    • För att källfördela läckage av kväve och fosfor till Östersjön och Västerhavet behövs en stor mängd geografisk information. Kartunderlaget är därför av mycket stor betydelse. Rapporten syftar till att ge en detaljerad fördjupning i hur de olika kartbaserade underlagsdata till PLC6 togs fram. Särskilt för markanvändningen fördjupas även analysen av resultaten. Hydrologiska vattenförekomstområden har tagits fram baserat på vattenmyndigheternas klassning av vattenförekomster och SVAR-databasen för delavrinningsområden. De ca 23 100 vattenförekomstområdena har grupperats till ca 1100 redovisningsområden, samt vidare till huvudavrinningsområden och havsbassängernas avrinningsområden.Regionsindelning för olika beräkning av läckage från jordbruksmark, skog och öppen mark i olika delar av Sverige har tagits fram baserat på PLC5- indelningen och med ett tillägg av två nya regioner för läckaget från skog och öppen mark. En ny markanvändningskarta har tagits fram, baserad på vägkartan, Jordbruksverkets jordbruksblock och stödsökta grödor, SCB:s tätortskarta, Skogsstyrelsens data på faktiskt avverkad areal, och Corine-data för Norge och Finland. Inom tätorter har SMD-kartan använts för en mer högupplöst klassning till beräkningen av dagvattenbelastning. Jordbruksmarkens jordartsfördelning, fosforhalt i marken och lutning har beräknats baserats på nya kartdata.Beräkningar av varje vattenförekomstområdes centrumkoordinater och medelhöjd har genomförts.Läckageregionernas målavrinning och kvävedeposition har beräknats baserat på vattenförekomstområdenas avrinning respektive rasterdata för depositionen.

  • Autonomous last-mile delivery robots: a literature review

    • This literature review investigates how self-driving autonomous delivery robots (ADRs) impact last-mile deliveries, add value to the logistics and transport industry, and contribute to creating competitive business models. Autonomous vehicles are still a developing technology and ADRs could possibly be one of the solutions to the last-mile problem, in particular in cities and for urban freight with an increasing number of parcels to deliver. Last-mile delivery is also changing as e-commerce and more demanding customers emerge. Such development, however, faces challenges regarding infrastructure, externalities such as CO2 emissions, and shorter delivery-time requirements. This review, focused on ADRs, reveals four major themes (operations, infrastructure, regulations, and acceptance) through which we explain the barriers and benefits of using ADRs for last-mile deliveries. The review shows that the operations of ADRs can impact last-mile deliveries by lowering costs, optimising the use of time, and reducing externalities. The review also shows that the foundation of last-mile infrastructure would have to change if ADRs are to be used to a greater extent. Regulations for ADRs are still not yet in place, which makes the market somewhat confused. The acceptance of ADRs in society is another challenge because the innovation of ADRs is still new and unfamiliar. Altogether, the use of ADRs for last-mile deliveries shows great potential, based on the promising results of the articles reviewed. However, most studies on ADRs have been theoretical in nature, such as models, which highlights the need for real-world case studies and implementations.

  • The genome of Haemoproteus tartakovskyi and its relationship to human malaria parasites

    • The phylogenetic relationships among hemosporidian parasites, including the origin of Plasmodium falciparum, the most virulent malaria parasite of humans, have been heavily debated for decades. Studies based on multiple-gene sequences have helped settle many of these controversial phylogenetic issues. However, denser taxon sampling and genome-wide analyses are needed to confidently resolve the evolutionay relationships among hemosporidian parasites. Genome sequences of several Plasmodium parasites are available but only for species infecting primates and rodents. To root the phylogenetic tree of Plasmodium, genomic data from related parasites of birds or reptiles are required. Here, we use a novel approach to isolate parasite DNA from microgametes and describe the first genome of a bird parasite in the sister genus to Plasmodium, Haemoproteus tartakovskyi Similar to Plasmodium parasites, H. tartakovskyi has a small genome (23.2 Mb, 5,990 genes) and a GC content (25.4%) closer to P. falciparum (19.3%) than to Plasmodium vivax (42.3%). Combined with novel transcriptome sequences of the bird parasite Plasmodium ashfordi, our phylogenomic analyses of 1,302 orthologous genes demonstrate that mammalian-infecting malaria parasites are monophyletic, thus rejecting the repeatedly proposed hypothesis that the ancestor of Laverania parasites originated from a secondary host shift from birds to humans. Genes and genomic features previously found to be shared between P. falciparum and bird malaria parasites, but absent in other mammal malaria parasites, are therefore signatures of maintained ancestral states. We foresee that the genome of H. tartakovskyi will open new directions for comparative evolutionary analyses of malarial adaptive traits.

  • Decreased startle modulation during anticipation in the postpartum period in comparison to late pregnancy

    • Knowledge about healthy women's psychophysiological adaptations during the large neuroendocrine changes of pregnancy and childbirth is essential in order to understand why these events have the potential to disrupt mental health in vulnerable individuals. This study aimed to compare startle response modulation, an objective psychophysiological measure demonstrated to be influenced by anxiety and depression, longitudinally across late pregnancy and the postpartum period. The acoustic startle response modulation was assessed during anticipation of affective images and during image viewing in 31 healthy women during gestational weeks 36-39 and again at 4 to 6 weeks postpartum. No startle modulation by affective images was observed at either time point. Significant modulation during anticipation stimuli was found at pregnancy assessment but was reduced in the postpartum period. The women rated the unpleasant images more negative and more arousing and the pleasant images more positive at the postpartum assessment. Self-reported anxiety and depressive symptoms did not change between assessments. The observed postpartum decrease in modulation of startle by anticipation suggests a relatively deactivated defense system in the postpartum period.

  • Exploring in-shoe plantar pressure differences at the metatarsal pad position for patients with diabetes and toe deformities: A pilot study

    • Patients with diabetes are often prescribed insoles with metatarsal pads intended to prevent and offload forefoot ulcers. However, to our knowledge, the plantar pressures specifically at the metatarsal pad had not previously been researched. The aim of this study was therefore to explore plantar pressures at the metatarsal pad position by comparing three toe deformity groups, claw toe (CT)/hammer toe (HT) and hallux valgus (HV), with a control group (NONE). In-shoe plantar pressures were measured using F-ScanTM and a polygon region of interest at the metatarsal pad position was created. Mean peak pressure (MPP), pressure-time integral (PTI) and percentage foot-insole contact area at the metatarsal pad position (%CA) were analysed from 24 patients (48 feet) in the metatarsal pad area. No significant difference was found between the groups regarding MPP, PTI or contact area. For MPP, the highest value was found in CT/HT (140kPa ± 61) and the lowest in HV (112kPa ± 33). In PTI, the highest value was for CT/HT (2.0 Ns/cm2 ± 1.0) and the lowest for HV (1.3 Ns/cm2 ± 0.4). The %CA was highest in CT/HT/HV (95%) and lowest in NONE (70%). A new quantitative approach was presented, assessing the metatarsal pad pressure effect on its corresponding plantar area of the foot. Further studies are needed to identify the conditions in which a metatarsal pad affects different foot statuses with regard to plantar pressure.

Primate malaria parasites can synthesize vitamin B1 (thiamine) de novo but rodent malaria and other genome-sequenced apicomplexans cannot, as the three essential genes responsible for this pathway are absent in their genomes. The unique presence of functional thiamine synthesis genes in primate malaria parasites and their sequence similarities to bacterial orthologues, have led to speculations that this pathway was horizontally acquired from bacteria. Here we show that the genes essential for the de novo synthesis of thiamine are found also in avian Plasmodium species. Importantly, they are also present in species phylogenetically basal to all mammalian and avian Plasmodium parasites, i.e. Haemoproteus. Furthermore, we found that these genes are expressed during the blood stage of the avian malaria infection, indicating that this metabolic pathway is actively transcribed. We conclude that the ability to synthesize thiamine is widespread among haemosporidians, with a recent loss in the rodent malaria species.

    • IntroductionSickness insurance systems and their processes have been studied in terms of transparency, comprehensibility and fairness, highlighting the importance of just procedures that make sense to clients. Related research demonstrates differences between groups of clients, pointing towards a social gradient. The concept of social insurance literacy and the Social Insurance Literacy Questionnaire (SILQ) was recently developed and serves as a measure for client's ability to obtain, understand and act on information in a sickness insurance system, relating to the comprehensibility of the information that the system provides.ObjectiveThe purpose of this study was to investigate social insurance literacy among clients on sick leave and its associations with perceived justice, being granted sickness benefits and background factors.MethodsThis was a questionnaire study with clients on sick leave in Sweden. In the selection process 3993 clients were invited, of which 1173 recently had their sickness benefits withdrawn. Those who answered the SILQ (n = 1152) also answered a perceived justice measure and accepted sharing register data from the Swedish Social Insurance Agency. Data were analyzed through regression analysis.ResultsThe findings demonstrate that clients' perceptions of system comprehensibility and the status of their sick leave case was significantly associated with perceived justice, and being granted sickness benefits, while their individual abilities to obtain, understand, and act on information had lesser influence.ConclusionsThe system's ability to provide understandable information seems more important than clients' abilities to comprehend it. From a client perspective, a just system seems to be related to their experiences of the sick leave process (i,e., whether they had an ongoing or closed case) rather than their skills to obtain the correct information.

  • Early-life gut dysbiosis linked to juvenile mortality in ostriches

    • Background: Imbalances in the gut microbial community (dysbiosis) of vertebrates have been associated with several gastrointestinal and autoimmune diseases. However, it is unclear which taxa are associated with gut dysbiosis, and if particular gut regions or specific time periods during ontogeny are more susceptible. We also know very little of this process in non-model organisms, despite an increasing realization of the general importance of gut microbiota for health. Methods: Here, we examine the changes that occur in the microbiome during dysbiosis in different parts of the gastrointestinal tract in a long-lived bird with high juvenile mortality, the ostrich (Struthio camelus). We evaluated the 16S rRNA gene composition of the ileum, cecum, and colon of 68 individuals that died of suspected enterocolitis during the first 3 months of life (diseased individuals), and of 50 healthy individuals that were euthanized as age-matched controls. We combined these data with longitudinal environmental and fecal sampling to identify potential sources of pathogenic bacteria and to unravel at which stage of development dysbiosis-associated bacteria emerge. Results: Diseased individuals had drastically lower microbial alpha diversity and differed substantially in their microbial beta diversity from control individuals in all three regions of the gastrointestinal tract. The clear relationship between low diversity and disease was consistent across all ages in the ileum, but decreased with age in the cecum and colon. Several taxa were associated with mortality (Enterobacteriaceae, Peptostreptococcaceae, Porphyromonadaceae, Clostridium), while others were associated with health (Lachnospiraceae, Ruminococcaceae, Erysipelotrichaceae, Turicibacter, Roseburia). Environmental samples showed no evidence of dysbiosis-associated bacteria being present in either the food, water, or soil substrate. Instead, the repeated fecal sampling showed that pathobionts were already present shortly after hatching and proliferated in individuals with low microbial diversity, resulting in high mortality several weeks later. Conclusions: Identifying the origins of pathobionts in neonates and the factors that subsequently influence the establishment of diverse gut microbiota may be key to understanding dysbiosis and host development.   • Host transcriptional responses to high-and low-virulent avian malaria parasites

    • The transcriptional response of hosts to genetically similar pathogens can vary substantially, with important implications for disease severity and host fitness. A low pathogen load can theoretically elicit both high and low host responses, as the outcome depends on both the effectiveness of the host at suppressing the pathogen and the ability of the pathogen to evade the immune system. Here, we investigate the transcriptional response of Eurasian siskins (Spinus spinus) to two closely related lineages of the malaria parasite Plasmodium relictum. Birds were infected with either the high-virulent lineage P. relictum SGS1, the low-virulent sister lineage P. relictum GRW4, or sham-injected (controls). Blood samples for RNA sequencing were collected at four time points during the course of infection, totaling 76 transcriptomes from 19 birds. Hosts infected with SGS1 experienced up to 87% parasitemia and major transcriptome shifts throughout the infection, and multiple genes showed strong correlation with parasitemia. In contrast, GRW4-infected hosts displayed low parasitemia (maximum 0.7%) with a minor transcriptional response. We furthermore demonstrate that the baseline gene expression levels of hosts prior to infection were irrelevant as immunocompetence markers, as they could not predict future pathogen load. This study shows that the magnitude of the host transcriptional response can differ markedly from related parasites with different virulence, and it enables a better understanding of the molecular interactions taking place between hosts and parasites.

  • Major shifts in gut microbiota during development and its relationship to growth in ostriches

    • The development of gut microbiota during ontogeny is emerging as an important process influencing physiology, immunity and fitness in vertebrates. However, knowledge of how bacteria colonize the juvenile gut, how this is influenced by changes in the diversity of gut bacteria and to what extent this influences host fitness, particularly in nonmodel organisms, is lacking. Here we used 16S rRNA gene sequencing to describe the successional development of the faecal microbiome in ostriches (Struthio camelus, n = 66, repeatedly sampled) over the first 3 months of life and its relationship to growth. We found a gradual increase in microbial diversity with age that involved multiple colonization and extinction events and a major taxonomic shift in bacteria that coincided with the cessation of yolk absorption. Comparisons with the microbiota of adults (n = 5) revealed that the chicks became more similar in their microbial diversity and composition to adults as they aged. There was a five-fold difference in juvenile growth during development, and growth during the first week of age was strongly positively correlated with the abundance of the genus Bacteroides and negatively correlated with Akkermansia. After the first week, the abundances of six phylogenetically diverse families (Peptococcaceae, S24-7, Verrucomicrobiae, Anaeroplasmataceae, Streptococcaceae, Methanobacteriaceae) were associated with subsequent reductions in chick growth in an age-specific and transient manner. These results have broad implications for our understanding of the development of gut microbiota and its associations with animal growth.

  • The Avian Transcriptome Response to Malaria Infection.

    • Malaria parasites are highly virulent pathogens which infect a wide range of vertebrates. Despite their importance, the way different hosts control and suppress malaria infections remains poorly understood. With recent developments in next generation sequencing techniques, however, it is now possible to quantify the response of the entire transcriptome to infections. We experimentally infected Eurasian siskins (Spinus spinus) with avian malaria parasites (Plasmodium ashfordi), and used high-throughput RNA-sequencing to measure the avian transcriptome in blood collected before infection (day 0), during peak parasitemia (day 21 post infection), and when parasitemia was decreasing (day 31). We found considerable differences in the transcriptomes of infected and uninfected individuals, with a large number of genes differentially expressed during both peak and decreasing parasitemia stages. These genes were overrepresented among functions involved in the immune system, stress response, cell death regulation, metabolism, and telomerase activity. Comparative analyses of the differentially expressed genes in our study to those found in other hosts of malaria (human and mouse), revealed a set of genes that are potentially involved in highly conserved evolutionary responses to malaria infection. By using RNA-sequencing we gained a more complete view of the host response, and were able to pinpoint not only well documented host genes, but also unannotated genes with clear significance during infection, such as microRNAs. This study shows how the avian blood transcriptome shifts in response to malaria infection, and we believe it will facilitate further research into the diversity of molecular mechanisms that hosts utilize to fight malaria infections.

    • Malaria parasites (Plasmodium spp.) include some of the world's most widespread and virulent pathogens. Our knowledge of the molecular mechanisms these parasites use to invade and exploit hosts other than mice and primates is, however, extremely limited. It is therefore imperative to characterize transcriptome-wide gene expression from non-model malaria parasites and how this varies across host individuals. Here, we used high-throughput Illumina RNA-sequencing on blood from wild-caught Eurasian siskins experimentally infected with a clonal strain of the avian malaria parasite Plasmodium ashfordi (lineage GRW2). By using a multi-step approach to filter out host transcripts, we successfully assembled the blood-stage transcriptome of P. ashfordi. A total of 11 954 expressed transcripts were identified, and 7 860 were annotated with protein information. We quantified gene expression levels of all parasite transcripts across three hosts during two infection stages – peak and decreasing parasitemia. Interestingly, parasites from the same host displayed remarkably similar expression profiles during different infection stages, but showed large differences across hosts, indicating that P. ashfordi may adjust its gene expression to specific host individuals. We further show that the majority of transcripts are most similar to the human parasite Plasmodium falciparum, and a large number of red blood cell invasion genes were discovered, suggesting evolutionary conserved invasion strategies between mammalian and avian Plasmodium. The transcriptome of P. ashfordi and its host-specific gene expression advances our understanding of Plasmodium plasticity and is a valuable resource as it allows for further studies analysing gene evolution and comparisons of parasite gene expression.
