    • CONTEXT: Studies on clinical and immunological features of Autoimmune Addison's disease (AAD) are needed to understand the disease burden and increased mortality.OBJECTIVE: To provide upgraded data on autoimmune comorbidities, replacement therapy, autoantibody profiles and cardiovascular risk factors.DESIGN, SETTING AND PARTICIPANTS: Cross sectional, population-based study. 660 AAD patients were included utilizing the Swedish Addison Registry (SAR) 2008-2014. When analyzing cardiovascular risk factors, 3,594 individuals from the population-based survey in Northern Sweden, MONICA (MONItoring of Trends and Determinants of CArdiovascular Disease), served as controls.MAIN OUTCOME MEASURE: Prevalence of autoimmune comorbidities and cardiovascular risk factors. Autoantibodies against 13 autoantigens were determined.RESULTS: Sixty percent of the SAR cohort consisted of females. Mean age at diagnosis was significantly higher for females than for males (36.8 vs. 31.1 years). The proportion of 21-hydroxylase autoantibody positive patients was 83% and 62% of patients had one or more associated autoimmune diseases, more frequently coexisting in females (p<0.0001). AAD patients had lower BMI (p<0.0001) and prevalence of hypertension (p=0.027) compared with controls. Conventional hydrocortisone tablets were used by 89% of patients; with the mean dose 28.1±8.5 mg/day. The mean hydrocortisone equivalent dose normalized to body surface was 14.8±4.4 mg/m(2)/day. Higher hydrocortisone equivalent dose was associated with higher incidence of hypertension (p=0.046).CONCLUSIONS: Careful monitoring of AAD patients is warranted to detect associated autoimmune diseases. Contemporary Swedish AAD patients do not have increased prevalence of overweight, hypertension, T2DM or hyperlipidemia. However, high glucocorticoid replacement doses may be a risk factor for hypertension.

    • A proposed EU regulation requires the automotive industry to develop technologies that will substantially decrease the risk for vulnerable road users such as pedestrians when hit by a vehicle. Automatic brake assist systems, activated by a suitable sensor, will reduce the speed of the vehicle before the impact. Far InfraRed (FIR) detectors are ideal candidates for such sensing systems. In order to enable high volume serial installation, the main development must be focused on cost reduction. Optimizing all aspects of the system, including sensor size, production yield and 3D wafer level vacuum packaging will lower today's "high end" FIR product costs by an order of magnitude. A low-cost FIR infrared bolometer is developed in the Eureka labeled PIMS (Pedestrian Injury Mitigation System) project. In the paper the background and the actual design of the first demonstrator to be finished in 2008 are described in detail.

    • F-18-(E)-N-(3-iodoprop-2-enyl)-2 beta-carbofluoroethoxy-3 beta-(4'-methylphenyl) nortropane (F-18-FE-PE2I) was recently developed and has shown adequate affinity and high selectivity for the dopamine transporter (DAT). Previous studies have shown promising results for F-18-FE-PE2I as a suitable radioligand for DAT imaging. In this study, we investigated the whole-body biodistribution and dosimetry of F-18-FE-PE2I in healthy volunteers to support its utility as a suitable PET imaging agent for the DAT. Methods: Five healthy volunteers were given a mean activity of 2.5 MBq/kg, and 3 PET scans, head to thigh, were performed immediately after injection followed by 4 whole-body PET/CT scans between 0.5 and 6 h after injection. Blood samples were drawn in connection with the whole-body scans, and all urine was collected until 6 h after injection. Volumes of interest were delineated around 17 organs on all images, and the areas under the time-activity curves were calculated to obtain the total number of decays in the organs. The absorbed doses to organs and the effective dose were calculated using the software IDAC. Results: The highest activity concentration was observed in the liver (0.9%-1.2% injected activity/100 g) up to 30 min after injection. At later time points, the highest concentration was seen in the gallbladder (1.1%-0.1% injected activity/100 g). The activity excreted with urine ranged between 23% and 34%, with a mean of 28%. The urinary bladder received the highest absorbed dose (119 mu Gy/MBq), followed by the liver (46 mu Gy/MBq). The effective dose was 23 mu Sv/MBq (range, 19-28 mu Sv/MBq), resulting in an effective dose of 4.6 mSv for an administered activity of 200 MBq. Conclusion: The effective dose is within the same order of magnitude as other commonly used PET imaging agents as well as DAT agents. The reasonable effective dose, together with the previously reported favorable characteristics for DAT imaging and quantification, indicates that F-18-FE-PE2I is a suitable radioligand for DAT imaging.

  • Low-cost far infrared bolometer camera for automotive use

    • A new low-cost long-wavelength infrared bolometer camera system is under development. It is designed for use with an automatic vision algorithm system as a sensor to detect vulnerable road users in traffic. Looking 15 in in front of the vehicle it can in case of an unavoidable impact activate a brake assist system or other deployable protection system. To achieve our cost target below epsilon 100 for the sensor system we evaluate the required performance and can reduce the sensitivity to 150 mK and pixel resolution to 80 x 30. We address all the main cost drivers as sensor size and production yield along with vacuum packaging, optical components and large volume manufacturing technologies. The detector array is based on a new type of high performance thermistor material. Very thin Si/SiGe single crystal multi-layers are grown epitaxially. Due to the resulting valence barriers a high temperature coefficient of resistance is achieved (3.3%/K). Simultaneously, the high quality crystalline material provides very low 1/f-noise characteristics and uniform material properties. The thermistor material is transferred from the original substrate wafer to the read-out circuit using adhesive wafer bonding and subsequent thinning. Bolometer arrays can then be fabricated using industry standard MEMS process and materials. The inherently good detector performance allows us to reduce the vacuum requirement and we can implement wafer level vacuum packaging technology used in established automotive sensor fabrication. The optical design is reduced to a single lens camera. We develop a low cost molding process using a novel chalcogenide glass (GASIR (R) 3) and integrate anti-reflective and anti-erosion properties using diamond like carbon coating.

    • Electrochemical reactions depend on the electrochemical interface between the electrode surfaces and the electrolytes. To control and advance electrochemical reactions there is a need to develop realistic simulation models of the electrochemical interface to understand the interface from an atomistic point-of-view. Here we present a method for obtaining thermodynamic realistic interface structures, a procedure we use to derive specific coverages and to obtain ab initio simulated cyclic voltammograms. As a case study, the method and procedure is applied in a matrix study of three Cu facets in three different electrolytes. The results have been validated by direct comparison to experimental cyclic voltammograms. The alkaline (NaOH) cyclic voltammograms are described by H* and OH*, while in neutral medium (KHCO3) the CO3* species are dominating and in acidic (KCl) the Cl* species prevail. An almost one-to-one mapping is observed from simulation to experiments giving an atomistic understanding of the interface structure of the Cu facets. Atomistic understanding of the interface at relevant eletrolyte conditions will further allow realistic modelling of electrochemical reactions of importance for future eletrocatalytic studies.

    • GH responsiveness guides GH dosing during the catch-up growth (CUG) period; however, little is known regarding GH dosing during the prepubertal maintenance treatment period.To evaluate if standard deviation score (SDS) channel parallel growth with normal height velocity can be maintained following CUG by reducing GH dose by 50% in children receiving doses individualized based on estimated GH-responsiveness during the catch-up period.and settings: Prepubertal children (n=98; 72 boys) receiving GH during CUG (GH-deficient (n=33); non-GH-deficient (n=65)), were randomized after 2-3 years to either a 50% reduced individualized (GHRID; n=27; 20 boys) or unchanged individualized dose (GHUID; n=38; 27 boys). Another 33 children (25 boys) continued on a standard weight-based dose, 43 µg/kg/day (GHFIX).The primary endpoint was the proportion of children with ΔheightSDS within ±0.3 at 1 year after GH-dose reduction, versus two control groups: GHUID and GHFIX. The hypothesis was that heightSDS could be maintained within ±0.3 with a reduced individualized GH dose.For the intention-to-treat population at 1 year, 85% of the GHRIDgroup maintained ΔheightSDS within ±0.3 versus 41% in the GHUIDgroup, p=0.0055 and 48% in the GHFIXgroup, p=0.0047. ΔIGF-ISDS in the GHRIDgroup was (mean±SD) -0.75±1.0 at 3 months, p=0.003 and at 1 year -0.72±1.2, compared to the GHUIDgroup 0.15±1.2, p=0.005, and for the GHFIXgroup 0.05±1.0, p=0.02.Channel parallel growth, i.e. normal height velocity, and IGFSDS levels within ±2 were maintained after completed CUG using a 50% lower individualized dose than used during the CUG period.

  • Influence of impact velocity and angle in a detailed reconstruction of a bicycle accident

    • Bicycle accidents have become the most common cause of serious injury in the traffic during the last couple of years in Sweden. The objective of this study was to investigate the effect of the input variables, initial velocity and head orientation, of a bicycle accident reconstruction on the strain levels in the brain using a detailed FE head model. The accident involved a non-helmeted 68 year old male who sustained a linear skull fracture, contusions, acute subdural hematoma, and small bleeding at the swelling (subarachnoid blood). The orientation of the head just before impact was determined from the swelling appearing in the computer tomography (CT) scans. The head model used in this study was developed at the Royal Institute of Technology in Stockholm. The stress in the cranial bone, first principal strain in the brain tissue and acceleration were determined. The model was able to predict a strain pattern that correlated well with the medical images from the victim. The variation study showed that the tangential velocity had a large effect on the strain levels in the studied case. The strain pattern indicated larger areas of high strain with increased tangential velocity especially at the more superior sections.

    • PURPOSE To present long-term ocular complications and electroretinographic (ERG) findings in children with long-chain 3-hydroxyacyl-CoA dehydrogenase (LCHAD) deficiency - a life-threatening metabolic disease - and the relation to age at diagnosis, treatment and other clinical parameters. METHODS Ten children with LCHAD deficiency underwent repeated ophthalmological evaluations including ERG. RESULTS All 10 children developed chorioretinal pathology. Regardless of age at diagnosis, initiation of treatment and age at examination, inter-individual differences were present. Profound chorioretinal atrophy, severe visual impairment and progressive myopia had developed in two teenagers. Milder chorioretinopathy with or without subnormal visual acuity was present in all other children. ERG was pathological in seven children. The chorioretinopathy often started in the peripapillary or perimacular areas. In one patient, unilateral visual impairment was associated with fibrosis. CONCLUSION Early diagnosis and adequate therapy might delay but not prevent the progression of retinal complications. Late diagnosis with severe symptoms at diagnosis, neonatal hypoglycaemia and frequent decompensations may increase the progression rate of the chorioretinopathy. LCHAD deficiency, a potentially lethal disease, is sometimes difficult to diagnose. Unusual chorioretinal findings should alert the ophthalmologist to the long-chain 3-hydroxyacyl-CoA dehydrogenase deficiency, especially if there is a history of neonatal hypoglycaemia or failure to thrive.

    • PURPOSE: The aim of this study was to evaluate AZD2995 side by side with AZD2184 as novel PET radioligands for imaging of amyloid-β in Alzheimer's disease (AD).METHODS: In vitro binding of tritium-labelled AZD2995 and AZD2184 was studied and compared with that of the established amyloid-β PET radioligand PIB. Subsequently, a first-in-human in vivo PET study was performed using [(11)C]AZD2995 and [(11)C]AZD2184 in three healthy control subjects and seven AD patients.RESULTS: AZD2995, AZD2184 and PIB were found to share the same binding site to amyloid-β. [(3)H]AZD2995 had the highest signal-to-background ratio in brain tissue from patients with AD as well as in transgenic mice. However, [(11)C]AZD2184 had superior imaging properties in PET, as shown by larger effect sizes comparing binding potential values in cortical regions of AD patients and healthy controls. Nevertheless, probably due to a lower amount of nonspecific binding, the group separation of the distribution volume ratio values of [(11)C]AZD2995 was greater in areas with lower amyloid-β load, e.g. the hippocampus.CONCLUSION: Both AZD2995 and AZD2184 detect amyloid-β with high affinity and specificity and also display a lower degree of nonspecific binding than that reported for PIB. Overall [(11)C]AZD2184 seems to be an amyloid-β radioligand with higher uptake and better group separation when compared to [(11)C]AZD2995. However, the very low nonspecific binding of [(11)C]AZD2995 makes this radioligand potentially interesting as a tool to study minute levels of amyloid-β. This sensitivity may be important in investigating, for example, early prodromal stages of AD or in the longitudinal study of a disease modifying therapy.

  • Information Needs for Water Resource and Risk Management : Hydro-Meteorological Data Value and Non-Traditional Information

    • Data availability is extremely important for water management. Without data it would not be possible to know how much water is available or how often extreme events are likely to occur. The usually available hydro-meteorological data often have a limited representativeness and are affected by errors and uncertainties. Additionally, their collection is resource-intensive and, thus, many areas of the world are severely under-monitored. Other areas are seeing an unprecedented – yet local – wealth of data in the last decades. Additionally, the spread of new technologies together with the integration of different approaches to water management science and practice have uncovered a large amount of soft information that can potentially complement and expand the possibilities of water management.This thesis presents a series of studies that address data opportunities for water management. Firstly, the hydro-meteorological data needs for correctly estimating key processes for water resource management such as precipitation and discharge were evaluated. Secondly, the use of non-traditional sources of information such as social media and human behaviour to improve the efficiency of flood mitigation actions were explored. The results obtained provide guidelines for determining basic hydro-meteorological data needs. For instance, an upper density of 24 rain gauges per 1000 km2 for spatial precipitation estimation beyond which improvements are negligible was found. Additionally, a larger relative value of discharge data respect to precipitation data for calibrating hydrological models was observed. Regarding non-traditional sources of information, social memory of past flooding events was found to be a relevant factor determining the efficiency of flood early warning systems and therefore their damage mitigation potential. Finally, a new methodology to use social media data for probabilistic estimates of flood extent was put forward and shown to achieve results comparable to traditional approaches.This thesis significantly contributes to integrated water management by improving the understanding of data needs and opportunities of new sources of information thus making water management more efficient and useful for society.

    • Autoimmune polyglandular syndrome type I (APS I) is an autosomal recessive disorder characterized by a combination of autoimmune manifestations affecting endocrine and non-endocrine organs. APS I usually presents in childhood. The three most common manifestations are chronic mucocutaneous candidiasis, hypoparathyroidism and Addison's disease. At least two of these must be present to fulfill the diagnostic criteria of this syndrome. The spectrum of other associated diseases includes gonadal insufficiency, alopecia, vitiligo and chronic active hepatitis. APS I is caused by a mutation in the AIRE-gene (autoimmune regulator) located on chromosome 21. Analysis of specific autoantibodies against intracellular enzymes, particularly enzymes in the synthesis of steroids and neurotransmittors, can be used in the diagnosis of APS I and to predict different manifestations of the disease.

    • Children with long-chain 3-hydroxyacyl-CoA dehydrogenase deficiency (LCHAD) have a defect in the degradation of long-chain fatty acids and are at risk of hypoketotic hypoglycemia and insufficient energy production as well as accumulation of toxic fatty acid intermediates. Knowledge on substrate metabolism in children with LCHAD deficiency during fasting is limited. Treatment guidelines differ between centers, both as far as length of fasting periods and need for night feeds are concerned. To increase the understanding of fasting intolerance and improve treatment recommendations, children with LCHAD deficiency were investigated with stable isotope technique, microdialysis, and indirect calometry, in order to assess lipolysis and glucose production during 6 h of fasting. We found an early and increased lipolysis and accumulation of long chain acylcarnitines after 4 h of fasting, albeit no patients developed hypoglycemia. The rate of glycerol production, reflecting lipolysis, averaged 7.7 ± 1.6 µmol/kg/min, which is higher compared to that of peers. The rate of glucose production was normal for age; 19.6 ± 3.4 µmol/kg/min (3.5 ± 0.6 mg/kg/min). Resting energy expenditure was also normal, even though the respiratory quotient was increased indicating mainly glucose oxidation. The results show that lipolysis and accumulation of long chain acylcarnitines occurs before hypoglycemia in fasting children with LCHAD, which may indicate more limited fasting tolerance than previously suggested.

    • Centrally located lumbar disc herniations have been reported to be of predictive value for poor post-operative clinical outcome. One hundred and fifty patients undergoing lumbar disc herniation surgery were prospectively included. Herniation-related parameters, including the grading of contours, were assessed from pre-operative computed tomography (CT) and magnetic resonance imaging (MRI) images using a new three-dimensional grading system. The radiological findings were compared with outcome parameters two years post-operatively (patient-assessed pain, function/health scores and evaluation by an independent observer). An intra- and inter-observer validation of the classification was performed in a subgroup of patients. High intra-observer and good inter-observer reliability for both CT and MRI was seen. In the study population, no relation between the distribution or size of the herniations and outcome at 2-year follow-up were found. The distribution and size of the lumbar disc herniations with the three-dimensional classification were not found to be of importance for the clinical outcome.

  • Electrochemical Interface during Corrosion of Copper in Anoxic Sulfide-Containing Groundwater-A Computational Study

    • Corrosion of copper is an expensive degradation process of materials in engineered infrastructures and in various technical applications. It is also an important factor in the geological disposal of spent nuclear fuel, where sulfide-induced corrosion is expected to be the predominant chemical degradation process of copper canisters used for encapsulation and isolation of the radioactive material from the biosphere. One aspect of the corrosion process that is still under intense research is the corrosion morphology and how it might be affected by the composition of the groundwater. Using density functional theory, we investigate the electrochemical interface of corroding copper in aqueous solutions containing sulfides, with and without the presence of Cl-, HCO3-, and SO42- anions. Through state-of-the-art electrochemical models, we account for the effects of pH, concentrations, and potential on the interfacial structure and composition. Results are presented for the Cu(110) surface facet and compared to the (110) and (001) facets of chalcocite (Cu2S), i.e., the main product of sulfide-induced corrosion. It is found that at low potentials, H dominates on all surfaces, and at high potentials, sulfides. In the intermediate ranges, the surfaces differ with sulfides prevailing on Cu, while adsorbed H2O, Cl, or H dominate on Cu2S. The results are summarized as surface Pourbaix diagrams and are generally applicable in corrosion science and electrochemistry. The implications of the study are discussed in light of the expected conditions of planned spent nuclear fuel repositories in Sweden, Finland, and Canada.

  • Uncovering the electrochemical interface of low-index copper surfaces in deep groundwater environments

    • Using a combination of a sophisticated modeling protocol and well-established electrochemical techniques, we unravel the chemical composition of the low-index surfaces of copper in groundwater environments at different ion concentrations, pHs, and redox potentials. By carefully linking density functional theory (DFT) and cyclic voltammetry (CV), we are able to extract fundamental information on interfaces of broad significance. Herein, we focus on the case of groundwater found in deep geological environments of importance to the planned constructions of disposal repositories for spent nuclear fuel around the world. Within the error margins of DFT, we can assign adsorption structures and compositions to the current peaks of the CVs. It is found that among the groundwater ions of main interest (i.e. sulfide, bisulfide, sulfate, chloride and bicarbonate), sulfides (HS-, S2-) bind strongest to the surface, and are likely to dominate at the interfaces under the deep geological conditions relevant for repositories of spent nuclear fuel.

    • An increasing number of fatty acid oxidation defects are being detected owing to diagnostic improvements and a greater awareness among clinicians. The metabolic block leads to energy disruption, fatty infiltration, and toxic effects on organ functions exerted by β-oxidation metabolites. This investigation was undertaken to assess the influence of long-chain 3-hydroxyacyl-CoA dehydrogenase (LCHAD) deficiency on lipolysis and energy turnover. We addressed the question whether the lipolysis and glucose production rates would be altered in the fasting state in a child with this disease. Lipolysis, glucose production and resting energy expenditure (REE) were studied in a 17-month-old girl with LCHAD deficiency and her healthy twin sister. Lipolysis and glucose production were determined after a 4–6 h fast by constant-rate infusion of [1,1,2,3,3-2H5]glycerol and [6,6-2H2]glucose and analysis by gas chromatography–mass spectrometry. REE was estimated by indirect calorimetry. The affected girl showed 50% higher lipolysis than did her sister, whereas the glucose production rates were similar. Plasma levels of dicarboxylic acids of 6–12 carbon atoms chain length, 3-hydroxy fatty acids of 6–18 carbon atoms chain length, total free fatty acids, and acylcarnitines were increased in the patient, as was REE. Since glucose production rates and plasma glucose levels were similar in the two girls, the increased lipolysis observed in the patient probably represents a compensatory mechanism for energy generation. This is achieved at the price of an augmented risk for fatty acid infiltration and toxic effects of β-oxidation intermediates. This highlights the importance of avoiding fasting in these patients.

  • Cloudburst catastrophe modelling : Case study – Jönköping municipality, Sweden

    • In Europe, flash floods are one of the most significant natural hazards, causing serious risk to life and destruction of buildings and infrastructure. The intense rain causing those floods has a few different names, however, with very similar meaning. The term chosen in this study, ‘cloudburst’, was introduced by Woolley (1946) as “…a torrential downpour of rain which by its spottiness and relatively high intensity suggests the bursting and discharge of the whole cloud at once”. While these events play an important role in the ongoing flood risk management discussion, they are under-represented among flood models.The main aim of this study is to demonstrate an approach by showing how methods and techniques can be integrated together to construct a catastrophe model for flash flooding of Jönköping municipality in Sweden. The model is developed in the framework of the ‘Oasis Loss Modelling Framework’ platform, jointly with end-users from the public sector and the insurance industry. Calibration and validation of the model were conducted by comparisons against three historical cloudburst events and corresponding insurance-claim data.The analysis has shown that it is possible to get acceptable results from a cloudburst catastrophe model using only rainfall data, and not surface-water level as driving variable. The approach presented opens up for such loss modelling in places where complex hydraulic modelling cannot be done because of lacking data or skill of responsible staff. The Swedish case study indicates that the framework presented can be considered as an important decision making tool, by establishing an area for collaboration between academia; insurance businesses; and local authorities, to reduce long-term disaster risk in Sweden.

    • AIMS:The association between drunk driving (DD) and/or risky driving (RD) offences and subsequent hospitalization and mortality was studied during a 25-year period.METHODS:Information about drinking habits and psychosocial factors for the 8122 conscripts from Stockholm County in 1969-1970 was linked to register data on hospitalization, mortality, DD and RD.RESULTS:Analyses comparing background characteristics of DD and RD groups showed that the prevalence of problem behaviour and drug use was highest in the RD group. The relative risk (RR) for hospitalization after DD and/or RD was significantly elevated in multivariate logistic regression analysis for all the studied diagnostic categories (alcohol diagnoses, narcotic diagnoses, suicide attempts, psychoses, E-codes, all diagnoses), and was especially high for alcohol (RR = 7.2) and diagnosis of drug misuse (RR = 9.2). The RR of all hospitalization was 1.5 for the DD group, 1.8 for the RD group, and 1.9 for those who had been sentenced both for drunk driving and risky driving (DRD), all of which were significantly increased. The RR of death was significantly elevated in all three groups.CONCLUSIONS:The results show a significantly increased risk of hospitalization and mortality both in the DD and the RD group. From a public health and traffic safety perspective, this implies a need for developing and implementing better prevention strategies.

  • Cloudburst-disaster modelling : A new open-source catastrophe model

    • Cloudburst flash floods cause big casualties and economic losses. This study primarily investigated if a cloudburst catastrophe (cat) model could be constructed to meaningfully assess such a hazard, exposure and vulnerability in Swedish urban context. Rainfall intensity was used directly as hazard measure, bypassing hydraulic water-level modelling, to predict vulnerability. The Splash (Swedish pluvial modelling analysis and safety handling) cloudburst-disaster model was constructed using the Oasis Loss Modelling Framework, and was based on individual property values and building locations, property-level insurance-loss data, high-resolution geographical data, and rainfall data from a dense municipal gauge network in the city of Jönköping. One major cloudburst event was used to derive a vulnerability curve. The following two events were used for validation and supported the hypothesis that the vulnerability curve changed with time because of municipal flood-risk-reduction measures after the first event. A faulty rain gauge during the first event, replaced by a trustworthy private gauge, clarified the very high sensitivity to cloudburst input. Given the limited amount of loss data, our results were uncertain but they pointed towards possible ways to further this study with other loss data at other locations, possibly using more easily available aggregated loss data. We concluded that a cat model based only on rainfall intensity provided acceptable results, thus providing an opening for future, simplified cloudburst cat models applicable in most geographical contexts where reliable cloudburst data are available, especially in cities with limited topographic data and hydraulic-modelling capacity.

    • Controlled electrochemical oxidation of hydrocarbons to desired products is an attractive approach in catalysis. Here we study the electrochemical propene oxidation under operando conditions using Pd L-edge X-ray absorption spectroscopy (XAS) as a sensitive probe to elucidate surface processes occurring during catalysis. Together with ab initio multiple-scattering calculations, our XAS results enable assignment of characteristic changes of the Pd L-edge intensity and energy position in terms of a mechanistic understanding of the selective oxidation of propene. The results, supported by electrochemical density functional theory DFT simulations, show that in the potential range of 0.8–1.0 V vs. the reversible hydrogen electrode (RHE), selective oxidation of propene to acrolein and acrylic acid occurs on the metallic Pd surface. These reactions are proposed to proceed via the Langmuir–Hinshelwood mechanism. In contrast, for the potential range of 1.1–1.3 V vs. RHE, selective oxidation of propene to propylene glycol takes place on a Pd oxide surface.

    • CONTEXT: Weight-based GH dosing results in a wide variation in growth response in children with GH deficiency (GHD) or idiopathic short stature (ISS). OBJECTIVE: The hypothesis tested was whether individualized GH doses, based on variation in GH responsiveness estimated by a prediction model, reduced variability in growth response around a set height target compared with a standardized weight-based dose. SETTING: A total of 153 short prepubertal children diagnosed with isolated GHD or ISS (n = 43) and at least 1 SD score (SDS) below midparental height SDS (MPH(SDS)) were included in this 2-yr multicenter study. INTERVENTION: The children were randomized to either a standard (43 microg/kg.d) or individualized (17-100 microg/kg.d) GH dose. MAIN OUTCOME MEASURE: We measured the deviation of height(SDS) from individual MPH(SDS) (diffMPH(SDS)). The primary endpoint was the difference in the range of diffMPH(SDS) between the two groups. RESULTS: The diffMPH(SDS) range was reduced by 32% in the individualized-dose group relative to the standard-dose group (P < 0.003), whereas the mean diffMPH(SDS) was equal: -0.42 +/- 0.46 and -0.48 +/- 0.67, respectively. Gain in height(SDS) 0-2 yr was equal for the GH-deficient and ISS groups: 1.31 +/- 0.47 and 1.36 +/- 0.47, respectively, when ISS was classified on the basis of maximum GH peak on the arginine-insulin tolerance test or 24-h profile. CONCLUSION: Individualized GH doses during catch-up growth significantly reduce the proportion of unexpectedly good and poor responders around a predefined individual growth target and result in equal growth responses in children with GHD and ISS.

    • Evaporation of water intercepted by vegetation represents an important (sometimes major) part of evapotranspiration in temperate regions. Interception evaporation is an important process where insufficient measurement techniques hamper progress in knowledge and modeling. An ideal technique to study the interception evaporation process should monitor intercepted mass (and its vertical distribution) and interception loss with high accuracy (0.1 mm) and time resolution (1 min), and give correct area estimates. The method should be inexpensive, require minor supervision during extended periods, and work in dense forests. Net precipitation techniques, in which interception evaporation is determined from the difference between gross precipitation (measured with funnels) and throughfall (measured with funnels, troughs, or plastic sheet net-rainfall gauges) fulfill many of the requirements but usually have a too-low accuracy and time resolution for process studies. Precipitation measurements are normally affected by distortion of the wind field around gauges as well as by adhesive and evaporative losses. Throughfall measurements with precipitation funnels, troughs, or plastic sheet net-rainfall gauges, manually emptied or combined with tipping buckets, usually have too-low accuracy and time resolution for process studies and are impaired by adhesive losses. A new loadcell-based system to determine interception evaporation from gross and net precipitation is presented. A weighing gauge with minimal wind loss is used for precipitation, and weighing troughs are used for throughfall measurements. The weighing troughs minimize adhesive-loss errors and react instantaneously. Preliminary results with the method confirm that it can be used for process studies with a high accuracy (0.1 mm) and a high time resolution (1 min).

    • It is a major challenge in modem science to decrease the uncertainty in predictions of global climate change. One of the largest uncertainties in present-day global climate models resides with the understanding of processes in the soil-vegetation-atmosphere-transfer (SVAT) system. Continuous, long-term data are needed in order to correctly quantify balances of water, energy and CO2 in this system and to correctly model it. It is the objective of this paper to demonstrate how a combined system of existing sensor, computer, and network technologies could be set up to provide continuous and reliable long-term SVAT-process data from a forested site under almost all environmental conditions. The Central Tower Site (CTS) system was set up in 1993-1994 in a 25 m high boreal forest growing on a highly heterogeneous till soil with a high content of stones and blocks. It has successfully monitored relevant states and fluxes in the system, such as atmospheric fluxes of momentum, heat, water vapour and CO2, atmospheric profiles of temperature, water vapour, CO2, short-and long-wave radiation, heat storage in soil and trees, sap-dow and a variety of ecophysiological properties, soil-water contents and tensions, and groundwater levels, rainfall and throughfall. System uptime has been more than 90% for most of its components during the first 5 years of operation. Results from the first 5 years of operation include e.g., budgets for energy, water and CO2, information on important but rarely occurring events such as evaporation from snow-covered canopies, and reactions of the forest to extreme drought. The carbon budget shows that the forest may be a sink of carbon although it is still growing. The completeness of the data has made it possible to test the internal consistency of SVAT models. The pioneering set-up at the CTS has been adopted by a large number of SVAT-monitoring sites around the world. Questions concerning tower maintenance, long-term calibration plans, maintenance of sensors and data-collection system, and continuous development of the computer network to keep it up to date are, however, only partly of interest as a research project in itself. It is thus difficult to get it funded from usual research-funding agencies. The full value of data generated by the CTS system can best be appreciated after a decade or more of continuous operation. Main uses of the data would be to evaluate how SVAT models handle the natural variability of climate conditions, quantification of water. carbon and energy budgets during various weather conditions, rind development of new parameterisation schemes in global and regional climate models.

    • PURPOSE: Current positron emission tomography (PET) radioligands for detection of Aβ amyloid in Alzheimer's disease (AD) are not ideal for quantification. To improve the signal to noise ratio we have developed the radioligand [11C]AZD2184 and report here the first clinical evaluation.METHODS: Eight AD patients and four younger control subjects underwent 93-min PET measurements with [11C]AZD2184. A ratio approach using the cerebellum as reference region was applied to determine binding parameters.RESULTS: Brain uptake of [11C]AZD2184 peaked within 1 min at 3-4% of injected radioactivity. AD patients had high radioactivity in cortical regions while controls had uniformly low radioactivity uptake. Specific binding peaked within 30 min at which time standardized uptake value ratios (SUVR) ranged between 1.19 and 2.57.CONCLUSION: [11C]AZD2184 is a promising radioligand for detailed mapping of Aβ amyloid depositions in Alzheimer's disease, due to low non-specific binding, high signal to background ratio and reversible binding as evident from early peak equilibrium.

    • Lack of discharge data for model calibration is challenging for flood prediction in ungauged basins. Since establishment and maintenance of a permanent discharge station is resource demanding, a possible remedy could be to measure discharge only for a few events. We tested the hypothesis that a few flood-event hydrographs in a tropical basin would be sufficient to calibrate a bucket-type rainfall-runoff model, namely the HBV model, and proposed a new event-based calibration method to adequately predict floods. Parameter sets were chosen based on calibration of different scenarios of data availability, and their ability to predict floods was assessed. Compared to not having any discharge data, flood predictions improved already when one event was used for calibration. The results further suggest that two to four events for calibration may considerably improve flood predictions with regard to accuracy and uncertainty reduction, whereas adding more events beyond this resulted in small performance gains.

    • Concentration times in small and medium-sized basins (similar to 10-1000 km(2)) are commonly less than 24 h. Flood-forecasting models are thus required to provide simulations at high temporal resolutions (1 h-6 h), although time-series of input and runoff data with sufficient lengths are often only available at the daily temporal resolution, especially in developing countries. This has led to study the relationships of estimated parameter values at the temporal resolutions where they are needed from the temporal resolutions where they are available. This study presents a methodology to treat empirically model parameter dependencies on the temporal resolution of data in two small basins using a bucket-type hydrological model, HBV-light, and the generalised likelihood uncertainty estimation approach for selecting its parameters. To avoid artefacts due to the numerical resolution or numerical method of the differential equations within the model, the model was consistently run using modelling time steps of one-hour regardless of the temporal resolution of the rainfall-runoff data. The distribution of the parameters calibrated at several temporal resolutions in the two basins did not show model parameter dependencies on the temporal resolution of data and the direct transferability of calibrated parameter sets (e.g., daily) for runoff simulations at other temporal resolutions for which they were not calibrated (e.g., 3 h or 6 h) resulted in a moderate (if any) decrease in model performance, in terms of Nash-Sutcliffe and volume-error efficiencies. The results of this study indicate that if sub-daily forcing data can be secured, flood forecasting in basins with sub-daily concentration times may be possible with model-parameter values calibrated from long time series of daily data. Further studies using more models and basins are required to test the generality of these results.

    • The performance of hydrological models is affected by uncertainty related to observed climatological and discharge data. Although the latter has been widely investigated, the effects on hydrological models from different starting times of the day have received little interest. In this study, observational data from one tropical basin were used to investigate the effects on a typical bucket-type hydrological model, the HBV, when the definitions of the climatological and discharge days are changed. An optimization procedure based on a genetic algorithm was used to assess the effects on model performance. Nash-Sutcliffe efficiencies varied considerably between day definitions, with the largest dependence on the climatological-day definition. The variation was likely caused by how storm water was assigned to one or two daily rainfall values depending on the definition of the climatological day. Hydrological models are unlikely to predict high flows accurately if rainfall intensities are reduced because of the day definition.

    • Discharge observations and reliable rainfall forecasts are essential for flood prediction but their availability and accuracy are often limited. However, even scarce data may still allow adequate flood forecasts to be made. Here, we explored how far using limited discharge calibration data and uncertain forcing data would affect the performance of a bucket-type hydrological model for simulating floods in a tropical basin. Three events above thresholds with a high and a low frequency of occurrence were used in calibration and 81 rainfall scenarios with different degrees of uncertainty were used as input to assess their effects on flood predictions. Relatively similar model performance was found when using calibrated parameters based on a few events above different thresholds. Flood predictions were sensitive to rainfall errors, but those related to volume had a larger impact. The results of this study indicate that a limited number of events can be useful for predicting floods given uncertain rainfall forecasts.

    • Floods pose a threat to society that can cause large socio-economic damages and loss of life in many parts of the world. Flood-forecasting models are required to provide simulations at temporal resolutions higher than a day in basins with concentration times smaller than 24 h. However, data at such resolutions are commonly limited or not available, especially in developing or low-income countries. This thesis covers issues related to the scarcity and lack of high temporal-resolution hydro-meteorological data and explores methods where the value of existing data is maximised to improve flood prediction.By varying the starting time of daily records (the day definition), it was shown that this definition had large implications on model calibration and runoff simulation and therefore, should be considered in regionalisation and flood-forecasting applications. A method was developed to treat empirically model-parameter dependencies on the temporal resolution of data. Model parameters seemed to become independent of the temporal resolution of data when the modelling time-step was sufficiently small. Thus, if sub-daily forcing data can be secured, flood forecasting in basins with sub-daily concentration times may be possible using model-parameter values calibrated from time series of daily data. A new calibration method using only a few event hydrographs could improve flood prediction compared to a scenario with no discharge data. Two event hydrographs may be sufficient for calibration, but accuracy and reduction in uncertainty may improve if data on more events can be acquired. Using flood events above a threshold with a high frequency of occurrence for calibration may be as useful for flood prediction as using only extreme events with a low frequency of occurrence. The accuracy of the rainfall forecasts strongly influenced the predictive performance of a flood model calibrated with limited discharge data. Between volume and duration errors of the rainfall forecast, the former had the larger impact on model performance.The methods previously described proved to be useful for predicting floods and are expected to support flood-risk assessment and decision making during the occurrence of floods in data-scarce regions. Further studies using more models and basins are required to test the generality of these results.

    • The interaction of water vapor and sulfur dioxide (SO2) with single crystal cuprous oxide (Cu2O) surfaces of (100) and (111) termination was studied by photoelectron spectroscopy (PES) and density functional theory (DFT). Exposure to near-ambient pressures of water vapor, at 5 × 10-3 %RH and 293 K, hydroxylates both Cu2O surfaces with OH coverage up to 0.38 copper monolayers (ML) for (100) and 0.25 ML for (111). O 1s surface core level shifts indicate that the hydroxylation lifts the (3,0;1,1) reconstruction of the clean (100) surface. On both clean Cu2O terminations, SO2 adsorbs to unsaturated surface oxygen atoms to form SO3 species with coverage, after a saturating SO2 dose, corresponding to 0.20 ML on the Cu2O(100) surface and 0.09 ML for the Cu2O(111) surface. Our combined DFT and PES results suggest that the SO2 to SO3 transformation is largely facilitated by unsaturated copper atoms at the Cu2O(111) surface. SO3-terminated surfaces exposed to low doses of water vapor (=100 langmuirs) in ultrahigh vacuum show no adsorption or reaction. However, during exposure to near-ambient pressures of water vapor, the SO3 species dissociate, and sulfur replaces a Cu2O lattice oxygen in a reaction that forms Cu2S. The hydroxylation of the Cu2O surfaces is believed to play a central role in the reaction.

    • Pyruvate dehydrogenase (PDH) deficiency is an inherited disorder of carbohydrate metabolism, resulting in lactic acidosis and neurological dysfunction. In order to provide energy for the brain, a ketogenic diet has been tried. Both the disorder and the ketogenic therapy may influence energy production. The aim of the study was to assess hepatic glucose production, lipolysis and resting energy expenditure (REE) in an infant, given a ketogenic diet due to neonatal onset of the disease. Lipolysis and glucose production were determined for two consecutive time periods by constant-rate infusions of [1,1,2,3,3-2H5]-glycerol and [6,6-2H2]-glucose. The boy had been fasting for 2.5 h at the start of the sampling periods. REE was estimated by indirect calorimetry. Rates of glucose production and lipolysis were increased compared with those of term neonates. REE corresponded to 60% of normal values. Respiratory quotient (RQ) was increased, indicating a predominance of glucose oxidation. Blood lactate was within the normal range. Several mechanisms may underlie the increased rates of glucose production and lipolysis. A ketogenic diet will result in a low insulin secretion and reduced peripheral and hepatic insulin sensitivity, leading to increased production of glucose and decreased peripheral glucose uptake. Surprisingly, RQ was high, indicating active glucose oxidation, which may reflect a residual enzyme activity, sufficient during rest. Considering this, a strict ketogenic diet might not be the optimal choice for patients with PDH deficiency. We propose an individualised diet for this group of patients aiming at the highest glucose intake that each patient will tolerate without elevated lactate levels.

    • Introduction: Due to the rise in the number of patients with dementia the imperative for finding new diagnostic and treatment options becomes ever more pressing. While significant progress has been made in PET imaging of A beta aggregates both in vitro and in vivo, options for imaging tau protein aggregates selectively are still limited. Based on the work previously published by researchers from the Tohoku University, Japan, that resulted in the development of [F-18]THK-5351, we have undertaken an effort to develop a carbon-11 version of the identical structure - [C-11]THK-5351. In parallel, THK-5351 was also labeled with tritium ([H-3]THK-5351) for use in in vitro autoradiography (ARG). Methods: The carbon -11 labeling was performed starting with di-protected enantiomeric pure precursor-tertbutyl 5-(6-( (2S)-3-fluoro-2-(tetrahydro-2H-Pyran-2-yloxy)proPoxy)quinolin-2-yl)pyridin-2-yl carbamate, which was reacted with [C-11]MeI, using DMF as the solvent and NaH as base, followed by deprotection with trifluoroacetic acid/water mixture, resulting in enantiomerically pure carbon-11 radioligand,[C-11]THK-5351 (S)-1-fluoro-3-(2-(6-([C-11]methylamino)pyridin-3-yl)quinolin-6-yloxy)propan-2-ol. Tritium labeling and purification of [H-3]THK-5351 were undertaken using similar approach, resulting in [H-3]THK-5351 with RCP >99.8% and specific radioactivity of 13 GB q/mu mol. Results: [C-11]THK-5351 was produced in good yield (1900 +/- 355 MBq), specific radioactivity (SRA) (361 +/- 119 GBq/mu rnol at EOS + 20 min) and radiochemical purity (RCP) (>99.8%), with enantiomeric purity of 98.7%. [H-3]TM-5351 was evaluated for ARG of tau binding in post-mortem human brain tissue using cortical sections from one AD patient and one control subject. [H-3]THK-5351 binding density was higher in the AD patient compared to the control subject, the binding was displaced by unlabeled THK-5351 confirming specific [H-3] THK-5351 binding.     • The degree of belief we have in predictions from hydrologic models depends on how well they can reproduce observations. Calibrations with traditional performance measures such as the Nash-Sutcliffe model efficiency are challenged by problems including: (1) uncertain discharge data, (2) variable importance of the performance with flow magnitudes, (3) influence of unknown input/output errors and (4) inability to evaluate model performance when observation time periods for discharge and model input data do not overlap. A new calibration method using flow-duration curves (FDCs) was developed which addresses these problems. The method focuses on reproducing the observed discharge frequency distribution rather than the exact hydrograph. It consists of applying limits of acceptability for selected evaluation points (EPs) of the observed uncertain FDC in the extended GLUE approach. Two ways of selecting the EPs were tested – based on equal intervals of discharge and of volume of water. The method was tested and compared to a calibration using the traditional model efficiency for the daily four-parameter WASMOD model in the Paso La Ceiba catchment in Honduras and for Dynamic TOPMODEL evaluated at an hourly time scale for the Brue catchment in Great Britain. The volume method of selecting EPs gave the best results in both catchments with better calibrated slow flow, recession and evaporation than the other criteria. Observed and simulated time series of uncertain discharges agreed better for this method both in calibration and prediction in both catchments without resulting in overpredicted simulated uncertainty. An advantage with the method is that the rejection criterion is based on an estimation of the uncertainty in discharge data and that the EPs of the FDC can be chosen to reflect the aims of the modelling application e.g. using more/less EPs at high/low flows. While the new method is less sensitive to epistemic input/output errors than the normal use of limits of acceptability applied directly to the time series of discharge, it still requires a reasonable representation of the distribution of inputs. Additional constraints might therefore be required in catchments subject to snow. The results suggest that the new calibration method can be useful when observation time periods for discharge and model input data do not overlap. The new method could also be suitable for calibration to regional FDCs while taking uncertainties in the hydrological model and data into account.

  • Observational Uncertainties in Water-Resources Modelling in Central America : Methods for Uncertainty Estimation and Model Evaluation

    • Knowledge about spatial and temporal variability of hydrological processes is central for sustainable water-resources management, and such knowledge is created from observational data. Hydrologic models are necessary for prediction for time periods and areas lacking data, but are affected by observational uncertainties. Methods for estimating and accounting for such uncertainties in water-resources modelling are of high importance, especially in regions such as Central America. Observational uncertainties were addressed in three ways in this thesis; quality control, quantitative estimation and development of model-evaluation techniques that addressed unquantifiable uncertainties. A first step in any modelling study should be the quality control and concurrent analysis of the representativeness of the observational data. In the characterisation of the precipitation regime in the Choluteca River basin in Honduras, four different quality problems were identified and 22% of the daily data had to be rejected. The monitoring network was found to be insufficient for a comprehensive characterisation of the high spatiotemporal variability of the precipitation regime. Quantitative estimations of data uncertainties can be made when sufficient information is available. Discharge-data uncertainties were estimated with a fuzzy regression for time-variable rating curves and from official rating curves for 35 stations in Honduras. The uncertainties were largest for low flows, as a result of measurement uncertainties and natural variability. A method for calibration with flow-duration curves was developed which enabled calibration to the whole flow range, accounting for discharge uncertainty and calibration with non-overlapping time periods for model input and evaluation data. The method compared favourably to traditional calibration in a test using two models applied in basins with different runoff-generation processes. A post-hoc analysis made it possible to identify potential model-structure errors and periods of disinformative data. Flow-duration curves were regionalised and used for calibration of a Central-American water-balance model. The initial model uncertainty for the ungauged basins was reduced by 70%. Non-representative precipitation data were found to be the main obstacle to comprehensive regional water-resources modelling in Central America. These methods bridged several problems related to observational uncertainties in water-balance modelling. Estimates of prediction uncertainty are an important basis for all types of decisions related to water-resources management.

Few studies have investigated the uncertainties related to non-stationarity in the stage-discharge relationship. We calculated discharge and the associated uncertainty with a weighted fuzzy regression of rating curves applied within a moving time window, based on estimated uncertainties in the observed rating data. An 18-year-long dataset with unusually frequent ratings (1268 in total) was the basis of this study. A large temporal variability in the stage-discharge relationship was found especially for low flows. The time-variable rating curve resulted in discharge estimate differences of -60 to +90% for low flows and +/- 20% for medium to high flows when compared to a constant rating curve. The final estimated uncertainty in discharge was substantial and the uncertainty limits varied between -43 to +73% of the best discharge estimate.
