    • We report the detection of the ground state N, J = 1, 3/2 -> 1, 1/2 doublet of the methylidyne radical CH at similar to 532 GHz and similar to 536 GHz with the Herschel/ HIFI instrument along the sight-line to the massive star-forming regions G10.6-0.4 (W31C), W49N, and W51. While the molecular cores associated with these massive star-forming regions show emission lines, clouds in the diffuse interstellar medium are detected in absorption against the strong submillimeter background. The combination of hyperfine structure with emission and absorption results in complex profiles, with overlap of the different hyperfine components. The opacities of most of the CH absorption features are linearly correlated with those of CCH, CN, and HCO+ in the same velocity intervals. In specific narrow velocity intervals, the opacities of CN and HCO+ deviate from the mean trends, giving rise to more opaque absorption features. We propose that CCH can be used as another tracer of the molecular gas in the absence of better tracers, with [CCH]/[H2] similar to 3.2 +/- 1.1 x 10-8. The observed [CN]/[CH], [CCH]/[CH] abundance ratios suggest that the bulk of the diffuse matter along the lines of sight has gas densities nH = n(H) + 2n(H2) ranging between 100 and 1000 cm-3).

    • Context. C-3 is the smallest pure carbon chain detected in the dense environment of star-forming regions, although diatomic C-2 is detected in diffuse clouds. Measurement of the abundance of C-3 and the chemistry of its formation in dense star-forming regions has remained relatively unexplored. Aims. We aim to identify the primary C-3 formation routes in dense star-forming regions following a chemical network producing species like CCH and c-C3H2 in the star-forming cores associated with DR21(OH), a high-mass star-forming region. Methods. We observed velocity resolved spectra of four ro-vibrational far-infrared transitions of C-3 between the vibrational ground state and the low-energy nu(2) bending mode at frequencies between 1654-1897 GHz using HIFI on board Herschel, in DR21(OH). Several transitions of CCH and c-C3H2 were also observed with HIFI and the IRAM 30 m telescope. Rotational temperatures and column densities for all chemical species were estimated. A gas and grain warm-up model was used to obtain estimates of densities and temperatures of the envelope. The chemical network in the model was used to identify the primary C-3 forming reactions in DR21(OH). Results. We detected C-3 in absorption in four far-infrared transitions, P(4), P(10), Q(2), and Q(4). The continuum sources MM1 and MM2 in DR21(OH), though spatially unresolved, are sufficiently separated in velocity to be identified in the C-3 spectra. All C-3 transitions are detected from the embedded source MM2 and the surrounding envelope, whereas only Q(4) and P(4) are detected toward the hot core MM1. The abundance of C-3 in the envelope and MM2 is similar to 6 x 10(-10) and similar to 3 x 10(-9), respectively. For CCH and c-C3H2, we only detect emission from the envelope and MM1. The observed CCH, C-3 and c-C3H2 abundances are most consistent with a chemical model with n(H2) similar to 5 x 10(6) cm(-3), a post-warm-up dust temperature T-max = 30 K, and a time of similar to 0.7-3 Myr. Conclusions. Post-warm-up gas phase chemistry of CH4 released from the grain at t similar to 0.2 Myr and lasting for 1 Myr can explain the observed C-3 abundance in the envelope of DR21(OH), and no mechanism involving photodestruction of PAH molecules is required. The chemistry in the envelope is similar to the warm carbon chain chemistry found in lukewarm corinos. We interpret the observed lower C-3 abundance in MM1 as compared to MM2 and the envelope to be due to the destruction of C-3 in the more evolved MM1. The timescale for the chemistry derived for the envelope is consistent with the dynamical timescale of 2 Myr derived for DR21(OH) in other studies.

  • Functional and symptom impact of trametinib versus chemotherapy in BRAF V600E advanced or metastatic melanoma : quality-of-life analyses of the METRIC study

    • We report the first quality-of-life assessment of a MEK inhibitor in metastatic melanoma from a phase III study. Trametinib prolonged progression-free survival and improved overall survival versus chemotherapy in patients with BRAF V600 mutation-positive melanoma. Less functional impairment, smaller declines in health status, and less exacerbation of symptoms were observed with trametinib.In a randomized phase III study, trametinib prolonged progression-free survival and improved overall survival versus chemotherapy in patients with BRAF V600 mutation-positive melanoma. Patients' quality of life (QOL) was assessed at baseline and follow-up visits using the European Organisation for Research and Treatment of Cancer Core QOL questionnaire. In the primary efficacy population (BRAF V600E+, no brain metastases) from baseline to weeks 6 and 12, patients' global health status scores worsened by 4-5 points with chemotherapy but improved by 2-3 points with trametinib. Rapid and substantive reductions in QOL functionality (e.g. role functioning, 8-11 points at weeks 6 and 12) and symptom exacerbation (e.g. fatigue, 4-8 points; nausea and vomiting, 5 points, both at weeks 6 and 12) were observed in chemotherapy-treated patients. In contrast, trametinib-treated patients reported small improvements or slight worsening from baseline at week 12, depending on the functional dimension and symptom. The mean symptom-scale scores for chemotherapy-treated patients increased from baseline (symptoms worsened) for seven of eight symptoms at week 6 (except insomnia) and six of eight symptoms at week 12 (except dyspnea and insomnia). In contrast, at weeks 6 and 12, the mean symptom-scale scores for trametinib decreased from baseline (symptoms improved) for pain (11-12 points), insomnia (10-12 points), and appetite loss (1-5 points), whereas those for diarrhea worsened (15-16 points). Mixed-model repeated-measures analyses showed significant (P < 0.05) and/or clinically meaningful improvements (small to moderate) from baseline in favor of trametinib for global health; physical, role, and social functioning; fatigue; pain; insomnia; nausea and vomiting; constipation; dyspnea; and appetite at weeks 6 and/or 12. QOL results for the intent-to-treat population were consistent. This first QOL assessment for a MEK inhibitor in metastatic melanoma demonstrated that trametinib was associated with less functional impairment, smaller declines in health status, and less exacerbation of symptoms versus chemotherapy.

    • We present a comparative study of Finnish and Swedish free-text nursing narratives from intensive care. Although the two languages are linguistically very dissimilar, our hypothesis is that there are similarities that are important and interesting from a language technology point of view. This may have implications when building tools to support producing and using health care documentation. We perform a comparative qualitative analysis based on structure and content, as well as a comparative quantitative analysis on Finnish and Swedish Intensive Care Unit (ICU) nursing narratives. Our findings are that ICU nursing narratives in Finland and Sweden have many properties in common, but that many of these are challenging when it comes to developing language technology tools.

    • Correlating the electronic structure and magnetic response with the morphology and crystal structure of the same single ferromagnetic nanoparticle has been up to now an unresolved challenge. Here, we present measurements of the element-specific electronic structure and magnetic response as a function of magnetic field amplitude and orientation for chemically synthesized single Fe nanocubes with 18 nm edge length. Magnetic states and interactions of monomers, dimers, and trimers are analyzed by X-ray photoemission electron microscopy for different particle arrangements. The element-specific electronic structure can be probed and correlated with the changes of magnetic properties. This approach opens new possibilities for a deeper understanding of the collective response of magnetic nanohybrids in multifunctional materials and in nanomagnetic colloidal suspensions used in biomedical and engineering technologies. © 2011 American Chemical Society.

    • We present high resolution [N II] 205 μm (3^P_1 − 3^P_0) spectra obtained with Herschel-HIFI towards a small sample of far-infrared bright star forming regions in the Galactic plane: All sources display an emission line profile associated directly with the H II regions themselves. For the first time we also detect absorption of the [N II] 205 μm line by extended low-density foreground material towards W 31C and W 49N over a wide range of velocities. We attribute this absorption to the warm ionised medium (WIM) and find N(N+) ≈ 1.5×10^17 cm^-2 towards both sources. This is in agreement with recent Herschel-HIFI observations of [C II] 158 μm, also observed in absorption in the same sight-lines, if ≈7–10% of all C + ions exist in the WIM on average. Using an abundance ratio of [N]/[H] = 6.76×10^-5 in the gas phase we find that the mean electron and proton volume densities are ∼0.1–0.3 cm^-3 assuming a WIM volume filling fraction of 0.1–0.4 with a corresponding line-of-sight filling fraction of 0.46–0.74. A low density and a high WIM filling fraction are also supported by RADEX modelling of the [N II] 205 μm absorption and emission together with visible emission lines attributed mainly to the WIM. The detection of the 205 μm line in absorption emphasises the importance of a high spectral resolution, and also offers a new tool for investigation of the WIM.

    • As a part of the Herschel key programme PRISMAS, we have used the Herschel-HIFI instrument to observe interstellar nitrogen hydrides along the sight-lines towards eight high-mass star-forming regions in order to elucidate the production pathways leading to nitrogen-bearing species in diffuse gas. Here, we report observations towards W49N of the NH N = 1 - 0, J = 2 - 1, and J = 1 - 0, ortho-NH2 N_(Ka, K_c) J = 1_(1,1) 3/2 - 0_(0,0) 1/2, ortho-NH3 J_K = 1_0 - 0_0 and 2_0 - 1_0, para-NH3 J_K = 2_1 - 1_1 transitions, and unsuccessful searches for NH+. All detections show absorption by foreground material over a wide range of velocities, as well as absorption associated directly with the hot-core source itself. As in the previously published observations towards G10.6-0.4, the NH, NH2 and NH3 spectra towards W49N show strikingly similar and non-saturated absorption features. We decompose the absorption of the foreground material towards W49N into different velocity components in order to investigate whether the relative abundances vary among the velocity components, and, in addition, we re-analyse the absorption lines towards G10.6-0.4 in the same manner. Abundances, with respect to molecular hydrogen, in each velocity component are estimated using CH, which is found to correlate with H2 in the solar neighbourhood diffuse gas. The analysis points to a co-existence of the nitrogen hydrides in diffuse or translucent interstellar gaswith a high molecular fraction. Towards both sources, we find that NH is always at least as abundant as both o-NH2 and o-NH3, in sharp contrast to previous results for dark clouds. We find relatively constant N(NH)/N(o-NH3) and N(o-NH2)/N(o-NH3) ratios with mean values of 3.2 and 1.9 towards W49N, and 5.4 and 2.2 towards G10.6-0.4, respectively. The mean abundance of o-NH4 is ~2x10^-9 towards both sources. The nitrogen hydrides also show linear correlations with CN and HNC towards both sources, and looser correlations with CH. The upper limits on the NH+ abundance indicate column densities

    • Purpose: During our efforts to develop tumor-infiltrating lymphocyte (TIL) therapy to counter the devastating recurrence rate in patients with primary resectable pancreatic ductal adenocarcinoma (PDA), we found that PDA TILs can readily be expanded in vitro and that the majority of resulting TIL cultures show reactivity against the autologous tumor. However, the fraction of tumor-reactive T cells is low. We investigated to which extent this was related to the in vitro expansion. Experimental Design: We compared the clonal composition of TIL preparations before and after in vitro expansion using T-cell receptor (TCR) deep sequencing. Our findings for PDA were benchmarked to experiments with melanoma TILs. Results: We found that the TIL TCR repertoire changes dramatically during in vitro expansion, leading to loss of tumor-dominant T-cell clones and overgrowth by newly emerging T-cell clones that are barely detectable in the tumor. These changes are primarily driven by differences in the intrinsic in vitro expansion capacity of T-cell clones. Single-cell experiments showed an association between poor proliferative capacity and expression of markers related to antigen experience and dysfunction. Furthermore, we found that spatial heterogeneity of the TIL repertoire resulted in TCR repertoires that are greatly divergent between TIL cultures derived from distant tumor samples of the same patient. Conclusions: Culture-induced changes in clonal composition are likely to affect tumor reactivity of TIL preparations. TCR deep sequencing provides important insights into the factors that govern the outcome of in vitro TIL expansion and thereby a path toward optimization of the production of TIL preparations with high therapeutic efficacy.

  • Five-Year Outcomes With Nivolumab in Patients With Wild-Type BRAF Advanced Melanoma.

    • The CheckMate 066 trial investigated nivolumab monotherapy as first-line treatment for patients with previously untreated BRAF wild-type advanced melanoma. Five-year results are presented herein.In this multicenter, double-blind, phase III study, 418 patients with previously untreated, unresectable, stage III/IV, wild-type BRAF melanoma were randomly assigned 1:1 to receive nivolumab 3 mg/kg every 2 weeks or dacarbazine 1,000 mg/m2 every 3 weeks. The primary end point was overall survival (OS), and secondary end points included progression-free survival (PFS), objective response rate (ORR), and safety.Patients were followed for a minimum of 60 months from the last patient randomly assigned (median follow-up, 32.0 months for nivolumab and 10.9 months for dacarbazine). Five-year OS rates were 39% with nivolumab and 17% with dacarbazine; PFS rates were 28% and 3%, respectively. Five-year OS was 38% in patients randomly assigned to dacarbazine who had subsequent therapy, including nivolumab (n = 37). ORR was 42% with nivolumab and 14% with dacarbazine; among patients alive at 5 years, ORR was 81% and 39%, respectively. Of 42 patients treated with nivolumab who had a complete response (20%), 88% (37 of 42) were alive as of the 5-year analysis. Among 75 nivolumab-treated patients alive and evaluable at the 5-year analysis, 83% had not received subsequent therapy; 23% were still on study treatment, and 60% were treatment free. Safety analyses were similar to the 3-year report.Results from this 5-year analysis confirm the significant benefit of nivolumab over dacarbazine for all end points and add to the growing body of evidence supporting long-term survival with nivolumab mono-therapy. Survival is strongly associated with achieving a durable response, which can be maintained after treatment discontinuation, even without subsequent systemic therapies.

  • Nivolumab in Previously Untreated Melanoma without BRAF Mutation

    • BACKGROUND Nivolumab was associated with higher rates of objective response than chemotherapy in a phase 3 study involving patients with ipilimumab-refractory metastatic melanoma. The use of nivolumab in previously untreated patients with advanced melanoma has not been tested in a phase 3 controlled study. METHODS We randomly assigned 418 previously untreated patients who had metastatic melanoma without a BRAF mutation to receive nivolumab (at a dose of 3 mg per kilogram of body weight every 2 weeks and dacarbazine-matched placebo every 3 weeks) or dacarbazine (at a dose of 1000 mg per square meter of body-surface area every 3 weeks and nivolumab-matched placebo every 2 weeks). The primary end point was overall survival. RESULTS At 1 year, the overall rate of survival was 72.9% (95% confidence interval [CI], 65.5 to 78.9) in the nivolumab group, as compared with 42.1% (95% CI, 33.0 to 50.9) in the dacarbazine group (hazard ratio for death, 0.42; 99.79% CI, 0.25 to 0.73; P < 0.001). The median progression-free survival was 5.1 months in the nivolumab group versus 2.2 months in the dacarbazine group (hazard ratio for death or progression of disease, 0.43; 95% CI, 0.34 to 0.56; P < 0.001). The objective response rate was 40.0% (95% CI, 33.3 to 47.0) in the nivolumab group versus 13.9% (95% CI, 9.5 to 19.4) in the dacarbazine group (odds ratio, 4.06; P < 0.001). The survival benefit with nivolumab versus dacarbazine was observed across prespecified subgroups, including subgroups defined by status regarding the programmed death ligand 1 (PD-L1). Common adverse events associated with nivolumab included fatigue, pruritus, and nausea. Drug-related adverse events of grade 3 or 4 occurred in 11.7% of the patients treated with nivolumab and 17.6% of those treated with dacarbazine. CONCLUSIONS Nivolumab was associated with significant improvements in overall survival and progression-free survival, as compared with dacarbazine, among previously untreated patients who had metastatic melanoma without a BRAF mutation.

  • Selective adsorption, bound states, and potential parameters for He, Ne, and Ar interacting with a Cu(110) surface

    • Using nozzle beams of He, Ne, and Ar, we have measured diffractive selective adsorption resonances from a Cu(110) surface kept at 20 K. Bound state energies of the atom-surface potentials have been determined from plots of the measured resonance energies versus incident angle and their fits to calculated kinematical dispersion relations. For 3He and 4He we have found a unique level assignment that is compatible with a single gas-surface potential curve with a well depth of 6.05 meV of the He–Cu(110) potential. This value is about 10% larger than the prediction of 5.55 meV from the current physisorption theory. The Ne and Ar data reveal a large number of closely spaced levels with level separations and estimated van der Waals coefficients that are compatible with available theoretical data.

    • This analysis provides long-term follow-up in patients with BRAF wild-type advanced melanoma receiving first-line therapy based on anti-programmed cell death 1 receptor inhibitors.To compare the 3-year survival with nivolumab vs that with dacarbazine in patients with previously untreated BRAF wild-type advanced melanoma.This follow-up of a randomized phase 3 trial analyzed 3-year overall survival data from the randomized, controlled, double-blind CheckMate 066 phase 3 clinical trial. For this ongoing, multicenter academic institution trial, patients were enrolled from January 2013 through February 2014. Eligible patients were 18 years or older with confirmed unresectable previously untreated stage III or IV melanoma and an Eastern Cooperative Oncology Group performance status of 0 or 1 but without a BRAF mutation.Patients were treated until progression or unacceptable toxic events with nivolumab (3 mg/kg every 2 weeks plus dacarbazine-matched placebo every 3 weeks) or dacarbazine (1000 mg/m2 every 3 weeks plus nivolumab-matched placebo every 2 weeks).Overall survival.At minimum follow-ups of 38.4 months among 210 participants in the nivolumab group (median age, 64 years [range, 18-86 years]; 57.6% male) and 38.5 months among 208 participants in the dacarbazine group (median age, 66 years [range, 25-87 years]; 60.1% male), 3-year overall survival rates were 51.2% (95% CI, 44.1%-57.9%) and 21.6% (95% CI, 16.1%-27.6%), respectively. The median overall survival was 37.5 months (95% CI, 25.5 months-not reached) in the nivolumab group and 11.2 months (95% CI, 9.6-13.0 months) in the dacarbazine group (hazard ratio, 0.46; 95% CI, 0.36-0.59; P<.001). Complete and partial responses, respectively, were reported for 19.0% (40 of 210) and 23.8% (50 of 210) of patients in the nivolumab group compared with 1.4% (3 of 208) and 13.0% (27 of 208) of patients in the dacarbazine group. Additional analyses were performed on outcomes with subsequent therapies. Treatment-related grade 3/4 adverse events occurred in 15.0% (31 of 206) of nivolumab-treated patients and in 17.6% (36 of 205) of dacarbazine-treated patients. There were no deaths due to study drug toxic effects.Nivolumab led to improved 3-year overall survival vs dacarbazine in patients with previously untreated BRAF wild-type advanced melanoma.ClinicalTrials.gov identifier: NCT01721772.

    • Budget effectiveness in multinational companies receives research interest because of globalisation. Research to date has used contingency-theory methods which have limitations. A systems-fit approach, which views organisations and systems holistically, is a complementary alternative. Compared with reductionistic contingency framework methods using regression analysis, it emphasises holistic methods related to effectiveness and deals with the inherent complexity of multinationals. This study examines the effectiveness of budget control systems in a Finnish multinational company using the systems approach. The tests examined the correlation of the fit of different profiles with effectiveness of the budget control system for both domestic and foreign subunits under conditions of high and low environmental uncertainty. No significant difference between domestic and foreign subunits is detected. Reliance on budget control is not inherently dysfunctional. The study demonstrates the viability of the systems-fit research approach.

  • New environmental metabarcodes for analysing soil DNA: potential for studying past and present ecosystems

    • Metabarcoding approaches use total and typically degraded DNA from environmental samples to analyse biotic assemblages and can potentially be carried out for any kinds of organisms in an ecosystem. These analyses rely on specific markers, here called metabarcodes, which should be optimized for taxonomic resolution, minimal bias in amplification of the target organism group and short sequence length. Using bioinformatic tools, we developed metabarcodes for several groups of organisms: fungi, bryophytes, enchytraeids, beetles and birds. The ability of these metabarcodes to amplify the target groups was systematically evaluated by (i) in silico PCRs using all standard sequences in the EMBL public database as templates, (ii) in vitro PCRs of DNA extracts from surface soil samples from a site in Varanger, northern Norway and (iii) in vitro PCRs of DNA extracts from permanently frozen sediment samples of late-Pleistocene age (similar to 16 00050 000 years bp) from two Siberian sites, Duvanny Yar and Main River. Comparison of the results from the in silico PCR with those obtained in vitro showed that the in silico approach offered a reliable estimate of the suitability of a marker. All target groups were detected in the environmental DNA, but we found large variation in the level of detection among the groups and between modern and ancient samples. Success rates for the Pleistocene samples were highest for fungal DNA, whereas bryophyte, beetle and bird sequences could also be retrieved, but to a much lesser degree. The metabarcoding approach has considerable potential for biodiversity screening of modern samples and also as a palaeoecological tool.

    • Over the last twenty years, accounting diversity between countries has been an important issue in international accounting research. A driving force has been the growth of multinational companies across borders. The development has created new challenges in consolidating financial statements across borders to a common group accounting report. The actual behaviour at the headquarter levels and in the subsidiaries abroad when consolidating vertically to a group report has not been empirically examined. The main research question in the study is if Swedish multinational enterprises make the accounting adjustments that would be necessary to comply with the Swedish accounting standards in their group reports. The vertical adjustments can take place either at the parent company (group level) or at subsidiary levels. The empirical investigations were conducted as three survey investigations of companies on the Stockholm Stock Exchange (1990, 1992 and 1999) and case studies in twelve Swedish groups and their subsidiaries in Denmark, the US, and Brazil (1991-1992). The objective of the empirical study was to investigate how Swedish multinational groups listed on the Stockholm Stock Exchange deal with accounting diversity among foreign subsidiaries in the consolidation processes.The study focused on accounting method choices in the adjustment process from local subsidiary reports to the group accounts as well as the issue in which currency the adjustments were made. The effects of currency translation methods can have a material impact on reported profits. Factors that were related to the frequency of adjustments were: the number of stock exchanges the Swedish groups were listed on, proportion of foreign activity in the group, inflation rate in the subsidiary countries, accounting diversity the group faced, main industry of the group, size of the group and income smoothing behaviour especially on subsidiary levels.An unexpected result was that some of the groups declared in their annual reports that they had followed actual Swedish accounting standards even if they made accounting method choices that did not comply with Swedish standards.

  • Diversity of the tardigrade communities in the Norwegian forests

    • Background: Tardigrades are common in most habitats, however few studies have focused on large faunistic survey, specifically on tardigrade diversity in forests. Up to now, only 61 species have been recorded in different types of forest in Norway with an additional 25 found in limnic environments in forests. Although little is known about the ecological preferences of many species, previous studies have found that tardigrade diversity and community composition are significantly affected by ecological variables. In this study we associate georeferenced tardigrade species records with forest type, substrate type and substrate composition in order to see if tardigrade diversity and species communities can be associated with ecological characteristics of Norwegian forests. Methods: In total 390 moss, lichen and litter samples were collected from 12 forests in central and southern Norway in the summers of 2017 and 2018 and later stored in paper envelopes. For the identification modern literature and keys forspecific genera and groups of species were used. For statistical analyses, moss and lichen substrate of each sample was classified according to the main species, life form, growth forms and habitat of substrate and associated with each tardigrade identification and sample metadata. Results: A total of 17 407 specimens were identified, encompassing in total 132 species (including some new species). Species richness increases with precipitation, but does not change with temperature or precipitation seasonality. The distribution of species richness between life forms and forest types showed considerable variation within and among the variables. Disregarding variables with low sample numbers, among life forms only acrocarpous moss samples appeared to deviate with respect to species richness, containing less species than substrates with other life forms. Conclusions: Tardigrades in Norwegian forest are extremely abundant, frequent and diverse. Moreover, it appears that that certain species and/or entirecommunities prefer specific microhabitats.

Although little is known on their ecological preferences, previous studies found diversity and community composition significantly affected by various variables. This study associated tardigrade species’ records with climatic variables, forest type, and substrate categories exploring tardigrade diversity and species communities to find associations with ecological characteristics of Norwegian forests. A total of 17 473 specimens were identified, encompassing 131 species (including putatively new species) from 305 samples of different substrates (leaf litter, bryophytes, and lichens). Bryophytes and lichens of samples were classified according to the main species, and growth form, and associated with tardigrade species and sample metadata. Tardigrade species’ richness was related to climatic variables and forest type, increased with precipitation, decreased with summer temperature, and not varied with precipitation seasonality. Although there was an unbalanced representation of substrate categories in the different forest types, some tendencies were detectable. Mixed oak and birch forests reached the highest species’ richness. Tardigrade community composition varied between substrate categories and, to a lesser degree, between forest types, but not with climatic variables. Our study highlights the importance of large-scale variables on tardigrade diversity, and substrate categories for tardigrade community composition.

    • Aim: We were able to show that targeted Next-Generation Sequencing is well suited to be applied in clinical routine diagnostics, substantiating the ongoing paradigm shift from low- to high-throughput genomics in medicine. By means of our atlas of the genetics of human DCM, we aspire to soon be able to apply our findings to the individual patient with cardiomyopathy in daily clinical practice. Numerous genes are known to cause dilated cardiomyopathy (DCM). However, until now technological limitations have hindered elucidation of the contribution of all clinically relevant disease genes to DCM phenotypes in larger cohorts. We now utilized next-generation sequencing to overcome these limitations and screened all DCM disease genes in a large cohort. Methods and results: In this multi-centre, multi-national study, we have enrolled 639 patients with sporadic or familial DCM. To all samples, we applied a standardized protocol for ultra-high coverage next-generation sequencing of 84 genes, leading to 99.1% coverage of the target region with at least 50-fold and a mean read depth of 2415. In this well characterized cohort, we find the highest number of known cardiomyopathy mutations in plakophilin-2, myosin-binding protein C-3, and desmoplakin. When we include yet unknown but predicted disease variants, we find titin, plakophilin-2, myosin-binding protein-C 3, desmoplakin, ryanodine receptor 2, desmocollin-2, desmoglein-2, and SCN5A variants among the most commonly mutated genes. The overlap between DCM, hypertrophic cardiomyopathy (HCM), and channelopathy causing mutations is considerably high. Of note, we find that >38% of patients have compound or combined mutations and 12.8% have three or even more mutations. When comparing patients recruited in the eight participating European countries we find remarkably little differences in mutation frequencies and affected genes. Conclusion: This is to our knowledge, the first study that comprehensively investigated the genetics of DCM in a large-scale cohort and across a broad gene panel of the known DCM genes. Our results underline the high analytical quality and feasibility of Next-Generation Sequencing in clinical genetic diagnostics and provide a sound database of the genetic causes of DCM.

    • This study examines the interaction between two dimensions of budgetary participation and environmental dynamism on perceptions of budget effectiveness in multinational companies. Unique features are dichotomies of: (i) domestic and foreign subunits; (ii) two dimensions of budget participation, communication and influence; and (iii) two facets of effectiveness perceptions, higher level involvement and outcomes of budget process. Results show that benefits of budgetary participation are limited and not consistent. Significant differences between foreign and domestic subunits are apparent. The primary perceived benefits are associated with communication. Budget influence is associated with positive perceptions of top-level involvement in foreign subunits but negative perceptions of outcomes of budget process in both domestic and foreign subunits.

    • In the framework of the Herschel/PRISMAS guaranteed time key program, the line of sight to the distant ultracompact H ii region W51e2 has been observed using several selected molecular species. Most of the detected absorption features are not associated with the background high-mass star-forming region and probe the diffuse matter along the line of sight. We present here the detection of an additional narrow absorption feature at ~70 km s-1 in the observed spectra of HDO, NH3 and C3. The 70 km s-1 feature is not uniquely identifiable with the dynamic components (the main cloud and the large-scale foreground filament) so-far identified toward this region. The narrow absorption feature is similar to the one found toward low-mass protostars, which is characteristic of the presence of a cold external envelope. The far-infrared spectroscopic data were combined with existing ground-based observations of 12CO, 13CO, CCH, CN, and C3H2 to characterize the 70 km s-1 component. Using a non-LTE analysis of multiple transitions of NH3 and CN, we estimated the density (n(H2) ~ (1-5) × 105 cm-3) and temperature (10-30 K) for this narrow feature. We used a gas-grain warm-up based chemical model with physical parameters derived from the NH3 data to explain the observed abundances of the different chemical species. We propose that the 70 km s-1 narrow feature arises in a dense and cold clump that probably undergoes collapse to form a low-mass protostar, formed on the trailing side of the high-velocity filament, which is thought to be interacting with the W51 main cloud. While the fortuitous coincidence of the dense clump along the line of sight with the continuum-bright W51e2 compact H ii region has contributed to its nondetection in the continuum images, this same attribute makes it an appropriate source for absorption studies and in particular for ice studies of star-forming regions.Based on data acquired with Herschel and IRAM observatories.     • Reporting of environmental information along with financial information has become an important research topic. Research to date has focused on the nature of the information reported by companies. This study extends prior research by examining the inclusion of environmental information by financial analysts in their research reports on companies in the chemical and in the oil and gas industries. Both companies and financial analysts are divided into subsets by geographic region, Europe and North America. Results show that only 35 per cent of financial analysts' reports have environmental information. Those reports that do have such information have more environmental information for North American companies than for European companies and analysts tend to report more information for companies in their regions.     • We have used the Herschel-HIFI instrument to observe both nuclear spin symmetries of amidogen (NH2) towards the high-mass star-forming regions W31C (G10.6-0.4), W49N (G43.2-0.1), W51 (G49.5-0.4) and G34.3+0.1. The aim is to investigate the ratio of nuclear spin types, the ortho-to-para ratio (OPR), of NH2. The excited NH2 transitions are used to construct radiative transfer models of the hot cores and surrounding envelopes in order to investigate the excitation and possible emission of the ground state rotational transitions of ortho-NH2 N_(K_a,K_c} J=1_(1,1) 3/2 - 0_(0,0) 1/2 and para-NH2 2_(1,2) 5/2 - 1_(0,1) 3/2$ used in the OPR calculations. Our best estimate of the average OPR in the envelopes lie above the high temperature limit of three for W49N, specifically 3.5 with formal errors of \pm0.1, but for W31C, W51, and G34.3+0.1 we find lower values of 2.5\pm0.1, 2.7\pm0.1, and 2.3\pm0.1, respectively. Such low values are strictly forbidden in thermodynamical equilibrium since the OPR is expected to increase above three at low temperatures. In the translucent interstellar gas towards W31C, where the excitation effects are low, we find similar values between 2.2\pm0.2 and 2.9\pm0.2. In contrast, we find an OPR of 3.4\pm0.1 in the dense and cold filament connected to W51, and also two lower limits of >4.2 and >5.0 in two other translucent gas components towards W31C and W49N. At low temperatures (T \lesssim 50 K) the OPR of H2 is 20-25 K, depending on time, and values above three at lower temperatures.

    • Introduction: One of the major research dimensions of the Identification and prevention of Dietary- and lifestyle-induced health EFfects In Children and infantS (IDEFICS) study involved the development, implementation and evaluation of a setting-based community-oriented intervention programme for primary prevention of childhood obesity. In this supplement of Obesity Reviews, a compilation of key results of the IDEFICS intervention is packaged in a series of complementary papers. Objective: This paper describes the overall design and methods of the IDEFICS intervention in order to facilitate a comprehensive reading of the supplement. In addition, some 'best practice' examples are described. Results: The IDEFICS intervention trial was conducted to assess whether the IDEFICS intervention prevented obesity in young children aged 2 to 9.9years. The study was a non-randomized, quasi-experimental trial with one intervention matched to one control region in each of eight participating countries. The intervention was designed following the intervention mapping framework, using a socio-ecological theoretical approach. The intervention was designed to address several key obesity-related behaviours in children, parents, schools and community actors; the primary outcome was the prevalence of overweight/obesity according to the IOTF criteria based on body mass index. The aim was to achieve a reduction of overweight/obesity prevalence in the intervention regions. The intervention was delivered in school and community settings over a 2-year period. Data were collected in the intervention and control cohort regions at baseline and 2 years later. Conclusion: This paper offers an introductory framework for a comprehensive reading of this supplement on IDEFICS intervention key results. © 2015 World Obesity.

  • Magnetic field sensing with the kinetic inductance of a high-Tc superconductor

    • We carry out an experimental feasibility study of a magnetic field sensor based on the kinetic inductance of the high critical temperature (high-T c ) superconductor yttrium barium copper oxide. We pattern thin superconducting films into radio-frequency resonators that feature a magnetic field pick-up loop. At 77 K and for film thicknesses down to 75 nm, we observe the persistence of screening currents that modulate the loop kinetic inductance. We report on a device with a magnetic field sensitivity of 4 pT/Hz, an instantaneous dynamic range of 11 μT, and operability in magnetic fields up to 28 μT. According to the experimental results the device concept appears attractive for sensing applications in ambient magnetic field environments.

    • The kynurenine pathway (KP) has been identified as a potential mediator linking acute illness to cognitive dysfunction by generating neuroactive metabolites in response to inflammation. Delirium (acute confusion) is a common complication of acute illness and is associated with increased risk of dementia and mortality. However, the molecular mechanism underlying delirium, particularly in relation to the KP, remain elusive.We undertook a multi-center observational study with 586 hospitalized patients (248 with delirium) and investigated associations between delirium and KP metabolites measured in cerebrospinal fluid (CSF) and serum by targeted metabolomics. We also explored associations between KP metabolites and markers of neuronal damage and one-year mortality.In delirium, we found concentrations of the neurotoxic metabolite quinolinic acid in CSF (CSF-QA, OR 2.26 [1.78, 2.87], p<0.001) to be increased, as well as increases in several other KP metabolites in serum and CSF. In addition, CSF-QA was associated with the neuronal damage marker neurofilament light chain (NfL, β 0.43, p<0.001) and was a strong predictor of one-year mortality (HR 4.35 [2.93, 6.45] for CSF-QA ≥ 100 nmol/L, p<0.001). The associations between CSF-QA and delirium, neuronal damage, and mortality remained highly significant following adjustment for confounders and multiple comparisons.Our data identified how systemic inflammation, neurotoxicity, and delirium are strongly linked via the KP, and should inform future delirium prevention and treatment clinical trials that target enzymes of the KP.Norwegian Health Association and the South-Eastern Norway Regional Health Authorities.
