  • A low-gluten diet induces changes in the intestinal microbiome of healthy Danish adults

    • © 2018, The Author(s). Adherence to a low-gluten diet has become increasingly common in parts of the general population. However, the effects of reducing gluten-rich food items including wheat, barley and rye cereals in healthy adults are unclear. Here, we undertook a randomised, controlled, cross-over trial involving 60 middle-aged Danish adults without known disorders with two 8-week interventions comparing a low-gluten diet (2 g gluten per day) and a high-gluten diet (18 g gluten per day), separated by a washout period of at least six weeks with habitual diet (12 g gluten per day). We find that, in comparison with a high-gluten diet, a low-gluten diet induces moderate changes in the intestinal microbiome, reduces fasting and postprandial hydrogen exhalation, and leads to improvements in self-reported bloating. These observations suggest that most of the effects of a low-gluten diet in non-coeliac adults may be driven by qualitative changes in dietary fibres.

    • From exploratory studies and theoretical expectations it is known that simplifying approximations in spectroscopic analysis (local thermodynamic equilibrium (LTE), 1D) lead to systematic biases of stellar parameters and abundances. These biases depend strongly on surface gravity, temperature and, in particular, for LTE versus non-LTE (NLTE), on metallicity of the stars. Here we analyze the [Mg/Fe] and [Fe/H] plane of a sample of 326 stars, comparing LTE and NLTE results obtained using 1D hydrostatic models and averaged models. We show that compared to the NLTE benchmark, the other three methods display increasing biases toward lower metallicities, resulting in false trends of [Mg/Fe] against [Fe/H], which have profound implications for interpretations by chemical evolution models. In our best NLTE model, the halo and disk stars show a clearer behavior in the [Mg/Fe]-[Fe/H] plane, from the knee in abundance space down to the lowest metallicities. Our sample has a large fraction of thick disk stars and this population extends down to at least [Fe/H] ∼ -1.6 dex, further than previously proven. The thick disk stars display a constant [Mg/Fe] ≈ 0.3 dex, with a small intrinsic dispersion in [Mg/Fe] that suggests that a fast SN Ia channel is not relevant for the disk formation. The halo stars reach higher [Mg/Fe] ratios and display a net trend of [Mg/Fe] at low metallicities, paired with a large dispersion in [Mg/Fe]. These indicate the diverse origin of halo stars from accreted low-mass systems to stochastic/inhomogeneous chemical evolution in the Galactic halo.

    • The ArmazoNes high Dispersion Echelle Spectrograph (ANDES) is the optical and near-infrared high-resolution echelle spectrograph envisioned for the Extremely Large Telescope (ELT). We present a selection of science cases, supported by new calculations and simulations, where ANDES could enable major advances in the fields of stars and stellar populations. We focus on three key areas, including the physics of stellar atmospheres, structure, and evolution; stars of the Milky Way, Local Group, and beyond; and the star-planet connection. The key features of ANDES are its wide wavelength coverage at high spectral resolution and its access to the large collecting area of the ELT. These features position ANDES to address the most compelling questions and potentially transformative advances in stellar astrophysics of the decades ahead, including questions which cannot be anticipated today.

    • Atomic diusion and mixing processes in stellar interiors influence the structure and the surface composition of stars. Some of these processes cannot yet be modelled from the first principles, and they require calibrations. This limits their applicability in stellar models used for studies of stellar populations and Galactic evolution. Aims. Our main goal is to put constraints on the stellar structure and evolution models using new refined measurements of the chemical composition in stars of a Galactic open cluster. Methods.We used medium-resolution, 19 200 R 21 500, optical spectra of stars in the open cluster NGC2420 obtained within the Gaia-ESO survey. The sample covers all evolutionary stages from the main sequence to the red giant branch. Stellar parameters were derived using a combined Bayesian analysis of spectra, 2MASS photometry, and astrometric data from Gaia DR2. The abundances of Mg, Ca, Fe, and Li were determined from non-local thermodynamic equilibrium (NLTE) synthetic spectra, which were computed using one-dimensional (1D) and averaged three-dimensional (3D) model atmospheres. We compare our results with a grid of Code d'Evolution Stellaire Adaptatif et Modulaire (CESTAM) stellar evolution models, which include atomic diusion, turbulent, and rotational mixing. Results. We find prominent evolutionary trends in the abundances of Fe, Ca, Mg, and Li with the mass of the stars in the cluster. Furthermore, Fe, Mg, and Ca show a depletion at the cluster turn-o, but the abundances gradually increase and flatten near the base of the red giant branch. The abundance trend for Li displays a signature of rotational mixing on the main sequence and abrupt depletion on the sub-giant branch, which is caused by advection of Li-poor material to the surface. The analysis of abundances combined with the CESTAM model predictions allows us to place limits on the parameter space of the models and to constrain the zone in the stellar interior, where turbulent mixing takes place.

  • Reliability of orofacial quantitative sensory testing for pleasantness and unpleasantness

    • BACKGROUND: Quantitative sensory testing protocols for perceptions of pleasantness and unpleasantness based on the German Research Network on Neuropathic Pain protocol were recently introduced. However, there are no reliability studies yet published.AIM: To evaluate the intra-examiner (test-retest) and inter-examiner reliability for orofacial pleasantness and unpleasantness quantitative sensory testing protocols.METHODS: Sixteen healthy participants from Aarhus University (11 women and five men, mean age 24, range 21-26 years) contributed. Two examiners were trained in performing the entire quantitative sensory testing protocols for pleasantness and unpleasantness, which included the additional dynamic tactile stimulation test using a goat-hair brush. Each participant underwent examination of both protocols by each examiner (inter-examiner reliability) on day 1. They returned at least 8 days following the testing to be re-examined by one examiner (intra-examiner reliability). All testing was performed on the skin of the right mandibular mental region. The intraclass correlation (ICC) was used to determine reliability.RESULTS: For the protocol investigating pleasantness, the majority of parameters had good to excellent intra-examiner (11/14: Intraclass correlation 0.67-0.87) and inter-examiner (13/14: Intraclass correlation 0.62-0.96) reliabilities. Similarly, the protocol investigating unpleasantness had good to excellent intra-examiner (intraclass correlation 0.63-0.99) and inter-examiner (intraclass correlation 0.65-0.98) reliabilities for most (13/15) of the parameters.CONCLUSION:Intra and inter-examiner reliabilities in the majority of quantitative sensory testing parameters (apart from the summation ratio) investigating pleasantness and unpleasantness are acceptable when assessing somatosensory function of the orofacial region.Trial registration: NA.
