data by providing continuity of physical and biological observations during this key transitional period. We show examples of synergies between the extensive MOSAiC remote sensing observations and numerical modeling, such as estimating the skill of ice drift forecasts and evaluating coupled system modeling. The MOSAiC DN has been proven to enable analysis of local to mesoscale processes in the coupled atmosphere-ice-ocean system and has the potential to improve model parameterizations of important, unresolved processes in the future.

  • Antiviral action of a functionalized plastic surface against human coronaviruses

    • Viruses may persist on solid surfaces for long periods, which may contribute to indirect transmission. Thus, it is imperative to develop functionalized surfaces that will lower the infectious viral load in everyday life. Here, we have tested a plastic surface functionalized with tall oil rosin against the seasonal human coronavirus OC43 as well as severe acute respiratory syndrome coronavirus 2. All tested non-functionalized plastic surfaces showed virus persistence up to 48 h. In contrast, the functionalized plastic showed good antiviral action already within 15 min of contact and excellent efficacy after 30 min over 90% humidity. Excellent antiviral effects were also observed at lower humidities of 20% and 40%. Despite the hydrophilic nature of the functionalized plastic, viruses did not adhere strongly to it. According to helium ion microscopy, viruses appeared flatter on the rosin-functionalized surface, but after flushing away from the rosin-functionalized surface, they showed no apparent structural changes when imaged by transmission electron microscopy of cryogenic or negatively stained specimens or by atomic force microscopy. Flushed viruses were able to bind to their host cell surface and enter endosomes, suggesting that the fusion with the endosomal membrane was halted. The eluted rosin from the functionalized surface demonstrated its ability to inactivate viruses, indicating that the antiviral efficacy relied on the active leaching of the antiviral substances, which acted on the viruses coming into contact. The rosin-functionalized plastic thus serves as a promising candidate as an antiviral surface for enveloped viruses.IMPORTANCEDuring seasonal and viral outbreaks, the implementation of antiviral plastics can serve as a proactive strategy to limit the spread of viruses from contaminated surfaces, complementing existing hygiene practices. In this study, we show the efficacy of a rosin-functionalized plastic surface that kills the viral infectivity of human coronaviruses within 15 min of contact time, irrespective of the humidity levels. In contrast, non-functionalized plastic surfaces retain viral infectivity for an extended period of up to 48 h.     • Objectives: Antinuclear antibodies (ANA) are important for the diagnosis of various autoimmune diseases. ANA are usually detected by indirect immunofluorescence assay (IFA) using HEp-2 cells (HEp-2 IFA). There are many variables influencing HEp-2 IFA results, such as subjective visual reading, serum screening dilution, substrate manufacturing, microscope components and conjugate. Newer developments on ANA testing that offer novel features adopted by some clinical laboratories include automated computer-assisted diagnosis (CAD) systems and solid phase assays (SPA).Methods: A group of experts reviewed current literature and established recommendations on methodological aspects of ANA testing. This process was supported by a two round Delphi exercise. International expert groups that participated in this initiative included (i) the European Federation of Clinical Chemistry and Laboratory Medicine (EFLM) Working Group “Autoimmunity Testing”; (ii) the European Autoimmune Standardization Initiative (EASI); and (iii) the International Consensus on ANA Patterns (ICAP).Results: In total, 35 recommendations/statements related to (i) ANA testing and reporting by HEp-2 IFA; (ii) HEp-2 IFA methodological aspects including substrate/conjugate selection and the application of CAD systems; (iii) quality assurance; (iv) HEp-2 IFA validation/verification approaches and (v) SPA were formulated. Globally, 95% of all submitted scores in the final Delphi round were above 6 (moderately agree, agree or strongly agree) and 85% above 7 (agree and strongly agree), indicating strong international support for the proposed recommendations.Conclusions: These recommendations are an important step to achieve high quality ANA testing.

DeCarolis et al. articulate the benefits of forming collaborative teams with a wide array of disciplinary and domain expertise to conduct analysis with macro-energy system models. Open-source models, tools, and datasets underpin such efforts by enabling transparency, accessibility, and replicability among team members and with the broader modeling community.

  • Associations of physical activity, sedentary time, and diet quality with biomarkers of inflammation in children

    • We investigated the associations of physical activity (PA), sedentary time (ST), and diet quality with biomarkers of inflammation in 390 children (192 girls, 198 boys) aged 6–8 years. PA energy expenditure (PAEE), light PA, moderate PA (MPA), vigorous PA (VPA), moderate-to-vigorous PA (MVPA), and ST were assessed by combined movement and heart rate sensor. Finnish Children Healthy Eating Index was calculated using data from 4 d food records. Body fat percentage (BF%) was measured by dual-energy X-ray absorptiometry. High-sensitivity C-reactive protein (Hs-CRP), leptin, interleukin-6 (IL-6), adiponectin, tumour necrosis factor-α, and glycoprotein acetyls were measured from fasting blood samples. PAEE, MPA, VPA, and MVPA were inversely associated with hs-CRP (β=−191 to −139, 95% CI=−0.294 to −0.024), leptin (β=−0.409 to −0.301, 95% CI=−0.499 to −0.107), IL-6 (β=−0.136 to −0.104, 95% CI=−0.240 to −0.001) and PAEE, MPA, and MVPA were inversely associated with glycoprotein acetyls (β=−0.117 to −0.103, 95% CI=−0.213 to −0.001). ST was directly associated with hs-CRP (β=0.170, 95% CI=0.070–0.269), leptin (β=0.355, 95% CI=0.265–0.445), and IL-6 (β=0.105, 95% CI=0.005–0.205). VPA was inversely associated with hs-CRP, leptin, and IL-6 in children with higher BF% (β=−0.344 to −0.181, 95% CI=−0.477 to −0.033) but not among children with lower BF% (β=−0.007–0.033, 95% CI=−0.183–0.184). In conclusion, PA was inversely and ST directly associated with circulating levels of biomarkers of inflammation among children. Furthermore, we observed that PA was inversely associated with these biomarkers for inflammation in children with a higher BF%. Highlights Systemic inflammation, as indicated by increased circulating concentrations of biomarkers for inflammation, may be important in causal pathways leading to insulin resistance, sub-clinical atherosclerosis, and eventually clinical manifestations of cardiovascular diseases. Higher levels of physical activity and lower levels of sedentary time were associated with more favourable inflammatory profile. Body fat percentage modified these associations and especially vigorous intensity physical activity was inversely associated with biomarkers of inflammation on children with higher body fat percentage but not in children with lower body fat percentage.

  • Carbon dioxide balance of a fen ecosystem in northern Finland under elevated UV-B radiation

    • The effect of elevated UV-B radiation on CO2 exchange of a natural flark fen was studied in open-field conditions during 2003-2005. The experimental site was located in SodankylA in northern Finland (67 degrees 22'N, 26 degrees 38'E, 179 m a.s.l.). Altogether 30 study plots, each 120 cm x 120 cm in size, were randomly distributed between three treatments (n=10): ambient control, UV-A control and UV-B treatment. The UV-B-treated plots were exposed to elevated UV-B radiation level for three growing seasons. The instantaneous net ecosystem CO2 exchange (NEE) and dark respiration (R-TOT) were measured during the growing season using a closed chamber method. The wintertime CO2 emissions were estimated using a gradient technique by analyzing the CO2 concentration in the snow pack. In addition to the instantaneous CO2 exchange, the seasonal CO2 balances during the growing seasons were modeled using environmental data measured at the site. In general, the instantaneous NEE at light saturation was slightly higher in the UV-B treatment compared with the ambient control, but the gross photosynthesis was unaffected by the exposure. The R-TOT was significantly lower under elevated UV-B in the third study year. The modeled seasonal (June-September) CO2 balance varied between the years depending on the ground water level and temperature conditions. During the driest year, the seasonal CO2 balance was negative (net release of CO2) in the ambient control and the UV-B treatment was CO2 neutral. During the third year, the seasonal CO2 uptake was 43 +/- 36 g CO2-C m(-2) in the ambient control and 79 +/- 45 g CO2-C m(-2) in the UV-B treatment. The results suggest that the long-term exposure to high UV-B radiation levels may slightly increase the CO2 accumulation to fens resulting from a decrease in microbial activity in peat. However, it is unlikely that the predicted development of the level of UV-B radiation would significantly affect the CO2 balance of fen ecosystems in future.

    • To study the effects of elevated ozone concentration on methane dynamics and a sedge species, Eriophorum vaginatum, we exposed peatland microcosms, isolated by coring from an oligotrophic pine fen, to double ambient ozone concentration in an open-air ozone exposure field for four growing seasons. The field consists of eight circular plots of which four were fumigated with elevated ozone concentration and four were ambient controls. At the latter part of the first growing season (week 33, 2003), the methane emission was 159 +/- 14 mg CH4 m(-2) day(-1) (mean +/- SE) in the ozone treatment and 214 +/- 8 mg CH4 m(-2) day(-1) under the ambient control. However, towards the end of the experiment the ozone treatment slightly, but consistently, enhanced the methane emission. At the end of the third growing season (2005), microbial biomass (estimated by phospholipid fatty acid biomarkers) was higher in peat exposed to ozone (1975 +/- 108 nmol g(-1) dw) than in peat of the control microcosms (1589 +/- 115 nmol g(-1) dw). The concentrations of organic acids in peat pore water showed a similar trend. Elevated ozone did not affect the shoot length or the structure of the sedge E. vaginatum leaves but it slightly increased the total number of sedge leaves towards the end of the experiment. Our results indicate that elevated ozone concentration enhances the general growth conditions of microbes in peat by increasing their substrate availability. However, the methane production did not reflect the increase in the concentration of organic acids, probably because hydrogenotrophic methane production dominated in the peat studied. Although, we used isolated peatland microcosms with limited size as study material, we did not find experimental factors that could have hampered the basic conclusions on the effects of ozone.

  • Ridged sea ice characteristics in the arctic from a coupled multicategory sea ice model

    • In this study, a multicategory sea ice model with explicit ice classes for ridged and rafted ice was used to examine the evolution of deformed ice during the period 1980-2002. The results show that (1) ridged ice comprises roughly 45-60% of Arctic sea ice volume and 25-45% of the sea ice area, (2) most of the perennial ice consists of ridged ice, and (3) ridged ice exhibits a small seasonal variability. Our results also show an increase in mean ridged ice thickness of 4-6 cm yr(-1) during the summer in an area north of the Canadian Archipelago and a corresponding decrease in the East Siberian Sea and Nansen Basin. At the same time, Arctic sea ice age has been observed to decline and ice drift speed to increase during the simulation period. We connect these findings with a modeled regional increase in the production rate of ridged ice. Comparison of the multicategory model and a two category reference model shows a substantially increased ice production rate due to a more frequent occurrence of leads, resulting in an ice thickness increase of up to 0.8 m. Differences in ice physics between the multicategory and reference models also affect the freshwater content. The sum of liquid and solid freshwater content in the entire Arctic Ocean is about 10% lower and net precipitation (P-E) is about 7% lower as compared to the reference model.

    • In this study a commercial particle analyzer was used to image and help sorting microplastic particles (MPs) dispersed in filtrated and de-aerated tap water. The device provides a relatively easy and fast procedure for obtaining ultra-high-definition imaging, allowing the determination of shape, size, and number of 2D-projections of solid particles. The image analysis revealed clear differences among the studied different MPs originating from the grinding of five common grades of plastic sheets as they affect the image rendering differently, principally due to the light scattering either at the surface or in the volume of the microplastics. The high-quality imaging of the device also allows the discrimination of the microplastics from air bubbles with well-defined spherical shapes as well as to obtain an estimate of the size of MPs in a snapshot. We associate the differences among the shapes of the identified MPs in this study depending on the plastic type with known physical properties, such as brittleness, crystallinity, or softness. Furthermore, as a novel method we exploit a parameter based on the light intensity map from moving particles in cuvette flow to sort MPs from other particles, such as, wood fiber, human hair, and air bubbles. Using the light intensity map, which is related to the plastic-water refractive index ratio, the presence of microplastics in water can be revealed among other particles, but not their specific plastic type.

In this study, we (1) report the creation of a comprehensive reference library of DNA barcodes for the arthropods of an entire country (Finland), (2) publish this library, and (3) deliver a new identification tool for insects and spiders, as based on this resource. The reference library contains mtDNA COI barcodes for 11,275 (43%) of 26,437 arthropod species known from Finland, including 10,811 (45%) of 23,956 insect species. To quantify the improvement in identification accuracy enabled by the current reference library, we ran 1000 Finnish insect and spider species through the Barcode of Life Data system (BOLD) identification engine. Of these, 91% were correctly assigned to a unique species when compared to the new reference library alone, 85% were correctly identified when compared to BOLD with the new material included, and 75% with the new material excluded. To capitalize on this resource, we used the new reference material to train a probabilistic taxonomic assignment tool, FinPROTAX, scoring high success. For the full-length barcode region, the accuracy of taxonomic assignments at the level of classes, orders, families, subfamilies, tribes, genera, and species reached 99.9%, 99.9%, 99.8%, 99.7%, 99.4%, 96.8%, and 88.5%, respectively. The FinBOL arthropod reference library and FinPROTAX are available through the Finnish Biodiversity Information Facility (www.laji.fi) at https://laji.fi/en/theme/protax. Overall, the FinBOL investment represents a massive capacity-transfer from the taxonomic community of Finland to all sectors of society.

    • All-cellulose composites (ACCs) are manufactured using only cellulose as a raw material. Biobased materials are more sustainable alternatives to the petroleum-based composites that are used in many technical and life-science applications. In this study, an aquatic NaOH-urea solvent system was used to produce sustainable ACCs from wood-based woven textiles with and without the addition of TEMPO-oxidized nanocellulose (at 1 wt.-%). This study investigated the effects of dissolution time, temperature during hot press, and the addition of TEMPO-oxidized nanocellulose on the mechanical and thermal properties of the composites. The results showed a significant change in the tensile properties of the layered textile composite at dissolution times of 30 s and 1 min, while ACC elongation was the highest after 2 and 5 min. Changes in hot press temperature from 70 °C to 150 °C had a significant effect: with an increase in hot press temperature, the tensile strength increased and the elongation at break decreased. Incorporating TEMPO-oxidized nanocellulose into the interface of textile layers before partial dissolution improved tensile strength and, even more markedly, the elongation at break. According to thermal analyses, textile-based ACCs have a higher storage modulus (0.6 GPa) and thermal stabilization than ACCs with nanocellulose additives. This study highlights the important roles of process conditions and raw material characteristics on the structure and properties of ACCs.

    • All-cellulose composites (ACCs) have been fabricated by using a variety of cellulosic sources, versatile technologies, and are sustainable alternatives for traditional composites. In this study, nonwoven-woven ACC laminates were created from wood-based Spinnova short fibers and Lyocell fabrics via partial dissolution and an NaOH-urea solvent system. The less-known wood-based Spinnova fiber is created for the textile industry, but it also has great potential for the composite industry. To identify the mechanical properties of ACCs—which greatly influence the range of material application—tensile, impact, and flexural tests were conducted. The mechanical properties indicated only moderate properties, which are influenced by high porosity and weak fiber bonding. Despite this, valuable information on the nonwoven-woven structured ACCs was obtained. To improve the ACC laminate’s ability to resist moisture, biobased coatings (e.g., commercially available birch bark betulin and suberin acid mixture) were applied on the surface of ACCs and it successfully improved the wetting resistance. The results of contact angle analyses demonstrated that the highest contact angle of 128° was measured for betulin-coated laminates and the best stable hydrophobicity calculated a minute after the beginning of the experiment were observed at 109° for the uncommercial pressurized hot ethanol (PHE) extract of birch bark.
