    • There is an urgent need to apply effective, data-driven approaches to reliably predict engineered nanomaterial (ENM) toxicity. Here we introduce a predictive computational framework based on the molecular and phenotypic effects of a large panel of ENMs across multiple in vitro and in vivo models. Our methodology allows for the grouping of ENMs based on multi-omics approaches combined with robust toxicity tests. Importantly, we identify mRNA-based toxicity markers and extensively replicate them in multiple independent datasets. We find that models based on combinations of omics-derived features and material intrinsic properties display significantly improved predictive accuracy as compared to physicochemical properties alone.

    • Globally, ciguatera fish poisoning (CFP) avoidance efforts rely primarily on local knowledge of the fish being consumed, its collection location, and association with illnesses. In 2016, several fish that appeared to be hybrids between a local commercially prized species, Ocyurus chrysurus, and a regionally prohibited species Lutjanus apodus (due to CFP concerns), were caught nearshore in United States Virgin Islands waters, leading to confusion regarding the safety of consuming the fish. The hybrid status of the fish was verified as O. chrysurus (male) x L. apodus (female) by comparing two sets of gene sequences (mitochondrial CO1 and nuclear S7). Using an in vitro mouse neuroblastoma (N2a) assay, one of the hybrid fish exhibited a composite cytotoxicity of 0.038 ppb Caribbean ciguatoxin-1 (C-CTX-1) equivalents (Eq.); a concentration below the US Food and Drug Administration (FDA) guidance level for safety in fish products for CFP (0.1 ppb C-CTX-1 Eq.) but approximately 2x above the maximum described in the commercially prized parent species (0.019 ppb C-CTX-1 Eq./g). C-CTX-1 was confirmed in the hybrid sample by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The second hybrid fish tested negative for CTXs. This research confirms hybridization between two species with contrasting commercial statuses, discusses CTX accumulation implications for hybridization, and provides a methodology for future studies into novel CFP vectors, with the goal of providing critical information for fishermen and consumers regarding CFP risk management.

    • The workshop was organised to discuss the validity and limitations of existing functional markers of Se status in human subjects and to identify future research priorities in this area. Studies presented as part of this workshop investigated: the bioavailability of Se from different dietary sources; potential functional markers of Se status; individual variation in response to Se; the effect of marginal Se status on immune function. The workshop highlighted the need to define the relationship between functional markers of Se status and health outcomes.

    • Garnets in continentally-derived high-pressure (HP) rocks of the Sesia Zone (Western Alps) have three different chemical zonation patterns, depending on sample locality. Comparison of observed garnet zonation patterns with thermodynamically modelled patterns shows that the different patterns are caused by differences in the water content of the subducted rocks during prograde metamorphism. Zonation patterns of garnets from water-saturated host rocks show typical prograde chemical zonations with steadily increasing pyrope content and increasing XMg, together with bell-shaped spessartine patterns. In contrast, garnets from water-undersaturated rocks have more complex zonation patterns with a characteristic decrease in pyrope and XMg between core and inner rim. In some cases, garnets show an abrupt compositional change in core-to-rim profiles, possibly due to water-undersaturation prior to HP metamorphism. Garnets from both water-saturated and water-undersaturated rocks show signs of intervening growth interruptions and core resorption. This growth interruption results from bulk-rock depletion caused by fractional garnet crystallisation. The water content during burial influences significantly the physical properties of the subducted rocks. Due to enhanced garnet crystallisation, water-undersaturated rocks, i.e. those lacking a free fluid phase, become denser than their water-saturated equivalents, facilitating the subduction of continental material. Although water-bearing phases such as phengite and epidote are stable up to eclogite-facies conditions in these rocks, dehydration reactions during subduction are lacking in water-undersaturated rocks up to the transition to the eclogite facies, due to the thermodynamic stability of such hydrous phases at high P-T conditions. Our calculations show that garnet zonation patterns strongly depend on the mineral parageneses stable during garnet growth and that certain co-genetic mineral assemblages cause distinct garnet zonation patterns. This observation enables one to interpret complex garnet growth zonation patterns in terms of garnet-forming reactions and water content during HP metamorphism, as well as to determine detailed P-T paths.

    • Numerical thermodynamic modelling of mineral composition and modes for specified pressure-temperature paths reveals the strong influence of fractional garnet crystallisation, as well as water fractionation, on garnet growth histories in high pressure rocks. Disequilibrium element incorporation in garnet due to the development of chemical inhomogeneities around porphyroblasts leads to pronounced episodic growth and may even cause growth interruptions. Discontinuous growth, together with pressure- and temperature-dependent changes in garnet chemistry, cause zonation patterns that are indicative of different degrees of disequilibrium element incorporation. Chemical inhomogeneities in the matrix surrounding garnet porphyroblasts strongly affect garnet growth and lead to compositional discontinuities and steep compositional gradients in the garnet zonation pattern. Further, intergranular diffusion-controlled calcium incorporation can lead to a characteristic rise in grossular and spessartine contents at lower metamorphic conditions. The observation that garnet zonation patterns diagnostic of large and small fractionation effects coexist within the same sample suggests that garnet growth is often controlled by small-scale variations in the bulk rock chemistry. Therefore, the spatial distribution of garnet grains and their zonation patterns, together with numerical growth models of garnet zonation patterns, yield information about the processes limiting garnet growth. These processes include intercrystalline element transport and dissolution of pre-existing grains. Discontinuities in garnet growth induced by limited element supply can mask traces of the thermobarometric history of the rock. Therefore, thermodynamic modelling that considers fractional disequilibrium crystallisation is required to interpret compositional garnet zonation in terms of a quantitative pressure and temperature path of the host rock

  • Multicenter analytical evaluation of a high-sensitivity troponin T assay

    • Background: High-sensitivity cardiac troponin assays are being introduced clinically for earlier diagnosis of acute myocardial infarction (AMI). We evaluated the analytical performance of a high-sensitivity cardiac troponin T assay (hscTnT. Roche Diagnostics) in a multicenter, international trial. Methods: Three US and 5 European sites evaluated hscTnT on the Modular (R) Analytics E170, cobas (R) 6000, Elecsys 2010, and cobas (R) e 411. Precision, accuracy, reportable range, an inter-laboratory comparison trial, and the 99th percentile of a reference population were assessed. Results: Total imprecision (CVs) were 4.6-36.8% between 3.4 and 10.3 ng/L hscTnT. Assay linearity was up to 10,000 ng/L and the limit of blank and detection were 3 and 5 ng/L, respectively. The 99th percentile reference limit was 14.2 ng/L (n = 533). No significant differences between specimen types, assay incubation time, or reagent lots existed. A substantial positive bias (76%) exists between the 4th generation and hscTnT assays at the low end of the measuring range (< 50 ng/L). hscTnT serum pool concentrations were within 2SD limits of the mean of means in the comparison trial, indicating comparable results across multiple platforms and laboratories. Conclusion: The Roche hscTnT assay conforms to guideline precision requirements and will likely identify additional patients with myocardial injury suspicious for AMI.

    • Engineered nanomaterials (ENMs) are widely present in our daily lives. Despite the efforts to characterize their mechanism of action in multiple species, their possible implications in human pathologies are still not fully understood. Here we performed an integrated analysis of the effects of ENMs on human health by contextualizing their transcriptional mechanism-of-action with respect to drugs, chemicals and diseases. We built a network of interactions of over 3,000 biological entities and developed a novel computational tool, INSIdE NANO, to infer new knowledge about ENM behavior. We highlight striking association of metal and metal-oxide nanoparticles and major neurodegenerative disorders. Our novel strategy opens possibilities to achieve fast and accurate read-across evaluation of ENMs and other chemicals based on their biosignatures.
