  • Attitudes towards Sustainable Development. Priorities, Responsibility, Empowerment

    • The UN conference in Rio on Environment and Development in 1992 was the starting point for the local Agenda 21 work and several Swedish municipalities began their local Agenda 21 processes in 1993. This thesis aims at investigating how people perceive sustainable development in four Swedish municipalities. What is important and who is responsible? Individual influence and strategies for implementation, feelings of empowerment as well as the connection between personality and environmental concern, were analysed. Based on the Agenda 21 document, 20 statements including social, environmental and participation-/implementation issues, were developed. 160 persons (politicians, officials, environmental representatives and the public) were interviewed in four municipalities in the south east of Sweden. The respondents were asked to sort the statements according to the importance to family, municipality and world, and to the responsibility of the individual, local politicians and governments, EU and UN. The results showed that sustainable development issues were experienced as more important to the world than to the municipality or to the family, and that people felt worried and anxious when they were facing global problems. Most important were clean air, protecting fresh water, and health. What was important to the family was also perceived as important to the world however the connection to issues important in the municipalities was much less pronounced. The responsibility for sustainable development was mostly allocated to governments and global organizations, whereas the individual responsibility was perceived as low. Although the municipalities used information, education and local projects in order to increase the awareness, the public felt less individual responsibility and less influence than the other groups. Environmental concern was investigated among childcare personnel, who perceived their influence on environmental issues to be small. Still they felt that their actions would be meaningful if only they were provided with a scope of action and the possibility to influence. To increase the feelings of empowerment and improve the understanding of the global situation, more international co-operation projects on the local level are suggested. It is important to make local problems visible and explain the connection to global issues. If people were to perceive more control the prerequisites for treating problems in a constructively way might increase considerably.

  • Environmental preferences among steel stakeholders

    • Emissions of carbon dioxide, dioxins, nitrogen oxides and particulate matter as well as use of non-renewable resources and energy are some important sustainability challenges for the Swedish steel industry. Much effort has been made, mainly by technical solutions, which to a high degree have decreased the emissions during the last 30 years.Technical solutions however will not be sufficient to reach sustainable development, stakeholder involvement is also necessary. Stakeholder theory states that stake­holder involvement must include a dialog between the stakeholders involved and the operation. The first step in this process is to identify which key issues the stakeholders find most important and then the organisation needs to start interact with its stakeholders. This thesis deals with such issues.Stakeholder preferences for environmental issues were assessed with conjoint analysis, Q-methodology and focus group discussions. The theory of planned behaviour was used to assess how attitudes were connected to background factors and a potential pro-environmental behaviour.Five studies have been carried out in the framework of this thesis. The studies include: a literature review, method evaluation, evaluation of environ­mental objectives in stakeholder groups, screening of relevant factors, evaluation of steel environmental characteristics, identification of barriers to the introduction of new materials and the im­pact of worry and risk perception on strategic environmental decisions.It can be concluded that the methods applied in the studies work well in eliciting preferences. It has been possible to show how different stakeholder groups as well as individuals prioritise environmental objectives and sustaina­bility issues. Since individuals within a stakeholder group vary considerably in preferences, the results from this thesis show the importance of illustrating results on an individual level instead of the traditional group level. Also, a method has been tested where the results were brought back to the respondents in order to stimulate discussions between different stakeholder groups.

En viktig pusselbit för att nå dit är att veta hur individer och grupper ställer sig till olika strategiska miljöfrågor. Sådan kunskap kan användas för att anpassa informationskampanjer, skräddarsy utbildningspaket eller undanröja faktorer som riskerar att omintetgöra miljöarbetet.Metoderna som beskrivs i den här handboken kan användas inom många fler områden än miljö, t.ex. inom marknadsföring eller för att identifiera kunskaps- eller informationsluckor bland den egna personalen eller för att undersöka attityder och synsätt hos olika intressentgrupper.Handboken är framtagen inom forskningsprogrammet Stålkretsloppet och utgiven av Linnéuniversitetet i Kalmar med stöd från Jernkontoret och Mistra.

  • Why Aren't Advanced High–Strength Steels More Widely Used? : Stakeholder Preferences and Perceived Barriers to New Materials

    • Advanced high-strength steels may reduce the use of nonrenewable resources and energy given that the amount of material needed is smaller, compared to traditional steel grades. Still, advanced steel grades are not utilized to the extent that could be expected. This study examines stakeholders’ preferences of steel characteristics and perceived barriers to the introduction of new materials. Focus group interviews were used to identify steel characteristics and perceived barriers. Stakeholder preferences of steel characteristics were evaluated through a conjoint analysis; the results showed that low weight was given the highest priority, followed by high impact strength and low price. Low chromium content was the steel characteristic of least interest. Perceived barriers to the introduction of high-strength steel were categorized as technical barriers, knowledge barriers, scrap management barriers, suitability barriers, and cost barriers.

  • The Challenge of Transdisciplinary Research : A Case Study of Learning by Evaluation for Sustainable Transport Infrastructures

    • While transdisciplinary (TD) research is desired in order to solve real world sustainability issues, this may be challenging for both academic and non-academic participants. Supporting learning through evaluation, we analyzed a project aiming at sustainable transport infrastructures. After developing a TD research framework as a benchmark, two external independent evaluators interviewed all project researchers, representatives for end-users, and donors. The evaluators compared results with the framework, and evaluators and participants critically reflected on the results together. There were three inconsistencies relative to the framework: (1) limited understanding of TD research among project management, end-users, and most of the researchers; (2) no structured learning process among end-users; instead, they expressed very diverse opinions about what they expected from the project; (3) project leaders had limited understanding of the special challenges of TD research, did not fully understand the status of the project's social system, and thus did not act as facilitators of the required collaborative learning process. Non-academic participants saw themselves as customers and not as partners in the knowledge production process. We conclude that TD problem-solving research requires much time and needs facilitation and training. A preparatory phase with a lower level of funding would be helpful in preparing for TD processes.

  • The Effects of Wind Power on Human Interests : A Synthesis

    • PrefaceThere is a great need for knowledge concerning the impacts of wind power on humans, landscapes, the marine environment, birds, bats and other mammals.Previous studies of these environmental impacts have lacked an overall view of the effects. This has led to deficiencies in the processes surrounding the establishment of new wind farms. Vindval is a knowledge programme undertaken as a collaboration between the Swedish Energy Agency and the Swedish Environmental Protection Agency. Its aim is to gather and communicate scientific knowledge about the impacts of wind power on people and the natural environment. The programme continues until 2013.Vindval comprises some 30 individual research projects, together with four synthesis projects. Syntheses are prepared by experts, who compile and assess overall research results and experience regarding the effects of wind power in four different areas – humans, birds/bats, marine life and terrestrial mammals.The results of this research and synthesis work will provide a basis for environmental impact assessments and for the planning and permitting processes associated with wind power installations. Vindval requires high standards in the review and approval of research proposals, in order to ensure high-quality reports.     • More and more time is spent on recreational activities, but few risk assessments focus specifically on these situations and exposure factor data are often scarce. To assess exposure to contaminants at a public bathing place in an urban environment, we have compiled literature data, conducted observation studies, and analyzed water and sediment samples. The levels of anthropogenic contaminants are high in urban environments and traffic frequently plays an important role. In this study, to characterize variability and uncertainty, the deterministic exposure calculations for metal pollutants were supplemented by a probability bounds analysis for the polycyclic aromatic hydrocarbons (PAH). The results from these calculations show that oral intake is the major exposure route for metals, while skin absorption, with present assumptions, is more important for the PAH. The presently measured levels of contaminants, at this public bathing place, cannot be anticipated to cause any significant adverse influence on public health.

    • The European Landscape Convention indicates that assessment of different dimensions that exist in landscapes should be taken into account in planning. In this study, we first investigated ecological, cultural and social values as perceived by the local people in a highway-planning process in Sweden. Next, we explored which factors influenced the local people's participation in the road-planning process. We used questionnaires, planning documents and the Theory of Planned Behavior to investigate the relations between different factors and local people's participation in the planning process. The results showed that people presumed the ecological values in the landscape to be adversely affected by the new road, while the social values would remain the same. Landowners had heard of the participatory-process, but few participated. Those who lived within 300m of the road were more active in the planning process than people living further away. The findings suggest that people living within a few hundred metres of the road should be treated as key stakeholders in the planning process. The involvement of other stakeholders, and when in the public participation process stakeholders should be involved, is also discussed.

    • The aims were to identify sustainable development priorities and perceived responsibility of sustainable development issues. Differences in opinions between politicians, officials, environmental representatives and the public in four Swedish municipalities were analysed. Sustainable development issues were perceived as more important on the global level than on the municipal and family levels. Among the most important issues on both the global and family levels were clean air, fresh water, and health, whereas decision-making, co-operation, and participation were considered most important on the municipal level. The highest responsibility for sustainable development issues was placed on governments and global organizations and the lowest on the individual. The public felt less individual responsibility and less possibility to influence than did the other groups. The correlation between importance and responsible on municipal and world levels was high but low on the individual level. The responsibility for the most important issue on the family level (clean air) was placed on governments and global organizations. An attitude change is needed to increase responsibility for issues ranked low on individual responsibility. To further raise the awareness of global sustainable development issues, an increased concentration on locally based international co-operation projects is suggested.

  • Social-Psychological Factors in Public Support for Local Biodiversity Conservation

    • This case study investigates the role of social-psychological variables in the generalpublic’s intention to support conservation of five local biotopes. A questionnairesurvey of 245 people was carried out in Kristianstad Municipality, Sweden. Inhierarchical regression analyses, public intention to support conservation was partlypredicted by attitude toward biodiversity conservation and by personal norm. Therelationship between these variables and intention differed depending on the ecologicalcontent of the biotope. It is concluded that when communicating the importanceof conserving biotopes to the public, the message should be tailored according to theecological content of the biotope as well as with regard to public attitudes andpersonal norms.

    • Fusion genes are potent driver mutations in cancer. In this study, we delineate the fusion gene landscape in a consecutive series of 195 paediatric B-cell precursor acute lymphoblastic leukaemia (BCP ALL). Using RNA sequencing, we find in-frame fusion genes in 127 (65%) cases, including 27 novel fusions. We describe a subtype characterized by recurrent IGH-DUX4 or ERG-DUX4 fusions, representing 4% of cases, leading to overexpression of DUX4 and frequently co-occurring with intragenic ERG deletions. Furthermore, we identify a subtype characterized by an ETV6-RUNX1-like gene-expression profile and coexisting ETV6 and IKZF1 alterations. Thus, this study provides a detailed overview of fusion genes in paediatric BCP ALL and adds new pathogenetic insights, which may improve risk stratification and provide therapeutic options for this disease.

  • Environmental Effects of Agricultural Expansion in the Upper Amazon : A study of river basin geochemistry and hydrochemistry, and farmers' perceptions

    • In this thesis natural science is combined with environmental psychology in order to determine how deforestation and subsequent agricultural expansion in the Peruvian highland jungle has affected the natural environment and rural livelihoods. This region is part of one of the most biodiverse areas on Earth and is also exposed to high pressure from deforestation that threatens the ecosystems as well as the well-being of local populations. The problem stretches beyond the upper Amazon since the region constitutes headwaters to theAmazon Riverand is part of the most important forest ecosystem of the world.This study evaluates the relative controls of human induced land-cover change and natural factors on the chemical status of soils, stream waters, and sediments, mainly through a spatial sampling design. The field work was located to two adjacent river basins underlain by sedimentary rocks. Streams of 48 independent sub-basins, the two main rivers, 80 upland soil sites (weakly developed soils on sandstone and siltstone) and four vertical profiles of floodplain sediments were sampled and analysed for major and trace elements, including nutrients and potentially toxic metals. Further, perceptions of environmental changes were investigated through a combination of quantitative and qualitative interview data collected from 51 smallholder farmers.Soils of primary forests were found to be chemically similar to those of regenerated forests and agricultural land-covers (pastures and coffee plantations), and differences in chemical concentrations between streams draining areas to varying degrees covered by forest were assigned to natural variability. In addition, the chemical composition of alluvial deposits was similar in the two drainage basins despite a substantial difference in exploitation degree (30 % versus 70 % cleared from forest). Thus, no evidence was found of long-term changes in the geochemistry of the Subandean river basins as a result of the conversion of primary forest to agricultural land-uses. The farmers, however, perceived an overall increase in environmental degradation as well as a change towards drier and warmer climatic conditions. The climate change was reported to be the main factor responsible for a negative trend in life quality (rural livelihoods). The results may be used in the work of identifying priorities and key factors necessary for environmental and socioeconomic sustainability in the upper Amazon.

    • Amazon ecosystem degradation profoundly impacts life supporting processes of global importance such as climate regulation, as well as localconditions for livelihoods. In Peru’s highland jungle, an expanding deforestationfront of forest conversion to agriculture has vastly transformed the landscape.Small-scale farming, the main driver of forest degradation, and consequently household natural resource management affect ecosystem functionality.To investigate farmers’ attitudes and priorities to services provided by the ecosystems (ES) we interviewed 51 farmers, both local and colonists. They strongly agreed that over the last three decades, local conditions for livelihoods have deteriorated following forest degradation and climate change. The latterwas reported the primary contributor to an impaired life quality and their greatest concern. Overall, local farmers perceived greater than did colonists who were also more positive towards intensive agricultureand forestry. This should be considered in environmental conservation efforts in the upper Amazon.

Here, we propose connectivity as a conceptual tool to include different dimensions into landscape and spatial planning. We present a short review of the connectivity concept in relation to ecological, social and cultural dimensions and illustrate it by examples from a real landscape planning case.

  • Clonal competition within complex evolutionary hierarchies shapes AML over time

    • Clonal heterogeneity and evolution has major implications for disease progression and relapse in acute myeloid leukemia (AML). To model clonal dynamics in vivo, we serially transplanted 23 AML cases to immunodeficient mice and followed clonal composition for up to 15 months by whole-exome sequencing of 84 xenografts across two generations. We demonstrate vast changes in clonality that both progress and reverse over time, and define five patterns of clonal dynamics: Monoclonal, Stable, Loss, Expansion and Burst. We also show that subclonal expansion in vivo correlates with a more adverse prognosis. Furthermore, clonal expansion enabled detection of very rare clones with AML driver mutations that were undetectable by sequencing at diagnosis, demonstrating that the vast majority of AML cases harbor multiple clones already at diagnosis. Finally, the rise and fall of related clones enabled deconstruction of the complex evolutionary hierarchies of the clones that compete to shape AML over time.

    • Mutated Nucleophosmin 1 (NPM1) is the most common genetic alteration in acute myeloid leukemia (AML), found in approximately 30% of cases. Although mutations in this gene are considered favorable according to current risk stratification guidelines, a large fraction of patients will suffer from relapse, demonstrating the urgent need for new treatment options. Therefore, we aimed to identify cell surface proteins specifically expressed on NPM1)-mutated AML cells, allowing for potential targeting with antibody-based therapies. Herein, we performed an arrayed flow cytometry-based screen directed to 362 cell surface markers. Comparing the cell surface expression on NPM1-mutated AML cells to primitive (CD34+ CD38-) normal bone marrow cells, we identified the complement receptor C3AR as specifically expressed in NPM1-mutated AML. By flow cytometry and single cell RNA-sequencing, we further show that normal hematopoietic stem and progenitor cells lack detectable C3AR gene and protein expression, making it particularly suitable as a target for antibody therapy. We also demonstrate that C3AR in combination with GPR56 distinguishes the leukemic stem cells (LSCs) in NPM1-mutated AML from the normal hematopoietic stem cells, defining the LSC population, as shown by transplantation into immunodeficient mice. Mechanistically, stimulation of C3AR-expressing cells with C3a, the ligand of C3AR, leads to activation of ERK1/2 and increased survival of AML cells, suggesting that this is an important signaling axis in this subtype of AML. Finally, we show that antibodies directed against C3AR efficiently elicit NK cell-mediated killing of primary AML cells ex vivo, highlighting C3AR as a candidate therapeutic target in NPM1-mutated AML.
