    • Although model-driven engineering (MDE) is now an established approach for developing complex software systems, it has not been universally adopted by the software industry. In order to better understand the reasons for this, as well as to identify future opportunities for MDE, we carried out a week-long design thinking experiment with 15 MDE experts. Participants were facilitated to identify the biggest problems with current MDE technologies, to identify grand challenges for society in the near future, and to identify ways that MDE could help to address these challenges. The outcome is a reflection of the current strengths of MDE, an outlook of the most pressing challenges for society at large over the next three decades, and an analysis of key future MDE research opportunities.

  • Biomarkers of inflammation in workers exposed to compost and sewage dust

    • The association between exposure during handling of sewage and compost and the serum concentration of inflammatory biomarkers was studied. A total of 44 workers exposed to sewage dust, 47 workers exposed to compost dust and 38 referents from the administrative staff participated. Microbial aerosols were collected by personal inhalable samplers. The concentrations of bacterial cells, spores from fungi and bacteria (actinomycetes) and endotoxins were determined by fluorescence and scanning electron microscopy and the Limulus assay. Fibrinogen, D-dimer, ICAM-1, VCAM-1 and IL-6 were determined by ELISA and C-reactive protein (CRP) by HS-MicroCRP assay in blood samples collected post-shift. The exposure to dust ranged from 0.02 to 11 mg/m(3), endotoxins from 1 to 3160 EU/m(3) and bacteria from 0 to 209 x 10(6) cells/m(3). Fungal (0-41 x 10(6) spores/m(3)) and actinomycetes spores (0-590 x 10(6) actinomycetes spores/m(3)) were observed only at compost plants. The exposed workers had significantly higher fibrinogen (arithmetic mean 3.3 mg/ml) and CRP (geometric mean 1.5 mg/L) compared to the referents (2.8 and 1.0 mg/L, respectively). The serum concentration of CRP was negatively associated with forced expiratory volume in one second (FEV1) in % of predicted. Exposure to inhalable dust and bacteria was positively associated with the serum concentration of ICAM-1. This study suggests that exposure to bacteria and dust when handling sewage and compost may initiate an inflammation shown by an increase in serum concentration of ICAM-1. The higher concentrations of fibrinogen and CRP in exposed workers compared to the referents may reflect a low-grade systemic inflammation.

  • Catching up with Method and Process Practice: An Industry-Informed Baseline for Researchers

    • Software development methods are usually not applied by the book. Companies are under pressure to continuously deploy software products that meet market needs and stakeholders' requests. To implement efficient and effective development processes, companies utilize multiple frameworks, methods and practices, and combine these into hybrid methods. A common combination contains a rich management framework to organize and steer projects complemented with a number of smaller practices providing the development teams with tools to complete their tasks. In this paper, based on 732 data points collected through an international survey, we study the software development process use in practice. Our results show that 76.8% of the companies implement hybrid methods. Company size as well as the strategy in devising and evolving hybrid methods affect the suitability of the chosen process to reach company or project goals. Our findings show that companies that combine planned improvement programs with process evolution can increase their process' suitability by up to 5%.

  • Short-term side effects and attitudes towards second donation: A comparison of related and unrelated haematopoietic stem cell donors

    • The Nordic Register of Haematopoietic Stem Cell Donors (NRHSD) has registered related and unrelated donors from 10 transplant centres in Sweden, Norway, Finland and Denmark since 1998. We present a prospective, observational study of 1,957 donors, focusing mainly on the differences between related and unrelated donors. Related donors are reported to have more comorbidities, but similar side effects compared with unrelated donors. Side effects after BM or PBSC donation are generally of short duration and in this study no deaths, myocardial infarctions, splenic ruptures, or thromboembolic events are reported. Interestingly, related donors express more hesitancy towards donating again when asked 1 month after donation.

  • The CD34+ Cell Dose Matters in Hematopoietic Stem Cell Transplantation with Peripheral Blood Stem Cells from Sibling Donors

    • The effect of CD34+ cell dose in allogeneic hematopoietic stem cell transplantation (HSCT) on overall survival (OS) and incidence of acute and chronic graft-versus-host disease (GvHD) has not been established and few studies have been performed. Our single center analysis included 189 patients with hematological malignancies who received peripheral blood stem cell (PBSC) grafts from sibling donors. Myeloablative conditioning was used in 88 cases and 101 received reduced intensity conditioning. The median CD34+ cell dose was 5.6 × 106/kg (0.6–17.0). In the multivariate analysis, a CD34 cell dose of 6–7 × 106/kg was associated with better OS and lower transplant-related mortality (TRM), while a dose of <5 × 106/kg led to increased relapse and reduced chronic GVHD (cGVHD). A high CD34 cell-dose (>6.5 × 106/kg) correlated with less acute GVHD (aGVHD) II–IV. We conclude that the CD34 cell dose has an impact on the outcome of HSCT from sibling donor PBSCs.

  • Pneumoproteins and markers of inflammation and platelet activation in the blood of grain dust exposed workers

    • Purpose: To investigate if blood biomarkers could indicate early signs of lung damage or cardiovascular risk due to exposure to grain dust. Materials and methods: Pneumoproteins and markers of inflammation and platelet activation were analysed in blood samples of 102 grain elevator and compound feed mill workers. Differences between exposed (n = 67) and controls (n = 35), and associations with exposure measurements and respiratory health were investigated by multiple linear regression analyses. Results: Concentrations of CC-16 and IL-6 were higher in exposed workers compared with controls (p < 0.001 for both), whereas fibrinogen was lower (p = 0.005). Concentrations of CRP, TNF-alpha, sCD40L and sP-selectin were similar in both groups. Serum CC-16 was significantly higher in workers with farm childhood, regardless of exposure. The impact of farm childhood on CC-16 interacted with smoking. None of the biomarkers were associated with exposure measurements or any of the tested respiratory health parameters. Conclusion: Dust exposure induced inflammatory and anti-inflammatory reactions, but did not induce systemic inflammation and had no effect on platelet activation. No cause-effect relationship could be established in spite of relatively high exposure levels, particularly to endotoxin. Whether increased serum CC-16 is an early sign of lung damage or a reversible defense reaction remains unclear.
