    • Six DNA regions were evaluated as potential DNA barcodes for Fungi, the second largest kingdom of eukaryotic life, by a multinational, multilaboratory consortium. The region of the mitochondrial cytochrome c oxidase subunit 1 used as the animal barcode was excluded as a potential marker, because it is difficult to amplify in fungi, often includes large introns, and can be insufficiently variable. Three subunits from the nuclear ribosomal RNA cistron were compared together with regions of three representative protein-coding genes (largest subunit of RNA polymerase II, second largest subunit of RNA polymerase II, and minichromosome maintenance protein). Although the protein-coding gene regions often had a higher percent of correct identification compared with ribosomal markers, low PCR amplification and sequencing success eliminated them as candidates for a universal fungal barcode. Among the regions of the ribosomal cistron, the internal transcribed spacer (ITS) region has the highest probability of successful identification for the broadest range of fungi, with the most clearly defined barcode gap between inter- and intraspecific variation. The nuclear ribosomal large subunit, a popular phylogenetic marker in certain groups, had superior species resolution in some taxonomic groups, such as the early diverging lineages and the ascomycete yeasts, but was otherwise slightly inferior to the ITS. The nuclear ribosomal small subunit has poor species-level resolution in fungi. ITS will be formally proposed for adoption as the primary fungal barcode marker to the Consortium for the Barcode of Life, with the possibility that supplementary barcodes may be developed for particular narrowly circumscribed taxonomic groups.

  • Status of the ESS elliptical cryomodules at CEA saclay

    • The first ESS prototype cryomodule with medium beta cavities named M-ECCTD is being assembled at CEA Saclay. The Q curves of the 4 cavities mounted inside the cryomodule are presented, and the four power couplers have been conditioned at high power before their assembly onto the cavity string. Completion of the M-ECCTD assembly outside clean room is in progress as well as the finalization of the RF power test stand preparation. RF power tests of the M-ECCTD will be performed during summer 2017. CEA is preparing the production of the ESS medium and high beta cryomodules of the series before the test of the M-ECCTD and the contracts for the procurement of the most critical components have already been signed.

  • Very compact FTTH Diplexer design using advanced wafer level fabrication methods

    • FTTH networks require implementing a diplexer at each user termination. According to most of the standards, this diplexer detects a download signal beam at 1.49ÎŒm and emits an upload signal beam at 1.31ÎŒm on the same single mode fibre. Both signals exhibit datarate speed below 2.5Gbps. Today, most of the diplexers are obtained by actively aligning a set of individual optoelectronic components and micro-optics. However, new manufacturing solutions satisfying very low cost and mass production capability requirements of this market would help to speed the massive spreading of this technology. In this paper, we present an original packaging design to manufacture Diplexer Optical Sub-Assembly for FTTH application. A dual photodiode is stacked over a VCSEL and detects both the download signal beam at 1.49ÎŒm passing through the laser and one part of the upload signal beam at 1.31ÎŒm for monitoring. To satisfy this approach, an innovative VCSEL has been designed to have a very high transmission at 1.49ÎŒm. All these components are mounted on a very small circuit board on glass including also integrated circuits such as transimpedance amplifier. So, the device combines advanced optoelectronic components and highly integrated Multi-Chip-Module on glass approach using collective wafer-level assembling technologies. For the single mode fibre optical coupling, active and passive alignment solutions are considered.

    • Autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) is a rare autoimmune disease causing a wide spectrum of autoimmune dysfunction potentially including diabetes of an autoimmune etiology. We have previously described a pair of discordant APECED siblings and pointed to a possible role of 5' insulin variable number of tandem repeats (VNTR) locus IDDM2 in the appearance of diabetes within this disease. In vitro studies have previously suggested that class I VNTR alleles were associated with decreased fetal thymic insulin expression. We genotyped the 5' INS VNTR locus and several flanking 11p15.5 markers in 5' Finnish APECED subjects and explored the possible contribution of IDDM2 in the development of diabetes. The shorter 5' INS VNTR class I alleles (<35 repeats) were more prevalent in the diabetic Finnish APECED subjects than in non-diabetic APECED subjects. Logistic regression analysis revealed that having 1 short (<35) VNTR allele did not increase the risk of developing diabetes (95% CI 0.6-27.0), whereas having 2 short alleles conferred a 43.5-fold increased risk (95% CI 3.0-634.6). We conclude that short 5' INS VNTR class I alleles play a role in susceptibility to autoimmune diabetes in the context of APECED.

    • In order to fit with the present and future needs of the Datacom transceiver market, newly designed high rate transmitter optical subassemblies (TOSAs) have to be compact, low cost and compatible with mass production. We propose here an innovative design strategy that reaches all these targets by integrating a 10Gbps 850nmVCSEL laser diode and its laser driver with other functionalities (e.g. power monitoring and thermal monitoring) in a small form factor package. Taking advantages of the optical properties of the VCSEL and using flip-chip techniques, the transmitter exhibits excellent hyperfrequency performances and compatibility with mass production due to the use of collective manufacturing technologies and passive optical alignment. This versatile approach is also applicable to high rate receivers, parallel optics emitters, and singlemode low cost transmitter integrating long wavelength VCSELs.

    • IODP Expedition 350 was the first to be drilled in the rear part of the Izu-Bonin, although severalsites had been drilled in the arc axis to fore-arc region; the scientific objective was to understand theevolution of the Izu rear arc, by drilling a deep-water volcaniclastic section with a long temporalrecord (Site U1437). The Izu rear arc is dominated by a series of basaltic to dacitic seamount chainsup to ~100-km long roughly perpendicular to the arc front. Dredge samples from these aregeochemically distinct from arc front rocks, and drilling was undertaken to understand this arcasymmetry. Site U1437 lies in an ~20-km-wide basin between two rear arc seamount chains, ~90-kmwest of the arc front, and was drilled to 1804 m below the sea floor (mbsf) with excellent recovery.We expected to drill a volcaniclastic apron, but the section is much more mud-rich than expected(~60%), and the remaining fraction of the section is much finer-grained than predicted from itsposition within the Izu arc, composed half of ashes/tuffs, and half of lapilli tuffs of fine grain size(clasts <3 cm). Volcanic blocks (>6.4 cm) are only sparsely scattered through the lowermost 25% ofthe section, and only one igneous unit was encountered, a rhyolite peperite intrusion at~1390 mbsf. The lowest biostratigaphic datum is at 867 mbsf (~6.5 Ma), the lowest palaeomagneticdatum is at ~1300 mbsf (~9 Ma), and the rhyolite peperite at ~1390 mbsf has yielded a U–Pb zirconconcordia intercept age of (13.6 + 1.6/−1.7) Ma. Both arc front and rear arc sources contributed tothe fine-grained (distal) tephras of the upper 1320 m, but the coarse-grained (proximal) volcani-clastics in the lowest 25% of the section are geochemically similar to the arc front, suggesting arcasymmetry is not recorded in rocks older than ~13 Ma.

    • In this communication, we will review the problems caused by cell-mediated gene therapy, taking skeletal muscle as a physiological model. In particular we have utilised vectors transferring telomerase under the control of retroviral promoters into human satellite cells. The set of results presented here has several implications regarding gene therapy trials. Nevertheless, more experiments will be required to fully validate this cellular model and to use telomerase to safely extend the lifespan of putative gene therapy vectors.

    • DNA phylogenetic comparisons have shown that morphology-based species recognition often underestimates fungal diversity. Therefore, the need for accurate DNA sequence data, tied to both correct taxonomic names and clearly annotated specimen data, has never been greater. Furthermore, the growing number of molecular ecology and microbiome projects using high-throughput sequencing require fast and effective methods for en masse species assignments. In this article, we focus on selecting and re-annotating a set of marker reference sequences that represent each currently accepted order of Fungi. The particular focus is on sequences from the internal transcribed spacer region in the nuclear ribosomal cistron, derived from type specimens and/or ex-type cultures. Re-annotated and verified sequences were deposited in a curated public database at the National Center for Biotechnology Information (NCBI), namely the RefSeq Targeted Loci (RTL) database, and will be visible during routine sequence similarity searches with NR_prefixed accession numbers. A set of standards and protocols is proposed to improve the data quality of new sequences, and we suggest how type and other reference sequences can be used to improve identification of Fungi.

    • The thermal component of Mercury's electron population has never been measured. One scientific objective of the Plasma Wave Investigation consortium, PWI, is to determine the influence of the thermal plasma upon the formation and dynamics of the planetary magnetosphere, as a function of solar activity. The Active Measurement of Mercury's Plasma experiment, AM(2)p, has been proposed as part of PWI, to monitor the density and temperature of the thermal electron population, during the whole mission of the Mercury Magnetospheric Orbiter of BepiColombo. These two physical parameters will be deduced from the measurements of the self- and mutual-impedances of the MEFISTO (Mercury Electric Field In Situ Tool) double-sphere antenna, in a frequency range comprising the expected plasma frequency. The in situ measurement of the antenna impedance is also essential for calibrating the electric antenna which measures the natural waves; it will allow, in particular, the effective length of the antenna to be calculated as a function of frequency and plasma conditions. The purpose of this paper is to define the scientific objectives of AM(2)p, to explain the principle of the measurement, to describe the electronic device, and to show the ability of AM 2p to make reliable and accurate measurements of the thermal plasma density and temperature in the Hermean magnetosphere, as well as in the solar wind at heliocentric distances of 0.31-0.47 AU. The potential performance of this instrument has been evaluated using both an analytical approach and numerical simulations.

  • RPC-MIP : The mutual impedance probe of the Rosetta Plasma Consortium

    • The main objective of the Mutual Impedance Probe (MIP), part of the Rosetta Plasma Consortium (RPC), is to measure the electron density and temperature of Comet 67P/Churyumov-Gerasimenko's coma, in particular inside the contact surface. Furthermore, MIP will determine the bulk velocity of the ionised outflowing atmosphere, define the spectral distribution of natural plasma waves, and monitor dust and gas activities around the nucleus. The MIP instrumentation consists of an electronics board for signal processing in the 7 kHz to 3.5 MHz range and a sensor unit of two receiving and two transmitting electrodes mounted on a 1-m long bar. In addition, the Langmuir probe of the RPC/LAP instrument that is at about 4 m from the MIP sensor can be used as a transmitter (in place of the MIP ones) and MIP as a receiver in order to have access to the density and temperature of plasmas at higher Debye lengths than those for which the MIP is originally designed.
