  • Precision electroweak measurements on the Z resonance

    • We report on the final electroweak measurements performed with data taken at the Z resonance by the experiments operating at the electron-positron colliders SLC and LEP. The data consist of 17 million Z decays accumulated by the ALEPH, DELPHI, L3 and OPAL experiments at LEP, and 600 thousand Z decays by the SLID experiment using a polarised beam at SLC. The measurements include cross-sections, forward-backward asymmetries and polarised asymmetries. The mass and width of the Z boson, m(Z) and Gamma(Z), and its couplings to fermions, for example the p parameter and the effective electroweak mixing angle for leptons, are precisely measured: m(Z) = 91.1875 +/- 0.0021 GeV, Gamma(Z) = 2.4952 +/- 0.0023 GeV, rho(l) = 1.0050 +/- 0.0010, sin(2)theta(eff)(lept) = 0.23153 +/- 0.00016. The number of light neutrino species is determined to be 2.9840 +/- 0.0082, in agreement with the three observed generations of fundamental fermions. The results are compared to the predictions of the Standard Model (SM). At the Z-pole, electroweak radiative corrections beyond the running of the QED and QCD coupling constants are observed with a significance of five standard deviations, and in agreement with the Standard Model. Of the many Z-pole measurements, the forward-backward asymmetry in b-quark production shows the largest difference with respect to its SM expectation, at the level of 2.8 standard deviations. Through radiative corrections evaluated in the framework of the Standard Model, the Z-pole data are also used to predict the mass of the top quark, m(t) = 173(+10)(+13) GeV, and the mass of the W boson, m(W) = 80.363 +/- 0.032 GeV. These indirect constraints are compared to the direct measurements, providing a stringent test of the SM. Using in addition the direct measurements of m(t) and m(W), the mass of the as yet unobserved SM Higgs boson is predicted with a relative uncertainty of about 50% and found to be less than 285 GeV at 95% confidence level.   • Electroweak measurements in electron positron collisions at W-boson-pair energies at LEP

    • Electroweak measurements performed with data taken at the electron positron collider LEP at CERN from 1995 to 2000 are reported. The combined data set considered in this report corresponds to a total luminosity of about 3 fb(-1) collected by the four LEP experiments ALEPH, DELPHI, 13 and OPAL, at centre-of-mass energies ranging from 130 GeV to 209 GeV. Combining the published results of the four LEP experiments, the measurements include total and differential cross-sections in photon-pair, fermion-pair and four-fermion production, the latter resulting from both double-resonant WW and ZZ production as well as singly resonant production. Total and differential cross-sections are measured precisely, providing a stringent test of the Standard Model at centre-of-mass energies never explored before in electron positron collisions. Final-state interaction effects in four-fermion production, such as those arising from colour reconnection and Bose Einstein correlations between the two W decay systems arising in WW production, are searched for and upper limits on the strength of possible effects are obtained. The data are used to determine fundamental properties of the W boson and the electroweak theory. Among others, the mass and width of the W boson, m(w) and Gamma(w), the branching fraction of W decays to hadrons, B(W -> had), and the trilinear gauge-boson self-couplings g(1)(Z), K-gamma and lambda(gamma), are determined to be: m(w) = 80.376 +/- 0.033 GeV Gamma(w) = 2.195 +/- 0.083 GeV B(W -> had) = 67.41 +/- 0.27% g(1)(Z) = 0.984(-0.020)(+0.018) K-gamma - 0.982 +/- 0.042 lambda(gamma) = 0.022 +/- 0.019.     • A direct search has been made for magnetic monopoles produced in e(+)p collisions at a centre of mass energy of 300 GeV at HERA. The beam pipe surrounding the interaction region in 1995-1997 was investigated using a SQUID magnetometer to look for stopped magnetic monopoles. During this time an integrated luminosity of 62 pb(-1) was delivered. No magnetic monopoles were observed and charge and mass dependent upper limits on the e(+)p production cross section are set.

    • Cross sections for the production of two isolated muons up to high di-muon masses are measured in ep collisions at HERA with the H1 detector in a data sample corresponding to an integrated luminosity of 71 pb(-1) at a centre of mass energy of roots = 319 GeV. The results are in good agreement with Standard Model predictions, the dominant process being photon-photon interactions. Additional muons or electrons are searched for in events with two high transverse momentum muons using the full data sample corresponding to 114 pb(-1), where data at roots = 301 GeV and roots = 319 GeV are combined.   • A general search for new phenomena in ep scattering at HERA

    • A model-independent search for deviations from the Standard Model prediction is performed in e(+) p and e(-) p collisions at HERA using H1 data corresponding to an integrated luminosity of 117 pb(-1). For the first time all event topologies involving isolated electrons, photons, muons, neutrinos and jets with high transverse momenta are investigated in a single analysis. Events are assigned to exclusive classes according to their final state. A statistical algorithm is developed to search for deviations from the Standard Model in the distributions of the scalar sum of transverse momenta or invariant mass of final state particles and to quantify their significance. A good agreement with the Standard Model prediction is observed in most of the event classes. The most significant deviation is found for a topology containing an isolated muon, missing transverse momentum and a jet, consistent with a previously reported observation.

    • A narrow resonance in D*(-)p and D*(+)(p) over bar invariant mass combinations is observed in inelastic electron-proton collisions at centre-of-mass energies of 300 GeV and 320 GeV at HERA. The resonance has a mass of 3099 +/- 3(stat.) +/- 5(syst.) MeV and a measured Gaussian width of 12 +/- 3(stat.) MeV, compatible with the experimental resolution. The resonance is interpreted as an anti-charmed baryon with a minimal constituent quark composition of uudd (c) over bar, together with the charge conjugate. (C) 2004 Elsevier B.V.     • Inclusive production cross sections are measured in deep inelastic scattering at HERA for meson states composed of a charm quark and a light antiquark or the charge conjugate. The measurements cover the kinematic region of photon virtuality 2 < Q(2) < 100 GeV2, inelasticity 0.05 < y < 0.7, D meson transverse momenta p(t)( D) greater than or equal to 2.5 GeV and pseudorapidity |eta( D)| less than or equal to 1.5. The identification of the D-meson decays and the reduction of the combinatorial background profit from the reconstruction of displaced secondary vertices by means of the H1 silicon vertex detector. The production of charmed mesons containing the light quarks u, d and s is found to be compatible with a description in which the hard scattering is followed by a factorisable and universal hadronisation process.

  • Measurement of anti-deuteron photoproduction and a search for heavy stable charged particles at HERA

    • The cross section for anti-deuteron photoproduction is measured at HERA at a mean centre-of-mass energy of W-gammap=200 GeV in the range 0.2 < p(T)/M < 0.7 and y < 0.4, where M, p(T) and y are the mass, transverse momentum and rapidity of the anti-deuteron in the HERA laboratory frame, respectively. The numbers of anti-deuterons per event are found to be similar in photoproduction to those in central proton-proton collisions at the CERN ISR but much lower than those in central Au-Au collisions at RHIC. The coalescence parameter B-2, which characterizes the likelihood of anti-deuteron production, is measured in photoproduction to be 0.010+/-0.002+/-0.001, which is much higher than in Au-Au collisions at a similar nucleon-nucleon centre-of-mass energy. No significant production of particles heavier than deuterons is observed and upper limits are set on the photoproduction cross sections for such particles.

    • Triple differential dijet cross sections in e(+/-)p interactions are presented in the region of photon virtualities 2 < Q(2) < 80 GeV2, inelasticities 0.1 < y < 0.85, jet transverse energies E*(T1) > 7GeV, E*(T2) > 5 GeV, and pseudorapidities - 2.5 < η(1)*, η(2)* < 0. The measurements are made in the gamma*p centre-of-mass frame, using an integrated luminosity of 57 pb(-1). The data are compared with NLO QCD calculations and LO Monte Carlo programs with and without a resolved virtual photon contribution. NLO QCD calculations fail to describe the region of low Q(2) and low jet transverse energies, in contrast to a LO Monte Carlo generator which includes direct and resolved photon interactions with both transversely and longitudinally polarised photons. Initial and final state parton showers are tested as a mechanism for including higher order QCD effects in low E-T jet production.

  • Measurement of prompt photon cross sections in photoproduction at HERA

    • Results are presented on the photoproduction of isolated prompt photons, inclusively and associated with jets, in the gammap center of mass energy range 142 < W < 266 GeV. The cross sections are measured for the transverse momentum range of the photons 5 < E. T < 10 GeV and for associated jets with E-T(jet) > 4.5 GeV. They are measured differentially as a function of E-T(gamma), E-T(jet), the pseudorapidities eta(gamma) and eta(jet) and estimators of the momentum fractions x(gamma) and x(p) of the incident photon and proton carried by the constituents participating in the hard process. In order to further investigate the underlying dynamics, the angular correlation between the prompt photon and the jet in the transverse plane is studied. Predictions by perturbative QCD calculations in next to leading order are about 30% below the inclusive prompt photon data after corrections for hadronisation and multiple interactions, but are in reasonable agreement with the results for prompt photons associated with jets. Comparisons with the predictions of the event generators PYTHIA and HERWIG are also presented.

QED Compton events are used to access the kinematic range of very low virtualities of the exchanged photon, Q(2), down to 0.5 GeV2, and Bjorken x up to similar to 0.06, a region which has not been covered previously by inclusive measurements at HERA.     • A search for scalar top quarks in R-parity violating supersymmetry is performed in e(+)p collisions at HERA using the H1 detector. The data, taken at roots = 319 GeV and 301 GeV, correspond to an integrated luminosity of 106 pb(-1). The resonant production of scalar top quarks t in positron quark fusion via an R-parity violating Yukawa coupling lambda' is considered with the subsequent bosonic stop decay t --> BW. The R-parity violating decay of the sbottom quark b --> dv(e) and leptonic and hadronic W decays are considered. No evidence for stop production is found in the search for bosonic stop decays nor in a search for the direct R-parity violating decay t --> eq. Mass dependent limits on lambda' are obtained in the framework of the minimal supersymmetric Standard Model. Stop quarks with masses up to 275 GeV can be excluded at the 95% confidence level for a Yukawa coupling of electromagnetic strength. (C) 2004 Elsevier B.V. All rights reserved.

    • A search for gravitinos produced in e(+/-)p collisions is performed using the H1 detector at HERA. The data were taken at a centre-of-mass energy of 319 GeV and correspond to an integrated luminosity of 64.3 pb(-1) for e(+)p collisions and 13.5 pb(-1) for e(-)p collisions. If R-parity is not conserved, the t-channel exchange of a selectron can produce a neutralino, which, in models where the gravitino is the lightest supersymmetric particle, subsequently decays into a photon and a light gravitino. The resulting event signature, which involves an isolated photon, a jet and missing transverse energy, is analysed for the first time at HERA. No deviation from the Standard Model is found. Exclusion limits on the cross section and on R-parity-violating Yukawa couplings are derived in a Gauge Mediated Supersymmetry Breaking scenario. The results are independent of the squark sector. Neutralinos and supersymmetric partners of the left-handed electron with masses up to 112 GeV and 164 GeV, respectively, can be ruled out at the 95% confidence level for R-parity-violating couplings lambda' equal to 1, in some parts of the parameter space of the considered model.

The resonant production of squarks via a Yukawa coupling lambda' is considered, taking into account direct and indirect R-parity violating decay modes. No evidence for squark production is found in the multi-lepton and multi-jet final state topologies investigated. Mass dependent limits on lambda' are obtained in the framework of the Minimal Supersymmetric Standard Model. In addition, the results are interpreted in terms of constraints on the parameters of the minimal Supergravity model. At the 95% confidence level squarks of all flavours with masses up to 275 GeV are excluded in a large part of the parameter space for a Yukawa coupling of electromagnetic strength. For a coupling strength 100 times smaller, masses up to 220 GeV can be ruled out.

    • Measurements are presented of inclusive charm and beauty cross sections in e(+)p collisions at HERA for values of photon virtuality Q(2) > 150 GeV2 and of inelasticity 0.1 < y < 0.7. The charm and beauty fractions are determined using a method based on the impact parameter, in the transverse plane, of tracks to the primary vertex, as measured by the H1 vertex detector. The data are divided into four regions in Q(2) and Bjorken x, and values for the structure functions F-2(c (c) over bar) and F-2(b (b) over bar) are obtained. The results are found to be compatible with the predictions of perturbative quantum chromodynamics.

  • Measurement of dijet cross sections in ep interactions with a leading neutron at HERA

    • Measurements are reported of the production of dijet events with a leading neutron in ep interactions at HERA. Differential cross sections for photoproduction and deep inelastic scattering are presented as a function of several kinematic variables. Leading order QCD simulation programs are compared with the measurements. Models in which the real or virtual photon interacts with a parton of an exchanged pion are able to describe the data. Next-to-leading order perturbative QCD calculations based on pion exchange are found to be in good agreement with the measured cross sections. The fraction of leading neutron dijet events with respect to all dijet events is also determined. The dijet events with a leading neutron have a lower fraction of resolved photon processes than do the inclusive dijet data.

    • Over the past few decades, neuroimaging has become a ubiquitous tool in basic research and clinical studies of the human brain. However, no reference standards currently exist to quantify individual differences in neuroimaging metrics over time, in contrast to growth charts for anthropometric traits such as height and weight1. Here we assemble an interactive open resource to benchmark brain morphology derived from any current or future sample of MRI data (http://www.brainchart.io/). With the goal of basing these reference charts on the largest and most inclusive dataset available, acknowledging limitations due to known biases of MRI studies relative to the diversity of the global population, we aggregated 123,984 MRI scans, across more than 100 primary studies, from 101,457 human participants between 115 days post-conception to 100 years of age. MRI metrics were quantified by centile scores, relative to non-linear trajectories2 of brain structural changes, and rates of change, over the lifespan. Brain charts identified previously unreported neurodevelopmental milestones3, showed high stability of individuals across longitudinal assessments, and demonstrated robustness to technical and methodological differences between primary studies. Centile scores showed increased heritability compared with non-centiled MRI phenotypes, and provided a standardized measure of atypical brain structure that revealed patterns of neuroanatomical variation across neurological and psychiatric disorders. In summary, brain charts are an essential step towards robust quantification of individual variation benchmarked to normative trajectories in multiple, commonly used neuroimaging phenotypes.

    • Microorganisms in marine subsurface sediments substantially contribute to global biomass. Sediments warmer than 40 degrees C account for roughly half the marine sediment volume, but the processes mediated by microbial populations in these hard-to-access environments are poorly understood. We investigated microbial life in up to 1.2-kilometer-deep and up to 120 degrees C hot sediments in the Nankai Trough subduction zone. Above 45 degrees C, concentrations of vegetative cells drop two orders of magnitude and endospores become more than 6000 times more abundant than vegetative cells. Methane is biologically produced and oxidized until sediments reach 80 degrees to 85 degrees C. In 100 degrees to 120 degrees C sediments, isotopic evidence and increased cell concentrations demonstrate the activity of acetate-degrading hyperthermophiles. Above 45 degrees C, populated zones alternate with zones up to 192 meters thick where microbes were undetectable.

    • Plasma pTau181 concentrations are elevated specifically in patients diagnosed with Alzheimer's disease compared to those diagnosed with frontotemporal lobar degeneration or elderly controls, supporting its further development as a blood-based biomarker for AD. With the potential development of new disease-modifying Alzheimer's disease (AD) therapies, simple, widely available screening tests are needed to identify which individuals, who are experiencing symptoms of cognitive or behavioral decline, should be further evaluated for initiation of treatment. A blood-based test for AD would be a less invasive and less expensive screening tool than the currently approved cerebrospinal fluid or amyloid beta positron emission tomography (PET) diagnostic tests. We examined whether plasma tau phosphorylated at residue 181 (pTau181) could differentiate between clinically diagnosed or autopsy-confirmed AD and frontotemporal lobar degeneration. Plasma pTau181 concentrations were increased by 3.5-fold in AD compared to controls and differentiated AD from both clinically diagnosed (receiver operating characteristic area under the curve of 0.894) and autopsy-confirmed frontotemporal lobar degeneration (area under the curve of 0.878). Plasma pTau181 identified individuals who were amyloid beta-PET-positive regardless of clinical diagnosis and correlated with cortical tau protein deposition measured by F-18-flortaucipir PET. Plasma pTau181 may be useful to screen for tau pathology associated with AD.

    • An ambidentate dicarboxylic acid bipyridine ligand, (4,5-diazafluoren-9-ylidene) malonic acid (dfm), was synthesized for coordination to Ru(II) and mesoporous nanocrystalline (anatase) TiO2 thin films. The dim ligand provides a conjugated pathway from the pyridyl rings to the carbonyl carbons of the carboxylic acid groups. X-ray crystal structures of [Ru(bpy)(2)(dfm)]Cl-2 and the corresponding diethyl ester compound, [Ru(bpy)(2)(defm)](PF6)(2), were obtained. The compounds displayed intense metal-to-ligand charge transfer (MLCT) absorption bands in the visible region (epsilon > 11,000 M-1 cm(-1) for [Ru(bpy)(2)(dfm)](PF6)(2) in acetonitrile). Significant room temperature photo-luminescence, PL, was absent in CH3CN but was observed at 77 K in a 4:1 EtOH:MeOH (v:v) glass. Cyclic voltammetry measurements revealed quasi-reversible Ru-III/(II) electrochemistry. Ligand reductions were quasi-reversible for the diethyl ester compound [Ru(bpy)(2)(defm)](2+), but were irreversible for [Ru(bpy)(2)(dfm)](2+). Both compounds were anchored to TiO2 thin films by overnight reactions in CH3CN to yield saturation surface coverages of 3 X 10(-8) mol/cm(2). Attenuated total reflection infrared measurements revealed that the [Ru(bpy)(2)(dfm)](2+) compound was present in the deprotonated carboxylate form when anchored to the TiO2 surface. The MLCT excited states of both compounds injected electrons into TiO2 with quantum yields of 0.70 in 0.1 M LiClO4 CH3CN. Micro- to milli- second charge recombination yielded ground state products. In regenerative solar cells with 0.5 M LiI/0.05 M I-2 in CH3CN, the Ru(bpy)(2)(dfm)/TiO2 displayed incident photon-to-current efficiencies of 0.7 at the absorption maximum. Under the same conditions, the diethylester compound was found to rapidly desorb from the TiO2 surface.

    • Objective: To study the effects of different doses of platelet activating factor receptor antagonist WEB 2170 on animal survival, haemodynamics, reperfusion and ultrastructural changes in the ischaemic liver in rats undergoing 90-min total hepatic inflow interruption (THII). Design: Sixty-five rats were divided into five groups. All animals underwent 90-min THII. Group 1 served as controls. Group 2 underwent THII alone. Group 3 received an intravenous injection of 1 mg/kg WEB 2170 prior to THII. Group 4 received a bolus injection of 3 mg/kg WEB 2170 before THII. Group 4 received a bolus injection of 3 mg/kg WEB 2170 before THII. Group 5 received 3 mg/kg WEB 2170 before, during and after THII. The liver reperfusion index using laser Doppler flowmetry, the time of ischaemic liver initiative reperfusion, and scanning electron microscopy were performed to evaluate the results of different dose schedules. Setting: Lund University Hospital, Lund, Sweden. Results: Animal survival rate, liver reperfusion index, the time of ischaemic liver initiative reperfusion and ultrastructural damage of the ischaemic liver were markedly improved in the groups treated with WEB 2170 compared with the non-treated 90-min THII group. The best result was obtained in the group receiving the three separate doses. Conclusion: In the 90-min THII model, WEB 2170 protects the liver from ischaemia-reperfusion injury and the spread of damage to the post-stasis splanchnic organs. These beneficial effects may be extended to hepatic transplantation or major resections of the liver.

    • Plasma tau phosphorylated at threonine 217 (p-tau217) and plasma tau phosphorylated at threonine 181 (p-tau181) are associated with Alzheimer's disease tau pathology. We compared the diagnostic value of both biomarkers in cognitively unimpaired participants and patients with a clinical diagnosis of mild cognitive impairment, Alzheimer's disease syndromes, or frontotemporal lobar degeneration (FTLD) syndromes.In this retrospective multicohort diagnostic performance study, we analysed plasma samples, obtained from patients aged 18-99 years old who had been diagnosed with Alzheimer's disease syndromes (Alzheimer's disease dementia, logopenic variant primary progressive aphasia, or posterior cortical atrophy), FTLD syndromes (corticobasal syndrome, progressive supranuclear palsy, behavioural variant frontotemporal dementia, non-fluent variant primary progressive aphasia, or semantic variant primary progressive aphasia), or mild cognitive impairment; the participants were from the University of California San Francisco (UCSF) Memory and Aging Center, San Francisco, CA, USA, and the Advancing Research and Treatment for Frontotemporal Lobar Degeneration Consortium (ARTFL; 17 sites in the USA and two in Canada). Participants from both cohorts were carefully characterised, including assessments of CSF p-tau181, amyloid-PET or tau-PET (or both), and clinical and cognitive evaluations. Plasma p-tau181 and p-tau217 were measured using electrochemiluminescence-based assays, which differed only in the biotinylated antibody epitope specificity. Receiver operating characteristic analyses were used to determine diagnostic accuracy of both plasma markers using clinical diagnosis, neuropathological findings, and amyloid-PET and tau-PET measures as gold standards. Difference between two area under the curve (AUC) analyses were tested with the Delong test.Data were collected from 593 participants (443 from UCSF and 150 from ARTFL, mean age 64 years [SD 13], 294 [50%] women) between July 1 and Nov 30, 2020. Plasma p-tau217 and p-tau181 were correlated (r=0·90, p<0·0001). Both p-tau217 and p-tau181 concentrations were increased in people with Alzheimer's disease syndromes (n=75, mean age 65 years [SD 10]) relative to cognitively unimpaired controls (n=118, mean age 61 years [SD 18]; AUC=0·98 [95% CI 0·95-1·00] for p-tau217, AUC=0·97 [0·94-0·99] for p-tau181; pdiff=0·31) and in pathology-confirmed Alzheimer's disease (n=15, mean age 73 years [SD 12]) versus pathologically confirmed FTLD (n=68, mean age 67 years [SD 8]; AUC=0·96 [0·92-1·00] for p-tau217, AUC=0·91 [0·82-1·00] for p-tau181; pdiff=0·22). P-tau217 outperformed p-tau181 in differentiating patients with Alzheimer's disease syndromes (n=75) from those with FTLD syndromes (n=274, mean age 67 years [SD 9]; AUC=0·93 [0·91-0·96] for p-tau217, AUC=0·91 [0·88-0·94] for p-tau181; pdiff=0·01). P-tau217 was a stronger indicator of amyloid-PET positivity (n=146, AUC=0·91 [0·88-0·94]) than was p-tau181 (n=214, AUC=0·89 [0·86-0·93]; pdiff=0·049). Tau-PET binding in the temporal cortex was more strongly associated with p-tau217 than p-tau181 (r=0·80 vs r=0·72; pdiff<0·0001, n=230).Both p-tau217 and p-tau181 had excellent diagnostic performance for differentiating patients with Alzheimer's disease syndromes from other neurodegenerative disorders. There was some evidence in favour of p-tau217 compared with p-tau181 for differential diagnosis of Alzheimer's disease syndromes versus FTLD syndromes, as an indication of amyloid-PET-positivity, and for stronger correlations with tau-PET signal.     • Two series of Ru-II polypyridyl compounds with formulas [(bpy)(2)RuL](PF6)(2) and [(deeb)(2)RuL](PF6)(2), where bpy is 2,2-bipyridine, deeb is 4,4-diethylester-2,2-bpy, and L is one of several substituted 9-(1,3-dithiole-2-ylidene)-4,5-diazafluorene ligands, were studied as potential photosensitizers for TiO2. These compounds possess notably high extinction coefficients (40000M(-1)cm(-1) @470 nm) which are shown by time-dependent density functional theory (TD-DFT) calculations to result from overlapping metal-to-ligand charge transfer (MLCT) and ligand-localized transitions. Low-temperature absorption and photoluminescence measurements were suggestive of a short-lived MLCT excited state. When adsorbed onto TiO2 thin films, both the free ligands (L) and their corresponding [(deeb)(2)RuL](2+) complexes exhibited rapid excited-state electron injection into TiO2; in the case of the complexes, this was followed by rapid (k>10(8) s(-1)) hole transfer from Ru-III to the 1,3-dithiole ring of the L ligand. Observation of diffusion-limited reductive quenching of the [Ru(bpz)(3)](2+)* (bpz is 2,2-bipyrazine) excited state by the L ligands in solution supported the occurrence of intramolecular hole transfer following electron injection by the TiO2-anchored complexes.

    • Background: Cortical α-synuclein pathology plays a role in the development of cognitive dysfunction in both Parkinson's disease and dementia with Lewy bodies, although the causative cellular lesions have remained unclear. We aimed to address causal links between α-synuclein-driven pathology in the cerebral cortex and the development of cognitive impairments using new experimental models. Methods: Neuronal overexpression of human α-synuclein was induced in the rat medial prefrontal cortex using viral vectors. This was combined with inoculations of preformed fibrils of human α-synuclein in some animals. Rats were evaluated with tests probing prefrontal cognitive functions (delayed matching/nonmatching to position and 5-choice serial reaction time task). Patterns of neuropathology were characterized immunohistochemically. Results: Neither α-synuclein overexpression nor the fibril seeds alone yielded any behavioral phenotype. In contrast, combining the 2 approaches produced significant impairments in working memory, attention, and inhibitory control. All animals injected with α-synuclein vectors exhibited high immunoreactivity for human α-synuclein in the medial prefrontal cortex and its primary projection targets. However, only when this overexpression was combined with fibril inoculations did animals exhibit large, proteinase K-resistant and Ser129-phosphorylated α-synuclein intraneuronal inclusions in the medial prefrontal cortex and its closely interconnected brain regions. The inclusions were associated with distorted dendritic morphologies and partial neuronal loss in the targeted cortical areas. Conclusions: Cortical overexpression of human α-synuclein is not sufficient to produce cognitive dysfunction, whereas combining this overexpression with fibril seeds yields both cognitive and histopathological phenotypes that are relevant to human Lewy body disease.

    • The report is the result of the external Panel ex post evaluation of the JRC activities under H2020 and Euratom 2014-2020. It provides the independent assessment requested in the Council Decisions concerning the specific programmes to be carried out by means of direct actions by the Joint Research Centre implementing the Horizon 2020 Framework Programme (2014-2020) of the European Commission and of the European Atomic Energy Community (Euratom). The evaluation has been conducted by a panel of independent external experts under the chairmanship of Dr Rolf-Dieter Heuer. In this report the Panel concludes positively on the effectiveness of the JRC as the Commission’s science service in support of Euratom and EU policies. Besides a number of recommendations for incremental improvement of the JRC, the Panel has flagged that the JRC is in a unique position as a provider of independent scientific evidence inside the European Commission, but, because of this, the JRC and its research work are less visible to the outside world than they merit. The Panel also flags that the JRC and its stakeholders, internal and external to the Commission, would also benefit from more communication and interactions. The Panel has particularly appreciated the meetings with the stakeholders that gave much insight into the cooperation between the JRC and the other parts of the Commission, supporting our positive assessment and our suggestions for improvement.

    • This report provides an assessment of the European Commission's Joint Research Centre (JRC) under Horizon Europe and the Euratom research and training programme 2021-2025, prepared by a Panel of nine independent experts for the two programme parts separately, with common conclusions and observations on the JRC's performance and future direction as a whole. The Panel's assessment will support the evaluation of the JRC's contribution to the overall research programmes and as the European Commission's in-house science service. The assessment follows almost immediately after the ex post evaluation was completed in 2022. Thus the Panel was asked to evaluate activities under the current research programmes building on the results of the previous programmes. In line with its mandate, this Panel has also looked into what changes the JRC has implemented and what effects they have already had or are expected to have, and what could be further developed to enhance the agility, impact and efficiency of the JRC’s policy support and to ensure EU decision-makers get the best possible scientific advice also in the future.

    • Background: Thyroid hormones (TH) are essential for brain development and function. The TH transporters monocarboxylate transporter 8 (MCT8) and organic anion transporter1 C1 (OATP1C1) facilitate the transport of TH across the blood-brain barrier and into glia and neuronal cells in the brain. Loss of MCT8 function causes Allan-Herndon-Dudley syndrome (AHDS, OMIM 300523) characterized by severe intellectual and motor disability due to cerebral hypothyroidism. Here, the first patient with loss of OATP1C1 function is described. The patient is a 15.5-year-old girl with normal development in the first year of life, who gradually developed dementia with spasticity and intolerance to cold. Brain imaging demonstrated gray and white matter degeneration and severe glucose hypometabolism. Methods: Exome sequencing of the patient and parents was performed to identify the disease-causing mutation, and the effect of the mutation was studied through a panel of in vitro experiments, including thyroxine uptake studies, immunoblotting, and immunocytochemistry. Furthermore, the clinical effects of treatment with the triiodothyronine analogue triiodothyroacetic acid (Triac) are described. Results: Exome sequencing identified a homozygous missense mutation in OATP1C1, changing the highly conserved aspartic acid 252 to asparagine (D252N). In vitro, the mutated OATP1C1 displays impaired plasma membrane localization and decreased cellular thyroxine uptake. After treatment with Triac, the clinical condition improved in several domains. Conclusions: This is the first report of human OATP1C1 deficiency compatible with brain-specific hypothyroidism and neurodegeneration. © Copyright 2018, Mary Ann Liebert, Inc., publishers 2018.
