    • The folate metabolism enzyme MTHFD2 (methylenetetrahydrofolate dehydrogenase/cyclohydrolase) is consistently overexpressed in cancer but its roles are not fully characterized, and current candidate inhibitors have limited potency for clinical development. In the present study, we demonstrate a role for MTHFD2 in DNA replication and genomic stability in cancer cells, and perform a drug screen to identify potent and selective nanomolar MTHFD2 inhibitors; protein cocrystal structures demonstrated binding to the active site of MTHFD2 and target engagement. MTHFD2 inhibitors reduced replication fork speed and induced replication stress followed by S-phase arrest and apoptosis of acute myeloid leukemia cells in vitro and in vivo, with a therapeutic window spanning four orders of magnitude compared with nontumorigenic cells. Mechanistically, MTHFD2 inhibitors prevented thymidine production leading to misincorporation of uracil into DNA and replication stress. Overall, these results demonstrate a functional link between MTHFD2-dependent cancer metabolism and replication stress that can be exploited therapeutically with this new class of inhibitors. Helleday and colleagues describe a nanomolar MTHFD2 inhibitor that causes replication stress and DNA damage accumulation in cancer cells via thymidine depletion, demonstrating a potential therapeutic strategy in AML tumors in vivo.

  • Stress in children: how fifth-year pupils experience stress in everyday life.

    • In many cases, stress in children takes the form of psychosomatic symptoms. The prevalence of headache in children has increased, as has stomach pain and self-reported psychological problems. Many children also experience stress without exhibiting any symptoms, as children perceive, understand and evaluate stress in a variety of different ways. The aim of this study was to describe how fifth-year pupils experience stress in everyday life. An explorative and descriptive design based on phenomenology was chosen to investigate children's lifeworld and lived experience of the phenomenon of stress. The data analysis was based on Giorgi's method. The study comprised 29 children, who were interviewed regarding their experience of stress in everyday life. The results revealed five key constituents: 'fear of being late', 'not having sufficient time', 'physical and mental consequences', 'both a negative and a positive feeling' and 'experiencing significant others' stress'. Together, these key constituents form the overall result. This study demonstrates that stress is a part of children's everyday life. The children described stress that was often acute and linked to situations that occur in everyday life. The results can assist in identifying the factors behind the stress experienced by children. This in turn will facilitate observation and intervention when a child exhibits stress symptoms.

  • Tobacco cesssation - as we want it! : an interview study with young people

    • Background: Smoking among teenagers and young people is still common and about every third young smoker states that they want to quit smoking. There are no obvious evidence-based methods for tobacco cessation for young people, and therefore, the same methods are used for young people as well as adults. The aim was to study young people's views about what support they would like for quit smoking, with the intention of developing a model for tobacco cessation for young people through interviews with the target groups.Methods: A qualitative study using a descriptive and exploratory design was conducted. Young smokers 16 - 29 years old (n=25) were interviewed in five focus groups in southern Sweden. Data were analyzed by content analysis.Results: The results illuminate young people's suggestions to develop a model for tobacco cessation. Four main categories emerged; Existing support rejected - existing apps, support- by SMS, and self-help brochures did not appeal to young people. Agreement between friends - To sign an agreement between two friends which included a fee on relapse. Adult support - To organize the model with duo groups and give adapted support. Celebrate and competition - Lift the positive with the freedom of smoke and create the opportunity to win simple prizes to celebrate the achieved interim goals.Conclusions: The young people suggested that the model for tobacco cessation would contain adult support and involves an agreement between friends and has elements of celebration and competition. The challenge will be to implement this model into environments where young people live.

  • Adolescent Girls' Experiences of Underlying Social Processes Triggering Stress in Their Everyday Life : A grounded theory study

    • The aim of this study was to generate a theoretical model of underlying social processes that trigger stress in adolescent girls’ everyday life. In-depth interviews regarding the experiences of stress at home, school and during leisure time were conducted with 14 17-year-old schoolgirls. Data were analysed by means of the grounded theory method. Stress was triggered in the interaction between responsibility and the way in which the girls were encountered. Triggered emotional reactions took the form of four dimensions of stress included ambivalence, frustration, despair and downheartedness. These reactions were dependent on whether the girls voluntary assumed responsibility for various situations or whether they were forced, or felt they were being forced, to assume responsibility in interaction with an encounter characterized by closeness or distance. These forms of stress reactions could appear in one dimension and subsequently shift to another. From the public health perspective, the generated stress model can be used in the planning and implementation of future actions to prevent stress and promote well-being related to stress in adolescent girls.

  • Defining the genetics of systemic autoimmunity in mouse models of lupus

    • Systemic Lupus Erythematosus (SLE) is a chronic multi-organ autoimmune disease considered a prototype for autoantibody and immune complex-mediated tissue injury. Although autoantibodies against a wide diversity of self-antigens are characteristically found in this disease, an important hallmark is the presence of autoantibodies to nuclear antigens. Despite this common clinical feature, individual patients vary widely in the organ systems afflicted, disease severity, disease course, and response to treatment. These characteristics make clinical management of SLE challenging and highlight the need for effective and less toxic therapeutic interventions. Susceptibility to lupus has been shown in both human studies and mouse models to be dependent on genetic predisposition. Therefore, it is likely that knowledge of the genetic basis of SLE will be required before full understanding of SLE pathogenesis can be achieved. In this thesis, studies to define the genetic basis of lupus in an induced and two spontaneous models of the disease are presented. These studies encompass mapping, characterization of interval congenic mice, and cloning of the Lmb3 locus gene. In the first study, a genomewide mapping study was performed to define the genetic basis for resistance of the DBA/2 mice to mercury-induced autoimmunity. On chromosome 1, a single quantitative trait was linked with resistance to HgIA. These results linked the locus Hmr1 to a late stage of lupus with GN. Interval congenic mice are important tools to define and characterize the roles of different loci in lupus-like diseases. The second paper identifies the effect of NZB and NZW Lbw2 alleles on lupus susceptibility by using BWF1 mice with none, one or two copies of the lupus-predisposing NZB.Lbw2 locus. The lack of the NZB locus significantly reduced mortality, GN and B cell activation. IgM anti-chromatin levels in genome-wide mapping was linked only to Lmb2 and none of the known B cell hyperactivity-promoting genes were present in this location, which might indicate a novel B cell activation gene. The third study used reciprocal single locus interval-specific congenic mice to characterize the contribution of Lmb1-4 on the MRL-Faslpr and B6-Faslpr backgrounds. The Lmb3 locus on chromosome 7 was found to have the most prominent phenotype with clear effects on lymphoproliferation, GN and mortality. In the fourth paper the Lmb3 was cloned and shown to be a spontaneous nonsense mutation in the Coro1a gene that encodes an actin-binding and -regulatory protein. Upon further characterization, this genetic alteration was discovered to be a new lupus suppressing mutation that reduced T cell migration, activation, and survival. Our findings highlight the complexity of the genetics of lupus, and further suggest that genes involved in controlling the actin cytoskeleton might be potential targets for autoimmune therapeutics.

    • OBJECTIVE: The aim of this study was to evaluate a school-based adolescent health promotion programme with focus on well-being related to stress. STUDY DESIGN: Interventional and evaluative with tests before and after the intervention. The study was performed in two secondary schools in a town on the west coast of Sweden. METHODS: A health promotion programme comprising massage and mental training was implemented for a single academic year in one school (intervention school, 153 participants) in order to strengthen and maintain well-being. No intervention was implemented in the other school (non-intervention school, 287 participants). A questionnaire was developed and tested, resulting in 23 items distributed across the following six areas: self-reliance; leisure time; being an outsider; general and home satisfaction; school satisfaction; and school environment. RESULTS: A pre- and postintervention comparison of the six areas was made within each school. In the intervention school, the boys maintained a very good or good sense of well-being related to stress in all six areas, while the girls' sense of well-being was maintained in five areas and deteriorated in one area. In the non-intervention school, the boys maintained a very good or good sense of well-being related to stress in four areas and deteriorated in two areas, while the girls' sense of well-being was maintained in two areas and deteriorated in four areas. CONCLUSION: Massage and mental training helped to maintain adolescents' very good or good sense of well-being related to stress. A questionnaire with acceptable validity and reliability was developed and tested in order to evaluate the health promotional approach. However, there is a need for further study to develop both the intervention and the questionnaire for young people.

  • "Happy when they arrive, happy when they go home" : Focusing on promoting children's mental health creates a sense of trust at preschools

    • A need for research on young children’s mental health has been identified. Moreover, there is a need to enhance teachers’ skills regarding health promotion work in preschool. The aim of the study is to examine the impact of a specific course on mental health promotion among Swedish preschool teachers. Data were gathered through interviews and documentation. A qualitative content analysis was used. Three categories: ‘attention’, ‘belonging’ and ‘personal growth’ – and six subcategories emerged from the analysis. The latent content of these categories is described in the theme: ‘Increased awareness, in thoughts and actions, of promoting mental health may create a sense of trust at preschools’. The results show that a specific course to promote mental health enhanced the teachers’ awareness of mental health which improved their work with children. This contributed to the development of a sense of trust at preschools, which is important to children’s mental health.

To address these issues, we consider it is important that how we conceive learning include opportunities for learning outside the classroom. This study aimed to explore pupils’ experiences of being taught science subjects using outdoor learning, including the learning they achieved. The research was conducted using an ethnographic research design, a useful method for research in this type of environment where a greater understanding of teaching and learning processes is sought, enabling researchers to explore events more deeply. The study took place during one school year and was conducted for pupils in grade 6 in a school on Sweden’s west coast. This study provides unique insights into how these teacher efforts to offer outdoor learning opportunities have improved pupils’ willingness to participate. Likewise, how the pupils have developed knowledge of and used scientific concepts and skills in different contexts became visible when the pupils were allowed to try, explore, be creative, and observe in real life contexts in various places. In addition, the pupils have been given opportunities to ask, discuss and reflect on questions about various phenomena and processes in nature and humans. From the pupils’ point of view, this has contributed to their easier understanding of the content of the science subjects. These results significantly improve science literacy for science subjects in a school context (Holbrook & Rannikmae, 2009). © The Author(s) 2024.

    • Stress is a widespread phenomenon in society today, not least among children and adolescents. Stress-related ill-health has increased in this population and affects girls to a greater extent than boys. Against this background, it is important to acquire knowledge about measures that prevent stress, especially in girls. The aim of this study was therefore to illuminate adolescent girls' experiences and reflections about what makes everyday life less stressful. An explorative design, qualitative content analysis, was used. In-depth interviews were conducted with fifteen 17-year-old girls. The analysis comprised both manifest and latent content and revealed the girls' own experiences of and reflections about what makes everyday life less stressful. Three categories, ‘Enjoyment and Recovery’, ‘Trust’ and ‘Insight and Influence’, and nine subcategories emerged. The latent content of these categories is described by the theme ‘access to sources of strength’. It is essential that persons in the girls' surroundings are aware of all sources that provide the strength to resist and prevent stress in everyday life. A climate has to be created in all arenas of the girls' everyday life in which they can access these sources of strength. Utilizing the girls' experiences and views about what needs to be done is the first step towards a preventive and promotive mode of working on their own circumstances and wishes. This approach is consistent with the Ottawa Charter for Health Promotion, which emphasizes the importance of involving the target group.

    • Here, we show that a lupus-suppressing locus is caused by a nonsense mutation of the filamentous actin-inhibiting Coronin-1A gene. This mutation was associated with developmental and functional alterations in T cells including reduced migration, survival, activation, and Ca2+ flux. T-dependent humoral responses were impaired, but no intrinsic B cell defects were detected. By transfer of T cells, it was shown that suppression of autoimmunity could be accounted for by the presence of the Coro1a(Lmb3) mutation in T cells. Our results demonstrate that Coronin-1A is required for the development of systemic lupus and identify actin-cytoskeleton regulatory proteins as potential targets for modulating autoimmune diseases.

    • Scholars recommend that health promotion researchers engage practitioners in the analysis and reporting phase and expand their ability to share their research beyond academia. The purpose of this study was to draw benefit from physical education (PE) teachers’ discussions and reflections of the implementation of a health promotion intervention in school during research circle meetings. The health promotion intervention ‘Pulse for Health and Learning’ (PuLH) focused on moderate-to-vigorous physical activity, incorporating a child-centred coaching approach. This study has an action research approach. The research circle consisted of PE teachers (N = 22, approximately 18 per meeting) from eight primary and middle schools (from grades 4 to 9) in eight municipalities in Sweden and three researchers. The theory of ‘practice architectures’ was employed to interpret, discuss, and clarify what enables and constrain PE teachers’ implementation of the health promotion intervention. During the analysis, three discourses were identified: technical-rational discourse, participating discourse, and steering and supporting discourse. The practice architectures both enabled and constrained the implementation of PuLH. The research circle meetings stimulated critically conscious acting and decision-making through collaboration between PE teachers and together with researchers which improved the implementation of PuLH and contributed to PE teachers’ professional development.

  • Research circle in a health promotion intervention : Developing physical education teacher’s child-centred coaching approach

    • Background: Research has shown that professional development positively influences teachers’ capacity to reflect on new knowledge and practice. One way of working with collegial learning in schools is through so-called research circles, in which teachers and researchers collaborate on specific content.Purpose: This presentation aims to illuminate how a research circle can develop and deepen physical education teachers’ (PE) critical reflection on a child-centred coaching approach.Methods: The research circle was conducted within the health promotion intervention. The intervention focused on moderate-to-vigorous physical activity (MVPA), incorporating a child-centred coaching approach. The research circle consisted of three full days during a school year with PE teachers (N=22, approximately 18 per meeting) from eight primary and middle schools (from grades 4 to 9) in eight municipalities in Sweden, and three researchers. The data consist of group discussions audio-recorded and transcribed verbatim in the research circle meetings.Findings: During a thematic analysis, three themes were identified: Creating an inclusive environment, Challenges to motivate all pupils, and Adapting MVPA activities.Conclusion: The research circle contributed to critical reflections that improved actions in the intervention and everyday practice, as the teachers learned from each other and the researchers when critically reflecting on a childcentred perspective on practice.

  • Myocardial strains from 3D DENSE magnetic resonance imaging

    • The ability to measure and quantify myocardial motion and deformation provides a useful tool to assist in the diagnosis, prognosis and management of heart disease. The recent development of magnetic resonance imaging methods, such as harmonic phase and displacement encoding with stimulated echoes (DENSE), make detailed non-invasive 3D transmural kinematic analyses of human myocardium possible in the clinic and for research purposes. As data acquisition technologies improve, quantification methods for cardiac kinematics need to be adapted and validated on the new types of data. In the present paper, a previously presented polynomial method for cardiac strain quantification is extended to quantify 3D strains from DENSE magnetic resonance imaging data. The method yields accurate results when validated against an analytical standard, and is applied to in vivo data from a healthy  human heart. The polynomial field is capable of resolving the measured material positions from the in vivo data, and the obtained in vivo strains agree

    • BackgroundThe ability to measure and quantify myocardial motion and deformation provides a useful tool to assist in the diagnosis, prognosis and management of heart disease. The recent development of magnetic resonance imaging methods, such as harmonic phase analysis of tagging and displacement encoding with stimulated echoes (DENSE), make detailed non-invasive 3D kinematic analyses of human myocardium possible in the clinic and for research purposes. A robust analysis method is required, however.MethodsWe propose to estimate strain using a polynomial function which produces local models of the displacement field obtained with DENSE. Given a specific polynomial order, the model is obtained as the least squares fit of the acquired displacement field. These local models are subsequently used to produce estimates of the full strain tensor.ResultsThe proposed method is evaluated on a numerical phantom as well as in vivo on a healthy human heart. The evaluation showed that the proposed method produced accurate results and showed low sensitivity to noise in the numerical phantom. The method was also demonstrated in vivo by assessment of the full strain tensor and to resolve transmural strain variations.ConclusionsStrain estimation within a 3D myocardial volume based on polynomial functions yields accurate and robust results when validated on an analytical model. The polynomial field is capable of resolving the measured material positions from the in vivo data, and the obtained in vivo strains values agree with previously reported myocardial strains in normal human hearts.

    • A quantitative analysis of myocardial mechanics is fundamental to the understanding of cardiac function, diagnosis of heart disease and assessment of therapeutic intervention. In the clinical situation, where limited scan time often is important, a detailed analysis of the myocardium in a specific region might be more applicable than a full 3D measurement of the entire left ventricle. This paper presents a method to obtain temporal evolutions of transmural 3D Lagrangian strains from two intersecting 2D planes of slice followed cine displacement encoding with stimulated echoes (DENSE) data using a bilinear-cubic polynomial element to resolve strain from the displaced myocardial positions. The method demonstrates accurate results when validated in an analytical model, and has been applied to in vivo data acquired on a 3 T magnetic resonance (MR) system from a healthy volunteer to quantify systolic strains at the anterior-basal region of left ventricular myocardium. The in vivo results agree within experimental accuracy with values reported in the literature.

    • The development of non-genotoxic therapies that activate wild-type p53 in tumors is of great interest since the discovery of p53 as a tumor suppressor. Here we report the identification of over 100 small-molecules activating p53 in cells. We elucidate the mechanism of action of a chiral tetrahydroindazole (HZ00), and through target deconvolution, we deduce that its active enantiomer (R)-HZ00, inhibits dihydroorotate dehydrogenase (DHODH). The chiral specificity of HZ05, a more potent analog, is revealed by the crystal structure of the (R)-HZ05/DHODH complex. Twelve other DHODH inhibitor chemotypes are detailed among the p53 activators, which identifies DHODH as a frequent target for structurally diverse compounds. We observe that HZ compounds accumulate cancer cells in S-phase, increase p53 synthesis, and synergize with an inhibitor of p53 degradation to reduce tumor growth in vivo.     • This paper highlights a current phenomenon reported from preschools placed in multilingual areas in Sweden, namely that some preschoolers with mutually different language backgrounds sometimes use English as lingua franca instead of Swedish during play. The data stems from a study of language environments in Swedish preschools situated in both monolingual and multilingual areas. The analyses reveal that many children are influenced by the English language in both areas, but to a much greater extent in multilingual areas. An interesting situation arises when the majority language of society, which is also the language of education and lingua franca of the preschool, acquires a less prevailing role in children's accomplishment of everyday practices. Data show that the participating children are exposed to and speak English to a varying extent. They learn and teach each other English, and speak English in an array of pragmatic purposes; to position themselves in the social hierarchies of the preschool group, to create meaning within their shared peer culture and for the purpose of exclusion of intruders. English is also used as a secret language of friendship.

    • In order to improve the learning conditions and health of schoolchildren, the Pulse for Learning and Health [PuLH] program in Sweden has introduced additional mandatory moderate to vigorous physical activity [MVPA] that lasts for 30 min three times a week. The PE teachers used a child-centered coaching approach to support all pupils. The aim of this study was to explore pupils' perception and experience of PuLH that has been implemented in primary and middle schools in Sweden. We have taken into account children's rights perspectives and adopted an exploratory and interpretive approach. In total, 73 pupils (34 girls, 39 boys, grades 4-9) were recruited through purposive sampling. 13 focus group interviews (n = 71) and individual interviews (n = 2) were carried out. All interviews were analyzed using qualitative content analysis. The analysis resulted in three major themes: 'promotes academic performance and a learning school environment', 'promotes health and well-being', and 'individual and structural barriers'. From children's perspective, the results highlight the importance of teachers and principals taking into account the interests and needs of all pupils, to have a well-planned MVPA intervention and to deal with issues regarding body ideals.

    • To improve academic performance and health in schoolchildren, the program Pulse for learning and health [PuLH] in Sweden has introduced moderate to vigorous physical activities [MVPA] three times a week (á 30 minutes). The teachers used a child-centered coaching approach, which has the child’s best interests in mind and focused on having fun. The objective of this contribution is to describe what Swedish pupils convey, using their own voices, about their experiences of participating in the school-based program PuLH. We have taken on children’s perspectives in order to be able to understand the children’s own culture. Eight schools in the Region of Jönköping, Sweden, have implemented PuLH. In total, 73 pupils (34 girls, 39 boys, grades 4-9) were recruited through purposive sampling. Thirteen focus group interviews (n=71) and individual interviews (n=2) were carried out. An explorative design was employed, in which data were categorized using qualitative content analysis. The analysis resulted in four major themes, which we classify as ‘promotes academic performance’, ‘promotes a learning school environment’, ‘promotes health and well-being’, and ‘individual and structural barriers’. The pupils stated that they increased their working capacity, the classroom environment became more calm and peaceful, they got more cohesion and new friends, their needs and interest were in focus, they felt more alert and got in a better mood, they increased their physical competence, their habits became healthier and illness decreased. However, some of the pupils revealed individual and structural barriers, which reduced pleasure and motivation to participate in MVPA due to; poor planning and that they felt time pressure, felt uncomfortable showing their bodies and digital challenges. We recommend that schools take pupils’ interests and needs into consideration when planning for and implementing MVPA activities and not use MVPA as a 'quick fix' i.e. to solve complex problems regarding pupil's learning.

    • De främsta miljöerna där barn/ungdomar vistas är i hemmet, skolan och fritiden. Dessa miljöer har därmed stor betydelse för ungdomars hälsa och välbefi nnande. Under sin uppväxt tillbringar t.ex. barn mer än 15 000 timmar i skolan!Det är viktigt att ställa sig frågorna; Vad kan göras för att bevara, stärka och främja hälsan och välbefi nnandet för alla ungdomar? Vilka generella insatser har betydelse oavsett om det gäller i hemmet, i skolan eller på fritidsarenan? Vad kan göras för att förebygga och komma tillrätta med den ohälsa som vi kan se drabbar ungdomarna idag? Vilka specifi ka insatser har betydelse? Det är viktigt med konkreta insatser i många fall omgående, men det är också viktigt att tänka långsiktigt, att våga satsa tidigt och tänka efter före. Att även satsa på forsknings- och utvecklingsprojekt behövs för att på ett vetenskapligt sätt kunna följa upp konsekvenserna av olika insatser för att insatserna i framtiden skall bli så rätta som möjligt. Detta behöver göras för både generella och specifi ka insatser.Hälsa och välbefi nnande är ett välfärdsmått och därmed en politisk fråga på alla plan, lokalt, regionalt, nationellt och globalt. Barn och ungdomars hälsa och välbefi nnande är också en angelägenhet för alla, vår framtid, våga därför satsa nu! Delaktighet och infl ytande har positiv effekt på både hälsa och välbefinnande, låt därför barn och ungdomar i än större omfattning vara med och bestämma i de frågor som rör dem. Att utgå från Barnkonventionen ”att barns bästa ska sättas i främsta rummet samt att barn och unga har rätt till bästa uppnåeliga hälsa” i alla beslut som fattas är ett utmärkt utgångsläge – för framtiden!

    • Schools are recognized worldwide as settings for health promotion, and leadership has a bearing on schools’ ability to be health promoting. School managers have a great influence on what is prioritized in school, which in turn affects students’ school performance and health. There is lack of research into school managers’ views on health promotion, and what they consider to be central to health promotion. The aim was therefore to examine school managers’ views about what health promotion in schools include. An explorative design, qualitative content analysis, was performed. In-depth interviews were conducted with all 13 school managers of a middle-sized municipality in central Sweden. The analysis had both manifest and latent content and three categories: ‘Organization and Collaboration’, ‘Optimize the arena’ and ‘Strengthen the individual’, and 10 subcategories emerged. The theme, ‘Opportunities for learning and a good life’, describes the latent content of these categories. Taking into account the views of school managers are important because these views help form a more complete picture of how school managers work with health promotion and what is needed to enhance health promotion to improve students’ opportunities for learning and a good life. The Ottawa Charter for Health promotion is thereby transformed into practice.

Qualitative contentanalysis was used.Results: Two categories emerged from the analysis:‘psychosocial climate’, which included the subcategories‘adults’ roles and responsibilities’ and ‘classmates’ normsand values’; ‘influence’, which included the subcategories‘changes in the physical environment’ and ‘flexible learn-ing’. The categories are seen as important to increaseschool satisfaction and improve social relations amongpeers at school.Conclusion: Examining children’s opinions is requestedand promoted by the UN convention on the Rights of theChild. The findings contribute to the field by showing howschool satisfaction and social relations might be improved,if the child perspective is considered in the planning ofhealth promotion activities in school

    • Type I IFN and nucleic acid-sensing TLRs are both strongly implicated in the pathogenesis of lupus, with most patients expressing IFN-induced genes in peripheral blood cells and with TLRs promoting type I IFNs and autoreactive B cells. About a third of systemic lupus erythematosus patients, however, lack the IFN signature, suggesting the possibility of type I IFN-independent mechanisms. In this study, we examined the role of type I IFN and TLR trafficking and signaling in xenobiotic systemic mercury-induced autoimmunity (HgIA). Strikingly, autoantibody production in HgIA was not dependent on the type I IFN receptor even in NZB mice that require type I IFN signaling for spontaneous disease, but was dependent on the endosomal TLR transporter UNC93B1 and the endosomal proton transporter, solute carrier family 15, member 4. HgIA also required the adaptor protein-3 complex, which transports TLRs from the early endosome to the late endolysosomal compartments. Examination of TLR signaling pathways implicated the canonical NF-kappa B pathway and the proinflammatory cytokine IL-6 in autoantibody production, but not IFN regulatory factor 7. These findings identify HgIA as a novel type I IFN-independent model of systemic autoimmunity and implicate TLR-mediated NF-kappa B proinflammatory signaling from the late endocytic pathway compartments in autoantibody generation.
