  • Functional Polymorphisms of FAS and FASL Gene andRisk of Breast Cancer – Pilot Study of 134 Cases

    • Fas/Fas ligand (FasL) system is one of the key apoptotic signaling entities in the extrinsic apoptotic pathway. De-regulation of this pathway, i.e. by mutations may prevent the immune system from the removal of newly-formed tumor cells, and thus lead to tumor formation. The present study investigated the association between −1377 G/A (rs2234767) and −670 A/G (rs1800682) polymorphisms in Fas as well as single nucleotide polymorphisms INV2nt −124 A/G (rs5030772) and −844 C/T (rs763110) in FasL in a sample of Iranian patients with breast cancer. This case-control study was done on 134 breast cancer patients and 152 normal women. Genomic DNA was extracted from whole blood samples. The polymorphisms were determined by using tetra-ARMS-PCR method. There was no significant difference in the genotype distribution of FAS rs2234767 polymorphism between cases and controls. FAS rs1800682, FASL rs5030772, and FASL rs763110 genotypes showed significant associations with an increasing risk of breast cancer (odds ratio OR = 3.18, P = 0.019; OR = 5.08, P = 0.012; OR = 2.40, P = 0.024, respectively). In conclusion, FAS rs2234767 was not associated with breast cancer risk. Though, FAS rs1800682, FASL rs5030772, and FASL rs763110 polymorphisms were associated with the risk of breast cancer in the examined population.

  • Mevalonate Cascade Inhibition by Simvastatin Induces the Intrinsic Apoptosis Pathway via Depletion of Isoprenoids in Tumor Cells

    • The mevalonate (MEV) cascade is responsible for cholesterol biosynthesis and the formation of the intermediate metabolites geranylgeranylpyrophosphate (GGPP) and farnesylpyrophosphate (FPP) used in the prenylation of proteins. Here we show that the MEV cascade inhibitor simvastatin induced significant cell death in a wide range of human tumor cell lines, including glioblastoma, astrocytoma, neuroblastoma, lung adenocarcinoma, and breast cancer. Simvastatin induced apoptotic cell death via the intrinsic apoptotic pathway. In all cancer cell types tested, simvastatin-induced cell death was not rescued by cholesterol, but was dependent on GGPP-and FPP-depletion. We confirmed that simvastatin caused the translocation of the small Rho GTPases RhoA, Cdc42, and Rac1/2/3 from cell membranes to the cytosol in U251 (glioblastoma), A549 (lung adenocarcinoma) and MDA-MB231( breast cancer). Simvastatin-induced Rho-GTP loading significantly increased in U251 cells which were reversed with MEV, FPP, GGPP. In contrast, simvastatin did not change Rho-GTP loading in A549 and MDA-MB-231. Inhibition of geranylgeranyltransferase I by GGTi-298, but not farnesyltransferase by FTi-277, induced significant cell death in U251, A549, and MDA-MB-231. These results indicate that MEV cascade inhibition by simvastatin induced the intrinsic apoptosis pathway via inhibition of Rho family prenylation and depletion of GGPP, in a variety of different human cancer cell lines.

  • The global burden of adolescent and young adult cancer in 2019: a systematic analysis for the Global Burden of Disease Study 2019

    • Background In estimating the global burden of cancer, adolescents and young adults with cancer are often overlooked, despite being a distinct subgroup with unique epidemiology, clinical care needs, and societal impact. Comprehensive estimates of the global cancer burden in adolescents and young adults (aged 15-39 years) are lacking. To address this gap, we analysed results from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2019, with a focus on the outcome of disability-adjusted life-years (DALYs), to inform global cancer control measures in adolescents and young adults. Methods Using the GBD 2019 methodology, international mortality data were collected from vital registration systems, verbal autopsies, and population-based cancer registry inputs modelled with mortality-to-incidence ratios (MIRs). Incidence was computed with mortality estimates and corresponding MIRs. Prevalence estimates were calculated using modelled survival and multiplied by disability weights to obtain years lived with disability (YLDs). Years of life lost (YLLs) were calculated as age-specific cancer deaths multiplied by the standard life expectancy at the age of death. The main outcome was DALYs (the sum of YLLs and YLDs). Estimates were presented globally and by Socio-demographic Index (SDI) quintiles (countries ranked and divided into five equal SDI groups), and all estimates were presented with corresponding 95% uncertainty intervals (UIs). For this analysis, we used the age range of 15-39 years to define adolescents and young adults. Findings There were 1.19 million (95% UI 1.11-1.28) incident cancer cases and 396 000 (370 000-425 000) deaths due to cancer among people aged 15-39 years worldwide in 2019. The highest age-standardised incidence rates occurred in high SDI (59.6 [54.5-65.7] per 100 000 person-years) and high-middle SDI countries (53.2 [48.8-57.9] per 100 000 person-years), while the highest age-standardised mortality rates were in low-middle SDI (14.2 [12.9-15.6] per 100 000 person-years) and middle SDI (13.6 [12.6-14.8] per 100 000 person-years) countries. In 2019, adolescent and young adult cancers contributed 23.5 million (21.9-25.2) DALYs to the global burden of disease, of which 2.7% (1.9-3.6) came from YLDs and 97.3% (96.4-98.1) from YLLs. Cancer was the fourth leading cause of death and tenth leading cause of DALYs in adolescents and young adults globally. Interpretation Adolescent and young adult cancers contributed substantially to the overall adolescent and young adult disease burden globally in 2019. These results provide new insights into the distribution and magnitude of the adolescent and young adult cancer burden around the world. With notable differences observed across SDI settings, these estimates can inform global and country-level cancer control efforts. Copyright (C) 2021 The Author(s). Published by Elsevier Ltd.

  • Pathways to electrochemical solar-hydrogen technologies

    • Solar-powered electrochemical production of hydrogen through water electrolysis is an active and important research endeavor. However, technologies and roadmaps for implementation of this process do not exist. In this perspective paper, we describe potential pathways for solar-hydrogen technologies into the marketplace in the form of photoelectrochemical or photovoltaic-driven electrolysis devices and systems. We detail technical approaches for device and system architectures, economic drivers, societal perceptions, political impacts, technological challenges, and research opportunities. Implementation scenarios are broken down into short-term and long-term markets, and a specific technology roadmap is defined. In the short term, the only plausible economical option will be photovoltaic-driven electrolysis systems for niche applications. In the long term, electrochemical solar-hydrogen technologies could be deployed more broadly in energy markets but will require advances in the technology, significant cost reductions, and/ or policy changes. Ultimately, a transition to a society that significantly relies on solar-hydrogen technologies will benefit from continued creativity and influence from the scientific community.

    • The proposed methodology combines a Daubechies (Db2) wavelet transform and the statistical parameters, skewness and kurtosis for the detection of microcalcifications in mammography. The effeciency of the discrete algorithm is heavily relied on the order of performing wavelet approximation and the computation of the statistical moments. The significant of wavelet and statistical parameter approaches, as well as the ordering issue in performing the analysis, are justified through implementing numerical examples for some clinical data. Finally, a ROC curve summarizing the performance of the CAD scheme is also presented.

  • Application of remote sensing techniques and machine learning algorithms in dust source detection and dust source susceptibility mapping

    • The aim of this research was to develop a method to produce a Dust Source Susceptibility Map (DSSM). For this purpose, we applied remote sensing and statistical-based machine learning algorithms for experimental dust storm studies in the Khorasan Razavi Province, in north-eastern Iran. We identified dust sources in the study area using MODIS satellite images during the 2005–2016 period. For dust source identification, four indices encompassing BTD3132, BTD2931, NDDI, and D variable for 23 MODIS satellite images were calculated. As a result, 65 dust source points were identified, which were categorized into dust source data points for training and validation of the machine learning algorithms. Three statistical-based machine learning algorithms were used including Weights of Evidence (WOE), Frequency Ratio (FR), and Random Forest (RF) to produce DSSM for the study region. We used land use, lithology, slope, soil, geomorphology, NDVI (Normalized Difference Vegetation Index), and distance from river as conditioning variables in the modelling. To check the performance of the models, we applied the Area Under the Curve (AUC) of the Receiver Operating Characteristic (ROC). As for the AUC success rate (training), the FR and WOE algorithms resulted in 82 and 83% accuracy, respectively, while the RF algorithm resulted in 91% accuracy. As for the AUC predictive rate (validation), the accuracy of all three models, FR, WOE, and RF, were 80, 81, and 88%, respectively. Although all three algorithms produced acceptable susceptibility maps of dust sources, the results indicated better performance of the RF algorithm.

    • Autophagy and apoptosis are basic physiologic processes contributing to the maintenance of cellular homeostasis. Autophagy encompasses pathways that target long-lived cytosolic proteins and damaged organelles. It involves a sequential set of events including double membrane formation, elongation, vesicle maturation and finally delivery of the targeted materials to the lysosome. Apoptotic cell death is best described through its morphology. It is characterized by cell rounding, membrane blebbing, cytoskeletal collapse, cytoplasmic condensation, and fragmentation, nuclear pyknosis, chromatin condensation/fragmentation, and formation of membrane-enveloped apoptotic bodies, that are rapidly phagocytosed by macrophages or neighboring cells. Neurodegenerative disorders are becoming increasingly prevalent, especially in the Western societies, with larger percentage of members living to an older age. They have to be seen not only as a health problem, but since they are care-intensive, they also carry a significant economic burden. Deregulation of autophagy plays a pivotal role in the etiology and/or progress of many of these diseases. Herein, we briefly review the latest findings that indicate the involvement of autophagy in neurodegenerative diseases. We provide a brief introduction to autophagy and apoptosis pathways focusing on the role of mitochondria and lysosomes. We then briefly highlight pathophysiology of common neurodegenerative disorders like Alzheimer's diseases, Parkinson's disease, Huntington's disease and Amyotrophic lateral sclerosis. Then, we describe functions of autophagy and apoptosis in brain homeostasis, especially in the context of the aforementioned disorders. Finally, we discuss different ways that autophagy and apoptosis modulation may be employed for therapeutic intervention during the maintenance of neurodegenerative disorders.

    • A complex of two S100 EF-hand calcium-binding proteins S100A8/A9 induces apoptosis in various cells, especially tumor cells. Using several cell lines, we have shown that S100A8/A9-induced cell death is not mediated by the receptor for advanced glycation endproducts (RAGE), a receptor previously demonstrated to engage S100 proteins. Investigation of cell lines either deficient in, or over-expressing components of the death signaling machinery provided insight into the S100A8/A9-mediated cell death pathway. Treatment of cells with S100A8/A9 caused a rapid decrease in the mitochondrial membrane potential (ΔΨm) and activated Bak, but did not cause release of apoptosis-inducing factor (AIF), endonuclease G (Endo G) or cytochrome c. However, both Smac/DIABLO and Omi/HtrA2 were selectively released into the cytoplasm concomitantly with a decrease in Drp1 expression, which inhibits mitochondrial fission machinery. S100A8/A9 treatment also resulted in decreased expression of the anti-apoptotic proteins Bcl2 and Bcl-XL, whereas expression of the pro-apoptotic proteins Bax, Bad and BNIP3 was not altered. Over-expression of Bcl2 partially reversed the cytotoxicity of S100A8/A9. Together, these data indicate that S100A8/A9-induced cell death involves Bak, selective release of Smac/DIABLO and Omi/HtrA2 from mitochondria, and modulation of the balance between pro- and anti-apoptotic proteins.

    • The complex formed by two members of the S100 calcium-binding protein family, S100A8/A9, exerts apoptosis-inducing activity against various cells, especially tumor cells. Here, we present evidence that S100A8/A9 also has cell growth-promoting activity at low concentrations. Receptor of advanced glycation end product (RAGE) gene silencing and cotreatment with a RAGE-specific blocking antibody revealed that this activity was mediated via RAGE ligation. To investigate the signaling pathways, MAPK phosphorylation and NF-κB activation were characterized in S100A8/A9-treated cells. S100A8/A9 caused a significant increase in p38 MAPK and p44/42 kinase phosphorylation, and the status of stress-activated protein kinase/JNK phosphorylation remained unchanged. Treatment of cells with S100A8/A9 also enhanced NF-κB activation. RAGE small interfering RNA pretreatment abrogated the S100A8/A9-induced NF-κB activation. Our data indicate that S100A8/A9-promoted cell growth occurs through RAGE signaling and activation of NF-κB.

    • The complex formed by two members of the S100 calcium-binding protein family, S100A8/A9, exerts apoptosis-inducing activity in various cells of different origins. Here, we present evidence that the underlying molecular mechanisms involve both programmed cell death I (PCD I, apoptosis) and PCD II (autophagy)-like death. Treatment of cells with S100A8/A9 caused the increase of Beclin-1 expression as well as Atg12-Atg5 formation. S100A8/A9-induced cell death was partially inhibited by the specific PI3-kinase class III inhibitor, 3-methyladenine (3-MA), and by the vacuole H+-ATPase inhibitor, bafilomycin-A1 (Baf-A1). S100A8/A9 provoked the translocation of BNIP3, a BH3 only pro-apoptotic Bcl2 family member, to mitochondria. Consistent with this finding, ΔTM-BNIP3 overexpression partially inhibited S100A8/A9-induced cell death, decreased reactive oxygen species (ROS) generation, and partially protected against the decrease in mitochondrial transmembrane potential in S100A8/A9-treated cells. In addition, either ΔTM-BNIP3 overexpression or N-acetyl-L-cysteine co-treatment decreased lysosomal activation in cells treated with S100A8/A9. Our data indicate that S100A8/A9-promoted cell death occurs through the cross-talk of mitochondria and lysosomes via ROS and the process involves BNIP3.

    • Type 1 von Willebrand disease (VWD) is characterized by a personal and family history of bleeding coincident with reduced levels of normal plasma von Willebrand factor (VWF). The molecular basis of the disorder is poorly understood. The aims of this study were to determine phenotype and genotype and their relationship in patients historically diagnosed with type 1 VWD. Families were recruited in 9 European countries based on previous type 1 VWD diagnosis. Bleeding symptoms were recorded, plasma phenotype analyzed, and VWF mutation analysis performed in all index cases (ICs). Phenotypic and molecular analysis stratified patients into those with or without phenotypes suggestive of qualitative VWF defects (abnormal multimers) and with or without mutations. A total of 105 of 150 ICs (70%) had mutations identified. A subgroup with abnormal multimers (38% of ICs, 57 of 150) showed a high prevalence of VWF gene mutations (95% of ICs, 54 of 57), whereas in those with qualitatively normal VWF, fewer mutations were identified (55% of ICs, 51 of 93). About one third of the type I VWD cases recruited could be reconsidered as type 2. The remaining group could be considered "true" type 1 VWD, although mutations were found in only 55%.

    • A novel cyber physical method is proposed and experimentally verified for reliable distributed estimation of vehicle longitudinal velocity, robustly to road friction condition variations. In this method, the vehicle speed estimated at each of the four corners of the vehicle, using a linear parameter-varying observer in the physical layer, and speed data measured by a conventional low-cost GPS are incorporated in a distributed structure (in the cyber layer) to enhance the reliability of the estimate. The method minimizes a cost function quantizing the effect of disturbances on each corner's estimation and adversaries due to occasional GPS signal drops. A fault-tolerant estimation policy is integrated to deal with large deviations in corner estimations, which have unexpectedly high levels of confidence. The main advantages of the proposed method are increased reliability on various road surface conditions and robustness to faults, as confirmed by road tests. Several experimental tests, including lane change and low-excitation maneuvers, with various powertrain configurations on dry and slippery roads demonstrate the efficiency of the algorithm.

    • Rhizopus oryzae is a filamentous fungus, belonging to the order Mucorales. It can ferment a wide range of carbohydrates hydrolyzed from lignocellulosic materials and even cellobiose to produce ethanol. However, R. oryzae also produces lactic acid as a major metabolite, which reduces the yield of ethanol. In this study, we show that significant reduction of lactic acid production could be achieved by short (25nt) synthetic siRNAs targeting the ldhA gene. The average yield of lactic acid production by R. oryzae during the batch fermentation process, where glucose had been used as a sole carbon source, diminished from 0.07gm/gm in wild type to 0.01gm/gm in silenced samples. In contrast, the average yield of ethanol production increased from 0.39gm/gm in wild type to 0.45gm/gm in silenced samples. These results show 85.7% (gm/gm) reduction in lactic acid production as compared with the wild type R. oryzae, while an increase of 15.4% (gm/gm) in ethanol yield.

  • Association between PD-1 and PD-L1 polymorphisms and the risk of cancer : a meta analysis of case-control studies

    • A number of case-control studies regarding the association of the polymorphisms in the programmed cell death 1 (PD-1) and programmed cell death ligand 1 (PD-L1) genes with the risk of cancer have yielded inconsistent findings. Therefore, we have conducted a comprehensive, updated meta-analysis study to identify the impact of PD-1 and PD-L1 polymorphisms on overall cancer susceptibility. The findings revealed that PD-1 rs2227981 and rs11568821 polymorphisms significantly decreased the overall cancer risk (Odds Ratio (OR) = 0.82, 95% CI = 0.68–0.99, p = 0.04, TT vs. CT+CC; OR = 0.79, 95% CI = 0.67–0.94, p = 0.006, AG vs. GG, and OR = 0.82, 95% CI = 0.70–0.96, p = 0.020, AG+AA vs. GG, respectively), while PD-1 rs7421861 polymorphism significantly increased the risk of developing cancer (OR = 1.16, 95% CI = 1.02–1.33, p = 0.03, CT vs. TT). The PD-L1 rs4143815 variant significantly decreased the risk of cancer in homozygous (OR = 0.62, 95% CI = 0.41–0.94, p = 0.02), dominant (OR = 0.70, 95% CI = 0.50–0.97, p = 0.03), recessive (OR = 0.76, 95% CI = 0.60–0.96, p = 0.02), and allele (OR = 0.78, 95% CI = 0.63–0.96, p = 0.02) genetic models. No significant association between rs2227982, rs36084323, rs10204525, and rs2890658 polymorphisms and overall cancer risk has been found. In conclusions, the results of this meta-analysis have revealed an association between PD-1 rs2227981, rs11568821, rs7421861, as well as PD-L1 rs4143815 polymorphisms and overall cancer susceptibility.

  • Association of CASP8 polymorphisms and cancer susceptibility: A meta-analysis

    • Caspase-8 plays is an essential enzyme in apoptosis pathway. Several investigation have been done to identify the relation between CASP8 polymorphisms and different human cancers, but, the findings are still debated. The aim of the current investigation is to assess if CASP8 rs3834129 (- 652 6N insertion/deletion), rs1045485 G > C, rs3769818 G > A, rs6723097 A > C, rs3769821 T > C, rs13113 T > A, rs3769825 G > A, rs2293554 A > C, and rs10931936 C > T polymorphisms are linked to susceptibility of cancer. Our team has extracted the eligible studies up to July 4, 2019, from different sources. Pooled odds ratios (ORs) with corresponding 95% confidence intervals (CIs) were estimated to quantitatively evaluate the association between CASP8 polymorphisms and cancer susceptibility. Our results showed that the rs3834129 and rs1045485 polymorphisms meaningfully reduced the risk of cancer, while the rs3769818, rs3769821 and rs3769825 polymorphisms considerably increased cancer susceptibility. No association of rs6723097, rs13113, rs2293554 and rs10931936 polymorphisms was observed with cancer susceptibility. The CASP8 rs3834129 polymorphism reduced the risk of gastrointestinal, digestive tract, colorectal, breast and lung cancers. Furthermore, the cancer risk was decreased in Asian and Caucasian populations as well as population- and hospital-based studies due to this polymorphism. There was not any relation between this gene polymorphism and the risk of prostate and cervical cancer development. Regarding the CASP8 rs1045485 polymorphism, the reduced breast cancer risk along with the risk of cancer in Caucasians, population- and hospital-based studies were observed.

    • Zinc is an essential trace element with cofactor functions in a large number of proteins of intermediary metabolism, hormone secretion pathways, immune defence mechanisms, and as a cofactor of transcription factors it is also involved in the control of gene expression. Our study demonstrates that the modulation of intra and extracellular zinc alone is sufficient to induce metabolic changes or even apoptosis in two model human breast cancer cell lines MCF-7 and MDA-MB468. Treatment of breast cancer cells with different concentrations of a cell membrane permeable zinc chelator, N,N,N'N'-tetrakis(2-pyridylmethyl)ethylenediamine (TPEN) and the membrane impermeable zinc chelator, diethylenctriaminepentacetic acid, (DTPA) resulted in a significant increase of cell death. Features of apoptosis, such as chromatin condensation and nuclear fragmentation accompanied the DTPA and TPEN-induced cell death. A significant increase in the activity of caspase-9 was observed in both cell lines; whereas, caspase-3 activity was only increased in MDA-MB468 cells since caspase-3 is not expressed in MCF-7 cells. Caspase-8 activation was negligible in both cell lines. Addition of Zn2+ or Cu2+ prevented DTPA and TPEN-induced cytotoxicity, indicating that both bivalent cations can be replaced functionally to a certain extent in our experimental system. Interestingly, addition of Ca2+, or Mg2+ had no effect. The antioxidant N-Acetyl-L-Cysteine inhibited the cytotoxic effect of DTPA and TPEN, indicating that oxidative stress is the likely mediator of Zn-deficiency-related cell death.     • Vinasse and whey are wastewaters that are produced in large quantities in the sugar-to-ethanol and dairy industries, respectively. They pose a considerable threat to the environment due to the high concentration of nutrients and COD. In this study, the potential of producing protein-rich fungal biomass and biomethane from vinasse and whey through a two-stage biorefinery was examined. In the first stage, an edible and safe for human filamentous fungus, Neurospora intermedia, was cultivated on these wastewaters. To maximize the fungal biomass yield, the cultivation parameters, i.e., pH, vinasse to whey ratio, incubation time, and nutrients supplementation, were optimized. The highest yield of 12.0 g biomass per L of wastewaters was obtained by cultivation at pH 6.5 and vinasse to whey ratio of 25:75 (v/v) for 96 h with nitrogen source supplementation. The N. intermedia biomass contained about 45% protein and noticeable essential amino acid contents, comparable to commercial sources of protein for aquatic feed such as soybean meal and fishmeal. In the second stage, the effluent of fungal cultivation was anaerobically digested to produce 425 mL/g VS biomethane. Overall, 1 m3 of wastewater yielded 5.4 kg crude protein and 10.3 m3 methane, accompanied by 93.3% COD removal.

    • Many factors are involved in urban heat island development such as lack of urban green spaces, improper choice of building materials, air pollution, densification, and other human activities. The aim of this research was to quantify the effects of land use/land cover (LU/LC) changes on urban land surface temperature (LST) during a 25-year period (1993-2018) for the semiarid Shiraz City in southern Iran using Landsat data (TM, ETM+, and OLI/TIRS) and machine learning algorithms. Five main LU/LC classes such as orchard, vegetation, bare surface, asphalt cover, and built-up areas were identified using a support vector machine algorithm. Landsat images were used to retrieve normalized difference vegetation index (NDVI) and normalized difference built-up index (NDBI). The LST and linear regression analysis were used to quantify the relationship between NDVI and NDBI. The results showed that the mean LST over the entire study domain increased considerably between 1993 and 2018, due to urbanization, decrease of green areas, increasing industrial areas, and other human activities. Built-up areas increased considerably by 25.8% from 80 to 100.6 km2 between 1993 and 2018, while vegetation cover decreased dramatically by 69.3%. Mean LST increased from 38.4 to 40.2°C during the 25-year period with a significant increase of 3.9°C between 2013 and 2018. In addition, the Urban heat island Ratio Index (URI) displayed a substantial upward trend during the 25-year period.

    • From 1985 to 2016, the prevalence of underweight decreased, and that of obesity and severe obesity increased, in most regions, with significant variation in the magnitude of these changes across regions. We investigated how much change in mean body mass index (BMI) explains changes in the prevalence of underweight, obesity, and severe obesity in different regions using data from 2896 population-based studies with 187 million participants. Changes in the prevalence of underweight and total obesity, and to a lesser extent severe obesity, are largely driven by shifts in the distribution of BMI, with smaller contributions from changes in the shape of the distribution. In East and Southeast Asia and sub-Saharan Africa, the underweight tail of the BMI distribution was left behind as the distribution shifted. There is a need for policies that address all forms of malnutrition by making healthy foods accessible and affordable, while restricting unhealthy foods through fiscal and regulatory restrictions.

    • This paper proposes an event-triggered add-on safety mechanism to adjust the control parameters for timely braking in a networked vehicular system while maintaining maneuverability. Passenger vehicle maneuverability is significantly affected by the combined slip friction effect, in which larger longitudinal tire slips result in considerable drop in lateral tire forces. This is of higher importance when unexpected dangerous situations occur on the road and immediate actions, such as braking, need to be taken to avoid collision. Harsh braking can lead to high-slip and loss of maneuverability; hence, timely braking is essential to reduce high-slip scenarios. In addition to the vehicles own active safety systems, the proposed event-triggered add-on safety is activated upon being informed about dangers by the road-side infrastructure. The aim is to incorporate the add-on safety feature to adjust the automatic control parameters for smooth and timely braking such that a collision is avoided while vehicle's maneuverability is maintained. We study two different wireless technologies for communication between the infrastructure and the vehicles, the Long-Term Evolution (LTE) and the fifth generation (5G) schemes.     • Optimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was <1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.

    • In this paper, we study the joint design of Doppler robust transmit sequence and receive filter to improve the performance of an active sensing system dealing with signal-dependent interference. The signal-to-noise-plus-interference (SINR) of the filter output is considered as the performance measure of the system. The design problem is cast as a max-min optimization problem to robustify the system SINR with respect to the unknown Doppler shifts of the targets. To tackle the design problem, which belongs to a class of NP-hard problems, we devise a novel method (which we call DESIDE) to obtain optimized pairs of transmit sequence and receive filter sharing the desired robustness property. The proposed method is based on a cyclic maximization of SINR expressions with relaxed rank-one constraints, and is followed by a novel synthesis stage. We devise synthesis algorithms to obtain high quality pairs of transmit sequence and receive filter that well approximate the behavior of the optimal SINR (of the relaxed problem) with respect to target Doppler shift. Several numerical examples are provided to analyze the performance obtained by DESIDE.

    •  In this paper, we study the problem of code design to improve the detection performance of multi-static radar in the presence of clutter (i.e., a signal-dependent interference). To this end, we briefly present a discrete-time formulation of the problem as well as the optimal detector in the presence of Gaussian clutter. Due to the lack of analytical expression for receiver operation characteristic (ROC), code design based on ROC is not feasible. Therefore, we consider several popular information-theoretic criteria including Bhattacharyya distance, Kullback-Leibler (KL) divergence, J-divergence, andmutual information (MI) as design metrics. The codeoptimization problems associated with different information-theoretic criteria are obtained and cast under a unified framework. We propose two general methods based on Majorization-Minimization to tackle the optimization problems in the framework. The first method provides optimal solutions via successive majorizations whereas the second one consists of a majorization step, a relaxation, and a synthesis stage. Moreover, derivations of the proposed methods are extended to tackle the code design problems with a peak-to-average ratio power (PAR) constraint. Usingnumerical investigations, a general analysis of the coded system performance, computational efficiency of the proposed methods, and the behavior of the information-theoretic criteria is provided.

    • There is over 15,600 km of track in the Swedish railroad network. This network is vital for the transportation of people and goods across the country. It is important that this network is monitored and maintained to ensure good function and safety. A tool for monitoring and measuring ground deformation over a large area remotely with high frequency and accuracy was developed in recent decades. This tool is known as Interferometric Synthetic Aperture Radar (InSAR), and is used by researchers, geo-technicians, and engineers.The purpose of this study has been to evaluate the use and feasibility of the InSAR technique for track condition monitoring and compare it to conventional track condition monitoring techniques. Malmbanan, which is primarily used to transport iron-ore from mines in Sweden to the ports of Luleå, Sweden and Narvik, Norway, is used as a case study for this project; specifically, the section between Kiruna and Riksgränsen. Coordinate matching of measurements from the provided Persistent Scatterer Interferometry (PSI) InSAR data and Optram data from survey trains were performed. Then measured changes over different time spans within the two systems were overlapped and classified with different thresholds to see if there is correlation between the two systems. An extensive literature review was also conducted in order to gain an understanding of InSAR technologies and uses.The literature review showed that there is a large potential and a quickly growing number of applications of InSAR to monitor railways and other types of infrastructure, and that the tools and algorithms for this are being improved. The case study, on the other hand, shows that it can be difficult to directly compare measurement series from different tools, each working on different resolutions in terms of both time and space. InSAR is thus not about to replace techniques such as those behind Optram (using measurement trains). Instead, the approaches offer complementary perspectives, each highlighting different types of issues.We find that InSAR offers a good way to identify locations with settlements or other types of ground motions. Especially transition zones between settlements and more stable ground can be challenging from a maintenance point of view and can clearly be identified and monitored using InSAR. With the rollout of national InSAR-data, and the large increase in data accessibility, we see a considerable potential for future studies that apply the technique to the railway area.

    • This paper is concerned with the network-theoretic properties of so-called k-nearest neighbor intelligent vehicular platoons, where each vehicle communicates with k vehicles, both in front and behind. The network-theoretic properties analyzed in this paper play major roles in quantifying the resilience and robustness of three generic distributed estimation and control algorithms against communication failures and disturbances, namely resilient distributed estimation, resilient distributed consensus, and robust network formation. Based on the results for the connectivity measures of the k-nearest neighbor platoon, we show that extending the traditional platooning topologies (which were only based on interacting with nearest neighbors) to k-nearest neighbor platoons increases the resilience of distributed estimation and control algorithms to both communication failures and disturbances.

    • Baker's yeast production industry generates large quantities of high chemical oxygen demand (COD) wastewater. The integration of baker's yeast wastewater (BYW) for an innovative two-step waste biorefinery process by producing protein-rich fungal biomass and biogas along with COD and nutrients removal was the main object of the present research. In the first step, fungal biomass production from BYW was investigated using four species of filamentous fungi. The maximum biomass yield of 5.13 g/L BYW containing 43.8% mycoprotein and 36.3% COD removal was achieved by A. oryzae. In the second step, to produce biogas and further remove organic matter, the effluent of fungal fermentation was subjected to anaerobic digestion and COD removal between 22.4 and 44.2% was obtained. Overall, 1 m3 of BYW yielded 5.13 kg of protein-rich biomass and 1.42 m3 of methane. Additionally, pigment production using N. intermedia was investigated, and 1.54 mg carotenoids/g biomass was produced.

    • The widespread use of nanomaterials has raised the threat of nanoparticles (NPs) infection of soils and groundwater resources. This research aims to investigate three parameters including flow velocity, ionic strength (IS), and initial particle concentration effects on transport behavior and retention mechanism of functionalization form of graphene oxide with polyvinylpyrrolidone (GO-PVP). The transport of GO-PVP was investigated in a laboratory-scale study through saturated/unsaturated (Saturation Degree = 0.91) sand columns. Experiments were conducted on flow velocity from 1.20 to 2.04 cm min−1, initial particle concentration from 10 to 50 mg L−1, and IS of 5–20 mM. The retention of GO-PVP was best described using the one-site kinetic attachment model in HYDRUS-1D, which accounted for the time and depth-dependent retention. According to breakthrough curves (BTCs), the lower transport related to the rate of mass recovery of GO-PVP was obtained by decreasing flow velocity and initial particle concentration and increasing IS through the sand columns. Increasing IS could improve the GO-PVP retention (based on katt and Smax) in saturated/unsaturated media; katt increases from 2.81 × 10−3 to 3.54 × 10−3 s−1 and Smax increases from 0.37 to 0.42 mg g−1 in saturated/unsaturated conditions, respectively. Our findings showed that the increasing retention of GO-PVP through the sand column under unsaturated condition could be recommended for the reduction of nanoparticles danger of ecosystem exposure.

    • To focus the incident light, both convex metalenses with hyperbolic phase profile and autofocused Airy (AFA) beams are widely used. To obtain AFA-based bifocal metalenses, we propose two methods in this article. One is to bring the two conjugate focal points of an AFA beam into the real space by applying a proper convex lens phase profile. The other is to use the inefficient central space between the two launched Airy beams with zero amplitude and constant phase profile as an independent metalens. We also show that combining the two introduced methods, leads the design to upgrade to a multifocal structure.

    • Fossil and archaeological evidence indicates that hominin dispersals into Southwest Asia occurred throughout the Pleistocene, including the expansion of Homo sapiens populations out of Africa. While there is evidence for hominin occupations in the Pleistocene in Iran, as evidenced by the presence of Lower to Upper Paleolithic archaeological sites, the extent to which humid periods facilitated population expansions into western Asia has remained unclear. To test the role of humid periods on hominin dispersals here we assess Paleolithic site distributions and paleoenvironmental records across Iran. We developed the first spatially comprehensive, high-resolution paleohydrological model for Iran in order to assess water availability and its influence on hominin dispersals. We highlight environmentally mediated routes which likely played a key role in Late Pleistocene hominin dispersals, including the expansion of H. sapiens and Neanderthals eastwards into Asia. Our combined analyses indicate that, during MIS 5, there were opportunities for hominins to traverse a northern route through the Alborz and Kopet Dagh Mountains and the Dasht-I Kavir desert owing to the presence of activated fresh water sources. We recognize a new southern route along the Zagros Mountains and extending eastwards towards Pakistan and Afghanistan. We find evidence for a potential northern route during MIS 3, which would have permitted hominin movements and species interactions in Southwest Asia. Between humid periods, these interconnections would have waned, isolating populations in the Zagros and Alborz Mountains, where hominins may have continued to have had access to water.

    • High blood cholesterol is typically considered a feature of wealthy western countries1,2. However, dietary and behavioural determinants of blood cholesterol are changing rapidly throughout the world3 and countries are using lipid-lowering medications at varying rates. These changes can have distinct effects on the levels of high-density lipoprotein (HDL) cholesterol and non-HDL cholesterol, which have different effects on human health4,5. However, the trends of HDL and non-HDL cholesterol levels over time have not been previously reported in a global analysis. Here we pooled 1,127 population-based studies that measured blood lipids in 102.6 million individuals aged 18 years and older to estimate trends from 1980 to 2018 in mean total, non-HDL and HDL cholesterol levels for 200 countries. Globally, there was little change in total or non-HDL cholesterol from 1980 to 2018. This was a net effect of increases in low- and middle-income countries, especially in east and southeast Asia, and decreases in high-income western countries, especially those in northwestern Europe, and in central and eastern Europe. As a result, countries with the highest level of non-HDL cholesterol—which is a marker of cardiovascular risk—changed from those in western Europe such as Belgium, Finland, Greenland, Iceland, Norway, Sweden, Switzerland and Malta in 1980 to those in Asia and the Pacific, such as Tokelau, Malaysia, The Philippines and Thailand. In 2017, high non-HDL cholesterol was responsible for an estimated 3.9 million (95% credible interval 3.7 million–4.2 million) worldwide deaths, half of which occurred in east, southeast and south Asia. The global repositioning of lipid-related risk, with non-optimal cholesterol shifting from a distinct feature of high-income countries in northwestern Europe, north America and Australasia to one that affects countries in east and southeast Asia and Oceania should motivate the use of population-based policies and personal interventions to improve nutrition and enhance access to treatment throughout the world.

Anti-HCV NS3 and anti-HBs antibodies were also used to confirm infection. We performed triple blind counting of events to determine the co-incidence of autophagy (LC3β punctuate), apoptosis (cleaved caspase-3), and unfolded protein response (GRP78) with HBV and HCV infection in hepatocytes. All statistical analyses were performed using SPSS software for Windows (Version 16 SPSS Inc, Chicago, IL, United States). P-values < 0.05 were considered statistically significant. Statistical analyses were performed with Mann-Whitney test to compare incidence rates for autophagy, apoptosis, and UPR in HBV- and HCV-infected cells and adjacent non-infected cells.RESULTS: Our results showed that infection of hepatocytes with either HBV and HCV induces significant increase (P < 0.001) in apoptosis (cleavage of caspase-3), autophagy (LC3β punctate), and UPR (increase in GRP78 expression) in the HCV- and HBV-infected cells, as compared to non-infected cells of the same biopsy sections. Our tissue microarray immunohistochemical expression analysis of LC3β in HBVNeg and HBVPos revealed that majority of HBV-infected hepatocytes display strong positive staining for LC3β. Interestingly, although XBP splicing in HBV-infected cells was significantly higher (P < 0.05), our analyses show a slight increase of XBP splicing was in HCV-infected cells (P > 0.05). Furthermore, our evaluation of patients with HBV and HCV infection based on stage and grade of the liver diseases revealed no correlation between these pathological findings and induction of apoptosis, autophagy, and UPR.CONCLUSION: The results of this study indicate that HCV and HBV infection activates apoptosis, autophagy and UPR, but slightly differently by each virus. Further studies are warranted to elucidate the interconnections between these pathways in relation to pathology of HCV and HBV in the liver tissue.

    • The cholesterol biosynthesis pathway, also known as the mevalonate (MVA) pathway, is an essential cellular pathway that is involved in diverse cell functions. The enzyme 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase (HMGCR) is the rate-limiting step in cholesterol biosynthesis and catalyzes the conversion of HMG-CoA to MVA.Given its role in cholesterol and isoprenoid biosynthesis, the regulation of HMGCR has been intensely investigated. Because all cells require a steady supply of MVA, both the sterol (i.e. cholesterol) and non-sterol (i.e. isoprenoid) products of MVA metabolism exert coordinated feedback regulation on HMGCR through different mechanisms. The proper functioning of HMGCR as the proximal enzyme in the MVA pathway is essential under both normal physiologic conditions and in many diseases given its role in cell cycle pathways and cell proliferation, cholesterol biosynthesis and metabolism, cell cytoskeletal dynamics and stability, cell membrane structure and fluidity, mitochondrial function, proliferation, and cell fate.The blockbuster statin drugs (‘statins’) directly bind to and inhibit HMGCR, and their use for the past thirty years has revolutionized the treatment of hypercholesterolemia and cardiovascular diseases, in particular coronary heart disease. Initially thought to exert their effects through cholesterol reduction, recent evidence indicates that statins also have pleiotropic immunomodulatory properties independent of cholesterol lowering.In this review we will focus on the therapeutic applications and mechanisms involved in the MVA cascade including Rho GTPase and Rho kinase (ROCK) signaling, statin inhibition of HMGCR, geranylgeranyltransferase (GGTase) inhibition, and farnesyltransferase (FTase) inhibition in cardiovascular disease, pulmonary diseases (e.g. asthma and chronic obstructive pulmonary disease (COPD)), and cancer.

  • Graphene Oxide Tablets for Sample Preparation of Drugs in Biological Fluids : Determination of Omeprazole in Human Saliva for Liquid Chromatography Tandem Mass Spectrometry

    • In this study, a novel sort of sample preparation sorbent was developed, by preparing thin layer graphene oxide tablets (GO-Tabs) utilizing a mixture of graphene oxide and polyethylene glycol on a polyethylene substrate. The GO-Tabs were used for extraction and concentration of omeprazole (OME) in human saliva samples. The determination of OME was carried out using liquid chromatography-tandem mass spectrometry (LC-MS/MS) under gradient LC conditions and in the positive ion mode (ESI+) with mass transitions of m/z 346.3 -> 198.0 for OME and m/z 369.98 -> 252.0 for the internal standard. Standard calibration for the saliva samples was in the range of 2.0-2000 nmol L-1. Limits of detection and quantification were 0.05 and 2.0 nmol L-1, respectively. Method validation showed good method accuracy and precision; the inter-day precision values ranged from 5.7 to 8.3 (%RSD), and the accuracy of determinations varied from -11.8% to 13.3% (% deviation from nominal values). The extraction recovery was 60%, and GO-Tabs could be re-used for more than ten extractions without deterioration in recovery. In this study, the determination of OME in real human saliva samples using GO-Tab extraction was validated.
